This paper describes the polymerization of 2-methyl-(3,6-di-O-benzyl- 1,2-dideoxy-α-D -glucopyrano)-[2,1-d]-2-oxazoline ( 1 ) with an acid catalyst. The polymerization proceeds involving stereoregular glycosylation to give polysaccharide 2 . The polymer structure, 2-acetamido-3,6-di-O-benzyl-2-deoxy-(l→4)-β-D -glucopyranan was determined by means of 1H NMR, 13C NMR, and IR spectra as well as elemental analysis. The molecular weight was at most 4900 (degree of polymerization ≈ 13).  相似文献

ABSTRACT

The stepwise synthesis of methyl α-D-glucopyranosyl-(1→3)-α-L-rhamnopyranosyl-(1→3)-α-L-rhamnopyranoside (EBC-OMe, 1), methyl α-L-rhamnopyranosyl-(1→2)-[α-D-glucopyranosyl-(1→3)]-α-L-rhamnopyranosyl-(1→3)-α-L-rhamnopyranoside (A(E)BC-OMe, 2), and methyl 2-acetamido-2-deoxy-β-D-glucopyranosyl-(1→2)-α-L-rhamnopyranosyl-(1→2)-[α-D-glucopyranosyl-(1→3)]-α-L-rhamnopyranosyl-(1→3)-α-L-rhamnopyranoside (DA(E)BC-OMe, 3) is described. Compounds 1, 2 and 3 constitute the methyl glycosides of fragments of the O-specific polysaccharide of Shigella flexneri serotype 5a. Methyl 2,4-di-O-benzoyl-α-L-rhamnopyranosyl-(1→3)-2,4-di-O-benzoyl-α-L-rhamnopyranoside was an appropriate BC precursor for the synthesis of 1. For the synthesis of the branched targets 2 and 3, a benzyl group was best suited at position 2 of rhamnose C. Thus, methyl 4-O-benzyl-α-L-rhamnopyranosyl-(1→3)-2,4-di-O-benzyl-α-L-rhamnopyranoside was the key intermediate to the BC portion. In all cases, 2,3,4,6-tetra-O-benzyl-α-D-glucopyranosyl fluoride was a convenient E precursor, when used in combination with titanium tetrafluoride. All along, attention was paid to steric hindrance as a factor of major impact on the condensation steps outcome. Therefore, based on previous experience, 2-O-acetyl-3,4-di-O-allyl-α-L-rhamnopyranosyl trichloroacetimidate and 3,4,6-tri-O-acetyl-2-deoxy-2-trichloroacetamido-α-D-glucopyranosyl trichloroacetimidate were used as donors. Both suited all requirements when used as key precursors for residues A and D in the synthesis of 3, respectively.  相似文献

Abstract

Using methyl 2,2-bis(ethylthio)propionate as acetalating agent and triflic acid-sulfuryl chloride as catalyst, synthesis of 2,3-trans diequatorial pyruvate ketal was achieved. Starting from D-galactose and L-rhamnose derivatives, methyl 2,3,4,6-tetra-O-benzyl-α-D-galactopyranosyl-(1→4)-6-O-benzyl-2,3-O-(1-methoxycarbonyl)ethylidene- α-D-galactopyranosyl-(1→3)-2,4-di-O-benzyl-α-L-rhamnopyranoside and methyl 4,6-di-O-benzyl-2,3-O-(1-methoxy-carbonyl)ethylidene-α-D-galactopyranosyl-(1→3)-2,4-di-O-benzyl-α-L-rhamnopyranoside were synthesized. Removal of the protecting groups from the former, afforded the trisaccharide repeating unit of the K-antigen from E.coli O101:K103:H? in the form of its methyl glycoside methyl ester.  相似文献

Abstract

A carboxylate-containing pentasaccharide, methyl O-(β-d-galactopyranosyl)-(1→4)-O-(β-d-glucopyranosyl)-(1→6)-O-{3-O-[(S)-1-carboxyethyl]-β-d-galactopyranosyl-(1→4)-O}-(2-acetamido-2-deoxy-β-d-glucopyranosyl)-(1→3)-β-d-galactopyranoside (27) was synthesized by block condensation of suitably protected donors and acceptors. Phenyl 3-O-benzyl-4,6-di-O-chloroacetyl-2-deoxy-2-phthalimido-1-thio-β-d-glucopyranoside (17) was condensed with methyl 2,4,6-tri-O-benzyl-β-d-galactopyranoside (4) to afford a disaccharide, methyl O-(3-O-benzyl-4,6-di-O-chloroacetyl-2-deoxy-2-phthalimido-β-d-glucopyranosyl)-(1→3)-2,4,6-tri-O-benzyl-β-d-galactopyranoside (18). Removal of chloroacetyl groups gave 4,6-diol, methyl 0-(3-O-benzyl-2-deoxy-2-phthalimido-β-d-glucopyranosyl)-(1→3)-2,4,6-tri-O-benzyl-β-d-galactopyranoside (19), in which the primary hydroxy group (6-OH) was then selectively chloroacetylated to give methyl O-(3-O-benzyl-6-O-chloroacetyl-2-deoxy-2-phthalimido-β-d-glucopyranosyl)-(1→3)-2,4,6-tri-O-benzyl-β-d-galactopyranoside (20). This acceptor was then coupled with 2,4,6-tri-O-acetyl-3-O-[(S)-1-(methoxycarbonyl)ethyl]-α-d-galactopyranosyl trichloroacetimidate (14) to afford a trisaccharide, methyl O-{2,4,6-tri-O-acetyl-3-O-[(S)-l-(methoxycarbonyl)ethyl]-β-d-galactopyranosyl}-(1→4)-O-(3-O-benzyl-6-O-chloroacetyl-2-deoxy-2-phthalimido-β-d-glucopyranosyl)-(1→3)-2,4,6-tri-O-benzyl-β-d-galactopyranoside (21). Removal of the 6-O-chloroacetyl group in 21 gave 22, which was coupled with 4-O-(2,3,4,6-tetra-O-acetyl-β-d-galactopyranosyl)-2,3,6-tri-O-acetyl-α-d-glucopyranosyl trichloroacetimidate (23) to yield protected pentasaccharide 24. Standard procedures were used to remove acetyl groups and the phthalimido group, followed by N-acetylation, and debenzylation to yield pentasaccharide 27 and a hydrazide by-product (28) in a 5:1 ratio, respectively. Compound 27 contains a complete repeating unit of the capsular polysaccharide of type III group B Streptococcus in which terminal sialic acid is replaced by an (S)-1-carboxyethyl group.  相似文献

A facile synthesis of the trisaccharide α-D-mannopyranosyl-(1→2)-α-D-mannopyranosyl-(1→6)-α-D-mannopyranose and the tetrasaccharide α-D-mannopyranosyl-(1→2)-α-D-mannopyranosyl-(1→6)-α-D-mannopyranosyl-(1→6)-D-mannopyranose, the repeating units of fungal cell-wall polysaccharide from Microsporum gypseum and Trychophyton, was achieved using α-(1→2)-linked disaccharide imidate as the donor. The disaccharide imidate was prepared from the self-condensation of 3,4,6-tri-O-benzoyl-1,2-O-allyloxyethylidene-β-D-mannopyranose.  相似文献

Four new triterpenoid saponins were isolated from the roots of Adina rubella Hance. They were characterized as adinaic acid 3β-O-[α-L-rhamnopyranosyl(l→2)-β-D-glucopyranosyl(l→2)-β-D-glucurono-pyranoside-6-O-methyl ester]-28-O-β-D)-glucopyranoside, adinaic acid 3β-O-[α-L-rham-nopyranosyl(l→2)-β-D-glucopyranosyl(l→2)-β-D-glucuronopyranoside-6-O-butyl ester]-28-O-β-D-glu-copyranoside, adinaic acid 3β-O-[β-D-glucopyranosyl(l→2)-β-D-glucopyranosyl]-(28→1)-β-D-gluco-pyranosyl(l→6)-β-D-glucopyranosyl ester, 27-hydroxyursolic acid 3β-O-[α-L-rhamnopyranosyl (l→2)-β-O-glucopyranosyl(l→2)-β-D)-glucuronopyranoside-6-O-methyl ester]-28-O-β-D)-glucopyranoside. Their structures were elucidated by spectral methods, especially with the aid of 2D NMR techniques. Their complete assignments of the 1H and 13C NMR signals were carried out.  相似文献

A branched hexasaccharide fragment of type Ia group B streptococcal polysaccharide, α-NeuAc(2→3)-β-D-Gal(1→4)-β-D-GlcNAc(1→3)-[β-D-Glc(1→4)]-β-D-Gal(1→4)-β-D-Glc-OMe (13), has been synthesized by chemical-enzymatic procedures. Chemical synthesis of a pentasaccharide, β-D-Gal(1→4)-β-D-GlcNAc(1→3)-[β-D-Glc(1→4)]-β-D-Gal(1→4)-β-D-Glc-OMe (12), was achieved from glycosyl donor, 4-O-(2,3,4,6-tetra-O-acetyl-β-D-galactopyranosyl)-3,6-di-O-acetyl-2-deoxy-2-phthalimido-β-D-glucopyranosyl trichloroacetimidate (9), and acceptor, methyl O-(2,3,4,6-tetra-O-acetyl-β-D-glucopyranosyl)-(1→4)-O-(2,6-di-O-benzyl-β-D-galactopyranosyl)-(1→4)-2,3,6-tri-O-benzyl-β-D-glucopyranoside (6), by block condensation in 41% yield. Following enzymatic sialylation of 12 at the 3-O-position of its terminal galactopyranosyl residue using recombinant α-(2→3)-sialyltransferase and CMP-NeuAc afforded 13 in 59% yield.  相似文献

A variety of sialyl-α-(2→3)-neolactotetraose (IV3NeuAcnLcOse4 or IV3NeuGcnLcOse4) derivatives (23, 31–37, 58–60) modified at C-2 of the GlcNAc residue have been synthesized. The phthalimido group at C-2 of GlcNAc in 2-(trimethylsilyl)ethyl (3,6-di-O-benzyl-2-deoxy-2-phthalimido-β-d-glucopyranosyl)-(1→3)-(2,4,6-tri-O-benzyl-β-d-galactopyranosyl)-(1→4)-2,3,6-tri-O-benzyl-β-d-glucopyranoside (5) was systematically converted to a series of acylamino groups, to give the per-O-benzylated trisaccharide acceptors (6–11). On the other hand, modification of the hydroxyl group at C-2 of the terminal Glc residue in 2-(trimethylsilyl)ethyl (4,6-O-benzylidene-β-d-glucopyranosyl)-(1→3)-(2,4,6-tri-O-benzyl-β-d-galactopyranosyl)-(1→4)-2,3,6-tri-O-benzyl-β-d-glucopyranoside (42) gave three different kinds of trisaccharide acceptors containing D-glucose (49), N-acetyl-d-mannosamine (50), and D-mannose (51) instead of the GlcNAc residue. Totally ten trisaccharide acceptors (5–11 and 49–51) were each coupled with sialyl-α-(2→3)-galactose donor 12 to afford the corresponding pentasaccharides (14–21 and 52–54) in good yields, respectively, which were then transformed into the target compounds. Acceptor specificity of the synthetic sialyl-α-(2→3)-neolactotetraose probes for the human α-(1→3)-fucosyltransferases, Fuc-TVII and Fuc-TVI, was examined.  相似文献

The stereocontrolled synthesis of methyl α-D-glucopyranosyl-(1→4)-α-L-rhamnopyranoside (EC, 1), methyl α-L-rhamnopyranosyl-(1→3)-[α-D-glucopyranosyl-(1→4)]-α-L-rhamnopyranoside (B(E)C, 3) and methyl α-D-glucopyranosyl-(1→4)-α-L-rhamnopyranosyl-(1→3)-2-acetamido-2-deoxy-β-D-glucopyranoside (ECD, 4) is described; these constitute the methyl glycosides of branched and linear fragments of the O-specific polysaccharide of Shigella flexneri serotype 2a. Emphasis was put on the construction of the 1,2-cis EC glycosidic linkage resulting in the selection of 2,3,4,6-tetra-O-benzyl-α-D-glucopyranosyl fluoride (8) as the donor. Condensation of methyl 2,3-O-isopropylidene-4-O-trimethylsilyl-α-L-rhamnopyranoside (11) and 8 afforded the fully protected αE-disaccharide 20, as a common intermediate in the synthesis of 1 and 3, together with the corresponding βE-anomer 21. Deacetalation and regioselective benzoylation of 20, followed by glycosylation with 2,3,4-tri-O-benzoyl-α-L-rhamnopyranosyl trichloroacetimidate (15) afforded the branched trisaccharide 25. Full deprotection of 20 and 25 afforded the targets 1 and 3, respectively. The corresponding βE-disaccharide, namely, methyl β-D-glucopyranosyl-(1→4)-α-L-rhamnopyranoside (βEC, 2) was prepared analogously from 21. Two routes to trisaccharide 4 were considered. Route 1 involved the coupling of a precursor to residue E and a disaccharide CD. Route 2 was based on the condensation of an appropriate EC donor and a precursor to residue D. The former route afforded a 1:2 mixture of the αE and βE condensation products which could not be separated, neither at this stage, nor after deacetalation. In route 2, the required αE-anomer was isolated at the disaccharide stage and transformed into 2,3,4,6-tetra-O-benzyl-α-D-glucopyranosyl-(1→4)-2,3-di-O-benzoyl-α-L-rhamnopyranosyl trichloroacetimidate (48) as the EC donor. Methyl 2-acetamido-2-deoxy-4,6-O-isopropylidene-β-D-glucopyran-oside (19) was preferred to its benzylidene analogue as the precursor to residue D. Condensation of 19 and 48 and stepwise deprotection of the glycosylation product afforded the target 4.  相似文献

Abstract

2-Azido-4-O-benzoyl-2,6-dideoxy-3-O-methyl-D-allopyranose, needed as one of the building blocks for construction of a novel cyclodextrin-like compound, was prepared in the form of crystalline β-anomer 6 from methyl 2-azido-4,6-O-benzylidene-2-deoxy-α-D-allopyranoside 1. As a model of α-glycosidation necessary for formation of a cyclic structure, 6 was converted into the corresponding β-glycosyl trichloroacetimidate and coupled with methyl 6-O-benzyl-2,3-di-O-methyl-α-D-glucopyranoside 8, giving α(1→4)-linked disaccharide derivative 9.  相似文献

The disaccharide methyl (4-O-benzoyl-3-O-benzyl-2-O-acetyl-α-L-rhamno pyranosyl)-(1→4)-(allyl 2,3-di-O-benzyl-β-D-galactopyranosid)uronate (13) was obtained in an excellent yield of 88% using methyl (allyl 2,3-di-O-benzyl-β-D-galactopyranosid)uronate ((12) as the glycosyl acceptor and a slight excess of the 1,2-di-O-acetyl-rhamnoglycosyl donor 5a. Disaccharide 13 is a key intermediate that can be used either as a glycosyl acceptor or glycosyl donor for the preparation of rhamnogalacturonan fragments. Here, introduction of the trichloroacetimidate function at the anomeric center gave the disaccharide glycosyl donor 28, which could be applied in a blockwise glycosylation reaction to form the L-Rha-α(1→4)-D-GalA-α(1→4)-D-GalA trisaccharide 29. Generally, on condition that no neighboring group effect influenced the reaction at the anomeric center of the α-trichloroacetimidate galacturonate glycosyl donors (20–22, 28), α-glycosidic linkages were nearly exclusively formed, except in the case of the 4-O-methylgalactopyranosyluronate 22.  相似文献

Abstract

1,6-Anhydro-2-deoxy-3,4-di-O-benzyl-2-phthalimido-β-d- glucopyranose (5) was synthesized from 1,6-anhydro-β-d-mannopyranose (1) in five steps. Compound 5 was polymerized under cationic conditions and selectively yielded glucosamine oligomers (degree of polymerization 5-7). Copolymerization of 5 with 1,6-anhydro-2,3,4-tri-O-benzyl-β-d-glucopyranose indicated the low reactivity of 5 with the active cation derived from 5. Deprotection of 2-deoxy-3,4-di-O-benzyl-2-phthalimido-(1→6)-β-d-glucopyranan (7) and N-acetylation gave 2-acetamido-2-deoxy-(1→6)-β-d-glucopyranan (9).  相似文献

A systematic study is presented for the most common methods used for the preparation of the disaccharide benzyl O-(3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-β-D-glucopyranosyl)-(1→'4)-3,6-di-O-benzoyl-2-deoxy-2-phthalimido-β-D-glucopyranoside (9) from “standard 2-amino-2-deoxyglucopyranosyl donors” 1-6 and benzyl 3,6-di-O-benzoyl-2-deoxy-2-phthalimido-β-D-glucopyranoside (7) as an acceptor. It was found that the highest yield was obtained when the trichloroacetimidate derivative 1 was coupled to the 4 position of acceptor 7. In an analogous manner, the disaccharides allyl O-(3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-β-D-glucopyranosyl)-(1→'4)-3,6,-di-O-benzoyl-2-deoxy-2-phthalimido-β-D-glucopyranoside (10), benzyl O-(3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-β-D-glucopyranosyl)-(1→'4)-3,6-di-O-benzoyl-2-deoxy-2-phthalimido-β-D-galactopyranoside (12), and allyl O-(3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-β-D-glucopyranosyl)-(1→'3)-4,6-O-benzylidene-2-deoxy-2-phthalimido-β-D-glucopyranoside (14) were prepared.  相似文献

ABSTRACT

The four derivatives of β-maltosyl-(1→4)-trehalose have been synthesized, which are monodeoxygenated at the site of one of the primary hydroxyl groups. The tetrasaccharides were constructed in [2+2] block syntheses. Thus, 6′″-deoxy-β-maltosyl-(1→4)-trehalose was prepared by selective iodination of allyl 2,3,6,2′,3′-penta-O-acetyl-β-maltoside (3) followed by catalytic hydrogenolysis and coupling with 2,3-di-O-benzyl-4,6-O-benzylidene-α-D-glucopyranosyl 2′,3′,6′-tri-O-benzyl-α-D-glucopyranoside (9), and 6″-deoxy-β-maltosyl-(1→4)-trehalose by selective iodination of allyl 4′,6′-O-isopropylidene-β-maltoside (14), coupling with 9, and one-step hydrogenolysis at the tetrasaccharide level. For the synthesis of 6′-deoxy-β-maltosyl-(1→4)-trehalose, the diol 2,3-di-O-benzyl-4,6-O-benzylidene-α-D-glucopyranosyl 2′,3′-di-O-benzyl-α-D-glucopyranoside (22) was selectively iodinated and glycosylated with acetobromomaltose followed by catalytic hydrogenolysis. The 6-deoxy-β-maltosyl-(1→4)-trehalose was obtained upon selective iodination of a tetrasaccharide diol.  相似文献

Polymerization 1,6-anhydro-2,3,4-tri-O-benzyl-β-D -mannopyranose at ?60°C with phosphorus pentafluoride (0.9 mole-%) gives stereoregular 2,3,4-tri-O-benzyl-[1 → 6]-α-D -mannopyranan with substantially higher viscosity ([η] = 2.8 dl/g) than the corresponding gluco- and glactopyranan derivatives prepared similarly. Debenzylation with sodium in liquid ammonia produces stereoregular [1 → 6]-α-D -mannopyranan of viscosity up to [η] = 0.54 dl/g. Stereoregular 2,3,4-tri-O-acetyl-[1 → 6]-α-D -glycopyranans are most simply prepared by acetylation of the corresponding crude [1 → 6]-α-D -glycopyranans obtained directly from the debenzylation reaction. The galactan is extremely difficult to acetylate by conventional methods if isolated in a pure form. Physical and spectral properties of these highly stereoregular synthetic 2,3,4-tri-O-acetyl-[1 → 6]-α-D -glycopyranans are presented. Optical rotary dispersion curves of 2,3,4, tri-O-acetyl-[1 → 6]-α-D -glycopyranans show small Cotton effects in the 200–230 nm region, superimposed on strong background rotation. Circular dichroism spectra show a single n →* acetate absorption band for each polymer. The sign of the band appears to be determined largely by the C-2 configuration. Stereoregular 2,3,4-tri-O-acetyl-[1 → 6]-α-D -glycopyranans in 2,2,2-trifluoroethanol solution are likely to possess a random rather than helical conformation.  相似文献

Two routes to trisaccharide 1 were considered. Route 1 involved the coupling of a precursor to residue A and a disaccharide EB, whereas route 2 was based on the condensation of a precursor to residue E and a disaccharide AB. Rather surprisingly, the latter afforded the β-anomer of 1, namely methyl α-L-rhamnopyranosyl-(1→2)-[β-D-glucopyranosyl-(1→3)]-α-L-rhamnopyranoside as the major product. Route 1 was preferred. Overall, several observations made during this study suggested that, for the construction of higher fragments, a suitable precursor to rhamnose A would require protecting groups of low bulkiness at position 3 and 4. Therefore, the 2-O-acetyl-3,4-di-O-allyl-α-L-rhamnopyranosyl trichloroacetimidate (35) was the precursor of choice to residue A in the synthesis of the tetrasaccharide 2. The condensation product of 35 and methyl 2,3,4,6-tetra-O-benzyl-α-D-glucopyranosyl-4-O-benzyl-α-L-rhamnopyranoside was selectively deacylated and condensed to 2-trichloroacetamido-3,4,6-tri-O-acetyl-2-deoxy-α-D-glucopyranosyl trichloroacetimidate to afford the corresponding fully protected tetrasaccharide 45. Controlled stepwise deprotection of the latter proceeded smoothly to afford the target 2. It should be emphasised that the preparation of 45 was not straightforward, several donors and coupling conditions that were tested resulted only in the complete recovery of the acceptor. Distortion of several signals in the 13C NMR spectra of the fully or partially protected tetrasaccharide intermediates suggested that steric hindrance, added to the known low reactivity of HO-2 of rhamnosyl acceptors, probably played a major role in the outcome of the glycosidation attempts.  相似文献

A first total synthesis of gangliosides GD1c and GT1a containing Neu5Acα(2→8) Neu5Acα(2→3)Gal residue in their non-reducing terminal is described. Condensation of methyl O-[methyl 5-acetamido-8-O-(5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-D-glycero-α-D-galacto-2-nonulopyranosylono-11,9-lactone) -4,7- di-O-acetyl-3,5-dideoxy-D-glycero-α-D-galcto-2-nonulopyranosyranosylanate]-(2→3)-2,4,6-tri-O-benzoyl-1-thio-β-D-gala-ctopyranoside (1) with 2-(trimethylsilyl)ethyl O-(2-acetamido-4,6-O-benzylidene-2-deoxy-β-D-galactopyranosyl)- (1→4) -O -(2,3,6-tri-O-benzyl-β-D-galactopyranosyl)-(1→4)-2,3,6-tri-O-benzyl-β-D-glucopyranoside (2) or 2-(trimethylsilyl)ethyl O-(2-acetamido-6-O-benzyl-2-deoxy-β-D-galactopyranosyl)-(1→4)-(9-[methyl 5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-D-glycero-α-D-galacto-2-nonulopyranosylonate)-(2→3)]-O-(2,6-di-O-benzyl-β-D-galactopyranosyl) - (1→4) - 2,3,6-tri-O-benzyl-β-D-glucopyranoside (3) in the presence of dimethyl(methylthio)sulfonium triflate (DMTST) gave the corresponding hexa-and heptasaccharide derivatives 4 and 5, respectively. These oligosaccharides were converted into the α-trichloroacetimidates 10 and 11 via reductive removal of the benzyl groups and/or benzylidene group, O-acetylation, selective removal of the 2-(trimethylsilyl)ethyl group and treatment with trichloroacetonitrile, which, on coupling with 2-azidosphingosine derivatives 12 or 13, gave the β-glycosides 14 and 15, respectively. Finally, 14 and 15 were transformed, via selective reduction of the azido group, coupling with octadecanoic acid and removal of all protecting groups, into the title gangliosides GD1c 18 and GT1a 19.  相似文献

A tetrasaccharide, α-D-mannopyranosyl-(1→2)-α-D-mannopyranosyl-(1→6)-α-D-mannopyranosyl-(1→6)-D-mannopyranose (1), the repeating unit of the cell-wall mannans of Microsporum gypseum and related species of Trychophyton, was synthesized using 6-O-acetyl-2,3,4-tri-O-benzoyl-α-D-mannopyranosyl trichloroacetimidate (5) and 2-O-acetyl-3,4,6-tri-O-benzoyl-α-D-mannopyranosyl trichloroacetimidate (13) as the glycosyl donors in “the inverse Schmidt” procedure.  相似文献

ABSTRACT

The partially deprotected trisaccharide 17 has been synthesized as an analogue of the repeating unit of the backbone of rhamnogalacturonan I. The trisaccharide 17 was obtained after prior selective derivatization of HO-3 and HO-4 of a rhamnopyranose cyanoethylidene glycosyl donor, followed by coupling with a tritylated galactopyranosyluronic acceptor (11), selective removal of the acetyl group at the O-2' position of the formed disaccharide 12, and glycosylation of the HO-2' position with methyl (ethyl 2,3-di-O-benzyl-4-O-p-methoxybenzyl-1-thio-β-D-galactopyranosid)uronate (14) providing methyl (methyl 2,3-di-O-benzyl-4-O-p-methoxybenzyl-α-D-galactopyranosyluronate)-(1→2)-(4-O-benzoyl-3-O-benzyl-α-L-rhamnopyranosyl)-(1→4)-(allyl 2,3-di-O-benzyl-β-D-galactopyranosid)uronate (15). Finally, palladium chloride catalyzed deallylation (16) and hydrogenolysis over Pd-C resulted in methyl (methyl α-D-galactopyranosyluronate)-(1→2)-(4-O-benzoyl-α-L-rhamnopyranosyl)-(1→4)-α/β D-galactopyranuronate (17).  相似文献
