An Unusual Aminal Derivative from a Misdirected Gabriel Synthesis

N-(2-Bromoethyl)phthalimide (1) was reacted with sodium imidazolate in DMF to give the novel aminal N-[1-(1H-imidazol-1-yl)ethyl]phthalimide (4a) as well as N-vinylphthalimide (3) and the desired Gabriel intermediate 2. Aminal 4a as well as heterologues 4b - d form directly from reaction of 3 with the appropriate heterocyclic sodium salt.

以对叔丁基苯酚为起始原料,把酯基和具有荧光性质的喹啉环引入杯芳烃大环分子中,通过缩合,两步取代反应,获得了具有荧光性质的新型含喹啉环杯[4]芳烃衍生物。其结构经1H NMR,IR,MS和元素分析表征。     

Novel Synthesis of Agaritine,a 4-Hydrazinobenzyl-Alcohol Derivative Occurring in Agaricaceae

The 4-hydrazinobenzyl alcohol ( 3 was prepared (58%)) by diiobutylaluminiumhydride reduction of methyl 4-hydrazinobenzoate ( 4 ), whereas LiA1H₄ or LiBh₄ reduction of 4 proceeded further to yield (via intermediate 3 ) (4-tolyl)hydrazine ( 5 ). The alcohol 3 was stable under O₂-free conditions and exhibited no tendency to eliminate H₂O, neither thermally nor with H⁺ catalysis. Oxidation of 3 with SeO₂ yielded 4-(hydroxymethyl)benzine-diazonium ion ( 8 ), identified by its azo coupling product 9 with 2-naphthol. Condensation of 3 with 1-benzyl 5-Hydrogen N-(benzyloxycarbonyl)-L-glutamate ( 10 ) in presence of dicyclohexylcarbodiimide afforded 81% of N²-(benzyloxycarbonyl)-L- glutamic acid 1-(benzyl-ester) 5-{2-[4-(hydroxymethyl)phenyl]hydrazide} ( 11 ) which upon controlled hydrogenolysis (quinoline-sulfur-poisoned Pd/C catalyst) gave 82% of L-Glutamic acid 5-{2-[4-(hydroxymethyl)phenyl] hydrazide} ( 1 ), i. e. agaritine, a metabolite of Agaricus bisporus. Without poisoning of the catalyst, hydrogenolysis of ( 11 ) yielded L-glutamic acid 5-[2-(4-tolyl)hydrazide] ( 12 ).

以沙美特罗的缩酮保护产物为原料,经长链仲氨的叔丁氧羰基保护、分子内酯交换反应选择性保护氨基间位的苄羟基、脱缩酮保护、与昔萘酸甲酯的Friedel-Crafts反应后再经碱水解合成了一个新型的沙美特罗衍生物——1-羟基-4-【2-羟基-5-【1-羟基-2-{[6-(4-苯丁氧基)]己氨基}乙基】苯甲基】-2-萘甲酸,总收率16.3%,其结构经1H NMR和MS表征。     

一种新的四硫富瓦烯衍生物的合成

经偶联、缩合等反应合成了一种新的用于分子组装的底物2,3-(二硫甲基)-6,7(二硫丁氧基对苯亚甲基巴比妥)四硫富瓦烯,产物用元素分析,IR和^1H NMR进行了表征,并初步探讨了其电化学行为。     

苯甲酰苯并咪唑衍生物的合成

以3,5-二甲基-1H-吡咯-2-甲酸乙酯(2)为起始原料,与新型Vilsmeier试剂反应得4-甲酰基-3,5-二甲基-1H-吡咯-2-甲酸乙酯(3);3与3,4-二氨基二苯甲酮环合制得4-(5-苯甲酰-1H-苯并咪唑-2-基)-3,5-二甲基-1H-吡咯-2-甲酸乙酯(4);4水解后与2-吡唑啉经脱水反应合成了一个苯并咪唑衍生物,总收率54%,其结构经1H NMR,13C NMR和MS确证。     

新型萘酰亚胺衍生物的合成

N-(N’,N’-二甲基氨基乙基)-4-硝基-1,8-萘酰亚胺和2,3-二甲基苯并噻唑碘化盐在痕量水存在下经亲核取代反应合成了一个新型的萘酰亚胺衍生物——N-(N’,N’-二甲基氨基乙基)-4-[2-(2,3-二氢-2,3-二甲基苯并噻唑)氧基]-1,8-萘酰亚胺,其结构经1H NMR,13C NMR和ESI-MS表征。并对反应机理进行了初步研究。     

新型邻菲啰啉衍生物的合成

以2-［2-（2-甲氧基乙氧基）乙氧基］乙基-4-甲基苯磺酸为原料,经2步反应制得中间体2,2′-【2,5-二｛2-［2-（2-甲氧基乙氧基）乙氧基］｝1,4-二（4,4,5,5-四甲基）-1,3,2-二氧硼基】苯（6）; 3-溴-1,10-邻菲啰啉和6经Suzuki偶联反应合成了一个新型的邻菲啰啉衍生物--3,3′-【2,5-二｛2-［2-（2-甲氧基乙氧基）乙氧基］｝-1,4-二（1,10-菲啰啉基）】苯,其结构经¹H NMR和MALDI-TOF-MS表征。     

一种胞苷衍生物的合成

以胞苷为原料,首先通过单乙酰化反应保护-NH2;再分别用二对甲氧基三苯甲基氯和氯甲氧基三异丙基硅烷保护5-′OH和2-′OH;被保护的胞苷经磷酰化反应合成了一种用于制备RNA长链的胞苷衍生物单体——N-乙酰基-5′-O-二对甲氧基三苯甲基-2′-O-三异丙基硅氧甲基-3′-O-双(N,N-二异丙基氨基)(2-氰乙氧基)磷基胞苷(1),总收率29.8%。1及其中间体的结构经1H NMR,MS和元素分析表征。     

四硫脲基杯[4]芳烃衍生物的合成

对叔丁基杯[4]-1,3-二乙酸乙酯与过量的二乙基三胺反应,生成含有游离氨基的杯芳烃酰胺衍物(2),2再与异硫氰酸苯酯反应合成了含4个硫脲基的杯[4]芳烃衍生物,其结构经1H NMR,IR,MS和元素分析表征。     

Total Synthesis of a Dimeric Thymol Derivative Isolated from Arnica sachalinensis

The total synthesis of a dimeric thymol derivative (thymarnicol) isolated from Arnica sachalinensis was accomplished in 6 steps. A key biomimetic Diels–Alder dimerization was found to occur at ambient temperature and the final oxidative cyclization occurs when the substrate is exposed to air and visible light. These results indicate that this natural product is likely the result of spontaneous (non‐enzyme‐mediated) reactivity.     

Synthesis and Structure of a 1,3-alternate-Thiacalix[4]arene diamide Derivative

A novel receptor possessing two complexation sites and bearing 1,3-alternate conformation based on thiacalix[4]arene, confirmed by single crystal X-ray analysis, was prepared. The tetrathiacalix[4]arene diamide shows strong intramolecular hydrogen bonding. The binding behavior towards K⁺ and halides has been examined by ¹H NMR titration experiments.

从对叔丁基杯[4]芳烃出发,对其下沿的酚羟基进行修饰,先后在1,3-和2-酚羟基位点引入冠醚环和乙基;最后在NaH/THF中与3-溴丙烯酸丙酯反应制得聚合单体——新型杯[4]冠醚衍生物,其结构经1H NMR和MS表征。     

四对-乙酰胺苯磺酰氧基杯[4]芳烃的合成

以对叔丁基苯酚为起始原料,通过缩合、取代反应合成了25,26,27,28-四对-乙酰胺苯磺酰氧基对叔丁基杯[4]芳烃,其结构经FT—IR,^1H NMR和FAB-MS表征。
