Abstract

Suitably protected 1-deoxynojirimycin (l, 5-dideoxy-l, 5-imino-D-glucitol; DNJ) and its 2-acetamido derivative, i.e., 2, 3, 6-tri-O-benzyl-.N-benzyloxycarbonyl-l, 5-dideoxy-1, 5-imino-D-glucitol (6) and 2-acetamido-3, 6-di-O-benzyl-N-benzyloxycarbonyl-1, 2, 5-trideoxy-l, 5-imino-D-glucitol (14) were each coupled with methyl 2, 3, 4, 6-tetra-O-acetyl-1-thio-β-D-galactopyranoside (15) in the presence of dimethyl(methylthio)-sulfonium triflate (DMTST) as a promoter, to give 16 and 18, which were converted to the novel disaccharides (20, 21) related to lactose and lactosamine. Coupling of 14with methyl 3, 4, 6-tri-O-acetyl-2-deoxy-2-phthalimido-l-thio-β-D-glucopyranoside (22) gave achitobiose analog (25). O-(β-D-Galactopyranosyl)-(l→3)-DNJ derivatives (38, 39) and O-(β-D-glucopyranosyl)-(l→3)-DNJ (45) were also synthesized. Conformational analysis of a variety of DNJ derivatives, based on the 1H NMR data, is also discussed.  相似文献

ABSTRACT

The behavior of 3,4- and 4,6-cyclic sulfates derived from benzyl 2,6- and 2,3-di-O-benzyl-β-D-galactopyranosides toward hydrolysis has been studied using aqueous sodium hydroxide under various conditions. Starting from benzyl 2,6-di-O-benzyl-3,4-O-sulfuryl-β-D-galactopyranoside (5), the reaction with aq NaOH in THF gave both 3- and 4-monosulfates 7 and 8 (83%, in 68:32 ratio), while the reaction in DMF led unexpectedly to the 4-deoxy-3-keto derivative 10 in 77% yield after acidic hydrolysis of the intermediate enolester 9. On the other hand, when benzyl 2,3-di-O-benzyl-4,6-O-sulfuryl-β-D-galactopyranoside (6) was treated with aq NaOH in THF, a mixture of benzyl 2,3-di-O-benzyl-6-deoxy-4-O-(sodium sulfonato)-α-L-arabino-hex-5-enopyranoside (11) and benzyl 2,3-di-O-benzyl-4-deoxy-6-O-(sodium sulfonato)-α-L-threo-hex-4-enopyranoside (12) (in 65:35 ratio) was obtained in 93% yield, giving a new and rapid access to 11, a potential precursor of L-sugars derivatives. Alternatively, BzONBu4 gave a regiospecific opening reaction of 6 and led to the 6-O-benzoate 4-O-sulfate derivative (13) in excellent yield.  相似文献

ABSTRACT

The four derivatives of β-maltosyl-(1→4)-trehalose have been synthesized, which are monodeoxygenated at the site of one of the primary hydroxyl groups. The tetrasaccharides were constructed in [2+2] block syntheses. Thus, 6′″-deoxy-β-maltosyl-(1→4)-trehalose was prepared by selective iodination of allyl 2,3,6,2′,3′-penta-O-acetyl-β-maltoside (3) followed by catalytic hydrogenolysis and coupling with 2,3-di-O-benzyl-4,6-O-benzylidene-α-D-glucopyranosyl 2′,3′,6′-tri-O-benzyl-α-D-glucopyranoside (9), and 6″-deoxy-β-maltosyl-(1→4)-trehalose by selective iodination of allyl 4′,6′-O-isopropylidene-β-maltoside (14), coupling with 9, and one-step hydrogenolysis at the tetrasaccharide level. For the synthesis of 6′-deoxy-β-maltosyl-(1→4)-trehalose, the diol 2,3-di-O-benzyl-4,6-O-benzylidene-α-D-glucopyranosyl 2′,3′-di-O-benzyl-α-D-glucopyranoside (22) was selectively iodinated and glycosylated with acetobromomaltose followed by catalytic hydrogenolysis. The 6-deoxy-β-maltosyl-(1→4)-trehalose was obtained upon selective iodination of a tetrasaccharide diol.  相似文献

Ganglioside GM3 and KDN-ganglioside GM3, containing hexanoyl, decanoyl, and hexadecanoyl groups at the ceramide moiety have been synthesized. Selective reduction of the azido group in O-(methyl 5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-D-glycero-α-D-galacto-2-nonulopyranosylonate)-(2→3)-O-(2,4-di-O-acetyl-6-O-benzoyl-β-D-galactopyranosyl)-(1→4)-O-(3-O-acetyl-2,6-di-O-benzoyl-β-D-glucopyranosyl)-(1→1)-(2S,3R,4E)-2-azido-3-O-benzoyl-4-octadecene-1,3-diol (1) and O-(methyl 4,5,7,8,9-penta-O-acetyl-3-deoxy-D-glycero-α-D-galacto-2-nonulopyranosylonate)-(2→3)-O-(2,4-di-O-acetyl-6-O-benzoyl-β-D-galactopyranosyl)-(1→4)-O-(3-O-acetyl-2,6-di-O-benzoyl-β-D-glucopyranosyl)-(1→1)-(2S,3R,4E)-2-azido-3-O-benzoyl-4-octadecene-1,3-diol (2), coupling with hexanoic, decanoic, and hexadecanoic acids, O-deacylation, and de-esterification gave the title gangliosides GM3 (11→13) and KDN-GM3 (14→16) in good yields. On the other hand, O-deacylation of 1 and subsequent de-esterification gave 2-azido-sphingosine containing-GM3 analogue 17, which was converted into lyso-GM3, in which no fatty acyl group was substituted at the sphingosine residue, by selective reduction of the azido group.  相似文献

ABSTRACT

Two derivatives of β-maltosyl-(1→4)-trehalose monodeoxygenated at positions 4 or 4′″ have been synthesized in [2+2] block syntheses. After the preparation of precursors with only one free hydroxyl group the deoxy function was introduced by a Barton-McCombie reaction. Thus, glycosylation of 2,3,6-tri-O-benzyl-α-D-glucopyranosyl 2,3,6-tri-O-benzyl-α-D-glucopyranoside (4) with octa-O-acetyl-β-maltose (3) gave tetrasaccharide 5 with only one free hydroxyl group at the 4-position. The 4′-position of an allyl maltoside was available selectively after removal of a 4′,6′-cyclic acetal and selective benzoylation of the 6′-position. Reduction of this derivative 11 afforded allyl O-(2,3-di-O-acetyl-6-O-benzoyl-4-deoxy-α-D-glucopyranosyl)-(1→4)-2,3,6-tri-O-acetyl-β-D-glucopyranoside (14), which was deallylated, activated as an trichloroacetimidate, and coupled to 2,3-di-O-benzyl-4,6-O-benzylidene-α-D-glucopyranosyl 2′,3′,6′-tri-O-benzyl-α-D-glucopyranoside (20). Several compounds were fully characterized by 1H NMR spectroscopy. Deprotection furnished the monodeoxygenated tetrasaccharides 9 and 23.  相似文献

Abstract

We have determined the preferred conformers in solution by a detailed NMR analysis using COSY and HETCOR experiments of three inositol isomers: myo (1), scyllo (2) and epi (3) plus sixteen derivatives of myo-inositol: 1,2,3,4,5,6-hexa-O-acetyl-myo-inositol (4), 1,2,-O-isopropylidene-myo-inositol (5), 1,2:4,5-di-O-isopropylidene-myo-inositol (6), 3,4,5,6-tetra-O-acetyl-1,2-O-isopropylidene-myo-inositol (7), 3,4,5,6-tetra-O-acetyl-myo-inositol (8), 1,2-O-isopropylidene-3,6-di-O-tosyl-myo-inositol (9), 1,2-O-isopropylidene-3,4,6-tri-O-tosyl-myo-inositol (10), 1,2:4,5-di-O-isopropylidene-3-O-tosyl-myo-inositol (11), 3,6-di-O-benzyl, 1,2:4,5-di-O-isopropylidene-myo-inositol (12), 3,6-di-O-benzyl-1,2-O-isopropylidene-myo-inositol (13), 3,6-di-O-benzyl-myo-inositol (14), 1,2-O-cyclohexylidene-myo-inositol (15), 1,2:4,5-di-O-cyclohexylidene-myo-inositol (16), 1,2:5,6-di-O-cyclohexylidene-myo-inositol (17), 1,3,5-O-(orthoformate)-myo-inositol (18) and 2-benzyl-1,3,5-O-(orthoformate)-myo-inositol (19). The X-ray diffraction structure of compounds 2, 6-8, 18 and 19 are reported.

Abstract

Two derivatives of β-maltosyl-(1→4)-trehalose monodeoxygenated at C-2′′ or C-2′′′ have been synthesized in [2+2] block syntheses. O-(2,3,4,6-Tetra-O-benzyl-α-D-glucopyranosyl)-(1→4)-3,6-di-O-benzyl-1,2-di-O-acetyl-β-D-glucopyranose (6), prepared from the respective orthoester, was coupled to the glycosyl acceptor 2,3-di-O-benzyl-4,6-O-benzylidene-α-D-glucopyranosyl 2,3,6-tri-O-benzyl-α-D-glucopyranoside. In the resulting tetrasaccharide 8, the only ester group was removed and replaced by a xanthate which was reduced in a Barton-McCombie reaction to afford the 2′′-deoxygenated tetrasaccharide 12. For the synthesis of a 2′′′-deoxygenated derivative, a maltose building block was assembled from two monosaccharides. The key building block was ethyl 2,3,6-tri-O-benzyl-1-thio-β-D-glucopyranoside (14) which was used i) as a glycosyl acceptor in a phenylselenyl chloride mediated coupling reaction with tri-O-benzyl-glucal and ii) after the first coupling as a glycosyl donor to react with glycosyl acceptor 7 to give tetrasaccharide 18. The phenylselenyl group was reduced with tributyltin hydride on the disaccharide level. Deprotection of 18 furnished the 2′′′-deoxy-maltosyl-(1→4)-trehalose 20.  相似文献

3-O-Sulfo glucuronyl neolactohexanosyl ceramide derivatives (heptasaccharides) have been synthesized. Condensation of 2-(trimethylsilyl)ethyl 2,4,6-tri-O-benzyl-β-D-galactopyranoside (2) with 4-O-acetyl-3,6-di-O-benzyl-2-deoxy-2-phthalimido-β-D-glucopyranosyl trichloroacetimidate (1) gave the desired β-glycoside 3, which was converted into 2-(trimethylsilyl)ethyl O-(2-acetamido-3,6-di-O-benzyl-2-deoxy-β-D-glucopyranosyl)-(1→3)-2,4,6-tri-O-benzyl-β-D-galactopyranoside (4) via removal of the O-acetyl and N-phthaloyl groups, followed by N-acetylation. Glycosylation of 4 with O-(methyl 4-O-acetyl-2-O-benzoyl-3-O-levulinoyl-β-D-glucopyranosyluronate)-(1→3)-2,4,6-tri-O-benzoyl-α-D-galactopyranosyl trichloroacetimidate (5) using trimethylsilyl trifluoromethanesulfonate gave the target tetrasaccharide 6, which was transformed via removal of the benzyl group, O-benzoylation, removal of the 2-(trimethylsilyl)ethyl group and imidate formation into the tetrasaccharide donor 9. Glycosylation of 2-(trimethylsilyl)ethyl O-(2-acetamido-3,6-di-O-benzyl-2-deoxy-β-D-glucopyranosyl)-(1→3)-O-(2,4,6-tri-O-benzyl-β-D-galactopyranosyl)-(1→4)-2,3,6-tri-O-benzyl-β-D-glucopyranoside (10) with the imidate donor 9 using trimethylsilyl trifluoromethanesulfonate gave the desired heptasaccharide 11, which was transformed into the heptasaccharide imidate donor 14. Glycosylation of (2S, 3R, 4E)-2-azido-3-O-benzoyl-4-octadecene-1,3-diol (15) with 14 gave β-glycoside 16, which was transformed into the four target compounds, via reduction of the azido group, coupling with octadecanoic acid or tetracosanoic acid, selective removal of the levulinoyl group, O-sulfation, hydrolysis of the methyl ester group and O-deacylation.  相似文献

An unexpected epimerization resulting from the reaction of α-D-glucopyranosyl derivatives with DAST is described. The reaction of 3,4-di-O-acetyl-1,6-di-O-trityl-β-D-fructofuranosyl 2,3,6-tri-O-acetyl-α-D-glucopyranoside (1), methyl 2,3-di-O-acetyl-6-O-trityl-α-D-glucopyranoside (6), 2,3-di-O-acetyl-6-O-trityl-α-D-glucopyranosyl 2,3-di-O-acetyl-6-O-trityl-α-D-glucopyranoside (13), and 2,3-di-O-acetyl-6-O-tert-butyldiphenylsilyl-α-D-glucopyranosyl 2,3,4,6-tetra-O-acetyl-α-D-glucopyranoside (14) with DAST at 0°C did not give the expected C-4 fluorodeoxy galacto derivatives, but instead, the corresponding 4-O-acetyl-3-hydroxy-α-D-galactopyranosides in yields of 52–78%. When the treatment of 6 was carried out at ?25°C for ~5 min the corresponding diastereomeric 4-O-diethylaminosulfinates (9a,b) were isolated as the major products (40%). Evidence suggests that the epimerization reaction most probably resulted from an intramolecular displacement of the intermediate diethylaminosulfur difluoride ester or diethylaminosulfinyl ester by the neighbouring acetoxy groups.  相似文献

Stereoselective α-D-galactosylation at the position 3 of 4,6-O-substituted derivatives of methyl 2-acetamido-2-deoxy-α-D-glucopyranoside is described. Glycosyl chlorides derived from 3,4,6-tri-O-acetyl-2-O-benzyl- and 2-O-(4-methoxybenzyl)-D-galactopyranose have been used as glycosyl donors. Methyl 2-acetamido-4,6-di-O-acetyl-2-deoxy-3-O-(3,4,6-tri-O-acetyl-α-D-galactopyranosyl)-α-D-glucopyranoside (27) and methyl 2-acetamido-4,6-di-O-benzyl-2-deoxy-3-O-(3,4,6-tri-O-acetyl-α-D-galactopyranosyl)-α-D-glucopyranoside (31) have been prepared.  相似文献

3,4-Di-O-acetyl-2-deoxy-2-nitroso-α-D-xylo-pentopyranosyl chloride (2) reacts with pyrazole to afford 1-[3,4-di-O-acetyl-2-deoxy-2-(Z)-hydroxyimino-α- (3) and β-D-threo-pentopyranosyl]pyrazole (4). The products of condensation were modified at C-2 or C-3 to give pyrazole derivatives with 3-azido-2,3-dideoxy-2-hydroxyimino-pentopyranosyl (5,7,8,9,10), 2-acetoxyimino-2,3-dideoxy-β-D-glycero-pentopyranosyl (12,13), β-D-lyxo- (14), β-D-xylopentopyranosyl (15) structures and 2,3-dihydro-2-pyrazol-1-yl-6H-pyran-3-one oximes (6,11). The conformation of the sugar residue and configuration at the anomeric centre and of the hydroxyimino group were established on the basis of 1H NMR and polarimetric data.  相似文献

Treatment of methyl 2,3-di-O-benzyl-α-D-glucopyranoside (1), methyl 2,3-di-O-acetyl-α-D-glucopyranoside (4), 3-O-benzyl-1,2-O-(1-methylethylidene)-α-D-glucofuranose (6), 3-O-acetyl-1,2-O-(1-methylethylidene)-α-D-glucofuranose (9), 1,2-O-(1-methylethylidene)-α-D-xylofuranose (11) and methyl 2,3-di-O-acetyl-α-D-galactopyranoside (15) with diisopropylazodicarboxylate-triphenylphosphine in tetrahydrofuran led to the corresponding dioxaphosphoranes, which were opened by trimethylsilyl azide affording the silylated primary azidodeoxysugars. When the same reaction was performed on methyl 2,3-di-O-benzyl-α-D-galactopyranoside (20), an inversion of the regioselectivity of the dioxaphosphorane opening was observed, leading mainly to the 4-azido-4-deoxy-α-D-glucopyranoside derivative 27.  相似文献

Abstract

Conformational investigations using 1D TOCSY and ROESY 1H NMR experiments on 1,3,4,6-tetra-O-acetyl-2-C-(4,6-di-O-acetyl-2,3-dideoxy-α-D-erythro-hexopyranosyl)-2-deoxy-β-D-glucopyranose (8) and related disaccharides showed that for steric reasons the C-linked hexopyranosyl ring occurs in the usually unfavoured 1C4 conformation and reconfirmed the structure of 1,3,4,6-tetra-O-acetyl-2-C-(4,6-di-O-acetyl-2,3-dideoxy-α-D-erythro-hex-2-enopyranosyl)-2-deoxy-β-D-glucopyranose (5). Glycosylation of 2,3,6-tri-O-benzyl-α-D-glucopyranosyl 2,3-di-O-benzyl-4,6-(R)-O-benzylidene-α-D-glucopyranoside (13) with acetate 8 using trimethylsilyl triflate as a catalyst afforded the α-D-linked tetrasaccharide 14. A remarkable side product in this reaction was the unsaturated tetrasaccharide 2,3,6-tri-O-benzyl-4-O-[4,6-di-O-acetyl-2,3-dideoxy-2-C-(4,6-di-O-acetyl-2,3-dideoxy-β-D-erythro-hexopyranosyl)-α-D-erythro-hex-2-enopyranosyl]-α-D-glucopyranosyl 2,3-di-O-benzyl-4,6-(R)-O-benzylidene-α-D-glucopyranoside (16) where in the C-linked hexopyranosyl ring an isomerization to the β-anomer had taken place to allow for the favoured 4C1 conformation. The tetrasaccharide 14 was deacetylated and hydrogenolyzed to form the fully deprotected tetrasaccharide 18. The 1 C 4 conformation of the C-glycosidic pyranose of this tetrasaccharide was maintained as shown by an in depth NMR analysis of its peracetate 19.  相似文献

Abstract

Selective acetolysis of methyl 2, 3, 4, 6-tetra-O-benzyl-α-D-manno-pyranoside (2) allows for easy preparation of 1-acetates of 2, 3,4, 6-tetra-O-benzyl (5), 6-O-acetyl-2, 3, 4, tri-O-benzyl-(6), 4, 6-di-O-acetyl-2,3-di-O-benzyl-(7), 3, 4, 6-tri-O-acetyl-2-O-benzyl-(8), and 2, 4, 6-tri-O-acetyl-3-O-benzyl-D-mannopyranoside (9). 8 and 9 formed are separated by preparative HPLC in 30-60g scale. The time course of previously described acetolyses of 3, 4, 6-tri-O-benzyl- 1, 2-O-(1-methoxyethyidene)-β-D-mannopyranose (3), and methyl 2, 3-dt-O-benzyl-4, 6-O-benzylldene-α-D-mannopyranoside (4) giving 9, 1, 2, 6-tri-O-acetyl-3, 4-di-O-benzyl-(10), and 1, 2-di-O-acetyl-3, 4, 6-tri-O-benzyl-(11) α-D-mannopyranose as well 7 have been studied.  相似文献

Abstract

α-Stereocontrolled, glycoside synthesis of trimeric sialic acid is described toward a systematic approach to the synthesis of sialoglycoconjugates containing an α-sialyl-(2→8)-α-sialyl-(2→8)-sialic acid unit α-glycosidically linked to O-3 of a galactose residue in their oligosaccharide chains. Glycosylation of 2-(trimethylsilyl)ethyl 6-O-benzoyl-β-d-galactopyranoside (4) or 2-(trimethylsilyl)ethyl 2,3,6,2′,6′-penta-O-benzyl-β-lactoside (5), with methyl [phenyl 5-acetamido-8-O-[5-acetamido-8-O-(5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-d-glycero-α-d-galacto-2-nonulopyranosylono-1”, 9′-lactone)-4,7-di-O-acetyl-3,5-dideoxy-d-glycero-α-d-galacto-2-nonulopyranosylono-1′, 9-lactone]-4,7-di-O-acetyl-3,5-dideoxy-2-thio-d-glycero-d-galacto-2-nonulopyranosid]onate (3), using N-iodosuccinimide-trifluoromethanesulfonic acid as a promoter, gave the corresponding α-glycosides 6 and 8, respectively. The glycosyl donor 3 was prepared from trimeric sialic acid by treatment with Amberlite IR-120 (H+) resin in methanol, O-acetylation, and subsequent replacement of the anomeric acetoxy group with phenylthio. Compounds 6 and 8 were converted into the per-O-acyl derivatives 7 and 9, respectively.  相似文献

Abstract

A systematic synthesis of sulfatide (I) and novel sulfatide analogs (II-VI) carrying 2-(tetradecyl)hexadecyl group as a ceramide substitute is described. The 3-O-, 4-O- and 3,4-di-O-levulinoyl derivatives of galactopyranosyl trichloroacetimidates (1, 12, and 13) were coupled with (2S,3R,4E)-3-O-acetyl-2-octadecanamido-4-octadecene-1,3-diol or 2-(tetradecyl)hexadecan-1-ol. The resulting glycolipids (2, 4, 14, and 15) were each transformed, by selective removal of the levulinoyl group(s), and successive sulfation and de-O-acylation, into the 3-sulfates (I, II), 4-sulfate (III), and 3,4-disulfate (IV). The 6-sulfate (V) was prepared from 2-(tetradecyl)hexadecyl β-D-galactopyranoside (21) via the 6-O-t-butyldimethylsilyl derivative, while the 3′-sulfate of 2-(tetradecyl)hexadecyl β-D-lactoside (VI) was synthesized from 2-(trimethylsilyl)ethyl 3′-O-benzyl-β-D-lactoside (26). The structures of the sulfated glycolipids (I-VI) were characterized by ion-spray MS, MS/MS, and 1H NMR spectrometry.

The synthesis of the glycosyl donors 2,3-di-O-acetyl- and 2,3-di-O-chloroacetyl- 4,6-O-ethylidene-ß-D-glucopyranose (10β) and (11β) and their use for the glycosidation of 4'-O-benzyloxycarbonyl- and 4'-O-chloroacetyl-4'-O-demethyl-4-epi-podophyllotoxins (12) and (13) is described. Starting from benzyl β-D-glucopyranoside (6), benzyl 2,3-di-O-acetyl- and 2,3-di-O-chloroacetyl-4,6-O-ethylidene-ß-D-glucopyranoside (8) and (9) were prepared. Hydrogenolysis of the benzyl group in 8 or 9 afforded the β-hydroxy glucopyranose donors 10β and 11β. Condensation of 10β or 11β with 4'-O-Z-epi-podophyllotoxin 12 in the presence of BF3-etherate gave selectively the 4-O-(2,3-di-O-acetyl- or -2,3-di-O-chloroacetyl-4,6-O-ethylidene-β-D-glucopyranosyl)-epi-podophyllotoxins 14β and 15β, respectively. The β-glycoside 16 was prepared in the same manner starting from 11β and 4'-O-chloroacetyl-epi-podophyllotoxin 13. By deblocking (Dowex 1X8, 3:2 methanol-chloroform) of the chloroacetyl groups in 15β and the following hydrogenolysis of the benzyloxycarbonyl group in 17 etoposide 1 was obtained. The deacylation of 16 afforded 1 in a one step procedure.  相似文献

Four sialyl and sulfo Lex analogs containing glucose in place of N-acetylglucosamine, and a ceramide or 2-(tetradecyl)hexadecyl residue, have been synthesized. Condensation of O-(methyl 5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-d-glycero-α-d-galacto-2-nonulopyranosylonate)-(2→3)-O-(4-O-acetyl-2,6-diO-benzoyl-β-d-galactopyranosyl)-(1→4)-O-[(2,3,4-tri-O-acetyl-α-L-fucopyranosyl)-(1→3)]-2,4-di-O-benzoyl-α-d-glucopyranosyl trichloroacetimidate (1) with (2S,3R,4E)-2-azido-3-O-benzoyl-4-octadecene-1,3, diol (2) or 2-(tetradecyl)-hexadecyl-1-ol (3) gave the corresponding β-glycosides 4 and 7. Compound 4 was converted into the ganglioside 6 via selective reduction of the azido group, coupling with octadecanoic acid, O-deacylation, and saponification of the methyl ester group. Hydrolysis of the O-acyl groups in 7 followed by saponification of the methyl ester, gave sialyl Lex ganglioside analog 8 containing a branched fatty alkyl residue. On the other hand, glycosylation of O-(4-O-acetyl-2,6-di-O-benzoyl-3-O-levulinyl-β-d-galactopyranosyl)-(1→4)-[O-(2,3,4-tri-O-acetyl-α-L-fucopyranosyl)-(1→3)]-2,6-di-O-benzoyl-α-d-glucopyranosyl trichloroacetimidate (13), prepared from 2-(trimethylsilyl)ethyl O-(2,6-di-O-benzoyl-β-d-galactopyranosyl)-(1→4)-O-[(2,3,4-tri-O-benzyl-α-L-fucopyranosyl)-(1→3)]-2,6-di-O-benzoyl-β-d-glucopyranoside (9) via selective 3-O-levulinylation, acetylation, removal of the 2-(trimethylsilyl)ethyl group, with 2 or 3, gave the desired β-glycosides 14 and 19. Selective reduction of the axido group in 14 followed by coupling with octadecanoic acid gave the ceramide derivative 16. Removal of the levulinyl group in 16 and 19, treatment with sulfur trioxide pyridine complex and subsequent hydrolysis of the protecting groups yielded the corresponding sulfo Lex analogs 18 and 21.  相似文献

Acetyl protected 1,2-O-(1-methoxyethylidene)-disaccharides 1 of maltose, cellobiose, and lactose, respectively were converted via the corresponding benzyl protected couterparts 2, the benzyl protected phenyl 2-O-acetyl- 3 and 2-O-unprotected 1-thio-glycoside disaccharides 4 into 2-O-succinoylated disaccharides 5. The latter were esterified with benzyl 2-O-benzoyl-4,6-di-O-benzylidene-α-D-glucopyranoside (6) to afford succinyl linked derivatives 7 the benzylidene groups of which were regioselectively opened to give prearranged glycoside trisaccharides 8. Intramolecular glycosylation of the latter with N-iodosuccinimide resulted in exclusive formation of the corresponding α-(1→4)-linked trisaccharides 9. No influence of the donor moiety on the diastereoselectivity of the intramolecular glycosylation was observed.  相似文献

Synthesis of sugar phosphate derivatives by means of phosphite triester method is described. Seven glucose phosphotriester derivatives have been prepared, i.e. dimethyl, methyl n-propyl, and methyl isopropyl (1, 2:5, 6-di-O-isopropylidene-α-D-glucofuranose-3-) phosphate (5, 7 and 8); methyl bis-(1, 2:5, 6-di-O-isopropylidene-α-D-glucofuranose-3-) phosphate (6); methyl bis-(1, 2, 3, 4-tetra-O-acetyl-β-D-glucopyranose-6-) phosphate (9); methyl bis-(1, 2-O-isopropylidene-3,5-O-benzylidene-α-D-glucofuranose-6-) phosphate (10); and methyl (1, 2, 5, 6-di-O-iso-propylidene-α-D-glucofuranose-3-) (1, 2, 3, 4-tetra-O-acetyl-β-D-glucopyranose-6-) [phosphate (11). The results of the displacement of second chlorine atom of the reagent by different alcohols showed that methanol, n-propanol, isopropanol and as well as the glucose derivatives reacted normally to give the expected phosphite esters which yield the expected phosphate products after oxidation, but not the t-butanol. Removal of methyl group from a phosphotriester linkage can be easily achieved by the action of t-butyl amine and thus, t-butyl ammonium bis-(1, 2:5, 6-di-O-isopropylidene-α-D-glucofuranose-3-) phosphate t-butyl amine salt (12) has been obtained from its parent phosphotriester in nearly quantitative yield. The mass spectra data of di-O-isopropylideneglucose phosphate reveals that the cleavage of these compounds follows a general pattern and can be used for their characterization.  相似文献
