Abstract

A novel aromatic compound, grandiuvarone B (5-acetoxy-3-benzoyloxymethyl-5H-oxepin-4-one), along with a known compound grandiuvarone A (5-acetoxy-6-benzoyloxymethyl-5H-oxepin-4-one) were isolated from methanol extracts of Desmos chinensis leaves. Their structures were determined by various spectroscopic techniques including nuclear magnetic resonance (NMR), high-resolution electrospray ionisation mass spectrometry (HR-ESI-MS) and circular dichroism (CD). Grandiuvarone A and grandiuvarone B are isomers and the S configuration of grandiuvarone B was reported for the first time. We then determined their antifungal activity against Aspergillus flavus. Results revealed that grandiuvarone B exhibited better antifungal activity against A. flavus, with MIC values of 0.01?mg/mL compared to grandiuvarone A (MIC values of 0.02?mg/mL). In the presence of each active compound at 160?μg/g of aquafeed, A. flavus growth was completely inhibited. Grandiuvarone B also showed antibacterial activity against the plant pathogen Ralstonia solanacearum.  相似文献

Microbial resistance is a major drawback in chemotherapy of microbial or fungal infection disease. In this study, the antifungal activity of ethanol extract of a selected plant (Sarcococca saligna) has been investigated against clinical isolates of Aspergillus niger, Aspergillus treus, Aspergillus flavus, and Aspergillus fumigatus. Also, the enhancement of the antifungal activity of fluconazole by this extract was further evaluated against mentioned test strains. Conventional disk diffusion method was used to assay the antifungal activity of S. saligna ethanol extract in the absence and presence of fluconazole. The highest antifungal activity was observed against A. treus. The ethanol extract of S. saligna enhanced the antifungal activity of fluconazole against A. niger and A. treus and A. flavus. At the highest tested contents (4 mg/disk), 1.15-, 0.64-, and 2.47-fold increases in inhibition zone surface area were observed for A. niger, A. treus, and A. flavus, respectively. However, no enhancing effect was observed for this plant extract against Aspergillus fumigates at tested contents (0.5, 1, 2, 3, and 4 mg/disk). In a separate experiment, the general cytotoxicity of the ethanol extract of S. saligna was examined with brine shrimp assay. This plant extract showed low cytotoxicity against Artemia salina (LC50 = 186 μg/ml).  相似文献

Three compounds, toosendanin (1), kulactone (2) and scopoletin (3), were isolated from either the root bark and/or the stem bark of Melia volkensii. Their structures were determined on the basis of spectroscopic data generated and by comparison with data from the literature. 1 and 2, isolated for the first time from M. volkensii, exhibited significant (p < 0.05) activity against Escherichia coli with minimum inhibitory concentration of 12.5 μg/mL, close to that of neomycin (6.25 μg/mL). The compounds also exhibited high activity against Aspergillus niger (MIC 6.25 μg/mL compared to 2.5 μg/mL for clotrimazole). Dichloromethane and methanol seed, hexane stem bark and methanol root bark extracts exhibited activities towards Escherichia coli, Staphylococcus aureus, Aspergillus niger and Plasmodium falciparum, respectively. Antimicrobial activity of the plant towards A. niger, P. falciparum and S. aureus is reported for the first time in the current work.  相似文献

Abstract

A novel Mn(III) complex with γ-diketone having general formula [Mn(hdo)3], where hdo?=?hexane-2,5-dione ligand, has been synthesized and characterized using UV–vis, FT-IR, ESI-mass and EPR spectra, elemental analysis, powder X-ray diffraction, SEM, and magnetic susceptibility measurements. The X-ray diffraction studies reveal that it has monoclinic lattice system with C2/C space group and the unit cell dimensions are a?=?9.92245 Å, b?=?10.50696 Å, and c?=?9.80835 Å. The particle size of this complex has been found to be 32.1?nm. The complex was evaluated for its antifungal activity against Aspergillus flavus, Aspergillus niger, and Aspergillus terreus fungal species. The results indicate that the minimum inhibitory concentration of the synthesized complex was 8?ppm for A. niger while for A. flavus and A. terreus it was 6?ppm. β-Diketone Mn(III) complexes inhibit the fungal growth only partially. This communication is the first report of transformation of a keto-group to an ester group catalyzed by a metal complex.  相似文献

A new furan derivative named 5-acetoxymethylfuran-3-carboxylic acid (2), together with a known furan compound, 5-hydroxymethylfuran-3-carboxylic acid (1), were isolated from the fermentation of Aspergillus flavus, endophytic fungi in Cephalotaxus fortunei. The structures of 1 and 2 were elucidated by NMR, IR, UV and MS data, as well as compared with literature data. The compounds 1 and 2 exhibited potent antibacterial activity against Staphylococcus aureus with MIC values of 31.3 and 15.6 μg/mL, respectively. The compound 2 showed moderate antioxidant activity.  相似文献

Neutral complexes of Co(II), Ni(II), Cu(II), and Zn(II) have been synthesized from the Schiff bases derived from 3-nitrobenzylidene-4-aminoantipyrine and aniline (L1)/p-nitro aniline (L2)/p-methoxy aniline (L3) in the molar ratio 1 : 1. The structural features have been determined from microanalytical, IR, UV-Vis, 1H-NMR, mass, and ESR spectral data. The Cu(II) complexes are square planar, while Co(II), Ni(II), and Zn(II) complexes are tetrahedral. Magnetic susceptibility measurements and molar conductance data provide evidence for the monomeric and neutral nature of the complexes. The X-band ESR spectrum of Cu(II) complexes at 300 and 77 K were recorded. The electrochemical behavior of the complexes in MeCN at 298 K was studied. The in vitro biological screening effects of the investigated compounds were tested against the bacterial species Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, Proteus vulgaris, and Pseudomonas aeruginosa and fungal species Aspergillus niger, Rhizopus stolonifer, Aspergillus flavus, Rhizoctonia bataicola, and Candida albicans by the well-diffusion method. Comparison of the inhibition values of the Schiff bases and their complexes indicate that the complexes exhibit higher antimicrobial activity.  相似文献

This study was designed to assess the susceptibility of various microorganisms and inhibition on heat-induced protein denaturation against diosgenin and santonin, isolated from Polygonatum verticillatum rhizomes. Both diosgenin and santonin showed significant zone of inhibition when studied against various Gram-positive (Bacillus subtilis, Bacillus cereus, Staphylococcus aureus and Staphylococcus epidermidis) and Gram-negative bacteria (Escherichia coli and Salmonella typhi). In antifungal assay, only santonin exhibited profound sensitivity against various fungi (Aspergillus flavus, Aspergillus niger, Trichoderma harzianum and Fusarium oxysporum) used in the test. Both diosgenin and santonin also exhibited marked attenuation on heat-induced protein denaturation in a concentration-dependent manner with EC50 values of 375 and 310 μg/mL, respectively. In conclusion, both the isolated compounds have antimicrobial potential supported by strong inhibition on protein denaturation and thus support the antimicrobial uses of plant in traditional system of treatment.  相似文献

Cu(II) complexes have been synthesized from the Schiff base ligands derived from furfurlyidene-4-aminoantipyrine and aniline (L1)/p-nitroaniline (L2)/p-hydroxyaniline (L3). They were characterized using analytical and spectral techniques. All the Cu(II) complexes exhibit square planar geometry. The X-band ESR spectra of the copper complexes in DMSO solution at 300 and 77 K were recorded and their salient features are reported. The in vitro biological screening effects of the investigated compounds were tested against the bacterial species, Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, Proteus vulgaris and Pseudomonas aeruginosa and fungal species, Aspergillus niger, Rhizopus stolonifer, Aspergillus flavus, Rhizoctonia bataicola and Candida albicans by serial dilution method. A comparative study of inhibition values of the Schiff base ligands and their complexes indicate that the complexes exhibit higher antimicrobial activity than the Schiff base ligands. Superoxide dismutase and reducing power activities of the copper complexes have also been studied. Depending on the molecular structure, the [CuL2(OAc)2] complex possess promising SOD mimetic activities.  相似文献

In continuing our efforts to find new effective antimicrobial agents for overcoming the problem of microbial resistance, a new series of functionalized 5-hetarylthiazoles have been designed and synthesized starting from readily accessible 1-(2-allylamino-4-methylthiazol-5-yl)ethanone (3). The structures of newly synthesized compounds were confirmed by elemental analyses, spectral data, and chemical transformations. The synthesized compounds were evaluated in vitro for their antimicrobial activity against some human pathogenic bacterial and fungal strains. The compounds 7, 18, and 24 exhibited higher antibacterial activity with minimum inhibitory concentration (MIC) values ranging from (0.03–0.06?µg/mL) than ampicillin (MIC, 0.12?µg/mL) against Streptococcus pneumoniae. Whereas compounds 4, 22, and 24 revealed higher antifungal potency than amphotericin B against Aspergillus fumigatus. The structure and antimicrobial activity relationship was also discussed.  相似文献

The reaction of S-benzyl dithiocarbazate (SBDTC) with 2,4,5-trimethoxybenzaldehyde afforded a bidentate NS Schiff base 1 (benzyl-3-N-(2,4,5-trimethoxyphenylmethylenehydrazine carbodithioate), which on further reaction with M(II) (where M(II) = nickel(II), zinc(II), palladium(II) and copper(II)) in ethanol under reflux yielded bis-chelated inner complexes [ML2] 25 with deprotonated L. The ligand and its complexes were characterized by physicochemical techniques, viz., molar conductance, magnetic susceptibility measurement, IR, NMR, UV–Vis and mass spectroscopic techniques. The crystal structures of 1 and 5 were also determined by single-crystal X-ray crystallography. The crystal structure analysis showed that the ligand exists in its thione tautomeric form. In the complexes, each of the two deprotonated ligands chelated the metal ions through the β-nitrogen and the thione sulfur forming five-membered rings. The copper(II) complex (5) exhibited a square-planar geometry, where the two N2S2 chromophores are arranged trans. All the compounds showed strong antibacterial activity against S.-β-hemolyticus, Klebsiella pneumoni, and Escherichia coli. The compounds also showed strong antifungal activity against Aspergillus fumigatus, Aspergillus niger, Aspergillus flavus, and Candida albicans with the exception of the palladium(II) complex (4) which showed no activity, while all the compounds showed no activity against Fusarium vasinfectum.  相似文献

Gastropods comprise approximately 80% of molluscans, of which land snails are used variably as food and traditional medicines due to their high protein content. Moreover, different components from land snails exhibit antimicrobial activities. In this study, we evaluated the antifungal activity of soft tissue extracts from Helix aspersa against Candida albicans, Aspergillus flavus, and Aspergillus brasiliensis by identifying extract components using liquid chromatography-tandem mass spectrometry (LC-MS-MS). Two concentrations of three extracts (methanol, acetone, and acetic acid) showed antifungal activity. Both acetone (1 g/3 mL) and acetic acid extracts (1 g/mL) significantly inhibited C. albicans growth (p = 0.0001, 5.2 ± 0.2 mm and p = 0.02, 69.7 ± 0.6 mm, respectively). A. flavus and A. brasiliensis growth were inhibited by all extracts at 1 g/mL, while inhibition was observed for acetic acid extracts against A. brasiliensis (p = 0.02, 50.3 ± 3.5 mm). The highest growth inhibition was observed for A. flavus using acetic acid and acetone extracts (inhibition zones = 38 ± 1.7 mm and 3.1 ± 0.7 mm, respectively). LC-MS-MS studies on methanol and acetone extracts identified 11-α-acetoxyprogesterone with a parent mass of 372.50800 m/z and 287.43500 m/z for luteolin. Methanol extracts contained hesperidin with a parent mass of 611.25400 m/z, whereas linoleic acid and genistein (parent mass = 280.4 and 271.48900 m/z, respectively) were the main metabolites.  相似文献

In the quest for bioactive natural products of fungal origin, Aspergillus flavus was isolated from rhizosphere of Mentha piperita using Potato Dextrose Agar (PDA) and Czapec Yeast Broth (CYB) nutrient media for metabolites production. In total, three different metabolites were purified using HPLC/LCMS and the structures were established using 500 Varian NMR experiments. Further the isolated metabolites in different concentrations (10, 100, 1000 μg/mL) were tested for herbicidal activity using Completely Randomized design (CRD) against the seeds of Silybum marianum and Avena fatua which are major threats to wheat crop in Pakistan. Among the isolated metabolites, one compound was found active against the test weed species whose activity is reported in the present work. The chemical name of the compound is 2-(1, 4-dihydroxybutan-2-yl)-1, 3-dihydroxy-6, 8-dimethoxyanthracene-9, 10(4aH, 9aH)-dione with mass of 388. Results showed that all seeds germinated in control treatment; however, with the metabolite treated, the growth was retarded to different levels in all parts of the weeds. At a dose of 1000 μg/mL of the pure compound, 100% seeds of S. marianum and 60% seeds of A. fatua were inhibited. Interestingly, the pure compound exhibited less inhibition of 10% towards the seeds of common wheat (Triticum aestivum).  相似文献

Reaction of tin dichloride and tin tetrachloride with cyclohexylamine-N-thiohydrazide (ChaThz) [L1] and 1,3-propanediamine-N-thiohydrazide (PdaThz) [L2] results in [Sn(ChaThz)2] (1), Sn(ChaThz)2Cl2] (2), [Sn(PdaThz)2] (3), and [Sn(PdaThz)2Cl2] (4), in which the thiohydrazide coordinates to tin through imine nitrogen and thioamide sulfur. The ratio metal?:?ligand was 1?:?2 for all complexes. The tin(IV) thiohydrazide complexes were characterized by elemental analysis, IR, UV-Vis, 1H-NMR, 119Sn NMR, and mass spectral studies. Using the disc diffusion method, the ligands and metal complexes were screened for in vitro antibacterial activities against four pathogenic bacteria, Escherichia coli, Staphylococcus aureus, P. aeruginosa, and Bacillus cereus and for antifungal activities against Aspergillus flavus, A. carbonarius, A. niger, and A. fumigatus. While the tin(IV) complexes exhibited moderate antifungal activities, their parent ligands showed much higher and long-lasting broad spectrum of bioactivity against fungal growth. This was particularly the case for L1 whose fungal inhibitory activity by the end of the experimental period was comparable and, for the most part, more pronounced than that of AmB. This higher activity of L1 was maintained specifically against S. Aureus but in general, bacteria were more susceptible to complexes than ligands.  相似文献

A microwave-induced technique has been developed for the synthesis of a series of novel spiro indoline-based heterocycles (7a–g) at atmospheric pressure in an open vessel using an environmentally benign procedure. This rapid method produces pure products in good yields within a few minutes in comparison to the conventional procedure developed for the synthesis of these heterocycles. The synthesized compounds have been screened in vitro for antifungal activity against two fungal strains (i.e., Aspergillus niger and Aspergillus flavus). All compounds have shown significant activity against these pathogens.

Four Melodinus species with antifungal activity were found in survey of the floral resources, in Shiwan Mountain Natural Reserve, Guangxi Province, China. Crude methanolic extracts of the twigs and leaves of Melodinus suaveolens exhibited potent antifungal activities against the plant pathogenic fungi Colletotrichum musae, Colletotrichum graminicola, Colletotrichum gloeosporioides and Alternaria musae, and the ethyl acetate fraction inhibited these pathogens at rates of 85.37, 91.47, 72.77 and 89.87%, respectively (5 mg/mL). A new compound, (2R, 3S, 5S, 6R)-1-O-methyl- chiro-inositol was isolated from the ethyl acetate fraction, along with 15 known compounds. The antifungal activities of compounds (116) were evaluated for the first time. Compound (4) had potent antifungal activity against C. gloeosporioides, C. graminicola and A. musae.  相似文献

A new series of compounds containing thiazole nucleus as Rhodanine analogues have been synthesized. The new compounds were prepared from the reactions of the thiosemicarbazones ( 3a,b ) with a series of α‐halo carbonyl compounds to give the corresponding Rhodanine analogues. The thiosemicarbazones derivatives ( 3a,b ) were reacted also with hydrazonoyl chlorides to afford the corresponding tri‐substituted and tetra‐substituted thiazoles. The structures of the newly synthesized compounds were confirmed by elemental analysis and spectral data. The biological activities of the new synthesized Rhodanine analogues' were evaluated for their antimicrobial activities. The results showed that some of these compounds showed excellent activity against two fungal strains, including Aspergillus niger and Aspergillus flavus, in addition to three yeast strains, including Saccharomyces cervesi, Candida albicans NRRL Y‐477, and Candida Pathological specimen compared with the ketoconazol, as the reference drug.  相似文献

Complexes of Cu(II), Fe(II) and Fe(III) have been synthesized with 2-nitro-3,3′-benzylidene bis[4-hydroxycoumarin]/4-chloro-3,3′-benzylidene bis[4-hydroxycoumarin]/4-hydroxy-3,3′-benzylidene bis[4-hydroxycoumarin]. They have been characterized using 1H-NMR, 13C-NMR, IR spectra, electronic spectra, magnetic measurements, elemental analyses and screened for their in-vitro biocidal activity against Bacillus cereus, Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Salmonella typhi, and Serratia marcescens bacterial strains and for their in-vitro antifungal activity against Aspergillus niger, Aspergillus flavus and Lasiodiplodia theobromae. The metal complexes exhibit good activity against bacterial strains compared to parent compounds, but no significant antifungal activity against fungal strains. In-vitro nuclease activity has been carried out using agarose gel electrophoresis. The synthesized compounds show effective nuclease activity.  相似文献

Schiff-base copper(II) complexes were prepared using macrocyclic ligands, synthesized by condensation of diethylmalonate with Schiff bases derived from o-phenylenediamine and Knoevenagel condensed β-ketoanilides (obtained by the condensation of acetoacetanilide and substituted benzaldehydes). The ligands and their copper complexes were characterized by microanalytical, mass, UV–Vis, IR, 1H-NMR, ESR and CV studies, as well as conductivity data. Microanalytical, mass and magnetic moment analyses are consistent with formation of monomeric [CuL]Cl2. Spectral studies indicate square-planar geometry around copper. The smaller grain sizes found from XRD data suggest that these complexes are polycrystalline with nanosized grains. The SEM images of [CuL1]Cl2 have leaf-like morphology. The in vitro biological screening of the investigated compounds against the bacteria Escherichia coli, Klebsiella pneumoniae, Salmonella typhi, Pseudomonas aeruginosa and Staphylococcus aureus and fungi Aspergillus niger, Rhizopus stolonifer, Aspergillus flavus, Rhizoctonia bataicola and Candida albicans were tested by the well diffusion method to assess growth inhibition. A comparative study of MIC values of the Schiff-base ligands and their complexes indicate that the complexes exhibit higher antimicrobial activity than the free ligands.  相似文献

A furanone (1), (S)-methyl 2-(2-hydroxy-3,4-dimethyl-5-oxo-2,5-dihydrofuran-2-yl)acetate, was isolated from the edible mushroom Grifola frondosa. Mass spectrometry and NMR analyses were used to elucidate the structure of this compound, and its absolute configuration was determined using circular dichroism spectroscopy. Compound 1 exhibited specific antifungal activity against the plant pathogens, Fusarium oxysporum, Gibberella zeae and Piricularia oryzae and the opportunistic human pathogen, Pseudallescheria boydii, resulting in minimum inhibitory concentration values of 2.5, 2.5, 1.25 and 0.15 μg/mL, respectively. In contrast, the furanone showed only weak activity towards Aspergillus spp., Candida albicans and several other fungal strains tested as well as no appreciable antibacterial activity.  相似文献
