A specific adsorbent for extraction of methidathion from olive oil was developed. The design of the molecularly imprinted polymer (MIP) was based on the results of the computational screening of the library of polymerisable functional monomers. MIP was prepared by thermal polymerisation using N,N’-methylene bisacrylamide (MBAA) as a functional monomer and ethylene glycol dimethacrylate (EGDMA) as a cross-linker. The polymers based on the itaconic acid (IA), methacrylic acid (MAA) and 2-(trifluoromethyl)acryl acid (TFMAA) functional monomers and one control polymer which was made without functional monomers with cross-linker EGDMA were also synthesised and tested. The performance of each polymer was compared using corresponding imprinting factor. As it was predicted by molecular modelling the best results were obtained for the MIP prepared with MBAA. The obtained MIP was optimised in solid-phase extraction coupled with high performance liquid chromatography (MISPE-HPLC-UV) and tested for the rapid screening of methidathion in olive oil. The proposed method allowed the efficient extraction of methidathion for concentrations ranging from 0.1 to 9 mg L−1 (r2 = 0.996). The limits of detection (LOD) and quantification (LOQ) in olive oil were 0.02 mg L−1 and 0.1 mg L−1, respectively. MIPs extraction was much more effective than traditional C18 reverse-phase solid phase extraction.  相似文献

The feasibility of biomimetic molecular sensing of homocysteine, an independent risk factor for cardiovascular diseases, was studied. The sensing approach coupled fluorescent derivatization of dl-homocysteine by a thiol-specific fluoro-tagging agent, N-(1-pyrenyl)maleimide, with molecular recognition by a molecularly imprinted polymer (MIP) matrix. The non-covalent MIP was fabricated using the N-(1-pyrenyl)maleimide-dl-homocysteine (PM-H) adduct as template. The PM-H-MIP was found to possess outstanding analyte-specific affinity for PM-H with binding constant, KB, of 9.28±1.6×105 M−1 and density of recognition sites, Bmax, of 11.9±0.8 nmol/g dried MIP. Following in situ fluorescent derivatization, luminescent response of the MIP was found to correlate linearly with concentration of dl-homocysteine in the range corresponding to realistic total homocysteine concentration in blood plasma. Besides being a passive recognition matrix for the binding of the fluoro-tagged analyte, the PM-H-MIP material was found to be able to specifically enhance the rate of derivatization reaction between dl-homocysteine and N-(1-pyrenyl)maleimide. In a sense, the MIP transformed a fluoro-tagging agent, which is generally reactive towards a broad spectrum of thiol-containing species, into a dl-homocysteine-specific derivatizing agent. The mechanism of such analyte-specific enhancement of derivatization rate and its advantages to the biomimetic molecular sensing are discussed.  相似文献

This paper reports the synthesis and testing of a molecularly imprinted polymer membrane for digoxin analysis. Digoxin-specific bulk polymer was obtained by the UV initiated co-polymerisation of methacrylic acid and ethylene glycol dimethacrylate in acetonitrile as porogen. After extracting the template analyte, the ground polymer particles were mixed with plasticizer polyvinyl chloride to form a MIP membrane. A reference polymer membrane was prepared from the same mixture of monomers but with no template. The resultant membrane morphologies were examined by scanning electron microscopy. The imprinted membrane was tested as the recognition element in a digoxin-sensitive fluorescence sensor; sensor response was measured using standard solutions of digoxin at concentrations of up to 4 × 10−3 mg L−1. The detection limit was 3.17 × 10−5 mg L−1. Within- and between-day relative standard deviations RSD (n = 5) were in the range 4.5-5.5% and 5.5-6.5% respectively for 0 and 1 × 10−3 mg L−1 digoxin concentrations. A selectivity study showed that compounds of similar structure to digoxin did not significantly interfere with detection for interferent concentrations at 10, 30 and 100 times higher than the digoxin concentration. This simply manufactured MIP membrane showed good recognition characteristics, a high affinity for digoxin, and provided satisfactory results in analyses of this analyte in human serum.  相似文献

A pyrimethanil-imprinted polymer (P1) was prepared by iniferter-mediated photografting a mixture of methacrylic acid and ethylene dimethacrylate onto homemade near-monodispersed chloromethylated polydivinylbenzene beads. The chromatographic behaviour of a column packed with these imprinted beads was compared with another column packed with irregular particles obtained by grinding a bulk pyrimethanil-imprinted polymer (P2). The comparison was made using the kinetic model of non-linear chromatography, studying the elution of the template and of two related substances, cyprodinil and mepanipyrim. Extension of the region of linearity, capacity factors for the template and the related substances, column selectivity, binding site heterogeneity, apparent affinity constant (K) and lumped kinetic association (ka) and dissociation rate constant (kd) were studied during a large interval of solute concentration, ranging between 1 and 2000 μg/ml. From the experimental results obtained, in the linearity region of solute concentration column selectivity and binding site heterogeneity remained essentially the same for the two columns, while column capacity (at 20 μg/ml, P1 = 23.1, P2 = 11.5), K (at 20 μg/ml, P1 = 8.3 × 106 M−1, P2 = 2.5 × 106 M−1) and ka (at 20 μg/ml, P1 = 3.5 μM−1 s−1, P2 = 0.47 μM−1 s−1) significantly increased and kd (at 20 μg/ml, P1 = 0.42 s−1, P2 = 0.67 s−1) decreased for the column packed with the imprinted beads. These results are consistent with an influence of the polymerisation method on the morphology of the resulting polymer and not on the molecular recognition properties due to the molecular imprinting process.  相似文献

Silica particles have been used as supports for the preparation of three different propazine-imprinted polymer formats. First format refers to grafting of thin films of molecularly imprinted polymers (MIPs) using an immobilised iniferter-type initiator (inif-MIP). The other two new formats were obtained by complete filling of the silica pores with the appropriate polymerisation mixture leading to a silica-MIP composite material (c-MIP) followed by the dissolution of the silica matrix resulting in spherical MIP beads (dis-MIP). These techniques offer a mean of fine-tuning the particle morphology of the resulting MIP particles leading to enhanced capacity in chromatographic applications. Porous silica (specific surface area S = 380 m2 g−1, particle size ps = 10 μm, pore volume Vp = 1.083 ml g−1 and pore diameter dp = 10.5 nm), methacrylic acid and ethylenglycol dimethacrylate were used for the preparation of the materials. All the MIP formats imprinted with propazine have been characterised by elemental analysis, FT-IR spectroscopy, nitrogen adsorption and scanning electron microscopy. Further, the materials were assessed as stationary phases in HPLC. Capacity factors, imprinting factors and theoretical plate numbers were calculated for propazine and other related triazines in order to compare the chromatographic properties of the three different stationary phases. For the inif-MIPs the column efficiency depended strongly on the amount of grafted polymer. Thus, only the polymers grafted as thin films of ca. 1.3 nm average thickness show imprinting effects and the highest column efficiency giving plate numbers (N) of 1600 m−1 for the imprinted propazine. The performance of the c-MIP stationary phase decreases as result of the complete pore filling after polymerisation and increases again after the removal of the silica matrix due to a better mass transfer in the porous mirror-image resulting polymer. From this study can be concluded that the inif-MIP shows the best efficiency for use as stationary phase in HPLC for the separation of triazinic herbicides.  相似文献

A novel voltammetric sensor for O,O-dimethyl-(2,4-dichlorophenoxyacetoxyl)(3′-nitrophenyl)methinephosphonate (Phi-NO2) based on molecularly imprinted polymer (MIP) film electrode is constructed by using sol-gel technology. The sensor responds linearly to Phi-NO2 over the concentration range of 2.0 × 10−5 to 1.0 × 10−8 mol L−1 and the detection limit is 1.0 × 10−9 mol L−1 (S/N = 3). This sensor provides an efficient way for eliminating interferences from coexisting substances in the solution. The high sensitivity, selectivity and stability of the sensor demonstrates its practical application for a simple and rapid determination of Phi-NO2 in cabbage samples.  相似文献

We have investigated the effect of buffer solution composition and pH during the preparation, washing and re-loading phases within a family of acrylamide-based molecularly imprinted polymers (MIPs) for bovine haemoglobin (BHb), equine myoglobin (EMb) and bovine catalyse (BCat). We investigated water, phosphate buffer saline (PBS), tris(hydroxymethyl)aminomethane (Tris) buffer and succinate buffer. Throughout the study MIP selectivity was highest for acrylamide, followed by N-hydroxymethylacrylamide, and then N-iso-propylacrylamide MIPs. The selectivity of the MIPs when compared with the NIPs decreased depending on the buffer conditions and pH in the order of Tris > PBS > succinate. The Tris buffer provided optimum imprinting conditions at 50 mM and pH 7.4, and MIP selectivities for the imprinting of BHb in polyacrylamide increased from an initial 8:1 to a 128:1 ratio. It was noted that the buffer conditions for the re-loading stage was important for determining MIP selectivity and the buffer conditions for the preparation stage was found to be less critical. We demonstrated that once MIPs are conditioned using Tris or PBS buffers (pH7.4) protein reloading in water should be avoided as negative effects on the MIP's imprinting capability results in low selectivities of 0.8:1. Furthermore, acidifying the pH of the buffer solution below pH 5.9 also has a negative impact on MIP selectivity especially for proteins with high isoelectric points. These buffer conditioning effects have also been successfully demonstrated in terms of MIP efficiency in real biological samples, namely plasma and serum.  相似文献

The synthesis and performance of a molecularly imprinted polymers (MIPs) as a selective solid phase extraction sorbent for the preconcentration of the carbamate pirimicarb from water samples is described. The MIP was prepared using pirimicarb as the template, methacrylic acid as the functional monomer and ethylene glycol dimethacrylate as the cross-linking monomer, and using chloroform as the solvent. The detection of pirimicarb was carried out by differential pulse voltammetry (DPV) at a hanging mercury drop electrode (HMDE) in 0.1 mol l−1 HCl. Solvents of different polarities were checked for the polymer synthesis, and different experimental variables (sample pH, selection of the eluent used, eluent volume, analyte and eluent flow rates and sample volume) associated with the rebinding/extraction process were optimised. For a 25 ml sample, the process took about 13 min and resulted in a nominal enrichment factor of 50 (eluent MeOH:H2O:HAc, 7:2:1; 0.5 ml) for pirimicarb. A limit of detection of 4.1 μg l−1 was obtained, and a good reproducibility of the measurements using different MIP microcolumns was found. Furthermore, the MIP selectivity was evaluated by checking several substances with similar and different molecular structures to that of pirimicarb. As an application, pirimicarb was determined in water samples of diverse origin which were spiked at a concentration level of 71.5 μg l−1.  相似文献

Two molecularly imprinted polymers (MIPs), in the physical form of well-defined polymer microspheres, were synthesised via precipitation polymerisation (PP) using an antiepileptic drug, carbamazepine (CBZ), as template molecule, methacrylic acid as functional monomer and either divinylbenzene 80 (DVB-80) or a mixture of DVB-80 and ethylene glycol dimethacrylate (EGDMA) as crosslinking agents. The MIP obtained using DVB-80 alone as crosslinking agent (MIP A) had a narrow particle size distribution (9.5 ± 0.5 μm) and a well-developed permanent pore structure (specific surface area in the dry state = 758 m2 g−1), whereas when a mixture of DVB-80 and EGDMA (MIP B) were used as crosslinking agents, the polymer obtained had a broader particle size distribution (6.4 ± 1.8 μm) and a relatively low specific surface area (23 m2 g−1). The molecular recognition character of both polymers was evaluated by means of LC and then a molecularly imprinted solid-phase extraction (MISPE) protocol; CBZ was recognised by both polymers, and useful cross-selectivity for oxcarbazepine (OCBZ), which is the main metabolite of CBZ, also observed. In a detailed bioanalytical study, MIP A was selected in preference to MIP B since MIP A enabled a high volume of sample to be extracted such that lower limits of detection were achievable using this polymer. High recoveries of CBZ and OCBZ were obtained in a MISPE protocol when 50 mL of human urine spiked at 0.2 mg L−1 were percolated through MIP A (90% and 83%, respectively).  相似文献

The high-temperature polymorphs of two photocatalytic materials, BiNbO4 and BiTaO4 were synthesized by the ceramic method. The crystal structures of these materials were determined by single-crystal X-ray diffraction. BiNbO4 and BiTaO4 crystallize into the triclinic system P1¯ (No. 2), with a=5.5376(4) Å, b=7.6184(3) Å, c=7.9324(36) Å, α=102.565(3)°, β=90.143(2)°, γ=92.788 (4)°, V=326.21 (5) Å3, Z=4 and a=5.931 (1) Å, b=7.672 (2) Å, c=7.786 (2) Å, α=102.94 (3)°, β=90.04 (3)° γ=93.53 (3)°, V=344.59 (1) Å3 and Z=4, respectively. The structures along the c-axis, consist of layers of [Bi2O2] units separated by puckered sheets of (Nb/Ta)O6 octahedra. Photocatalytic studies on the degradation of dyes indicate selectivity of BiNbO4 towards aromatics containing quinonic and azo functional groups.  相似文献

Molecularly imprinted polymers (MIPs) selective for lysozyme were prepared on SPR sensor chips by radical co-polymerization with acrylic acid and N,N′-methylenebisacrylamide. Gold-coated SPR sensor chips were modified with N,N′-bis(acryloyl)cystamine, on which MIP thin films were covalently conjugated. The presence of NaCl during the polymerization and the re-binding tests affected the selectivity and the optimization of NaCl concentration in the pre-polymerization mixture and the re-binding buffer could enhance the selectivity in the target protein sensing. When the lysozyme-imprinted polymer thin films were prepared in the presence of 40 mM NaCl, the selectivity factor (target protein bound/reference protein bound) of MIP in the re-binding buffer containing 20 mM NaCl was 9.8, meanwhile, that of MIP in the re-binding buffer without NaCl was 1.2. A combination of SPR sensing technology with protein-imprinted thin films is a promising tool for the construction of selective protein sensors.  相似文献

Efficient aerobic oxidation of benzylic compounds has been achieved under no irradiation using a new organocatalytic system in the presence of acridine yellow and N-hydroxyphthalimide with assistance of a catalytic amount of molecular bromine. Various substrates, especially alkylaromatics, were effectively oxygenated to the corresponding carbonyl compounds with molecular oxygen as oxidant under mild conditions. For instance, indan was oxidized with 92% conversion and 79% selectivity for 1-indanone under 0.3 MPa of O2 at 75 °C.  相似文献

An easy-to-use colorimetric test-system for the efficient detection of creatinine in aqueous samples was developed. The test-system is based on composite molecularly imprinted polymer (MIP) membranes with artificial receptor sites capable of creatinine recognition. A thin MIP layer was created on the surface of microfiltration polyvinylidene fluoride (PVDF) membranes using method of photo-initiated grafting polymerization. The MIP layer was obtained by co-polymerization of a functional monomer (e.g. 2-acrylamido-2-methyl-1-propanesulfonic acid, itaconic acid or methacrylic acid) with N, N′-methylenebisacrylamide as a cross-linker. The choice of the functional monomer was based on the results of computational modeling. The creatinine-selective composite MIP membranes were used for measuring creatinine in aqueous samples. Creatinine molecules were selectively adsorbed by the MIP membranes and quantified using color reaction with picrates. The intensity of MIP membranes staining was proportional to creatinine concentration in an analyzed sample. The colorimetric test-system based on the composite MIP membranes was characterized with 0.25 mM detection limit and 0.25–2.5 mM linear dynamic range. Storage stability of the MIP membranes was estimated as at least 1 year at room temperature. As compared to the traditional methods of creatinine detection the developed test-system is characterized by simplicity of operation, small size and low cost.  相似文献

An approach using systematic optimization for the formation of an albumin molecularly imprinted polymer (MIP), able to separate albumin from proteins in solution, has been prepared by imprinting albumin using a copolymer comprising 3-dimethylaminopropyl methacrylate and tetraethylene glycol dimethacrylate in a mole ratio of 1 to 8. Cytochrome c, lysozyme and myoglobin were used in competitive re-binding experiments to compete with the polymer's native template with all protein species present at 0.0004 g mL− 1. The effects of: monomer to crosslinker mole ratio, polymerization temperature and time were investigated. It was found that the addition of water 6.04%, into the pre-polymerization albumin-monomer complex enhanced the adsorption capacity and selectivity of the resulting MIP from 2.18 × 10− 3 to 6.02 × 10− 3 g g-MIP− 1 and 83.5% to 98.7%, respectively. These results also showed that the MIP possessed high selectivity and adsorption capacity with respect to albumin in comparison with interfering species also present in solution. Polymerization temperature, time and the water content of the pre-polymerization mixture were all shown to have significant effects on the resulting albumin-MIP's performance. However, their influence on the polymer's affinity for the potentially interfering species was negligible. Additionally, higher polymerization temperatures (> 38 °C) and extended polymerization times (> 60 h) increased monomer conversion as determined by HPLC, but decreased the selectivity and adsorption capacity of the MIP. An optimized MIP, with very high selectivity and 6.37 × 10− 3 g g-MIP− 1 template re-adsorption capacity was obtained using the following polymerization conditions: 0.125 mole ratio of monomer to crosslinker, 6.04 wt.% water content with respect to the mass of the monomer complex, 60 h polymerization time at 38 °C, and with 0.47% albumin in the pre-polymerization monomer complex. Finally, the functions of polymerization temperature, time and the significance of the water content in the albumin-monomer complex are also discussed.  相似文献

Seven tweezer-type copper(II) ion-selective ionophores; that is, 3α,12α-bis[[[N-(R)thiocarboxamino]acetyl]oxy]-N,N-dioctyl-5β-cholan-24-amides and 3α,12α-bis[[[N-(R)thiocarboxaminomethyl]acetyl]oxy]-N,N-dioctyl-5β-cholan-24-amides (R = alkyl and phenyl), were newly designed and synthesized. Their potentiometric evaluation of the poly(vinyl chloride) (PVC) membranes showed excellent affinity and selectivity to copper(II) ions over those of other transition metal ions and alkali/alkaline earth metal ions. These membranes exhibited super-Nernstian responses toward copper(II) ions (34-36 mV/decade), with detection limits of 10−6-10−7 M.  相似文献

The syntheses, physical characterization and crystal structures of two new molecular copper(II) complexes of composition [Cu(C5H5N)2(C7F5O2)2] (1) and [Cu(C5H5N)2(C7F5O2)2(H2O)] (2) (C5H5N = py = pyridine and C7F5O2 = pfb = pentafluorobenzoate) are reported. Single-crystal X-ray structure determinations revealed that in 1, the Cu2+ ion, which lies on a crystallographic inversion centre, is coordinated to two py molecules and two oxygen atoms from two monodentate pfb anions, resulting in a trans-CuN2O2 square planar geometry. In 2, the Cu2+ ion is also coordinated to two py and two pfb species in addition to a water molecule in the apical site of a distorted CuN2O3 square pyramid. In the crystal packing, both 1 and 2 show segregated aromatic π-π stacking interactions in which (py + py) and (pfb + pfb) ring-pairings are seen, but no (py + pfb) pairings occur. This work reports the preparation of a molecularly imprinted polymer (MIP) for selective catalytic detection of serotonin (5-hydroxytryptamine, 5-HT). The process is based on the synthesis of polymers with hemin introduced as the catalytic center to mimic the active site of peroxidase. The copolymer MIP, containing artificial recognition sites for 5-HT, has been prepared by bulk polymerization using methacrylic acid (MAA) and hemin as the functional monomers, and ethylene glycol dimethacrylate (EGDMA) as the cross-linker. For the determination of 5-HT, a flow injection analysis system coupled to an amperometric detector was optimized using multivariate analysis. The effects of different parameters, such as pH, buffer flow rate, buffer nature, peroxide concentration and sample volume were evaluated. After optimizing the experimental conditions, a linear response range from 1.0 up to 1000.0 μmol L−1 was obtained with a sensitivity of 0.4 nA/μmol L−1. The detection limit was found to be 0.30 μmol L−1, while the precision values (n = 6) evaluated by relative standard deviation (R.S.D.) were, respectively, 1.3 and 1.7% for solutions of 50 and 750 μmol L−1 of 5-HT. No interference was observed by structurally similar compounds (including epinephrine, dopamine and norepinephrine), thus validating the good performance of the imprinted polymer. The method was applied for the determination of 5-HT in spiked blood serum samples.  相似文献

The affinity of a 2,4-dichlorophenoxyacetic acid (2,4-D) molecularly imprinted polymer (MIP), which was synthesised directly in an aqueous organic solvent, for its template (2,4-D) was studied and compared with the affinity exhibited by two other reference (control) polymers, NIPA and NIPB, for the same analyte. Zonal chromatography was performed to establish the optimal selectivity, expressed as imprinting factor (IF), under chromatographic conditions more aqueous than those described so far in the literature. Frontal analysis (FA) was performed on columns packed with these polymers, using an optimized mobile phase composed of methanol/phosphate buffer (50/50, v/v), to extract adsorption isotherm data and retrieve binding parameters from the best isotherm model. Surprisingly, the template had comparable and strong affinity for both MIP (K = 3.8 × 104 M−1) and NIPA (K = 1.9 × 104 M−1), although there was a marked difference in the saturation capacities of selective and non-selective sites, as one would expect for an imprinted polymer. NIPB acts as a true control polymer in the sense that it has relatively low affinity for the template (K = 8.0 × 102 M−1). This work provides the first frontal chromatographic characterization of such a polymer in a water-rich environment over a wide concentration range. The significance of this work stems from the fact that the chromatographic approach used is generic and can be applied readily to other analytes, but also because there is an increasing demand for well-characterised imprinted materials that function effectively in aqueous media and are thus well-suited for analytical science applications involving, for example, biofluids and environmental water samples.  相似文献

Flavonoids are natural products having several biological and physiological properties depending upon their molecular configurations. Flavonoids with similar configuration cannot be separated by traditional separation method and membrane separation technology whose selectivity is lower. This work investigates composite membranes with structural and functional molecular recognition properties prepared according to molecular imprinted technology. Functional silica sol was synthesized by taking luteolin as the template (or imprinting) molecule, γ-aminopropyltriethoxysilane (γ-APTS) as the functional monomer, and tetraethoxysilane (TEOS) as the cross-linker. The resultant functional silica sol was coated on Al2O3 microporous substrate followed by the removal of the template molecule. Scanning electron microscope micrographs showed a 5 μm thickness composite membrane with uniformly distributed porosity. Steady state flux was reached at ∼70 min at 215 L m−2 h−1 for the composite membrane, while a lower value of 168 L m−2 h−1 was measured for the blank membrane (i.e. non-templated). Further, in an aqueous mixture containing similar template molecules, the selectivity factor of luteolin to rutin was 14.1, thus suggesting that the imprinting process allowed for preferential permeance and affinity selectivity to the template molecule (i.e. luteolin). These results strongly suggest the formation of cavities, which are joined by channels to deliver the percolative effect for the permeation of luteolin. In addition to structural formation, further site recognition properties were accomplished by the functional silica sol in the composite matrix by electrovalent bonds. Considering the percolative effect in tandem with electrovalent bonds and under the influence of a concentration gradient (i.e. driving force), a mechanism of molecular recognition was proposed based on the molecular bond, followed by bond cutting and jumping to another site to form another molecular bond. The preparation method of the composite membrane was applied to other template molecules, and the template molecules can selectively permeate the membrane. So the method was universal for other substance. So it made it possible for the separation of the natural products exactly and efficiently. At the same time, it had great potential for the resolution of the chiral drugs and the preparation of the new membrane reactor.  相似文献
