The directly suspended droplet microextraction (DSDME) technique coupled with the capillary gas chromatography-flame ionization detector (GC-FID) was used to determine BTEX compounds in aqueous samples. The effective parameters such as organic solvent, extraction time, microdroplet volume, salt effect and stirring speed were optimized. The performance of the proposed technique was evaluated for the determination of BTEX compounds in natural water samples. Under the optimal conditions the enrichment factors ranged from 142.68 to 312.13, linear range; 0.01-20 μg mL−1, limits of detection; 0.8-7 ng mL−1 for most analytes. Relative standard deviations for 0.2 μg mL−1 of BTEX in water were in the range 1.81-2.47% (n = 5). The relative recoveries of BTEX from surface water at spiking level of 0.2 μg mL−1 were in the range of 89.87-98.62%.  相似文献

The herbicides amitrole and glyphosate, and its metabolite aminomethyl-phosphonic acid (AMPA), in water samples have been directly analysed by high-performance liquid chromatography using an electrochemical (EC) detector. Limits of detection of 0.3 μg mL−1 for glyphosate, 0.05 μg mL−1 for AMPA and 0.03 μg mL−1 for amitrole were comparable to those obtained by other authors using EC and also by liquid chromatography coupled to mass spectrometry, but the latter method requires derivatisation and pre-concentration of the sample whereas EC methods show similar sensitivity without the need of any derivatisation. The method was specifically designed to analyse extracts from passive samplers used for monitoring of polar herbicide residues in waters. To this purpose, three types of Empore® disks were tested for their ability to adsorb and desorb these ionic, polar analytes. A procedure for their extraction from the membranes and reducing the interferences from other substances present in natural waters (i.e. humic acids) is described. The method is simple, does not require sophisticated equipment and is valid for the analysis and monitoring of herbicides residues using passive samplers.  相似文献

A novel solid-phase microextraction (SPME) fiber coated with multiwalled carbon nanotubes/polypyrrole (MWCNTs/Ppy) was prepared with an electrochemical method and used for the extraction of pyrethroids in natural water samples. The results showed that the MWCNTs/Ppy coated fiber had high organic stability, and remarkable acid and alkali resistance. In addition, the MWCNTs/Ppy coated fiber was more effective and superior to commercial PDMS and PDMS/DVD fibers in extracting pyrethroids in natural water samples. Under optimized conditions, the calibration curves were found to be linear from 0.001 to 10 μg mL−1 for five of the six pyrethroids studied, the exception being fenvalerate (which was from 0.005 to 10 μg mL−1), and detection limits were within the range 0.12-0.43 ng mL−1. The recoveries of the pyrethroids spiked in water samples at 10 ng mL−1 ranged from 83 to 112%.  相似文献

A novel solid-phase microextraction (SPME) fiber coated with multiwalled carbon nanotubes (MWCNTs)/Nafion was developed and applied for the extraction of polar aromatic compounds (PACs) in natural water samples. The characteristics and the application of this fiber were investigated. Electron microscope photographs indicated that the MWCNTs/Nafion coating with average thickness of 12.5 μm was homogeneous and porous. The MWCNTs/Nafion coated fiber exhibited higher extraction efficiency towards polar aromatic compounds compared to an 85 μm commercial PA fiber. SPME experimental conditions, such as fiber coating, extraction time, stirring rate, desorption temperature and desorption time, were optimized in order to improve the extraction efficiency. The calibration curves were linear from 0.01 to 10 μg mL−1 for five PACs studied except p-nitroaniline (from 0.005 to 10 μg mL−1) and m-cresol (from 0.001 to 10 μg mL−1), and detection limits were within the range of 0.03–0.57 ng mL−1. Single fiber and fiber-to-fiber reproducibility were less than 7.5 (n = 7) and 10.0% (n = 5), respectively. The recovery of the PACs spiked in natural water samples at 1 μg mL−1 ranged from 83.3 to 106.0%.  相似文献

A simple, sensitive, and specific method with gas chromatography-mass spectrometry was developed for simultaneous extraction and derivatization of amphetamines (APs) and 3,4-methylenedioxyamphetamines (MDAs) in human urine by using a monolithic silica spin column. All the procedures, such as sample loading, washing, and elution were performed by centrifugation. APs and MDAs in urine were adsorbed on the monolithic silica and derivatized with propyl chloroformate in the column. Methamphetamine-d5 was used as an internal standard. The linear ranges were 0.01-5.0 μg mL−1 for methamphetamine (MA) and 3,4-methylenedioxymethamphetamine (MDMA) and 0.02-5.0 μg mL−1 for amphetamine (AP) and 3,4-methylenedioxyamphetamine (MDA) (coefficient of correlation ≧0.995). The recovery of APs and MDAs in urine was 84-94%, and the relative standard deviation of the intra- and interday reproducibility for urine samples containing 0.1, 1.0, and 4.0 μg mL−1 of APs and MDAs ranged from 1.4% to 13.6%. The lowest detection limit (signal-to-noise ratio ≧ 3) in urine was 5 ng mL−1 for MA and MDMA and 10 ng mL−1 for AP and MDA. The proposed method can be used to perform simultaneous extraction and derivatization on spin columns that have been loaded with a small quantity of solvent by using centrifugation.  相似文献

In the present work, a new ligandless-dispersive liquid-liquid microextraction (LL-DLLME) method has been developed for preconcentration trace amounts of copper as a prior step to its determination by flame atomic absorption spectrometry. In the proposed approach 1,2-dicholorobenzene and ethanol were used as extraction and dispersive solvents, respectively. Some factors influencing on the extraction efficiency of copper and its subsequent determination were studied and optimized, such as the extraction and dispersive solvent type and volume, pH of sample solution, extraction time and salting out effect. Under the optimal conditions, the calibration curve was linear in the range of 1.0 ng mL−1-0.6 μg mL−1 of copper with R2 = 0.9985. Detection limit was 0.5 ng mL−1 in original solution (3Sb/m) and the relative standard deviation for seven replicate determination of 0.2 μg mL−1 copper was ±1.4%. The proposed method has been applied for determination of copper in standard and water samples with satisfactory results.  相似文献

The reactions of 4-chloro-7-nitrobenzofurazan (NBD-Cl) with glyphosate (GLY) and with its main metabolite, aminomethylphosphonic acid (AMPA), have been studied. The resolution of binary mixtures of glyphosate and aminomethylphosphonic acid has been accomplished by partial least squares (PLS) multivariate calibration. The method of determination is based on the fluorescence emission of the derivatives formed in presence of NBD-Cl at 90 °C, in methanol and in basic medium. The dynamic ranges of the methods were comprised between 10 and 150 μg l−1 for GLY and between 10 and 200 μg l−1 for AMPA, being the detection limits 2 and 5.4 μg l−1 for GLY and AMPA, respectively. The total luminiscence information of the derivatives has been used to optimize the spectral data set to perform the calibration, by analysis of the three-dimensional excitation-emission matrices. A comparison between the predictive ability of the multivariate calibration method, partial least squares type 1 (PLS-1), on two spectral data sets, emission and synchronous spectra, has been performed. The PLS-1 method, applied to the emission spectra, has been selected as optimum. The proposed method has been applied to the simultaneous determination of GLY and AMPA in river water. For concentrations ranging from 100 to 600 μg l−1 of each compound in the samples, analytical recoveries range from 83 to 94% for GLY and from 104 to 120% for AMPA.  相似文献

In pH 4.2-4.8 HAc-NaAc buffer solution, folic acid (FA) could react with uranium (VI) to form a 2:1 anionic chelate which further reacted with some basic triphenylmethane dyes (BTPMD) such as Ethyl Violet (EV), Methyl Violet (MV) and Crystal Violet (CV) to form 1:2 ion-association complexes. As a result, not only the absorption spectra were changed, but also the intensities of resonance Rayleigh scattering (RRS) were enhanced greatly and the new RRS spectra were observed. The maximum RRS wavelengths were located at 328 nm for EV system, 325 nm for MV system and 328 nm for CV system. The fading degree (ΔA) and RRS intensities (ΔI) of three systems were different. Under given conditions, the ΔA and ΔI were all directly proportional to the concentration of FA. The linear ranges and the detection limits of RRS methods were 0.0039-5.0 μg mL−1 and 1.2 ng mL−1 for EV system, 0.0073-4.0 μg mL−1 and 2.2 ng mL−1 for MV system, 0.014-3.5 μg mL−1 and 4.7 ng mL−1 for CV system. The RRS methods exhibited higher sensitivity, so they are more suitable for the determination of trace FA. The optimum conditions, the influencing factors and the effects of coexisting substances on the reaction were investigated. The method can be applied to the determination of FA in serum and urine samples with satisfactory results. The structure of the ternary ion-association complex and the reaction mechanism were discussed in this work.  相似文献

Enriching the surface density of immobilized capture antibodies enhances the detection signal of antibody sandwich microarrays. In this study, we improved the detection sensitivity of our previously developed P-Si (porous silicon) antibody microarray by optimizing concentrations of the capturing antibody. We investigated immunoassays using a P-Si microarray at three different capture antibody (PSA – prostate specific antigen) concentrations, analyzing the influence of the antibody density on the assay detection sensitivity. The LOD (limit of detection) for PSA was 2.5 ng mL−1, 80 pg mL−1, and 800 fg mL−1 when arraying the PSA antibody, H117 at the concentration 15 μg mL−1, 35 μg mL−1, and 154 μg mL−1, respectively. We further investigated PSA spiked into human female serum in the range of 800 fg mL−1 to 500 ng mL−1. The microarray showed a LOD of 800 fg mL−1 and a dynamic range of 800 fg mL−1 to 80 ng mL−1 in serum spiked samples.  相似文献

Orthogonal array design (OAD) was utilized for the first time to optimize the experimental conditions of ultrasound-assisted emulsification–microextraction (USAEME) for determining chlorinated phenoxyacetic acids (CPAs) in river water samples. The use of ultrasound facilitates the mass transfer of CPAs from an aqueous phase into a water-immiscible organic extraction solvent (dichloromethane, DCM) without adding dispersive solvent to form numerous microdroplets. The water-immiscible extractant was collected by centrifugation, dried under low pressure, reconstituted in methanol–water mixture (1:1), and injected into a HPLC system for the determination of CPAs. The linear range was 2–1000 ng mL−1 (2, 5, 10, 50, 200, 500 and 1000 ng mL−1) for each analyte and the relative standard deviations of CPAs among the seven different concentrations were in the range of 1.5–17.0% (n = 3). The detection limits (signal-to-noise ratio of 3) of CPAs ranged from 0.67 to 1.50 ng mL−1. The ranges of intra-day precision (n = 3) for CPAs at the levels of 5 and 200 ng mL−1 were 3.6–11.9% and 5.3–9.5%, respectively. The range of inter-day precision (n = 3) at 5 and 200 ng mL−1 were 1.4–7.7% and 8.5–12.2%, respectively. The applicability of USAEME for environmental analysis was demonstrated by determining CPAs in river water. The recoveries of CPAs from five-spiked river water samples at 10 and 200 ng mL−1 were 96.3–112.5% and 94.8–109.4%, respectively. The maximum contaminant level (MCL) of 2,4-D in drinking water and the tolerance of residues in food for p-CPA are 70 and 200 μg L−1, respectively, according to the US EPA regulations. These contaminant levels fall in the linear range investigated in this study. In addition, this USAEME method provided detection limits lower than their contaminant levels, which made USAEME an effective sample preparation method for determining organic environmental contaminants, such as CPAs, in river water samples with little consumption of organic solvent.  相似文献

In this article, a new ligandless dispersive liquid-liquid microextraction method has been developed for preconcentration of trace quantities of silver as a prior step to its determination by flame atomic absorption spectrometry. In the proposed approach, carbon tetrachloride and ethanol were used as extraction and dispersive solvents. Several factors that may be affected on the extraction process, like, extraction solvent, disperser solvent, the volume of extraction and disperser solvent, pH of the aqueous solution and extraction time were optimized. Under the optimal conditions, the calibration curve was linear in the range of 5.0 ng mL−1 to 2.0 μg mL−1 of silver with R2 = 0.9995 (n = 9) and detection limit based on three times the standard deviation of the blank (3Sb) was 1.2 ng mL−1 in original solution. The relative standard deviation for eight replicate determination of 0.5 μg mL−1 silver was ±1.5%. The high efficiency of dispersive liquid-liquid microextraction to carry out the determination of silver in complex matrices was demonstrated. The proposed method has been applied for determination of trace amount of silver in standard and water samples with satisfactory results.  相似文献

The present article reports on the application of modified multiwalled carbon nanotubes (MMWCNTs) as a new, easily prepared and stable solid sorbent for the preconcentration of trace rhodium ion in aqueous solution. Rhodium ions were complexed with 1-(2-pyridylazo)-2-naphthol (PAN) in the pH range of 3.2-4.7 and then the formed Rh-PAN complex was adsorbed on the oxidized MWCNTs. The adsorbed complex was eluted from MWCNTs sorbent with 5.0 mL of N,N-dimethylformamide (DMF). The rhodium in eluted solution was determined by flame atomic absorption spectrometry (FAAS). Linear range for the determination of rhodium was maintained between 0.16 ng mL−1 and 25.0 μg mL−1 in initial solution. Relative standard deviation for the 10 replicated determination of 4.0 μg mL−1 of rhodium was ±0.97%. Detection limit was 0.010 ng mL−1 in initial solution (3Sbl, n = 10) and preconcentration factor was 120. Sensitivity for 1% absorbance of rhodium (III) was 0.112 μg mL−1. The sorption capacity of oxidized MWCNTs for Rh (III) was 6.6 mg g−1. The effects of the experimental parameters, including the sample pH, flow rates of sample and eluent solution, eluent type, breakthrough volume and interference ions were studied for the preconcentration of Rh3+. The proposed method was successfully applied to the extraction and determination of rhodium in different samples.  相似文献

The study details the development of a fully validated, rapid and portable sensor based method for the on-site analysis of microcystins in freshwater samples. The process employs a novel lysis method for the mechanical lysis of cyanobacterial cells, with glass beads and a handheld frother in only 10 min. The assay utilises an innovative planar waveguide device that, via an evanescent wave excites fluorescent probes, for amplification of signal in a competitive immunoassay, using an anti-microcystin monoclonal with cross-reactivity against the most common, and toxic variants. Validation of the assay showed the limit of detection (LOD) to be 0.78 ng mL−1 and the CCβ to be 1 ng mL−1. Robustness of the assay was demonstrated by intra- and inter-assay testing. Intra-assay analysis had % C.V.s between 8 and 26% and recoveries between 73 and 101%, with inter-assay analysis demonstrating % C.V.s between 5 and 14% and recoveries between 78 and 91%. Comparison with LC–MS/MS showed a high correlation (R2 = 0.9954) between the calculated concentrations of 5 different Microcystis aeruginosa cultures for total microcystin content. Total microcystin content was ascertained by the individual measurement of free and cell-bound microcystins. Free microcystins can be measured to 1 ng mL−1, and with a 10-fold concentration step in the intracellular microcystin protocol (which brings the sample within the range of the calibration curve), intracellular pools may be determined to 0.1 ng mL−1. This allows the determination of microcystins at and below the World Health Organisation (WHO) guideline value of 1 μg L−1. This sensor represents a major advancement in portable analysis capabilities and has the potential for numerous other applications.  相似文献

A rapid and sensitive method for the determination of carbendazim (methyl benzimidazole-2-ylcarbamate, MBC) and thiabendazole (TBZ) in water and soil samples was developed by using dispersive liquid-liquid microextraction (DLLME) coupled with high performance liquid chromatography with fluorescence detection. The water samples were directly used for the DLLME extraction. For soil samples, the target analytes were first extracted by 0.1 mol L−1 HCl. Then, the pH of the extract was adjusted to 7.0 with 2 mol L−1 NaOH before the DLLME extraction. In the DLLME extraction method, chloroform (CHCl3) was used as extraction solvent and tetrahydrofuran (THF) as dispersive solvent. Under the optimum conditions, the enrichment factors for MBC and TBZ were ranged between 149 and 210, and the extraction recoveries were between 50.8 and 70.9%, respectively. The linearity of the method was obtained in the range of 5-800 ng mL−1 for water sample analysis, and 10-1000 ng g−1 for soil samples, respectively. The correlation coefficients (r) ranged from 0.9987 to 0.9997. The limits of detection were 0.5-1.0 ng mL−1 for water samples, and 1.0-1.6 ng g−1 for soil samples. The relative standard deviations (RSDs) varied from 3.5 to 6.8% (n = 5). The recoveries of the method for MBC and TBZ from water samples at spiking levels of 5 and 20 ng mL−1 were 84.0-94.0% and 86.0-92.5%, respectively. The recoveries for soil samples at spiking levels of 10 and 100 ng g−1 varied between 82.0 and 93.4%.  相似文献

Ultrasound-assisted dispersive liquid–liquid microextraction coupled with high-performance liquid chromatography-fluorescence detection was used for the extraction and determination of three biogenic amines including octopamine, tyramine and phenethylamine in rice wine samples. Fluorescence probe 2,6-dimethyl-4-quinolinecarboxylic acid N-hydroxysuccinimide ester was applied for derivatization of biogenic amines. Acetonitrile and 1-octanol were used as disperser solvent and extraction solvent, respectively. Extraction conditions including the type of extraction solvent, the volume of extraction solvent, ultrasonication time and centrifuging time were optimized. After extraction and centrifuging, analyte was injected rapidly into high-performance liquid chromatography and then detected with fluorescence. The calibration graph of the proposed method was linear in the range of 5–500 μg mL−1 (octopamine and tyramine) and 0.025–2.5 μg mL−1 (phenethylamine). The relative standard deviations were 2.4–3.2% (n = 6) and the limits of detection were in the range of 0.02–5 ng mL−1. The method was applied to analyze the rice wine samples and spiked recoveries in the range of 95.42–104.56% were obtained. The results showed that ultrasound-assisted dispersive liquid–liquid microextraction was a very simple, rapid, sensitive and efficient analytical method for the determination of trace amount of biogenic amines.  相似文献

Dispersive liquid–liquid microextraction (DLLME) has been proved to be a powerful tool for the rapid sample treatment of liquid samples providing at the same time high enrichment factors and extraction recoveries. A new, simple and easy to handle one step in-syringe set-up for DLLME is presented and critically discussed in this paper. The novel approach avoids the centrifugation step, typically off-line and time consuming, opening-up a new horizon on DLLME automation. The suitability of the proposal is evaluated by means of the determination of non-steroidal anti-inflammatory drugs in urine by liquid chromatography/ultraviolet detection. In the presented approach an ionic liquid is used as extractant. The target drugs can be determined in urine within the concentration range 0.02–10 μg mL−1, allowing their determination at therapeutic and toxic levels. Limits of detection were in the range from 8.3 ng mL−1 (indomethacin) to 32 ng mL−1 (ketoprofen). The repeatability of the proposed method expressed as RSD (n = 5) varied between 2.5% (for ketoprofen) and 8.6% (for indomethacin).  相似文献

Four simple, rapid, accurate, precise, reliable and economical spectrophotometric methods have been proposed for simultaneous determination of salbutamol sulphate (SS), bromhexine hydrochloride (BH) and etofylline (ET) in pure and commercial formulations without any prior separation or purification. They were first derivative zero crossing spectrophotometry (method 1), simultaneous equation method (method 2), derivative ratio spectra zero crossing method (method 3) and double divisor ratio spectra derivative method (method 4). The ranges for SS, BH and ET were found to be 1-35 μg mL−1, 4-40 μg mL−1 and 5-80 μg mL−1. For methods 1 and 2, the values of limit of detection (LOD) were 0.2314 μg mL−1, 0.4865 μg mL−1 and 0.2766 μg mL−1 and the values of limit of quantitation (LOQ) were 0.7712 μg mL−1, 1.6217 μg mL−1 and 0.9221 μg mL−1 for SS, BH and ET, respectively. For method 3, LOD values were 0.3297 μg mL−1, 0.2784 μg mL−1 and 0.7906 μg mL−1 and LOQ values were 0.9325 μg mL−1, 0.9282 μg mL−1 and 2.6352 μg mL−1 for SS, BH and ET, respectively. For method 4, LOD values were 0.3161 μg mL−1, 0.2495 μg mL−1 and 0.2064 μg mL−1 and LOQ values were 0.9869 μg mL−1, 0.8317 μg mL−1 and 0.6879 μg mL−1 for SS, BH and ET. The precision values were less then 2% R.S.D. for all four methods. The common excipients and additives did not interfere in their determinations. The results obtained by the proposed methods have been statistically compared by means of Student t-test and by the variance ratio F-test.  相似文献

A simple, rapid and efficient method, dispersive liquid-liquid microextraction (DLLME), has been developed for the extraction and preconcentration of polybrominated diphenyl ethers (PBDEs) in water samples. The factors influencing microextraction efficiencies, such as the kind and volume of extraction and dispersive solvent, the extraction time and the salt effect, were optimized. Under the optimum conditions (sample volume: 5 mL; extraction solvent: tetrachloroethane, 20.0 μL; dispersive solvent: acetonitrile, 1.00 mL; extraction time: below 5 s and without salt addition), the enrichment factors and extraction recoveries were high and ranged from 268 to 305 and 87.0 to 119.1%, respectively. Linearity was observed in the range 0.05-50 ng mL−1 for BDE-28 and BDE-99, and 0.1-100 ng mL−1 for BDE-47 and BDE-209, respectively. Coefficients of correlation (r2) ranged from 0.9995 to 0.9999. The repeatability study was carried out by extracting the spiked water samples at concentration levels of 50 ng mL−1 for BDE-28 and BDE-99, and 100 ng mL−1 for BDE-47 and BDE-209, respectively. The relative standard deviations (R.S.D.s) varied between 3.8 and 6.3% (n = 5). The limits of detection (LODs), based on signal-to-noise ratio (S/N) of 3, ranged from 12.4 to 55.6 pg mL−1 (the wavelength of detector at 226 nm). The relative recoveries of PBDEs from tap, lake water and landfill leachate samples at spiking levels of 5, 10 and 50 ng mL−1 were in the range of 89.7-107.6%, 114.3-119.1% and 87.0-90.9%, respectively. As a result, this method can be successfully applied for the determination of PBDEs in landfill leachate and environmental water samples.  相似文献

Although there are a number of existing assays for monitoring the activity of both isopenicillin N synthase (IPNS) and deacetoxycephalosporin C synthase (DAOCS), none have demonstrated the qualities required for screening a mutant library. Hence, enzyme-linked immunosorbent assays (ELISAs) for IPNS and DAOCS were developed based on the detection of the catalytic turnover products isopenicillin N and cephalexin/phenylacetyl-7-aminodeacetoxycephalosporanic acid (G-7-ADCA), respectively. These assays are relatively fast compared to existing assays, such as the hole-plate bioassay, and are amenable with high-throughput screening. Both the IPNS and DAOCS-ELISAs were optimised for use with crude protein extracts rather than purified protein, thereby eliminating any additional time required for purification. The ELISA developed for the detection of cephalexin had an IC50 value of 154 ± 9 ng mL−1 and LOD of 7.2 ± 2.2 ng mL−1 under conditions required for the assay. Good recoveries and correlation was observed for spiked samples when the concentration of crude protein was kept below 1 mg mL−1. The DAOCS-ELISA was found to have increased sensitivity compared to the hole-plate bioassay (10.3 μg mL−1). The IPNS-ELISA did not significantly increase the sensitivity (approximately 5 μg mL−1) compared to that of the hole-plate bioassay (16 μg mL−1) for isopenicillin N. The minimum amount of crude protein extract required for producing detectable amounts of product for both assays was below 0.5% of the maximum amount of protein that the assay could contain without any effect on the ELISA. This suggests that when screening a mutant library, mutants producing low amounts of the product could still be detected using these assays.  相似文献

A novel fluorescent probe for Cu2+ determination based on the fluorescence quenching of glyphosate (Glyp)-functionalized quantum dots (QDs) was firstly reported. Glyp had been used to modify the surface of QDs to form Glyp-functionalized QDs following the capping of thioglycolic acid on the core–shell CdTe/CdS QDs. Under the optimal conditions, the response was linearly proportional to the concentration of Cu2+ between 2.4 × 10−2 μg mL−1 and 28 μg mL−1, with a detection limit of 1.3 × 10−3 μg mL−1 (3δ). The Glyp-functionalized QDs fluorescent probe offers good sensitivity and selectivity for detecting Cu2+. The fluorescent probe was successfully used for the determination of Cu2+ in environmental samples. The mechanism of reaction was also discussed.  相似文献
