An asymmetric synthesis of the 3-substituted Δ3,4-unsaturated prolines 7a-e and 3-substituted 4-methylene prolines 14a-c starting from the corresponding γ,δ-unsaturated α-amino acids 4a-e and 11a-c, respectively, is described. Amino acid derivatives 4a-e and 11a-d were obtained through aminoalkylation of the corresponding sulfoximine substituted allyl titanium(IV) complexes 2a-e and 10a-d, respectively, with the N-tert-butylsulfonyl imino ester 3. Activation of sulfoximines 4a-e and 11a-c through methylation of the sulfoximine group followed by a KF mediated isomerization of the vinyl aminosulfoxonium salts 5a-e and 12a-c, respectively, to the corresponding allyl aminosulfoxonium salt 6a-e and 13a-c, respectively, and a subsequent intramolecular nucleophilic substitution of the allylic aminosulfoxonium group afforded the enantio- and diastereomerically pure proline derivatives in medium to high yields.  相似文献

Selective addition of the chiral, sulfonimidoyl substituted bis(allyl)titanium complexes 5a-d, which are configurationally labile in regard to the Calpha-atoms, to N-toluenesulfonyl (Ts)-, N-2-trimethylsilylethanesulfonyl (SES)-, and N-tert-butylsulfonyl (Bus) alpha-imino ester (9a-c) in the presence of Ti(OiPr)(4) and ClTi(OiPr)(3) afforded with high regio- and diastereoselectivities in good yields the (syn, E)-configured beta-alkyl-gamma,delta-unsaturated alpha-amino acid derivatives 2a-g, which carry a chiral, electron-withdrawing nucleofuge at the delta-position and a cyclohexyl, an isopropyl, a phenyl, and a methyl group at the beta-position. Addition of the cyclic bis(allyl)titanium complex 14 to N-Bus alpha-imino ester 9c afforded with similar high regio- and diastereoselectivities the (E)- and (Z)-configured amino acid derivatives (E)-8 and (Z)-8. Reaction of complexes 5a-d with alpha-imino esters 9a-c in the presence of Ti(OiPr)(4) occurs stepwise to give first the mono(allyl)titanium complexes containing 2a-g as ligands, which react in the presence of ClTi(OiPr)(3) with a second molecule of 9a-c with formation of two molecules of 2a-g. Formation of (S,R,E)-configured homoallylic amines 2a-g entails Si,Re,E processes of alpha-imino esters 9a-c with the (R,R)-configured bis(allyl)titanium complexes (R,R)-5a-d and (R)-configured mono(allyl)titanium complexes (R)-17a-d, both of which are most likely in rapid equilibrium with their (S,S)-diastereomers and (S)-diastereomers, respectively. Interestingly, in the reaction of 5a-d with aldehydes, the (S,S)-configured complexes (S,S)-5a-d are the ones which react faster. Reaction of the N-titanated amino acid derivatives Ti-2a and Ti-2b with N-Ts alpha-imino ester 9a led to the highly diastereoselective formation of imidazolidinones 15a and 15b, respectively. Cleavage of the sulfonamide group of the N-Bus amino acid derivative 2d with CF(3)SO(3)H gave quantitatively the sulfonimidoyl functionalized amino acid H-2d. A Ni-catalyzed cross-coupling reaction of the amino acid derivative 2e with ZnPh(2) led to a substitution of the sulfonimidoyl group by a phenyl group and furnished the enantiomerically pure protected alpha-amino acid Bus-1. Two new N-sulfonyl alpha-imino esters, the SES and the Bus alpha-imino esters 9b and 9c, respectively, have been synthesized from the corresponding sulfonamides by the Kresze method in medium to good yields. The N-SES alpha-imino ester 9b and the N-Bus alpha-imino ester 9c should find many synthetic applications, in particular, in cases where the N-Ts alpha-imino ester 9a had been used before.  相似文献

Variable-temperature (1)H and (13)C NMR spectroscopy of the sulfonimidoyl-substituted allyltitanium(IV) complexes E-1a-c and Z-1a-c, which carry diethylamino groups at the Ti atom, revealed a fast 1,3-C,N-shift of the Ti atom, leading to an equilibrium between the epimeric Calpha-titanium allyl complexes A and C and the N-titanium allyl aminosulfoxonium ylide B. Based on these findings a model for the reactions of E-1a-c and Z-1a-c with aldehydes is proposed, which features regio- and diastereoselective reactions of the N-titanium ylide B at the alpha-position and the Calpha-titanium complex A at the gamma-position. Model ab initio calculations of the methylene and allyl (dimethylamino)sulfoxonium ylides 10 and 14, respectively, revealed short Calpha-S bonds, a stabilization by both electrostatic interaction and negative hyperconjugation, and a low Calpha-S rotational barrier. The ylides preferentially adopt Calpha-S and Calpha-N conformations in which the lone pair orbital at the Calpha atom is periplanar to the S=O bond and that at the N atom periplanar to the Calpha-Ph bond. Variable-temperature NMR spectroscopy of the sulfonimidoyl-substituted alkyltitanium(IV) complex 16, which carries diethylamino groups at the Ti atom, revealed a dynamic behavior leading to a complete topomerization of all four methylene hydrogens of the Calpha-ethyl groups. Two fast processes are held responsible for the topomerization of the hydrogens of 16. The first one is a reversible intramolecular beta-hydride elimination/alkene-Ti-H insertion with the intermediate formation of a complex between (Et(2)N)TiH and a 1-alkenyl sulfoximine, and the second one consists of a reversible 1,3-C,N-shift of the Ti atom in combination with a Calpha-S bond rotation. Interestingly, the room-temperature NMR spectra of the corresponding sulfonimidoyl-substituted alkyltitanium(IV) complex 17, which carries isopropoxy groups at the Ti atom, give no indication of a similar dynamic behavior of this complex.  相似文献

Asymmetric allylidenation of aldehydes with sulfur ylides is possible with proper substitution of the initial sulfide, to avoid the [2,3] sigmatropic rearrangement of the unsaturated ylides. One-pot reaction of (2R,5R)-dimethylthiolane with allyl halides, aldehydes, and sodium hydroxide in tert-butyl alcohol affords vinyl oxiranes in good yields. Enantiomeric excesses up to 90% and trans selectivities have been achieved with methallyl-type halides.  相似文献

Bis-BINOLs 1a,b in which two BINOL units are tethered by o- and m-phenylenebis(ethynyl) groups form stable dinuclear bis-BINOLate titanium(IV) complexes 2a,b by treatment with titanium tetraisopropoxide. In the presence of excess titanium tetraisopropoxide, 2a and 2b (2–20 mol %) catalyze diethylzinc addition to aromatic and aliphatic aldehydes in an efficient manner to give the ethylation products with high enantioselectivities. While more than 1 equiv of titanium tetraisopropoxide (with respect to a substrate aldehyde) is generally employed for obtaining high turnover frequency and selectivity in reactions catalyzed by a parent (BINOLate)Ti(OiPr)2, the amount can be reduced as low as 0.2 equiv in the reactions catalyzed by 2a,b.  相似文献

A new bifunctional chiral Al(III) complex of BINOL derivative, which contained tert-amine at 3,3′-position of the BINOL, has been developed for the effective enantioselective hydrophosphonylation of aldehydes. A variety of aromatic, heteroaromatic, condensed-ring, α,β-unsaturated, and aliphatic aldehydes were found to be suitable substrates for the reaction, and the desired α-hydroxy phosphonate were obtained in good to excellent yields (up to 99%) with moderate to good enantioselectivities (up to 87% ee) under mild conditions (at 0 °C). A possible catalytic cycle based on the experimental results was proposed.  相似文献

The diastereoselective syntheses of Zn(II) bis(dipyrrinato) helicates is reported, involving ligands templated by the incorporation of homochiral binol within the linker joining the two dipyrrinato units. The most diastereoselective formation of dinuclear bis(dipyrrinato) helicates to date is reported. The formation of either mononuclear or dinuclear helicates can be tuned by varying the length of the linker between the dipyrrinato units and by varying the complexation procedure. The neutral dipyrrinato helicates were readily analyzed by HPLC to ascertain diastereoselectivity, and circular dichroism studies revealed the helical nature of the complexes. The molar ellipticities of the helicates produced by diastereoselective complexation are very large in the visible region and typically correspond to binol moieties in the UV region. Extensive X-ray crystallographic investigations further confirmed the helicity of the mononuclear Zn(II) helicates and identified significant interlayer displacement and bending within crystals.  相似文献

Asymmetric syntheses of the iodomethyl-substituted bicyclic tetrahydrofuran 22 and the chloro-amino sulfone 30 from the allylic sulfoximine 15 and the alpha-hetero aldehydes 2 and 23, respectively, are described. Further examples for the asymmetric synthesis of chloromethyl tetrahydrofurans and chloro-amino sulfones are given. The synthesis of 30 features as key step the stereoselective Cl-substitution of a hydroxy group under neighboring group participation by an aminosulfoxonium group which is converted to a sulfonyl group. We have shown that Me-DuPHOS monoxide (BozPHOS) is a very effective ligand in the copper-catalyzed addition of dialkylzinc to N-phosphinoylimines providing access to alpha-chiral amines. The new ligand is particularly effective for the addition of the lesser reactive dimethylzinc. The major advantages of this process are high yields, broad substrate scope, and high enantioselectivities with low catalyst loading (3 mol %).  相似文献

Enantioselective catalytic trimethylsilylcyanations of aldehydes with 48% to 92% e.e. have been studied using the novel Ti-chiral Schiff base complexes. We have found that the catalyst led to high enantoselectivity when the molar ratio of the Schiff base 1 to Ti(O-i-Pr)4 was 2:1.  相似文献

Titanium(IV)(salen) and vanadium(V)(salen) complexes are both known to form catalysts for asymmetric cyanohydrin synthesis. When a mixture of titanium and vanadium complexes derived from the same or different salen ligands is used for the asymmetric addition of trimethylsilyl cyanide to benzaldehyde, the absolute configuration of the product and level of asymmetric induction can only be explained by in situ formation of a catalytically active heterobimetallic complex, and is not consistent with two monometallic species acting cooperatively. Combined use of complexes containing chiral and achiral salen ligands demonstrates that during the asymmetry inducing step of the mechanism, the aldehyde is coordinated to the vanadium rather than the titanium ion. The titanium complexes also catalyse the asymmetric addition of ethyl cyanoformate to aldehydes, a reaction in which vanadium(V)(salen) complexes are not active. For this reaction, use of a mixture of titanium and vanadium(salen) complexes results in a complete loss of catalytic activity, a result which again can only be explained by in situ formation of a heterometallic complex. Both the titanium and vanadium based catalysts also induce the asymmetric addition of potassium cyanide/acetic anhydride to aldehydes. For this reaction, combined use of chiral and achiral complexes indicates that during the asymmetry inducing step of the mechanism, the aldehyde is coordinated to titanium rather than vanadium, a result which contrasts with the observed results when trimethylsilyl cyanide is used as the cyanide source.  相似文献

The asymmetric cyclopropanation of a vinylphosphonate using optically active sulfonium and selenonium ylides derived from (−)-menthol and (+)-limonene was developed. The ylides were generated in situ by the reaction of the corresponding sulfonium or selenonium salt in the presence of potassium carbonate or DBU as a base. The transfer of the CHPh and CHCO2Et groups into the cyclopropane ring showed moderate diastereoselectivity and excellent enantioselectivity (up to 99:1) for the trans- and cis-products. The absolute configuration of phenyl cyclopropyl was assigned based on comparison to their tolyl analogues.  相似文献

The first application of chiral auxiliaries synthesized from levoglucosenone (a biomass-derived anhydrosugar) in asymmetric 1,3-dipolar cycloaddition reactions of azomethine ylides is herein reported. The corresponding pyrrolidinic cores were obtained in excellent levels of regio and stereocontrol, good to excellent π-facial selectivities, and could be isolated enantiomerically pure by column chromatography. Unexpected NMR observations coupled with DFT calculations allowed the stereochemical assignment of the synthesized adducts. The stereochemical assignment performed in silico was further unambiguously validated by structural X-ray diffraction analysis.  相似文献

Reaction of bis(amide) sodium Na2[(1R,2R)-(−)-1,2-(NSiMe3)2-C6H10] (Na2[L1]) with Ti(OiPr)2Cl2 in different conditions gave mixed-ligand complexes [Ti(OiPr)Cl][L1] (1) or [Ti(OiPr)2Cl]2[L1] (2); 2 is a dinuclear titanium example in which Ti atoms are bridged by nitrogen and oxygen atoms simultaneously forming a distorted rhombic core. Reaction of the amine-amidinate ligand (1R,2R)-(−)-1-Li[NC(Ph)N(SiMe3)]-2-(NHSiMe3)-C6H10(Li[L2]) or rarely linked bis(amidinate) ligand Li2[(1R,2R)-(−)-1,2-{NC(Ph)N(SiMe3)}2-C6H10](Li2[L3]) with ZrCl4 yielded the unbridged and bridged bis(amidinate) complexes ZrCl2[L2]2 (3) and [ZrCl2(THF)][L3] (4), respectively; Moreover, the reaction of (1R,2R)-(−)-1-Li[NC(Ph)N(SiMe3)]-2-Li(NSiMe3)C6H10(Li2[L2]) with Ti(OiPr)2Cl2 gave a new type of tridentate amido-amidinate product [Ti(OiPr)2][L2] (6), which is a distinct model compared to [Ti(OiPr)2Cl][L2] (5) yielded from Li[L2]. All the products have been characterized by X-ray crystallography and the structural studies are presented detailedly comparing with relevant compounds.  相似文献

Described is an asymmetric synthesis of new Delta(3a,4)-unsaturated, fused bicyclic proline analogues from cyclic bis(allylsulfoximine)titanium complexes and N-tert-butylsulfonyl imino ethyl ester. Treatment of the enantiomerically pure five-, six-, seven-, and eight-membered cyclic bis(allylsulfoximine)titanium complexes with the imino ester gave mixtures of the corresponding (E,syn)- and (Z,syn)-configured, delta-sulfoximine substituted, cyclic gamma,delta-unsaturated alpha-amino acid esters with high regio- and diastereoselectivities in good yields. Activation of the N-methyl sulfoximine group of these amino acid derivatives through methylation with Me(3)OBF(4) afforded in nearly quantitative yields the corresponding (dimethylamino)sulfoxonium salts. A novel migratory cyclization of these salts with DBU gave via an isomerization to the corresponding allylic (dimethylamino)sulfoxonium salts and an intramolecular substitution of the (dimethylamino)sulfoxonium group the enantio- and diastereomerically pure, bicyclic, N-tert-butylsulfonyl protected proline analogues having a six- and eight-membered unsaturated carbocyclic ring. Cyclization of the alkenyl (dimethylamino)sulfoxonium salts was independent of the configuration of the double bond. N,N-Dimethylphenylsulfinamide of > or =99% ee was obtained in good yield as a further reaction product. Conversion of the sulfinamide to N,S-dimethyl-S-phenylsulfoximine of > or =99% ee, the starting material for the synthesis of the allylic sulfoximines, had been accomplished previously. Finally, cleavage of the tert-butylsulfonyl protecting group with anhydrous acid furnished the fused bicyclic proline analogue containing an unsaturated six-membered ring in high yield.  相似文献
