The chemical composition of the functional surfaces of substrates used for microarrays is one of the important parameters that determine the quality of a microarray experiment. In addition to the commonly used contact angle measurements to determine the wettability of functionalized supports, X-ray photoelectron spectroscopy (XPS) and time-of-flight secondary ion mass spectrometry (ToF-SIMS) are more specific methods to elucidate details about the chemical surface constitution. XPS yields information about the atomic composition of the surface, whereas from ToF-SIMS, information on the molecular species on the surface can be concluded. Applied on printed DNA microarrays, both techniques provide impressive chemical images down to the micrometer scale and can be utilized for label-free spot detection and characterization. Detailed information about the chemical constitution of single spots of microarrays can be obtained by high-resolution XPS imaging. The chemical analysis of microparticles is challenging due to the need to mount the particles on a substrate for analysis; double-sided adhesive tape is often used (sometimes conductive), however that is usually coated with poly (dimethyl siloxane) (PDMS) that is often used as a release agent. PDMS is a common surface contamination that can mask surface chemistries and hinder material performance where it is dependent on this contaminated interface. It is known that PDMS contains a very mobile oligomeric fraction that readily diffuses across surfaces resulting in the contamination of mounted particulate samples before and during surface chemistry analysis. This makes it impossible to determine whether the PDMS has arisen from the analysis procedure or from the sample itself. A new sample preparation method is proposed where polymer microparticles are mounted on a poly (hydroxyethyl methacrylate) (pHEMA) polymer solution, which we compare with particles that have been mounted on adhesive discs using time-of-flight secondary ion mass spectrometry (ToF-SIMS) and 3D OrbiSIMS analysis. Particles mounted on the pHEMA substrate results in a reduction of PDMS signal by 99.8% compared with microparticles mounted on adhesive discs. This illustrates how a simple, quick and inexpensive polymer solution can be used to adhere particles for analysis by ToF-SIMS, or other surface chemical analysis techniques such as X-ray photoelectron spectroscopy (XPS), without introduction of large amounts of silicone contaminant.  相似文献

Epoxy resin composites reinforced with E-glass (E), 3D glass (3D) and carbon fibre (CF) were subjected to an intense UV and high temperature accelerated degradation environment. X-ray photoelectron spectroscopy (XPS) and time-of-flight secondary ion mass spectrometry (ToF-SIMS) were used to provide a molecular characterisation of the surface of the degraded composites. The response at the surface of the epoxy resin composites to oxidative degradation is influenced by the composite reinforcement type and characteristics. XPS results indicate that 3D resin composites exhibit more surface oxidation as a result of the accelerated degradation in comparison with E and CF composites. Principal components analysis (PCA) of the ToF-SIMS positive ion spectra showed that E and 3D resin composites suffered chain scission while CF composites suffered chain scission and cross-linking reactions as a result of the intense UV exposure. The extent of the surface oxidation, cross-linking/condensation reaction and loss of low molecular weight (lower than C4Hx) aliphatic hydrocarbons may be indicated using PCA of both the ToF-SIMS positive and negative ion spectra. PCA also provides insight for proposing epoxy resin chain scission and oxidation reaction mechanisms.  相似文献

应用傅里叶转换红外光谱(FTIR)和X射线光电子能谱(XPS), 研究了3种提纯方法得到的竹木质素及其化学反应产物的化学结构特性. 确定竹木质素C1s的电子结合能分别为283.52 (C—H或C—C), 284.58~285.72 (C—OR或C—OH), 286.10~286.44 (C=O或HO—C—OR), 287.65~287.72 (O—C=O) eV. O1s的电子结合能分别为530.31(羟基氧原子), 531.45~531.72(醛或酮的羰基氧原子), 532.73~533.74(酯键或羧酸中的羰基氧原子) eV. 竹木质素中的结构单元之间主要是通过醚键和碳碳单键连接, 慈竹磨木木质素结构单元中醚键、碳碳单键、酯键、羰基和烯双键的比例为100∶63∶32∶40∶32 (49.3∶31.0∶16.0∶19.9∶16.0)  相似文献

David Briggs was a surface analysis pioneer. Starting in 1970 and continuing throughout his career, Dave used his expertise, vision, and ability to quickly master new surface analysis methods and solve important industrial problems. It certainly helped that he was an outstanding fundraiser in both industrial and academic settings, which ensured he always had an impressive array of the latest, most advanced surface analysis instrumentation at his disposal. He insisted on doing surface analysis correctly, and through his publications, databases, and books, he provided the community with the needed guidelines and methods to do so. In the 1970s, Dave's research was largely focused on X-ray photoelectron spectroscopy (XPS, also known as electron spectroscopy for chemical analysis [ESCA]) characterization of polymers and catalysts. He added secondary ion mass spectrometry (SIMS) to his instrumentation arsenal in the 1980s and provided many of the key, pioneering publications that described how to use this method to characterize polymer surfaces. He also did some of the first surface analysis imaging experiments in the 1980s. In the 1990s, he continued his XPS and SIMS research on polymers and advanced the surface analysis community's ability to properly interpret surface analysis data through databases and advanced data processing methods. Dave continued to publish polymer and catalysis surface analysis papers in the 2000s, but also expanded his surface analysis studies to several other topics.  相似文献

XPS was used to characterize the chemical changes occurring after drying or applying a heat‐treatment to beech wood samples. Our results indicate that the surface of this air‐exposed material could be strongly affected either by the ambient atmosphere during storage or by the complex atmosphere in the oven during drying or heat‐treatment. However, the O/C ratio measured after removal of a thin slice of a few millimetres of an untreated sample is in reasonable agreement with that calculated from the well‐established chemical composition of beech. Through this methodology (equivalent to scraping for hard materials) it is expected to get a realistic characterization of the wood. The reliability and repeatability of the XPS measurements have been checked and the method applied to the study of the chemical changes of the beech samples subjected to heat‐treatment. Heating at 240 °C induces a significant decrease of the O/C ratio from 0.55 before to 0.44 after the treatment. Heat‐treatment induces also a decrease of the C2 carbon contribution (carbon atom bound to a single non‐carbonyl oxygen) associated with an increase of the C1 carbon contribution (carbon atoms bound only to carbon or hydrogen atoms), in agreement with chemical modifications reported previously in the literature. Thanks to the small analysed area of the equipment used in this study, different spots were analysed to demonstrate the presence or absence of a gradient of chemical composition due to thermal degradation or migration of extractives from within the wood structure to its surface. The secondary ion mass spectrometry method (ToF-SIMS) has been applied to the analysis of some mineral and organic species on the surface layers of airborne urban particulates. Particulate matter was collected on aluminum sheets of the 8-stage Anderson type cascade impactor from three different sampling sites in the city of Lodz, Poland, which were different in terms of aerosol source apportionment. The obtained results show that vehicle combustion products occur mainly in fine fraction with aerodynamic diameters below 1μm. The main contribution to the mineral components comes from direct and traffic related soil resuspension.   相似文献

Time-of-flight secondary ion mass spectrometry (ToF-SIMS) was employed for an analysis of the rare earth element Ce in pure iron and steel. Secondary ion images showed that the amount of the segregation of Ce and its oxides was much less as the concentration of Ce decreased to about 11 ppm in the pure iron. In terms of depth profiling from the surface to the matrix, variation of the CeO+/Ce+ ratio in a Ce oxides sample and a pure Ce sample exhibited difference between oxidized state and metallic state. It was demonstrated that metallic Ce was presented when the ratio of CeO+/Ce+ decreased to approximately 2 and even smaller. Oxygen content with concentration of tens of ppm may influence the ratio obviously. Such ToF-SIMS analysis was also feasible to discern the states of cerium in a Q345E steel with concentration in ppm level.  相似文献

Time-of-flight secondary ion mass spectrometry (ToF-SIMS) is a powerful tool for surface analysis, but fragmentation of molecular species during the SIMS process may lead to complex mass spectra. While the fragmentation pattern is typically characteristic for each compound, industrial samples are engineered materials, and, thus, may contain a mixture of many compounds, which may result in a variety of overlapping peak patterns in ToF-SIMS spectra. Consequently, the process of data evaluation is challenging and time-consuming. Principal component analysis (PCA) can be used to simplify data analysis for complex sample systems. Especially, correlation loadings were observed as an ideal tool to identify relevant signals in PCA results, which induce the separation of different sample groups. This is because correlation loadings show the relevance of signals independent from their intensity in the raw data. In correlation loadings, however, fragmentation patterns are no longer observed and the identification of peaks' sum formulas is challenging. In this study, a new approach is presented, which simplifies peak identification and assignment in ToF-SIMS spectra after PCA is performed. The approach uses a mathematical transformation that projects PCA results, in particular loadings and correlation loadings, in the direction of specific sample groups. The approach does not change PCA results but rather presents them in a new way. This method allows to visualize characteristic spectra for specific sample groups that contain only relevant signals and, additionally, visualize fragmentation patterns. Data analysis is simplified and helps the user to focus on data interpretation rather than processing.  相似文献

Time-of-flight secondary ion mass spectrometry (ToF-SIMS) equipped with a gold ion gun was used to image mouse embryo sections and differentiate tissue types (brain, spinal cord, skull, rib, heart and liver). Embryos were paraffin-embedded and then deparaffinized. The robustness and repeatability of the method was determined by analyzing ten tissue slices from three different embryos over a period of several weeks. Using principal component analysis (PCA) to reduce the spectral data generated by ToF-SIMS, histopathologically identified tissue types of the mouse embryos can be differentiated based on the characteristic differences in their mass spectra. These results demonstrate the ability of ToF-SIMS to determine subtle chemical differences even in fixed histological specimens.  相似文献

采用XPS对六枝(Liuzhi)和遵义(Zunyi)原煤及其固定床热解半焦表面的含碳官能团和硫迁移行为进行了研究。在氮气气氛下,在400℃~700℃遵义煤焦表面O=C-O, C-H 和 C-C含量与原煤相比,呈现下降趋势,而C=O和O-C-O、C-O含量却呈现增加趋势; 六枝煤焦表面的O=C-O、C=O和O-C-O、C-O含量的变化规律与遵义煤相似,而C-H和C-C含量的变化却与遵义煤相反。在氢气气氛下, 遵义煤焦表面不仅O=C-O含量与原煤相比下降, 而且C=O和O-C-O、C-O含量也下降,但C-H和C-C含量却增加; 六枝煤除了C=O 和O-C-O含量下降外, 其他官能团的变化与其在氮气气氛下相似。两种煤焦表面S/C比在氮气气氛下随着温度的升高而升高,氢气气氛下在400℃~600℃随着温度的升高却下降,700℃又显著升高。这说明在热解过程中硫在两种气氛下都能从体相向表面迁移,但是氢气能够和含硫自由基反应,所以氢气气氛下煤焦表面的S/C比率低于其氮气气氛下的。  相似文献

Size-fractionated aerosol particles were collected with a MOUDI 10-stage cascade impactor from an urban roadside place in a downtown area of Hong Kong. Fine aerosol particulate samples from stage 6 (aerodynamic particle diameter between 0.56 and 1 μm) and stage 9 (aerodynamic particle diameter between 0.10 and 0.18 μm) were pretreated at a chosen temperature, including −100°C, −50°C, 25°C, and 60°C, in a load lock chamber and then analyzed using time-of-flight secondary ion mass spectrometry (ToF-SIMS) at the same temperature (−100°C). Principal component analysis (PCA) was applied to further analyze ToF-SIMS spectra of aerosol particles with different pretreatment temperatures from two selected stages. ToF-SIMS results showed that the intensities of aliphatic hydrocarbon ions such as C4H7+ and C4H9+ and amine ions such as C2H8N+ and C4H12N+ decreased with an increase of the pretreatment temperature under ultrahigh vacuum conditions. We have shown that analyses of this type of aerosol particles using ToF-SIMS should not be conducted at ambient temperature but at low temperature (eg, −50°C). In addition, we also developed a procedure that can be used to analyze aerosol particle samples under ultrahigh vacuum environment.  相似文献

Inverse gas chromatography (IGC) was used to determine the dispersive component of the free energy as well as the acid-base properties of cellulose fibre surfaces, before and after modification by corona treatment. It was found that the corona treatment increases both the dispersive contribution to surface energy and its acidic character, whereas only a slight increase in its basicity was observed. It was also found that some chemical degradation of the surface occurs at high corona currents. The extent of modification of the surface properties, as revealed by IGC, was correlated to the surface chemical composition deduced from XPS analysis as well as with the electrical conductance and the pH of the water suspensions of the cellulose fibres.  相似文献

Cisplatin has been clinically used for treatment of solid tumors such as non–small-cell lung cancer for decades. However, tumor resistance may be acquired with losing the antitumor activity of cisplatin. As cellular membrane is the first barrier that cisplatin has to overcome before its further action inside the cells, the membrane composition must play a vital role in the cisplatin uptake and excretion, which further influences cisplatin sensitivity. In this work, we applied time-of-flight secondary ion mass spectrometry (ToF-SIMS) surface analysis combined with principle component analysis to distinguish the differences of cell membrane composition between non–small-cell lung cancer cells (A549) and its cisplatin resistant counterpart A549/DDP cells. The decreased phosphatidylcholine content and more abundant cholesterol were observed in the drug resistant cell surfaces, indicating the decreased membrane fluidity of A549/DDP cells. Moreover, we further compared membrane composition of A549 and A549/DDP cells after being treated with different concentrations of cisplatin. A higher composition level of proteins was discovered on all groups of A549/DDP cell membranes. The altered surface chemistry of cellular membranes induced by cisplatin indicates the significance of membrane structures in the drug resistance, which deserves further investigations to this regard.  相似文献

Artificial ageing of polymeric insulation jackets is routinely performed in order to assess end-of-life material characteristics. Practical constraints including high temperatures/short times ageing treatments lead to strong influence of diffusion-limited oxidation (DLO) resulting in unreliable life-time predictions. This study proposes a new experimental approach to the investigation of cable insulation ageing, exploiting analytical techniques capable of resolving chemistry at the length scale relevant for DLO (nano-microscale). When studying the potential effects of DLO using time of flight secondary ion mass spectrometry (ToF-SIMS) sample preparation becomes crucial. This paper presents the development of a methodology to generate suitable specimens to investigate the DLO effect using ToF-SIMS. A reference polymeric material has been thermally aged in various DLO conditions. Cross sections of aged samples were generated using three different methods. In order to assess the most suitable approach for this study, cross-section topography were scanned using a profilometer and the surface chemistry was investigated using ToF-SIMS together with multivariate analysis methods.  相似文献

In this paper the microfabrication of ITO (tin‐doped indium oxide) films by the sol–gel process combined with chemical modification is presented. The microfabricated ITO thin film could be obtained through a one‐step process that combines film patterning with film leaching. The morphology and chemical components of the patterned ITO thin films were assessed by microscopy and XPS, respectively. In recent years developments in the capabilities of techniques such as XPS and static SIMS have led to wider application of these methods for the characterisation of industrial materials. After a brief discussion of recent key developments of these techniques, this paper presents a selection of results from research work at our laboratory in the characterisation of organic film-forming corrosion inhibitors on a range of metal substrates which illustrate the nature of the information available. The inherent advantage ofsurface sensitivity and the advantages of a multi-technique approach for the evaluation of surface molecular structure from complex organic compounds is discussed. The additional benefits for analysis offered by the latest developments in technique are also demonstrated.  相似文献

Model samples of the interface of an adhesive joint containing small levels of aminopropyl triethoxysilane (APS) have been prepared in order to examine the interface formed with an aluminium substrate. X‐ray photoelectron spectroscopy (XPS) and time of flight secondary ion mass spectrometry (ToF‐SIMS) have been used to analyse and image the interface region in between the aluminium and an epoxy adhesive in order to ascertain the reactions by the organosilane which is present as a minor component within the system. It was found that APS was present at the interface between the adhesive and the substrate and that it had reacted with the substrate forming a covalent bond and was also crosslinked within the adhesive. 综述木材中五氯苯酚的检测方法。高效液相色谱法不需要复杂的衍生化操作,具有良好的选择性与较高的灵敏度,是高沸点、热稳定性较差的五氯苯酚较理想的分析方法,该方法简便,检测成本较低,易于在检测机构及企业推广应用;采取乙酰衍生化,应用毛细管色谱柱进行分离的气相色谱法(电子捕获检测器)是目前五氯苯酚常用的检测方法,该方法简单,但易存在假阳性结果且无法识别;电极法可避免使用有机溶剂,减少环境污染,但苦味酸根离子有明显干扰;气相色谱-质谱法可以准确地进行定性定量分析,有效地去除基质干扰,该方法选择性好,灵敏度高,检出限低,具有良好的发展前景。  相似文献
