A catalyst-free water-mediated N-Boc deprotection of N-Boc-amines is reported. In the absence of any additional reagents, the free amines were formed from a variety of aromatic and aliphatic N-Boc-amines as well as from some N-Boc-amino acid derivatives.  相似文献

In this Letter, we describe the unexpected reaction pattern of N,NN″-tri-Boc-guanidine (TBG) with amines at room temperature and under reflux conditions affording N-substituted guanidines and amidinoureas, potentially important compounds with extensive applications in medicinal chemistry. This investigation shows that TBG is an excellent, readily available common starting material for the synthesis of various N-alkyl guanidines as well as N-alkyl-N′-substituted amidinoureas by simply manipulating the reaction conditions.  相似文献

The geometry of N,N′-diphenyl-p-phenylenediamine (DPPD), N-phenyl-N′-(1′-methylbenzyl)-p-phenylenediamine (SPPD), N-phenyl-N′-(1,3-dimethyl-butyl)-p-phenylenediamine (6PPD), N-phenyl-N′-isopropyl-p-phenylenediamine (IPPD), and N-(1-methyl-1-phenylethyl)-N′-phenyl-p-phenylenediamine (CPPD) as well as of their dehydrogenation products has been optimized at B3LYP/6-31G level of theory. Our results support the idea of formation of stable ketimine Ph-NC structures (instead of quinonediimine structures) during consecutive dehydrogenation of SPPD, 6PPD, and IPPD antioxidants despite the formation of tertiary carbon-centered radicals in the first dehydrogenation step is energetically preferred for SPPD only.  相似文献

We report the synthesis and photophysical properties of N-alkyl- or N,N-dialkyl-pyrene-1-carboxamide. These derivatives, as well as pyrene, exhibited blue emission. N-Alkyl-type derivatives exhibited strong fluorescence emission (Φfl = 0.61 in EtOH) in both nonpolar and polar solvents. On the other hand, N,N-dialkyl-type derivatives showed weak fluorescence emission (Φfl <0.01) due to vibrational deactivation. However, in highly viscous solvents such as glycerin, the quantum efficiencies of N-alkyl-type (Φfl = 0.91) and N,N-dialkyl-type (Φfl = 0.082) derivatives were increased. We also investigated the fluorescence mechanism of these compounds using time-dependent density-functional theory (TD-DFT). From these results, we find that highly fluorescent pyrene-1-carboxamide derivatives can be designed by introducing an appropriate functional group at the nitrogen atom of the amide. Thus, N,N-dialkyl-type pyrene-1-carboxamide has considerable potential for use in applications such as environmental response sensors and probes.  相似文献

The stereoselective synthesis of N-acyl- and N-Boc-protected pyrrolidines via Pd-catalyzed reactions of γ-(N-acylamino) alkenes and γ-(N-Boc-amino) alkenes with aryl bromides is described. These reactions effect formation of two bonds in a single operation and proceed with generally high levels of diastereoselectivity. In contrast to previously described reactions of γ-(N-arylamino) alkenes, these transformations proceed in high yield and high regioselectivity with both electron-rich and electron-deficient aryl bromides as well as vinyl bromide substrates.  相似文献

The synthesis of two N-aryl substituted 2-silaimidazolidenes 9a, b by metal-reduction of the appropriate silicon(IV) heterocycles is reported. Structural as well as spectroscopic data obtained for the N-aryl substituted N-heterocyclic silylenes (NHSi) are very close to those obtained previously for their N-alkyl substituted counterparts. NHSis 9a, b are used as starting materials for the synthesis of a series of dichalcogenadisiletanes 19-24 and for of a mono silylene tungsten complex 29. The reactivity studies revealed only marginally differences between the N-aryl substituted NHSis 9a, b and previously described N-alkyl substituted silylenes.  相似文献

Codeine was synthesized with 90% enrichment of the N-methyl group with carbon-13. N-Demethylation of this substrate by cell-free extracts of Cunninghamella bainieri in an n.m.r, tube gave norcodeine and 13C-labelled formaldehyde. Fourier-transform 13C-n.m.r. spectroscopy was used to observe the N-demethylation process at selected temperatures. The labelled formaldehyde liberated was trapped with sodium sulphite, and the sulphite adduct, as well as intermediates, were located in the n.m.r, spectrum at each temperature. Intermediate resonances assignable to codeine-N-oxide were not detected during these enzyme-transformation studies. These data suggest that the observed 13C-n.m.r. signals correspond to the chemically labile carbinolamine intermediate formed during N-demethylation. A methine 13C signal was not observed. Thus, N-demethylation of codeine by Cunninghamella bainieri occurs by direct C-oxidation and not via an N-oxide intermediate.  相似文献

Diphenylphosphoryl azide (DPPA) mediated one-pot synthesis of Nα-Fmoc/Bsmoc/Boc/Z-protected ureidopeptides and peptidyl ureas as well as phenyl/succinimidyl (Nα-urethane protected) methyl carbamates starting from Nα-protected amino acids is reported. The formation of an azide, its rearrangement and coupling with an amino component is accomplished in a sequence of one-pot operations. The photophysical properties and photoreduction of N-acetylphthalimide (AcP) and N-benzoylphthalimide (BzP), N-3,4,5-trimethoxybenzoylphthalimide (trimethoxyBzP) and N-4-nitrobenzoylphthalimide (nitroBzP) were studied by steady-state and transient techniques. Radicals and their precursor triplet states were detected by flash photolysis. The triplet state properties of AcP and BzP were characterized. In contrast, no triplet absorption was observed with ns-detection for trimethoxyBzP and nitroBzP. Specific products are formed upon electron transfer from triethylamine to the photoexcited acylphthalimides. In addition, H-atom transfer from 2-propanol or other alcohols to the triplet state takes place. The properties of several radical intermediates involved in photoreduction of the acylphthalimides as well as some structure-function relationships are described.  相似文献

A facile synthetic route to N-polyfluoro(trimethylsilyl)ethyl azole derivatives was developed starting from N-bromo(chloro)polyfluoroethyl-substituted azoles. The silanes thus obtained were reacted with various electrophiles in the presence of the fluoride ion to yield the corresponding fluorinated carbinols, ketones, carboxylic acids, and methyl dithiocarboxylates as well as N-pentafluoroethylbenzimidazole.  相似文献

1-Substituted-5-alkylsulfanyl-1H-tetrazoles are well known class of organic substances with various applications in medicinal chemistry or photographic industry. Their selenium analogues, 1-substituted-5-alkylselanyl-1H-tetrazoles are, however, much less explored because of the lack of suitable methods for their preparation. In this work we investigated the synthesis of 1-alkyl/aryl-5-alkylselanyl-1H-tetrazoles from synthetically available alkyl/arylisoselenocyanates. One-pot reactions of arylisoselenocyanates with sodium azide and alkylating agent led to the target 5-alkylselanyl-1-aryl-1H-tetrazoles but also to interesting side products, namely N-alkyl-N-arylcyanamides and (Z)-Se-alkyl-N-cyano-N,N′-diarylisoselenoureas. Nevertheless, when alkylisoselenocyanates were utilized as the substrates, the reactions led exclusively to the formation of 1-alkyl-5-alkylselanyl-1H-tetrazoles in good yields. This simple one-pot procedure brings new possibilities for the preparation of variously substituted selenium compounds. It also opens the way to further investigations of selenium isosteres of the widely utilized 5-thiotetrazole moiety in biomedical applications.  相似文献

Over the past decade, sulfur-based fluoromethyl containing compounds have been exhaustively investigated as versatile fluoroalkylating reagents by our research laboratory as well as many others. Lately, we have designed a novel electrophilic difluoromethylating protocol employing in situ prepared N,N-dimethyl-S-difluoromethyl-S-phenylsulfoximinium salt. The present reagent provides excellent reactivity toward a broad spectrum of nucleophilic species (N-, P-, S-, and O-nucleophiles) to yield the corresponding difluoromethylated products with high efficacy under mild conditions. Additional studies have been performed to elucidate the mechanistic fundamentals of the reactions.  相似文献

An efficient method for the synthesis of N-acyl-benzotriazoles from a wide variety of protected amino acids, as well as from compounds frequently used in drug discovery such as biotin and N-Fmoc polyethylene glycol, has been developed. The reaction of carboxylic acid derivatives with benzotriazole in the presence of T3P® yielded the corresponding N-acyl-benzotriazoles, which were obtained in high purity following a simple work-up procedure, in most cases via precipitation of the desired product in water.  相似文献

A catalyst-free reaction of 1-(N-acylamino)alkyltriphenylphosphonium tetrafluoroborates with silyl enolates was developed to prepare β-amino carbonyl compounds. The reported method is a useful approach for the preparation of N-protected β-amino esters as well as N-protected β-amino ketones. The starting 1-(N-acylamino)alkyltriphenylphosphonium tetrafluoroborates are readily available from N-protected α-amino acids. Therefore, the presented approach can be considered a new method for the α-homologation of N-protected α-amino acids to prepare β-amino acid derivatives.  相似文献

A new class of S-linked 1,3,4-oxadiazole-tethered Nα-protected peptidomimetics is designed and synthesized by a reaction of Nα-Z/Boc-protected 1,3,4-oxadiazole-2-thiol with α-bromo ester derived from amino acid. The protocol has also been employed for the synthesis of glycosylated amino acids and N,N′-orthogonally protected dipeptidomimetics bearing S-linked 1,3,4-oxadiazole mimetics as well. The intermediate 5-alkyl amino-1,3,4-oxadiazole-2-thiols have been isolated as stable compounds. Further, the chain extension at both N- and C-termini of N-protected S-linked 1,3,4-oxadiazole tethered dipeptidomimetics was also demonstrated.  相似文献

We report the divergent effects of a 3a-methyl and 3a-phenyl substituent on the chemoselectivity and stereoselectivity of reduction of the enamide moiety of N-Boc-hexahydro-1H-indolin-5(6H)-ones. Under ionic reduction conditions (triethylsilane/trifluoroacetic acid) the enamide group of 3a-methyl-N-Boc-hexahydro-1H-indolin-5(6H)-one was reduced to afford exclusively a cis ring-fused product. For the 3a-phenyl substituted analogue more forcing conditions (sodium cyanoborohydride at pH 2-2.5) were required and resulted in the selective reduction of the enamide group to give a trans ring-fused product as well as reduction of the ketone group.  相似文献
