The interactions of nabumetone (NAB) with -cyclodextrin (-CD) and-cyclodextrin (-CD) were studied in aqueous solution by meansof phase-solubility analysis. Solid dispersions of NAB with -cyclodextrin(-CD), -cyclodextrin (-CD), methyl- (M-CD),hydroxypropyl-cyclodextrin (HP-CD) were prepared by coevaporationand kneading and also by coprecipitation in the case of -CD. X-ray diffractometry, thermal analysis and infrared spectroscopy (FTIR) were used to study the possibility of complexation of the drug with the different cyclodextrins. Solid dispersions of nabumetone with -CD showed a remarkable improvement in the dissolution rate of nabumetone.  相似文献

Aim of the present work was to investigate the effect of somecyclodextrins (CDs) on the solubility and ocular bioavailability of rufloxacin base (RUF), with theultimate goal of developing an ophthalmic formulation. Phase solubility studies of RUF inpH 7.4 buffer were carried out in the presence of -cyclodextrin (-CD),hydroxypropyl--cyclodextrin (HP--CD) and -cyclodextrin(-CD). The effect of hydroxypropyl methylcellulose (HPMC) on RUF solubility was evaluated after heating the solutionscontaining HP--CD at 120 °C.A significant enhancement of RUF solubility was achieved by associatingthe drug with CDs, particularly HP--CD. This CD formed with RUF a less stablecomplex than that formed by -CD, but did not suffer the solubility limitations ofthe parent CD, and showed a higher solubilizing capacity than -CD. Addition of 0.25%(w/v) HPMC to solutions containing HP--CD increased the solubilizing effect of this CD,thus allowing reduction of the amount necessary for solubilization of 0.3% (w/v) RUF.Preliminary pharmacokinetic data in rabbits indicated that theocular bioavailability of 0.3% (w/v) RUF solubilized by HP--CD was higher when compared witha 0.3% (w/v) RUF suspension used as reference.  相似文献

The photo-oxidation reaction of phenothiazine has been studied in the presence of-cyclodextrin (-CD) and 2-hydroxypropyl-cyclodextrin (HP -CD). The influence of these organized media on the formation of the oxidation photoproduct upon UV irradiation has been investigated. Phenothiazine forms an inclusion complex with the cyclodextrins. The stoichiometry and formation constant of the complex formed with 2-hydroxypropyl -CD have been calculated using the changes of the fluorescence emission signal and of the absorbance of the drug upon inclusion. An increase of the fluorescence intensity of the photogenerated product is attained when it becomes included inside the cyclodextrin cavity.  相似文献

Summary The effect of tri-O-methyl--cyclodextrin(methyl--CD) on the partition coefficients of drugs, such as p-nitrophenol, salicylic acid, benzoic acid, and aspirin, was studied at 25°C. The partition coefficients of these drugs were increased linearly with methyl--CD concentration. The increase of partition coefficients was interpreted by the 11 complex formation between methyl--CD and the drug in CHCl3 phase.The interaction between p-nitrophenol and methyl--CD in solution was studied by UV and PMR spectroscopies. It was concluded that p-nitrophenol is included in the cavity of methyl--CD in both aqueous solution and CHCl3 solution.Inclusion compounds of these drugs with methyl--CD in the solid state were studied by X-ray diffractometry, IR spectroscopy, and DSC measurements. 11 crystalline inclusion compounds were obtained from hot water. It is also suggested that amorphous inclusion compound was obtained by the grinding of drug with methyl--CD.The dissolution rate and the bioavailability of ketoprofen were significantly increased in the presence of methyl--CD. The bioavailability of ketoprofen after oral administration with methyl--CD to rats was 3.7 times that of ketoprofen alone.  相似文献

The solubility of nimodipine was measured in aqueous solutions of the following cyclodextrins: -cyclodextrin (-CD), hydroxypropyl--CD (HP--CD), -cyclodextrin (-CD), random substituted methyl--CD (M--CD), three hydroxypropyl--CDs (HP--CD) with mutually different average degree of substitution, and hydroxypropyl--cyclodextrin (HP--CD). From the determined linear solubility diagrams the values of the binding constant K11 of the inclusion complexes of nimodipine with the respective CDs were evaluated. The -CDs efficiently solubilized sparingly soluble nimodipine, the highest value of K11 was found for M--CD (1680 M-1), followed by -CD (550 M-1) and HP--CDs, where the higher degree of substitution lowered K11. Only slight solubilization of nimodipine was observed in the solutions of the -CDs and HP--CD.  相似文献

Inclusion complexation of all-trans-retinol, retinal and retinoic acid with -cyclodextrin (-CD) and heptakis(2,6-O-dimethyl)--cyclodextrin (DM--CD) were investigated by means of UV-vis spectroscopy. The association constants (K a) obtained for vitamin A with DM--CD is greater than with -CD. Menthone on reduction with sodium dithionite,showed a good amount of mentholformation in the water/DMF system, withincreasing -cyclodextrin (-CD)concentration from 47.0% for 0.1 equivalent of-CD to 93.5% for 1 equivalentof -CD. Increasing hydroxypropyl--cyclodextrin(HP-CD) gave higher menthol/neomenthol (M/N) ratiosfrom 2.8 to 3.5. In the case of pulegone,increasing -CD showed an increase in the formationof menthols in thewater/DMF system from 13.3% for 0.1 equivalent of-CD to 78.1% for 1equivalent of -CD with greater proportions ofneomenthol and neoisomenthol.However, HP-CD which showed only marginalenhancement in the formationof menthol from menthone (32.1–41%), exhibiteda greater proportion of mentholformation in the case of pulegone (55.1%).However, the phase-transfer capabilityof HP-CD was not found to be significant.  相似文献

In this work the potential of MIMS (Membrane Introduction Mass Spectrometry) for studying the inclusion of small organic guest molecules into cyclodextrin hosts in aqueous medium was investigated. MIMS profiles showed that the inclusion of benzene in cyclodextrins is favored in the following order: -CD HO-propyl--CD> -CD> -CD with equilibrium constants of K -CD = 404; KHO-propyl-CD = 395, K -CD = 335 and K -CD = 210 M-1 at 25 °C. Kinetic experiments suggested that under the conditions employed the inclusion process has a pseudo first-order dependence on the guest benzene concentration with the following order: -CD > -CD HO-propyl--CD> -CD. MIMS inclusion profiles of chlorobenzene and toluene showed that the presence of substituents in benzene makes the inclusion in -CD more difficult. Experiments with ferrocene--CD have also been carried out, showing that the complex rapidly dissociates in water and the resulting free -CD can complex with benzene present in the solution.  相似文献

The inclusion complex of the anti-inflammatory drug, loxoprofen, with -cyclodextrin-(CD), sulfated -CD, and glycerol ether -CD was studied by UV-VIS absorption and 1H-NMR spectroscopy in solution. The inclusion complex of loxoprofen with -CDs was prepared by freeze-drying, and then characterized in the solid state by thermal analysis, X-ray diffraction, FT-IR and FT-Raman spectroscopy, and scanning electron microscopy (SEM). Furthermore, a physical mixture of loxoprofen/-CD (1/1, mol-%) in the solid state was also characterized. The solubility of the loxoprofen increased on addition of -CDs. The solubility enhancement of the loxoprofen with -CDs is in the following order: glycerol ether -CD > sulfated -CD > -CD.  相似文献

Two pyridinium groups were introduced into -cyclodextrin (-CD) at the A and E glucose units to make a molecular flask for controlling the stereo-selectivity of photodimerization of 2-anthracenecarboxylic acid. When the photodimerization of 2-anthracenecarboxylic acid was carried out in the presence of bispyridinio-appended -CD, the relative yield of one of the configurational isomers was increased 1.5-fold compared to the corresponding yield in aqueous solution. The optical yields of the photodimerization reaction products also increased more than 10-fold by the addition of bispyridinio-appended -CD.  相似文献

Anionic N-acetylated -aminoacids (AcTrp-, AcPhe-, AcLeu- and AcVal-) are bound to protonatedheptakis(6-amino-6-deoxy)--cyclodextrin(per-NH3 +--CD) by a cooperative work of inclusion and Coulomb interactions.Such complexation occurs enantioselectively ((S)-selective)and is accompanied bypositive entropy changes. Similar (S)-selectivecomplexation occurs in the oppositelycharged system. Namely, cationic -aminoacid methyl esters are enantioselectivelybound to dissociatedheptakis(6-carboxymethylthio-6-deoxy)--cyclodextrin(per-COO---CD). In order to obtain thegeneral mechanism for complexationof a charged host with an oppositely charged guest,we examined the 1H NMR spectra oncomplexation of simple carboxylate anions suchas p-methylbenzoate anion andalkanoate anions with per-NH3 +--CD.Both Coulomb interactions andinclusion are essential to form stable complexesof these carboxylate anions. In allcases, positive entropy changes promote thecomplexation between the carboxylateanions and per-NH3 +--CD. Dehydrationfrom both charged host and guestis the origin of entropic gains. The mechanism forcomplexation of a charged host withan oppositely charged guest involving the cooperativework of inclusion and Coulombinteractions and positive entropy change due todehydration upon complexation isgenerally applied for related systems such asenantioselective complexation ofRu(phen)3 2+ with per-COO---CDand of Ru(phen)3 2+with DNA.  相似文献

The solubility of carbaryl increased with increasing concentrations of-CD, G2--CD, and M--CD. The result suggests theformation of soluble inclusion complex. Solubility increase was highestin M--CD-carbaryl, being 18.4 fold higher than that of carbaryl when 100 mM M--CD was used. The apparent formation constant for the complex calculated from phase solubility diagram was 223.18 M-1. The preparation of the complex in solid form for characterization was successful by kneading andfreeze-drying. The DSC curves for kneading and freeze-drying mixture didnot show the endothermic peak characteristic of carbaryl, but a small new endothermic peak was observed. FTIR analysis showed a shift of the major peak of carbonyl group in carbaryl molecule from 1717 to 1744 and 1734 cm-1 in kneading and freeze-dried mixtures, respectively. M--CD-carbaryl complex demonstrated higher dissolution rate, higher thermal and UV stability but lower toxicity than its parent carbaryl compound.  相似文献

6-Hydrocinnamoyl -cyclodextrin(6-HyCiO--CD), 6-hydrocinnamoyl -cyclodextrin(6-HyCiO--CD), 6-cinnamoyl -cyclodextrin(6-CiO--CD), and 6-cinnamoyl -cyclodextrin (6-HyCiO--CD) have been prepared.6-HyCiO--CD formed an intramolecular complex in an aqueous solution. 6-HyCiO--CD formed weak intermolecular complexes.6-CiO--CD formed intermolecular complexes to give supramolecular oligomers. 6-CiO--CD formed insoluble supramolecular complexes in the solid state. An anthocyanidin, pelargonidin (PG), loses its color with time in acidic media. The rate determining step of the discoloration reaction at pH 1–4 is the nucleophilic attack of the OH- ion on PG to give a hemiacetal, which readily isomerizes to the corresponding -diketone. Native, branched, and methylated cyclodextrins (CDs) form inclusion complexes with PG to retard the discoloration. The inhibitory effect (copigmentation) of CDs on the PG discoloration is slight in -CD, significant in - and -CDs, and the largest in heptakis(2,6-dimethyl)--CD (DM--CD). The -CD and DM--CD include the phenyl moiety of PG, whereas -CD includes the benzopyrylium moiety of PG. The CD cavities protect the reaction site of included PG from the attack of the OH- ion.  相似文献

The fluorescence quenching of naphthalene, 1-methylnaphthalene, and acenaphthene by trimethylamine (TMA) was studied in aqueous -cyclodextrin (-CD) solutions to know the structural requirements for guest molecules to form three-component complexes. The apparent rates for the fluorescence quenching of the naphthalene derivatives by TMA markedly increased in the presence of -CD. The fluorescence quenching of 1-methylnaphthalene and acenaphthene by TMA was accelerated by -CD more efficently than that of naphthalene. These results suggest the structures of the three-component complexes as the arene-capped -CD including TMA in its cavity.  相似文献

In neutral and alkaline solutions, tetrakis(4-carboxyphenyl)porphyrin (TCPP) and tetrakis(4-sulfonatophenyl)porphyrin (TSPP) form 1:1 and 2:1 host–guest inclusion complexes with -cyclodextrin (-CD), -cyclodextrin (-CD), and 6-deoxy-6-diethylamino--CD (DEA--CD), except for DEA--CD in alkaline solution. On the other hand, TCPP and TSPP form only 1:1 inclusion complexes with 6-deoxy-6-dihexylamino--CD (DHA--CD). The limited solubilities of DEA--CD in alkaline solution and DHA--CD are likely responsible for no observation of the 2:1 inclusion complex containing DEA--CD in alkaline solution and that containing DHA--CD. The equilibrium constants (Ks) of TCPP and TSPP for the formation of the inclusion complexes have been estimated from the absorption and/or fluorescence intensity changes in neutral and alkaline solutions. The K2 values, which are the equilibrium constants for the formation of the 2:1 host–guest inclusion complex from the 1:1 inclusion complex, are about one tenth the corresponding K1 values, except for the -CD–TSPP system in alkaline solution. In neutral solution, where DEA--CD and DHA--CD are in protonated forms, the electrostatic force operates between DEA--CD (DHA--CD) and TCPP (TSPP), leading to the greater K values than those in alkaline solution, where DEA--CD and DHA--CD exist as neutral species.  相似文献

The possibility of the formation of inclusion complexes based on methyl-4-isobutylphenyl ketone, -cyclodextrin (-CD), and o-hydroxy propylated -cyclodextrin in an aqueous phosphate buffer is demonstrated by methods of NMR spectroscopy and cyclic voltammetry. Preparative electroreduction of acetophenone and methyl-4-isobutylphenyl ketone is performed on a lead electrode in aqueous phosphate and acetate buffers in the absence and presence of -CD and the composition of products of electrochemical reactions is ascertained. It is shown that the products of reduction of ketones are corresponding alcohols and pinacones. The possibility of asymmetric electrochemical hydrogenation of ketones in the presence of -CD, which yields relevant chiral alcohols, is shown.  相似文献
