In this paper, an off-line combination method of supercritical fluid extraction and supercritical fluid chromatography was developed for the selective extraction and isolation of diphenylheptanes and flavonoids from Alpinia officinarum Hance. The enrichment of target components was successfully achieved using supercritical fluid extraction with the following conditions (8% ethanol as co-solvent at 45°C and 30 MPa for 30 min). Taking full advantage of the complementarity of supercritical fluid chromatography stationary phases, a two-step preparative supercritical fluid chromatography strategy was constructed. The extract was firstly divided into seven fractions on a Diol column (250 × 20 mm internal diameter, 10 μm) within 8 min by gradient elution increasing from 5% to 20% modifier (methanol) at 55 ml/min and 15 MPa. Then the seven fractions were separated by using a 1-AA or a DEA column (250 × 19 mm internal diameter, 5 μm) at 50 ml/min and 13.5 MPa. This two-step strategy showed superior separation ability for structural analogs. As a result, seven compounds, including four diphenylheptanes and three flavonoids with high purity, were successfully obtained. The developed method is also helpful for the extraction and isolation of other structural analogs of traditional Chinese medicines.  相似文献

应用高速逆流色谱法(HSCCC)分离纯化了高良姜中3种二苯基庚烷类化合物。以正己烷-乙酸乙酯-甲醇-水(2:3:1.75:1, v/v/v/v)为两相溶剂系统,下相为固定相,上相为流动相,在主机转速为858 r/min、流速1.5 mL/min的条件下,从122.20 mg高良姜石油醚萃取物中经一步HSCCC分离可制备得到5R-羟基-7-(4-羟基-3-甲氧基苯基)-1-苯基-3-庚酮(7.37 mg)、7-(4-羟基-3-甲氧基苯基)-1-苯基-4E-烯-3-庚酮(9.11 mg)和1,7-二苯基-4E-烯-3-庚酮(15.44 mg),经高效液相色谱分析,纯度均大于93%,各化合物的结构由质谱和核磁共振氢谱、碳谱鉴定确证。该方法简便、快速、高效,可用于高良姜中二苯基庚烷类化合物的快速分离制备。  相似文献

Phytochemical investigation of methanol extract of the rhizomes of Alpinia officinarum Hance afforded four known diarylheptanoids 1,7-diphenylhept-4-en-3-one (1), 5-hydroxy-1,7-diphenyl-3-heptanone (2), 5-hydroxy-7-(4″-hydroxy-3″-methoxyphenyl)-1-phenyl-3-heptanone (3), and 7-(4″-hydroxy-3″-methoxyphenyl)-1-phenyl heptan-3-one (4).The acetate derivative of (4), 7-(4″-actetate-3″-methoxy phenyl)-1-phenyl heptan-3-one (5), was prepared. These diarylheptanoids exhibited promising in vitro and ex vivo antitubercular activity for the first time against dormant Mycobacterium tuberculosis H37Ra with the IC50 values between 0.3447.69 and 0.13–22.91 μM, respectively. All compounds showed comparable activity against Mycobacterium bovis BCG (dormant phage) and did not show any activity against two gram + ve and two gram –ve bacterial strains. These compounds were also weakly cytotoxic up to 300 μM against three human cancer cell lines THP-1, Panc-1 and A549.  相似文献

Alpinia officinarum is used for its anti‐inflammatory activity historically in China. Diarylheptanoids isolated from A. officinarum play important biological roles in the prevention and treatment of inflammatory disorders. Seven diarylheptanoids (1–7) were isolated from A. officinarum. The cell viabilities and anti‐inflammatory activities of diarylheptanoids were evaluated by MTT assay and tumor necrosis factor‐α production in Kdo2‐lipid A‐stimulated RAW 264.7 cells in vitro. The relationships between their anti‐inflammatories and structure‐activities are discussed. The results indicated that compounds 1 and 3–7 had significant anti‐inflammatory activities. The relationships between inflammation and phospholipids metabolism were elucidated by multivariate data analysis. Twenty‐two potential biomarkers were identified in inflammatory group vs. blank group, and 11 potential biomarkers were identified for inflammatory group vs. drug‐treatment groups. Ten common phospholipids were characterized. On the basis of a previous study in our laboratory, we found that phosphatidylethanolamine (18:0/18:1) might be the important glycerophospholipid biomarker in inflammation. In this study, we firstly combined anti‐inflammatory activities and glycerophospholipids changes of traditional Chinese medicine. This work suggests that the anti‐inflammatory activities of diarylheptanoids might be significantly related to glycerophospholipids and could provide a useful database for investigating the anti‐inflammatory effects of traditional Chinese medicine.  相似文献

Direct analysis of polar solutes by supercritical fluid chromatography (SFC) has been somewhat limited because the mobile phases in common use are all relatively nonpolar. Derivatization is demonstrated as a viable means for facilitating the SFC separation of sugars. A novel dimeric diarylheptanoid, (5R,5′R)‐7,7′‐(6,6′‐dihydroxy‐5,5′‐dimethoxy[1,1′‐biphenyl]‐3,3′‐diyl)bis[5‐methoxy‐1‐phenylheptan‐3‐one] ( 1 ), and two new diarylheptanoids, (4E,6R)‐6‐hydroxy‐7‐(4‐hydroxy‐3‐methoxyphenyl)‐1‐phenylhept‐4‐en‐3‐one ( 2 ) and (4E,6R)‐6‐hydroxy‐1,7‐diphenylhept‐4‐en‐3‐one ( 3 ), together with seven known diarylheptanoids, were isolated from the rhizomes of Alpinia officinarum. The enantiomeric separation of triadimenol and triadimefon on a Chiralpak AD column using supercritical fluid chromatography, was studied in this work. The effect of different modifiers (methanol, ethanol and 2-propanol) was tested, with methanol and ethanol providing the best results for the enantiomeric separation of the two compounds. The enantioseparation of a mixture of triadimenol and triadimefon (six stereoisomers) was achieved in only 15 min using a gradient of ethanol, 200 bar, 35 degrees C and a flow-rate of 2 ml/min. The separation of triadimenol diastereoisomers on different achiral columns (diol, silica and ODS) was also investigated. In this case, the type of organic modifier to be used depended on the stationary phase, the Spherex Diol being the column that gave the best separation. Using this column, resolutions higher than 3 were obtained in analysis times of 5 min with any of the modifiers checked.  相似文献

Alpinia officinarum has been used as a folk medicine and contains diarylheptanoids that have various biological activities. However, their antiviral activities are less elucidated. We examined the antiviral activities of nine diarylheptanoids isolated from A. officinarum against respiratory syncytial virus (RSV), poliovirus, measles virus, and herpes simplex virus type 1 (HSV-1) using a plaque reduction assay. The 50% inhibitory concentrations of seven of the nine diarylheptanoids for RSV were moderately but significantly lower than their 50% cytotoxic concentrations, as determined by a trypan blue exclusion assay. Four diarylheptanoids with anti-RSV activity also showed anti-poliovirus and anti-measles virus activities and three of the four exhibited anti-HSV-1 activity. Thus, seven of the nine diarylheptanoids examined exhibited potential antiviral activity against RSV, and most of the diarylheptanoids with anti-RSV activity, including two diarylheptanoids without anti-RSV activity, were effective against poliovirus, measles virus, and/or HSV-1 in vitro. Diarylheptanoids were suggested to have a broad spectrum of antiviral activity.  相似文献

Two amino acid phosphoramidates with high antiviral activity were used as model compounds and separated using supercritical fluid chromatography on an achiral Hypersil BDS cyano column (250 × 4.6 mm, 5 µm). Supercritical CO2 was used as a mobile phase with different co-solvents, including methanol, ethanol, and 2-propanol. Several key processing parameters were evaluated, including the type and concentration of alcohol modifier (5 to 15%), backpressure (100 to 200 bar), and column temperature (30 to 45 ?C) in terms of their impact on retention factor, diastereoselectivity, and resolution. The optimized chromatographic conditions allowed for a complete separation of mixture of two diastereomers with good resolution over a short time (5–9 min). At a backpressure of 150 bar and a temperature of 33°C, the diastereomers of phenylalanine methyl ester phenyl 5′-phosphoamidate of d4T (d4T-P-N-PheOMe) and alanine methyl ester phenyl 5′-phosphoamidate of d4T (d4T-P-N-AlaOMe) were separated with resolutions of 4.43 and 2.55 using 10.0% and 5.0% (v/v) methanol, respectively.  相似文献

Summary In supercritical fluid chromatography (SFC), the solubilizing nature of the mobile phase enables the use of composition gradients (gradient elution) like in liquid chromatography. With this gradient technique, oligomer separations have been achieved over wide molecular weight ranges. For separating oligomers which absorb only at low wavelengths, a carbon dioxide/acetonitrile mobile phase was used which allows UV detection down to 200 nm. As an ideally suited detection assembly for the chromatography of oligomers, a combination of mass-sensitive and evaporative light-scattering detection is suggested.

Background: Alpinia officinarum (A. officinarum) is known to exhibit a beneficial effect for anti-inflammatory, anti-oxidant, and anti-hyperlipidemic effects. However, no sufficient research data are available on the cardiovascular effect of A. officinarum. Thus, in this study, we investigate whether A. officinarum extract has direct effects on vascular reactivity. Methods: To examine whether A. officinarum extract affects vascular functionality, we measured isometric tension in rat mesenteric resistance arteries using a wire myograph. After arteries were pre-contracted with high-K+ (70 mM), phenylephrine (5 µM), or U46619 (1 µM), A. officinarum extract was treated. Results: A. officinarum extract induced vasodilation in a concentration-dependent manner, and this effect was endothelium independent. To further investigate the mechanism, we incubated arteries in a Ca2+-free and high-K+ solution, followed by the cumulative addition of CaCl2 (0.01–2.5 mM) with or without A. officinarum extract (30 µg/mL). Pre-treatment of A. officinarum extract reduced the contractile responses induced by cumulative administration of Ca2+, which suggests that extracellular Ca2+ influx was inhibited by the treatment of A. officinarum extract. These results were associated with a reduction in phosphorylated MLC20 in VSMCs treated with A. officinarum extract. Furthermore, eucalyptol, an active compound of A. officinarum extract, had a similar effect as A. officinarum extract, which causes vasodilation in mesenteric resistance arteries. Conclusion: A. officinarum extract and its active compound eucalyptol induce concentration-dependent vasodilation in mesenteric resistance arteries. These results suggest that administration of A. officinarum extract could exert beneficial effects to treat high blood pressure.  相似文献

A method for separation and detection of divalent transition metal beta-diketonates by adduct formation/supercritical fluid chromatography (SFC) with an open-tubular capillary column and a FID detector is described. The crystal structures of copper (Cu)-hexafluoro-acetylacetone (HFA) and Cu bis(2,2,6,6-tetramethyl-3,5-heptanedionato) (THD) complexes have been determined by X-ray crystallography. The SFC behavior of Cu beta-diketonates shows a strong correlation with the structure of the complexes. The hydrated cu beta-diketonate complexes usually exhibit strong intermolecular interactions or decomposition in SFC. Formation of adducts with a neutral donor, such as tributyl phosphine oxide (TBPO), can greatly improve the SFC behavior and detection sensitivity of Cu(II) and Mn(II) beta-diketonates. The stoichiometry and thermal stability of the adducts Cu(II) and Mn(II) beta-diketonates with TBPO in supercritical CO(2) have also been investigated. The implications of utilizing adduct formation for supercritical fluid extraction (SFE) of divalent transition metals and for on-line coupled SFE/SFC analysis of divalent transition metals are discussed.  相似文献

Summary A rapid separation of salbutamol sulphate and six related impurities: 5-formyl-saligenin, salbutamol ketone, salbutamol bis ether, isopropyl salbutamol, desoxysalbutamol sulphate and salbutamol aldehyde, has been achieved by employing packed column supercritical fluid chromatography. The effects of temperature, pressure, additive concentration and identity on retention have been studied. The use of a basic additive is necessary in order to elute the compounds and improve the peak shape. The best results were obtained by using a diol column and a gradient of modifier (methanol with 0.5% of n-propylamine).  相似文献

Summary Two-step liquid chromatographic separation (LC) has been applied to soot extract and the identification of higher fullerenes has been accomplished by LC-MS measurements using an ESI interface. The first separation step is preparative-scale LC using a 50 mm i.d. column packed with monomeric octadecylisilica (ODS) because elution is mainly controlled by relative molecular mass. 39 batches of five fractions each were collected and then as the second separation step each fraction was analysed by analytical-scale LC using a conventional column of a polymeric ODS phase which can elute fullerenes according to shape and structure. This stationary phase can also separate many isomers of higher fullerenes, consequently the existence of several higher fullerenes larger than C86 has been confirmed and their UV-Vis spectra were obtained by the photodiode array detection system coupled to the analytical LC.  相似文献
