In this work,a kind of preparation method of zwitterionic ion chromatography(ZIC) stationary phase modified with phosphorylcholine(PC) was obtained by hydrolyzing after bonding phosphorylcholine dichloride to diol-silica to better explore the characteristics of the PC groups as ZIC stationary phase ligand in simultaneous separation of acidic proteins and basic proteins. The results showed that tw’o kinds of acidic proteins and three kinds of basic proteins can be separated completely,meanwhile,hen egg white was separated and purified and three kinds of egg white components ovalbumin,G2 ovoglobulin and ovotransfemin proteins were separated completely by one single step on PC-ZIC column,the purity of all proteins reached above 95%.PC-ZIC stationary phase was successfully improved with better separation capacity and selectivity than previously reported in this paper.  相似文献

A new β-cyclodextrin modified hyperbranched carbosilane stationary phase for gas chromatography was synthesized by substituting the -OH groups ofβ-cyclodextrin with hyperbranched carbosilane and was coated on the inner wall of fused silica capillary column for gas chromatography.The chromatographic behaviors of the stationary phase were studied.The initial testing results showed mat it possessed good separation abilities for several kinds of mixtures,such as benzenes,acrylates,ketones and alkylchlorides.  相似文献

A silica-bonded bovine serum albumin (BSA) chiral monolithic stationary phase for capillary electrochromatography(CEC) was introduced. An inorganic-organic hybrid monolithic column was firstly prepared by sol-gel chemistry with homogeneously distributed aminopropyl groups throughout the silica matrix. Then the chiral stationary phase was synthesized by the in situ covalent immobilization of BSA on the monolithic column activated with glutaraldehyde. The effects of pH value and concentration of phosphate buffer on the separation of D,L-tryptophan were investigated. The separation factor of D,L-tryptophan reached 3.37 on CEC mode.  相似文献

<正>A guanidinium ionic liquid,N,N,N',N'-tetrahexyl-N",N"-dimethylguanidinium bis(trifluoromethane)sulfonylimide(THDMGNTf_2), was synthesized and used as stationary phase for capillary gas chromatography.In comparison with imidazolium ionic liquid stationary phase,the present new stationary phase exhibits quite different selectivity and behaves more like a low polar stationary phase.The guanidinium ionic liquid of THDMG-NTf_2 exhibited better separation of Grab test mixture than imidazolium ionic liquid of 1-octyl-3-butylimidazolium bis(trifluoromethane)sulfonylimide(OBIM-NTf_2).Solvation parameter model was also used to evaluate the selectivity of THDMG-NTf_2.Additionally,essential oil of Magnolia biondii Pamp was analyzed to further evaluate the selectivity of THDMG-NTf_2 for a sample of complicated components.Satisfactory separation of the essential oil was achieved on a THDMG-NTf_2 column(10 m) while using a commercial column(30 m) as reference.The present study shows that the guanidinium ionic liquid possesses novel chromatographic selectivity and has great potential for wide applications.  相似文献

Graphene oxide(GO) has attracted extensive attention due to its unique properties and potential applications.Here,we report the investigation of GO nanosheets as a stationary phase for capillary gas chromatographic(GC) separations.The GO column,fabricated by a new one-step coating approach,showed average McReynolds constants of 308,suggesting the medium polar nature of the GC stationary phase.The GO stationary phase achieves good separation for analytes of different types with good peak shapes,especially for H-bonding analytes,such as alcohols and amines.The different retention behaviors of GO stationary phase from the conventional stationary phase may originate from its multiple interactions with analytes,involving H-bonding,dipole-dipole,π-π stacking and dispersive interactions.Moreover,GO column showed good separation reproducibility with relative standard deviation(RSD%) less than 0.24%(n = 5) on retention times of analytes.  相似文献

A novel chiral stationary phase (CSP1) for ligand-exchange chromatography (CLEC) was prepared by firstly using dimethyl- chlorosilane as an endcapping reagent for decreasing residual silanol group on the surface of silica gel,and then introducing L-Pro as a chiral selector and hydrophobic octyl group to the silica gel surface simultaneously.The enantioseparations of 14 DL-amino acids on CSP1 were achieved with the enantioselectivityαranging from 1.09 to 2.44 and the resolution Rs being between 0.8 and 6.3.The chromatographic performances of CSP1 with the bonded phase (CSP2) prepared using reference method were compared.The results showed that the column efficiency and resolution Rs of chiral stationary phase could be improved by using the above modifying method.  相似文献

β-Cyclodextrin 1 was directly oxidized to the corresponding monoaldehyde 2 on their primary faces by cyclized 2-iodoxybenzoic acid(IBX) in DMSO, followed by the synthesis of β-cyclodextrin derivatives bearing Schiff-base group 3. A new chiral stationary phase(BCDS 6) was then prepared by immobilization of β-cyclodextrin derivative with α-Schiff base group onto the surface of sillica gel. A series of compounds with amino groups were readily separated using this CSE Methanol and acetonitrile were tested as the mobile phase while the influence of temperature and the addition of aqueous triethylammonium acetate buffer to the mobile phase was also innvestigated. Considering all the kinds of interactions between solute and solvent, solute and stationary phase, solvent and stationary phase molecules as well as the competitional adsorption among various kinds of solvent molecules on the stationary phase, we present a stoichiometric displacement model of solute retention with four sets of parameters in liquid chromatography. This model was tested with data from both literature and experiments done by ourselves. These results show that this model may fit the experimental data for a liquid chromatography system with various kinds of mobile phases consisting of a complete range of multi-components and with different types of stationary phases.  相似文献

正A new ionic liquid-based high-performance liquid chromatography stationary phase is reported.A derivative of N-methyl pyrrolidinium tetrafluoroborate was covalently immobilized on the surface of silica particles to prepare silica-based N-methyl pyrrolidinium tetrafluoroborate(SilprMP BF4)stationary phase.The obtained ionic liquid-modified silica was evaluated and confirmed by elemental analysis,infrared spectroscopy,and thermogravimetric analysis.A column was packed with the modified particles.The retention behavior of aromatic compounds,alkyl benzenes,and acidic and basic compounds on the SilprMP BF4 stationary phase was studied under reversed-phase liquid chromatography conditions.The effect of the eluent pH on the separation of the acidic and basic compounds was also studied.The new stationary phase involves multiple retention mechanisms,such as electrostatic,hydrophobic,ion-dipole,and anion-exchange interactions,which might lead to multipurpose separation media.  相似文献

A new chiral chemically bonded stationary phase for direct resolution of optical isomers in HPLC was prepared. The experiemental data showed that the chiral stationary phase gave good resolution for the racemic α-amino acid derivatives and that there was no significant influence on the resolution as the eluting rate varied from 0.6 to 2.0 mL/min. The content of isopropanol in the eluent significantly affected the resolution of the enantiomers. The eluent consisting of 1% isopropanol in hexane gave most favorable result, but the retention times were a little longer. It is evident that the resolution was also influenced by the structure of the solute. In all cases, the resolution of N-Ac-DL-Phe-OMe is lower than that of N-Ac-DL-Leu-OMe and N-Ac-DL-Val-OMe. Although the capacity factor of N-Ac-DL-Tyr-Ome varied greatly in different eluting systems, the separation factor and resolution were very close. It was observed that D-amino acid derivatives eluted faster than the corresponding L-isomers. In the above chromatographic conditions N-acetyl-DL-α-phenylethylamine could not be resolved.  相似文献

Summary In preparative column liquid chromatography, it is necessary to work with conditions of mass overload to obtain high throughput per unit time. The influence of sample mass and of eluent composition on the separation of R,S-1-(1-naphthylethyl)propylamide on chiral 3,5-dinitrobenzoylphenylglycine-silica (Pirkle type stationary phase) was investigated. At a sample load of 200μg of racemate per gram of stationary phase, the column becomes overloaded. At higher sample loads the peaks become triangular; therefore the first peak is always pure up to loads of 10mg g−1, although resolution is low. The purity of the second peak depends on sample mass, but not on the strength (polarity) of the mobile phase. This is due to the effect that peak width, as well as resolution, decrease as the polarity of the eluent increases. In certain cases the polarimeter, used as a detector, can give more information on peak purity than the UV detector.  相似文献

The L-proline-functionalized polystyrene with 1-methyldecylene spacer was synthesized from 2% divinylbenzene-crosslinked polystyrene gel via 10-bromo-1-methyldecyl polystyrene intermediate.After complexed with copper(Ⅱ) ion,the polymer with L-proline ligand was used as the chiral stationary phase (CSP) for ligand exchange chromatography of amino acid racemates.The results showed that the CSP possessed powerful enantioselectivity and all racemates of the fifteen tried amino acids were completely separated.  相似文献

Based on the monodisperse poly(glycidyl methacrylate-co-ethylenedimethacrylate) beads (PGMA/EDMA) with macropore as a medium, a new hydrophilic medium cation exchange (MCX) stationary phase for HPLC was synthesized by a new chemically modified method. The stationary phase was evaluated with the property of ion exchange, separability, reproducibility, hydrophilicity, effect of salt concentration, salt types, column loading and pH on the separation and retention of proteins in detail. It was found that it follows ion exchange chromatographic (IEC) retention mechanism. The measured bioactivity recovery for lysozyme was (96 ± 5)%. The dynamic protein loading capacity of the synthesized MCX packings was 21.8 mg/g. Five proteins were almost completely separated within 6.0 min at a flow rate of 4 mL/min using the synthesized MCX resin. The MCX resin was also used for the rapid separation and purification of lysozyme from egg white with only one step. The purity and specific bioactivity of the purified lysozyme was found more than 95% and 70345 U/mg, respectively.  相似文献

Amylose tris(3,5-dimethylphenylcarbamate) (ADMPC) coated on a kind of small particle silica gel was prepared. On this ADMPC chiral stationary phase (CSP), the direct enantiomeric separation of six novel chiral transition metal tetrahedral clusters has firstly been achieved using n-hexane as the mobile phase containing various alcohols as modifiers. The effect of mobile phase modifiers and the structural variation of the solutes on their retention factors (k‘) and resolutions (Rs) were investigated. The result suggests that not only the structure and concentration of alcohol in mobile phase, but also the structural differences in racemates can have a pronounced effect on enantiomeric separation. ADMPC-CSP is a suitable CSP for the optical resolution of chiral tetrahedral cluster by HPLC.  相似文献

A smallbore chemically bonded alkyl stationary phase column of 2 mm I. D.(inner diameter) and 15 cm in length having an efficiency of 6400—7600 theoretical plates has been developed. The column officicncy is nearly the same as that of routine analytical column of 4.6—5.0 mm I. D. The amount of cluant used for the smallbore column, however, is less than about one fifth that for the ordinary columns.Using the reaction of monochlorosilane with 5 μm accumulated silica bead produced in Qingdao Ocean Chemical Factory instead of trichlorosilane and irregular silica gel, we have. prepared a nonpolar chemically bonded phase packing which has a higher carbon content and more obvious hydrophobicity. Moreover, the reproducibility of the preparation is better.The equipment and assembly necessary for getting high efficiency with the smallbore column are discussed briefly. A microcalorimetric technique based on the metabolic heat-output was explored to evaluate the inhibition of cyanide on the mitochondrial metabolism of aquatic animal, Cyprinus carpio. The power-time curves could be divided into four parts: lag phase, active recovery phase, stationary phase, and decline phase, and the corresponding thermokinetic parameters were obtained. The maximum heat production rate Pmax decreased in a linear manner with the increase of concentration of cyanide, however, such mitochondria of aquatic animal were still metabolized actively even under the action of high concentration of cyanide. All the observations suggested that the mitochondria of this aquatic animal should exhibit considerable ability of cyanide-resistant respiration.  相似文献

It was the intention of this paper to show some recent developments in thin layer chroma-togrphy aiming at increasing the separation efficiency by instrumental means and techniquesusing existing separation layers. Gradient elution in the normal phase is a most efficient wayto achieve this goal. Multi-dimensional separations by coupling gradient elution column liquid chromatography inthe reversed phase with AMD gradient elution in normal phase is suitable to achieve newdimensions of separation numbers. Assuming that the N numbers reported for HPLC translateinto separation numbers near 100, by coupling the two kinds of chromatography, separationnumbers around 500 become practically usable.  相似文献

<正>The present study describes guanidinium-based ionic liquids(GBILs) as stationary phases for capillary gas chromatography (CGC) and to the best of our knowledge,no related reports are available up to now.In this study,a hexaalkylguanidinium ionic liquid(DOTMG-NTf_2) was synthesized and coated statically onto capillary columns.Selectivity of the stationary phase was evaluated by separating Grob test mixture,test mixture,alcohols mixture,and fatty acid methyl esters mixture,and thermal stability was investigated as well.The present study demonstrates that GBILs as CGC stationary phases exhibit satisfactory selectivity and thermal stability and have a great potential as new candidates for CGC stationary phases.  相似文献
