A novel assay of ciprofloxacin with a sensitivity at the microgram level is proposed based on the measurement of enhanced fluorescence intensity signals resulting from the interaction of functionalized nano-CdS with ciprofloxacin. The CdS nanoparticles was synthesized by thioacetamide (TAA) and cadmium nitrate (Cd(NO(3))(2)) in the alkaline solution. At pH 7.4, the fluorescence signals of functionalized nano-CdS were greatly enhanced by ciprofloxacin with the increase concentration of ciprofloxacin. Linear relationship can be established between the enhanced fluorescence intensity and ciprofloxacin concentration in the range of (1.25-8.75)x10(-4) mg mL(-1) ((3.77-26.4) x 10(-4)mmol L(-1)) or (8.75-1200) x 10(-4)mg mL(-1) ((26.4-3625) x 10(-4) mmol L(-1)). The limit of detection is 7.64 x 10(-6) mg mL(-1) (2.31 x 10(-5)mmol L(-1)). Based on this, a new direct quantitative determination method for ciprofloxacin in human serum samples without separation of foreign substances was established. The contents of ciprofloxacin in human serum samples were determined with recovery of 95-105% and relative standard deviation (R.S.D.) of 1.5-2.5%. This method was proved to be very sensitive, rapid, simple and tolerance of most interfering substances.  相似文献

Derivative spectrophotometry was employed to develop a rapid and accurate method for simultaneous determination of indomethacin and 5-methoxy-2-methyl-3-indoleacetic acid as its possible impurity in Metindol injections. At the selected wavelengths, 233.04 and 284.65 nm, no interference between the components determined was observed. Under the established experimental conditions, recoveries of the particular components were from 96.14 to 98.17%. Linearity was maintained over a broad range of concentrations, from 11.88 x 10(-3) to 35.64 x 10(-3) mg/mL for indomethacin and 0.4 to 1.2 mg/mL for 5-methoxy-2-methyl-3-indoleacetic acid. The limit of detection was found to be 6.0 x 10(-3) mg/mL for indomethacin and 0.04 x 10(-3) mg/mL for 5-methoxy-2-methyl-3-indoleacetic acid. The limits of quantitation were found to be 10.0 x 10(-3) mg/mL and 0.20 x 10(-3) mg/mL, respectively.  相似文献

Manganese, nickel, cobalt, and zinc are elements that appear together in many real samples. In this work, a simple, rapid, and sensitive method has been used for the simultaneous separation and preconcentration of trace amounts of these elements using water-soluble polyacrylic acid on alumina as a sorbent in a glass column system. A solution containing Mn, Ni, Co, and Zn was passed through the column at pH 7, and desorption was carried with 5.0 mL of 0.03 M nitric acid. Linearity was maintained between 0.25-5.0 x 10(3), 0.04-6.0 x 10(3), 0.10-8.0 x 10(3), and 0.028-1.0 x 10(3) ng/mL for Ni, Mn, Co, and Zn, respectively, in the original solution. Eight replicate determinations of a mixture containing 1.0 microg/mL of each of the elements in the final solution gave relative standard deviations of 1.4, 1.5, 2.4, and 1.2% for Ni, Mn, Co, and Zn, respectively. The sorption capacities for Mn (mg/g sorbent) obtained were 16.0 for Mn, 9.0 for Ni, 12.0 for Co, and 10.0 for Zn. By using 5.0 mL elution solution, preconcentration factors of 400, 500, 700, and 460 could be obtained for Ni, Mn, Zn, and Co, respectively.  相似文献

建立了大鼠血浆中原儿茶酸含量测定的高效液相色谱方法。采用的色谱柱为DiamondsilTM C18 柱(150 mm×4.6 mm,5 μm);流动相为乙腈-水(体积比为9∶91,用H3PO4 调pH至2.5);流速1.2 mL/min;检测波长260 nm;内标为对羟基苯甲酸。原儿茶酸的线性范围为0.050~3.20 mg/L,线性相关系数为0.9978,最低定量限为0.050 mg/L,日内和日间测定的精密度(以相对标准偏差表示)均低于7.0%,准确度(以相对误差表示)为-1.4%~2.6%;在0.050,0.40,3.20 mg/L低、中、高3个添加浓度水平下,血浆样品的提取回收率分别为83.4%,87.3%,91.1%。该方法简便,灵敏,准确,适用于大鼠体内原儿茶酸的药物动力学研究。  相似文献

A composite nano-TiO2-chitosan with collagen artificial skin (NTCAS) showed promising characteristics of moderate water absorptivity (110 wt.-%), fine thickness (0.34 mm), low density (0.33 mg x mm(-3)), moderate biodegradability (<3.0%), and favorable time-dependent biodegradability (0.0521 mg x mL(-1) x h(-1) on Day 0; 0.0006 mg x mL(-1) x h(-1)on Day 3 post covering). More importantly, it showed uniquely potent bactericidal (sterilization) property with relatively large values of pseudo first-order kinetic coefficients (0.1736-0.0360 min(-1)) for TiO2 contents ranging from 2.63 x 10(-5) to 2.11 x 10(-4) mg x cm(-2). In wounded subjects, NTCAS showed a steady level of TNF-alpha, while the IL-6 level reached a peak on Day 7, although significantly lower than in the control and Duoderm groups. In the animal model, NTCAS showed better and faster recovery than the other groups, which can be attributed to the unique bactericidal effect of nano-TiO2 and immune-enhancing effect of chitosan. NTCAS is a promising artificial skin substitute and is superior to any other market product currently in use.  相似文献

A method using HPLC-CL linkage was developed for simultaneous determination of N,N-dimethylaniline and phenol in wastewater, based on the strong sensitive chemiluminescence of the luminol-K3Fe(CN)6 systems in alkaline medium. The separation was carried out on a Hypersil ODS column with a mobile phase of ethanol-0.01% triethylamine (2:1, v/v). The linear ranges for N,N-dimethylaniline determinations were 2.0 x 10(-7) - 2.5 x 10(-5) g/mL and 4.0 x 10(-5) - 1.5 x 10(-4) g/mL with a detection limit (3sigma) of 1.20 x 10(-8) g/mL; the relative standard deviation (3sigma) for 5.0 x 10(-6) g/mL N,N-dimethylaniline was 1.4% (n = 6). The range for phenol was from 5.1 x 10(-7) to 1.3 x 10(-4) g/mL, and a detection limit (3sigma) of 2.5 x 10(-8) g/mL could be obtained. The method can be useful for the determination of N,N-dimethylaniline and phenol in some environmental samples.  相似文献

A quick and sensitive reversed-phase high-performance liquid chromatography (HPLC) method has been developed in order to determine the concentration of Propofol (2,6 diisopropylphenol) in human serum. Propofol can be isolated from serum by adding 0.5 mL precipitating solution. This consists of an acetonitrile and perchloric acid (67:33, v/v) mixture, which also contains dibutylphthalate (2 mg/100 mL) as internal standard. The sample is then mixed for 1 min on a vortex-mixer. The endogenous serum substances precipitated by acetonitrile and perchloric acid are further separated by centrifugation. The supernatant is directly injected into the HPLC system. A 250- x 4.6-mm column, packed with 10-microns Spherisorb reversed-phase octadecylsilane particles (C18), is used for chromatographic separation. The mobile phase consists of an acetonitrile-water mixture (67:33 ratio) with 0.4 mL acetic acid (pH 4). Propofol is monitored by a UV-visible detector at 270 nm and 0.1-0.002 absorbance units full scale (AUFS). The detection limit of Propofol (in human serum) is 0.1 mg/L for a 20-microL injection volume. The time of the assay is less than 20 min, including sample preparation.  相似文献

A simple, sensitive and selective chemiluminescence (CL) method was developed for the direct determination of aluminum (Al). This method is based on that the weak CL of cerium (IV)-calcein can be greatly enhanced by Al(III). The calibration curve was linear over the range 2.0 x 10(-10) to 4.0 x 10(-8)g mL(-1) with a detection limit of 8 x 10(-11)g mL(-1) (3sigma). The R.S.D. was 2.5% by 11 replicated determinations of 1.0 x 10(-9)g mL(-1) Al(III). The proposed method has been used to determine the concentration of Al(III) in real water samples with satisfactory results. The mechanism of the CL reaction was also discussed.  相似文献

The existence of attractive interaction between CMC and BSA was evidenced in solution at pH higher than the protein isoelectric point by means of SAXS. Mixtures of BSA at 10 x 10(-3) g . mL(-1) and CMC at the concentration range of 1 x 10(-3) g . mL(-1) to 10 x 10(-3) g . mL(-1) were investigated. Upturns in the very low q range revealed the presence of aggregates when the CMC concentration was higher than 2 x 10(-3) g . mL(-1). The comparison between the calculated form factor with the experimental curves at intermediate and long q values indicated that the BSA molecules keep their native form in mixtures with CMC concentrations lower than 5 x 10(-3) g . mL(-1). Therefore, for CMC concentrations higher than 2 x 10(-3) g . mL(-1) the mixtures start to present aggregates and free BSA molecules coexisting in solution.  相似文献

A novel reaction of cyanide with 2,2-dihydroxy-1,3-indanedione in the presence of sodium carbonate is described. It is highly selective and sensitive, and suitable for the determination of hydrogen cyanide in the environment and free cyanide ions in water, blood, urine, serum, etc. As little as 1.25x10(-7) mol x L(-1) CN(-) (3.25x10(-9) g x mL(-1) cyanide) can be determined by use of this reaction. The color system obeys Beer's law in the range 10 ng x mL(-1) to 1.0 microg x mL(-1) at 510 nm. The molar absorptivity was 8.0x10(4) L x mol(-1) x cm(-1) for a solution of concentration 0.2 microg x mL(-1). All other important analytical properties of the reaction have been studied. It is proposed that the purple color produced under these reaction conditions is that of 2-cyano-1,2,3-trihydroxy-2 H indene.  相似文献

Using liquid chromatography (LC), a convenient and versatile method was developed for the assay of disinfectant products containing the commonly used phenolic agents, alone or in combination, including phenol, 2-phenylphenol, 4-t-amylphenol, 4-chloro-3,5-dimethylphenol (PCMX), 2-benzyl-4-chlorophenol, and triclosan. The procedure involves a simple dilution or dissolution of the product aliquot or portion followed by membrane filtration and LC. The method was applied to 19 different products representing 17 manufacturers and containing from one to 4 phenolic agents. Chromatography was performed using a Zorbax SB-C18 column, acetonitrile-0.05M KH2PO4 (55 + 45, v/v) pH 3.00, as mobile phase and UV detection at 280 nm. The intralaboratory precision of the product assays ranged form 0.34-5.35% (n = 5) and recoveries via fortification varied from 96.1-103.8%. The limits of quantitation (LOQ) and detection (LOD) ranged from 5.58 x 10(-5) to 2.50 x 10(-4) mg/mL and from 1.76 x 10(-5) to 0.67 x 10(-4) mg/mL, respectively, for the 6 analytes. The response for all analytes was observed to be linear for at least a 50-fold range in concentration (0.001-0.05 mg/mL). The proposed method provides an efficient means for the isolation and quantitation of these phenolic compounds.  相似文献

A sensitive and simple high-performance liquid chromatographic method with UV detection was developed and validated for the determination of andrographolide in rat whole blood. Carbamazepine was employed as internal standard and the blood sample was extracted with chloroform. Chromatographic separations were achieved on a Chromasil ODS column (250 x 4.6 mm, 5 microm). The mobile phase was consisted of methanol-water (52:48, v/v) and delivered at 0.8 mL/min. The detection wavelength was set at 225 nm. The calibration curve had a good linearity in the range 0.053-530 microg/mL in rat whole blood with its correlation coefficient being 0.996. The extraction recovery of andrographolide was ranged from 65.7 to 72.6%. The intra-day and inter-days repeatabilities were below 4.2% in terms of the percentage of relative standard deviation (RSD). The method was used to provide data on the pharmacokinetics of the drug in rats. The data obtained was processed using the 3P87 pharmacokinetic program. The results showed that the disposition of andrographolide after intravenous administration of liposomal andrographolide conformed to a two-compartment open model with alpha = 4.75 x 10(-2) +/- 2.41 x 10(-3) min(-1), beta = 3.16 x 10(-3) +/- 1.58 x 10(-4) min(-1), V(c) = 174.67 +/- 13.97 mL, k(21) = 1.60 x 10(-2) +/- 8.12 x 10(-4) min(-1), k(10) = 9.38 x 10(-3) +/- 5.62 x 10(-4) min(-1), k(12) = 2.53 x 10(-2) +/- 1.27 x 10(-3) min(-1) and AUC(0-infinity) = 1525.47 +/- 92.35 microg min/mL. For the intragastric administration of andrographolide tablets, the disposition of andrographolide followed a one-compartment open model with k(e) = 6.78 x 10(-3) +/- 3.53 x 10(-4) min(-1), k(a) = 3.69 x 10(-2) +/- 4.68 x 10(-3) min(-1), T(max) = 59.69 +/- 3.61 min, C(max) = 1.62 +/- 0.11 microg/mL, V(c) = 1056.90 +/- 83.42 mL, AUC(0-infinity) = 348.75 +/- 24.41 microg min/mL.  相似文献

This study describes the simultaneous determination of phosphonate, phosphate, and diphosphate by CE with direct UV detection, based on in-capillary complexation with Mo(VI). When a mixture of phosphonate, phosphate, and diphosphate was injected into a capillary containing 3.0 mM Mo(VI), 0.05 M malonate buffer (pH 3.0) and 45% v/v CH3CN, three well-defined peaks, due to the migration of the corresponding polyoxomolybdate anions, were separated. The respective calibration graphs were linear in the concentration range of 2 x 10(-6)-2 x 10(-4) M for phosphonate, 1 x 10(-6)-5 x 10(-5) M for phosphate, and 1 x 10(-6)-2 x 10(-4) M for diphosphate; the correlation coefficients were better than 0.9990. The present CE method is successfully applied to the simultaneous determination of phosphonate, phosphate, and diphosphate in tap water.  相似文献

A novel flow injection chemiluminescence (CL) method has been developed for the determination of three metal ions, namely Fe2+, Fe3+ and Cr3+, based on the second CL (SCL) signal of the mixture of luminal with KMnO4 in a sodium hydroxide medium by the catalysis of Fe2+, Fe3+ or Cr3+. The possible CL mechanism of the systems, the influencing factors, and the optimum conditions for the reactions were investigated based upon the kinetic curve of the CL reaction, CL spectra, UV-visible spectra and some other experiments. Under the optimum conditions, the SCL intensity was directly proportional to the concentration of these metal ions in solution in the range of 0.10 - 100.00 mg l(-1) for Fe(2+), 0.50 - 7.50 and 7.50 - 200.00 mg l(-1) for Fe3+, 0.01 - 0.25 and 0.25 - 10.00 mg l(-1) for Cr3+. The detection limits (3 sigma/s) were 9.87 x 10(-6) g l(-1), 2.71 x 10(-6) g l(-1) and 5.25 x 10(-7) g l(-1) for Fe2+, Fe3+ and Cr3+, respectively.  相似文献

A LC-MS method was developed for the determination of coumarin, vanillin, and ethyl vanillin in vanilla products. Samples were analyzed using LC-electrospray ionization (ESI)-MS in the positive ionization mode. Limits of detection for the method ranged from 0.051 to 0.073 microg mL(-1). Using the optimized method, 24 vanilla products were analyzed. All samples tested negative for coumarin. Concentrations ranged from 0.38 to 8.59 mg mL(-1) (x =3.73) for vanillin and 0.33 to 2.27 mg mL(-1) (x =1.03) for ethyl vanillin. The measured concentrations are compared to values calculated using UV monitoring and to results reported in a similar survey in 1988. Analytical results, method precision, and accuracy data are presented.  相似文献

A simple isocratic liquid chromatographic method was developed for the determination of abacavir from its related substances and assay for the first time. This method involves the usage of C18 (Intertsil octadecyl silane-3V, 150 mm x 4.6 mm, 5 microm) column. The method was validated over the range of 0.002-0.1 mg/mL for chloro impurity, 0.005-0.1 mg/mL for amino impurity and pyrimidine impurity, and 0.005-0.2 mg/mL for abacavir. The mobile phase consists of a mixture of 10 mM ammonium acetate buffer and ACN in the ratio of 90:10. The flow rate was set at 1.0 mL/min with UV detection monitored at 214 nm. The drug substance was subjected to stress conditions of hydrolysis, oxidation, photolysis, and thermal degradation. The developed method was validated for linearity, range, precision, accuracy, and specificity. This method can be conveniently used in a quality control laboratory for routine analysis of both related substances and assay.  相似文献

This study addresses the response-to-retention hypothesis, which states that the subendothelial retention of atherogenic lipoproteins is the necessary and sufficient condition for the initiation of atherosclerosis. Here we focus on the relationship between the generation of ceramide in the low-density lipoprotein (LDL) phospholipid monolayer and the resulting aggregation of LDL particles. This study provides the first measurement of neutral, Mg (2+)-dependent Sphingomyelinase (Smase)-mediated ceramide formation from LDL-sphingomyelin and does so for a range of enzyme concentrations (0-0.22 units Smase/mL). The kinetics of ceramide generation was measured using a fluorescence assay for the above enzyme concentrations with a fixed substrate concentration (0.33 mg LDL/mL). The kinetics of LDL aggregate formation was measured by dynamic light scattering (DLS, method of cumulants) for identical enzyme concentrations. Ceramide concentration profiles were fit with a modification of the Michaelis-Menten model ( k a = 1.11 x 10 (-1) microM (-1) min (-1), k -a = 6.54 x 10 (2) microM (-1) min (-1), k 1 = 3.33 x 10 (1) microM (-1) min (-1), k -1 = 1.41 x 10 (-2) min (-1), k cat = 8.05 x 10 (1) min (-1), K M = 2.418 microM, k deact = 4.66 x 10 (-2) microM (-1) min (-1)) that accounts for the effects of enzyme attachment to the LDL monolayer and for deactivation of Smase due to product inhibition. LDL aggregation is described by a mass action model as explained in previous studies. A key result of this work is the finding that LDL aggregate size depends directly on ceramide concentration and is independent of enzyme concentration. This study demonstrates how principles of colloid science are relevant to important biomedical problems.  相似文献

A hyperbranched aliphatic polyester has been functionalized with PEG chains to afford a novel water-soluble BH40-PEG polymer which exhibits unimolecular micellar properties, and is therefore appropriate for application as a drug-delivery system. The solubility of the anticancer drug paclitaxel was enhanced by a factor of 35, 110, 230, and 355 in aqueous solutions of BH40-PEG of 10, 30, 60, and 90 mg x mL(-1), respectively. More than 50% of the drug is released at a steady rate and release is almost complete within 10 h. The toxicity of BH40-PEG was assessed in vitro with A549 human lung carcinoma cells and found to be nontoxic for 3 h incubation up to a 1.75 mg x mL(-1) concentration while LD50 was 3.5 mg x mL(-1). Finally, it was efficiently internalized in cells, primarily in the absence of foetal bovine serum, while confocal microscopy revealed the preferential localization of the compound in cell nuclei. A rapid and specific HPLC method was developed and validated for simultaneous determination of diazepam and its main active metabolites, desmethyldiazepam, oxazepam and temazepam in human plasma. Plasma samples were extracted using toluene. HPLC system included a Chromolith Performance RP-18e 100 mm x 4.6mm column, using 10mM phosphate buffer (pH 2.5)-methanol-acetonitrile (63:10:27, v/v) as mobile phase running at 2 mL min(-1). UV detector (lambda=230 nm) was used. The calibration curves were linear in the concentration range of 2-800 ng mL(-1) for diazepam and 2-200 ng mL(-1) for the three metabolites (r(2)>0.99). The lower limit of quantification was 2 ng mL(-1) for all analytes. Within and between-day precisions in the measurement of QC samples were in the range of 1.8-18.0% for all analytes. The developed procedure was used to assess the pharmacokinetics of diazepam and its main metabolites following single dose administration of 10mg diazepam orally to healthy subjects.  相似文献

Two sensitive and simple spectrophotometric methods are developed for the determination of trazodone HCl, famotidine, and diltiazem HCl in pure and pharmaceutical preparations. The methods are based on the oxidation of the cited drugs with iron(III) in acidic medium. The liberated iron(II) reacts with 1,10-phenanthroline (method A) and the ferroin complex is colorimetrically measured at 510 nm against reagent blank. Method B is based on the reaction of the liberated Fe(II) with 2,2-bipyridyl to form a stable colored complex with lambda(max )at 520 nm. Optimization of the experimental conditions was described. Beer's law was obeyed in the concentration range of 1-5, 2-12, and 12-32 microg mL(-1) for trazodone, famotidine, and diltiazem with method A, and 1-10 and 8-16 microg mL(-1) for trazodone and famotidine with method B. The apparent molar absorptivity for method A is 1.06x10(5), 2.9x10(4), 1.2x10(4) and for method B is 9.4x10(4 )and 1.6x10(4), respectively. The suggested procedures could be used for the determination of trazodone, famotidine, and diltiazem, both in pure and dosage forms without interference from common excipients.  相似文献
