Starting from 6-aryl-4-oxo-2-thioxo-1,2,3,4-tetrahydropyrimidine-5-carbonitrile (4a-d), a series of mono- and dialkyl derivatives 5a-j and 6a, b was synthesized. Hydrazinolysis of 4a, b, d and 5d afforded the hydrazino derivatives 7a-c which were cyclised to give the triazolopyrimidinones 8a-c and the pyrimidotriazinones 9a-c through the reaction with formic acid and chloroacetyl chloride, respectively. Most of the newly synthesized compounds were evaluated for their in-vitro antitumor activity. Compounds 6a and b displayed promising anticancer activity against leukaemia, non-small cell lung, melanoma, and renal cancer. On the other hand, all compounds prepared were screened for their in-vitro antibacterial and antifungal activities. Compounds 5h and j showed significant activity against Staphylococcus aureus, while compounds 5e, 7c and 8c displayed moderate inhibitory activity against Candida albicans.  相似文献

In this research, synthesis, antimicrobial and antioxidant activities of a series of catecholthioethers having benzoxazole and tetrazole moieties are described. Antimicrobial activity was evaluated by minimum inhibitory concentration (MIC) assay. The synthesized compounds were tested in vitro against three Gram-positive bacteria including Staphylococcus aureus (clinical isolated), Staphylococcus aureus ATCC 25922, Enterococcus faecium (clinical isolated), and two Gram-negative bacteria including Klebsiella pneumoniae (clinical isolated) and Pseudomonas aeruginosa 27853 and the yeast Candida albicans in comparison with control drugs. Microbiological results indicated that the synthesized compounds possessed a broad spectrum of activity against the tested microorganisms at MIC values between 4-256 μg/ml. This shows compounds having tetrazole moiety were the most active against Gram-negative strains, whereas compounds having benzoxazole moiety were more active against Gram-positive ones. Also both of them showed significant antifungal activity against Candida albicans and had lower activity than the compared control drugs (Sulfamethoxazole and Fluconazole). The antioxidant activity was assessed using two methods, including, 1,1-biphenyl-2-picrylhydrazyl (DPPH) radical scavenging, and reducing power assays. Some of the catecholthioether derivatives showed antioxidant activity more than Trolox and butylated hydroxyanisole (BHA) as reference antioxidants.  相似文献

Zerumbone 1, having powerful latent reactivity and containing two conjugated double bonds and a double conjugated carbonyl group is the major component of the essential oil of wild ginger, Zingiber zerumbet Smith. The conjugation system plays an important role in the expression of biological activity. N-Bromosuccinimide (NBS) reaction of 1 gave high reactive intermediate 2 with an exo-methylene group, which was obtained from 1 quantitatively. Treatment of 2 with nucleophiles gave various zerumbone-pendant derivatives, including C–H, C–O, C–N, and C–C bond formation, maintaining the conjugation system through SN2′-type reaction. Almost all zerumbone-pendant derivatives showed a good value of IC50 against the suppressive effect of NO generation. Among them, amine derivative 5, binding with 2 mol of zerumbone, showed the strongest activity (IC50: 0.24 μM).  相似文献

Catalyst-free one-pot CN and CC bond formation is described as a simple and ecofriendly method for the synthesis of pyrazole-aniline linked coumarin derivatives. Employing this protocol, a series of derivatives were synthesized in good to excellent yields and tested against different bacterial strains as well as fungal strains. Most of the compounds exhibited potential antimicrobial activity against both Gram-positive and Gram-negative bacterial strains. Among them, the compounds 4b, 4e, 4?h, 4i and 4?k exhibited promising activity on all the tested bacterial strains with values ranging between 1.9 and 7.8?µg/mL. In addition, these compounds were tested against various fungal strains and were found to exhibit potential antifungal activity. Fascinatingly, among the tested derivatives, the compounds 4e, 4?h and 4i were found to be equipotent to miconazole (positive control) against some of the tested fungal strains. Moreover, these compounds showed promising bactericidal, Candida-cidal and biofilm inhibition activities. Further, mechanistic study was carried out with the most active derivative 4i indicated that these compounds inhibit the ergosterol biosynthesis pathway.  相似文献

The absorption and fluorescence properties of nifedipine (NPDHP), felodipine (CPDHP) and a series of structurally related 1,4-dihydropyridines were studied in aqueous solution and organic solvents of different properties. The absorption and fluorescence spectra were found to depend on the chemical nature of the substituents at the position 4 of the 1,4-dihydropyridine ring (DHP) and on solvent properties. In aqueous solution, the fluorescence spectra of 4-phenyl substituted compounds are blue-shifted with respect to the alkyl substituted compounds. The more fluorescent compound is CPDHP. Nifedipine is not fluorescent. All compounds, with the exception of CPDHP, present monoexponential fluorescence decay with very short lifetime (0.2-0.4 ns). CPDHP showed a biexponential emission decay with a long-lived component of 1.7 ns; this behavior is explained in terms of different conformers because of the hindered rotation of the phenyl group by the ortho-substitution. Analysis of the solvent effect on the maximum of the absorption spectrum by using the linear solvent-energy relation solvato-chromic equation indicates the redshifts are influenced by the polarizability, hydrogen bonding ability and the hydrogen bond acceptance of the solvent. Whereas, the fluorescence characteristics (spectra, quantum yields and lifetimes) are sensitive to the polarizabilty and hydrogen bond ability of the solvents. Photo-decomposition of nifedipine is dependent on the solvent properties. Faster decomposition rates were obtained in nonprotic solvents. The 4-carboxylic derivative goes to decarboxylation. Under similar conditions, the other DHP compounds did not show appreciable photodecomposition.  相似文献

A series of new thiazolyl chalcones, 1-[2-amino-4-methyl-1,3-thiazol-5-yl]-3-aryl-prop-2-en-1-one were prepared by piperidine mediated Claisen-Schmidt condensation of thiazolyl ketone with aromatic aldehyde. These chalcones on cyclisation gave 2-amino-6-(2-amino-4-methyl-1,3-thiazol-5-yl)-4-aryl-4H-pyridine-3-carbonitrile and 2-amino-6-(2-amino-4-methyl-1,3-thiazol-5-yl)-4-aryl-4H-pyran-3-carbonitrile. The result showed that the compounds exhibited marked potency as antimicrobial agents.  相似文献

In this Letter, a pair of novel naphthalimide derivatives with long-wavelength emission (>600 nm) and larger Stokes shift (>140 nm) have been developed through the photochemical cycloaromatization, in which intramolecular radical-induced 1,3-aromatic hydrogen transfer might be occurred. Mesoionic pyrimidinium-olate derivatives have been known to undergo photoreactions upon irradiation with UV light to form bis(beta-lactame) structures for about two decades. Here, new mono-, poly- and macrocyclic mesoions were prepared and their photochemical rearrangement behavior was investigated. The synthesized compounds were characterized by NMR, IR, UV/Vis spectroscopy, elemental analysis and mass spectrometry. The extension of the aromatic core leads to a significant red-shift of the absorption maximum from ∼380 to ∼430 nm, indicating a strong electronic coupling of the mesoionic base chromophore with the annellated aromatic subunit. The rigidity of the extended aromatic system prevents polycyclic mesoions from shifting to their bis(beta-lactame) isomers, while the alkyl bridge of the 16-membered macrocycle in an ansa-mesoion does not inhibit photochemical rearrangement.  相似文献

We have described the conventional and microwave method for the synthesis of N-(4-((2-chloroquinolin-3-yl)methylene)-5-oxo-2-phenyl-4,5-dihydro-1H-imidazol-1-yl)(aryl)amides 3a–l. It is observed that the solvent-free microwave thermolysis is a convenient, rapid, high-yielding, and environmental friendly protocol for the synthesis of quinoline based imidazole derivatives when compared with conventional reaction in a solution phase. Antimicrobial activity of the newly synthesized compounds is screened in vitro on the following microbial cultures: Escherichia coli (MTCC 443), Pseudomonas aeruginosa (MTCC 1688), Staphylococcus aureus (MTCC 96), Streptococcus pyogenes (MTCC 442), Candida albicans (MTCC 227), Aspergillus niger (MTCC 282), Aspergillus clavatus (MTCC 1323). All the synthesized bio-active molecules are tested for their in vitro antimicrobial activity by bioassay namely serial broth dilution. Among these compounds 3c, 3d, 3f, 3h and 3j show significant potency against different microbial strains. All the compounds have been characterized by IR, 1H NMR, 13C NMR and mass spectral data. On the basis of statistical analysis, it is observed that these compounds give significant co-relation.  相似文献

Increase in the number of infections caused by pathogenic microbes in cancer patients has prompted the searcher to invest in the development of agents having dual anticancer and antimicrobial properties. The present study is concerned with synthesis and screening for anticancer and antimicrobial activity of a series of 5-hydrazinyl-2-(2-(1-(thien-2-yl)ethylidene)hydrazinyl)thiazole derivatives. The structure elucidation of the synthesized hydrazinyl thiazole derivatives was illustrated by spectroscopic and elemental analysis. All the newly synthesized compounds 5a-p were evaluated for in-vitro cytotoxic activity against breast carcinoma (MCF-7 cell line), hepatocellular carcinoma (HePG-2) and colorectal cancer (HCT-116) cell lines using MTT assay method. Compounds 5 g, 5h showed broad spectrum activity against three cancer cell lines with IC50 ranged from 3.81 to 11.34 µM in compared to the reference drug Roscovitine (IC50 = 9.32 to 13.82 µM), while compounds 5 l and 5 m were found to be more selective against HePG-2 and HCT-116 cell line (IC50 = 9.29 and 8.93 µM respectively) and compound 5j was more selective against HePG-2 and MCF-7 cell lines (IC50 = 6.73 and 10.87 µM respectively). The inhibitory activity of the most promising compounds was tested against the EGFR and ARO enzymes and were further tested for apoptosis and Annexin V/PI staining. The results of enzyme-based tests revealed that the tested compound 5j has a dual inhibitory effect on the EGFR and ARO enzymes with IC50 = 82.8 and 98.6 nM respectively in compared to the reference drugs Erlotinib and Letrozole (IC50 = 62.4 and 79 nM respectively). Furthermore, the majority of the tested hydrazinyl thiazole derivatives exhibited significant antimicrobial activity against the used pathogenic microbes species. Compounds 4b, 5h, 5j and 5 m exerted a good antibacterial and antifungal activity against all tested pathogenic microbes. Therefore, it was concluded that compounds 5 h, 5j and 5 m proved to possess dual anticancer and antimicrobial agent and may serves as a useful lead compounds in search for further modification or derivatization to give more potent and selective agents.  相似文献

This review presents an overview of silver nanoparticles (Ag NPs) preparation by green synthesis approaches that have advantages over conventional methods involving chemical agents associated with environmental toxicity. Green synthetic methods include mixed-valence polyoxometallates, polysaccharide, Tollens, irradiation, and biological. The mixed-valence polyoxometallates method was carried out in water, an environmentally-friendly solvent. Solutions of AgNO(3) containing glucose and starch in water gave starch-protected Ag NPs, which could be integrated into medical applications. Tollens process involves the reduction of Ag(NH(3))(2)(+) by saccharides forming Ag NP films with particle sizes from 50-200 nm, Ag hydrosols with particles in the order of 20-50 nm, and Ag colloid particles of different shapes. The reduction of Ag(NH(3))(2)(+) by HTAB (n-hexadecyltrimethylammonium bromide) gave Ag NPs of different morphologies: cubes, triangles, wires, and aligned wires. Ag NPs synthesis by irradiation of Ag(+) ions does not involve a reducing agent and is an appealing procedure. Eco-friendly bio-organisms in plant extracts contain proteins, which act as both reducing and capping agents forming stable and shape-controlled Ag NPs. The synthetic procedures of polymer-Ag and TiO(2)-Ag NPs are also given. Both Ag NPs and Ag NPs modified by surfactants or polymers showed high antimicrobial activity against gram-positive and gram-negative bacteria. The mechanism of the Ag NP bactericidal activity is discussed in terms of Ag NP interaction with the cell membranes of bacteria. Silver-containing filters are shown to have antibacterial properties in water and air purification. Finally, human and environmental implications of Ag NPs to the ecology of aquatic environment are briefly discussed.  相似文献

The syntheses of new three phthalonitriles (1, 2 and 3), together with photophysical and photochemical properties of the resulting peripherally and non-peripherally tetrakis- and octakis 3,4-(methylendioxy)-phenoxy-substituted zinc phthalocyanines (4, 5 and 6) are described for the first time. Complexes 4, 5 and 6 have been synthesized and characterized by elemental analysis, IR, 1H NMR spectroscopy, electronic spectroscopy and mass spectra. Complexes 4, 5 and 6 have good solubility in organic solvents such as CHCl3, DCM, DMSO, DMF, THF and toluene and are mainly not aggregated (except for complex 6 in DMSO) within a wide concentration range. General trends are described for singlet oxygen, photodegradation, fluorescence quantum yields, triplet quantum yields and triplet life times of these complexes in DMSO and toluene. Complex 4 has higher singlet oxygen quantum yields, fluorescence quantum yields, triplet quantum yields and triplet life times than complexes 5 and 6. The effect of the solvents on the photophysical and photochemical parameters of the zinc(II) phthalocyanines (4, 5 and 6) are also reported.  相似文献

2-(p-Acetylaminobenzenesulfonylamido)-substituted benzothiazoles were prepared from 2-amino-substituted benzothiazoles and p-acetamidobenzenesulfonyl chloride using a mixture of pyridine and Ac2O, which formed an electrophilic N-acetyl- pyridinium complex facilitating condensation to give the desired products by removal of HCl. 2-[4-(Substituted benzothiazol-2-yl)aminosulfonylanilino]pyridine-3-carboxylic acids (synthesized from 2-chloropyridine-3-carboxylic acid and the corresponding substituted 2-(p-aminobenzenesulfonylamido)benzothiazole in 2-ethoxyethanol using Cu-powder and K2CO3) were then converted to acid chlorides, which on further reaction with piperazine and 4-methoxyphenylpiperazine yielded the corresponding 2-[4-(substituted benzothiazol-2-yl)amino-sulfonyl]anilino-3-(piperazinocarbonyl) pyridine and 2-[4-(substituted benzothiazol-2-yl)amino-sulfonyl]anilino-3-[(4-methoxyphenyl)piperazin-1-yl-carbonyl]pyridine. The structures of the new compounds have been established on the basis of their elemental analyses as well as IR, 1H NMR, and mass-spectral data. All the compounds have been screened for antimicrobial activity and found to possess considerable antibacterial activity.  相似文献

Abstract

A series of new 5-amino-7-bromoquinolin-8-ol sulfonate derivatives 5(aj) were synthesized from 8-hydroxyquinoline through multi-step process with high yields using mild, efficient and conventional methods. Chemoselectivity was observed during the transformation of 5-amino-7-bromoquinolin-8-ol to 5-amino-7-bromoquinolin-8-ol sulfonate with various sulfonylchlorides exclusively to afford sulfonate derivatives. Also, the products were investigated for their in vitro antimicrobial activities and compared with the standard drugs. Among all the synthesized compounds 5-amino-7-bromoquinolin-8-yl biphenyl-4-sulfonate (5b) and 5-amino-7-bromoquinolin-8-yl 2-hydroxy-5-nitrobenzenesulfonate (5g) have showed potent antibacterial activity, whereas 5-amino-7-bromoquinolin-8-yl biphenyl-4-sulfonate (5b) and 5-amino-7-bromoquinolin-8-yl 2-hydroxy-5-nitrobenzenesulfonate (5g) possessed potent antifungal activities among all the tested pathogens.  相似文献

A series of novel oxadiazole functionalized pyridopyrimidine derivatives prepared starting from 6-methyl/ethyl-2-oxo-4-(trifluoromethyl)-1,2-dihydropyridine-3-carbonitrile 1 . This compound 1 on reaction with sulfuric acid obtained compound 2 , further compound 2 on reaction with chloroacetamide followed by reaction with ethoxy methylene malonic diethyl ester coupling and further cyclization to obtain compound 5 . Compound 5 on reaction with hydrazide hydrate obtained hydrazide derivatives 6 . Compound 6 on reaction with diverse substituted aromatic acids to get oxadiazole derivatives 7a–l . All the final compounds 7a–l evaluated for anticancer activity against four human cancer cell lines such as HeLa—cervical cancer (CCL-2); COLO 205—colon cancer (CCL-222); HepG2—liver cancer (HB-8065); and MCF7—breast cancer (HTB-22) and promising compounds 7d and 7k have been identified and evaluated for molecular docking interactions.  相似文献
