The relationships between electromigration dispersion (EMD) and on-line isotachophoresis-capillary zone electrophoresis (ITP-CZE) are described for several basic model proteins and interleukin-6 (rhIL-6). During CZE separation of the highly concentrated analyte zones which were generated during the initial ITP step EMD evolves from intrinsic differences in conductivity between the focused ITP zones and the leading electrolyte. Nearly triangular peaks with a sharp front and diffuse rear side were observed. An electromigration dispersion factor (FEMD) was introduced to measure peak asymmetry. EMD of individual peaks was shown to increase with the absolute amount of the respective analyte injected and with analyte mobility. Good linearity was observed when FEMD was plotted against protein mobility (r > 0.95). The slope of the graphs describing this relationship increased with the amount of analyte injected. The influence of EMD on the separation efficiency of neighboring peaks appeared to be less pronounced than expected. Consecutive release from the ITP-stack during transition from ITP to CZE might be an explanation for this observation.  相似文献

Several metal-binding proteins, including albumin, carbonic anhydrase, conalbumin, cytochrome c, ferritin, hemoglobin, myoglobin, plasma amine oxidase, superoxide dismutase and transferrin were separated with capillary zone electrophoresis (CZE) in uncoated and coated capillaries. Phosphate and tetraborate buffers achieved complementary separation selectivities. Optimised pre-wash protocols for uncoated capillaries using 0.1 M HCl as a rinsing solution for the borate buffer and a combination of 0.1 M NaOH and 0.1 M HCl for the phosphate system improved the stability of migration times considerably with coefficients of variation between 0.10 and 0.77% (n=7) instead of up to 2.92% with inappropriate rinsing conditions. Capillaries coated with poly(vinyl alcohol) and equipped with a 150 μm i.d. bubble cell increased the signal-to-noise ratio by a factor three, additionally improving the resolution. For commercial protein standards, which gave several peaks in CZE with UV detection, MS data proved the presence of proteinaceous contaminants. Molecular weights (Mr) of proteins experimentally determined from MS data showed deviations from theoretical Mr as small as 0.002-0.021%. Applicability of the developed separation for clinical samples is shown for human serum.  相似文献

The properties and behavior of polyethyleneimine (PEI) covalently coated capillaries with respect to different background electrolytes used in capillary zone electrophoresis (CZE) are described. The coating stability and changes of inner surface charge in the capillary were followed by measurement of electroosmotic flow (EOF). Interest was focused mainly on conjugate bases of carboxylic acids as anionic background electrolyte components (acetate, citrate, malate, malonate, tartrate, and succinate). An interesting phenomenon was observed in PEI-coated capillaries: The direction (and the magnitude) of EOF depends on the composition of the background electrolyte and at a certain pH it can undergo reversible change. Ionic complex formation was suggested as a hypothesis to explain this behavior. With this knowledge, the PEI-coated capillary was used for the separation of basic proteins in the above-mentioned background electrolytes. A standard protein mixture of cytochrome c, ribonuclease A, and lysozyme at a concentration of 0.25 mg/mL each was chosen as model sample.  相似文献

The feasibility of using the affinity CE methodologies pre-equilibrium CZE and CE frontal analysis was tested on interaction systems exhibiting rapid on-and-off kinetics. Experimentally, the methodologies differ only with respect to the volume of sample introduced into the capillary. Pre-equilibrium CZE has been considered amendable to interactions with slow on-and-off kinetics only; however, it has recently been applied in studies of interactions with fast on-and-off kinetics. The effect of varying the sample volume introduced hydrodynamically into the capillary on the apparent degree of complexation was studied. For two different binding systems, the fraction of free analyte was found to be overestimated using pre-equilibrium CZE as compared to volumes providing plateau peak conditions as used with frontal analysis. Results indicate that frontal analysis conditions lead to more robust binding assays and thus more reliable data. The validity of data obtained by pre-equilibrium CZE may be low, thus the use of an experimental setup providing plateau peaks is highly recommended. It is suggested that the effect of altering the sample volume on the degree of binding should be investigated as part of method development and validation.  相似文献

Capillary zone electrophoresis (CZE) of serum proteins has become a well-accepted method for the separation of serum proteins and for the detection of monoclonal proteins in clinical laboratories. As CZE uses ultraviolet detection for direct protein quantification, exogenous ultraviolet-absorbing substances are a novel challenge, not present in conventional gel-based methods in which protein is quantified by dye binding. Poor sensitivity and low phase ratio are the main drawbacks of open tubular capillary electrochromatography (OTCEC). The poor sensitivity results from the use of narrow bore size capillary, whereas the low phase ratio, which limits the separation capability, is caused by the limited surface area of conventional capillary. Two strategies may be useful to overcome these disadvantages. First, an extended light path (ELP) capillary, which has a bubble cell at the detection point, is used to improve the sensitivity. Secondly, an etched capillary of a 1,000-fold increased surface area is used to enhance the phase ratio. In this work, use of an ELP capillary and an etched capillary in OTCEC was evaluated with a chiral stationary phase of avidin prepared with the physical adsorption method. With a 20 microm I.D. ELP capillary with a 150 microm bubble cell, the peak height was enhanced by 4-10-fold and the corrected peak area was increased by 12-fold relative to a 20 microm I.D. conventional capillary. However, the peak efficiency and resolution decreased noticeably. The phase ratio on the etched capillary was slightly enhanced, by a factor of 1.64 relative to an unetched capillary. Consequently, the separation capability was slightly improved. The increase in the phase ratio was much lower than that expected from the increase in surface area, the reason for which is probably the reduced density of surface silanol group and the generation of nitrogen-containing groups due to the etching process.  相似文献

The present paper deals with the role of the solvent on thermal peak broadening. One main solvent property that determines the magnitude of the temperature gradient due to the generation of Joule heat in capillary zone electrophoresis is the thermal conductivity. As organic solvents have lower thermal conductivity than water (methanol and acetonitrile, e.g., nearly by a factor of 3) it can be hypothesized that the temperature gradient inside the capillary is more pronounced in organic solvents compared to an aqueous solution. On the other hand, the temperature dependence of the ion mobility (which is responsible for the velocity profile and thus for thermal peak broadening) is smaller in organic solvents. To get insight into the thermal effect of the solvent, first the temperature of a solution in a cylindrical tube was calculated utilizing the heat balance equation. It was shown that the two theoretical models most common in the literature (based on the analytical solution or on an assumption of the parabolic temperature profile in the tube, respectively) give the same results. The latter model was chosen for the further calculations, adding a quadratic term to express the electric conductivity as a function of the temperature. The temperature at the inner capillary wall and center as function of the capillary dimensions and the electric power was computed for electrolytes with a given conductivity at 25.0 degrees C with water, methanol, and acetonitrile as solvents. Capillary cooling systems used were circulating liquid cooling, enforced air-cooling, and natural convection in still air. The mean temperature (averaged over the cross section) resulting from Joule heating was compared with experimentally determined temperatures established upon application of an electric field; the latter temperature was derived from the measurement of the electric conductance of the background electrolyte solution and its (measured) temperature dependence. All investigations were carried out with solutions of the same initial electric conductivity (about 0.5 S.m(-1) at 25.0 degrees C). Agreement is found for natural convection conditions, and the deviation between theoretical and experimental results for the forced air and circulated liquid cooling systems can be related to the poorly defined thermal conditions of the capillaries in commercial instrumentation (with a part in a thermostated cassette and a part outside). For given conditions the temperature gradients in the organic solvents exceed largely those in water, independent of the type of cooling. As a consequence, the thermal plate height is significantly larger in organic solvents, at least under conditions where the deviation from the Nernst-Einstein limiting case is not too high. However, even for the maximum applicable field strengths the thermal plate height contributions are negligible compared to longitudinal diffusion in all solvents.  相似文献

建立了毛细管区带电泳分析五步蛇毒蛋白C激活剂(protein C activator, PCA)与血浆蛋白C(protein C, PC)的相互作用。以未涂层毛细管(60.2 cm(有效长度50 cm)×75 μm)为分离柱,50 mmol/L Tris-HCl(pH 7.4)为运行缓冲液,于198 nm波长下检测。对影响五步蛇毒PCA分离的因素(如缓冲液、离子浓度)及在37.5 ℃下孵育不同时间的五步蛇毒PCA与PC的相互作用进行考察。方法的检出限(以信噪比为3计)为3 mg/L,线性范围为10~300 mg/L。五步蛇毒PCA迁移时间和峰面积的相对标准偏差分别为0.56%和3.8%(n=6)。等体积的五步蛇毒PCA(200 mg/L)与PC(60 mg/L)孵育5 min,其结合率达到最大,且谱图中未见有PC水解的肽链。该五步蛇毒PCA可改变PC空间构象直接激活PC。该方法简单,灵敏度和分辨率高,分析结果为今后快速检测五步蛇毒PCA及其活性提供了重要的理论依据。  相似文献

We report an electrophoretic mobility shift-based method to study the interactions between phospholipids and proteins by capillary electrophoresis with laser-induced fluorescence detection. A fluorogenic dye, 3-(2-furoyl)quinoline-2-carboxaldehyde (FQ), was used to label phosphatidylserine (PS). Then the FQ labeled PS (FQ-PS) was used as the fluorescent probe for monitoring the association between PS and bovine serum albumin (BSA). Two conjugates were observed to form between each PS species and BSA, indicating that two interactions exist between these PS species and BSA. We can also detect the competitive association with BSA between labeled PS and unlabeled PS. This method only needs a minute volume of sample. It is highly sensitive and can be used to detect the interaction between phospholipids and nanomolar concentrations of proteins, such as BSA.  相似文献

The interactions between granulocyte-macrophage colony-stimulating factor (GM-CSF) and heparin or low-molecular weight heparin (LMWH) were studied by CZE. It was found that GM-CSF could bind to both heparin and LMWH. The binding constants were calculated from Scatchard regression to be (6.5 +/- 0.8) x 10(5)/M and (11.2 +/- 0.7) x 10(5)/M, respectively. The specificity of the interaction between GM-CSF and heparin was also studied by employing another sulfated K carrageenan oligosaccharide as a control. Results showed that K carrageenan oligosaccharide could not interact with GM-CSF, indicating that GM-CSF could specifically interact with heparin.  相似文献

The paper describes a method of size separation of proteins by capillary sieving electrophoresis with cationic surfactant. Proteins are separated within 12 min with repeatability of migration times better than 0.2%. Some proteins achieve the separation efficiency of 200,000 theoretical plates. The method can be used for determination of protein relative molecular masses. The accuracy of the determined relative molecular masses and the limitation of the method were investigated by the analysis of more than 60 proteins. The method also allows separation of protein oligomers. Proteins can be quantitated after the electrokinetic injection in the concentration range 0.07-0.43?g/L. The average detection limit is about 2?mg/L.  相似文献

The use of quasi-isoelectric buffers consisting of narrow pH cuts of carrier ampholytes (NC) has been investigated to limit protein adsorption on capillary walls during capillary zone electrophoresis experiments. To quantify protein adsorption on the silica surface, a method derived from that of Towns and Regnier has been developed. alpha-Lactalbumin (14 kDa, pI 4.8) and alpha-chymotrypsinogen A (25 kDa, pI 9.2) have been used as model proteins. Acidic narrow pH cuts of carrier ampholytes (NC, pH 3.0) obtained from fractionation of Serva 4-9 carrier ampholytes were used as BGE in bare-silica capillaries, and allowed to decrease significantly protein adsorption, as compared to experiments performed with classical formate buffer. The use of NC as BGE appeared to be as efficient as the use of polydimethylacrylamide coating to prevent protein adsorption. This increase of protein recovery when using NC was attributed to the interaction of carrier ampholytes with the silica surface, leading to a shielding of the capillary wall.  相似文献

In CZE, the adsorption of the proteins on the capillary wall is a common problem. This paper describes the simple method of utilizing zwitterionic buffer additives to improve the separation of proteins in untreated fused silica capillaries at neutral pH. Three kinds of zwitterion are evaluated in the separation of acidic, neutral, and basic proteins, including their effect on protein efficiency, mobility, separation, and resolution; the difference between the effects of the different additives are also highlighted. The method has proved to be a possible means of reducing protein adsorption, especially for basic proteins.  相似文献

Platinum adducts are supposed to be the cytotoxic lesions in DNA after platinum-containing anticancer therapy. Various adducts are formed upon interaction of platinum complexes with nucleotides, but contribution of individual adducts to antitumor activity and toxicity of platinum complexes still remains to be examined. A capillary zone electrophoresis (CZE) method is described that is suitable to separate individual platinum adducts. We investigated the formation of adducts following the reaction of cis-diamminedichloroplatinum (II) (cisplatin) with various DNA nucleotides. Baseline separation of unmodified and modified nucleotides (adducts) was achieved using uncoated fused-silica capillaries and basic separation buffers. In order to elucidate the observed peak pattern, a coupled CZE-electrospray ionization-mass spectrometry (ESI)-MS approach was applied. After incubation of mononucleotides with cisplatin, monochloro, monoaqua and bifunctional adduct species were detected. Consequently, the migration order of nucleotides and individual platinum adducts could be determined. Moreover, the time-dependent conversion from monochloro to monoaqua and subsequently to bifunctional adducts was monitored. In conclusion, individual platinum adducts were separated by CZE and identified by CZE-ESI-MS. Formation and conversion of distinct species were confirmed. Potential applications comprise studies of novel platinum complexes, investigations of platinum-adduct formation with DNA, and determination of platinum-DNA adducts in cells.  相似文献

A method to extend the separation power of CZE is described. The method is based on the separation of sample components at two different pH values during one separation run, and involves dynamic buffering of the pH inside a separation capillary by controlling the flow of H+ ions from the anodic electrode chamber. By changing the anolyte in the chamber, a dynamic pH step is generated, which proceeds rapidly along the capillary and establishes the required new pH value. The use of the method has been demonstrated by the cationic separation of a model mixture of proteins.  相似文献

The use of capillary zone electrophoresis (CZE) and capillary zone electrophoresis/mass spectrometry (CZE/MS) has been demonstrated, in principle, for the separation of nicotine and nicotine metabolites. The buffer system developed for separation and detection by CZE/UV was modified for use in CZE/MS analysis. Several of the metabolites are isobaric and tandem mass spectrometric (MS/MS) techniques have been used to differentiate such analytes.  相似文献

We have recently described [1] a fast and simple method for the "adsorbed static" coating of capillaries in capillary zone electrophoresis (CZE) with epoxy-poly(dimethylacrylamide) (EPDMA). Protein CZE peaks in the EPDMA-coated capillaries exhibited a peak asymmetry similar to that obtained in capillaries with "covalent static" coating of polyacrylamide, suggesting a similar degree of adsorption of the protein onto the coating [2]. Instability of such coating at very low ionic strength and its stripping from the capillary in the presence of sodium dodecyl sulfate (SDS) also indicated a hydrophilic bonding of EPDMA to the silanol surface of the capillary, while its stripping in the CZE of "carboxylate-modified" polystyrene suggested a competition between carboxylate and EPDMA for the hydrophilic bonds to silanol. To test those propositions, a number of EPDMA-derived coating agents with increased hydrophilicity were synthesized. Of a number of the hydrophilic coating agents tested (Table 1) only two, 2% hydrolyzed EPDMA (HPDMA) hydrolyzed in sulfuric acid to effect the conversion of the epoxy groups to diols (Table 1, No. 38), and 20% EPDMA (Table 1, No. 44) exhibited for representative proteins a decreased peak asymmetry and width while the stability of the suppression of electroosmotic flow (EOF), and the stability of mobility in consecutive CZE runs was reduced relative to EPDMA. Coating agents which were more highly hydrophilic than those two (Table 1, No. 49) or less hydrophilic than 2% EPDMA (Table 1, Nos. 57, 53, 46) provided no stable static coating.  相似文献

Alpha-1-antitrypsin (AAT) and alpha-1-acid glycoprotein (AAG) are reported to be the main proteins contributing to the alpha-1-globulin capillary zone electrophoresis (CZE) zone, but the sum (AAT + AAG) showed lower than the alpha-1-globulin. We investigated the role of high-density lipoprotein (HDL), an additional protein migrating in the alpha-1-globulin zone, as a possible cause for such a gap. In a set of 98 sera we measured the alpha-1-globulin with a dedicated clinical capillary electrophoresis system, and AAT, AAG and apolipoprotein A-1 (ApoA) by immunonephelometry. The alpha-1-globulin were consistently higher than the sum (AAT + AAG), by (mean value +/- standard deviation) 1.70 +/- 0.88 g/L in 49 sera with low ApoA, and by 3.59 +/- 0.75 g/L in 49 sera with high ApoA. Corresponding figures in the comparison alpha-1-globulin/(AAT + AAG + ApoA) were reduced to 1.08 +/- 0.77 g/L and 1.67 +/- 0.70 g/L. It is concluded that HDL significantly contribute to the CZE alpha-1-globulin zone, but do not completely explain the differences between the electrophoretic and the immunochemical measurements. However, CZE alpha-1-globulin measurements give information about increases of the two major acute phase proteins comparable to specific protein measurements.  相似文献
