Quite unlike the reported facile ene reactions on the periphery of many related heterocyclic systems, similarly disposed moieties on the periphery of the chromen‐4‐one (=4H‐1‐benzopyran‐4‐one) system fail to undergo an ene reaction and display a rather unusual preference for an overall [1,5] shift of the allylic C‐atom. Thus, heating xylene solutions of 2‐(N‐allylanilino)‐, 2‐(N‐crotylanilino)‐, and 2‐(N‐cinnamylamino)‐substituted (E)‐(oxochromenyl)propenoates 9a – c and 2‐[allyl(benzyl)amino]‐, 2‐[benzyl(crotyl)amino]‐, and 2‐[benzyl(cinnamyl)amino]‐substituted (E)‐(oxochromenyl)propenoates 16a – c in a sealed tube at 220–230° leads to a [1,5] shift of the allylic moieties (allyl, crotyl, cinnamyl), which is followed by intramolecular cyclization involving the N‐atom and the ester function, to give the 3‐allyl‐3‐crotyl‐, and 3‐cinnamyl‐substituted‐1‐phenyl‐ or 1‐benzyl‐2H‐[1]benzopyrano[2,3‐b]pyridine‐2,5(1H)‐diones 10a – c and 17a – c . The anticipated carbonyl–ene reaction in the 2‐(N‐allylanilino)‐, 2‐(N‐crotylanilino)‐, 2‐(N‐cinnamylanilino)‐, 2‐[allyl(benzyl)amino]‐, 2‐[benzyl(crotyl)amino]‐, and 2‐[benzyl(cinnamyl)amino]‐substituted 4‐oxochromene‐3‐carboxaldehydes 8a – c and 15a – c is also not observed, and these molecules remain untransformed under identical conditions. No [1,5] shifts of benzyl, phenyl, or methyl groups are observed, even in the absence of allylic moieties, though facile [1,5]‐H shift occurs in 2‐(benzylamino)‐ and 2‐(phenylamino)‐substituted (E)‐(oxochromenyl)propenoates 23a , b , which is followed by a similar intramolecular cyclization leading to the 2H‐[1]benzopyrano[2,3‐b]pyridine‐2,5(1H)‐diones 24a , b .  相似文献

Reactions of benzyl bromide or benzyl cinnamate with N-(benzotriazol-1-ylmethyl)arylimidoyl chlorides (2a,b) in the presence of t-BuOK occur with opening of the benzotriazole ring affording 1,2,4-triazolo[1,5-a]quinoxalines (3a,b). A possible reaction mechanism is discussed.  相似文献

爱滋病(AIDS)是一种严重威胁人类健康的传染性疾病。研究和开发新的、高疗效的抗AIDS的药物已成了生物学家、化学家和药物学家的主要的研究方向之一。2',3'-双脱氢.2',3'-双脱氧胸苷(d4T)是HIV逆转录酶抑制剂,它们是美国FDA通过的可用于治疗AIDS的一类药物。由于d4T等本身对HIV无活性,只有经细胞代谢成5'-磷酸化产物后才对HIV产生抑制作用,因此很有必要对d4T进行结构改造,而对上述药物的结构进行改造是研制新的抗AIDS的化学药物最简捷的途径。因此,我们合成了一系列的d41、5'-亚磷酸酯的衍生物,并对它们的电喷雾质谱进行了详细的研究。研究发现了2',3'-双脱氢,2',3'双脱氧胸苷(d4T)-5'-氢亚磷酸苄酯的一种新颖的重排反应,并对其机理进行了进一步的研究。有关结果不仅有助于对这类化合物的结构分析,而且对它们的合成、药物性质以及药物代谢等方面的研究都具有重要意义。  相似文献

A very efficient method of preparation for 5-alkyl and 5-arylthiotetrazoles from the corresponding alkyl or aryl halides is described. The halides are first transformed into thiocyanates which further react with azide, yielding the corresponding tetrazoles with [2+3] polar cycloaddition. All synthetic transformations are performed under phase transfer catalytic conditions. The yields vary from good to excellent except for the preparation of 5-benzylthiotetrazole, where the reaction between benzyl thiocyanate and azide [2+3] cycloaddition is in competition with nucleophilic substitution, with benzyl azide as product.  相似文献

[structures: see text] A systematic study was performed to establish general synthesis protocols for forming enantiomerically pure macrocyclic dipeptide lactams. Focusing on macrocycles of 8-, 9-, and 10-membered rings, effective syntheses were achieved by a sequence featuring peptide coupling of allyl- and homoallyl-glycine building blocks followed by ring-closing metathesis. The 8-membered lactam-possessing cis-amide and cis-olefin geometry as well as 9-membered [corrected] lactams having trans-amide and cis-olefin [corrected] configurations were effectively prepared by a general strategy employing the respective protected dipeptide, the first generation Grubbs' catalyst, and temporary protection of the central amide as a benzyl derivative. The 10-membered macrocycle was synthesized possessing cis- or trans-olefin geometry by employing similar metathesis conditions in the presence or absence of temporary benzyl amide protection, respectively [corrected]  相似文献

A simple and practical synthesis of the benzyl, allyl, and 4-nitrobenzyl esters of N-[2-(Fmoc)aminoethyl]glycine is described starting from the known N-(2-aminoethyl)glycine. These esters are stored as stable hydrochloride salts and were used in the synthesis of peptide nucleic acid monomers possessing bis-N-Boc-protected nucleobase moieties on the exocyclic amino groups of ethyl cytosin-1-ylacetate, ethyl adenin-9-ylacetate and ethyl (O(6)-benzylguanin-9-yl)acetate. Upon ester hydrolysis, the corresponding nucleobase acetic acids were coupled to N-[2-(Fmoc)aminoethyl]glycine benzyl ester or to N-[2-(Fmoc)aminoethyl]glycine allyl ester in order to retain the O(6) benzyl ether protecting group of guanine. The Fmoc/bis-N-Boc-protected monomers were successfully used in the Fmoc-mediated solid-phase peptide synthesis of mixed sequence 10-mer PNA oligomers and are shown to be a viable alternative to the currently most widely used Fmoc/Bhoc-protected peptide nucleic acid monomers.  相似文献

A heretofore unknown LDA-induced conversion of benzyl 1-alkynyl sulfones to 1H-2-benzothiopyran-S,S-dioxides is demonstrated. A benzyl carbanion, accessible by two means, is thought to cyclize by temporary disruption of aromaticity. Key intermediates are observed by ReactIR analysis. Thiophene derivatives are also amenable to cyclization, forming 7H-thieno[2,3-c]thiopyran-S,S-dioxides.  相似文献

[reaction: see text] A series of 4-amino-1H-pyrrole-2-carboxylic acid benzyl esters has been synthesized in 61-84% yields on treatment of N-PhF-4-oxoproline benzyl ester and its 3-alkyl-substituted derivatives with different primary and secondary amines and a catalytic amount of TsOH in THF. 4-Hydroxy-1H-pyrrole-2-carboxylic acid benzyl esters were prepared in 59 and 70% yields by treatment of N-PhF-4-oxoproline benzyl esters with ammonium hydroxide in THF.  相似文献

The driving force for the self-assembly process is the molecular recognition that exists as a result of [N(+)-H.O] and [C-H.O] hydrogen bonds between an NH(2)(+) center in one Fréchet-type benzyl ether wedge and a dibenzo[24]crown-8 unit that links the other two such wedges.  相似文献

A novel thiacalix[4]arene ditopic receptor with 1,3-alternate conformation and possessing two complexation sites for hard and soft cations, 5,11,17,23-tetra-tert-butyl-25,27-bis[(N,N-diethylaminocarbonyl)methoxy]-26,28-bis[(pyridylmethyl)oxy]-2,8,14,20-tetrathiacalix[4]arene is prepared. Regioselective synthesis of distal-bis[(N,N-diethylaminocarbonyl)methoxy]thiacalix[4]arene is accomplished by a protection-deprotection method using benzyl groups as a protecting group. The deprotection of benzyl group was succeeded in the presence of solid superacid (Nafion-H) under refluxing benzene. Its complexation behavior is examined by 1H-NMR titration experiments. The formation of 1:2 homo- and heteronuclear complexes demonstrates that the preorganization, subtle conformational changes and affinity have a pronounced effect on the complexation of the receptor.  相似文献

The eleven-step preparation of the bicyclic 2-pyridone dipeptide mimetic 1 [(3S)-6-(benzyloxycarbonylamino)-5-oxo-1,2,3,5-tetrahydroindolizine-3-carboxylic acid] in optically active form (60% ee) is described. Key steps in the synthesis of 1 include the osmium-catalyzed asymmetric dihydroxylation of olefin 13 [(6-but-3-enyl-2-methoxypyridin-3-yl)carbamic acid benzyl ester] and the intramolecular cyclization of protected diol 19 [(3'R)-[6-[4'-(tert-butyldimethylsilanyloxy)-3'-hydroxybutyl]-2-methoxypyridin-3-yl]carbamic acid benzyl ester] to afford the pyridinium salt 20 [(3S)-[3-(tert-butyldimethylsilanyloxymethyl)-5-methoxy-2,3-dihydro-1H-indolizin-6-yl]carbamic acid benzyl ester trifuoromethanesulfonic acid salt]. Several alternate methods to prepare olefin 13 are also discussed.  相似文献

The efficient synthesis of a new family of 2,6-disulfanyl-9-selenabicyclo[3.3.1]nonanes in high yields has been developed based on 9-selenabicyclo[3.3.1]nonane-2,6-dithiolate anion generated from bis-isothiouronium salt of 2,6-dibromo-9-selenabicyclo[3.3.1]nonane. The derivatives of 2,6-disulfanyl-9-selenabicyclo[3.3.1]nonane containing alkyl, allyl and benzyl moieties have been prepared in 90–99% yields by nucleophilic substitution of 9-selenabicyclo[3.3.1]nonane-2,6-dithiolate anion with alkyl, allyl and benzyl halides. The reaction of nucleophilic addition of 9-selenabicyclo[3.3.1]nonane-2,6-dithiolate anion to alkyl propiolates afforded 2,6-di(vinylsulfanyl)-9-selenabicyclo[3.3.1]nonanes. The conditions for regio- and stereoselective addition of 9-selenabicyclo[3.3.1]nonane-2,6-dithiolate anion to a triple bond of alkyl propiolates have been found. To date, not a single representative of 2,6-disulfanyl-9-selenabicyclo[3.3.1]nonanes has been described in the literature.  相似文献

A convenient Pictet-Splengler approach for the synthesis of substituted pyrrolo[1,2-a]quinoxalines from aryl amine and 1-(2-aminoaryl)-pyrrole has been developed. This is the first domino protocol involving formation of benzaldehyde as an intermediate from benzyl amines and benzyl alcohols using 1,2-DNB as an oxidant, followed by its endo cyclization with 1-(2-aminophenyl)-pyrrole to furnish the pyrrolo[1,2-a]quinoxalines. Notably, this procedure exhibits good functional group tolerance as well as good to moderate yield of pyrrolo[1,2-a]quinoxaline.  相似文献

Bulky lateral carboxylate substituents were introduced at the benzylic position of 2,5-bis[4-(n-perfluoroheptyloctyloxy)benzoyloxy]toluene by esterification of the corresponding benzyl bromide with potassium carboxylates. In spite of the bulky lateral substituents, none of the 2,5-bis[4-(n-perfluoroheptyloctyloxy)benzoyloxy]benzyl carboxylates exhibit a nematic mesophase in addition to, or instead of, the smectic mesophases. All of the crystalline and SmC-SmA transition temperatures and, with the exception of the 9-anthracene carboxylate derivative, all of the isotropization temperatures of the resulting 2,5-bis[4-(n-perfluoroheptyloctyloxy)benzoyloxy]benzyl carboxylates are lower than those of the parent toluene compound. The SmC-SmA transition decreases the most, thereby stabilizing the SmA mesophase. In most cases, the SmC mesophase is destabilized from an enantiotropic to a monotropic mesophase. There is no correlation between any of the transition temperatures and the size of the lateral substituent.  相似文献

通过大分子反应,将苯甲醛(BA)和邻氨基苯酚(AP)形成的双齿席夫碱配基键合在交联聚甲基丙烯酸缩水甘油酯(CPGMA)微球表面,形成固载有席夫碱配基的载体微球BAAP-CPGMA,再通过与铜盐的配位螯合反应,制备了固载有席夫碱铜配合物的微球[Cu(BAAP)2]-CPGMA.将该固载化铜配合物与均相的2,2,6,6-四甲基哌啶氮氧自由基(TEMPO)构成共催化体系TEMPO/[Cu(BAAP)2]-CPGMA,应用于分子氧氧化苯甲醇的催化氧化过程.我们考察了该共催化体系的催化性能,并探索研究了催化氧化机理.实验结果表明,共催化体系TEMPO/[Cu(BAAP)2]-CPGMA可在温和条件下(室温、常压的氧气)高效地将苯甲醇氧化为苯甲醛(选择性100%,苯甲醛产率93%),并具有良好的循环使用性能.  相似文献

A potent thromboxane antagonist, 1-[2-(2-carboxyethyl)benzyl)]-2-benzenesulfonamidobicyclo-[2.2.1]heptane was synthesized from norcamphor in 8 steps. It was shown to be a very potent thromboxane antagonist by inhibition of platelet aggregation induced by U46,619 at nanomolar concentration. The key intermediate 3-[2-bromomethylphenyl]propyl tetrahydropyran ether may be useful for the synthesis of other interphenylene containing prostaglandin analogs.  相似文献

MD simulations using a combination of AMI and AMBER are performed to elucidate the origin of complexation between fullerenes and calix[n]arenes (n = 4,6,8). Only calix[6]arenes and calix[8]arenes are suitable for complex formation. By introducing benzyl substitutents on the upper rim of the calixarenes the interaction energy between host and guest is even improved. Analysis of the data also reveals a strong stabilization by t-butyl or benzyl substitutents beside π-π interactions between the arene units of the calixarene and fullerenes. The calculated data perfectly correlate with the experimentally observed equilibrium constants.  相似文献

Novel N-[[dialkylamino)ethoxy]benzyl]benzamide derivatives (II-1-51), derived from the structural modification of metoclopramide (I), were synthesized and examined for their pharmacological activities. Among them, N-[4-[2-(dimethylamino)ethoxy]benzyl]-3,4-dimethoxybenzamide (II-34) which exhibited well balanced gastrointestinal prokinetic and antiemetic activities was selected as a new type of gastrointestinal prokinetic agent.  相似文献
