The novel unsymmetrical 3,5-dialkoxycarbonyl-2,6-dimethyl-4-(7′,8′-dimethoxycoumarin-4′-yl)-1,4-dihydropyridines and 5-acetyl-3-alkoxycarbonyl-2,6-dimethyl-4-(7′,8′-dimethoxycoumarin-4′-yl)-1,4-dihydropyridines (coumarinyl-1,4-dihydropyridines) have been synthesized by Knoevenagel condensation of 4-formyl-7,8-dimethoxycoumarin with alkyl acetoacetates in the presence of AlCl3 followed by cyclization of the resulted Knoevenagel product with other alkyl acetoacetate or acetyl acetone and ammonium acetate. The structure of the intermediate Knoevenagel product and the cyclized unsymmetrical coumarinyl-1,4-dihydropyridines has been established on the basis of their spectral data analysis and single-crystal X-ray diffraction analysis. The observed conformation of the coumarinyl-1,4-dihydropyridines holds the key to promising calcium antagonistic activity of the synthesized coumarinyl-1,4-dihydropyridines.  相似文献

New fulleropyrrolidines endowed with chlorine-containing biological active 1,4-dihydropyridines (1,4-DHPs) have been synthesised from the respective formyl substituted 1,4-DHPs by following Prato's procedure. The presence of the chlorine atom on C2 of the 1,4-DHP ring brings about important spectroscopical and structural differences in compounds 10a-f related to the parent hydrogen-containing 11a. The mass spectroscopy study reveals different fragmentation patterns for fulleropyrrolidines 10a-f and their precursors 1,4-DHPs, as well as with 11a. Semiempirical calculations (AM1 and PM3) predict a most stable stereoisomer in all cases (RS for 10a-f) and the same RR for 11a. The presence of chlorine atom in 10a-f is responsible for the higher calculated conformational energy barriers in comparison with 11a. The geometry of the 1,4-DHP shows that the presence of fullerene unit does not significantly alter the required conformation for biological activity.  相似文献

A microwave-assisted sol-gel method was employed for the preparetion of nano-sized MgO particles using Mg(NO 3)2·6H2O as precursor and deionized water as solvent.The sample calcined at 500℃ had a high specific surface area of 243.2m2/g and particles sizes from 9.5to10.5nm.For comparison,MgO nanoparticles were also synthesized without microwave irradiation.X-ray diffraction (XRD) characterization showed the formation of smaller particles after microwave irradiation.The structure and morphology of the MgO particles were analyzed by N2 adsorption-desorption,XRD,scanning electron microscopy,and transmission electron microscopy.Their catalytic behavior was studied with the one-pot synthesis of Hantzsch1,4-dihydropyridines from the reaction of aromatic aldehydes,ethyl acetoacetate,and ammonium acetate.The MgO nanoparticles have high catalytic activity and gave the desired products in good to high yields.The catalyst can be easily recovered by filtration and was used at least three times with only a slight reduction in its catalytic activity.  相似文献

It has been shown that chemical oxidation of the methyl ester of 3,4,5-trimethoxycarbonyl-1,2,6-trimethyl-1,4-dihydropyridine to the pyridinium salt, requiring forcing experimental conditions, may be replaced by electrochemical oxidation. In this paper, detailed comparisons of the driving force in thermodynamics and intrinsic force in the kinetics of 1,2-dihydropyridine and 1,4-dihydropyridine isomers of PNAH, HEH, and PYH in hydride transfer reactions are made. For 1,2-PNAH and 1,4-PNAH, the values of the thermodynamic driving forces, kinetic intrinsic barriers, and thermo-kinetic parameters are 60.50 and 61.90 kcal/mol, 27.92 and 26.34 kcal/mol, and 44.21 and 44.12 kcal/mol, respectively. For 1,2-HEH and 1,4-HEH, the values of the thermodynamic driving forces, kinetic intrinsic barriers, and thermo-kinetic parameters are 63.40 and 65.00 kcal/mol, 31.68 and 34.96 kcal/mol, and 47.54 and 49.98 kcal/mol, respectively. For 1,2-PYH and 1,4-PYH, the order of thermodynamic driving forces, kinetic intrinsic barriers, and thermo-kinetic parameters are 69.90 and 72.60 kcal/mol, 33.06 and 25.74 kcal/mol, and 51.48 and 49.17 kcal/mol, respectively. It is not difficult to find that thermodynamically favorable structures are not necessarily kinetically favorable. In addition, according to the analysis of thermo-kinetic parameters, 1,4-PNAH, 1,2-HEH, and 1,4-PYH have a strong hydride-donating ability in actual chemical reactions.  相似文献

The photochemistry of some Hantzsch 4-phenyl-1,4-dihydropyridines bearing a substituent on the phenyl ring (the three isomeric chloro derivatives and the 4′-nitro derivative) has been studied. All of these compounds underwent inefficient aromatization to the corresponding pyridines (quantum yield <10−4 at 366 nm, <10−2 at 254 nm). This process is scarcely affected by molecular oxygen and is initiated by proton transfer (from C4-H), probably to the solvent, from the excited singlet. In turn, the thus formed pyridines were photoreactive with comparable or higher efficiency. Thus, the 4-(3′-chlorophenyl) and 4-(4′-chlorophenyl) Hantzsch pyridines underwent positional rearrangement to form two isomers each. The reaction occurs via Dewar benzene--prismane path. In the case of the minor isomer a further 1,3-shift take place at the Dewar benzene level. The 4-(2′-chlorophenyl) derivative underwent C-Cl bond homolysis, which led to cyclization of the phenyl group onto one of the ester groups forming a pyrane ring.  相似文献

The conformational mobility of the 1,4-dihydropyridine cycle has been studied by the molecular mechanics method. It was shown that the N(1) and C(4) atoms are displaced asymmetrically from the plane of the double bonds. The asymmetry is the result of conjugation between the nitrogen lone pair and the systems of the double bonds. The Pd-catalyzed functionalization of lactam-derived vinyl phosphates has become an important tool in the last decade for the synthesis of nitrogen-containing heterocycles. An efficient, clean, and environmental friendly ‘one-pot’ four-component reaction was developed for assembling a novel type of spirooxindole derivatives containing dihydropyridine and pyrrolidinone. The reaction could be easily performed in aqueous ethanol with 4-DMAP as catalyst in good yields. The high efficiency and the simple and mild condition of the reaction, in addition with its avoidance of time-consuming, costly syntheses, and tedious workup, endowed the reaction with application significance. A series of spirooxindole derivatives were therefore prepared in excellent yields by using this ‘one-pot’ four-component reaction method.  相似文献

Summary. In continuation of previous work some novel 3,5-diacetyl-1,4-dihydropyridine derivatives were synthesized and their photochemical behavior was studied under oxygen and argon atmosphere. Oxidation of the dihydropyridine ring and formation of pyridine derivatives was the result of the reaction. The presence of oxygen affects not only on the rate of oxidation, but also the formation of some unidentified by-products was observed on irradiation under this atmosphere.  相似文献
