Intramolecular 1,3-dipolar cycloaddition between an alkyne and an azide leads to a series of 1,2,3-triazolo-fused 1,4-benzodiazepines, 1,2,5-benzothiadiazepines, pyrrolobenzodiazepines and pyrrolobenzothiadiazepines (eight examples). The products are privileged structures in medicinal chemistry. The precursor azido alkynes are obtained, usually as transient intermediates, by treatment of the corresponding aldehydes (derived from α-amino acids) with the Bestmann-Ohira reagent.  相似文献

Alkyne homocoupling is commonly observed in cross coupling reactions; however, self trimerization and homotetramerization of alkynes to form branched products through cross-coupling reactions are rarely reported. We describe herein homotrimerization and homotetramerization of terminal alkynes under the Sonogashira reaction conditions that gave the corresponding modified enediynes. A study of diastereoselectivity in the cycloaddition reactions of a series of mono- and disubstituted alkenes with two chiral, internally H-bonded methylenenitrones has been carried out. The high degree of stereochemical control in the presence of anhydrous magnesium bromide has been explained in terms of a metal chelated transition state. Intramolecular cycloaddition involving a methylenenitrone containing an alkene moiety linked to a nitrogen gave a stereoselective addition product.  相似文献

The molecular mechanism for the 1,3-dipolar cycloaddition of nitrone with sulfonylethene chlorides has been studied using ab initio and DFT methods at the HF, MP2 and B3LYP levels together with the 6-31G* basis set. Relative rates, stereo and regioselectivity, have been analysed and discussed. For this cycloaddition four reactive channels associated with the formation of two pairs of diastereoisomeric regioisomers have been characterized. Analysis of the geometries of the corresponding transition structures shows that the cycloaddition takes place along a concerted but asynchronous mechanism. Activation energies as asynchronicity are dependent on the computation level. Thus, while HF calculations gave large barriers, MP2 calculations tend to underestimate them. DFT calculations gave reasonable values. These 1,3-dipolar cycloadditions present an endo stereoselectivity while the meta regioselectivity depends on the computational level. Thus, while HF and DFT calculations predict meta path, in agreement with the experimental results, MP2 calculation predict ortho regioselectivity. The frontier molecular orbitals analysis shows that the reaction is controlled by the (HOMOdipole–LUMOdipolarophile) interaction in agreement with the charge transfer analysis carried out at the transition structures. Inclusion of diffuse functions at the B3LYP/6-31+G* level increases the energy barriers about 4 kcal/mol, giving a similar endo/meta selectivity. Solvent effects have been taken into account, by means of self-consistent reaction field.  相似文献

The spiroisoxazolidinyl-benzoisothiazoline derivatives were synthesized through a highly diastereoselective 1,3-dipolar cycloaddition of benzoisothiazole-2,2-dioxide-3-ylidenes with nitrones. The regiochemistry of cycloaddition was assigned on the 1H NMR chemical shift of methane-H in 3 and 5. The relative stereochemistry of cycloadducts was determined from the single-crystal X-ray crystallography.  相似文献

Summary Nitrile oxides add regioselectively to the carbon-carbon double bond of 3,3-methylene-5,5-dimethyl-2-pyrrolidinone (1), giving exclusively spiro-isoxazolines: 3-aryl-6-oxo-8,8-dimethyl-1-oxa-2,7-diazaspiro[4.4]non-2-enes3 a–l. A variety of 4′-(4-R-phenyl)-4-(methylthio)-2′-phenyl-2′H-spiro[chromene-2,5′-[1,2,3]thiadiazole] 5ad and 5′-(4-R-phenyl)-4-(methylthio)-3′-phenyl-3′H-spiro[chromene-2,2′-[1,3,4]thiadiazole] 6ad were synthesized regioselectively through the reaction of 4-(methylthio)-2H-chromene-2-thione 2 with diarylnitrilimines under refluxing dry chloroform. Whilst the reaction of 4-(allylthio)-2H-chromene-2-thione 3 with diarylnitrilimines under similar reaction conditions afforded the corresponding 4-(allylthio)-5′-(4-R-phenyl)-3′-phenyl-3′H-spiro[chromene-2,2′-[1,3,4]thiadiazole] 7ad and 5-(4-R-phenyl)-2′-methyl-3-phenyl-2′,3′-dihydro-3H-spiro[[1,3,4]thiadiazole-2,4′-thieno[3,2-c]chromene] 9ad as two unisolable diastereoisomeric forms. The structures of the obtained spiro cycloadduct thiadiazoles have been assigned by means of spectroscopic measurements.  相似文献

In this paper we report the first synthesis of isoxazolidine and isoxazoline-POSS macromers by 1,3-dipolar cycloaddition reactions of vinyl- and styryl-POSS with N-methyl-C-ethoxycarbonylnitrone and ethoxycarbonyl nitrile oxide, promoted by microwave irradiation. The nature of the resulting cycloadducts has been determined by NOE experiments and supported by computational studies at PM3 and DFT B3LYP/6-31G* levels.  相似文献

2-O- and 4-O-Allyl and -propargyl glucose and the corresponding oxime derivatives were prepared from readily available glucose dithioacetals. Intramolecular 1,3-dipolar cycloaddition of the N-benzyl and N-methyl nitrones of the above acyclic 2-O-allyl glucose derivatives led to the diastereoselective formation of chiral isoxazolidines incorporating the tetrahydrofuran ring. The EI mass spectra revealed a characteristic cleavage of the C-alkyl group adjacent to the furan oxygen atom. An enantiopure trisubstituted tetrahydrofuran was obtained by the reductive cleavage of the isoxazolidine ring of one of the cycloadducts. In contrast, the nitrile oxide cycloaddition of the 2-O-allyl derivatives afforded diastereomeric mixtures of the corresponding dihydroisoxazolines, the stereochemistry of which was tentatively assigned on the basis of the principle of optical superposition. The exclusive formation of a tetrahydrofuran ring from pentaallyl nitrone or nitrile oxide demonstrated the preferred formation of a five-membered ring to that of six or seven-membered rings. The nitrile oxide generated from a 3,4,5,6,7-pentaallyloxy-1-nitroheptane derivative obtained from pentaallylglucose underwent diastereoselective cycloaddition to give an isoxazoline fused to a pyran ring. Enantiopure isoxazoles containing tetrahydrofuran and oxepane rings were also prepared in good yields by the nitrile oxide cycloaddition of the 2-O- and 4-O-propargyl derivatives.  相似文献

A new method has been demonstrated to solve the aggregation issue of metal complex catalysts; Steric protection of the metal center of catalysts was effective in the cases of DBFOX/Ph-transition metal complexes by structural modification of chiral ligand. The new ligands have been successfully applied to the nitrone cycloadditions to a variety of α,β-unsaturated aldehydes. Excellent enantioselectivities up to 99% ee have been demonstrated in the reactions at room temperature with a catalytic loading of 2 mol %.  相似文献

Stereoselective intramolecular 1,3-dipolar cycloaddition of homochiral N-(alkenylglycosyl)nitrones, prepared by allylation of C-(glycosyl)nitrones and subsequent oxidation, is described. The previously described 2-aza-Cope rearrangement was not observed for these substrates, but evidences of E/Z isomerism during the cycloaddition were obtained. The obtained cycloadducts can serve as key precursors of imino disaccharide analogues. This is exemplified by a short route to a protected 2-furanosyl-4-hydroxy-6-phenyl piperidine.  相似文献

A series of unexpected cycloadducts along with normal cycloadducts have been isolated from the 1,3-dipolar cycloaddition of 3,4-dehydromorpholine N-oxide to piperidides of cinnamic acid and para-substituted cinnamic acids and these were analyzed by X-ray crystallography to reveal novel solid-state structures. At first, 1:1 cycloadducts were formed which underwent in situ nucleophilic attack by another reduced nitrone moiety. A plausible iminium-oxonium ion mechanism has been proposed.  相似文献

Herein described was a straightforward method for the highly regioselective synthesis of 5-trifluoromethyl-1,2,3-triazole nucleoside analogues, which featured the utilization of tert-butyldimethylsilyl (TBDMS) group as the directing group in the 1,3-dipolar cycloaddition reactions. 4-tert-Butyldimethylsilyl-5-trifluoromethyl-1,2,3-triazole nucleoside analogues were generated as the only cycloaddition products in moderate yields (15-79%) via the treatment of glycosyl azides with 3,3,3-trifluoro-1-tert-butyldimethylsilylpropyne 1 in toluene at 85 °C. Removal of TBS groups in these triazole cycloadducts with tetrabutylammonium fluoride (TBAF) smoothly afforded the various 5-trifluoromethyl-1,5-disubstituted 1,2,3-triazole nucleoside analogues in good yields (40-88%).  相似文献

Stereoselective 1,3-dipolar cycloadditions of exo-methylenesugars 1a-c to a sugar nitrone 2 were carried out with refluxing a toluene solution and afforded the corresponding cycloadducts, ketosyl spiro-isoxazolidine disaccharides 3a and 4a-c. Followed by reductive cleavage of the N-O bond of the isoxazolidine ring with the treatment of Zn-AcOH-Ac2O or with catalytic hydrogenation (Pd(OH)2/C), the cycloadduct 3a could be readily converted into a novel amino-C-disaccharide possessing a ketose form, providing an access to a novel amino-C-ketosyl disaccharides.  相似文献

The discovery of a novel synthesis of new fused bicyclic isoxazoles, for example, N-methyl-3-phenyl-5,6-dihydro-4H-isoxazolo[3,4-c]azepin-8-amine (2a), N-methyl-3-phenyl-4,5-dihydroisoxazolo[3,4-c]pyridin-7-amine (2b) and N-methyl-3-phenyl-4H-pyrrolo[3,4-c]isoxazol-6-amine (2d) in high yield is reported. 1,3-Dipolar cycloaddition of in situ generated C,N-diaryl nitrilimines to 2-methyl-2-vinyl oxirane gives rise to the regioselective formation of novel 5-(2-methyloxiranyl)-4,5-dihydropyrazole derivatives in moderate to good yields. Stereoselective synthesis of bicyclic pyrrolidine and pyrrolizidine grafted macrocycles has been accomplished through intramolecular 1,3-dipolar cycloaddition (1,3-DC) reaction. Alkenyl aldehydes 4ab derived from salicylaldehyde have been reacted with secondary aminoacids to generate azomethine ylides which in turn were trapped intramolecularly by suitably placed dipolarophiles to give the corresponding macrocycle in moderate yield. The structures were assigned by 2D NMR spectra and the regio- and stereochemical outcome of the cycloadducts was established by a single crystal X-ray analysis of one of the products.  相似文献
