Carbon-11 labeled isocyanates are efficiently prepared by dehydration of [(11) C]carbamate salts, which in turn are easily formed from cyclotron-produced [(11) C]CO(2) and amines in the presence of a CO(2) fixation agent. The [(11) C]isocyanates are useful radiosynthons for the synthesis of a variety of [carbonyl-(11) C]-labeled asymmetrical ureas and carbamate esters. The method is well suited to incorporate any isotope of carbon, and is especially useful for positron emission tomography (PET) radiotracers for in vivo imaging. This is demonstrated by using the method to make [carbonyl-(11) C]-6-hydroxy-[1,1'-biphenyl]-3-yl cyclohexylcarbamate which is a novel radiotracer for PET imaging of fatty acid amide hydrolase.  相似文献

Using either amines, amino alcohols, or alcohols in selenium-mediated synthesis with [(11)C]carbon monoxide, 3 ureas, 6 carbamates, and 1 carbonate were labeled. Tetrabutylammonium fluoride ((TBA)F) was discovered to form a soluble and reactive complex with selenium and drastically increase the radiochemical yields. Of the selected carbamoyl compounds, one was a receptor ligand, one was an enzyme inhibitor, and one was a muscular relaxant pharmaceutical. The (11)C-target compounds were obtained in radiochemical yields ranging from low to almost quantitative and with specific radioactivity up to 1300 GBq/micromol. The radiochemical purity of the final products exceeded 98%. In one case, the corresponding (13)C-substituted compound was produced to verify the position of the (11)C-label. In a typical experiment starting with 16.4 GBq [(11)C]carbon monoxide, 7.0 GBq of LC-purified 5-phenyl-1,3-oxazolidin-[2-(11)C]-2-one was obtained within 20 min from start of the carbonylation reaction (84% decay-corrected radiochemical yield). The presented approach is an interesting alternative to the use of [(11)C]phosgene in labeling chemistry.  相似文献

Novel radiolabeled O(6)-benzylguanine (O(6)-BG) derivatives, 6-O-[(11)C]-[(methoxymethyl)benzyl]guanines ([(11)C]p-O(6)-MMBG, 1a; [(11)C]m-O(6)-MMBG, 1b; [(11)C]o-O(6)-MMBG, 1c), 2-amino-6-O-[(11)C]-[(methoxymethyl)benzyloxy]-9-methyl purines ([(11)C]p-O(6)-AMMP, 2a; [(11)C]m-O(6)-AMMP, 2b; [(11)C]o-O(6)-AMMP, 2c), 2-amino-6-O-[(11)C]-[(methoxymethyl)benzyloxy]-9-benzyl purines ([(11)C]p-O(6)-AMBP, 3a; [(11)C]m-O(6)-AMBP, 3b; [(11)C]o-O(6)-AMBP, 3c), 2-amino-6-O-benzyloxy-9-[(11)C]-[(methoxycarbonyl)methyl]purine ([(11)C]ABMMP, 4), and 2-amino-6-O-benzyloxy-9-[(11)C]-[(4'-methoxycarbonyl)benzyl]purine ([(11)C]ABMBP, 5), have been synthesized for evaluation as potential novel positron emission tomography tumor imaging agents. The ability of O(6)-BG analogs to penetrate the blood--brain barrier in brain tumor could be due, at least in part, to their lipophilicity. In this paper, we measured lipophilicity coefficients (log P) of compounds 1--5 by the C(18) HPLC method. These log P values were compared, and correlations between lipophilicity and biological activity of selected analogs were made. The results suggest the appropriate level of lipophilic character in this class of compounds as useful imaging agents appears to be in the range of 1.4--2.6.  相似文献

New reactive polymers with pendant halomethyl groups were successfully synthesized by polyaddition reactions of bis(epoxide)s with bis(chloroacetoxy)ester such as 1,4-bis [(chloroacetoxy)methyl]benzene (BCAMB) or 1,4-bis[(bromoacetoxy)methyl]benzene (BBAMB) using quaternary onium salts or crown ether complexes as catalysts. The polyaddition reaction of diglycidyl ether of bisphenol A (DGEBA) with BCAMB proceeded very smoothly with high yields (83–96%) by the addition of quaternary onium salts such as tetrabutylphosphonium bromide (TBPB) or crown ether complexes such as 18-crown-6/KBr as catalysts to produce high molecular weight polymers, although the reaction occurred without any catalyst to give low molecular weight polymer in low yield at 90°C for 48 h. It was also found that the reaction proceeded smoothly in aprotic polar solvents such as N-methyl-2-pyrrolidone (NMP) and N,N-dimethylacetamide (DMAc) to produce high molecular weight polymers. Polyaddition reactions of DGEBA or digylcidyl ether of ethylene glycol (DGEEG) with BBAMB, other bis(chloroacetoxy)esters or bis(bromoacetoxy)esters using TBPB in DMAc also proceeded smoothly to give the corresponding polymers. The resulting poly(ether-ester)s contain reactive halomethyl groups as side chains, which were introduced during main chain formation. © 1997 John Wiley & Sons, Inc. Photoinitiated radical carbonylation with [(11)C]carbon monoxide at low concentration was employed in syntheses of carbonyl-(11)C-labeled amides using alkyl iodides and amines as precursors. Eleven (11)C-amides were synthesized in up to 74% decay-corrected radiochemical yields with reaction times of 400 s and with up to 95% conversion of carbon monoxide. Starting with 26.3 GBq of [(11)C]carbon monoxide, 10.6 GBq of 1-cyclohexane [(11)C]carbonyl-4-phenyl-piperazine (15) was obtained within 35 min from the end of bombardment (33 microA) and with a specific radioactivity of 192 GBq/micromol at the same time point. The influence of solvents was investigated. The described procedure extends the range of accessible labeling methods. The method may also be useful for preparation of (13)C- and (14)C-substituted compounds.  相似文献

A general methodology for the rapid purification of carbon-11 positron emission tomography (PET) radiotracers from radiolabeling reaction mixtures has been developed. Preparative HPLC and solid-phase extraction (SPE) techniques are described which can separate some commonly used radiopharmaceuticals such as [(11)C]raclopride, [(11)C]beta-CFT and [(11)C]choline from their unlabeled precursors.  相似文献

Reaction of [Cp*TaCl4] (Cp*=eta5-C5Me5) with a sixfold excess of LiBH(4)thf followed by BH3thf in toluene at 100 degrees C led to the isolation of hydrogen-rich metallaboranes [(Cp*Ta)2B4H10] (1), [(Cp*Ta)2B5H11] (2), [(Cp*Ta)2B5H10(C6H4CH3)] (3), and [(Cp*TaCl)2B5H11] (4) in modest yield. Compounds 1-3 are air- and moisture-sensitive but 4 is reasonably stable in air. Their structures are predicted by the electron-counting rules to be a bicapped tetrahedron (1), bicapped trigonal bipyramids (2, 3), and a nido structure based on a closo dodecahedron 4. Yellow tantalaborane 1 has a nido geometry with C2v symmetry and is isostructural with [(Cp*M)2B4H8] (M=Cr and Re); whereas 2 and 3 are C3v-symmetric and isostructural with [(Cp*M)2B5H9] (M=Cr, Mo, W) and [(Cp*ReH)2B5Cl5]. The most remarkable feature of 4 is the presence of a hydride ligand bridging the ditantalum center to form a symmetrical tantalaborane cluster with a long Ta--Ta bond (3.22 A). Cluster 4 is a rare example of electronically unsaturated metallaborane containing four TaHB bonds. All these new metallaboranes have been characterized by mass spectrometry, 1H, 11B, and 13C NMR spectroscopy, and elemental analysis, and the structural types were unequivocally established by crystallographic analysis of 1-4.  相似文献

Palladium(0)-mediated carbonylation reactions using aryl triflates, amines, and a low concentration of [(11)C]carbon monoxide were used in the syntheses of 13 (11)C-labeled amides. Lithium bromide was used as an additive to facilitate the reaction. The (11)C-labeled products were obtained with decay-corrected radiochemical yields in the range of 2-63%. The radiochemical purity of the final products exceeded 98%. As an example, a reaction starting with 1.79 GBq [(11)C]carbon monoxide gave 0.38 GBq of LC-purified N-isopropyl-4-nitro-[(11)C]benzamide within 27 min from the start of the carbonylation reaction (54% decay-corrected radiochemical yield). The specific radioactivity of this compound was 191 GBq/micromol, 35 min after the end of a 10 microAh bombardment. N-Benzylisoquinoline-1-((13)C)carboxamide was prepared and analyzed by NMR for confirmation of the labeling position. The triflates 16, 20, 21, and 22 were synthesized from the corresponding alcohols and trifluoromethanesulfonic anhydride. The reference compounds 30a and 30b were prepared from the corresponding carboxylic acids and benzylamine. The other nine reference compounds 32a to 32i were synthesized from the respective acid chlorides and amines. The presented report shows that the sometimes more easily obtainable aryl triflates can be a useful alternative to the commonly used aryl halides in palladium(0)-mediated synthesis of (11)C/(13)C-amides.  相似文献

By judicious selection of crystallization conditions, it has been possible to obtain the salts of a common building block, [(RNC)4Rh(I)]+, in single-crystal form suitable for X-ray diffraction. Salts that contain a single type of cation include deep green [(C6H11NC)12Rh(I)3](SbF6)3, deep green [(C6H11NC)12Rh(I)3](AsF6)3, and straw yellow [(C6H11NC)8Rh(II)2Cl2](BF4)2 (in addition to the previously isolated trimeric deep green [(i-PrNC)12RhI3]Cl3 x 4.5 H2O, monomeric, [(C6H11NC)4 Rh(I)](BPh4), and [(i-PrNC)4Rh(I)](BPh4) (both yellow), and red, dimeric [(C6H11NC)8Rh(I)2]Cl2 x 0.5C6H6 x 2H2O). Ordered crystals of [(C6H11NC)12Rh(I)3](SbF6)3 contain linear Rh3 units, while those of [(C6H11NC)12Rh(I)3](AsF6)3 show disorder which is consistent with the presence of linear or bent Rh3 units. The formation of green [(C6H11NC)12Rh(V/III)3Cl2][(C6H11NC)12Rh(I)3]Cl6, and brown [(C6H11NC)12Rh(V/III)3Cl2][(C6H11NC)8Rh(I)2][(C6H11NC)4RhI]Cl6 x 16H2O x 3C6H6 along with unidentified red-brown cubes from an air-exposed solution of [(C6H11NC)4Rh(I)]Cl is reported. As their formulas indicate, green [(C6H11NC)12Rh(V/III)3Cl2][(C6H11NC)12Rh(I)3]Cl6, and brown [(C6H11NC)12Rh(V/III)3Cl2][(C6H11NC)8Rh(I)2][(C6H11NC)8Rh(I)]Cl6 x 16H2O x 3C6H6 contain two or three chemically distinct cations, respectively, but again are built from a common precursor, [(C6H11NC)4Rh(I)]+.  相似文献

Microfluidic technology has been used to perform [(11)C]carbonylation reactions using solutions containing [(11)C]CO in the form of the complex, copper(i)tris(3,5-dimethylpyrazolyl)borate-[(11)C]carbonyl (Cu(Tp*)[(11)C]CO). The synthesis of the model compound [(11)C]N-benzylbenzamide and the known tracer molecule [(11)C]trans-N-[5-(2-flurophenyl)-2-pyrimidinyl]-3-oxospiro[5-azaisobenzofurane-1(3H),1'-cyclohexane]-4'-carboxamide ([(11)C]MK-0233), a ligand for the neuropeptide Y Y5 receptor, have been performed using this technique. Following semi-preparative HPLC purification and reformulation, 1262 ± 113 MBq of [(11)C]MK-0233 was produced at the end of the synthesis with a specific activity of 100 ± 30 GBq μmol(-1) and a >99% radiochemical purity. This corresponds to a decay corrected radiochemical yield of 7.2 ± 0.7%. Using a 3 mL vial as the reaction vessel, and following semi-preparative HPLC purification and reformulation, 1255 ± 392 MBq of [(11)C]MK-0233 was produced at the end of the synthesis with a specific activity of 100 ± 15 GBq μmol(-1) and a >99% radiochemical purity. This corresponds to a decay corrected radiochemical yield of 7.1 ± 2.2%.  相似文献

Reactions of lithium salts of the bulky guanidinate ligands, [ArNC(NR2)NAr](-) (NR2 = N(C6H11)2 (Giso-) and cis-NC5H8Me2-2,6 (Pipiso-); Ar = C6H3Pri2-2,6), with GeCl2.dioxane afforded the heteroleptic germylenes, [(Giso)GeCl] and [(Pipiso)GeCl], the former of which was structurally characterised. The further reactivity of these and the related complexes, [(Piso)GeCl] and [(Priso)GeCl] (Piso- = [ArNC(Bu(t))NAr]-, Priso- = [ArNC(NPri2)NAr]-) has been investigated. Salt elimination reactions have yielded the new monomeric complexes, [(Piso)Ge(NPri2)] and [(Piso)GeFeCp(CO)2], whilst a ligand displacement reaction afforded the heterometallic species, [(Piso)Ge(Cl)(W(CO)5)]. Chloride abstraction from [(Priso)GeCl] with GaCl3 has given the structurally characterised contact ion pair, [(Priso)Ge][GaCl4]. In addition, the inconclusive outcome of a number of attempts to reduce the germanium halide complexes are discussed.  相似文献

Ointment-like poly(ortho esters) were synthesized for the first time from the reaction of 3,9-bis(methylene)-2,4,8,10-tetraoxaspiro[5. 5] undecane with poly(ethylene glycol)-400, N,N-bis(2-hydroxyethyl)-n-hexadecylamine and N,N-bis(2-hydroxyethyl) palmitamide, respectively. The obtained polymers were characterized by 1H NMR spectra, 13C NMR spectra, elemental analyses, light scattering, and measurements of intrinsic viscosity. The influence of catalyst on the intrinsic viscosity of polymers was investigated. The 9-[(1,3-dihydroxy-2-propoxy) methyl] guanine controlled release profiles of hydrophobic ointment-like polymers such as polymer PII in vitro were also discussed.  相似文献

The reactions of Na[C(5)(CN)(5)] (Na[1]) with group 11 phosphine complexes [(P)(n)MCl] (M = Cu, Ag, Au, P = Ph(3)P; M = Cu, P = dppe (Ph(2)PCH(2)CH(2)PPh(2))] give a range of compounds containing the pentacyanocyclopentadienide ligand, [C(5)(CN)(5)](-) (1). The new complexes [(Ph(3)P)(2)M{1}](2) [M = Cu (3); M = Ag (5)], [(Ph(3)P)(3)Ag{1}] (4), [(dppe)(3)Cu(2){1}(2)] (6) and [Au(PPh(3))(2)][1] (7) include the first complete series of group 11 complexes of any cyclopentadienide ligand to be structurally characterised.  相似文献

The remarkable, vapor-induced transformation of the yellow polymorphs of [(C(6)H(11)NC)(2)Au(I)](AsF(6)) and [(C(6)H(11)NC)(2)Au(I)](PF(6)) into the colorless forms are reported along with related studies of the crystallization of these polymorphs. Although the interconversion of these polymorphs is produced by vapor exposure, molecules of the vapor are not incorporated into the crystals. Thus, our observations may have broad implications regarding the formation and persistence of other crystal polymorphs where issues of stability and reproducibility of formation exist. Crystallographic studies show that the colorless polymorphs, which display blue luminescence, are isostructural and consist of linear chains of gold(I) cations that self-associate through aurophilic interactions. Significantly, the yellow polymorph of [(C(6)H(11)NC)(2)Au(I)](AsF(6)) is not isostructural with the yellow polymorph of [(C(6)H(11)NC)(2)Au(I)](PF(6)). Both yellow polymorphs exhibit green emission and have the gold cations arranged into somewhat bent chains with significantly closer Au···Au separations than are seen in the colorless counterparts. Luminescence differences in these polymorphs clearly enhance the ability to detect and monitor their phase stability.  相似文献

New ester salts [R3NH]+[F5SC(SO2F)C(O)OR′] where RH, CH3CH2 and R′CH3,(CH3)2CH have been prepared from corresponding esters and amines. The sodiumsalt Na[F5SC(SO2F)C(O)OCH(CH3)2] was used to prepare the following -substitutedderivatives: SF5CX(SO2F)C(O)OCH(CH3)2, XBr, Cl. The crystal structure of[(C2H5)3NH]+[F5SC(SO2F)C(O)OCH3] was determined and is monoclinic: P21/n;a=8.758(2) Å, b=9.645(2) Å and c=19.167(4) Å; β=97.92(3)°; V=1603.6 Å3; Z=4.  相似文献

Lithium and potassium trifluoromethyl-, pentafluoroethyl- and [(diethylphosphinyl)difluoromethyl]trialkoxyborates were synthesized by reaction of either perfluoroalkyllithium or (perfluoroalkyl)trimethylsilane/F with trialkoxyboranes. Treatment of perfluoroalkyltrialkoxyborates with methanesulfonyl chloride, methyl triflate or methyl tosylate furnished the hitherto unknown trifluoromethyl-, pentafluoroethyl- and [(diethylphosphinyl)difluoromethyl]boronic esters.  相似文献

Addition of the carbene 1,3-dimesitylimidazol-2-ylidene (IMes) to a toluene solution of Ag[closo-CB(11)H(12)] results in the formation of the complex [(IMes)(2)Ag](2)[Ag(2)[closo-CB(11)H(12)](4)], the anionic component of which contains two silver(I) centers bridged by two carboranes in addition to one terminally bound carborane on each metal, in the solid-state. Comparison of the observed (11)B[(1)H] NMR chemical shifts of [(IMes)(2)Ag](2)[Ag(2)[closo-CB(11)H(12)](4)] or Ag[closo-CB(11)H(12)] with [NBu(4)][closo-CB(11)H(12)] in CD(2)Cl(2) demonstrates that the silver ion interacts significantly with the cage in solution. Theoretical investigations using the ab initio/GIAO/NMR method of [closo-CB(11)H(12)](-) and Na[closo-CB(11)H(12)] as model geometries for the silver salts support experimental evidence for these Ag...[BH] interactions in solution.  相似文献

A general synthesis of 1,2,3-thiadiazole-4-thiolates 1 and their derivatives 2–3 by an extension of the Hurd-Mori 1,2,3-thiadiazole synthesis is described. Treatment of methyl (or ethyl) [1-(alkylthio)alkylidene]hydrazinocarboxylates 11 (thiocarbazonate esters) or other N-acylthiohydrazonate esters [Y = ureido ( 12 ) or arenesulfonyl ( 13 )] with thionyl chloride affords 2–3 efficiently. Intermediates 11–13 are readily obtained from the N2-thioacylcarbazates 8 , N3-thioacylsemicarbazides 9 , or N2-thioacyl-N1-(p-toluenesulfonyl)hydrazides 10 , respectively, by S-alkylation. Physicochemical properties of the 1,2,3-thiadiazoles 1–3 and N-acylthiohydrazonate esters 11–13 are also described.  相似文献

Sterically demanding triazapentadiene [N[(C3F7)C(Dipp)N]2] H has been synthesized in good yield. It features a W-shaped ligand backbone in the solid state. [N[(C3F7)C(Dipp)N]2]H reacts with copper(I) oxide in acetonitrile leading to [N[(C3F7)C(Dipp)N]2]CuNCCH3. This copper adduct serves as an excellent precursor to obtain thermally stable [N[(C3F7)C(Dipp)N]2]CuCNBut and [N[(C3F7)C(Dipp)N]2]CuCO. IR spectroscopic data of these copper(I) isocyanide (CN = 2176 cm(-1)) and copper(I) carbonyl (CO = 2109 cm(-1)) complexes indicate that the [N[(C3F7)C(Dipp)N]2]- ligand is a fairly weak donor.  相似文献
