The effect of different abiotic elicitors [jasmonic acid, copper sulphate (CuSO4) and salicyclic acid] at varying concentrations on the stimulation of biomass and bacoside production in in vitro Bacopa monnieri shoot culture was studied. A systematic study conducted over a period of 35 days indicated that the maximum bacoside production (6.74 mg g? 1 dry weight (DW)) was obtained after a lag of 7 days and thereafter, the content decreased gradually to again increase at 28 days (5.91 mg g? 1 DW). Therefore, elicitation experiments were carried out over a period of 3, 6 and 9 days. The shoot cultures treated with 45 mg L? 1 of CuSO4 exhibited the highest bacoside content of 8.73 mg g? 1 DW (~1.42-fold higher) than in control cultures (6.14 mg g? 1 DW). This study indicates the effectiveness of abiotic elicitation on bacoside production in in vitro shoot cultures of this medicinally important herb known for its memory-enhancing properties.  相似文献

Hypericum perforatum L. (St. John’s Wort) is an important medicinal plant which is widely used in the treatment for depression and irritable bowel syndrome. It is also used as a dietary supplement. Major bioactive phytochemicals of H. perforatum are phenolics and flavonoids. Quality of these phytochemicals is dramatically influenced by environmental and biological factors in the field grown plants. As an alternative, we have developed adventitious root cultures in large-scale bioreactors for the production of useful phytochemicals. Adventitious roots of H. perforatum were cultured in 500 l pilot-scale airlift bioreactors using half-strength Murashige and Skoog medium with an ammonium and nitrate ratio of 5:25 mM and supplemented with 1.0 mg l?1 indole butyric acid, 0.1 mg l?1 kinetin, and 3 % sucrose for the production of bioactive phenolics and flavonoids. Then 4.6 and 6.3 kg dry biomass were realized in the 500 l each of drum-type and balloon-type bioreactors, respectively. Accumulation of 66.9 mg g?1 DW of total phenolics, 48.6 mg g?1 DW of total flavonoids, 1.3 mg g?1 DW of chlorogenic acid, 0.01 mg g?1 DW of hyperin, 0.04 mg g?1 DW of hypericin, and 0.01 mg g?1 DW of quercetin could be achieved with adventitious roots cultured in 500 l balloon-type airlift bioreactors. Our findings demonstrate the possibilities of using H. perforatum adventitious root cultures for the production of useful phytochemicals to meet the demand of pharmaceutical and food industry.  相似文献

Withanolides, the secondary metabolite from Withania species is used in Ayurvedic medicine and now proved to have potential use in treating cardiovascular, Alzheimer’s disease, etc. Its production in plants varies between genotypes with very low yield. For improved industrial commercialisation, there is a need to increase its production. Endophytic fungi are symbiotically associated with plants and can synthesise the same bioactive compounds and natural products as their host plant. There are no reports available on withanolide-producing (endophytic) fungi. The present study identified an endophytic fungus (Taleromyces pinophilus) from leaves of Withania somnifera which produces withanolides in the medium. The structure of withanolide was confirmed by 1H NMR, LC-MS analyses and quantified by HPLC analysis. The fungus produces high amount of withanolide when compared to leaf and root of W. somnifera. The fungus can be exploited to produce the withanolide to meet its demand.  相似文献

Four novel withanolide glycosides and a withanolide have been isolated from the leaves of Withania somnifera. The structures of the novel compounds were elucidated as physagulin D (1→6)-β-d-glucopyranosyl-(1→4)-β-d-glucopyranoside (1), 27-O-β-d-glucopyranosyl physagulin D (2), 27-O-β-d-glucopyranosyl viscosalactone B (3), 4,16-dihydroxy-5β, 6β-epoxyphysagulin D (4), and 4-(1-hydroxy-2,2-dimethylcyclo-propanone)-2,3-dihydrowithaferin A (5) on the basis of 1D-, 2D NMR and MS spectral data. In addition, seven known withanolides withaferin A (6), 2,3-dihydrowithaferin A (7), viscosalactone B (8), 27-desoxy-24,25-dihydrowithaferin A (9), sitoindoside IX (10), physagulin D (11), and withanoside IV (12) were isolated. These withanolides were assayed to determine their ability to inhibit cycloxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) enzymes and lipid peroxidation. The withanolides tested, except compound 9, showed selective COX-2 enzyme inhibition ranging from 9 to 40% at 100 μg/ml. Compounds 4, 10 and 11 also inhibited lipid peroxidation by 40, 44 and 55%, respectively. The inhibition of COX-2 enzyme by withanolides is reported here for the first time.  相似文献

In this article our aim was to evaluate mass cultivation of S. marianum hairy roots in a bioreactor to produce silymarin. The effects of methyl jasmonate (MJ) elicitation on the accumulation of silymarin and the extent of the MJ-induced oxidative damage were investigated in bioreactor hairy root cultures of S. marianum. The growth rate of the bioreactor hairy root cultures was higher than that of those in a shake flask after 3 weeks. Silymarin accumulation was increased from 0.13?mg?g?1 DW in non-treated hairy roots to 0.22?mg?g?1 DW in hairy roots 72?h after 100?μM?MJ treatment. Guaiacol peroxidase and ascorbate peroxidase were activated by MJ?72?h after treatment, being 3.2- and 1.3-fold higher, respectively, than that of the control. An increase in enzymatic activity suggests increased scavenging of reactive oxygen species, indicating the tolerance to MJ stress. These results suggest that MJ elicitation is beneficial for silymarin production using bioreactor hairy root cultures.  相似文献

Methanolic extracts from in vitro grown Scutellaria lateriflora shoots cultured on five Murashige and Skoog (MS) medium variants supplemented with different combinations of 6-benzylaminopurine (BAP) and α-naphthaleneacetic acid (NAA) under different light conditions (monochromatic light, white light and no light) were analysed by HPLC for three groups of metabolites: flavonoids (26 compounds), phenolic acids and their precursors (19+2) and phenylethanoid glycosides (2). The analyses revealed the presence of baicalein, baicalin, wogonin, wogonoside, 3,4-dihydroxyphenylacetic acid and verbascoside. There was clear evidence of the influence of plant growth regulators and light conditions on the accumulation of the analysed groups of secondary metabolites. The amounts of the compounds changed within a wide range—for the total flavonoid content, 30.2-fold (max. 1204.3 mg·100 g?1 dry weight (DW)); for 3,4-dihydroxyphenylacetic acid, 5.5-fold (max. 33.56 mg·100 g?1 DW); and for verbascoside, 1.5-fold (169.15 max. mg·100 g?1 DW). The best medium for the production of most of the compounds was the Murashige and Skoog variant with 1 mg l?1 BAP and 1 mg l?1 NAA. For verbascoside, the best ‘productive’ medium was the MS variant supplemented with 0.5 mg l?1 BAP and 2 mg l?1 NAA. The accumulation of the metabolites was stimulated to the greatest extent by blue light, under which the extracts were found to contain the highest total amount of flavonoids and the highest amounts of flavonoid glucuronides, baicalin and wogonoside, as well as of verbascoside. A sensitive, reliable, simple and rapid thin‐layer chromatographic method has been developed for routine analysis of withanolide S content for the purpose of quality control assessment of chemotype III of Withania somnifera. The new method was used first to compare the accumulation of withanolide S in different parts of the plant, which was found to be the highest in the leaves extract (0.21% w/w). Second, to investigate different extraction parameters that improve the extraction efficiency of withanolides from the leaves using conventional and ultrasound‐assisted extraction methods. Nine new steroidal lactones of the withanolide series (withanolides E-M) have been isolated from the leaves of Withania somnifera Dun (Solanaceae) growing in the southern coastal plane of Israel. This population of W. somnifera constitutes a new chemotype and is designated as chemotype III. An efficient plant regeneration protocol was established for an endangered ethnomedicinal plant Desmodium gangeticum (Linn.) DC. Morphogenic calli were produced from 96 % of the cultures comprising the immature leaf explants on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxyacetic acid (4.0 mg?l?1) in combination with 6-benzylaminopurine (BA; 0.8 mg?l?1). For callus regeneration, various concentrations of BA (1.0–5.0 mg?l?1) or thidiazuron (TDZ; 1.0–5.0 mg?l?1) alone or in combination with indole-3-acetic acid (IAA; 0.2–1.0 mg?l?1) were used. Highest response of shoot regeneration was observed on MS medium fortified with TDZ (4.0 mg?l?1) and IAA (0.5 mg?l?1) combination. Here, 100 % cultures responded with an average number of 22.3 shoots per gram calli. Inclusion of indole-3-butyric acid in half MS medium favored rooting of recovered shoots. Out of 45 rooted plants transferred to soil, 40 survived. Total DNA was extracted from the leaves of the acclimatized plants of D. gangeticum. Analysis of random amplified polymorphic DNA using 13 arbitrary decanucleotide primers showed the genetic homogeneity in all the ten plants regenerated from callus with parental plant, suggesting that shoot regeneration from callus could be used for the true-to-type multiplication of this plant.  相似文献

Daidzin (7-O-glucoside of daidzein) has several pharmacological benefits in herbal remedy, as antioxidant and shown antidipsotropic activity. Hairy root culture of Psoralea corylifolia L. was developed for biomass and enhanced daidzin production using signalling compounds such as jasmonic acid (JA) and acetyl salicylic acid (ASA). Best response of 2.8-fold daidzin (5.09% DW) with 1 μM JA treatment after second week and 7.3-fold (3.43% DW) with 10 μM JA elicitation after 10th week was obtained from hairy roots compared to untreated control. ASA at 10 μM promoted 1.7-fold increase in daidzin (1.49% DW) content after seventh week compared to control (0.83% DW). Addition of 25 μM ASA resulted in 1.44% DW daidzin (1.5-fold increase) with 0.91% DW in control after fifth week and 1.44% DW daidzin (2.3-fold increase) after eighth week when compared to untreated control (0.62% DW). Reduced biomass with increased daidzin content was facilitated by elicited hairy root cultures.  相似文献

Adventitious root cultures of Prunella vulgaris L. were established in shaking flask system for the production of biomass and secondary metabolites. Adventitious root cultures were induced from callus cultures obtained from leaf explants on solid Murashige and Skoog (MS) medium containing combination of 6-benzyladenine (BA; 1.0 mg l?1) and naphthalene acetic acid (NAA; 1.5 mg l?1). Thereafter, 0.49 g inoculum was transferred to liquid MS medium supplemented with different concentrations of NAA (0.5–2.0 mg l?1). Growth kinetics of adventitious roots was recorded with an interval of 7 days for 49 days period. Highest biomass accumulation (2.13 g/l) was observed in liquid medium containing 1.0 mg l?1 NAA after 21 days of inoculation. However, other concentrations of NAA also showed similar accumulation pattern but the biomass gradually decreases after 49 days of inoculation. Adventitious roots were collected and dried for investigation of total phenolics (TP), total flavonoids (TF), and antioxidant activities. Higher TPC (0.995 GAE mg/g-DRB) and TFC (6.615 RE mg/g-DRB) were observed in 0.5 mg l?1 NAA treated cultures. In contrast, higher antioxidant activity (83.53 %) was observed 1.5 mg l?1 NAA treated cultures. These results are helpful in up scaling of root cultures into bioreactor for secondary metabolites production.  相似文献

The phenolic composition of Feteasc? neagr?and B?beasc? neagr? grapes from Dealul Bujorului vineyard (south-east Romania) was studied using the spectrophotometric and high-performance liquid chromatographic (HPLC) methods. The results revealed significant differences between these cultivars. Total anthocyanins ranged from 0.22–5.98 mg g?1 DW berries in B?beasc? neagr? grapes to 0.54–18.54 mg g?1 DW berries in Feteasc? neagr?. Both cultivars were characterised by an interesting anthocyanin profile for winemaking with a prevalence of malvidin-3-glucoside. The skins and seeds both had small amounts of flavonoids. In contrast with B?beasc? neagr?, Feteasc? neagr? had more flavonoids in the seeds (69 %) than in the skins. Statistically significant correlations were observed between antioxidant activity and total anthocyanin content in both varieties. The antioxidant activity was also found to be highly correlated to the total phenolic compounds content.  相似文献

In this study, seedling, callus, cell, and adventitious root of Glycyrrhiza uralensis Fisch. have been established. In order to find the best one for producing G. uralensis active constituents, triterpenoid saponins and flavonoids in native root and tissue cultures were determined, and the contents in different G. uralensis materials were analyzed using cluster analysis. The contents of triterpenoid saponins and glycyrrhizic acid in tissue cultures were much lower than that in native G. uralensis. The total flavonoids content we determined in adventitious root was 6.31 mg?g?1, which was close to that of native root (9.82 mg?g?1). Based on the cluster analysis, we found that G. uralensis cultures were not suitable for production of glycyrrhizic acid, while adventitious root had a greater capability of flavonoids production comparing to seedling, callus, and cell.  相似文献

Abstract

The presented work reports anticholinesterase and antioxidant activities of extracts, fractions from aerial parts, fruits, flowers, roots and isolated compounds of roots from Leiotulus dasyanthus (bergapten, pimpinellin, umbelliferone, quercetin, rutin and kaempferol). Phenolic contents, antioxidant activities of samples were carried out by Folin-Ciocalteu, DPPH, TBA methods. Anticholinesterase activity was evaluated by Ellman’s method. The highest and lowest total phenolic content were detected in root MeOH extract (88.6?mg GAE g?1 DW) and aerial part (51.83?mg GAE g?1 DW), respectively. The highest antioxidant activity among isolated secondary metabolites got coumarins umbelliferone, bergapten and pimpinellin. Pimpinellin (66.55%) and umbelliferone (61.09%) demonstrated strong inhibition towards acetylcholinesterase and butyrylcholinesterase, respectively. Dichloromethane fraction of root demonstrated significant inhibition against AChE (49.66%) and BuChE (92.21%) at 20 µg/mL. Dichloromethane fractions of roots had a notableness antioxidant and anticholinesterase activities. The further studies on roots will be important for development use of this plant for pharmaceutical and food research needs.

Multiple shoot cultures of two experimental lines of Withania somnifera plants (RS-Selection-1 and RS-Selection-2) were established using nodal segments as explants. The hormonal combinations of benzyl adenine and kinetin not only influenced their morphogenetic response but also differentially modulated the level of biogeneration of withanolide A in the in vitro shoots of the two lines. Interestingly, withanolide-A, that was hardly detectable in the aerial parts of field-grown Withania somnifera (explant source), accumulated considerably in the in vitro shoot cultures of the plant. The productivity of withanolide A in the cultures varied considerably (ca. 10-fold, 0.014 to 0.14 mg per gram fresh weight) with the change in the hormone composition of the culture media as well as genotype used as source of the explant. The shoot culture of RS-Selection-1 raised at 1.00 ppm of BAP and 0.50 ppm of kinetin displayed the highest concentration of withanolide A in the green shoots of 0.238 g per 100 g dry weight tissue. This was a more analytical concentration keeping in view the isolation yields so far reported from the dried roots of the field-grown plant (ca. 0.015 g per 100 g dry weight), even if isolation losses are considered during purification. The enhanced de novo biogenesis of withanolide A in shoot cultures was corroborated with radiolabel incorporation studies using [2-(14)C] acetate as a precursor. Production of withaferin A was also found in the in vitro shoot cultures. As this compound is a predominant withanolide of native shoots as well and has been already reported to be accumulated in in vitro shoot cultures, its biogeneration observed in these shoot cultures is not discussed in detail.  相似文献

High yielding transformed callus culture of W. somnifera was established by infecting hypocotyls with Agrobacterium tumefaciens MTCC-2250. Maximum withaferin A content of 0.0875 mg/g dry cell weight and transformation efficiency of 80% were obtained. Confirmation of transformation was done on the basis of the presence of the ags gene by using polymerase chain reaction. Various abiotic elicitors (arachidonic acid, methyl jasmonate, calcium chloride, and copper sulfate) and biotic elicitors (cell extracts and culture filtrates of Alternia alternata, Fusarium solani, and Verticilium dahaliae) were tested at different concentrations to enhance withaferin A production in suspension culture of transformed cells. Maximum enhancements of 5.4 times and 9.7 times, respectively, were obtained when copper sulfate (100 microM) and the cell extract of V. dahaliae (5% v/v) were added separately to suspension cultures. The dual elicitation strategy by the combined addition of these two elicitors resulted in 13.8-fold enhancement of withaferin A content in comparison to control cultures (2.65 mg/L). The present study indicates the potential of this biotechnology-based methodology for the large-scale production of withaferin A.  相似文献

Biomagnification of butyltins (BTs) was examined in a simple food web including seawater, macroalgae (Alaria esculenta, Laminaria longicruris, Ulvaria obscura) and green urchin (Strongylocentrotus droebachiensis). The study was conducted in shallow waters of the St Lawrence Estuary (Canada) adjacent to two areas potentially contaminated by BTs. Levels of tri‐ (TBT), di‐ (DBT) and mono‐BT (MBT) were determined in seawater, green urchin (including faecal matter after sampling) and macroalgae surrounding the urchins at each sampling site. The concentrations of TBT in seawater from all stations were relatively low (3–7 ngSn l?1), and both the TBT and the total BTs (∑BT = MBT + DBT + TBT) concentrations decreased with increase in distance from the BT sources. The concentrations of TBT in algae were 0.35 ngSn g?1 dry weight (DW) in A. esculenta, 0.40 ngSn g?1 DW in L. longicruris and 3.58 ngSn g?1 DW in U. obscura. Following their location, green urchins feeding mainly on these algae accumulated BTs at levels ranging from 4 to 85 ngSn g?1 DW in gonads and from 35 to 334 ngSn g?1 DW in gut. The mean bioconcentration factor (BCF) calculated from seawater to algae ranged from 17 in A. esculenta to 151 in U. obscura, whereas the biomagnification factor (BMF) from algae to urchins ranged from 2 to 17 in gonads and from 10 to 67 in gut. The overall bioaccumulation factor of TBT between seawater and internal organs of urchins reached an average value of 1.2 × 103. These results are the first to illustrate high BT BCFs and BMFs in human‐edible macroalgae and urchins sampled from northern coastal areas with a low TBT contamination level. The adsorption of activated carbon prepared from Scenedesmus obliquus (algae) was evaluated through adsorption of Astrazon red. The adsorption efficiency of activated carbon was determined based on the specific surface area and pore size distribution. These results were compared with the results obtained with untreated algae. Approximately a 3-fold increase in the percentage of dye removal was observed for activated carbon compared to the untreated material. The primary reason for this observation may be the increase in specific surface area and total pore volume by chemical activation from 0.0136 to 423.7001?m2?g?1 and from 0.0012 to 0.1643?cm3?g?1, respectively. A pseudo-second-order model was fit with the kinetic data and the results indicate chemical adsorption. The maximum adsorption capacity of activated carbon was 181.82?mg?g?1 at 25°C according to Langmuir isotherm model.  相似文献

In this work, the effects of elicitor concentration and elicitation day on the growth and asiaticoside production of centella cells were investigated. The results showed that 2-hydroxybenzoic acid from 50?C200 ??M and yeast extract from 2?C5 g L?1 had different eliciting influences. The addition of 2-hydroxybenzoic acid and yeast extract to the cultures strongly enhanced asiaticoside production in centella cells. The increase in asiaticoside content induced by the addition of 100 ??M of 2-hydroxybenzoic acid and 4 g L?1 of yeast extract at day 10 of inoculation was about 5- and 3.5-fold, respectively, as compared with that of the reference cells. In general, 2-hydroxybenzoic acid (abiotic elicitor) was more effective in enhancing asiaticoside biosynthesis than yeast extract (biotic elicitor).  相似文献
