The asymmetric synthesis of 4-nitrocyclohexanone derivatives has been accomplished by enantioselective double Michael additions of γ,δ-unsaturated β-ketoesters to nitroalkenes using a catalytic amount of bifunctional thiourea and TMG. The three contiguous stereogenic centers of the obtained products were constructed with good to high diastereoselectivity and up to 92% ee. The biologically active natural product, (−)-epibatidine, has been synthesized from the intermediate 11a in seven steps in 30% overall yield.  相似文献

Successive treatment of γ,δ-unsaturated β-ketoesters and nitroalkenes with a bifunctional thiourea and TMG promoted the tandem Michael addition, giving rise to highly functionalized cyclohexanones in good yields. The three contiguous stereogenic centers of the obtained products were constructed with high diastereo- and enantioselectivity (up to >99% de and 92% ee). The reaction was successfully applied to the asymmetric synthesis of (−)-epibatidine, which was synthesized from the cyclohexanone derivative in seven steps in 30% overall yield.  相似文献

A novel method for the synthesis of 1,3,4-thiadiazole and 1,3,4-thiadiazine derivatives bearing a carbazole moiety is described. Carbazole was transformed into carbazole-9-thiocarbohydrazide in two steps. This compound was allowed to react with various electrophiles to yield 1,3,4-thiadiazole derivatives. The reaction with bifunctional electrophiles led to 1,3,4-thiadiazines. 2-(Carbazol-9-yl)-5,6-dihydro-4H-1,3,4-thiadiazin-5-one reacted with piperidine and formaldehyde to yield the 4-(piperidin-1-ylmethyl) derivative. The reaction with aromatic aldehydes led to the corresponding 6-arylidene derivatives, which were transformed into pyrimidino[4,5-e]-1,3,4-thiadiazines and pyrazolo[3,4-e]-1,3,4-thiadiazines by a reaction with guanidine, acetamidine, or phenylhydrazine, respectively. Structures of the products were confirmed by 1H NMR, 13C NMR, IR, and mass spectrometric measurements. Selected examples of products were screened for anticancer activity.  相似文献

The total synthesis of protein kinase C activator (-)-indolactam V (IL-V) has been successfully completed with two separate approaches: From known 4-nitrotryptophan derivative 3 in 8 steps (49% overall yield) and from L-glutamic acid in 12 steps (18% overall yield), where 4-nitrotryptophanol derivative 4 served as a key intermediate. Derivatives 3 and 4, both incorporating indole 4-substitution and the C-9 stereocenter in IL-V, were synthesized via the Pd-catalyzed indole synthesis from 3-nitro-2-iodoaniline 5 with aldehydes 6 and 7, respectively. Aldehyde 7 was, meanwhile, synthesized from l-glutamic acid in 5 steps (68% yield). Lactamization of the 9-membered ring was achieved using HATU in THF in good yield.  相似文献

A short and high-yielding synthesis has been devised to prepare C-linked 2-deoxy-2-acetamido-alpha-D-galactopyranose derivative 3. One of the main advantages of this approach is that it employs commercially available and inexpensive d-glucosamine as the starting material. The key steps include a highly stereoselective C-allylation followed by epimerization of the C-4 hydroxyl group. Building block 3 and orthogonally protected C-linked 2-deoxy-2-acetamido-alpha-D-galactopyranose derivative 2 were obtained in 44% overall yield (six steps) and 29% overall yield (eight steps), respectively. This represents a significant improvement over previously reported syntheses.  相似文献

A concise and practical strategy to obtain C3 reverse-prenylated pyrrolidinoindoline scaffold has been executed in 28.8% overall yield. The key conjugative step involved a Booker-Milburn-Feudoloff reaction involving an NCS-mediated activation of indole, followed by coupling to C5 dimethylallylalcohol. This linchpin step proceeded in 74% yield. The overall sequence proceeded in five steps from commercially available N-methyltryptamine with a single protection-deprotection operation and a single redox manipulation. Mechanistic insights of NCS activation, and an ensuing rearrangement of the isoprene unit were gained by rationally varying the C3 substituent.  相似文献

We report a short synthetic route to (±)‐horsfiline that provides the racemate in five steps from commercially available, substituted isatin in 41% overall yield. Efficient entry into the spiro[oxindole‐pyrrolidine] ring system is possible through the application of the cyclopropane opening/ring expansion chemistry we have described with MgI2 as a bifunctional catalyst.  相似文献

The Stoichiometry of thermal decomposition was studied for the following compounds: Ni(NCS)2(pip)4 (I), (pip=piperidine), Ni(NCS)2(pip)2py·H2O (II), (py=piridine), Ni(NCS)2(4-Mepip)3 (III), Ni(NCS)2(3-Mepip)3 (IV) and Ni(NCS)2(3.5-Me2pip)3 (V). In complexes I, II, III and IV the loss of the volatile ligands (on the TG curve to 300 °C) occurs in three steps and in complex V in two steps. The loss of the last molecules of volatile ligands is accompanied by the decomposition of NCS groups. Spectral data and magnetic moment values for the initial complexes I and II (together with the defined intermediates) indicated pseudooctahedral configuration while pentacoordination for complexes III, IV and V. Structural changes of the complexes studied in thermal decomposition reactions are discussed.  相似文献

Role of alkali and alkaline earth ions on the formation of calix[4]arene-amide derivatives through O-alkylation of the lower rim phenolic-OH groups in general and template action of K+ in particular have been explored. Na+ and K+ ions among alkali, and Ca2+ and Sr2+ ions among alkaline earth have shown tetra-amide derivatives bound to metal ion species. Among all these, potassium salts act as template and yields a K+ bound tetra-amide derivative where the charge is counter balanced by a calix[4] arene-monoanion and the product is crystallographically characterized. Change in the amide precursor used in these O-alkylation reactions has no effect on the type of the amide derivative formed. Also demonstrated is a direct one-step reaction for the preparation of 1,3-di-amide derivative in high yield and low reaction period using CsHCO3.  相似文献

A simple and direct approach for the synthesis of (S)-gizzerosine, an amino acid responsible for the disease, black vomit, and a potent histamine H2-receptor, has been developed in 10 steps and in 31% overall yield from l-aspartic acid. The key steps involved a two-carbon homologation of an l-aspartic acid semi-aldehyde and direct alkylation of unprotected histamine with a 6-hydroxynorleucine derivative.  相似文献

Sulfanyl radicals trigger a tandem addition-cyclization protocol in linalool or citronelene derivatives for the efficient construction of the iridane monoterpene skeleton. Best results in yields and diastereoselectivity were obtained when phenylethylsulfanyl was used as radical initiator. We have proved the utility of this protocol with the enantiospecific synthesis of natural iridane dehydroiridomyrmecin starting from a (−)-linalyl acetate ester derivative in five steps with a 28% overall yield.  相似文献

In the presence of 2 mol-equiv. of AcONa, both triglycosylborane 1 and dicyclohexylglycosylborane 2 readily react with bromine chloride generated in situ from bromide and N-chlorosuccinimide (NCS) to give the bromo-sugar 3 (75 and 60%, resp.). The use of the BH3-THF/bromide/NCS/2 AcONa procedure permits the rapid, face-specific synthesis of 6 (58% uptake of bromine) and face-selective synthesis of 8/9 (71% uptake of bromine), from vinyl ether derivatives 5 and 7 , respectively. The dicyclohexylborane/bromide/NCS/2 AcONa procedure leads to the fast and quantitative conversion of 11 to the bromosugar 12 (91%). Hydroboration-transmetallation sequences give access to C-mercuriated carbohydrates 13 (71%) and 14 (78%). The bis(glycosyl)mercury derivative 13 is spontaneously cleaved by reaction with one equivalent of bromide/chloramine-T/aqueous HCl-solution to give 3 (87%) and 14 (76%). Hydrostannylation of acetylenic sugar 15 gave the (E)-stannylvinyl derivative 16 as the major product. This latter precursor 16 is spontaneously cleaved by the bromide/chloramine-T/aqueous HCl-solution reagent to give the bromovinyl-sugar 17 (96%).  相似文献

The first steps of a novel approach to the total synthesis of 9, 11-dehydroestrone via tricyclo[3.3.0.02,8]octan-3-one (2) are described. One route involves a tandem-type transformation of the key intermediate 3 (A-CD unit) consisting of cyclopropane cleavage and ring B closure to afford C, 18-bisnor-13 α, 17 α-estradiol derivatives. E.g. the 3-methoxy-9 ζ-hydroxy-17 α-methanesulfonyloxy derivative (-)- 6 has been synthesized in 8 steps and 10% overall yield from 1,3-cyclohexadiene. As an alternative, the A-CD type intermediate 4b has been prepared and could be used for a ring C enlargement prior to cyclization.  相似文献

A possible inhibitor of proteases, which contains an indole core and an aromatic polar acetylene, was designed and synthesized. This indole derivative has a molecular architecture kindred to biologically relevant species and was obtained through five synthetic steps with an overall yield of 37% from the 2,2′-(phenylazanediyl)di(ethan-1-ol). The indole derivative was evaluated through docking assays using the main protease (SARS-CoV-2-Mpro) as a molecular target, which plays a key role in the replication process of this virus. Additionally, the indole derivative was evaluated as an inhibitor of the enzyme kallikrein 5 (KLK5), which is a serine protease that can be considered as an anticancer drug target.  相似文献

By formylation of calix[4]-1,3-aza-crown in hexamethylenetetramine/trifluoroacetic acid system, the first formylated calix[4]-aza-crown derivative 6 was synthesized in yield of 61%. Reacting compound 6 with salicyloyl hydrazine, 2,4-dinitrophenyl hydrazine, nicotinyl hydrazine or phenyl thiosemicarbazide afforded four novel calix[4]crown hydrazone derivatives 7ad in yields of 70–90%. By condensating compound 6 with series of bifunctional reagents, the novel mono-bridged biscalix[4]-aza-crown hydrazone derivatives 8ac were prepared in high yields. The extraction experiments showed that all new compounds were good receptors for metal cations, especially, for soft metal cations. The extraction results suggested that the hydrazone groups produced favorable influences for binding soft cations. The two calix[4]arene units in compounds 8ac could bind metal cations concurrently.  相似文献

New Tetrathiocyanato Diamine Chromiates with Tertiary Aliphatic Amines Three new Reinecke salt like complexes: [Cr(NCS)4(amine)2]? were obtained by means of substitution reactions of anhydrous K3[Cr(NCS)6] with triethyl-, tripropyl-, and tributylamine, respectively, without solvent. The formulae of the new complex anions were established by preparation of 63 derivatives with the hydrochlorides of some amines and alkaloides, with metal ions and cobalt(III) amine bases, respectively. The electronic and i.r. spectra of some derivatives were recorded. Some structural problems were discussed on the basis of spectral data.  相似文献

tert-Butyl 2-substituted 4,6-dioxo-1-piperidinecarboxylates 4 have been prepared in good yield starting from Boc-Asp-O(t)Bu and other beta-amino acids. By analogy with chiral tetramic acids, their reduction by NaBH(4) in CH(2)Cl(2)/AcOH afforded the corresponding cis-4-hydroxy delta-lactams in good yield and stereoselectivity (68-98% de). In the absence of the A(1,3) strain (reduction of 6-substituted 2,4-dioxo-1-piperidines 7), the cis-4-hydroxy isomer was still obtained as the major product but the de values were consistently lower. 4-Hydroxy-6-oxo-1,2-piperidinedicarboxylate 2a, readily accessible from Boc-Asp-O(t)Bu (three steps, 63% overall yield), has proven to be an excellent building block for the synthesis of cis- and trans-4-hydroxypipecolates 17 and 24 (52 and 36% overall yield, respectively) and for the synthesis of a protected 4-hydroxylysine derivative 29 (41% overall yield).  相似文献

This comprehensive report accounts the development of a highly diastereoselective samarium diiodide‐induced cascade reaction of substituted indolyl ketones. The complexity‐generating transformation with SmI2 allows the diastereoselective generation of three stereogenic centers including one quaternary center in one step. The obtained tetra‐ or pentacyclic dihydroindole derivatives are structural motifs of many monoterpene indole alkaloids, and their subsequent transformations gave way to one of the shortest approaches towards strychnine (14 % overall yield in ten steps, or 10 % overall yield in eight steps). During the course of this report we discuss the influence of substituents on the cyclization step, plausible mechanistic scenarios for the SmI2‐induced cascade reaction, diastereoselective reductive amination, and regioselective dehydratization protocols towards the pentacyclic core structure of strychnos alkaloids.  相似文献
