Stereoselective hydrogenation of thymol over Rh/alumina in the presence of various equivalents of -cyclodextrin (-CD) and itsderivatives in the solid state was studied. Hydrogenation of thymol in the absence of -CD gave 76.8% epimeric alcohols with a menthol/neomenthol (M/N) ratio of 5.6 and an alcohol/ketone ratio of 4.5, whereas the presence of 0.1 equivalent (to thymol) of -CD gave rise to 94.6% of epimeric alcohols with a M/N ratio of 6.3 and an alcohol/ketone ratio of 45.4. The effect of -cyclodextrin and its derivatives on the modification of the yield and the proportion of epimeric alcohols formed were found to be the salient features of this investigation. Inclusion complexation of thymol by -CD studied by UV-Visible spectroscopyindicated a 2:1 stoichiometry ofthymol: -CD complex with a binding constant value of 480 ±40 M-2.  相似文献

In an effort to modify the solubility and dissolution rate of the contraceptive steroid, 19-norprogesterone in order to improve its bioavailability, the cyclodextrin complexation approach was chosen. In solution, the complex formation with -cyclodextrin (-CD), hydroxyethyl -cyclodextrin (HE--CD) and hydroxypropyl -cyclodextrin (HP--CD) was confirmed by using solubility, UV, IR and 1H-NMR spectrophotometric techniques. The phase solubility diagrams were categorized as AL-type. The complexing affinity of the CDs investigated were ranked as follows: -CD > HP--CD > HE--CD. The complexation thermodynamic parameters were obtained from the temperature dependence of the dissociation constants. In the solid state, differential scanning calorimetery (DSC) and optical microscopy methods were utilized to characterize the complexes. Dissolution studies showed that such molecularly encapsulated forms offered a marked improvement in the dissolution rate compared to the parent drug.  相似文献

Inclusion complexation of all-trans-retinol, retinal and retinoic acid with -cyclodextrin (-CD) and heptakis(2,6-O-dimethyl)--cyclodextrin (DM--CD) were investigated by means of UV-vis spectroscopy. The association constants (K a) obtained for vitamin A with DM--CD is greater than with -CD. Effects of Cyclodextrins on Deodoration of ``Aging Odor'   总被引:2,自引:0,他引:2  
Hexenal, octenal and nonenal are known causes of unpleasant body odor and are present at markedly increased levels in the middle-aged and elderly. The odor of such unsaturated aldehydes is therefore called ``aging odor'. The present study investigated the effects of cyclodextrins (CDs) on deodoration of crotonaldehyde, pentenal, hexenal, heptenal, octenal, nonenal, decenal, undecenal and dodecenal. -, - and -CD formed inclusion complexes with the majority of unsaturated aldehydes in aqueous solution. The -CD inclusion complex contained the highest amount of guest molecule. One molecule of -CD was estimated to include 1 molecule of short chain aldehyde and 2 molecules of long chain aldehyde. Deodorant testing was conducted by headspace gas analysis using sealed vials. All CDs decreased the concentrations of unsaturated aldehyde. With nonenal, the deodorant power of parent CDs was -CD > -CD > -CD, and that of chemically modified CD was Me-CD > HP-CD > G2-CD > MCT-CD. CDs were demonstrated to reduce ``aging odor'. Me-CD was the most effective type of CD for the deodoration of ``aging odor'.  相似文献

Inclusion compounds of ferrocene(FcH) and its derivatives with cyclodextrins(CDs; -CD, -CD, and -CD) were prepared. CD-ferrocene inclusion compounds were obtained in a crystalline state in high yield. -CD and -CD formed 11 stoichiometric inclusion compounds with ferrocene and its derivatives. -CD formed 21 (CD:guest) complexes with ferrocene and the monosubstituted derivatives, but did not form complexes with 1,1-disubstituted derivatives, -CD-FcH and -CD-FcH complexes are thermally stable and do not liberate ferrocene on heating at 100°C in vacuo. The cyclodextrin inclusion compounds were characterized by1H-NMR, IR, UV, and CD spectra. A large positive induced Cotton effect was observed in the case of -CD-FcH complex, while the -CD-FcH complex showed a negative spectrum. The binding mode is discussed. -Cyclodextrin was found to form inclusion complexes in ethylene glycol, diethylene glycol, triethylene glycol, methyl cellosolve, ethyl cellosolve, methyl alcohol, and glycerine solutions. -CD also formed complexes in ethylene glycol solution. The binding of ferrocene by -CD is stronger in ethylene glycol than in dimethyl sulfoxide and dimethyl formamide.  相似文献

Aim of the present work was to investigate the effect of somecyclodextrins (CDs) on the solubility and ocular bioavailability of rufloxacin base (RUF), with theultimate goal of developing an ophthalmic formulation. Phase solubility studies of RUF inpH 7.4 buffer were carried out in the presence of -cyclodextrin (-CD),hydroxypropyl--cyclodextrin (HP--CD) and -cyclodextrin(-CD). The effect of hydroxypropyl methylcellulose (HPMC) on RUF solubility was evaluated after heating the solutionscontaining HP--CD at 120 °C.A significant enhancement of RUF solubility was achieved by associatingthe drug with CDs, particularly HP--CD. This CD formed with RUF a less stablecomplex than that formed by -CD, but did not suffer the solubility limitations ofthe parent CD, and showed a higher solubilizing capacity than -CD. Addition of 0.25%(w/v) HPMC to solutions containing HP--CD increased the solubilizing effect of this CD,thus allowing reduction of the amount necessary for solubilization of 0.3% (w/v) RUF.Preliminary pharmacokinetic data in rabbits indicated that theocular bioavailability of 0.3% (w/v) RUF solubilized by HP--CD was higher when compared witha 0.3% (w/v) RUF suspension used as reference.  相似文献

The influence of cyclodextrins (CDs) on the chemical stability of the contraceptive steroid progestin, ST1435, in aqueous solution has been studied using reversed phase high performance liquid chromatography. The effects of CD structure, temperature, and CD concentration on the rate of degradation were investigated. It was found that the drug degraded to different extents following a pseudo-first order reaction mechanism. The presence of the host molecules affected the degradation rate as a result of complexation which might result in protection of the labile moiety of the drug molecule against degradation. Hydroxypropyl--cyclodextrin (HP--CD) and hydroxyethyl--cyclodextrin (HE--CD) retarded the degradation in contrast to -cyclodextrin (-CD) which accelerated the steroid degradation. The stabilizing action of HP--CD is larger than that of HE--CD. The degradation rate increased upon increasing temperature and the Arrhenius equation is valid. Lineweaver-Burk equation analysis indicated that the steroid included inside the CD cavity degraded three times more slowly than did the free ST1435 in solution. This equation further supported the formation of a 1 : 1 inclusion complex between ST1435 and HP--CD with a stability constant of 934.5 M-1 at 65°C.  相似文献

The inclusion complexes of -CD with sodium octylsulfonate (C8As), sodium dodecyl sulfonate (C12As), andsodium hexadecyl sulfonate (C16As) in aqueous solutions havebeen studied by surface tension measurement at the air/water interfaceand 1H NMR spectroscopy at 323 K.At fixed concentrations of the surfactants, the surface tensions firstincrease with the increase of -CD concentrations,then they attain a maximum. The surface tension curves of the surfactantsin the presence of -CD are higher than those in the absence of-CD. The values increase with increasing -CD concentrations foreach surfactant. The apparent critical micelle concentrations (CMC) of thesurfactants vary linearly with -CD concentrations.A 1H NMR study shows that the signals of theinner H-3 and H-5 of -CDshift upfield upon addition of the surfactants.The magnitude of the chemicalshift changes (= CD obs)varies as a functionof the concentrations of the surfactants. From therelationships between the chemicalshift changes of H-3 or H-5 inside the -CD cavityand guest/host molar ratios, a 1:1 stoichiometry foreach inclusion complex is assumed. The associationconstants of the inclusion complexes have beendetermined by 1H NMR spectroscopy.  相似文献

The partition coefficients Kav between the solution phase and octyl- and phenyl-sepharose CL-4B were determined for three cyclodextrins (CDs), -CD, -CD and -CD in various aqueous solutions, as a measure of their interactions with the two hydrophobic ligands. Kav of the CDs increased in the order of -CD<-CD<-CD for octyl-sepharose CL-4B and -CD<-CD<-CD for phenyl-sepharose CL-4B. In all cases, Kav increased by increasing NaCl concentration in the aqueous solution phase and also by lowering temperature, but in the presence of NaBr and NaSCN, both chaotropic salts, Kav decreased markedly. The spontaneity of the transfer of the CDs from the aqueous solution phases to the gel phases was due to the enthalpy decrease. It was shown that discrete separation of the three CDs can be achieved by the hydrophobic chromatography on a short column (1×25cm) of octyl-sepharose CL-4B by adjusting the NaCl concentration and temperature.  相似文献

The effect of reoxidation temperature on TPR of 5% Rh/Al2O3 catalyst and its mechanical mixture with ZnO has been investigated. The results suggest bimodal character of supported rhodium and surprisingly high effect of this metal on ZnO reduction in the mixture.

The formation of inclusion complexes between 4-amino-1,8-naphthalimides and cyclodextrins (CDs) was investigated. The naphthalimides used in the study were 4-amino-1,8-naphthalimide (I) and4-(2-phosphonoethylamino)-N-(2-phosphonoethyl)-1,8-naphthalimide,tetraethylester (II). The CDs employed were -CD, -CD, -CD, HP--CD, HP--CD andHP--CD (HP = hydroxypropyl). Evidence for complex formation was obtained from the changes in the fluorescence spectra of the dyes in the presence of increasing amounts of the CDs. The most striking changes were observed with HP--CD and HP--CD. Treatment of the data using Benesi–Hildebrand plotswas consistent with a 1:1 inclusion model. The determined stabilityconstants were (Keq, M-1): 106 (I:HP--CD, pH = 2.0), 193 (I:HP--CD, pH = 7.0), 113 (I:HP--CD, pH = 7.0), 155(II:HP--CD, pH = 2.0), 121 (II:HP--CD,pH = 7.0), 301 (II:HP--CD, pH = 7.0). It can beconcluded that compound I forms a more stable complex with HP--CD than with HP--CD. Compound II, on the other hand, forms a more stable complex with HP--CD than with HP--CD.  相似文献

A mordenite-containing rock sample from the Tokaj Mountains (Hungary) and its various forms modified by chemical treatment have been tested for catalytic properties during isomerization of n-hexane. Addition of Pt to the catalyst leads to a sharp decrease in the rate of cracking and in the rate of deactivation and this results in a catalyst of high selectivity for hexane isomers.

A derivatograph was used for the thermal modification of the surface properties of different silica gels. Analogous investigations were carried out with an isothermal method. The possibility of using the thermal method of temperature programming to modify adsorbents is discussed.

Equilibrium constants for the formation of 1 : 1 inclusioncomplexes of -cyclodextrin (-CD) with neutral and anionic phenol derivatives (3- and 4-cyanophenols and 3- and 4-nitrophenols) have been evaluated at 5, 12, 25, and 35 °C by means of spectrophotometry. Similarly, the equilibrium constants have been determined for the inclusion complexes of-cyclodextrin (-CD) with the phenols. Enthalpy and entropy changes for the formationof the inclusion complexes have been estimated from the temperature dependences of theequilibrium constants. With -CD, the enthalpy andentropy changes for the anionic species have been found to be more negative than those for the neutral ones, except for 4-cyanophenol, suggesting that the inclusion complexes of the anionic species are more rigid than those of the neutral species. From analyses of chemical shift differences in 13C NMR spectra of 3- and 4-cyanophenolsand 3- and 4-nitrophenols in aqueous solutions with and without CDs, a nitro ora cyano group has been found to be first bound to the - and -CD cavities.  相似文献

The complexation behavior of zinc (II) tetraphenylporphyrin-viologen covalently linkedcompounds by a polymethylene chain (ZnPCnV2+; n = 4–9) withtri-O-methyl--cyclodextrin (TM--CD) was investigated by meansof 1H NMR and UV/Vis absorption spectrometry in acetonitrile-water (1:1, v/v).The 1H NMR spectra indicated that ZnPCnV2+ existed as a mixture ofa dimer and a monomer at high concentration (>1 × 10-3 M). The dimerwas degraded to the monomer upon complexation with TM--CD involving theformation of 1:1 and 1:2 (ZnPCnV2+: TM--CD) complexes, whichwas observed by UV/Vis spectrometry. Furthermore, 1H NMR spectra exhibitedthat there are at least two types of structures for these complexes, one is a fastexchanging complex, and another is a slow exchanging one. A slow exchanging complexcould be assigned as a rotaxane type structure in which the methylene chain moiety ofZnPCnV2+ penetrated into the cavity of TM--CD. Discrete formationconstants for these complexes were evaluated. The values were little affected by the spacermethylene chain length (n 7).  相似文献

FT-Raman spectra of 2-, 3-, and 4-chlorostyrene included in-cyclodextrin (CD),glycerol ether -CD, -CD,sulfated -CD, andglycerol ether -CD were recorded.In the inclusion complexes, the area of the vinyl(C=C) band decreased remarkably, whereasthe area of the phenyl (C=C) band increasedcompared to those of liquid 2-, 3-, and 4-chlorostyrene,respectively.
