An efficient synthesis of indolo[1,2-c]isoindolo[2,1-a]quinazolin-5(16aH)-one derivatives through palladium-catalyzed cyclocarbonylation reaction of 6-(2-bromoaryl)-5,6-dihydroindolo[1,2-c]quinazolines with the use of CO as a carbonyl source under atmospheric pressure has been disclosed. More intriguingly, the above-mentioned products can also be obtained directly from a one-pot two-step tandem reaction of 2-(1H-indol-2-yl)anilines and 2-bromobenzaldehydes, which are the precursors of 6-(2-bromoaryl)-5,6-dihydroindolo[1,2-c]quinazolines, under an atmospheric CO pressure in modest yields.  相似文献

A one-pot reductive amination of aldehydes and ketones with amines using α-picoline-borane as a reducing agent is described. The reaction has been carried out in MeOH, in H2O, and in neat conditions in the presence of small amounts of AcOH. This is a highly efficient and mild procedure that is applicable for a wide variety of substrates. In particular, this is the first successful demonstration that this type of reaction can be carried out in water and in neat conditions.  相似文献

Reaction of anomeric thiocyanates with triethyl phosphite, dimethyl phenylphosphonite and methyl diphenylphosphinite afforded the corresponding S-glycosyl thiophosphates, thiophosphonates and thiophosphinates in good yields. These derivatives were applied as glycosyl donors in the synthesis of benzyl glycosides and disaccharides with excellent stereoselectivity.  相似文献

A palladium-catalyzed cyclizative carbonylation of azobenzenes has been developed to access 1-acyl 2-aryl 3H-indazol-3-ones in moderate to good yields with good functional compatibility. This procedure proceeded with the sequential ortho-C–H carbonylation and cyclization, where formic acid served as the CO source. The practicability of this transformation was further increased by the employment of facilely available azobenzenes derivatives as one-handled starting materials.  相似文献

A novel series of conjugates of benzosuberone and 1,2,3-triazole i.e. 3-(4-phenyl-1H-1,2,3-triazol-1-yl)propyl-9-chloro-2,3-dimethyl-6,7-dihydro-5H-benzo[7]annulene-8-carboxylic acids (8a-j) were synthesized in good to excellent yields catalysed by CuSO4 under milder reaction conditions and evaluated for their anti-proliferative activity. The structural elucidation of the prepared compounds was carried out using IR, 1H NMR, 13C NMR and Mass spectral analysis. The newly synthesized derivatives (8a-j) were evaluated for their anti-proliferative activity against four human cell lines and the novel derivatives showed moderate to excellent activity. The obtained results suggest that these compounds can be considered as new hits for anti-proliferative drug development programme and further SAR studies can help obtain better anticancer agents.  相似文献

The application of immobilizing calixarene onto the surface of silica using a sol-gel method, as an efficient heterogeneous catalyst (CX4SO3HSi(n)) for multicomponent Povarov reaction has been described. Catalytic activity of the CX4SO3HSi(n) for the synthesis of julolidines under greener and environmentally benign conditions in simple and efficient method was explored. Notably, only 0.5 mol% of the catalyst is sufficient to catalyze the Povarov solvent-free reaction under microwave-assistance. Besides, this protocol allows the construction of four new C–C bonds and two C–N bonds in a single step. To the best of our knowledge, this consists the first silica support calix[4]arene as a heterogeneous catalyst for multicomponent synthesis of julolidines.  相似文献

A visible-light-mediated metal-free thiocyanate radical addition/ipso-cyclization/oxidation cascade reaction for the synthesis of thiocyanato-containing azaspirotrienediones from N-phenylpropynamides is described. Cheap and readily available ammonium thiocyanate was used as a precursor to the thiocyanate free radical, which undergoes a radical addition reaction with the alkyne, followed by selective ipso-cyclization and oxidation to afford the dearomatized products. No product of ortho-cyclization was detected. The reaction completes the synthesis of C–S, C–C, and CO bonds in one pot, with abundant and renewable air oxygen as the sole sacrificial reagent and oxygen source.  相似文献

Pd/Cu-catalyzed Heck type cross-coupling reaction of p-toluenesulfonylmethyl isocyanide (TosMIC) with various styrenes to access stilbenes in DMSO solvent under mild conditions is developed. Simple one pot procedure for the conversion of carboxylic acid to ketone is described. Various carboxylic acids were converted to the corresponding ketones in excellent manner in presence of N,N′-dicyclohexylcarbodiimide (DCC) and organometallic reagents. Aromatic, heteroaromatic and aliphatic acids were converted to the corresponding ketones smoothly under the optimum conditions using organolithium reagents. In this process, desired products have been isolated from the crude reaction mixtures in moderate yields during the purification process.  相似文献

In this article, a CaO-based catalyst was prepared by impregnating chloride salts on CaO to develop a highly efficient heterogeneous catalyst for the synthesis of glycerol carbonate (GC) from glycerol and dimethyl carbonate. LiCl/CaO exhibited a high catalytic activity under moderate reaction conditions. The effects of the LiCl loadings, the amount of catalyst and the calcination temperature on the catalytic activity were investigated. The highest yield of 94.19% glycerol carbonate was obtained at 65 °C on CaO loaded with 10% LiCl after 1 h, and the catalyst had high stability in reusing work. Scanning electron microscopy (SEM), X-ray diffraction (XRD), BET, CO2-TPD, XPS and thermalgravity (TG) were used to characterize the prepared catalyst. It was found that the high catalytic activity of CaO after modification with LiCl is associated with the structural aspects and the amount of basicity of the catalyst. The Li2O2 species, which is a strong basic site that is formed by the substitution of the Ca2+ in CaO lattice by Li+, has great activity for transesterification.  相似文献

Reaction sequences consisting of an initial intermolecular titanium-catalyzed hydroaminoalkylation of a suitably ortho-bromophenyl-substituted alkene and a subsequent intramolecular Buchwald-Hartwig amination are used for the synthesis of benzazepine, benzoxazepine, and benzothiazepine derivatives. While in the latter two cases, the hydroaminoalkylation products obtained from an allyl (2-bromophenyl) ether or an allyl (2-bromophenyl) thioether must be purified prior to the subsequent palladium-catalyzed amination step, both reactions can be combined to an efficient one-pot procedure for the synthesis of 2,3,4,5-tetrahydrobenzo[b]azepines when 4-(2-bromophenyl)-1-butene and various N-methylanilines are used as the starting materials.  相似文献

Development of a synthetic method for the preparation of homogeneous glycopeptides and glycoproteins is important for the elucidation of their structures and functions. Here, we report on the concise and facile synthesis of glycopeptides using Boc groups for the protection of carbohydrate hydroxyl groups. This method enables us to remove the protecting groups from peptide and carbohydrate moieties in a single-step process without undesirable any side reaction.  相似文献

Nanostarch is shown to be a highly efficient, eco-friendly and heterogenous organocatalyst for the synthesis of the diheteroaryl thioethers via one-pot reaction of methylcarbonyls, thiourea and iodine in DMSO. This method offers significant advantages such as available starting materials, higher purity and excellent yield of products, very easy reaction conditions and absence of any tedious purification. Furthermore, because of employing eco-friendly catalyst without using transition metal catalysts, this novel method emerges as a green-approach leading to less harmful residues. Moreover, a mechanism was proposed to rationalize the reaction and the role of starch nanoparticles was also investigated in these transformations.  相似文献

Lindenane oligomers isolated from Chloranthus pose significant challenges for chemical synthesis. The structure of a proposed biosynthetic monomer, lindenatriene, was recently called into question. Its attempted synthesis produced a compound whose 1H NMR spectrum differed significantly from the spectrum of a monomer produced by oligomer pyrolysis. Here we propose that the original structural assignment after pyrolysis was correct and instead the spectra of synthetic materials were misinterpreted. Reanalysis of 2D NMR data suggest that lindenatriene isomerizes (formal [1,7]-hydrogen shift) upon treatment with base.  相似文献

A convenient and highly efficient method for the synthesis of novel polyheterocyclic chromeno pyrimido[1,2-b]indazolone derivatives via a three-component condensation of 1H-indazol-3-amine, aldehydes and 4-hydroxycoumarin catalyzed by Bronsted acid ionic liquid [Et3NH][HSO4] under solvent-free reaction conditions is presented. This ionic liquid is air and water stable and easy to prepare from amine and acid. The main advantages of this protocol includes short reaction time, excellent yield, easy work-up, operational simplicity, a wide range of functional group tolerance and column chromatography-free method. The catalyst can be recovered and reused for at least four runs without any significant impact on the product yields.  相似文献

We designed new pyrazole derivatives as inhibitors of the cell cycle kinases and developed a simple environmentally sustainable synthesis process. We synthesized the pyrazolyl thiourea derivatives using rapid ultrasound mediated methods and confirmed their structures by NMR and IR spectra. The apoptosis and necrosis inducing effects of the new compounds were investigated. Cell cycle analysis and expression of genes involved in apoptosis, cell cycle and xenobiotic metabolism were studied. The compounds presented modest apoptotic effects in human cancer cells. The N-[[3-(4-bromophenyl)-1H-pyrazol-5-yl]carbamothioyl]-4-chloro-benzamide compound (4e) induced a significant increase of cells in G2/M phases in conjunction with an increased expression of cyclin A and cyclin B, emerging as a promising anticancer drug, to be further developed in animal models of cancer.  相似文献

1H-Pyrazolo[1,2-b]phthalazine-5,10-dione carbonitriles and carboxylates possess a broad range of applications as active compounds in the pharmacological and biological fields. We developed an efficient and ecofriendly silica gel-supported tungstic acid (STA)-catalyzed one-pot synthesis of 1H-pyrazolo[1,2-b]phthalazine-5,10-dione carbonitriles and carboxylates by a three-component reaction of an aldehyde, a malononitrile/ethyl cyanoacetate and a phthalhydrazide under solvent-free conditions. The major advantages of the present method are experimental simplicity, use of an inexpensive and ecofriendly reusable catalyst, good yields, and short reaction times.  相似文献

An Ir-mediated photocatalytic coupling of tertiary amines with Ugi-dehydroalanines was developed as an entry to medicinally important 2,4-diaminobutyric acid derivatives. In the process the 2,4-diaminobutyric acid framework is assembled directly embedded into a peptoide structure, via the construction of the C3(sp3)–C4(sp3) bond, through a CH functionalization. The photocatalyzed oxidation of the tertiary amine produce a free radical intermediate which reacts with the double bond present in the dehydroalanines. The complete protocol comprises an Ugi 4-CR followed by an elimination reaction and the photo-induced coupling. Using this strategy, 15 new diversely substituted unnatural α,γ-diamino acids peptide derivatives were prepared in low to good yields.  相似文献
