An efficient method for the preparation of 2,2-diarylvinyl halides from the corresponding 1,1-diarylethylenes has been developed. N-Halosuccinimides (N-bromosuccinimide or N-chlorosuccinimide) were used as the halogenation reagents. The practicability of this method is highlighted by its simple operation, broad substrate scope and capability for large-scale reaction.  相似文献

Three new dibromopyrrole alkaloids, 9-N-methylcylindradine A (1), 1-N-methylugibohlin (2), nemoechine H (3), together with one known dibromopyrrole alkaloid N-methyldibromoisophakellin (4) were isolated from the South China Sea sponge Agelas nemoechinata. Their structures were elucidated by comprehensive spectroscopic methods including HRESIMS and NMR, and the absolute configuration of compound 1 was further confirmed by comparison of optical rotation. Compound 3 exhibited moderate cytotoxic activity against K562 and L-02 with IC50 values of 6.1 μM and 12.3 μM, respectively.  相似文献

A variety of N, 2-diaryl-2-ureidoacetamide prepared from the condensation of N-aryl-α-aminoamide with potassium cyanate (KOCN) undergo efficient Hendrickson’s reagent-mediated nucleophilic cyclization to afford 2,5-diamine-4-aryloxazoles. The two-step synthesis provides seven target products in yields of 61–78% under mild conditions. This reaction involves an unusual pathway in which the electrophilic amide carbonyl carbon is activated by Hendrickson’s reagent and attacked by a nucleophilic ureido oxygen in a 5-exo-trig O-cycloisomerization.  相似文献

An efficient method of copper-catalyzed aryldifluoromethylenation of N-arylacrylamides is described to step-economic synthesis of the various biologically important biheterocyclic compounds linked by gem-difluoromethylene moiety through addition of benzo-1,3-azolic (oxa- or thia-)difluoromethyl radicals generated in situ to terminal of C–C double bond of N-arylacrylamides then cyclization cascade. This protocol has demonstrated to have broad substrate scope and excellent functional-group tolerance under mild reaction conditions.  相似文献

The composition of the ethanolic extract from the aerial parts of Crocus alatavicus Regel & Semen from southern Kazakhstan spontaneous flora was analyzed together with the determination of its antibacterial, antifungal, antiviral and anticancer activity. The phytochemical profile analysis by high-performance liquid chromatography-electrospray ionization-quadrupole-time of flight-mass spectrometry (HPLC/ESI-QTOF-MS) revealed the presence of multiple kaempferol derivatives. High-performance reverse-phase liquid chromatography combined with a photodiode-array detection (RP-HPLC/PDA) found that kaempferol 3-O-dihexoside and kaempferol 3-O-acyltetrahexoside accounted for 70.5% of the kaempferol derivatives. The minimum inhibitory concentration (MIC) values of the extract for all the tested reference microorganisms were high, reaching 10 mg/mL for yeasts and 20 mg/mL for bacteria. In contrast, antiviral activity was observed at 2 mg/mL, resulting in the inhibition of the HSV-1-induced cytopathic effect and the reduction in virus infectious titer by 1.96 log, as well as the viral load by 0.85 log. Among the tested prostate cancer cell lines, significant cytotoxic activity of the extract was noted only on the LNCaP cell line, with an IC50 value of 1.95 mg/mL. The LNCaP cell line treated with 2 mg/mL of the extract showed a noticeably reduced number of spindle-shaped cells with longer cellular projections, a significant increase in the peak corresponding to the population of apoptotic cells in the sub-G1 phase and a decreased intracellular glutathione (GSH) level, suggesting the prooxidative properties of the extract. The obtained data provide novel information about the flavonoids present in the aerial part of C. alatavicus and suggest its potential application as a source of the compounds active against HSV-1 and metastatic, androgen-sensitive prostate cancer.  相似文献

Recently, there has been an upsurge in the occurrence of the indazole motif in drug discovery. Accordingly, newer, milder and more efficient routes towards their synthesis have emerged in the literature. We recently reported the Mitsunobu-triggered cyclodehydration of salicylaldoximes to transient 1,2-benzisoxazoles, and salicylhydroxamic acids to their corresponding 3-hydroxybenzisoxazoles. We hypothesized subjecting ortho-aminobenzoximes to Mitsunobu conditions will likewise induce a cyclodehydration to deliver the corresponding 1H-indazoles. Indeed, secondary anilines afforded the predicted N1-substituted 1H-indazoles, and primary anilines, after activation with a Boc group, furnished the N1-Boc 1H-indazoles in good to excellent yields. This work further expands the chemical repertoire of the Mitsunobu reaction, representing its unprecedented use in the construction of the 1H-indazole nucleus.  相似文献

The rate of hydrolysis–condensation reaction of phenyltrichlorosilane in water-acetone solutions and the product yields were shown to significantly depend on the concentration of HCl (CHCl) in the solutions. The main product of the reaction was all-cis-(tetrahydroxy)(tetraphenyl)cyclotetrasiloxane. This was different from the earlier published results of analogous reactions of m-tolylSiCl3, m-ClPhSiCl3, and α-naphtylSiCl, in which some products of other types were formed. For example, trans-1,1,3,3-tetrahydroxy-1,3-di-α-naphtyldisiloxane was obtained in the case of α-naphtylSiCl3. All-cis-(tetrahydroxy)(tetraphenyl)cyclotetrasiloxane was treated in acetone with HCl to give the other three geometric isomers (cis-cis-trans-, cis-trans-, and all-trans-). The thermal self-condensation of these four isomers under “pseudo”-equilibrium conditions (under atmospheric pressure) was investigated in different solvents, in quartz or molybdenum glass flasks. The compositions of the products were monitored by APCI-MS and 29Si NMR spectroscopy. It was shown that all-cis- and cis-cis-trans-isomers in toluene or anisole mostly gave the cage-like Ph-T8,10,12,14 and uncompleted cage-like Ph-T10,12OSi(HO)Ph compounds. In contrast to these two isomers, the cis-trans–isomer in toluene mainly formed dimers with the loss of one or two molecules of water. However, in acetonitrile, significant amounts of Ph-T10,12 and Ph-T10,12OSi(HO)Ph species were formed along with the dimers. All-trans-isomer did not enter into the reaction at all.  相似文献

Photodynamic therapy of cancer (PDT) at early stages of various types of cancer is based on the use of the phototoxicity of photosensitizers (PS) which appears upon their excitation by light in tumor tissues. The review concentrates mainly on the data for the second and third generations of photosensitizers: the structure of PS, their photophysical and photochemical properties, and some results of theirin vitro andin vivo application. A carefully designed PS should exhibit the following properties: long-wave absorption, good singlet oxygen quantum yield, an intramolecular polarity axis, and lowin vivo (photooxidation) stability. Uncharged PS that are lipophilic, positively charged, and capable of binding with monoclonal antibodies are discussed as an example.  相似文献

The potential curve for rotation around the central bond in 1,3-butadiene has been estimated by ab initio calculations using Gaussian-type basis functions. The calculations, which also include limited geometry variation during rotation, suggest that in the SCF approximation the second stable form of the molecule is a gauche conformation rather than a cis. The predicted energy difference between the planar trans ground state and the stable gauche form is 2.7 kcal/mole and the barrier to internal rotation is found to be 6.0 kcal/mole using a (9,5) basis for carbon and 4s functions on hydrogen.  相似文献

β-(N-Benzoylamino)methacrylamide, a key intermediate for the preparation of [2-11C]thymine, was synthesized in three steps from ethyl α-formylpropionate and NH3. Reaction of the alkali metal salts of β-(N-benzoylamino)methacrylamide with [11C]phosgene gave [2-11C]thymine. The yield of [2-11C]thymine was 362 ± 53 MBq at EOS (n = 3) (18 MeV proton beam; 10 μA, 10 min). The total synthesis was accomplished in just 16 min from the end of bombardment.  相似文献

The electrical conductivities of aqueous solutions of sodium salts of trans-4-hydroxycinnamic acid (trans-p-coumaric acid), trans-3,4-dihydroxycinnamic acid (trans-caffeic acid), trans-4-hydroxy-3-methoxycinnamic acid, (trans-ferulic acid) and trans-3-phenylacrylic acid (trans-cinnamic acid) with α-cyclodextrin were measured in the temperature range of 288.15 K–318.15 K. For the first time in the literature, using the limiting molar conductivity (Λmo) obtained from conductivity measurements, the values of the complexation constants (Kf) of the salts of phenolic acid derivatives with α-cyclodextrin were determined using a modified low concentration chemical model (IcCM). An attempt was also made to analyze the individual thermodynamic functions ΔGo, ΔHo and ΔSo describing the complexation process as a function of temperature changes. The obtained results show that the process of formation of inclusion complexes is exothermic and is spontaneous.  相似文献

The rate of the thermal rearrangement of (S) 2 chloromethyl-1-ethylpyrrolidine [(S)-1a] to (R)-3-chloro-1-ethylpiperidine [(R) 2a] has been examined at three temperatures in benzene by PMR and polarimetry. The rearrangement was shown to be completely stereospecific and to obey a simple first order rate law. The calculated Ea ΔH3 and ΔS3 were 22 ± 2 kcalmole (25°), 21 ± 2.5 kcalmole (25°) and - 10 ± 2 e.u. (0°K) respectively. The effect of solvents having differing dielectric constants was also studied. A transition state 9'a and an ion pair intermediate 3a are suggested for the rearrangement. The stereochemical course of the reactions of (S)-1a, (R)-2a and (S)-2a with hydroxide and methoxide ions have been shown to be 100% stereospecific with an uncertainty of about 1%. The absolute configurations of all optically active reactants and products [(S)- and (R)-4a, (S)-4b (R)- and (S)-5a, (R)-5b, (S,S')-6a, (S,R')-7a and (R,R')-8a] were established by chemical correlations with known compounds or by ORD and chemical inference. The ring opening of both the primary and secondary aziridinium ion positions of 1-azonia-1-ethylbicyclo [3.1.0]hexane [(S)-3a] by nucleophiles proceeds entirely by SN2 processes. The conversion of (R)-1-ethyl-3-hydroxypiperidine [(R)-5a] to (S)-2a. HCl with thionyl chloride in chloroform proceeds by inversion with 4.8% racemization, whereas the thermal rearrangement of (S)-1a to (R)-2a occurs with complete retention of absolute configuration.  相似文献

The structure, stability, and bonding character of some exemplary LAr and L-ArBeO (L = He, Ne, Ar, N2, CO, F2, Cl2, ClF, HF, HCl, NH3) were investigated by MP2 and coupled-cluster calculations, and by symmetry-adapted perturbation theory. The nature of the stabilizing interactions was also assayed by the method recently proposed by the authors to classify the chemical bonds in noble-gas compounds. The comparative analysis of the LAr and L-ArBeO unraveled geometric and bonding effects peculiarly related to the σ-hole at the Ar atom of ArBeO, including the major stabilizing/destabilizing role of the electrostatic interactionensuing from the negative/positive molecular electrostatic potential of L at the contact zone with ArBeO. The role of the inductive and dispersive components was also assayed, making it possible to discern the factors governing the transition from the (mainly) dispersive domain of the LAr, to the σ-hole domain of the L-ArBeO. Our conclusions could be valid for various types of non-covalent interactions, especially those involving σ-holes of respectable strength such as those occurring in ArBeO.  相似文献

An attempt was made to evaluate the possibility of creating and assessing the stability of inclusion complexes of selected phenolic acids [trans-4-hydroxycinnamic acid (trans-p-coumaric acid), trans-3,4-dihydroxycinnamic acid (trans-caffeic acid), trans-4-hydroxy-3-methoxycinnamic acid, (trans-ferulic acid) and trans-3-phenylacrylic acid (trans-cinnamic acid)] with β-cyclodextrin and 2-HP-β-cyclodextrin in aqueous solutions in a wide temperature range 283.15 K–313.15 K. On the basis of the values of the limiting molar conductivity CDNaDod), calculated from the experimental data, the values of the formation constants and the thermodynamic functions of formation (standard enthalpy, entropy, and Gibs standard enthalpy) of the studied complexes were determined. It has been found that the stability of the studied complexes increases with lowering of the molar mass of cyclodextrin and lowering of the temperature.  相似文献

An efficient synthesis of N-Boc-tubuphenylalanine benzyl ester (N-Boc-Tup-OBn, 1a) and N-Boc-epi-tubuphenylalanine benzyl ester (N-Boc-epi-Tup-OBn, 1b) is reported herein. Regioselective aziridine 4 ring opening with carbon nucleophiles followed by hydroboration of 1,1-substituted aminoalkene 3 using 9-BBN and subsequent oxidation in an alkaline medium are used as the key steps to provide N-tosyl 1,4-aminoalcohols. The 1,4-aminoalcohols are successfully transformed into the desired products with an overall yield of 23% for 1a and 11% for 1b over 8 consecutive steps separately.  相似文献

New cultivars of lavender adapted to arid steppe conditions were developed by the Institute of Rice of Ukrainian National Academy of Agrarian Sciences (NAAS). This work is a part of the characterization process of the new cultivars. The chemical composition of the essential oil of the seven new Lavandula angustifolia and eight new Lavandula x intermedia cultivars was investigated and compared. In total, 71 different compounds were identified. Linalool and linalool acetate were the main components in both species in ranges of 26.14–57.07% and 9.08–24.45%, respectively. They were followed by terpinen-4-ol (2.16–22.44%), lavandulyl acetate (2.12–10.23%), and lavandulol (1.30–3.14) in the case of L. angustifolia and camphor (10.11–12.55%), borneol (5.49–8.71%), and eucalyptol (0.47–7.41%) in the case of L. x intermedia. The oils had a valuable terpene profile—a high linalool content and the substantial presence of lavandulol and its ester. Nevertheless, they did not comply with the industry standards, mostly due to high levels of terpinene-4-ol. Evidently, a high content of terpinen-4-ol is a characteristic feature of L. angustifolia oils bred in Ukraine. Additionally, the LA3 cultivar yielded an oil with some of the highest linalool contents reported in the literature. Statistical analysis and literature data allowed for the comparative analysis of the gathered data. MANOVA, PCA, and HCA marked caryophyllene oxide as another potential differentiating compound between studied species.  相似文献

Inhibitors of biotin protein ligase (BPL) are novel antimicrobial compounds with the potential to treat infections caused by bacteria resistant to current antibiotics. A novel BPL inhibitor (12, Ki 1.4 μM) was synthesized from biotin acetylene and an azide-functionalized analogue of fluorescent nitrobenzofurazan by Cu(I) catalysed cycloaddition and also by template guided synthesis using wild-type BPL from Staphylococcus aureus. LC/HRMS-based detection provides improved sensitivity over previous reports using a mutant BPL, with demonstrated applicability to other BPLs. Super-imaging fluorescence microscopy demonstrated the accumulation of 12 in the cytoplasm of S. aureus, but not Escherichia coli. This novel fluorescent probe can be used to gain new insights into the mechanism of uptake, efflux and metabolism of BPL inhibitors in S. aureus.  相似文献

A new class of flavonoids bearing cyclic polyethers involving a phenyl ring was conveniently provided by the intramolecular photochemical dimerization of 2-chromonecarboxylic esters. Irradiation of 2-chromonecarboxylate with a polyether tethered at both ends promoted intramolecular [2 + 2] cyclobutane formation leading to 14- to 27-membered cyclic polyethers. The efficiency depended on the substituted position of the phenyl ring, with ortho- and meta-substituted derivatives giving cycloadducts in good chemical yields and quantum efficiencies, whereas the para-derivatives were inert toward photolysis. X-ray crystallographic analysis revealed that the stereochemistry of the macrocyclic cycloadducts exhibited C2-symmetry.  相似文献
