采用毛细管电泳柱端安培检测对脂可平胶囊中的姜黄素进行测定。着重研究了缓冲溶液浓度和酸碱度、检测电位、进样时间和高压对分离测定的影响。以微Pt电极为工作电极,电极电位为 1.0 V,以V(甲醇)∶V(乙醇)∶V(水)=5∶2∶3为非水介质,磷酸二氢钾和硼砂(pH 9.5)为缓冲体系,并用二阶样条小波进行滤波处理,姜黄素在1.0~120 mg/L范围内,峰高与其质量浓度呈良好的线性关系,线性回归方程:Y=20.2 146ρ,检出限为0.02 mg/L。  相似文献

以未涂层融硅石英毛细管(50 cm×75μm)为分离柱,2 mmol/L NaAc+2mmol/L HAc+0.5 mmol/L Cu2+(pH 4.0)作为电泳运行液,分离电压15 kV,建立了亮氨酸对映体的高效毛细管电泳-电导分离检测的方法。对缓冲溶液的种类、浓度、分离电压、有机改性剂等因素对分离的影响进行了讨论。L-亮氨酸和D-亮氨酸的线性回归方程分别为:y=5.998ρ+40.677,y=3.605ρ+42.087。线性范围:L-亮氨酸4.0~160 mg/L;D-亮氨酸6.0~160 mg/L。检出限分别为:1.5和2.0 mg/L。  相似文献

Summary A capillary electrophoretic method for the determination of thiosulphate, bromide, Fe(III)-EDTA chelate and free EDTA in spent fixing solutions has been developed. Free EDTA was complexed with Ni(II) ions prior to analysis. The optimised separations were carried out in a fused silica capillary (57 cm×75 μm I.D.) filled with a borate buffer (100mmol L−1 borate, 0.2 mmol L−1 tetradecyltrimethylammonium hydroxide, pH 8.5; applied voltage, −30kV) using direct UV detection at 214 nm. All four anions were well separated in less than 4 min. The method was applied to the rapid monitoring of spent fixing solutions.  相似文献

We describe a new method for the determination of methylglyoxal in water and biological matrices, using o-phenylenediamine as derivatizing agent and solid-phase extraction followed by capillary zone electrophoresis with diode array detection. 25 mM sodium phosphate running buffers at pH 2.2, 30 kV, and 25 degrees C allowed the best instrumental conditions for the optimum separation of methylglyoxal in a suitable analytical time (< 10 min), using an uncoated fused-silica capillary of 75 microm inner diameter and an effective length of 45.1 cm with an extended light path and the wavelength set to 200 nm. Under optimized instrumental conditions, good reproducibility of the migration time (< 1.1%), precision (< 5%), an excellent linear dynamic range from 0.1 to 3.6 mg/L (r(2) = 0.9997), and low limits of detection (7.2 microg/L) were obtained for methylglyoxal measurements, using the internal standard methodology. Assays on laboratory-spiked tap and ground water samples allowed a remarkable accuracy, presenting yields of 95.0 +/- 4.3 and 94.0 +/- 1.1%, respectively, and good performance to determine methylglyoxal in beer and yeast cells suspensions matrices was also obtained at trace level. The present methodology is a cost-effective alternative for routine quality control analysis, showing to be reliable, sensitive, and with a low sample volume requirement to monitor methylglyoxal in water and biological matrices.  相似文献

在熔融石英毛细管(50μmi.d×50cm)中,以6mmol/LH3BO3 2mmol/L硼砂 体积分数0.02%TEPA(四乙烯五胺)为电泳运行介质,分离电压-15.0kV,采用毛细管电泳-电导检测法在4min内实现了氯霉素的分离检测。讨论了电渗流抑制剂的浓度、缓冲体系的组成和浓度等因素对检测结果的影响。线性检测范围为1~120mg/L,检出限为0.3mg/L。对市售氯霉素滴眼液和注射液中的氯霉素进行分析,加标回收率为93.8%~106.8%。  相似文献

The paper presents a rapid method for the determination of dioxopromethazine hydrochloride (DPZ), an antihistamine drug, by the capillary electrophoresis with electrochemiluminescene detection (CE–ECL) using tris(2,2′-bipyridyl)ruthenium(II) (Ru(bpy)32+) reagent. This CE–ECL detection method has high sensitivity, good selectivity and reproducibility for DPZ analysis. Under the optimized conditions: separation capillary, 38 cm length (25 μm i.d.); sample injection, 10 s at 8 kV; separation voltage, 12.5 kV; running buffer, 20 mmol L−1 sodium phosphate of pH 6.0; detection potential, 1.15 V; 50 mmol L−1 of phosphate buffer (pH 7.14) containing 5 mmol L−1 of Ru(bpy)32+ in ECL detection cell, the detection limit of DPZ was 0.05 μmol L−1 (S/N = 3). The linear range extended from 5 to 100 μmol L−1. The linear curve obtained was Y = 181.62 + 9.28X with a correlation coefficient of 0.9970. The relative standard deviations of the ECL intensity and the migration time for six continuous injections of 5 μmol L−1 DPZ were 3.7% and 0.92%, respectively. The CE–ECL method was applied to analyze DPZ in real samples including tablets, rat serum and human urine, and satisfactory results were obtained without interference from samples matrix. The CE–ECL technique was proved to be a potential method for the detection of DPZ in clinic analysis.  相似文献

The potential of CE with native fluorescence detection (Flu) for the profiling of the therapeutic protein erythropoietin (EPO) was studied. EPO is a highly heterogeneous glycoprotein comprising a large number of isoforms. CE was applied to induce separation among the various glycoforms. Native Flu of EPO provided high detection selectivity yielding good signal‐to‐noise ratios and stable baselines, particularly when compared to conventional UV absorbance detection. In order to enhance EPO isoform resolution, CE was performed using a capillary with a neutral coating in combination with a simple BGE of 2.0 M acetic acid (pH 2.1). CE‐Flu analysis of the EPO biological reference preparation of the European Pharmacopeia resulted in a highly detailed glycoform profile. Migration time RSDs for selected EPO isoforms were less than 0.22% and 0.80% for intraday and interday repeatability, respectively. RSDs for relative peak intensity of the major EPO isoforms were less than 3%. The achieved resolution, migration time stability, and sensitivity allowed discrimination of different EPO products (EPO‐α and EPO‐β) based on the recorded glycoform pattern. The developed CE‐Flu method is relatively straightforward, and shows potential for quality control in biopharmaceutical production.  相似文献

A simple method based on capillary electrophoresis with potential gradient detection was developed to separate and detect neomycin components within 4 min without a derivatization step. Satisfactory separation and good repeatability were obtained using a separation buffer composed of 1 mM ammonium citrate (pH 3.5). The linearity of the method ranged from 10 to 1000 ppm with a limit of detection for neomycin B of about 7 ppm. After a simple dilution and filtering pretreatment step, neomycin components in three real samples were successfully analyzed without any major interference. Due to its simplicity and reliability, this method could provide an excellent alternative to the assays currently listed in U.S. and European Pharmacopoeia. The experiments were performed on a portable capillary electrophoresis system and, hence, the method can be readily applied to field analysis and point-of-care testing. 采用毛细管电泳电化学检测法同时测定了烟草中的多元酚,即芦丁、绿原酸,槲皮素和咖啡酸。考察了工作电极的氧化电位、运行缓冲溶液浓度和pH值,分离电压和进样时间对分离和检测的影响。在优化条件下,以300μm直径的碳圆盘电极为工作电极,检测电位为+0.9 V(vs.SCE),在50 mmol/L硼酸盐(pH 8.4)的运行缓冲液中,被测物浓度与峰电流在三个数量级范围内呈良好线性,检出限为2×10-7或5×10-7g/mL。方法有着良好的重现性,被测组分的迁移时间和峰高的相对标准偏差(RSDs)小于4%(n=7)。单次测定可在16 min内完成,已用于实际样品多元酚的测定,样品处理简单,无须预富集。  相似文献

Summary Perchlorate is a groundwater contaminant originating from facilities that manufacture and test solid rocket fuel. A new technology, capillary electrophoresis, has the potential to measure perchlorate rapidly and inexpensively in water samples. With its speed and simplicity, this method would complement existing methods. The perchlorate anion is routinely detected in water samples using high performance ion exchange chromatography, a very sensitive yet time consuming and expensive method. In this work, the parameters for detection of perchlorate are optimized to permit detection of 0.400 mgL−1 perchlorate in a standard solution. The usefulness of this technology is demonstrated for measuring perchlorate in several ground-water samples from the Western United States. The results demonstrate that CE can be used to rapidly screen environmental samples for perchlorate at intermediate to high levels (greater than 0.400 mgL−1). This technique allows faster, easier screening of potential contamination sites and could complement the use of ion exchange chromatography for groundwater testing.  相似文献

Capillary zone electrophoresis with electrochemical detection (ED) has been employed for the separation and determination of adenine (A), guanine (G), theophylline (Thp), hypoxanthine (HX), xanthine (Xan) and uric acid (UA). Effects of several important factors such as the acidity and concentration of running buffer, separation voltage, injection time and detection potential were investigated to acquire the optimum conditions. The detection electrode was a 300 μm carbon disc electrode at a working potential of +0.95 V (versus saturated calomel electrode (SCE)). The six purine bases can be well separated within 14 min in a 40 cm length fused-silica capillary at a separation voltage of 10 kV in a 100 mmol/l borate buffer (BB, pH 10.0). The current response was linear over about three orders of magnitude with detection limits (S/N=3) ranging from 0.157×10−6 to 0.767×10−6 mol/l for all compounds. The proposed method was successfully applied to determine Thp in tea and aminophylline tablets, UA in human urine, and two purine bases in DNA.  相似文献

采用毛细管电泳-电化学检测法(CE-ED),对饲料中的6种磺胺类药物磺胺脒、磺胺二甲嘧啶、磺胺甲嘧啶、磺胺二甲氧嘧啶、磺胺嘧啶、磺胺甲恶唑进行了分离和测定。分别考察了工作电极电位、运行缓冲液的pH和浓度、分离电压和进样时间等实验参数对实验结果的影响。在优化的实验条件下,以直径300μm的碳圆盘电极为工作电极,检测电位为0.95 V(vs.SCE),在30 mmol/L硼砂-KH2PO4(pH7.6)的运行缓冲溶液中,6个分析物能够在16 min内实现很好的基线分离,被测物浓度与峰电流在3个数量级呈良好的线性,检出限(S/N=3)范围0.08~0.20μg/mL。该方法已应用于实际样品的分析。  相似文献

The present study illustrates the possibilities of capillary isotachophoresis (CITP) online coupled with capillary zone electrophoresis (CZE) and hyphenated with fiber-based spectrophotometric diode array detection (DAD) for the direct, highly reliable, and ultrasensitive determination of quinine (QUI) in real multicomponent ionic matrices (beverages). Here, the CITP provided an effective online sample pretreatment (preseparation and preconcentration) prior to the CZE separation. Due to the CITP sample preconcentration, a simple UV-visible absorbance spectrophotometric detection was sufficient for obtaining very low concentration limits of detection (~2.3 ng/mL). Enhanced separation selectivity due to the combination of different separation mechanisms (CITP vs. CZE) enabled to obtain a pure analyte zone, suitable for its detection and quantitation in the directly injected real samples. The spectrophotometric DAD, unlike single wavelength UV detection, enabled to characterize the purity (i.e. spectral homogeneity) of the analyte zone and preliminary data indicate structurally related compounds via characteristic spectra recorded in the interval of 200-600 nm. The proposed CITP-CZE-DAD method was characterized by favorable performance parameters (sensitivity, linearity, precision, recovery, accuracy, robustness, and selectivity) and successfully applied to the control of QUI and potential QUI impurities in commercial beverages. This method is proposed as a routine automatized method for the highly reliable quality food control.  相似文献

BRAF mutations are found in many human tumors, namely melanomas ( approximately 70%) and colon carcinomas ( approximately 15%). This paper presents a method for identification of exon 15 BRAF mutations by denaturant capillary electrophoresis (CE), an analysis method that is sensitive, cost-effective (involving only polymerase chain reaction (PCR) and electrophoresis) and capable of high-throughput screening. In total, we found 21 (70%) out of 30 melanoma cell lines with BRAF mutations in exon 15: two of which were the p.Val600Asp (c.1799-800TG>AT) mutation, one cell line contained the p.Val600Arg (c.1798-99GT>AG) mutation, and 18 cell lines contained the p.Val600Glu (c.1799T>A) mutation. Of the nine cell lines that did not contain a BRAF mutation, five contained an NRAS mutation at exon 2, and no mutations were detected in NRAS exon 1. There was no overlap of NRAS and BRAF mutations in the same cell line. In addition, we looked at 221 colon biopsy samples and identified one further BRAF mutation, the p.Asp594Gly (c.1781A>G) mutation, in seven samples. The p.Val600Glu mutation was identified in 11 of the colon biopsy samples. Using the four mutations of BRAF exon 15, we then constructed a denaturing CE standard capable of distinguishing between each of the mutations; therefore, sequencing does not need to be performed to confirm the mutation. In conclusion, this sensitive, cost-effective mutation assay for BRAF (and RAS) will provide the opportunity to detect and determine mutations without the need to purify samples for sequencing. Future large-scale studies will provide the clinical usefulness of such mutations.  相似文献

运用毛细管电泳-电导检测方法对4种四环素衍生物——土霉素(OTC)、金霉素(CTC)、强力霉素(DOC)和四环素(TC)的分离进行了研究。在3.5mmol/L三羟基氨基甲烷(Tris)-7.5mmol/L柠檬酸(Cit)pH4.0的运行缓冲液中,4种四环素衍生物在15min内获得完全分离。四环素衍生物的线性范围分别为5.0-500μg/mL OTC,3.6-420μg/mL CTC,4.5-470μg/mL DOC和2.5-400μg/mL TC。检测限(S/N=3)分别为OTC2.0μg/mL,CTC 1.8μg/mL,DOC2.5μg/mL和TC1.0μg/mL。采用本法对实际样品强力霉素片中强力霉素和土霉素片中土霉素进行测定,回收率分别为97.2%和96.4%。  相似文献

An electrophoretically mediated microanalysis method for detection of yoctomole (ymol) alkaline phosphatase (ALP) was developed by a combination of on-capillary enzyme-catalyzed reaction and electrochemical detection. In this method, ALP molecules were electrokinetically injected into a capillary of 10 μm i.d. and then electromigrated to the section of the capillary immersed in a warm water bath of 37 °C, where ALP reacted for a certain time with disodium phenyl phosphate as the enzyme substrate. ALP could be measured through determining the electroactive product phenol of the enzyme-catalyzed reaction by using electrochemical detection. The phenol concentration was proportional to the mass of ALP. As a catalyst, ALP was not consumed during the reaction, which provided amplification of signal with prolonged the reaction time. In order to enhance the signal-to-noise ratio, the detection end of the capillary was etched to a horn-shape and a single carbon fiber microcylinder electrode of 6 μm in diameter as the working electrode was inserted into the detection end of the capillary. Under these conditions, the mass of ALP as low as 1.2 × 10−22 mol (72 molecules) or 4.0 × 10−23 mol (24 molecules) could be detected for the on-capillary reaction time of 15 min or 2 h.  相似文献

A high-density 384-lane microfabricated capillary array electrophoresis device is evaluated for high-throughput single-strand conformation polymorphism (SSCP) analysis. A delayed back bias direct electrokinetic injection scheme is used to provide better than 10-bp resolution with an 8.0-cm effective separation length. Separation of a HaeIII digest of PhiX174 yielded theoretical plate numbers of 4.0 x 10(6). Using 5% PDMA containing 10% glycerol and 15% urea, 21 single-nucleotide polymorphisms (SNPs) from HFE, MYL2, MYL3, and MYH7 genes associated with hereditary hemochromatosis (HHC) and hereditary hypertrophic cardiomyopathy (HCM) are discriminated at two running temperatures (25 degrees C and 40 degrees C), providing 100% sensitivity. The data in this study demonstrate that the 384-lane microCAE device provides the resolution and detection sensitivity required for SSCP analysis, showing its potential for ultrahigh-throughput mutation detection.  相似文献

We present a fast detection of M467T, the major mutation causing cystinuria, by capillary electrophoresis version of single-strand conformation polymorphism (SSCP). The DNA fragment (317 bp) carrying the point mutation was amplified by polymerase chain reaction (PCR) on the exon 8 of the SLC3A1 gene, which encodes for the transmembrane glycoprotein rBAT, a part of the active cystine and dibasic amino acids transporter. The complementary strands of the fragment were labeled by fluorescein and TAMRA, respectively. Thus, the electromigration of both strands was recorded independently as a laser-induced fluorescence (LIF) signal, what enabled an effective optimization of separation conditions. The injected sample was denatured by immersing the inlet of the separation capillary into a vial with 0.1 M solution of NaOH prior to analysis. Under optimum conditions, the SSCP analysis in poly(vinyl alcohol) (PVA)-coated silica-fused capillary of an effective length of 15 cm, filled with 4% linear polyacrylamide (LPA) solution, was accomplished in approximately 6 min. The experimentally observed mobility shifts of single-stranded DNA (ssDNA) fragments were compared to the appearance of their calculated two-dimensional conformations using Version 3.0 of MFOLD software. The number of nucleotides involved in the duplex regions of theoretical structures correlates well with their real migration order in the sieving medium.  相似文献

A capillary electrophoretic method with UV detection for separation and quantitation of perfluorocarboxylic acids (PFCAs) from C6-PFCA to C12-PFCA has been developed. The optimization of measurement conditions included the choice of the most appropriate type and concentration of buffer in the background electrolyte (BGE), as well as the type and the content of an organic modifier. The optimal separation of investigated PFCAs was achieved with 50 mM phosphate buffer and 40% isopropanol in the BGE using direct UV detection. The optimum wavelength for direct UV detection was optimized at 190 nm. For indirect detection, several chromophores were studied. Five mM 3,5-Dinitrobenzoic acid (3,5-DNBA) in 20 mM phosphate buffer BGE and indirect UV detection at 280 nm gave the optimal detection and separation performance for the investigated PFCAs. The possibility of on-line preconcentration of solutes by stacking has been examined for indirect detection. The detection limits (LODs) determined for direct UV detection ranged from 2 microg/mL for C6-PFCA to 33 microg/mL for C12-PFCA. The LODs obtained for indirect UV detection were comparable to those obtained for direct UV detection.  相似文献
