The quantitative and confirmative performance of two different mass spectrometry (MS) techniques (high-resolution MS and tandem MS) was critically compared. Evaluated was a new extraction and clean-up protocol which was developed to cover more than 100 different veterinary drugs at trace levels in a number of animal tissues and honey matrices. Both detection techniques, high-resolution mass spectrometry (HRMS) (single-stage Orbitrap instrument operated at 50 000 full width at half maximum) and tandem mass spectrometry (MS/MS) (quadrupole technology) were used to validate the method according to the EU Commission Decision 2002/657/EEC. Equal or even a slightly better quantitative performance was observed for the HRMS-based approach. Sensitivity is higher for unit mass resolution MS/MS if only a subset of the 100 compounds has to be monitored. Confirmation of suspected positive findings can be done by evaluating the intensity ratio between different MS/MS transitions, or by accurate mass based product ion traces (no precursor selection applied). MS/MS relies on compound-specific optimized transitions; hence the second, confirmatory transition generally shows relatively high ion abundance (fragmentation efficacy). This is often not the case in single-stage HRMS, since a generic (not compound-optimized) collision energy is applied. Hence, confirmation of analytes present at low levels is superior when performed by MS/MS. Slightly better precision, but poorer accuracy (fortified matrix extracts versus pure standard solution) of ion ratios were observed when comparing data obtained by HRMS versus MS/MS.  相似文献

In order to deepen the understanding of the metabolism of Ganstigmine, a new acetylcholinesterase inhibitor under evaluation for the treatment of Alzheimer's disease, samples obtained by incubating the drug with female rat hepatocytes were investigated by low-resolution liquid chromatography/tandem mass spectrometry (LC/MS/MS). The results confirmed the formation of most of the phase I metabolites already demonstrated, but also three new species. The combination of high-resolution quadrupole time-of-flight (Q-TOF) LC/MS and LC/MS/MS measurements, and the evaluation of the more reasonable metabolic routes, allowed the identification of the new metabolites as Geneseroline-glucuronide and oxidized and rearranged Ganstigmine. Analogous investigations were made using hepatocytes from male rat and dog, and both gender monkeys and humans, to compare the metabolic patterns. The results did not indicate substantial differences in terms of numbers and abundances of detected metabolites among the considered species, and also between male and female hepatocytes within each species.  相似文献

The quantitative determination of nucleotides from DNA modified by styrene oxide is described using a combination of inductively coupled plasma high-resolution mass spectrometry (ICP-HRMS) and electrospray ionization mass spectrometry (ESI-MS), both interfaced to reversed-phase high-performance liquid chromatography (HPLC). LC/ICP-MS (resolution > 1500 to discriminate against 15N16O+ and 14N16OH+) was employed to determine quantitatively the content of modified nucleotides in standard solutions based on the signal of phosphorus; phosphoric acid served as an internal standard. By means of the standard addition technique the sensitivity of the LC/ESI-MS approach was subsequently determined. Since a comparison of UV, ICP and ESI-MS data suggested that in ESI-MS the ionization efficiency of the adducts is identical within the error limits, quantitative determination of all adducts is possible. For LC/ESI-MS with single ion monitoring, the detection limit for styrene oxide adducts of nucleotides was determined to be 20 pg absolute or 14 modified in 10(8) unmodified nucleotides in a 5 micrograms DNA sample, which comes close to the best methods available for the detection of chemical modifications in DNA.  相似文献

Quantitative determination was made of the iron-containing protein myoglobin in a range of different foods, including meat, processed meat, fish, and shellfish, by liquid chromatography coupled to a double-focusing sector field inductively coupled plasma mass spectrometry (ICP-MS). The concentration of myoglobin determined in the samples ranged from 0 to 6.5 mg/kg, and the analytical precision (coefficient of variation) for the analysis of 8 replicate raw steak extracts was 2.1%. By using a double-focusing ICP-MS instrument, direct on-line detection of the most abundant iron isotope 56Fe was possible without interference from a major polyatomic interference (40Ar16O). Separation of myoglobin from other iron-containing compounds was facilitated by use of a gel filtration column (TSK Gel G2000SW) and Tris buffer (pH 7.2). The chromatographic column was coupled directly to the nebulizer of the ICP-MS instrument by a short piece of PEEK tubing. To ensure sufficient quality control throughout the study, a raw beefsteak sample was developed as an in-house reference material. The concentration of the heme-iron-containing protein myoglobin in this sample was determined by the developed method and independently by a conventional spectrophotometric method. The agreement between the 2 analytical techniques was very good. The detection limit (3 times the signal/noise ratio for a blank) of the reported method for myoglobin was 0.85 ng Fe/L.  相似文献

The photodecomposition of imazamox, a herbicide of the imidazolinone family, was investigated in pure water. The main photoproducts from the photolysis were followed over time by liquid chromatography mass spectrometry and structures were proposed from exact mass determinations obtained by electrospray ionization Fourier transform ion cyclotron resonance mass spectrometry. The method comprised exact mass determination with better than 0.2 ppm mass accuracy and a corresponding structural visualization taking care of respective isotopes with an adapted van Krevelen diagram that enabled a systematic approach to the characterisation of the elementary composition of each photoproduct. By taking advantage of the high resolving power of FT-ICR MS to make precise formula assignments, the derived 2D van Krevelen diagram (O/C; H/C; m/z) enabled one to structurally differentiate the formed photoproducts and to propose a degradation pathway for imazamox. Some twenty cultured fish samples were analyzed for possible residues of veterinary drugs with high resolution mass spectrometry (single stage Orbitrap) coupled to ultra performance liquid chromatography. Quantitative analysis based on external standards covered 110 analytes. Some 116 additional compounds were monitored without having access to reference materials. Detection was based on calculated exact masses and narrow mass windows. Furthermore, a number of semi-targeted techniques were evaluated and compared to corresponding triple quadrupole precursor scan experiments. Single stage high resolution mass spectrometry was used to monitor compound specific product ions (without relying on a previous precursor selection). The capabilities of neutral loss searches based on exact masses were shown by detecting small concentrations of incurred oxytetracycline residues. High resolution mass spectrometry provided more sensitivity and selectivity than corresponding tandem quadrupole precursor and neutral loss scans. The currently limiting factor is the not adequate performance of the available software used for data mining. The high number of false positives that were produced, when searching for chlorine isotopic patterns, was clearly linked to the fact that the utilized software does not perform a peak deconvolution, but simply investigates one individual spectrum after another.  相似文献

Ion chromatography (IC) coupled with inductively coupled plasma mass spectrometry (ICP-MS) was systematically investigated for determining the speciation of chromium in environmental samples. Firstly, the stability of complexes formed by Cr(III) with various aminopolycarboxylic acids was studied by electrospray ionization mass spectrometry (ESI-MS). The results showed that [Cr(EDTA)] was stable in solution. Secondly, various mobile phases were examined to separate Cl from chromium species by IC to avoid Cl interference. The separation of [Cr(EDTA)] and Cr(VI) was achieved on a new anion-exchange column (G3154A/102) using a mobile phase containing 20 mM NH4NO3 and 10 mM NH4H2PO4 at pH 7.0 without Cl interference. Detection limits for chromium species were below 0.2 μg/L with a direct injection of sample and without prior removal of interferences from the matrix. Finally, the proposed method was used for the determination of chromium species in contaminated waters.  相似文献

Capillary liquid chromatography (LC) using a 320 microns column and a flow rate of 10 microL/min has been coupled to an ion trap mass spectrometer using electrospray ionisation (ESI) to enable the rapid and effective identification of metabolites in urine, following oral administration of a novel human neutrophil elastase inhibitor, GW311616. Metabolites were identified from their mass (MS) spectra and tandem (MS/MS) mass spectra using minimal sample (1 microL of urine) and no sample pretreatment. Sensitivity assessment has shown that both molecular weight and structural information is obtainable on as little as 5 pg of compound, making the capillary LC/ion trap system as described an ideal analytical tool for the detection and characterisation of low level metabolites in biofluids (particularly when sample volume is limited). This level of detection was unattainable using a triple quadrupole mass spectrometer operating in full-scan mode, although 200 fg on column was detected using selected reaction monitoring target analysis.  相似文献

A method based on ion chromatography (IC) and inductively coupled plasma MS (ICP-MS) was developed for the speciation of arsenic in water and soil extracts. An anion-exchange column (G3154A/101) was used to separate As(III), As(V), dimethylarsinic acid (DMA), and monomethylarsonic acid (MMA) with excellent resolution. Various ammonium salts, including NH4H2PO4, (NH4)2HPO4, (NH4)2CO3, and NH4HCO3, were examined as eluents to reduce matrix interference from chloride and to solve clogging problems. The best arsenic speciation was obtained within 9 min with excellent resolution and without interference from high chloride concentrations using an eluent containing 7.5 mM (NH4)2HPO4 at pH 7.9. The detection limits for the target arsenic species ranged from 0.1 to 0.4 microg/L with direct injection of sample without matrix elimination. The proposed method was effectively demonstrated by determining arsenic species in contaminated waters and soils of Bangladesh.  相似文献

The coupling of ultra-performance liquid chromatography, operating at elevated pressures, to a linear ion trap mass spectrometer provides a high-performance system suitable for drug metabolite characterisation. This system demonstrates improved chromatographic efficiency and sensitivity and at the same time provides diagnostic MSn data often critical for metabolite structural assignment. The linear ion trap was capable of dealing with the high chromatographic efficiencies and hence narrow peak widths associated with 1.7 microm particle-packed column separations. Polarity switching and data-dependent MSn data were generated with ease, and applied to the identification of metabolites found in human plasma.  相似文献

A precise and reliable method, using liquid chromatography combined with ion trap based mass spectrometry, for the determination of three endogenous estrogens, namely, estrone, estradiol, and estriol, and two synthetic estrogens, ethinyl estradiol and diethylstilbestrol, in environmental water samples was developed. Optimization of the parameter settings of the ion source and mass analyzer as well as evaluation of solvent composition were carried out by continuous introduction of standards through a syringe pump. In negative ion mode the electrospray ionization source gave acceptable results. The optimum solvent used consisted of water/acetonitrile, with no volatile bases or buffers added. A simple, off-line, manual solid-phase extraction method was developed for sample preparation of environmental water samples. Recoveries were over 86% for all compounds. The method was validated and found to be linear, selective, and robust. For analysis of a 50-mL sample, the limit of detection (LOD) ranged from 3.2 to 10.6 ng/L for all compounds, and the limit of quantitation (LOQ) from 10.6 to 35.0 ng/L. Within-day (n = 5) and total (n = 5) reproducibility were investigated at three different concentration levels and ranged from 6.2 to 9.5% and 9.4 to 12.1%, respectively. Resveratrol is an antitoxin secreted by plants such as Polygonum cuspidatum Sieb. et Zucc and Vitis vinifera L. when they are attacked by pathogens. In the present study, three methods were used to prepare biological samples, and then an efficient strategy based on ultra-high-performance liquid chromatography-linear ion trap-Orbitrap mass spectrometry (UHPLC-LTQ-Orbitrap MS) was developed to screen and identify resveratrol metabolites in rat urine, plasma and faeces. As a result, a total of 56 resveratrol metabolites were screened and characterized. Among them, 39 metabolites were found in rat urine, while 6 and 16 metabolites were characterized from rat plasma and faeces, respectively. In addition, 56, 12 and 15 metabolic products were screened by solid phase extraction (method I), methanol precipitation (method II) and acetonitrile precipitation (method III), respectively, indicating that method I could be adopted as the most acceptable method. The results also demonstrated that resveratrol mainly underwent glucuronidation, glucosylation, sulfation, hydroxylation, dehydrogenation, hydrogenation, methylation and their composite reactions. Moreover, these metabolic reactions occurred to form a possible metabolic network that is similar to a triangular pyramid model. In summary, this research provides an idea for the further study of drug metabolism.  相似文献

The identification and structure elucidation of drug metabolites is one of the main objectives in in vitro ADME studies. Typical modern methodologies involve incubation of the drug with subcellular fractions to simulate metabolism followed by LC-MS/MS or LC-MS(n) analysis and chemometric approaches for the extraction of the metabolites. The objective of this work was the software-guided identification and structure elucidation of major and minor buspirone metabolites using capillary LC as a separation technique and ion trap MS(n) as well as electrospray ionization orthogonal acceleration time-of-flight (ESI oaTOF) mass spectrometry as detection techniques.Buspirone mainly underwent hydroxylation, dihydroxylation and N-oxidation in S9 fractions in the presence of phase I co-factors and the corresponding glucuronides were detected in the presence of phase II co-factors. The use of automated ion trap MS/MS data-dependent acquisition combined with a chemometric tool allowed the detection of five small chromatographic peaks of unexpected metabolites that co-eluted with the larger chromatographic peaks of expected metabolites. Using automatic assignment of ion trap MS/MS fragments as well as accurate mass measurements from an ESI oaTOF mass spectrometer, possible structures were postulated for these metabolites that were previously not reported in the literature.  相似文献

Traditional Chinese medicine (TCM) is commonly considered to operate due to the synergistic effects of all the major and minor components in the medicines. Hence sensitive and comprehensive analytical techniques are needed to acquire a better understanding of the pharmacological basis of the herb and to enhance the product quality control. The present review mainly focuses on the phytochemical analysis of TCMs using high-performance liquid chromatography coupled with mass spectrometry (HPLC-MS). Atmospheric pressure chemical ionization (APCI) and electrospray ionization (ESI) are the two commonly used ion sources. Triple quadrupole, ion trap (IT), Fourier transform ion cyclotron resonance (FTICR) and time-of-flight (TOF) mass spectrometers are used as on-line analyzer. The relationship between structural features and fragmentation patterns should be investigated as thoroughly as possible and hence be applied in the on-line analysis to deduce the structures of detected peaks. Characteristic fragmentation behaviors of the reference standards, as well as information regarding polarity obtained from retention time data, on-line UV spectra, data from the literature and bio-sources of the compounds allowed the identification of the phytochemical constituents in the crude extracts. Although a mass spectrometer is not a universal detector, high-performance liquid chromatography coupled with multistage mass spectrometry (HPLC-MS(n)) technique was still proved to be a rapid and sensitive method to analyze the majority of the many constituents in herbal medicines, particularly for the detection of those present in minor or trace amounts. The methods established using HPLC-MS techniques facilitate the convenient and rapid quality control of traditional medicines and their pharmaceutical preparations. The combination of suppressed and non-suppressed cation-exchange chromatography with electrospray ionization mass spectrometry was demonstrated for the structural elucidation of unknown by-products (mostly quaternary ammonium compounds) in a new cholesterol-reducing drug. The suppressed mode using methanesulfonic acid and acetonitrile turned out to be unsuitable because of regenerant (tetrabutylammonium hydroxide) passing through the membrane of the suppressor into the eluent which led to a significant increase of spectral background in the mass spectrometer. Employing a mobile phase consisting of 200 mM formic acid and 60% (v/v) acetonitrile, the separation and detection of 8 unknown compounds was possible in the non-suppressed mode. The three most prominent compounds were selected for structural elucidation utilizing collision induced dissociation experiments. In a series of experiments the fragmentation behavior was investigated for different fragmentation voltages finally leading to structure proposals. Using gas chromatography hyphenated with mass spectrometry, additional information for the structure of the unknowns was collected and a possible way of their formation was proposed.  相似文献

A simple and efficient multiresidue method using dispersive solid phase extraction and liquid chromatography coupled with tandem mass spectrometry was developed for the targeted analysis of indaziflam and its five metabolites (indaziflam‐diaminotriazine, indaziflam‐carboxylic acid, indaziflam‐triazine indanone, indaziflam‐hydroxyethyl, and indaziflam‐olefin) in pitaya samples (including roots, plants, flowers, peels, pulp, and whole fruit). The analytes were extracted with acetonitrile, and the extracts were purified using multiwalled carbon nanotubes. The method was validated using pitaya samples spiked at 0.5, 5, and 50 µg/kg, and the average recoveries varied from 61.1 to 103.7% with relative standard deviations lower than 12.7% (= 5). This method exhibited sufficient linearity within the concentration range of 0.1–100 µg/L. The limits of detection and quantification were in the ranges of 0.001–0.1 and 0.003–0.3 µg/kg, respectively. The method was successfully applied to analyze pitaya samples in Nanning, and no indaziflam or its metabolites were detected in the samples analyzed.  相似文献

A strategy to identify metabolites of a marine biotoxin, 13-desmethyl spirolide C, has been developed using liquid chromatography coupled to high-resolution mass spectrometry (LC/HRMS). Metabolites were generated in vitro through incubation with human liver microsomes. A list of metabolites was established by selecting precursor ions of a common fragment ion characteristic of the spirolide toxin which was known to contain a cyclic imine ring. Accurate mass measurements were subsequently used to confirm the molecular formula of each biotransformation product. Using this approach, a total of nine phase I metabolites was successfully identified with deviations of mass accuracy less than 2 ppm. The biotransformations observed included hydroxylation, dihydroxylation, oxidation of a quaternary methyl group to hydroxymethyl or carboxylic acid groups, dehydrogenation and hydroxylation, as well as demethylation and dihydroxylation reactions. In a second step, tandem mass spectrometry (MS/MS) was performed to elucidate structures of the metabolites. Using the unique fragment ions in the spectra, the structures of the three major metabolites, 13,19-didesmethyl-19-carboxy spirolide C, 13,19-didesmethyl-19-hydroxymethyl spirolide C and 13-desmethyl-17-hydroxy spirolide C, were assigned. Levels of 13-desmethyl spirolide C and its metabolites were monitored at selected time points over a 32-h incubation period with human liver microsomes. It was determined that 13,19-didesmethyl-19-carboxy spirolide C became the predominant metabolite after 2 h of incubation. The stability plot of 13-desmethyl spirolide C showed first-order kinetics for its metabolism and the intrinsic clearance was calculated to be 41 μL/min/mg, suggesting first-pass metabolism may contribute to limiting oral toxicity of 13-desmethyl spirolide C.  相似文献

Humans can be exposed to mycotoxins through the food chain. Mycotoxins are mainly found as contaminants in food and could be subsequently excreted via biological fluids such as urine or human breast milk in native or metabolised form. Since breast milk is usually supposed as the only food for new-borns, the occurrence of mycotoxins in thirty-five human milk samples was evaluated by a newly developed method based on QuEChERS extraction and UHPLC–HRMS detection. The method described here allows the detection of target mycotoxins in order to determine the quality of this initial feeding. The method has been fully validated, with recoveries ranging from 64% to 93% and relative standard deviations (RSD, %) being lower than 20%. Using the method described, non-metabolised mycotoxins such as ZEA, NEO, NIV, ENA, ENA1, ENB, ENB1 and metabolites, such as ZEA metabolites, HT-2, DOM and T-2 triol were detected in human milk samples. Results obtained help to estimate the exposure of mothers and infants to mycotoxins. Moreover, to the best of our knowledge, this is the first work describing the simultaneous detection, quantification and screening of mycotoxins and their metabolites in human mature milk.  相似文献
