Photocyclization reactions were carried out on ethyl 2-(8-oxo-5,6,7,8-tetrahydro-1-naphthyloxy)acetates 1a-e and ethyl 2-(5-oxo-6,7,8,9-tetrahydro-5H-benzocyclohepten-4-yloxy)acetates 2a-e in acetonitrile. Irradiation of 1a-e gave naphtho[1,8-bc]furanols 3a-e and naphtho[1,8-bc]furans 4a-e in 33–83% yields and ethyl acrylates 5b-d were produced in 3–25% yields during irradiation of 1b-d . On the other hand, 2a-e afforded cyclohepta[ad|benzofuranols 6a-e and cyclohepta[ad]benzofurans 7a-e in 44–87% yields. Ethyl acrylates 8b-d were also produced in 7–43% yields from irradiation of 2b-d . Substituent effects on photocyclization and reaction pathways are discussed.  相似文献

Five new 1-(2-(5-nitrofuran-2-yl)-5-(aryl)-1,3,4-oxadiazol-3-(2H)-yl) ethanone compounds 5a-e were synthesized by cyclization of N-acylhydrazones 4a-e with acetic anhydride under reflux conditions. Their structures were fully characterized by IR, 1H-NMR, and 13C-NMR. Furthermore, evaluations of the antibacterial activity of the 1,3,4-oxadiazoles 5a-e and N-acylhydrazones 4a-e showed strong activity against several strains of Staphylococcus aureus, with MICs between 4 μg/mL to 32 μg/mL. In silico studies of the parameters of Lipinski's Rule of Five, as well as the topological polar surface area (TPSA), absorption percentage (% ABS), drug likeness and drug score indicate that these compounds, especially 4a and 5d, have potential to be new drug candidates.  相似文献

A one-pot synthesis of 1-methyl- and 1-phenylpyrazole-3(5)-ethyl esters 2,3a-e by the cyclocondensation of β-alkoxyvinyl trichloromethyl ketones 1a-e with methyl and phenyl hydrazine hydrochloride under mild conditions, is reported. A study using compounds 1a-e with different substituents proved that these are versatile building blocks for the synthesis of pyrazole derivatives, having a 3(5)-ethoxycarbonyl substituent in good yields (60–89%). The hydrazine and β-alkoxyvinyl trichloromethyl ketone substituent effects on the reaction regiochemistry on the formation of the 1,3- and 1,5-isomer were observed.  相似文献

Synthesis of the first oxaphosphirane chromium(0) and molybdenum(0) complexes of the type [{(R(1)PCH(R(2))-O}M(CO)(5)] (R(1) = C(5)Me(5)) (8a-e, 15a-e) and (R(1) = CH(SiMe(3))(2)) (9a-e, 16a-e) via reaction of dichloro(organo)- (1, 2, 10, 11) and chloro(organo)phosphane complexes (3,4,12) with lithium bases and aldehydes 7a-e is reported. Furthermore, bicyclic 1,3-oxaphospholane complexes 17 and 18 have been obtained via O-protonation of oxaphosphirane complexes 8a and 15a with HCl. All complexes were characterized by NMR, IR spectroscopic, mass spectrometric investigations and, in addition, single-crystal X-ray structures of complexes 8a-e, 9a,c, 15a,b,e, 16a-c, 17, 18 are presented and discussed.  相似文献

Reaction of methyl benzoylphosphonochloridate (3) with a secondary or primary series of amines yielded methyl benzoylphosphonamidates, 4a-e. The latter compounds reacted with hydroxylamine to yield a-hydroxyiminobenzylphosphonamidates (5a-e), largely as (E)-isomers. The structure of methyl (E)-a-hydroxyimino-benzyl-1-pyrrolidinylphosphinate (5b) was determined by single-crystal X-ray crystallography. Heating oximes 5a-e in boiling toluene caused them to undergo Beckmann rearrangement to N-benzoylphosphordiamidates 6a-e. © 1996 John Wiley & Sons, Inc.  相似文献

[structure: see text] A synthetic strategy promising the establishment of a new star-shaped and linear polycyclic aromatic hydrocarbons (PAHs) family with distinct molecular topologies has been developed. The Sonogashira reaction between the iodide derivatives 2a-e and phenylacetylene catalyzed with Pd(0) affords 3a-e in high yields. The Diels-Alder and decarbonylation reactions between 3a-e and tetraphenylcyclophentadiene following the oxidation by FeCl(3) produce the star-shaped and linear PAHs 5a-e containing a five-membered ring. The structural analysis and the optical properties of all new compounds are performed by a combination of MALDI-TOF mass spectrometry, UV-vis, and fluorescence spectrometry. The electronic and photophysical properties are studied by orthogonal comparisons of the absorption and fluorescence spectra in THF solutions, which not only give insight into the interactions among aromatic submoieties in each molecule and the effects of meta-conjugation and para-conjugation on electronic delocalization, but also indicate effective conjugation length variations from oligophenylacetylenes 3a-e to oligophenylene dendrimers 4a-e and PAHs 5a-e. The star-shaped 5c exhibits the highest aggregation in excited states compared with the other four hexa-peri-hexabenzocoronene (HBC) derivatives.  相似文献

Chemoselective SN2' condensation of primary enolates of alkyl methyl ketones 2a-e with dimethyl bromomethylfumarate (1) followed by highly diastereoselective NaBH4 reduction of the ketone function in the formed ketodiesters 3a-e and the regioselective in situ lactonization of the unisolable intermediates 4a-e exclusively furnished the cis-3,5-disubstituted gamma-butyrolactones (+/-)-5a-e in very good yields. Similarly, the face-selective coupling reaction of cyclohexanone enolate with 1 to form a mixture of diastereomers in an 8:2 ratio followed by a highly selective reductive cyclization of 9 plus 10 exclusively provided the cis-octahydrobenzofuran (+/-)-12 in 70% overall yield.  相似文献

6/3-(4-Chlorophenyl)-s-triazolo[3, 4-b][1, 3, 4]thiadiazoles (2,a-e) and (Sa-e) were synthesized respectively by intermolecular cyclization of 5-aryl / 4-chlorophenyl-4-amino-3- mercapto-1, 2, 4-triazoles (la-e) and (4) with 4-chlorobenzoic acid / aryl acids, which were condensed with piperazine under phase transfer catalyst TBAB to yield the corresponding free bases of monopiperazine derivatives and followed to form water-soluble salts (3a-e) and (6a-e) with hydrochloric acid in good yields. The in vitro biological results showed that piperazine group conjugated with the above fused heterocycles played an important role in antibacterial activity. The structures of novel compounds were confirmed by IR, 'H NMR, MS and elemental analysis.  相似文献

2,4-Bis(4-methoxyphenyl)-1,3,2,4-dithiadiphosphetane-2,4-disulfide (Lawesson's Reagent, LR, 1 ) reacts with cycloalkylidenecyanothioacetamides ( 2 and 3 ) to give 5'-cyano-2'-(4-methoxyphenyl)spiro [cyclopentane(cyclohexane)-1,6'-perhydro-[1,3,2]thiazaphosphixane]-2',4'-disulfide ( 4 and 5 ). The reaction of compounds 4 and 5 with f -halo compounds led to the formation of the substituted thio-compounds 6a-e and 7a-e , respectively, these compounds, upon treatment with sodium ethoxide, produce the corresponding thienothiazaphosphixine derivatives 8a-e and 9a-e respectively. Compounds 8a-e and 9a-e react with LR under different reaction conditions to give polyfused heterocyclic compounds 10a-d and 11a-d respectively. Treatment of compounds 8b and 9b with CS 2 and (CH 3 ) 2 SO 4 gave the corresponding dithiocarbamate methyl ester derivatives 12 and 13 , respectively, which on treating with hydrazine hydrate yielded compounds 14 and 15 respectively. Compounds 14 and 15 reacted with LR to yield compounds 16a , b and 17a , b respectively.  相似文献

Second-order rate constants have been determined spectrophotometrically for the reactions of 4-nitrophenyl X-substituted 2-methylbenzoates (2a-e) and Y-substituted phenyl 2-methylbenzoates (3a-e) with alicyclic secondary amines in 80 mol % H(2)O/20 mol % DMSO at 25.0 +/- 0.1 degrees C. The o-methyl group in the benzoyl moiety of 2a-e retards the reaction rate but does not influence the reaction mechanism. The Hammett plots for the reactions of 2a-e are nonlinear, while the corresponding Yukawa-Tsuno plots are linear with large r values (1.06-1.70). The linear Yukawa-Tsuno plots suggest that stabilization of the ground-state through resonance interaction between the electron donating substituent X and the carbonyl group is responsible for the nonlinear Hammett plots, while the large r values imply that the ground-state resonance interaction is significant. The reactions of 2a-e resulted in smaller rho(X) values but larger r values than the corresponding reactions of 4-nitrophenyl X-substituted benzoates (1a-e). The small rho(X) value for the reactions of 2a-e (e.g., rho(X) = 0.22) is suggested to be responsible for the large r value (e.g., r = 1.70). The reactions of 3a-e with piperidine are proposed to proceed in a stepwise manner with a change in the rate-determining step on the basis of the curved Br?nsted-type plot obtained. Microscopic rate constants associated with the reactions of 3a-e are also consistent with the proposed mechanism.  相似文献

Stannylacetylenes 7a-e react with Cp(2)Zr(H)Cl in THF at room temperature to give the alpha-zirconated vinylstannane intermediates 8a-e, which subsequently react with butyltellurenyl bromide (2.0 equiv) to give exclusively ketene stannyl(telluro) acetals 6a-e of E configuration. Similar reactions were performed using phenylselenenyl bromide (2.0 equiv) as the electrophile, but a mixture of products was formed including the expected ketene stannyl(seleno) acetals 12. Otherwise, the use of 1.4 equiv of Cp(2)Zr(H)Cl and 1.0 equiv of PhSeBr results in the exclusive formation of 12, in good yields. Treatment of ketene stannyl(telluro) acetals with iodine or NBS followed by reductive dehalogenation results in the formation of 1-iodo-1-telluroalkenes 4a-e and 1-bromo-1-telluroalkenes 5a-e, respectively, with total retention of the configuration.  相似文献

A kinetic study is reported for reactions of Y-substituted phenyl X-substituted cinnamates (1a-e and 3a-g) and benzoates (2a-e and 4a-g) with a series of alicyclic secondary amines in 80 mol % H2O/20 mol % DMSO at 25.0 +/- 0.1 degrees C. Reactions of 2,4-dinitrophenyl X-substituted cinnamates (1a-e) and benzoates (2a-e) with amines result in linear Yukawa-Tsuno plots. The rho(X) values are much smaller for the reactions of 1a-e than for those of 2a-e. A distance effect and the nature of the reaction mechanism (i.e., a concerted mechanism for 1a-e) have been suggested to be responsible for the small rho(X) values. The Br?nsted-type plots for the reactions of 2,4-dinitrophenyl X-substituted cinnamates (1a, 1c, and 1e) with amines are curved with a decreasing betanuc value from 0.65 to 0.3-0.4. The reactions of Y-substituted phenyl cinnamates (3a-g) with morpholine also result in a curved Br?nsted plot, while the corresponding reactions of Y-substituted phenyl benzoates (4a-e) exhibit a linear Br?nsted plot. It has been concluded that the curved Br?nsted plots found for the reactions of the cinnamates (1a, 1c, 1e, and 3a-g) are not due to a change in the rate-determining step (RDS) but due to a normal Hammond effect for a concerted mechanism, that is, an earlier transition state (TS) for a more reactive amine or substrate.  相似文献

1,5,6,10b-Tetrahydropyrrolo[2,1-alpha]isoquinolin-3(2H)-ones 17a,b, 17d,e, and 5,12b-dihydroisoindolo[1,2-alpha]isoquinolin-8(6H)-ones 22a-e were prepared by intramolecular cyclizations of 1-(2-arylethyl)-5-benzotriazolyl-pyrrolidin-2-ones 15a,b, 15d,e, and 3-benzotriazolyl-2-(2-arylethyl)-1-isoindolinones 20a-e, respectively, in the presence of titanium chloride. Hindered amino acids have been introduced into peptide chains by coupling N-(Cbz- and Fmoc-alpha-aminoacyl)benzotriazoles with amino acids, wherein at least one of the components was sterically hindered, to provide compounds 3a-e, (3c +3 c'), 5a-d, (5a + 5a'), 6a-c, (6b + 6b'), 8a-c, 9a-e, 10a-d, and (10a + 10a') in isolated yields of 41-95% with complete retention of chirality as evidenced by NMR and HPLC analysis. The benzotriazole activation methodology is a new route for the synthesis of sterically hindered peptides. [formula: see text] The bakers' yeast reduction of 3-(1,3-benzodioxol-5-yl)-6-propoxy-1H-inden-1-one 4 has been shown to give (S)-3-(1,3-benzodioxol-5-yl)-2,3-dihydro-6-propoxy-1H-indan-1-one 6 in 65% yield with high enantioselectivity (> 99.0% ee), a key intermediate for the synthesis of the endothelin receptor antagonist SB 217242. In addition, the substituted 3-arylinden-1-ones 10a-e gave equally high enantioselectivity for the 3-arylindan-1-one products 13a-e. Mechanistic studies of the reaction indicate the operative pathway to be an asymmetric conjugate reduction, wherein the hydride transfer from NAD(P)H occurs from the Re-face of the indenone substrate.  相似文献

In this study appropriate hydrazide compounds, furan-2-carboxylic acid hydrazide (1) and phenylacetic acid hydrazide (2) were converted into 1,4-substituted thiosemicarbazides 4a-e and 5a-e and 4-amino-5-(furan-2-yl or benzyl)-4H-1,2,4-triazole-3-thiols 7 and 10. The 1,4-substituted thiosemicarbazides were then converted into 5-(furan-2-yl or benzyl)-4-(aryl)-4H-1,2,4-triazole-3-thiols 8a-e and 9a-e. In addition, the azomethines 11a-d and 12a-d were prepared from the corresponding arylaldehydes and the 4-amino-5-(furan-2-yl or benzyl)-4H-1,2,4-triazole-3-thiols 7 and 10. The structures of all the synthesized compounds were confirmed by elemental analyses, IR, (1)H-NMR and(13) C-NMR spectra.  相似文献

A family of new coordination vanadium(IV) compounds supported by a terminal or bridged aryl imido ligand are reported. Reaction of V(NMe(2))(4) with anilines ArNH(2), where Ar = 2,6-i-Pr(2)-C(6)H(3), 2,6-Me(2)-C(6)H(3), Ph, 2,6-Cl(2)-C(6)H(3), and C(6)F(5), afforded the diamagnetic imido-bridged complexes [V(NAr)(NMe(2))(2)](2) (1a-e). Chlorination of 1a-e with trimethylchlorosilane afforded complexes 2a-e formulated as [V(=NAr)Cl(2)(NHMe(2))(x)()](n)(). One-pot reaction of V(NMe(2))(4) with ArNH(2) in the presence of an excess of trimethylchlorosilane gave the five-coordinate compound [V(=NAr)Cl(2)(NHMe(2))(2)] (3a-e). Reaction of 3a-e with pyridine, bipyridine (bipy), or N,N,N',N'-tetramethylethylenediamine (tmeda) gave respectively the six-coordinate tris- or bis(pyridine) adducts [V(=NAr)Cl(2)(Py)(3)] (4a-e) or [V(=NAr)Cl(2)(Py)(2)(NHMe(2))] (5a), bipyridine complexes [V(=NAr)Cl(2)(bipy)(NHMe(2))] (5a-e) and [V(=NAr)Cl(2)(bipy)(Py)] (9a), and tmeda adduct [V(=NAr)Cl(2)(tmeda)(NHMe(2))] (10a). Moreover, five-coordinate complexes free of NHMe(2) ligands, such as [V(=NAr)Cl(2)(Py)(2)] (5a), [V(=NAr)Cl(2)(bipy)] (8a), and [V(=NAr)Cl(2)(tmeda)] (11a), were directly prepared starting from precursors 2a-e. All compounds were totally characterized by spectroscopic methods (IR, (1)H NMR for diamagnetic complexes, and EPR for paramagnetic complexes), elemental analysis, magnetism, and single-crystal X-ray diffraction studies for 1b, 3a, 3d, 4b, 4d, 7c, 10a, and 11a.  相似文献

Nitration of 1-arylpyrroles 1a-c with acetyl nitrate, and 1-arylpyrroles, 1a-e and 1-(2-ethoxycarbonylbenzyl)pyrrole 4 with trifluoroacetyl nitrate gave the corresponding 2-nitro isomers 2a-e and 5, and 3-nitro isomers 3a-e and 6 . 3-Nitropyrroles 3d and 3e were further nitrated with a mixture of nitric and sulfuric acids to give compounds 10, 11 and 12 , respectively. Under the same conditions 1-(2-ethoxycarbonylbenzyl)-3-nitropyrrole 6 gave derivative 13 .  相似文献

A new route to C-6-selenenyl analogs of compound 1a from 5-alkyl-6-chlorouracils 6a-b has been described. A mild and highly efficient synthesis of 1-(alkoxymethyl)-5-alkyl-6-(arylselenenyl)uracils 8a-e has been accomplished from 6a-b in good yields using a two step procedure. Silylation of 5-alkyl-6-chlorouracils 6a-b using N,O-bis(trimethylsilyl)acetamide followed by regioselective alkylation of the silylated intermediate with ethyl or benzyl chloromethyl ether in dichloromethane afforded the desired 1-(alkoxymethyl)-5-alkyl-6-chlorouracils 7a-d in 88–94% yields. Compounds 7a-d readily underwent addition-elimination reaction with an appropriate arylselenol in the presence of ethanolic sodium hyroxide to produce the corresponding 1-(alkoxymethyl)-5-alkyl-6-(arylselenenyl)uracils 8a-e in excellent yields (94–99%).  相似文献
