The aldol-Tishchenko reaction of ketone aldols as enol equivalents has been developed as an efficient strategy to furnish differentiated 1,3-anti-diol monoesters in one step. The thermodynamically unstable ketone aldols undergo a facile retro-aldolization to yield a presumed zirconium enolate in situ, which then undergoes the aldol-Tishchenko reaction in typically high yields and with complete 1,3-anti diastereocontrol. Evaluation of a broad range of metal alkoxides as catalysts and optimization of the reaction protocol led to a modified zirconium alkoxide catalyst with attenuated Lewis acidity and dichloromethane as solvent, which resulted in suppression of the undesired acyl migration to a large extent. Various ketone aldols have been prepared and subjected to the general process, giving rise to a broad range of differently substituted 1,3-anti-diol monoesters, which may be hydrolyzed to the corresponding 1,3-anti-diols.  相似文献

A novel DABCO-catalyzed Ireland-Claisen rearrangement/Diels-Alder tandem reaction of propargylic acrylates 3 with dienophiles 4 was developed in the presence of an excess TMSCl and DBU with 1% hydroquinone as polymerization inhibitor in acetonitrile at refluxing temperature under air.This protocol gave cyclicα,β-unsaturated carboxylic acids 5 with complete regioselectivity.  相似文献

Here, we report a formal synthesis of the marine cytotoxic meroterpenoid actinoranone. Key steps include a semipinacol rearrangement/Wittig reaction sequence and a chiral pool approach for the syntheses of the tetralone and the ocatalin fragments, respectively. The presented route provides access to the natural product in 14 steps in the longest linear sequence.  相似文献

A facile and highly diastereoselective method for the construction of 2-quaternary 1,3-amino alcohols and 1,3-diols has been developed on the basis of the AlEt3/THF-promoted tandem rearrangement/reductive reaction of α-hydroxy (amino) aziridines (epoxides). The progressive achievement in this article included that both 2-epimers of the units could be constructed from the initially same substrate. A visible light-induced photocatalytic aerobic oxidation/semipinacol rearrangement has been successfully developed. This methodology allows for efficient conversion of a wide range of indoles into the synthetically significant 2,2-disubstituted indolin-3-ones, the core structure of pseudoindoxyl alkaloids, under mild reaction conditions.  相似文献

Summary The reaction of theophylline (1) with 2-methyl-3-butyn-2-ol and 1-butyn-3-ol under Mitsunobu conditions gave the respective 9-substituted derivatives 9-[2-(2-methyl-3-butynyl)]-theophylline (2) and 9-[2-(3-butynyl)]-theophylline (3). On reaction with 2-methyl-3-buten-2-ol, theophylline yielded in addition to the 9-[2-(2-methyl-3-butenyl)]-theophylline (4), two more cyclic products, identified as 1,5,5a,8-tetrahydro-1,3,8,8-tetramethyl-2H-pyrrolo[1,2-e]purine-2,4(3H)-dione (5) and 8a,9-dihydro-1,3,6,6-tetramethyl-1H-pyrrolo[2,1-f]purine-2,4(3H,6H)-dione (7).

Zusammenfassung Die Reaktion von Theophyllin (1) mit 2-Methyl-3-butin-2-ol und 1-Butin-2-ol unter Mitsunobu-Bedingungen führte zu den 9-substituierten Derivaten 9-[2-(2-Methyl-3-butinyl)]-theophyllin (2) bzw. 9-[2-(3-Butinyl)]-theophyllin (3). Bei der Reaktion mit 2-Methyl-3-buten-2-ol ergab Theophyllin außer 9-[2-(2-Methyl-3-butenyl)]-theophyllin (4) noch zwei weitere cyclisierte Produkte, die als 1,3,8,8-Tetramethyl-1,5,5a,8-tetrahydro-pyrrolo[1,2-e]purin-2,4(3H)-dion (5) und 1,3,6,6-Tetramethyl-8a,9-dihydro-1H,6H-pyrrolo[2,1-f]purin-2,4-dion (7) identifiziert wurden.

The direct alkylation of silyl enol ethers with para-methoxybenzylic alcohols or their corresponding acetates was efficiently catalyzed by Bi(OTf)3 in CH3NO2 as the solvent. The reaction provided the α-benzylated carbonyl compounds in high yields after short reaction times using 1-2.5 mol % of the catalyst. Benzylic acetates other than para-methoxybenzylic acetates also underwent the reaction. High facial diastereoselectivities were observed with acetates derived from chiral α-branched para-methoxybenzylic alcohols. In addition, a catalytic reduction with Et3SiH as the reducing agent is reported.  相似文献

The lanthanide catalysed para-Claisen-Cope rearrangement has been used as the key step in a short synthesis of the prenylated isoflavone, lupiwighteone. The Mitsunobu reaction was employed for the 5-O-prenylation of the acid/base sensitive acetylated isoflavone to afford the allyl aryl ether precursor, which was then rearranged under mild conditions in good yield. Rearrangement of the isoflavone gave the 8-prenylisoflavone as a single product, in good yield.  相似文献

Aliphatic aldehydes react with catalytic amount of Dibal-H in n-pentane to give the corresponding Tishchenko products in good to excellent yields. On contrary, α-silyloxy aldehydes give α-silyloxy ketones via Oppenauer oxidation under similar condition. Tishchenko reaction of ω-alkene aldehydes followed by RCM and hydrogenation affords a convenient method to prepare the 11-37 membered macrocyclic lactones.  相似文献

The catalytic asymmetric rearrangement of functionalised cyclohexene and cyclopentene oxides has been studied using sub-stoichiometric amounts of a chiral lithium amide in combination with a stoichiometric amount of three different lithiated imidazoles. 1-Methylimidazole that had been lithiated at the C-2 aryl position gave the highest enantioselectivity (82% ee). With 1,2-dimethylimidazole that had been lithiated at the C-2 methyl group, epoxide ring opening occurred as an unexpected and competing process. Ultimately, ring opening was suppressed using a more sterically hindered imidazole. In all catalytic examples, a racemic background reaction (presumably due to rearrangement by the lithiated imidazoles) was observed.  相似文献

Synthesis of dispiro[1,4-dithianes/dithiepanes]bisoxindoles from the reaction of 3-diazo-2-oxindoles and spiro-1,3-dithiolaneoxindole or -1,3-dithianeoxindole derivatives in the presence of rhodium(II) acetate dimer as a catalyst was demonstrated under mild conditions in a highly diastereoselective manner. These dispiro-compounds were obtained via intermolecular sulfonium ylides and followed by Stevens rearrangement.  相似文献
