Reaction of difluorocarbene with diarylmethanimines leads to the formation of gem-difluorinated NH-azomethine ylides, two types of competing transformations of which are found to be characteristic: a formal 1,2-H shift into N-(difluoromethyl)imines and 1,3-dipolar cycloaddition to electron-deficient multiple bonds. α,α,α-Trifluoroaceto-phenones are efficient dipolar traps for difluoro NH-ylides, the addition of which to the dipole proceeds regioselectively with the formation of 4-fluoro-2,5-dihydrooxazoles. According to the quantum-chemical calculations by the DFT B3LYP/6-31G* method, 1,3-dipolar cycloaddition of difluorinated NH-azomethine ylides to a C=O bond with the formation of 4-fluoro derivatives of oxazole has lower barrier of activation than the reaction, leading to another regioisomer; the formal 1,2-H shift in the ylide occurs intermolecularly with participation of an imine, a precursor of the ylide.  相似文献

Oxidation of aldehyde 4-chloro-2,3,5,6-tetrafluorophenylhydrazones (1) with [bis(acetoxy)iodo]benzene leads to the formation of nitrile imines (2) which reacted in situ with ethyl acrylate to produce 3-substituted-1-(4-chloro-2,3,5,6-tetrafluorophenyl)-5-ethoxycarbonyl-4,5-dihydropyrazoles (3) in moderate to good yields. The structures of new compounds were fully confirmed by their spectral data, elemental analyses and X-ray diffraction (XRD) analysis. A plausible reaction mechanism for the synthesis of title compounds is presented.  相似文献

An efficient method was developed for the asymmetric synthesis of 2′-alkyl-4′aryl-1H-spiro[indole-3,3′-pyrrolidin]-2-ones, which are potential inhibitors of the p53-MDM2 interaction. Our X-ray crystallographic analysis revealed that this 1,3-dipolar cycloaddition proceeds with high stereoselectivity but differently from previously published results.  相似文献

An efficient synthesis of fluorescent coumarin-nucleoside conjugates via Cu(I) catalysed Huisgen 1,3-dipolar cycloaddition is described. Starting from azidonucleosides and coumarin derivatives, products are obtained in good yields. A facile synthesis of novel spiroindane-1,3-diones has been achieved via 1,3-dipolar cycloaddition of an azomethine ylide, generated in situ from ninhydrin and 1,2,3,4-tetrahydroisoquinoline, with the conjugated double bond of chalcone derivatives. The regiochemistry and structures of the cycloadducts were determined with spectroscopic data and by X-ray crystal structure analysis.  相似文献

A series of new isoxazolidines was prepared by 1,3-dipolar cycloaddition of different mono-substituted styrenes with 1,3-dipolar compounds that were prepared by the reaction of N-methylhydroxylamine sulfate with aromatic carbonyl substances. This synthetic pathway for the preparation of isoxazolidines was an ideal process of green chemistry. The 1,3-dipolar cycloaddition reaction of azomethine ylides generated in situ from isatin and secondary amino acids with quinolinyl dipolarophiles in refluxing methanol afforded new class of quinolinyl dispiro heterocycles with multi hetero core units. The regio and stereochemistry of the product was unambiguously assigned by 1H, 13C, 2D NMR techniques and single crystal X-ray analysis. The structures of the compounds are stabilized through the presence of intermolecular hydrogen bonding and intramolecular non-covalent interactions.  相似文献

Adamantane-derived aldo- and ketonitrones react with maleimides giving corresponding isoxazolidines. In the case of aldonitrones reactions proceed giving the diastereomeric mixtures. The ratio of isomers is changing during the reaction process due to the reversibility of the cycloaddition reaction. The cytotoxic and virus-inhibiting activity against influenza virus was investigated for selected adducts.  相似文献

A study of diastereoselectivity in the cycloaddition reactions of a series of mono- and disubstituted alkenes with two chiral, internally H-bonded methylenenitrones has been carried out. The high degree of stereochemical control in the presence of anhydrous magnesium bromide has been explained in terms of a metal chelated transition state. Intramolecular cycloaddition involving a methylenenitrone containing an alkene moiety linked to a nitrogen gave a stereoselective addition product.  相似文献

1,3,5-Trisubstituted 3-thienylpyrazolines have been prepared in high yields by the reaction of substituted,N-(P;-nitrophenyl)-3-thiophenecarbohydrazonoyl chlorides with Et3N in CH2Cl2 in the presence of an excess of a monosubstituted olefin. Novel aza-bicyclic 2-isoxazolines, 4,5-dihydroisoxazole[5,4-b]pyrrolidines, and 4,5-dihydroisoxazole[5,4-b]piperidines were synthesized in a highly regioselective manner through a 1,3-dipolar cycloaddition reaction of 5- and 6-membered endocyclic enecarbamates and enamides with several nitrile oxides in good to excellent yields. Hydrogenolysis of 5- and 6-membered Cbz-cycloadducts led to secondary amines, which presented distinctive stabilities. 2-Isoxazoline bisamides were obtained in good yields through a N-benzoylation, followed by ammonolysis of the secondary amine, or directly from ammonolysis of the cycloadducts.  相似文献

Hydroxylamine hydrochloride adds via a Michael reaction on the acetonylidene moiety of (4Z)-(2-oxopropylidene)-1,2,4,5-tetrahydro-2H-1,5-benzodiazepin-2-one, leading only to 2-[(5-methyl-isoxazol-3-yl)]methyl]benzimidazoles. This report describes the difference of reactivity of C-phenyl-N-phenyl formohydrazonoyl chloride with benzimidazole. For the N-substituted benzimidazole, an unexpected opening of the azole ring occurs, which was confirmed by single crystal X-ray diffraction analysis.  相似文献

The spiroisoxazolidinyl-benzoisothiazoline derivatives were synthesized through a highly diastereoselective 1,3-dipolar cycloaddition of benzoisothiazole-2,2-dioxide-3-ylidenes with nitrones. The regiochemistry of cycloaddition was assigned on the 1H NMR chemical shift of methane-H in 3 and 5.
