The surface activity of two hydrophobin proteins, HFBII and SC3, at the solid–liquid, liquid–liquid and liquid–vapor interface has been investigated. Hydrophobins are fungal proteins that are known to adsorb and affect the physico-chemical properties of an interface. In this study, the surface activity was determined by measuring the interaction of hydrophobin molecules with various liquids, solid particles and films that are commonly used or produced in industrial processes. We found that a very low concentration of hydrophobin is required to facilitate the wet-in of hydrophobic solid particles, such as Teflon®, into aqueous solutions. It is also capable of stabilizing aqueous dispersions of Kevlar® nanopulp, reversing the wettability of hydrophobic films and stabilizing polyunsaturated fatty acid (PUFA) oil-in-water emulsions.  相似文献

The adsorption of surface-active protein hydrophobin, HFBII, and HFBII/surfactant mixtures at the solid-solution interface has been studied by neutron reflectivity, NR. At the hydrophilic silicon surface, HFBII adsorbs reversibly in the form of a bilayer at the interface. HFBII adsorption dominates the coadsorption of HFBII with cationic and anionic surfactants hexadecyltrimethyl ammonium bromide, CTAB, and sodium dodecyl sulfate, SDS, at concentrations below the critical micellar concentration, cmc, of conventional cosurfactants. For surfactant concentrations above the cmc, HFBII/surfactant solution complex formation dominates and there is little HFBII adsorption. Above the cmc, CTAB replaces HFBII at the interface, but for SDS, there is no affinity for the anionic silicon surface hence there is no resultant adsorption. HFBII adsorbs onto a hydrophobic surface (established by an octadecyl trimethyl silane, OTS, layer on silicon) irreversibly as a monolayer, similar to what is observed at the air-water interface but with a different orientation at the interface. Below the cmc, SDS and CTAB have little impact upon the adsorbed layer of HFBII. For concentrations above the cmc, conventional surfactants (CTAB and SDS) displace most of the HFBII at the interface. For nonionic surfactant C(12)E(6), the pattern of adsorption is slightly different, and although some coadsorption at the interface takes place, C(12)E(6) has little impact on the HFBII adsorption.  相似文献

The self-assembly of the protein hydrophobin, HFBII, and its self-assembly with cationic, anionic, and nonionic surfactants hexadecylterimethyl ammonium bromide, CTAB, sodium dodecyl sulfate, SDS, and hexaethylene monododecyl ether, C(12)E(6), in aqueous solution have been studied by small-angle neutron scattering, SANS. HFBII self-assembles in solution as small globular aggregates, consistent with the formation of trimers or tetramers. Its self-assembly is not substantially affected by the pH or electrolytes. In the presence of CTAB, SDS, or C(12)E(6), HFBII/surfactant complexes are formed. The structure of the HFBII/surfactant complexes has been identified using contrast variation and is in the form of HFBII molecules bound to the outer surface of globular surfactant micelles. The binding of HFBII decreases the surfactant micelle aggregation number for increasing HFBII concentration in solution, and the number of hydrophobin molecules bound/micelle increases.  相似文献

The pendant-drop method (with drop-shape analysis) and Langmuir trough are applied to investigate the characteristic relaxation times and elasticity of interfacial layers from the protein HFBII hydrophobin. Such layers undergo a transition from fluid to elastic solid films. The transition is detected as an increase in the error of the fit of the pendant-drop profile by means of the Laplace equation of capillarity. The relaxation of surface tension after interfacial expansion follows an exponential-decay law, which indicates adsorption kinetics under barrier control. The experimental data for the relaxation time suggest that the adsorption rate is determined by the balance of two opposing factors: (i) the barrier to detachment of protein molecules from bulk aggregates and (ii) the attraction of the detached molecules by the adsorption layer due to the hydrophobic surface force. The hydrophobic attraction can explain why a greater surface coverage leads to a faster adsorption. The relaxation of surface tension after interfacial compression follows a different, square-root law. Such behavior can be attributed to surface diffusion of adsorbed protein molecules that are condensing at the periphery of interfacial protein aggregates. The surface dilatational elasticity, E, is determined in experiments on quick expansion or compression of the interfacial protein layers. At lower surface pressures (<11 mN/m) the experiments on expansion, compression and oscillations give close values of E that are increasing with the rise of surface pressure. At higher surface pressures, E exhibits the opposite tendency and the data are scattered. The latter behavior can be explained with a two-dimensional condensation of adsorbed protein molecules at the higher surface pressures. The results could be important for the understanding and control of dynamic processes in foams and emulsions stabilized by hydrophobins, as well as for the modification of solid surfaces by adsorption of such proteins.  相似文献

Dispersing hydrophobin HFBII under air saturated with perfluorohexane gas limits HFBII aggregation to nanometer‐sizes. Critical basic findings include an unusual co‐adsorption effect caused by the fluorocarbon gas, a strong acceleration of HFBII adsorption at the air/water interface, the incorporation of perfluorohexane into the interfacial film, the suppression of the fluid‐to‐solid 2D phase transition exhibited by HFBII monolayers under air, and a drastic change in film elasticity of both Gibbs and Langmuir films. As a result, perfluorohexane allows the formation of homogenous populations of spherical, narrowly dispersed, exceptionally stable, and echogenic microbubbles.  相似文献

We report the remarkable surface behavior of class II hydrophobin proteins HFBI and HFBII from Trichoderma reesei and the resulting effect that these proteins have on the stability of air bubbles to the process of disproportionation. The surface properties were studied using surface tensiometry and surface shear rheology. Surface tensiometry data show that hydrophobins are very surface active proteins, reducing the surface tension to approximately 30 mN m-1. The rate at which the hydrophobins adsorb at the surface may also be related to the self-assembly behavior in aqueous solution. We further show that hydrophobins form air/water surfaces with high elasticity, the magnitude of which is well in excess of that of surface layers formed by other common proteins used as foam or emulsion stabilizers. The measured surface properties translate to the stability of bubbles with adsorbed hydrophobin, and in this study, we demonstrate the ability of hydrophobin to have a dramatic effect on the rate of disproportionation in some simple bubble dissolution studies.  相似文献

We have investigated the self-assembling properties of the class I hydrophobin Vmh2 isolated from the fungus Pleurotus ostreatus. Five different hydrophobin self assembled samples including monolayers, bilayers, and rodlets have been prepared by Langmuir technique and studied at the nanoscale. Local wettability and visco-elasticity of the different hydrophobins samples were obtained from atomic force spectroscopy experiments in dynamic mode performed at different, controlled relative humidity (RH) values. It was found that hydrophobins assembled either in rodlets or in bilayer films, display similar hydropathicity and viscoelasticity in contrast to the case of monolayers, whose hydropathicity and viscoelasticity depend on the adopted preparation method (Langmuir-Blodgett or Langmuir-Schaeffer). The comparison with monolayers properties evidences a rearrangement of the bilayers adsorbed onto solid substrates. It is shown that this rearrangement leads to the formation of a stable hydrophobic film, and that the rodlets structure consists in fragments of restructured proteins bilayers. Our results support the hypothesis that the observed variations in the viscoelastic properties could be ascribed to the localization of the large flexible loop, typical of Class I hydrophobins which appears free at the air interface for LB monolayers but not for the other samples. These findings should now serve future developments and applications of hydrophobin films beyond the archetypal monolayer.  相似文献

The equilibrium between free surfactant molecules in aqueous solution and adsorbed layers on structured solid surfaces is investigated by lattice Monte Carlo simulation. The solid surfaces are composed of hydrophilic and hydrophobic surface regions. The structures of the surfactant adsorbate above isolated surface domains and domains arranged in a checkerboard-like pattern are characterized. At the domain boundary, the adsorption layers display a different behavior for hydrophilic and hydrophobic surface domains. For the checkerboard-like surfaces, additional adsorption takes place at the boundaries between surface domains.  相似文献

The adsorption of the surface-active protein hydrophobin, HFBII, and the competitive adsorption of HFBII with the cationic, anionic, and nonionic surfactants hexadecyltrimethylammonium bromide, CTAB, sodium dodecyl sulfate, SDS, and hexaethylene monododecyl ether, C(12)E(6), has been studied using neutron reflectivity, NR. HFBII adsorbs strongly at the air-water interface to form a dense monolayer ~30 ? thick, with a mean area per molecule of ~400 ?(2) and a volume fraction of ~0.7, for concentrations greater than 0.01 g/L, and the adsorption is independent of the solution pH. In competition with the conventional surfactants CTAB, SDS, and C(12)E(6) at pH 7, the HFBII adsorption totally dominates the surface for surfactant concentrations less than the critical micellar concentration, cmc. Above the cmc of the conventional surfactants, HFBII is displaced by the surfactant (CTAB, SDS, or C(12)E(6)). For C(12)E(6) this displacement is only partial, and some HFBII remains at the surface for concentrations greater than the C(12)E(6) cmc. At low pH (pH 3) the patterns of adsorption for HFBII/SDS and HFBII/C(12)E(6) are different. At concentrations just below the surfactant cmc there is now mixed HFBII/surfactant adsorption for both SDS and C(12)E(6). For the HFBII/SDS mixture the structure of the adsorbed layer is more complex in the region immediately below the SDS cmc, resulting from the HFBII/SDS complex formation at the interface.  相似文献

Purification is a critical step to obtain hydrophobin HFBII for use in positive applications. In this study, hydrophobin HFBII was produced by Trichoderma reesei via submerged fermentation. Using the CO2-foam fractionation method yielded a fourfold increase in protein concentration. The foamate (αL-HFBII) was dried using a nano spray-dryer under optimal temperature. The gushing activity of the dried foamate (αS-HFBII) decreased. Addition of Tween 80 to the foamate before the drying process partially prevented the deactivation of hydrophobin HFBII. The purity of the powder was enhanced based on the theory of CO2-nanobubbles in a CO2-rich environment. The collected CO2-nanobubbles were added to an apolar–polar system and the interface of these two phases was collected. After evaporation of the apolar phase, the purity of the hydrophobins assembled on the surface of the liquid was significantly improved.  相似文献

Hydrophilicity improvement and bioactive surface design of poly(?-caprolactone) (PCL) grafts are of key importance for their application in tissue engineering. Herein, we develop a convenient approach for achieving stable hydrophilic surfaces by modifying electrospun PCL grafts with a class II hydrophobin (HFBI) coating. Static water contact angles (WCA) demonstrated the conversion of the PCL grafts from hydrophobic to hydrophilic after the introduction of amphiphilic HFBI. ATR-FTIR and XPS confirmed the presence of self-assembled HFBI films on the surface of the PCL nanofibers. The biocompatibility of the HFBI-modified PCL grafts was evaluated by cell proliferation in vitro, and by arteriovenous shunt (AV shunt) experiments ex vivo. Anti-CD31 antibody, which is specific for endothelial cells (ECs), was subsequently immobilized on the HFBI-coated PCL scaffolds through protein-protein interactions. This bioactive PCL graft was found to promote the attachment and retention of endothelial cells. These results suggest that this stepwise strategy for introducing cell-specific binding molecules into PCL scaffolds may have potential for development of vascular grafts that can endothelialize rapidly in vivo.  相似文献

The kinetics of adsorption of lysozyme and alpha-lactalbumin from aqueous solution on silica and hydrophobized silica has been studied. The initial rate of adsorption of lysozyme at the hydrophilic surface is comparable with the limiting flux. For lysozyme at the hydrophobic surface and alpha-lactalbumin on both surfaces, the rate of adsorption is lower than the limiting flux, but the adsorption proceeds cooperatively, as manifested by an increase in the adsorption rate after the first protein molecules are adsorbed. At the hydrophilic surface, adsorption saturation (reflected in a steady-state value of the adsorbed amount) of both proteins strongly depends on the rate of adsorption, but for the hydrophobic surface no such dependency is observed. It points to structural relaxation ("spreading") of the adsorbed protein molecules, which occurs at the hydrophobic surface faster than at the hydrophilic one. For lysozyme, desorption has been studied as well. It is found that the desorbable fraction decreases after longer residence time of the protein at the interface.  相似文献

利用六甲基二硅烷胺对平面玻璃光波导(高折射率透明导光薄膜介质)进行硅烷化处理, 得到水接触角大于90°的疏水表面. 然后使用时间分辨光波导分光光谱技术研究水溶液中的罗丹明6G (R6G)和亚甲基蓝(MB)分子在疏水玻璃表面的吸附行为, 并与亲水玻璃条件下测得的结果进行对比. 对利用疏水玻璃光波导测得的R6G的吸附-脱附动力学曲线进行Langmuir拟合得到了R6G的吸附速率常数, 脱附速率常数以及吸附自由能. 并且发现与亲水玻璃情况相比, 吸附速率常数增大, 脱附速率常数减小, 吸附自由能更负. 在疏水玻璃表面形成的R6G和MB吸附层的吸光度与亲水玻璃情况相比显著升高, 表明这两种分子更倾向于吸附在疏水玻璃表面. 实验结果还发现玻璃硅烷化处理能够有效抑制这两种染料分子在表面的聚合反应.  相似文献

The Pro-His-Ser-Arg-Asn (PHSRN) sequence in fibronectin is a second cell-binding site that synergistically affects Arg-Gly-Asp (RGD). The PHSRN peptide also induces cell invasion and accelerates wound healing. We report on the surface immobilization of PHSRN by spontaneous adsorption on polysiloxane thin films which have different surface free energy characteristics. Low-surface energy (hydrophobic) polysiloxane and the corresponding high-surface energy (hydrophilic) surfaces obtained by UV–ozone treatments were used as adsorbing substrates. The peptide adsorption process was investigated by quartz crystal microbalance with dissipation monitoring and atomic force microscopy. Both adsorption kinetics and peptide rearrangement dynamics at the solid interface were significantly different on the surface-modified films compared to the untreated ones. Fibroblast cells cultures at short times and in a simplified environment, i.e., a medium-free solution, were prepared to distinguish interaction events at the interface between cell membrane and surface-immobilized peptide for the two cases. It turned out that the cell-adhesive effect of immobilized PHSRN was different for hydrophobic compared to hydrophilic ones. Early signatures of cell spreading were only observed on the hydrophilic substrates. These effects are explained in terms of different spatial arrangements of PHSRN molecules immobilized on the two types of surfaces.  相似文献

Erythrocyte membrane permeability coefficients for a series of diols have been defined by the method developed. The method is based on the physical and mathematical modeling of hypotonic hemolysis process. There have been also determined membrane permeability coefficients for erythrocytes treated with p-chloromercuribenzenesulfonic acid monosodium salt (pCMBS), which is known to block aqueous protein channels. Permeating process is shown to be conditioned both by hydrophilic/hydrophobic properties of the molecules and their geometrical parameters. The obtained results propose that, when exceeding the molecules diameter over a value of 4 A, the permeability coefficient reduces due to decreasing of flow through the aqueous protein pores of a constant size. Permeability coefficients for comparatively hydrophobic molecules are almost directly proportional to the coefficients of partition between hydrophobic and hydrophilic phases, by pointing to a lipid way of permeation of these molecules through erythrocyte membranes.  相似文献

Hydrophobins are structural proteins produced by filamentous fungi that are amphiphilic and function through self-assembling into structures such as membranes. They have diverse roles in the growth and development of fungi, for example in adhesion to substrates, for reducing surface tension to allow aerial growth, in forming protective coatings on spores and other structures. Hydrophobin membranes at the air-water interface and on hydrophobic solids are well studied, but understanding how hydrophobins can bind to a polar surface to make it more hydrophobic has remained unresolved. Here we have studied different class II hydrophobins for their ability to bind to polar surfaces that were immersed in buffer solution. We show here that the binding under some conditions results in a significant increase of water contact angle (WCA) on some surfaces. The highest contact angles were obtained on cationic surfaces where the hydrophobin HFBI has an average WCA of 62.6° at pH 9.0, HFBII an average of 69.0° at pH 8.0, and HFBIII had an average WCA of 61.9° at pH 8.0. The binding of the hydrophobins to the positively charged surface was shown to depend on both pH and ionic strength. The results are significant for understanding the mechanism for formation of structures such as the surface of mycelia or fungal spore coatings as well as for possible technical applications.  相似文献

疏水蛋白是丝状真菌产生的一种外泌蛋白质, 它们可以在不同表面形成双亲性蛋白膜. 疏水蛋白也是一种优良的蛋白质固定化基质, 然而蛋白质在疏水蛋白表面吸附的驱动机制却是未知的. 本文系统研究了不同pH和离子浓度下蛋白质在疏水蛋白表面的吸附. 首先, 用石英晶体微天平技术研究了不同pH和离子浓度下, Ⅰ型疏水蛋白HGFI和Ⅱ型疏水蛋白HFBI在聚苯乙烯表面的吸附. 结果发现, pH和离子强度对HGFI在聚苯乙烯表面的吸附影响较大, 对HFBI的吸附影响与HGFI相比则较小; HGFI在聚苯乙烯表面主要形成的是弹性膜, 而HFBI在聚苯乙烯表面主要形成的是刚性膜. 随后又研究了不同pH和离子浓度下牛血清白蛋白(BSA)和亲和素(Avidin)在HGFI和HFB上吸附, 结果表明, pH和离子强度对BSA和Avidin在HGFI和HFB上吸附有显著影响, 说明BSA和Avidin在两种疏水蛋白上吸附的主要驱动力为静电力. 本文研究结果为实现疏水蛋白表面可控地固定蛋白质提供了理论指导.  相似文献

Pluronic triblock copolymers were entrapped on the surface of polyaniline (PANI) films by first immersing the latter in N-methylpyrrolidinone (NMP) solutions of one of the Pluronics for a short time. This softened the surface of the films and allowed the Pluronic molecules to entangle with PANI segments of the swollen film on the surface. Further, the films were taken out from the NMP solution and dipped into water, which is a nonsolvent for PANI. The rapid surface deswelling of PANI by the water resulted in the entrapment of the Pluronic on its surface, with the hydrophilic blocks toward water and the hydrophobic block imbedded in the PANI films. The modified PANI obtained was examined by X-ray photoelectron spectroscopy, water droplets contact angles, scanning electron microscopy, and wide angle X-ray diffraction. The surface of the Pluronic entrapped PANI films became more hydrophilic than the hydrophobic PANI films and decreased the amount of bovine serum albumin protein adsorbed on them. This means that, by reducing the biofouling, the life of the modified polyaniline film can be extended when the latter is employed as a biosensor.  相似文献
