3-Cyano-4,6-dimethyl-2-pyridone and 3-cyano-4-methyl-6-phenyl-2-pyridone were synthesized effectively by the reaction of readily available 1,3-diketone and malononitrile directly and in good yield. Upon photoexcitation, 3-cyano-4-methyl-6-phenyl-2-pyridone in ethanol shows strong blue emission. The ground- and excited-state geometries, charge distributions, and excitation energies of 2-pyridone derivatives were evaluated by ab initio calculations. Organic light-emitting diodes (OLED) made using 3-cyano-4-methyl-6-phenyl-2-pyridone as dopant showed blue light emission with a maximum electroluminescence (EL) emission at around 456 nm.  相似文献

4-Dicyanomethylene-2-phenyl-4H-1-benzopyran (1) reacts with primary amines under mild conditions to give 4-imino-3-alkyl-5-alkylimino-2-phenyl-3,4-dihydro-5H-[1]benzopyrano[3,4-c]-pyridine derivatives which, in turn, are hydrolyzed with acid to 4-imino-3-alkyl-2-phenyl-3,4-dihydro-5H-[1]benzopyrano[3,4-c]pyridin-5-ones. When more vigorous conditions are employed for the reactions of 1 with primary amines, Dimroth rearrangements take place and the products are derivatives of 4-alkyl- (or aryl)amino-5-alkyl- (or aryl)imino-2-phenyl-5H-[1]benzopyrano-[3,4-c]pyridine. The latter compounds are hydrolyzed by acid to the corresponding 5-pyridone derivatives. The reaction of 1 with piperidine gives 2-phenyl-4-piperidyl-5H-[1]benzopyrano-[3,4-c]pyridin-5-one. Sodium methoxide reacts with 1 to give 3-cyano-2-methoxy-4-(2-hydroxyphenyl)-6-phenylpyridine. Two benzologs of 1 have been allowed to react with primary and secondary amines and the products are analogous to those obtained from 1 .  相似文献

The Cope-Knoevenagel reaction of 2-phenylpropionaldehyde ( 7 ) with methyl cyanoacetate ( 8 ) produced methyl (E)-2-cyano-4-phenylpent-2-enoate ( 9 ) and the two highly fluorescent secondary products, 2-amino-3-carbomethoxy-6-phenyl-4-(1-phenylethyl)pyridine ( 10 ) and 3-cyano-6-phenyl-4-(1-phenylethyl)-2-pyridone ( 11 ). The structure of 10 was determined by X-ray crystallography while the structure of 11 was confirmed by the conversion of 9 into 11 . The mechanism of their formation is discussed. Fluorescence of 10 and 11 and the related compounds are also described.  相似文献

Abstract

By 1, 3-dipolar cycloaddition of benzonitrile oxide to 4, 6-di-O-acetyl-2, 3-dideoxy-D-erythro-hex-2-enono-1, 5-lactone (1), [3aR- (3aα, 6β,7α, 7aα)] - (2) and [3aS-(3aβ, 6β, 7α, 7aα)] -7- (acetyloxy) -6- (acetyloxymethyl) -3a, 6, 7, 7a-tetrahydro-3-phenyl-4H-pyrano [3, 4-d] isoxazole-4-one (3) were prepared in 58 and 7% yield respectively. From 2, [1′ R, 3aR-(3aα, 6β, 6aα)] -6-(1′, 2′-dihydroxymethyl)-6, 6a - dihydro-3-phenyl-furano [3, 4-d] isoxazole-4 (3aH) -one (5) was prepared by deacetylation. The structure of 3 was determined by X-ray analysis.  相似文献

The corresponding 2,3-substituted 4(3H)-quinazolinones were obtained in the reactions of 2-methyl- and 2-phenyl-4-oxo-3,1-benzoxazines with 1-amino-1,2,4-triazole, 4-amino-2,3-dimethyl-1-phenyl-5-pyrazolone, 2-amino-5-ethyl-1,3,4-thiadiazole, 3-amino-6,6-dimethyl-4-oxo-4,5,6,7-tetrahydroindazole, 1-amino-3-cyano-4,6-dimethyl-2-pyridone, and 1-amino-3-cyano-6-phenyl-4-trifluoromethyl-2-pyridone. The formation of N-benzolyanthranilamides in the reactions of 2-phenyl-4-oxo-3,1-benzoxazine with 2-amino-5-ethyl-1,3,4-thiadiazole and 1-amino-3-cyano-6-phenyl-4-trifluoromethyl-2-pyridones was exceptional. Malononitrile reacted with the title compound to give 6-amino-5-cyano-2-(3,3-dicyano-2-methylallylidene-4-methyl-2H-pyran (3). Treatment of 3 with hot 80% sulfuric acid yielded 4,7-dimethyl-56-hydroxy-2(1H)quinolone. With concentrated aqueous sodium hydroxide, 3 gave 5-amino-3,6-dicyano-4,7-dimethyl-2(1H)quinolone and 5-amino-6-carbamoyl-3-cyano-4,7-dimethyl-2(1H)quinolone. The reaction of 3 with hydrochloric in acetic acid gave a mixture of 6-amino-3,7-dicyano-2,8-dimethyl-4-quinolizone and 3-cyano-4-methyl-6-(3,3-dicyano-2-methylallyl)-2-pyrone. Compound 3 also reacted with methylamine, butylamine and piperidine to give 8-amino-5-cyano-4-methyl-2-pyridone, 6-bulylamino-5-cyano-4-methyl-2-pyridone and 5-eyano-4-methyl-6-piperidino-2-pyridone respectively.  相似文献

Abstract

In the present investigation, the authors could obtain a new series of spiranes (1) through the reaction of the high potential quinone tetrachloro-o-benzoquinone with 2-aryl-3-phenyl-3H-quinazoline-4-thiones. Thus, 2,3-diphenyl- (2a), 2-p-tolyl-3-phenyl- (2b) and 2-p-anisyl-3-phenyl- (2c)-3H-quinazoline-4-thiones react readily with tetrachloro-o-benzoquinone, in boiling toluene, to give the corresponding spiro-1,3-benzodioxole-2,4′-(3′H)-quinazolines (1a-c), respectively.  相似文献

4-Trifluoromethyl-6-phenyl-2-chloro-3-cyanopyridine was synthesized, and its molecular-crystal structure was established. The chlorine atom was replaced by amino, mercapto, thiocyanato, and azido groups. Condensation of 6-earbethoxy-4-hydroxy-2-pyridone or a silyl derivative of 5-earbomethoxy-4-hydroxy-2-pyridone with 2′,3′,5′-tri-O-benzoyl-D-ribofuranosyl halide has provided the 3-deaza analogs of orotidine and uridine-5-carboxylic acid. The corresponding amides have also been prepared in view of their possible structural relationship to l-β-D-ribohiranosyl nicotinamide. Tri-O-benzoyl-3-deazauridine was treated with N-bromosuccinimide to give, after deblocking, 3-bromo-4-hydroxy-1-(β-D-ribofuranosyl)-2-pyridone. The anomeric configuration of these nuclcosides was confirmed by pmr spectroscopy.  相似文献

Pirfenidone (5-methyl-1-phenyl-2(1H)-pyridone, PFD) is a small-molecule compound acting on multiple targets involved in pathological fibrogenesis and is effective to increase the survival of patients with fibrosis, such as idiopathic pulmonary fibrosis. However, PFD is not active enough, requiring a high daily dose. In this study, to keep the multiple target profiles, N?-substituted phenylhydroquinolinone derivatives, which retain the 1-phenyl-2(1H)-pyridone scaffold were designed and synthesized. The preliminary anti-fibrosis activities for all target compounds were evaluated on a NIH3T3 fibroblast cell line using MTT assay methods. Most compounds showed significant inhibition on NIH3T3 cell proliferation with a IC?? range of 0.09-26 mM, among which 5-hydroxy-1-(4'-bromophenyl)-5,6,7,8-tetrahydroquinolin-2(1H)-one (6j) displayed 13 times higher potency (IC?? = 0.3 mM) than that of AKF-PD (IC?? = 4.2 mM). These results suggest that N?-substituted phenylhydroquinolinone is a promising scaffold which can be applied for further investigation and for developing novel anti-fibrosis agents.  相似文献

Reactions of acetylenic ketones (Ia-f) with methyl α-naphthylacetate in the presence of sodium methoxide gave the corresponding 6-aryl-3-α-naphthyl-4-phenyl-2-pyrones (IIa-f) which upon refluxing with hydrazine hydrate in ethanol gave the corresponding 1-amino-2-pyridone derivatives (VIa-f). The spectrophotometric determination of vanadium(V) as a mixed thiocyanate-3-hydroxy-2-methyl-1-phenyl-4-pyridone (HX) complex and as a mixed thiocyanate-3-hydroxy-2-methyl-1-(4-tolyl)-4-pyridone (HY) complex is described. The extracted complexes in chloroform have a maximum absorbance at 450 and 650 nm. The optimal conditions for the extraction and spectrophotometric determination of vanadium(V) are determined. The solutions of the V-SCN-HX and V-SCN-HY complexes in chloroform obey Beer's law in the range 1–10 ppm of vanadium, and are stable for at least 24 hr. The molar absorptivity of the method is 6.8 × 103 liters mol?1 cm?1. The molar ratio V:SCN:HX (HY) of the extracted complex is 1:1:2.  相似文献

Nitration of 1-methyl-2-pyridone with 70% nitric acid or fuming nitric acid at 0–20°C proceeds in the 3 and 5 positions to give a mixture of mononitro isomers, the structures of which were proved by PMR spectroscopy. Three pathways were observed in the reactions of Schiff bases of Thiohydrazides with P(NR2)3. (a) MeS-R2N exchange: MeS-C([dbnd]S)-NHN[dbnd]CHPh (1) reacted with P(NR2)3 led to new Schiff bases, R2N-C([dbnd]S)-NHN[dbnd]CH = Ph (2). (b) Cleavage of C[dbnd]S bond and the formation of P[dbnd]S bond: H2N-C([dbnd]S)-NHN[dbnd]CH = Ph (3) reacted with P(NR2)3 gave rise to the thiophosphoric amide. (Et2N)2P([dbnd]S)-NH-CH = N-N[dbnd]CH-Ph (4). The splitting of the dihydropyridine ring of N-methyl-substituted 4-phenyl-5-oxo-4,5-dihydroindeno[1,2-b]pyridine in an acid medium takes place at the C-N bond.
