The plastic material known as cyclic olefin copolymer (COC) is a useful substrate material for fabricating microfluidic devices due to its low cost, ease of fabrication, excellent optical properties, and resistance to many solvents. However, the hydrophobicity of native COC limits its use in bioanalytical applications. To increase surface hydrophilicity and reduce protein adsorption, COC surfaces were photografted with poly(ethylene glycol) methacrylate (PEGMA) using a two-step sequential approach: covalently-bound surface initiators were formed in the first step and graft polymerization of PEGMA was then carried out from these sites in the second step. Contact angle measurements were used to monitor and quantify the changes in surface hydrophilicity as a function of grafting conditions. As water droplet contact angles decreased from 88 degrees for native COC to 45 degrees for PEGMA-grafted surfaces, protein adsorption was also reduced by 78% for the PEGMA-modified COC microchannels as determined by a fluorescence assay. This photografting technique should enable the use of COC microdevices in a variety of bioanalytical applications that require minimal nonspecific adsorption of biomolecules.  相似文献

The aim of this work is to join the advantages of two different kinds of stationary phases: monolithic columns and zirconia-based supports. On the one hand, silica monolithic columns allow a higher efficiency with a lower back-pressure than traditional packed columns. On the other hand, chromatographic stationary phases based on zirconia have a higher thermal and chemical stability and specific surface properties. Combining these advantages, a zirconia monolith with a macroporous framework could be a real improvement in separation sciences. Two main strategies can be used in order to obtain a zirconia surface on a monolithic skeleton: coating or direct synthesis. The coverage by a zirconia layer of the surface of a silica-based monolith can be performed using the chemical properties of the silanol surface groups. We realized this coverage using zirconium alkoxide and we further grafted n-dodecyl groups using phosphate derivatives. Any loss of efficiency was observed and fast separations have been achieved. The main advance reported in this paper is related to the preparation of zirconia monoliths by a sol-gel process starting from zirconium alkoxide. The synthesis parameters (hydrolysis ratio, porogen type, precursor concentration, drying step, etc.) were defined in order to produce a macroporous zirconia monoliths usable in separation techniques. We produced various homogeneous structures: zirconia rod 2 cm long with a diameter of 2.3 mm, and zirconia monolith inside fused silica capillaries with a 75 microm I.D. These monoliths have a skeleton size of 2 microm and have an average through pore size of 6 microm. Several separations have been reported.  相似文献

This article shows that there is great interest in using an electrochromatographic microchip made of hexyl acrylate (HA) based porous monolith cast within the channel of a cyclic olefin copolymer (COC) device. The monolith is simultaneously in situ synthesized and anchored to the inner walls of the channel in less than 10 min. By appropriate choice of light intensity used during the synthesis, the separation efficiency obtained for nonpolar solutes such as polycyclic aromatic hydrocarbons (PAH) is increased up to 250 000 plates/m. The performance of this HA‐filled COC microchip was investigated for a wide range of analytes of varying nature. The reversed‐phase separation of four aflatoxins is obtained in less than 2 min. The baseline separation of a mixture of neurotransmitters including six amino acids and two catecholamines is possible thanks to the superimposition of the differences in electrophoretic mobility on the chromatographic process. The durability of the system at pH 13 allows the separation of five biogenic amines and the quantitative determination of two of them in numerous wine samples. The feasibility of on‐line preconcentration is also demonstrated. Hydrophilic surface modification of COC channel via UV‐photografting with poly(ethylene glycol) methacrylate (PEGMA) before in situ synthesis of HA, is necessary to reduce the adsorption of very hydrophobic solutes such as PAH during enrichment. The detection limit of fluoranthene is decreased down to less than 1 ppb with a preconcentration of 4.5 h on the HA‐filled PEGMA functionalized COC microchip.  相似文献

As an alternative material to glass or silicon, microfluidic devices made from a cyclic olefin copolymer (COC) were fabricated. This material is of interest because of the relative ease of fabrication, low costs, and solvent resistance. However, as a result of the strong hydrophobic interactions normally present, COC surfaces are not suitable for protein separations. To reduce the protein adsorption and make COC suitable for protein separations, UV-initiated grafting of polyacrylamide was used to coat the surface of COC devices. The change in surface properties caused by different graft times was studied. The surface hydrophilicity and electroosmotic mobility were characterized by contact angle and electroosmosis measurements. Isoelectric focusing was performed to test protein separations in polyacrylamide-coated COC microchannels. A single protein, carbonic anhydrase, was used to analyze the focusing effects and peak capacities in uncoated and polyacrylamide-coated COC devices. Peak capacities ranging from 75 to 190 were achieved with a polyacrylamide-coated surface. A mixture of two proteins, conalbumin labeled with Alexa Fluor 488 and beta-lactoglobulin A labeled with Alexa Fluor 546, was used to test protein separations. Linear and rapid separation of proteins was achieved in the polyacrylamide-coated COC microfluidic device.  相似文献

A one-step nanocasting route has been demonstrated to prepare highly ordered single-crystal indium oxide nanowire (IONW) arrays with mesostructured frameworks. Unlike the reported multistep nanocasting process (synthesis of mesoporous materials, and then incorporation of precursors and formation of inorganic frameworks), a highly ordered mesostructured surfactant-silica monolith with low external surface serves as both the template and the reducing agent and makes the formation of single-crystal IONWs in its channels easily in one step by using normal In(NO(3))(3) as an inorganic precursor. After silica is removed, highly ordered uniform single-crystal IONW arrays with hexagonal (p6mm) or cubic (Ia3d) mesostructures are derived. These new materials are studied by XRD, SEM, TEM, N(2) adsoption, and UV spectrum. Furthermore, this one-step nanocasting synthesis route is a generalized method and can be used to synthesized a highly ordered mesoporous silica monolith with metal oxide nanocrystals in its channels. To the best of our knowledge, this is the first report of a single crystalline mesostructured In(2)O(3) framework.  相似文献

A new method of reproducible preparation of vinylic polymeric monolithic columns with a key step of covalently anchoring the monolith to PEEK surface is described. In order to chemically attach the polymer monolith to the tube wall, methacrylate functional groups were introduced onto PEEK surface by a three-step procedure, including surface etching, surface reduction and surface methacryloylation. The chemical state of the modified tubing surface was characterized by attenuated total reflectance infrared (ATR-IR) spectroscopy. It was found that the etching step is the key to successfully modifying the PEEK tubing surface. Poly(styrene-co-divinylbenzene) monoliths were in situ synthesized by thermally initiated free radical copolymerization within the confines of surface-vinylized PEEK tubings of dimensions close to ones conventionally used in HPLC and UHPLC (1.6 mm internal diameter, 10.0–12.5 cm length). Adhesion test was done by measuring the operating pressure drop, which the prepared stationary phases can withstand. Good pressure resistance, up to 140 bar/10 cm (flow rate 0.5 mL min−1, acetonitrile as a mobile phase), indicates strong bonding of monolith to the tubing wall. The monolithic material was proven to have a permeability of 1.7 × 10 −14 m2, applying acetonitrile–water 70:30 (v/v) as a mobile phase.  相似文献

Macroporous cross‐linked organic polymer based on N‐acryloxysuccinimide (NAS) and ethylene dimethacrylate (EDMA) was prepared inside 75 µm id fused silica capillary as a functionalizable monolithic stationary phase for chromatographic applications. Succinimide groups on the monolith surface provide reactive sites able to react readily through standard electrophile–nucleophile chemistry. Propargylamine was used to prepare alkyne functionalized poly(NAS‐co‐EDMA). Onto this azido‐reactive polymer surface was grafted β‐cyclodextrin (CD) via a triazole ring utilizing the copper(I)‐catalyzed 1,3‐dipolar cyclo‐addition reaction. Chemical characterization was performed in situ after each synthetic step by means of Raman spectroscopy. Good enantioseparations of flavanone enantiomers, chosen as test chiral compound, were achieved under reversed phase conditions by both capillary electrochromatography and nano‐liquid chromatography (nano‐LC) techniques. These results demonstrate the potentiality and usefulness of click chemistry in the preparation of β‐CD containing chiral organic polymer monolith.

In this work a new type of chiral monolith silica column was developed for the chiral separation by micro high-performance liquid chromatography (micro-HPLC). The chiral monolith column with a continuous skeleton and a large through-pore structure was prepared inside a capillary of 100 microm I.D. by a sol-gel process, and chemically modified with chiral selectors, such as L-phenylalaninamide, L-alaninamide and L-prolinamide, on the surface of the monolithic silica column. Based on the principle of ligand exchange, these chiral monolithic columns were successfully used for the separation of dansyl amino acid enantiomers, as well as hydroxy acid enantiomers by micro-HPLC. The chromatographic conditions, the enantioselectivity and the performance of columns are discussed.  相似文献

A microfluidic approach to generate hydrogel microstructures inside microchannels for controlled encapsulation of single cells was developed. The method was based on a modified microscope projection photolithography which allowed for the photopolymerization of poly(ethylene glycol) diacrylate (PEG-DA) inside microchannels. Uniform-sized hydrogel microstructures (~50 μm in diameter) were generated one by one with determined positions to encapsulate single cells without losing the viability. Cells of interest could be identified by any kinds of visible labels to be selectively encapsulated inside the formed hydrogel microstructures. Large-scale encapsulation of single cells was achieved with a relatively high efficiency of 80% and the viability of encapsulated cells could be guaranteed by removing the dead cells identified with Trypan blue. This method is simple, fast and convenient to pattern the microchannels with single cells for a wide range of cell-based applications. For demonstration, two intracellular enzyme assays of carboxylesterase were performed to investigate the distribution of enzyme concentrations and the kinetic information within the encapsulated single HepG2 cells.  相似文献

Polymer monolithic columns with controlled surface ligand density, providing stationary phase gradients within monolithic capillary columns, have been developed using photo-grafting through optical filters. Utilising commercially available cyclic olefin co-polymer (COC) films, the production of an optical filter capable of attenuating UV irradiation, in a tailored manner, was investigated. This novel optical filter was successfully applied to the surface modification of poly(BuMA-co-EDMA) monolithic columns in a multi-step grafting procedure. Fabricated columns were subjected to scanning capacitively coupled contactless conductivity (sC(4)D), to determine the distribution of the grafted functional groups, axially along the column. Further modification to produce a chelating stationary phase gradient of iminodiacetic acid (IDA) was demonstrated. To demonstrate the distribution of the IDA sites, a metal cation (Cu(2+)) was complexed to the IDA forming a chelate. Upon the formation of a complex of IDA with Cu(2+), an overall drop in conductive response was observed. The COC optical filter was also used in the fabrication of a grafted gradient of strong cation exchanger (SCX), sulphopropyl methacrylate (SPM) upon a polymer monolith, demonstrating the broader applicability of such a filter.  相似文献

榉木经高温热解转化为生物碳模板, 通过液相渗硅反应工艺制备了保持木材微观结构的多孔SiC陶瓷. 在生物形态多孔SiC陶瓷载体上采用原位沉积晶种-二次生长法在其微孔道内壁形成了一层5 μm厚的Silicalite-2分子筛膜. 利用XRD, SEM和BET对复合材料的相组成、微观结构和比表面积进行了表征, 研究了水热晶化温度对原位沉积晶种和二次生长成膜的影响. 经原位沉积(120 ℃, 36 h)晶种涂层后在载体孔道表面形成了一层球形颗粒堆积的连续晶种层, 经170 ℃, 36 h的二次生长, 晶种不断长大并交织生长形成连续致密单层分子筛膜. 在多孔SiC陶瓷微孔道中沿垂直于载体表面方向形成了一层对齐排列的Silicalite-2棒状晶体, 颗粒生长主要沿晶体的最长轴[101]方向进行. Silicalite-2/SiC复合孔结构材料的微孔体积为0.013 cm3/g, BET比表面积为43.2 m2/g, 而相应的分子筛负载量为9.5%.  相似文献

We report the self-assembly of insulin monomers into amyloid fibrils within microchannels. To demonstrate the microfluidic amyloid formation and fibril growth on a solid surface, we seeded the internal surfaces of the microchannels with insulin monomers via N-hydroxysuccinimide ester activation and continuously flushed a fresh insulin solution through the microchannels. According to our analysis using optical and fluorescence microscopy, insulin amyloid preferentially formed in the center of the microchannels and, after reaching a certain density, spread to the side walls of the microchannels. By using ex situ atomic force microscopy, we observed the growth of amyloid fibrils inside the microchannels, which occurred at a much higher rate than that in bulk systems. After 12 h of incubation, insulin formed amyloid spherulites having "Maltese cross" extinction patterns within the microchannels according to the polarized microscopic analysis. Microfluidic amyloid formation enabled low consumption of reagents, reduction of incubation time, and simultaneous observation of amyloid formation under different conditions. This work will contribute to the rapid analysis of amyloid formation associated with many protein misfolding diseases.  相似文献

Sample pretreatment is a required step in proteomics in order to remove interferences and preconcentrate the samples. Much research in recent years has focused on porous monolithic materials since they are highly permeable to liquid flow and show high mass transport compared with more common packed beds. These features are due to the micro-structure within the monolithic silica column which contains both macropores that reduce the back pressure, and mesopores that give good interaction with analytes. The aim of this work was to fabricate a continuous porous silica monolithic rod inside a heat shrinkable tube and to compare this with the same material whose surface has been modified with a C(18) phase, in order to use them for preconcentration/extraction of proteins. The performance of the silica-based monolithic rod was evaluated using eight proteins; insulin, cytochrome C, lysozyme, myoglobin, β-lactoglobulin, ovalbumin, hemoglobin, and bovine serum albumin at a concentration of 60 μM. The results show that recovery of the proteins was achieved by both columns with variable yields; however, the C(18) modified silica monolith gave higher recoveries (92.7 to 109.7%) than the non-modified silica monolith (25.5 to 97.9%). Both silica monoliths can be used with very low back pressure indicating a promising approach for future fabrication of the silica monolith inside a microfluidic device for the extraction of proteins from biological media.  相似文献

Building up longitudinal concentration gradients in shallow microchannels   总被引:1,自引:0,他引:1  
We demonstrate a compact and low consumption (60 nL) method for generating concentration gradients along microchannels with shallow parabolic cross-sections. The regimes of dispersion at work in such systems and the resulting concentration fields are described theoretically and experimentally. Experiments are performed in PDMS (polydimethylsiloxane) microchannels actuated by integrated valves. Detailed comparison between theory and experiment for the "short time" and "long time" regimes leads to excellent agreement. The system is used to successfully set up a series of isolated microchambers with mixtures of increasing solute concentrations, which may be a first step towards devices for screening.  相似文献

采用双模板法,向正硅酸甲酯的水解体系中同时引入聚乙二醇和三嵌段共聚物,成功制备出具有双连续大孔、同时孔壁中分布着有序介孔的复合孔结构硅胶独石材料. 产物的比表面积高达880 m2/g, 大孔孔径为0.2~5 μm, 介孔高度集中地分布在 5 nm. 结合物理吸附、扫描电镜、粉末X射线衍射和透射电镜等表征手段,发现合成条件如原料组成、反应温度和pH值等对反应体系中凝胶化转变和相分离发生的相对速度有重要影响,进而影响产物复合孔结构的生成. 此外,通过对合成条件的优化,一方面增强了无机骨架的强度,另一方面降低了湿凝胶干燥过程中的毛细管压力降,有效缓和了凝胶结构在干燥过程中的开裂和变形,使复合孔结构硅胶独石在厘米尺度内具有良好的整体性能.  相似文献

Chemical characterization was performed in situ after each synthetic step by means of Raman spectroscopy and grafting kinetics was investigated to ensure quantitative grafting of 1‐adamantanethiol. The as‐designed monolithic stationary phase exhibited typical reversed‐phase separation mechanism as evidenced by the linear increase of the logarithm of retention factor of neutral aromatic solutes with the increase of the aqueous buffer content in the mobile phase.  相似文献

Silyl glyoxylates are proposed here as high‐performance photoinitiators (PIs) for the hybrid polymerization of cationic and radical monomers. Recently, silyl glyoxylates were reported as a new class of high‐performance Type I photoinitiators for free radical polymerization under air upon exposure to different near‐UV and blue LEDs. In this article, we report this new class of photoinitiators to initiate cationic polymerization in combination with an iodonium salt. This system can also be used to initiate simultaneously free radical and cationic polymerizations, for example, for the free radical/cationic hybrid polymerization and for the synthesis of interpenetrating polymer networks. The system silyl glyoxylate/iodonium exhibits excellent polymerization performances and exceptional bleaching properties compared to other well established photoinitiators (e.g., camphorquinone). Furthermore, a hybrid monomer is also introduced in this article (2‐vinyloxyethoxyethyl methacrylate [VEEM]) leading to a huge improvement of the mechanical properties of the final polymer through hybrid polymerization. © 2019 Wiley Periodicals, Inc. Polymer monoliths in capillary format have been prepared as solid supports for the immobilisation of platinum/palladium bimetallic nano-flowers. Optimum surface coverage of nano-flowers was realised by photografting the monoliths with vinyl azlactone followed by amination with ethylenediamine prior to nano-particle immobilisation. Field emission SEM imaging was used as a characterisation tool for evaluating nano-particle coverage, together with BET surface area analysis to probe the effect of nano-particle immobilisation upon monolith morphology. Ion exchange chromatography was also used to confirm the nature of the covalent attachment of nano-flowers on the monolithic surface. In addition, EDX and ICP analyses were used to quantify platinum and palladium on modified polymer monoliths. Finally the catalytic properties of immobilised bimetallic Pd/Pt nano-flowers were evaluated in flow-through mode, exploiting the porous interconnected flow-paths present in the prepared monoliths (pore diameter~1–2?μm). Specifically, the reduction of Fe (III) to Fe (II) and the oxidation of NADH to NAD+ were selected as model redox reactions. The use of a porous polymer monolith as an immobilisation substrate (rather than aminated micro-spheres) eliminated the need for a centrifugation step after the reaction.

In this report, we are documenting the synthesis of peptide barbiturate through copper-catalyzed oxidation of peptide disulfide. Single disulfide-containing contryphans are used as models to access possibility of anchoring of barbituric acid (BRB) onto the peptide disulfide. Current method permits anchoring of two molecules of BRB onto the polypeptide and yield of peptide barbiturates varies from 59 to 84%. Formation of cysteine sulfenic acid (Cys-SOH) during oxidation of disulfide was confirmed using chemical probe of Cys-SOH dimedone. Mass spectrometric studies have confirmed the presence of cysteine barbiturate in anchored peptides. Based on the nature of reactive oxygen species involved in oxidation of peptide disulfide, possible mechanisms were proposed for anchoring of BRB onto the peptide disulfide through Cys-SOH. This is the first report on anchoring of two molecules of BRB onto the closely spaced cysteine residues of single polypeptide.  相似文献

UV-initiated grafting of plastic tubes and microfluidic chips with ethylene diacrylate followed by the preparation of porous polymer monoliths has been studied. The first step affords a thin grafted layer of polymer with a multiplicity of pendent double bonds that are then used in the second step for covalent attachment of the monolith to the wall. As clearly seen on scanning electron micrographs, this procedure prevents the formation of voids at the monolith-channel interface a problem that has always plagued approaches involving bulk polymerization in nontreated channels due to the shrinkage of the monolith during the polymerization process and its lack of compatibility with the material of the device. Irradiation with UV light through a photomask allows precise patterning specifying both the area subjected to surface modification and the location of the monolith within specific areas of the device.  相似文献
