An efficient solid-phase synthesis of 2-substituted 4-aminopyrido[2,3-d]pyrimidines 12 by cyclization-assisted cleavage from resin is reported. The procedure starts by solid supporting an ,-unsaturated acid 8 to the Wang resin 13 by using DCC and 4-DMAP in THF. The resulting ,-unsaturated ester 14 is converted to the Michael adduct by treatment with malononitrile in NaOMe/THF. Such Michael addition constitutes the first example of a Michael reaction with malononitrile in solid-phase. Finally, the Michael adducts 15 are treated with an amidine system in MeOH to yield the corresponding pyridopyrimidines 12. Compounds 12 present three diversity centers R1, R2 and G. Having validated the chemistry on solid support, a 40-membered combinatorial library was obtained using this protocol.  相似文献

A protocol for the synthesis of N-substituted 2-hydro-4-amino-pyrido[2,3-d]pyrimidin-7(8H)-ones (11) is described. Thus, the formylation of a 2-aminopyridone 12 in 85% formic acid/Ac2O, proceeding via in situ cyclization to the intermediate formamide 13, affords the corresponding 2-hydro-4-oxo-pyridopyrimidine 14, which is converted to a 4-chloro-pyridopyrimidine 15 upon treatment with POCl3. The subsequent transformation to the title compounds is carried by treatment with the corresponding amine in MeOH under microwave irradiation conditions.  相似文献

A series of pyrido[2,3-d]pyrimidine derivatives and related compounds were synthesized via the condensation reaction of an aldehyde, 2,6-diaminopyrimidine-4(3H)-one and either tetronic acid or 1,3-indanedione under ultrasonic irradiation without catalyst. This protocol has the advantages of higher yields, lower cost and convenient procedure.  相似文献

Under solvent-free high-speed ball milling, an I\(_{2}\)-promoted condensation/cyclization of easily available methyl ketones or 1,3-dicarbonyl compounds with 2-aminopyridines has been developed, which allows the quick assembly of 2,3-substituted imidazo[1,2-a]pyridines (IPs) with broad molecular diversity, including the antiulcer drug zolimidine. The advantages of high yields, good functional group compatibility, short reaction time (within 90 min), free use of heating, solvent and metal, employment of cheap starting materials, and simple work-up procedure make this protocol a very efficient alternative to traditional synthesis of IPs.  相似文献

采用不同方法制备了CO2-3替换的磷灰石固溶体,利用FTIR结合XRD对其进行了晶体化学研究,结果表明均相沉淀法制备的碳羟磷灰石(CHAP)属B型替换且替换方式是[CO3·OH]四面体替换[PO4]四面体;固相离子交换法制备的CHAP属A型替换且替换方式是[CO3]三角形配位体替换通道位置的OH-;固相反应法制备的碳氟磷灰石(CFAP)属B型替换,其替换方式是[CO3·F]四面体替换[PO4]四面体;sol-gel法制备的CHAP属AB混合型替换,其Ψ3分裂为Ψ3-1,Ψ3F,Ψ3-4.高斯函数法拟合表明Ψ3F峰是A型替换的Ψ3-2与B型替换的Ψ3-3的叠合.当WCO2-3<3.34%时,随CO2-3含量增加,A型替换量增大,且当WCO2-3=3.34%时达最大值,当3.34%<WCO2-3<7.52%时,随CO2-3含量增加,B型替换量增大,且当WCO2-3= 7.52%时亦到饱和.  相似文献

Summary The phosphorus ylide obtained from the reaction between 2-aminobenzo[d]isothiazol-3-one and dimethyl acetylenedicarboxylate in the presence of triphenylphosphine undergoes a smooth intramolecular Wittig-type reaction to produce, in a one-pot reaction, 3-H-benzo[d]pyrazolo[1,5-b]isothiazole-2,3a-dicarboxylic acid dimethyl ester, a novel functionalized heterocyclic compound.  相似文献

A microwave-assisted method is described for monoacylating 7-amino-5-aryl-6-cyanopyrido[2,3-d]pyrimidines using excess acid chlorides in pyridine. A diacylated intermediate is effectively deacylated to the product amide by a macroporous-Tris resin. A small library of 17 amides was prepared to validate the method. A practical protocol was developed for the synthesis of 2-arylamino substituted 4-amino-5,6-dihydropyrido[2,3-d]pyrimidin-7(8H)-ones from ??,??-unsaturated esters, malononitrile, and an aryl substituted guanidine via the corresponding 3-aryl-3,4,5,6- tetrahydropyrido[2,3-d]pyrimidin-7(8H)-ones. Such compounds are formed upon treatment of 2-methoxy-6-oxo-1,4,5,6-tetrahydropyridine-3-carbonitriles with an aryl substituted guanidine in 1,4-dioxane and are converted to the desired 4-aminopyridopyrimidines with NaOMe/MeOH through a Dimroth rearrangement. The overall yields of this three-step protocol are, generally speaking, higher than the multicomponent reaction, previously developed by our group, between an ??,??-unsaturated ester, malononitrile, and an aryl substituted guanidine.  相似文献

Two novel ESIPT molecules, 2-[4-(1,3-benzothiazol-2-yl)naphtho[1,2-d][1,3]oxazol-2-yl]phenol 9a and 4-[4-(1,3-benzothiazol-2-yl)naphtho[1,2-d][1,3]oxazol-2-yl]benzene-1,3-diol 9b were synthesized by condensing 1-amino-3-(1,3-benzothiazol-2-yl)naphthalen-2-ol with 2-hydroxybenzoic acid and 2,4-dihydroxybenzoic acid respectively. The novel compounds were characterized by FT-IR, (1)H NMR, Mass spectral and elemental analysis. Effect of polarity on photo physical properties, absorption and emission were studied. Compounds showed single absorption and dual emission due to ESIPT phenomenon. The structural changes due to ESIPT phenomenon in terms of bond angle, bond distances and geometry were investigated by using Gaussian 03 software. These two novel ESIPT molecules are thermally stable up to 200?°C.  相似文献

使用DFT的B3LYP方法对几种咪唑二氧杂环化合物的分子结构、红外光谱、生成焓、爆轰性能和化学/热稳定性进行了研究.四种不同含能基团-NO2,-NH2,-N3和-ONO2对该化合物各项性能的影响进行了比较.结果表明-NO2和-ONO2基团有效地增加了化合物的密度,而-N3基团极大地增加了化合物的生成焓.其中-NO2取代物爆轰性能接近1,3,5,7-tetranitro-1,3,5,7-tetraazacyclooctane,-ONO2取代物的爆轰性能低于hexahydro-1,3,5-trinitro-1,3,5-triazine.自然键轨道分析表明,这些化合物中相对较弱的键为取代基和咪唑环之间的键,以及二氧杂环中的C-O键.吸电子基团(-NO2, -N3和-ONO2)在分子中产生了明显的诱导效应,减弱了基团与咪唑环之间的链接,降低了对应键的键裂解能.研究表明基团的电负性与化合物的稳定性有关.考虑到化合物的爆轰性能和热稳定性,DNTNDI化合物满足高能量密度材料的要求.  相似文献

Reactions of 2,2-dialkylaldehydes with electron-rich 2-naphthols and para-substituted phenols in presence of catalytic amount of $p$ -TSA under closed vessel solvent-free microwave irradiation conditions resulted in formation of corresponding 2,2-dialkyl-1,2-dihydronaphtho[2,1- $b$ ]furans and 2,2-dialkyl-2,3-dihydrobenzofurans, respectively, in good to excellent yields. The effect of stoichiometry, temperature, and catalyst in reaction progress was systematically investigated. 14-Alkyl- $14H$ -dibenzo[ $a,j$ ]xanthenes was obtained as minor products when 2-naphthol and 6-bromo-2-naphthols were used as starting phenols. Simple phenols gave a lower yield of the 2,2-dialkyl-2,3-dihydrobenzofurans products than their electron-rich naphthalene counterparts. Also, xanthene-type products were not detected in case of simple phenols by GC–MS or column chromatography.  相似文献

A one-pot three-component cascade reaction for the green synthesis of a new class of 2-amino-5,8-dihydro-3H-pyrido[2,3-d]pyrimidin-4-ones was developed from the condensation of aromatic aldehydes with 2,6-diaminopyrimidin-4(3H)-one and acetophenone derivatives or various cyclic ketones in the presence of a catalytic amount of sodium carbonate in a mixture of water and ethanol at 60  \({^{\circ }}\hbox {C}\). This study describes the synthesis and anti-inflammatory effects of furo[3', 2':3,4]naphtho[1,2-d] imidazole derivatives. Among these furo[3', 2':3,4]naphtho[1,2-d]imidazole derivatives, 2-(4-methoxyphenyl)furo [3', 2':3,4]naphtho[1,2-d]imidazole (12) exhibited a strong inhibitory activity against LPS-induced PGE(2) production, with an IC(50) value of 47?nM. Compound 12 is then further examined for its inhibitory effects in the protein expression of COX-2 and microsomal prostaglandin E(2) synthase-1 (mPGES-1) in Raw 264.7 cells. Our results indicate that compound 12 was capable against inhibiting LPS-induced mPGES-1 protein expression at a concentration of 1.0?μM and no inhibitory effect in COX-2 expression. The sepsis-induced PGE(2) production in rat serum decreased ~250% by the pretreatment of 12 at 10?mg/kg. These results are especially important since compound 12 exhibited good oral bioavailability (72%) and was not cytotoxic at a concentration of 10.0?μM. Therefore, compound 12 is a highly selective mPGES-1 inhibitor that can serve as a lead for the development of novel oral anti-inflammatory drug candidates.  相似文献

利用原位红外光谱法研究标题化合作在N2 中的反应行为。发现它们在不同温度下能使丙酸分别转化为乙醛和戊酮。金属氧化物对丙酸的戊酮化反应活性顺序是 :Fe3O4 >MnO2 >Cr2 O3>Co2 O3>Ni2 O3,这顺序与下列簇合物对该反应的活性顺序类似 :[Fe2 MnOPH]>[Fe3OPH]>[Fe2 CrOPH]>[Fe2 CoOPH]>[Fe2 NiOPH]>[Cr3OPH][OPH =(μ3 O) (μ O2 CC2 H5) 6(H2 O) 3]。对于戊酮化反应 ,过渡金属氧化物的反应温度明显高于相应的 [Fe2 MOPH]的反应温度 ,这旁证了 [Fe2 MOPH]的活性组分不是其金属氧化物。还讨论了Fe3O4 ,Fe2 O3 对丙酸分别转化为乙醛和戊酮反应活性的明显差异。  相似文献

This study evaluated how the generation of pyrazines was promoted by high-intensity ultrasound (HIU) in a Maillard reaction (MR) model system of glucose-glycine. Carbohydrate module labeling (CAMOLA) technique was adopted using D-glucose-13C6 to elucidate the carbon skeleton of both intermediate and final MR products (MRPs). In the D-glucose-13C6-glycine HIU-MR model system, the concentration of 11 types of pyrazines was significantly higher than their counterparts in the thermal MR. Results of CAMOLA analysis showed that a significantly lower proportion of [M]+ in pyrazines with long-length side chains was observed when compared with the pyrazines generated in thermal MR. This phenomenon may suggest the aldol-type condensation was promoted by the HIU, which is a conversion from pyrazines with short-length side chains to those with long-length side chains involving carbonyl compounds. Furthermore, the analysis of isotopomers distribution in 2,3-dimethyl-quinoxaline as the o-phenylenediamine-derivatized 2,3-butanedione indicated that the increased proportion of [M + 4]+ in 2,3-dimethyl-quinoxaline (15.74% ± 0.11%) was attributed to a cleavage of D-glucose-13C6 promoted by the HIU. The above-mentioned findings elucidate that the aldol-type condensation and cleavage of D-glucose contribute to the promoted synthesis of pyrazines. The HIU would generate an extremely high temperature and pressure environment that is favored by the aldol-type condensation as a high-pressure favored reaction. The HIU, therefore, can be further developed as a promising technique to promote flavor generation through the MR.  相似文献
