2,4,6-trichloroanisole (TCA) is a microbial metabolite formed from chlorophenols through the activity of several natural fungal strains present on the cork oak bark. TCA is the primary compound responsible for the mousty/mould off-odour known as “cork taint” present in cork stoppers, wine, water and alcoholic beverages. Chromatographic and electrochemical methods are currently used for the determination of TCA, however its detection at low concentrations remains a technical challenge. The aim of this study was the development of a rapid novel biosensor system based on the Bioelectric Recognition Assay (BERA). The sensor measured the electric response of cultured membrane-engineered fibroblast cells suspended in an alginate gel matrix due to the change of their membrane potential in the presence of the analyte. Membrane-engineered cells were prepared by osmotic insertion of 0.5 μg/l of specific TCA antibodies into the membrane of the cells. The BERA-based sensor was able to detect TCA in a few minutes (3-5 min) at extremely low concentrations (10−1 ppt), thus demonstrating higher sensitivity than the human sensory threshold. In addition, the assay was quite selective against other haloanisoles and halophenols structurally related to or co-occurring with TCA. Finally the sensor was tested against real white wine samples from cork soaks. At this real test, the BERA sensor was able to detect TCA from cork soaks rapidly (3-5 min) at very low concentrations (1.02-12 ng/l), covering the whole range for the detection threshold for wines (1.4-10 ng/l). Therefore, this novel biosensor offers new perspectives for ultra-rapid, ultra-sensitive and low-cost monitoring of TCA presence in cork and wine and possibly also other food commodities.  相似文献

A rapid and simultaneous method for residue identification and quantification for seven pesticides in agricultural soils has been developed to study a realistic situation in vineyard. The target compounds are two insecticides, two herbicides and three fungicides, from different chemical families. The procedure is based on a pressurized liquid extraction (PLE) with acetone, before a multiresidue GC-MS analysis. The recovery of PLE is between 53.8 ± 2.4 and 99.9 ± 4.4% according to pesticide. A limit of detection (LOD) between 1.4 and 4.6 μg kg−1 of dry soil was obtained for five analytes. This procedure for testing soil contamination is sensitive and easy to perform.  相似文献

由于毛细管电泳(CE)和毛细管电色谱(CEC)具有所需样品体积小、分离效率高等特点,越来越多的学者已将它们应用到农药残留(简称农残)检测中,并将它们同各种不同的检测器以及样品浓缩方法相结合,以提高检测的灵敏度。本文对CE和CEC两种方法中所涉及的常见的样品预浓缩方法进行了简要的介绍。对各种不同类型的检测器(如紫外检测、荧光检测、电化学检测以及质谱检测等)的优缺点及其在农残检测中的应用情况进行了评述;同时对手性农药的CE和CEC分离检测情况进行了特别介绍;并对CE和CEC在农残分析与检测中的应用前景进行了展望。  相似文献

A method was described for simultaneous determination of nine organic heterocyclic pesticide residues by gas chromatography-mass spectrometry-selected ion monitoring. Atrazine, vinclozolin, procymidone, triflumizole, imazalil, buprofezin, propiconazole, fenarimol, and pyridaben were clearly separated from each other, extracted with acetone—hexane mixture, purified with graphitized carbon black cartridge and neutral Al2O3 cartridge, eluted with acetone—hexane mixture, simultaneously determined by GC-MS, and then quantified with an external standard method. Recoveries of pesticides ranged from 73 % to 116 % at the spiked level of 0.01–30 mg kg−1, while the relative standard deviation was between 3 % and 27 %. In addition, the limits of determination (0.01 mg kg−1 to 5.0 mg kg−1) and linearity (0.02–40 μg mL−1) revealed that simultaneous determination of multi-residues in Chinese teas (like Oolong tea, green tea, red tea, etc.) was possible. Furthermore, an interlaboratory study among 5 labs was conducted to further validate the method, and the results were satisfactory.  相似文献

建立了固相萃取(SPE)-反相高效液相色谱(RP-HPLC)同时测定橙子中痕量辛硫磷、二嗪农有机磷农药残留量的方法。样品经甲醇超声提取、C18固相萃取柱净化后,采用液相色谱柱分离,以乙腈-水(体积比为85∶15)为流动相等度洗脱,于254 nm下紫外检测。结果表明:在0.1~10.0 mg/L和0.4~10.0 mg/L范围内,辛硫磷、二嗪农的质量浓度与峰面积呈良好的线性关系;样品的加标平均回收率为87.3%~102.7%,相对标准偏差(RSD)为0.9%~4.9%。将该分析结果与用基质固相分散法(MSPD)处理样品所得的结果相比较,发现SPE对二嗪农的提取效果较好,而MSPD对辛硫磷的提取效果较好,但两种方法都能较好地净化样品,均能满足残留量的分析要求。  相似文献

在系统优化固相萃取吸附剂填料类型、洗脱溶剂种类及体积的基础上,建立了蔬菜和水果中193种农药残留的气相色谱-质谱(GC-MS)检测方法。样品经乙腈均质提取,C18/PSA固相萃取柱净化,乙腈洗脱,GC-MS选择离子监测(SIM)模式检测,以磷酸三苯酯内标法定量。结果表明,130种农药在10~1000 μg/L、34种农药在20~1000 μg/L、26种农药在50~1000 μg/L、3种农药在100~1000 μg/L范围内线性关系良好,相关系数为0.9967~1.0000,方法检出限(以信噪比为3计)为0.04~8.26 μg/kg,添加回收率为71.6%~117.9%,相对标准偏差为3.0%~11.8%。该方法样品处理简单快速,相比其他多残留分析方法净化效果好,其灵敏度和选择性明显提高,适用于日常检测工作。  相似文献

建立了果蔬中吡虫啉、咪鲜胺、苯醚甲环唑、嘧菌酯、噻虫嗪等16种常见农药多残留的液相色谱-串联质谱(LC-MS/MS)分析方法。以乙腈为提取剂,样品经高速匀浆方法提取后,提取液采用液-液萃取静置分层,取上清液进行净化处理。比较了石墨化碳-氨基复合固相萃取与QuEChERS两种不同净化技术的净化效果,最终确定采用QuEChERS方法为净化手段。即提取液经装有150 mg N-丙基乙二胺(PSA)填料、900 mg无水硫酸镁的净化管净化,除去样品中大部分的色素及有机酸等干扰基质,再经LC-MS/MS分析,有效地降低了样品中的复杂基质所带来的背景干扰。加标水平为5、10、20 μg/kg时,16种农药的平均回收率为75%~111%,相对标准偏差小于16%。16种农药的检出限为0.2~5 μg/kg。采用LC-MS/MS定性分析、基质匹配标准曲线法定量分析,线性关系和回收率结果均令人满意。实验证明,建立的QuEChERS净化与LC-MS/MS相结合的检测方法具有快速、准确、灵敏度高等优点,能够准确测定果蔬中16种农药残留。  相似文献

采用分散固相萃取-气相色谱-串联质谱(QuEChERS-GC-MS/MS)建立了蔬菜中107种农药残留量的分析方法。样品由含1%冰醋酸的正己烷饱和乙腈提取、分散固相萃取法净化,采用气相色谱-串联质谱方法在分时段选择反应监测模式下进行测定,外标法定量。所有农药在0.05~1 mg/L范围内线性关系均良好;所有农药的方法定量限(LOQ)均低于10 μg/kg;在10 μg/kg的添加水平下,大蒜、青刀豆、萝卜和菠菜4种基质中绝大多数农药的平均回收率处于60%~130%之间,相对标准偏差(RSD)不大于15.3%。该方法不仅能用于多种蔬菜基质中107种农药残留的检测,而且还能较好地解决本底成分相当复杂的大蒜基质极易出现的干扰问题。  相似文献

A multiresidue method was studied for determination of 323 pesticides representing a wide range of physicochemical properties in dry herbs (chamomile and parsley) based on QuEChERS method and LC-ESI-MS/MS analysis. In the current study, three different parameters were optimised for a higher accuracy and a lower matrix effects: extraction procedures, cleanup and matrix effect. Optimum extraction efficiency was obtained at sample hydration of 10 mL water on 2 g dry herbs and soaking time for 10 min. Use of different extraction techniques supported the use of mechanical shaker in comparison with ultrasound and handed shaking. As a way to remove interfering components from final extract, different cleanup techniques were studied: three dispersive solid phase extraction (D-SPE) (C18, primary secondary amine and graphitised carbon black) have been investigated; matrix effect was reduced, but several pesticides were lost during the cleanup process. Unlike the SPE (hydrophilic lipophilic balanced polymer) produced a good recovery with all substances without expected reduction in matrix effect. Moreover, sample dilution and LC-MS/MS injection volume were studied, 3 µL injection volume was found to be the best sensitive condition without sample dilution. The developed method was validated by performing recovery tests at 0.01, 0.05 and 0.1 mg/kg, the average recoveries ranged from 70% to 120%. The reproducibility expressed as relative standard deviation (RSD %) was ≤ 20%.  相似文献

分别采用电子轰击电离(EI)源和正化学电离(PCI)源两种离子源技术建立了气相色谱-质谱(GC-MS)同时测定白菜和苹果中苯丁锡、三苯锡和三环锡含量的方法,并对这两种方法进行了比较。样品经氢溴酸消解、丙酮-正己烷(1∶2,v/v)提取,四乙基硼化钠衍生后用Florisil固相萃取柱净化,分别在EI源和PCI源下以选择离子监测模式进行测定。结果表明,方法的检出限(S/N=3)分别为0.01~0.05 mg/kg(EI源)和0.01~0.02 mg/kg(PCI源),定量限(S/N=10)分别为0.03~0.16 mg/kg(EI源)和0.02~0.06 mg/kg(PCI源)。三苯锡、三环锡和苯丁锡分别在各自的线性范围内线性关系良好,相关系数(r~2)≥0.997。在50、100、200μg/kg 3个添加水平下,采用GC-EI/MS和GC-PCI/MS时,阴性样品中3种有机锡的平均回收率分别为59.24%~97.36%(苹果)、50.54%~94.54%(白菜)和65.38%~95.86%(苹果)、62.56%~90.44%(白菜),相对标准偏差(RSD)均不超过6.9%(n=6)。该方法简单、灵敏,PCI源的选择性优于EI源,两种方法可以相互结合提高检测结果的可靠性。  相似文献

评价了分析过程中不同条件下使用分散固相萃取(D-SPE)材料净化对农残分析定量结果可靠性的影响,表明通过条件优化,可明显减小定量误差,并取得满意的回收率。建立了蔬菜中112种农药的多反应监测-气相色谱-串联质谱(GC-MS/MS)检测方法。在pH 5~7范围内,样品经乙腈-甲苯(8:1, v/v)匀浆提取,每5 mL提取液加入0.8 g无水硫酸镁、0.05 g石墨化炭黑(GCB)、0.1 g乙二胺-N-丙基甲硅烷(PSA)、0.05 g C18粉末,分散固相萃取法净化,采用气相色谱-三重串联四极杆质谱(GC-QQQ-MS/MS)在多反应监测模式(MRM)下进行测定,内标法定量。分别对韭菜、黄瓜、紫甘蓝进行3个水平的加标回收试验(20、50、200 μg/kg),其回收率范围为53.1%~138.7%,其中86种农药的3个加标水平的回收率范围均为65.0%~120.0%,方法的相对标准偏差(RSD)小于12%,方法的定量限(LOQ)范围为1.6~13.4 μg/kg。对从市场采集的蔬菜样品进行了测定,检出了三唑磷、甲氰菊酯等农药残留。该方法样品前处理简单快速,灵敏度、准确度和精密度均符合农药多残留检测技术的要求,适用于蔬菜中112种农药残留的快速筛查测定。  相似文献

A new method was developed and validated for the simultaneous determination of 151 pesticide residues in strawberry by gas chromatography coupled to a triple quadrupole mass analyzer (GC/QqQ-MS/MS), mainly using the selected reaction monitoring (SRM) mode. The list of target compounds included various classes of pesticides such as organochlorine (OCPs), organophosphorus (OPPs), carbamates, pyrethroids, triazoles and dicarboximides. A single extraction of 10 g of sample with acetonitrile followed by liquid-liquid partition formed by the addition of 4 g of MgSO4 and 1 g of NaCl was applied in sample preparation. Cleanup of the extracts was carried out by applying dispersive solid-phase extraction (D-SPE) with primary secondary amine (PSA). The analysis time was 21 min. The method was subjected to a thorough validation procedure. The recovery data were obtained by spiking blank samples at two concentration levels (11.5 and 50 microg/kg), yielding recoveries in the range 70-110%. Precision values expressed as relative standard deviation (RSD) were lower than 18% and 22% for the intraday and interday precision, respectively. Linearity was studied in the range 10-200 microg/kg and determination coefficients (R(2)) were higher than 0.98% for all compounds. Limits of detection (LODs) and limits of quantification (LOQs) were established as 4 and 10 microg/kg, respectively. The overall uncertainty of the method was estimated at two different concentrations (11.5 and 50 microg/kg), being lower than 25% in both cases. According to the validation data and performance characteristics as well as the high sample throughput and low cost, the proposed method is suitable for routine application.  相似文献

建立了河豚鱼、鳗鱼和对虾中191种农药多残留的气相色谱-质谱分析方法。样品用乙酸乙酯-环己烷(体积比为1:1)均质提取,凝胶渗透色谱净化,收集26~44 min的流出液并进行在线浓缩,通过气相色谱柱(DB-1701)分离后在选择离子监测(SIM)模式下进行质谱检测。分别以最低定量限和4倍最低定量限为添加浓度对河豚鱼、鳗鱼和对虾样品进行了两个水平的添加回收率实验,方法的回收率范围为50.2%~120%,其中89.5%的农药的回收率为70%~120%,相对标准偏差范围为0.6%~21.6%。方法的最小检出限和最低定量限范围分别为0.002~0.3 mg/kg和0.007~1.2 mg/kg。该方法的灵敏度、准确度和精密度均符合农药多残留检测的技术要求,适用于河豚鱼、鳗鱼和对虾等动物源性水产品中多种农药残留的检测。  相似文献

建立了一种新型农药多残留检测方法。采用气相色谱-四极杆飞行时间质谱(GC-QTOF MS)结合Carbon NH2柱固相萃取净化可对不同类别的152种农药残留进行准确的鉴定。一级(MS1)精确质量数据库评价3~5个特征离子,以3个及以上特征离子检出且离子丰度比(Q/qi)合适作为一级质谱定性依据。对部分特征离子数不足及特征离子检出情况不符合条件的农药进行二级(MS2)确证,应用二级谱图库检索及镜像结果对比分析实现疑似农药确证。本文对梨、甘蓝、番茄3种代表性果蔬在5.0和10.0 μg/kg 2个添加水平的152种农药进行测定,回收率在70%~120%范围的比例分别为甘蓝91.45 %、94.08%,番茄88.20%、88.80%,梨86.84%、92.10%。本方法为常见果蔬中农药残留的准确判断提供了可靠依据,扩大了分析范围。  相似文献

A new procedure based on hydrophilic interaction chromatography coupled to tandem mass spectrometry (ionisation process by pneumatically assisted electrospray in negative ion mode), is developed and validated for the simultaneous determination of underivatised taurine and methionine in beverages rich in carbohydrates such as energy drinks. No initial clean-up procedure and no sample derivatisation are required. Satisfactory analysis was obtained on an Astec apHera NH2 (150 mm x 4.6 mm; 5 microm) column with methanol-water (60/40) as mobile phase. The method was validated in terms of specificity, detection limits, linearity, accuracy, precision and stability, using threonine as internal standard. The potential effects of matrix and endogenous amino acid content were also examined. The limits of detection in the beverage varied from 20 microg L(-1) for taurine to 50 micro L(-1) for methionine.  相似文献

A multiresidue method has been developed and validated for the simultaneous determination of organophosphorus insecticides and their toxic metabolites in sugarcane juice and refined sugar by gas chromatography with flame photometric detection. Limits of quantification of the method varied between 0.007 and 0.01 μg/g. Ethyl acetate based extraction followed by dispersive solid‐phase extraction cleanup with primary secondary amine yielded internationally acceptable recoveries of acephate, chlorpyrifos, dichlorvos, monocrotophos, malathion, malaoxon, phorate, phorate‐sulfoxide, phorate‐oxon, phorate‐sulfone, and quinalphos from selected matrices. The recoveries of target analytes from cane juice were 75.55 ± 0.5–102.57 ± 4.2, 77.45 ± 4.7–103.33 ± 3.3, and 80.55 ± 6.6–105.82 ± 9.8% at 0.01, 0.02, and 0.1 μg/g levels of fortification, respectively. The recoveries from cane sugar were 73.24 ± 3.5–104.47 ± 1.9, 75.23 ± 1.5–116.10 ± 3.7, and 70.75 ± 5.7–110.15 ± 2.7%, respectively at 0.01, 0.02, and 0.1 μg/g levels of fortification. Matrix effect and measurement uncertainty were within the permissible limit (less than 20%) as prescribed for pesticide residue analysis.  相似文献

建立了茶叶中氟虫腈、吡虫啉、啶虫脒、噻嗪酮、三唑酮、三唑醇、丙溴磷、哒螨酮共8种农药残留的液相色谱-串联质谱(HPLC-MS/MS)测定方法。样品用丙酮-二氯甲烷(1:1, v/v)混合溶剂加速溶剂提取,经石墨化炭黑/氨基(Carb/NH2)固相萃取小柱净化,采用Hypersil Gold C18色谱柱(150 mm×2.1 mm, 5 μm)分离,以乙腈-0.1%甲酸水溶液为流动相梯度洗脱,以电喷雾电离(ESI)、多反应监测(MRM)模式检测,采用基质标准曲线同位素内标法(吡虫啉、啶虫脒)或外标法(其余6种农药)定量。氟虫腈在1~100 μg/L、其余7种农药在5~200 μg/L范围内线性关系良好,方法的定量限(信噪比大于10)为氟虫腈2 μg/kg、其余7种农药10 μg/kg。氟虫腈在2、5、50 μg/kg、其余7种农药在10、50、100 μg/kg加标水平下的回收率为75.5%~115.0%,相对标准偏差为2.7%~7.7%。另外,按照JJF 1059-1999《测定不确定度评定与表示》中的有关规定,从标准溶液、样品称量、标准曲线、样品定容、仪器测定重复性、样品前处理等方面对测定结果的不确定度来源进行了评定。评定结果显示,测定结果的不确定度主要源于样品前处理、标准曲线及仪器测定的重复性。该方法的提取效果好、净化较彻底,灵敏度满足国外限量标准的要求,适合出口茶叶中农药残留的测定。  相似文献

HER2 (human epidermal growth factor receptor 2, erbB2, or neu) is overexpressed by a large number of tumor types and has been identified as an important target for cancer therapy. F5 is a single-chain human antibody fragment that recognizes HER2 receptor and is covalently conjugated to PEGylated lipid to form F5 conjugate (F5CG) in the product HER2 targeted STEALTH immunoliposome doxorubicin. Here we described the method development of a competitive enzyme-linked immunosorbent assay (ELISA) for the determination of total concentration of F5 conjugate in plasma samples. The method involved the biotinylation of F5CG, detergent treatment of plasma sample to solubilize F5CG into monomeric form, and competitive ELISA for solubilized F5CG competitively binding to anti-F5CG antibody with biotinylated F5CG for the determination of total F5CG in plasma. The detection range of this method was from 0.2 ng/mL to 125 ng/mL for F5CG in plasma. The lower limit of quantification (LLOQ) was 0.2 ng/mL. This method was established and used for the measurement of F5CG concentration to provide information about F5CG circulation after the administration of immunoliposome in preclinical studies.   相似文献

Hydroxymethylfurfural (HMF) and alpha-ketoglutaric acid (KG) have been recently investigated as potential cancer cell damaging agents. We herein report for the first time a validated quantitative assay for their simultaneous determination in human plasma which is amenable to be applied in the future screening of the target compounds in human probands in order to properly design a targeted chemotherapeutic regimen for certain types of malignant tumors.A simple liquid chromatographic method in conjunction to derivatization after a two-step optimized solid phase clean-up procedure is described. The method is based on the reaction of HMF and KG with 2-nitrophenylhydrazine or 2,4-dinitrophenylhydrazine in an aqueous environment. Reaction conditions were studied with respect to pH, reagent volume, reaction temperature and time. Exact testing of such parameters beside careful selection of the mobile phase composition rendered feasible the quantification of the chemically significantly differing analytes along a single chromatographic run. The formed derivatives could be separated isocratically by reversed-phase LC on a C8-column. Detection in the UV and in the visible range is possible. Results showed good recovery and reproducibility with detection limits (S/N = 3) down to 2 picomoles analyte on column. Resolution of the syn and anti geometric isomers of the HMF and KG derivatives is possible. The isomeric ratio in relation to the reaction pH is discussed.  相似文献
