The aim of this paper is to evaluate the ability of the mesoporous silica SBA-15 to adsorb polyphenols from red wine. The mesoporous molecular sieve silica SBA-15 was hydrothermally synthesized in acidic media and characterized by SAXRD, BET, EDX and SEM. The adsorption behavior of mesoporous silica SBA-15 was investigated at 5 °C for 24 h using an adsorbent dose of 8 g SBA-15 L−1 red wine. The total polyphenols content expressed as mg of gallic acid equivalents (GAE L−1) was estimated from the standard curve of gallic acid (absorbance at 280 nm). HPLC chromatograms of methanolic extract from mesoporous SBA-15 at 256, 280, 324, and 365 nm exhibits the strong retention of quercetin and cis-resveratrol and a reasonable retention of trans-resveratrol, catechin, epicatechin, rutin, and phenolic acids (meta- and para-hydroxybenzoic, vanillic, caffeic, syringic, salicylic and para-coumaric acids).  相似文献

This work presents an advantageous analytical procedure for the accurate determination of free trans-resveratrol in red and white wines. The proposed method involves solid-phase extraction (SPE), acetylation of the analyte in aqueous media and further determination by gas chromatography (GC) with mass spectrometry detection (MS). The use of a mixed-mode SPE sorbent provides an improvement in the selectivity of the extraction step; moreover, the presence of several intense ions in the electron impact mass spectra of its acetyl derivative guarantees the unambiguous identification of trans-resveratrol. Considering a sample intake of 10 mL, the method provides a limit of quantification (LOQ) of 0.8 ng mL−1 and linear responses for concentrations up to 2.5 μg mL−1, referred to wine samples. The average recovery, estimated with samples fortified at different concentrations in the above range, was 99.6% and the inter-day precision stayed below 8%. Trans-resveratrol levels in the analyzed wines varied from 3.4 to 1810 ng mL−1. Cis-resveratrol was also found in all samples. In most cases, equal or higher responses were measured for this latter form than for the trans-isomer. The reduced form of resveratrol, dihydro-resveratrol, was systematically identified in red wines.  相似文献

The present paper describes a direct procedure for the determination of catechin and epicatechin concentrations in red wines employing reverse-phase high performance liquid chromatography (RP HPLC) and detection by fluorescence. The method was performed using a sample volume of 10 µL without dilution. The separation process employed a Chromolith performance RP-18e (100 mm × 4.6 mm) column, and the mobile phase was composed of solvent A: methanol-acetic acid-water (90:8:2) and solvent B: water-acetic acid-methanol (10:2:88) at a flow rate of 1.0 mL min− 1. Linearity was observed in the range of 1 to 30 mg L− 1, with limits of detection and quantification of 0.27 and 0.89 mg L− 1, respectively, for catechin and 0.33 and 1.01 mg L− 1, respectively, for epicatechin. The precisions estimated by the relative standard deviation were 3.34 and 1.09% for catechin concentrations of 0.5 and 20 mg L− 1 respectively and 2.82 and 0.49% for epicatechin concentrations of 0.5 and 20 mg L− 1, respectively. The evaluation of the accuracy was done using an addition/recovery assay. Four wine samples were used, and the recoveries varied from 105 to 108% for catechin and from 97 to 119% for epicatechin. The method was applied to the analysis of red wine samples collected from the São Francisco region, Bahia State, Brazil. Nine samples were analyzed, and the catechin and epicatechin concentrations varied from 7.51 to 73.20 and from 5.08 to 43.32 mg L− 1, respectively. There has been considerable public interest and a growing number of scientific studies linking certain phenolic compounds in grapes and wines, particularly trans-resveratrol (trans-3,5,4′-trihydroxystilbene, TRA), to human health benefits. Typical TRA concentrations in wine are very low. It is a polar compound with very low volatility, which makes it difficult to extract and to separate on a gas chromatography (GC) column without derivatization. In this study, a new method for trace analysis of TRA was developed using solid-phase microextraction (SPME) with on-fiber silylation derivatization. Multidimensional GC equipped with a heartcut valve and cryogenic focusing was coupled with a mass-selective detector and used for improved separations and analysis. The effects of SPME fiber selection, extraction time, temperature, and desorption time were investigated. The derivatization conditions, time/temperature and the volume of derivatization reagent were also optimized. The calibration curve was linear over the concentration range from 10 ng L−1 to 5 mg L−1, with a correlation coefficient of 0.9996. The average recovery of TRA in red wine was 83.6 ± 5.6%. The method detection limit (MDL) for TRA in ethanol:water (12.5:87.5, v/v) solution in this study was 7.08 ng L−1 whereas the MDL for TRA in pure water was 2.85 ng L−1. The new method was used to test the TRA content in six selected Iowa red wine samples. Measured concentrations varied from 12.72 to 851.9 μg L−1.  相似文献

The absorption and fluorescence behaviour of trans-p-coumaric acid (trans-4-hydroxycinnamic acid) is investigated in buffered aqueous solution over a wide range from pH 1 to pH 12, in un-buffered water, and in some organic solvents. Absorption cross-section spectra, fluorescence quantum distributions, fluorescence quantum yields, and degrees of fluorescence polarisation are measured. p-Coumaric acid exists in different ionic forms in aqueous solution depending on the pH. There is an equilibrium between the neutral form (p-CAH2) and the single anionic form (p-CAH) at low pH (pKna ≈ 4.9), and between the single anionic and the double anionic form (p-CA2−) at high pH (pKaa ≈ 9.35). In the organic solvents studied trans-p-coumaric acid is dissolved in its neutral form. The fluorescence quantum yield of trans-p-coumaric acid in aqueous solution is ?F ≈ 1.4 × 10−4 for the neutral and the single anionic form, while it is ?F ≈ 1.3 × 10−3 for the double anionic form. For trans-p-coumaric acid in organic solvents fluorescence quantum yields in the range from 4.8 × 10−5 (acetonitrile) to 1.5 × 10−4 (glycerol) were measured. The fluorescence spectra are 7700–10,000 cm−1 Stokes shifted in aqueous solution, and 5400–8200 cm−1 Stokes shifted in the studied organic solvents. Decay paths responsible for the low fluorescence quantum yields are discussed (photo-isomerisation and internal conversion for p-CA2−, solvent-assisted intra-molecular charge-transfer or ππ to nπ transfer and internal conversion for p-CAH2 and p-CAH). The solvent dependence of the first ππ electronic transition frequency and of the fluorescence Stokes shift of p-CAH2 is discussed in terms of polar solute–solvent interaction effects. Thereby the ground-state and excite-state molecular dipole moments are extracted.  相似文献

A new automated spectrophotometric method for the determination of total sulfite in white and red wines is reported. The assay is based on the reaction of o-phthalaldehyde (OPA) and ammonium chloride with the analyte in basic medium under SI conditions. Upon on-line alkalization with NaOH, a blue product is formed having an absorption maximum at 630 nm. The parameters affecting the reaction - temperature, pH, ionic strength, amount concentration and volume of OPA, amount concentration of ammonium chloride, flow rate and reaction coil length - and the gas-diffusion process - sample and HCl volumes, length of mixing coil, donor flow rate - were studied. The proposed method was validated in terms of linearity (1-40 mg L−1, r = 0.9997), limit of detection (cL = 0.3 mg L−1) and quantitation (cQ = 1.0 mg L−1), precision (sr = 2.2% at 20 mg L−1 sulfite, n = 12) and selectivity. The applicability of the analytical procedure was evaluated by analyzing white and red wine samples, while the accuracy as expressed by recovery experiments ranged between 96% and 106%.  相似文献

The polyphenols, for example stilbenes and flavonoids, are an important family of compounds present in grapes and wines. Several studies have shown that stilbenes are antioxidants and cancer-preventing agents. For the first time, eight natural stilbenes (trans-resveratrol, trans-piceid, cis-piceid, trans-astringin, trans-piceatannol, (+)-trans-ε-viniferin, pallidol, and hopeaphenol), isolated and purified from Vitis vinifera, were simultaneously analysed by ultra-high-pressure liquid chromatography coupled with photodiode-array detection. Separation of the stilbenes by UHPLC was optimized with the assistance of “Quality-by-Design” commercial software. Four different reversed-phase columns packed with 1.5–1.7-μm particles were tested and compared for their retention behaviour and separation efficiency. On the basis of the performance characteristics determined, the VisionHT C18 HL column was selected for the stilbenes studied, because resolution of the critical pair was 1.5 with a peak width of 2–4 s. The optimized method resulted in highly repeatable retention times (RSD 0.03–0.07%), peak areas (RSD 3–6%), and linear ranges were between 0.005 and 50 mg L−1 for most of the compounds. All stilbenes, except trans-astringin, trans-piceatannol, and pallidol were identified and quantified in Burgundy red wines at different concentrations after direct injection of the wines.  相似文献

In this paper, we proposed a procedure for the determination of iron(II) and total iron in wine samples employing molecular absorption spectrophotometry. The ligand used is 2-(5-bromo-2-pyridylazo)-5-(diethylamino)-phenol (Br-PADAP) and the chromogenic reaction in absence or presence of ascorbic acid (reducing agent) allows the determination of iron(II) or total iron, respectively. The optimization step was performed using a multivariate technique (Box Behnken design) involving the factors pH, acid ascorbic concentration and reaction time.The method allows the determination of iron(II) and iron(III) in wine samples, with limits of detection and quantification 0.22 and 0.72 μg L−1, respectively. The precision expressed as relative standard deviation (R.S.D.) was 1.43 and 0.56% (both, n = 11) for content of iron(II) in wine samples of 1.68 and 4.65 mg L−1, and 1.66 and 0.87% (both, n = 11) for content of total iron in wine samples of 1.72 and 5.48 mg L−1.This method was applied for determination of iron(II) and total iron in six different wine samples. In these, the iron(II) content varied from 0.76 to 4.65 mg L−1 and from 1.01 to 5.48 mg L−1 for total iron. The results obtained in the determination of total iron by Br-PADAP method were compared with those that were performed after complete acid digestion in open system and determination of total iron employing FAAS. The method of regression linear was used for comparison of these results and demonstrated that there is no significant difference between the results obtained with these two procedures.  相似文献

A new procedure for determination of biogenic amines (BA): histamine, phenethylamine, tyramine and tryptamine, based on the derivatization reaction with 2-chloro-1,3-dinitro-5-(trifluoromethyl)-benzene (CNBF), is proposed. The amines derivatives with CNBF were isolated and characterized by X-ray crystallography and 1H, 13C, 19F NMR spectroscopy in solution. The novelty of the procedure is based on the pure and well-characterized products of the amines derivatization reaction. The method was applied for the simultaneous analysis of the above mentioned biogenic amines in wine samples by the reversed phase-high performance liquid chromatography. The procedure revealed correlation coefficients (R2) between 0.9997 and 0.9999, and linear range: 0.10–9.00 mg L−1 (histamine); 0.10–9.36 mg L-1 (tyramine); 0.09–8.64 mg L−1 (tryptamine) and 0.10–8.64 mg L−1 (phenethylamine), whereas accuracy was 97%–102% (recovery test). Detection limit of biogenic amines in wine samples was 0.02–0.03 mg L−1, whereas quantification limit ranged 0.05–0.10 mg L−1. The variation coefficients for the analyzed amines ranged between 0.49% and 3.92%. Obtained BA derivatives enhanced separation the analytes on chromatograms due to the inhibition of hydrolysis reaction and the reduction of by-products formation.  相似文献

Biogenic amines in wine usually are analyzed by high-performance liquid chromatography after direct derivatization. A method of isolation based on solid-phase extraction (SPE) with mixed-mode resins (Oasis MCX, reverse-phase and ion exchange) was developed. The different stages of the isolation process (loading, elution and washing) were optimized to obtain a simple procedure that yields a clean chromatogram. The relative standard deviation (%RSD) of the retention times and relative areas was less than 0.3% and 6%, respectively. Limits of quantification were lower than 0.16 mg L−1 for all the amines and the linear range of concentration was 0.16–8 mg L−1 for putrescine, cadaverine and tyramine, and up to 10 mg L−1 for histamine.  相似文献

A comparison of direct immersion solid-phase microextraction (DI-SPME) and stir bar sorptive extraction (SBSE) coupled to liquid chromatography (HPLC) with fluorimetric detection for the rapid analysis of resveratrol isomers is described. For DI-SPME, a polar Carbowax-template resin (CW/TPR) 50 μm fiber was the most efficient and optimum extraction conditions were 40 °C and an extraction time of 30 min, stirring in the presence of 5% (m/v) sodium chloride and 0.07 M acetate/acetic acid buffer (pH 6). Desorption was carried out using the static mode for 10 min. Linearity was obtained in the 5-150 and 2-150 ng mL−1 ranges for trans- and cis-resveratrol, with detection limits of 2 and 0.5 ng mL−1, respectively. When using SBSE, a polydimethylsiloxane (PDMS) twister provided best extraction by means of a derivatization reaction in the presence of acetic anhydride and potassium carbonate. The same time and temperature were used for the extraction step in the presence of 2.5% (m/v) sodium chloride, and liquid desorption was performed with 150 μL of a 50/50 (v/v) acetonitrile/1% (v/v) acetic acid solution in a desorption time of 15 min. Linearity was now between 0.5 and 50 ng mL−1 for trans-resveratrol with a detection limit of 0.1 ng mL−1, while cis-resveratrol could not be extracted. The proposed methods were successfully applied to determining the resveratrol isomer content of wine, must and fruit juices.  相似文献

Nitrate determination in waters is generally carried out with cadmium filings and carcinogenic reagents or by reaction with phenolic compounds in highly concentrated sulfuric acid medium. In this work, it was developed a green analytical procedure for nitrate determination in natural waters based on direct spectrophotometric measurements in ultraviolet, using a flow-injection system with an anion-exchange column for separation of nitrate from interfering species. The proposed method employs only one reagent (HClO4) in a minimum amount (equivalent to 18 μL concentrated acid per determination), and allowed nitrate determination within 0.50-25.0 mg L−1, without interference of up to 200.0 mg L−1 humic acid; 1.0 mg L−1 NO2; 200.0 mg L−1 PO43−; 75.0 mg L−1 Cl; 50.0 mg L−1 SO42− and 15.0 mg L−1 Fe3+. The detection limit (99.7% confidence level) and the coefficient of variation (n = 20) were estimated as 0.1 mg L−1 and 0.7%, respectively. The results obtained for natural water samples were in agreement with those achieved by the reference method based on nitrate reduction with copperized cadmium at the 95% confidence level.  相似文献

In this paper, the conversion of azoxystrobin in a strongly fluorescent degradation product by UV irradiation with quantitative purposes and its fluorimetric determination are reported for the first time. A multicommuted flow injection-solid phase spectroscopy (FI-SPS) system combined with photochemically-induced fluorescence (PIF) is developed for the determination of azoxystrobin in grapes, must and wine. Grape samples were homogenized and extracted with methanol and further cleaned-up by solid-phase extraction on C18 silica gel. Wine samples were solid-phase extracted on C18 sorbent using dichloromethane as eluent. Recoveries of azoxystrobin from spiked grapes (0.5-2.0 mg Kg−1), must (0.5-2.0 μg mL−1) and wine (0.5-2.0 μg mL−1) were 84.0-87.6%, 95.5-105.9% and 88.5-111.2%, respectively. The quantification limit for grapes was 0.021 mg Kg−1, being within European Union regulations, and 18 μg L−1 and 8 μg L−1 for must and wine, respectively.  相似文献

Potassium formate was extracted from airport storm water runoff by headspace solid-phase microextraction (HS-SPME) and analyzed by GC–MS. Formate was transformed to formic acid by adding phosphoric acid. Subsequently, formic acid was derivatized to methyl formate by adding methanol. Using sodium [2H]formate (formate-d) as an internal standard, the relative standard deviation of the peak area ratio of formate (m/z 60) and formate-d (m/z 61) was 0.6% at a concentration of 208.5 mg L−1. Calibration was linear in the range of 0.5–208.5 mg L−1. The detection limit calculated considering the blank value was 0.176 mg L−1. The mean concentration of potassium formate in airport storm water runoff collected after surface de-icing operations was 86.9 mg L−1 (n = 11) with concentrations ranging from 15.1 mg L−1 to 228.6 mg L−1.  相似文献

A sensitive fluorimetric method for determination of phytic acid in human urine samples was described. The method was based on a fluorimetric replacement reaction, in which the added phytic acid replaced the Cu2+ ion from Cu2+-gelatin complex, liberating the fluorescent gelatin molecule. The fluorescence of the solution was accordingly recovered proportionally to the amount of the foreign phytic acid. The excitation wavelength was 273.5 nm and the characteristic emission wavelength was 305.0 nm, respectively. The calibration graph was obtained by plotting the recovered fluorescent intensity at maximum 305.0 nm against the added standard phytic acid, and was divided into two sections. One section was linear over the range of 0.40-2.40 mg L−1 with a linear regression equation of If = −0.895 + 15.146c (R2 > 0.9993), and the other over the range of 2.40-9.20 mg L−1 with a linear regression equation of If = −29.526 + 26.113c (R2 > 0.9996), respectively. The relative standard deviation (R.S.D.) at 95% confidence degree for a 2.0 mg L−1 of standard phytic acid within 1 month was less than 1.26% (n = 5), indicating the procedure is reproducible. The detection and the quantification limits of phytic acid were estimated to be 0.23 and 0.40 mg L−1, respectively. The proposed method was applied to the determination of phytic acid in urine samples and the found concentrations of phytic acid in urine were in the range of 0.49-0.75 mg L−1 with recoveries of 96.2-108.8%. Comparison of the obtained results with the reported HPLC was performed, indicating the proposed method was reliable.  相似文献

In this work several pre-treatment methods were studied for metal (Na, K, Mg, Cu and Ca) determination in Orujo spirit samples using inductively coupled plasma atomic emission spectrometry (ICP-AES). Dilution, digestion, evaporation, and cryogenic desolvatation techniques were comparatively evaluated. Because of their analytical characteristics, digestion and evaporation with nitrogen current were found to be appropriate procedures for the determination of metals in alcoholic spirit samples. Yet, if simplicity and application time are to be considered, the latter—evaporation in a water bath with a nitrogen current—stands out as the optimum procedure for any further determinations in Orujo samples by ICP-AES. Low detection levels and wide linear ranges (sufficient to determine these metals in the samples studied) were achieved for each metal. The recoveries (in the 97.5-100.5% range) and the precision (R.S.D. lower than 5.6%) obtained were also satisfactory. The selected procedure was applied to determine the content of metals in 80 representative Galician Orujo spirit samples with and without a Certified Brand of Origin (CBO) which had been produced using different distillation systems. The metal concentrations ranged between 0.37 and 79.7 mg L−1 for Na, <LOD to 12.4 mg L−1 for K, 0.02-4.83 mg L−1 for Mg content, <LOD to 37.3 mg L−1 for Cu and 0.03-13.10 mg L−1 for Ca.  相似文献

This paper reports the synthesis and testing of a molecularly imprinted polymer membrane for digoxin analysis. Digoxin-specific bulk polymer was obtained by the UV initiated co-polymerisation of methacrylic acid and ethylene glycol dimethacrylate in acetonitrile as porogen. After extracting the template analyte, the ground polymer particles were mixed with plasticizer polyvinyl chloride to form a MIP membrane. A reference polymer membrane was prepared from the same mixture of monomers but with no template. The resultant membrane morphologies were examined by scanning electron microscopy. The imprinted membrane was tested as the recognition element in a digoxin-sensitive fluorescence sensor; sensor response was measured using standard solutions of digoxin at concentrations of up to 4 × 10−3 mg L−1. The detection limit was 3.17 × 10−5 mg L−1. Within- and between-day relative standard deviations RSD (n = 5) were in the range 4.5-5.5% and 5.5-6.5% respectively for 0 and 1 × 10−3 mg L−1 digoxin concentrations. A selectivity study showed that compounds of similar structure to digoxin did not significantly interfere with detection for interferent concentrations at 10, 30 and 100 times higher than the digoxin concentration. This simply manufactured MIP membrane showed good recognition characteristics, a high affinity for digoxin, and provided satisfactory results in analyses of this analyte in human serum.  相似文献

Sensitive, rapid and inexpensive chemiluminescence (CL) imaging has been developed based on molecular imprinted polymer (MIP) sensing elements. Imprinted uniform microspheres were synthesized by precipitation polymerization. Microtiter plates (96 wells) were coated with polymer microspheres imprinted with trans-resveratrol, which were fixed in place using poly(vinyl alcohol) (PVA) as glue. The amount of polymer-bound trans-resveratrol was quantified using imidazole (IMZ)-catalyzed peroxyoxalate chemiluminescence (PO-CL) reaction. The light produced was then measured with a high-resolution CCD camera. Calibration curve corresponding to analyte concentration ranging from 0.3 to 25 μg mL−1 was obtained with a limit of detection 0.1 μg mL−1. These results showed that the MIP-based CL imaging sensor can become a useful analytical tool for quick simultaneous detection of trans-resveratrol in a large number of real samples.  相似文献

The usefulness of the secondary line at 252.744 nm and the approach of side pixel registration were evaluated for the development of a method for sequential multi-element determination of Cu, Fe, Mn and Zn in soil extracts by high-resolution continuum source flame atomic absorption spectrometry (HR-CS FAAS). The influence of side pixel registration on the sensitivity and linearity was investigated by measuring at wings (248.325, 248.323, 248.321, 248.329, and 248.332 nm) of the main line for Fe at 248.327 nm. For the secondary line at 252.744 nm or side pixel registration at 248.325 nm, main lines for Cu (324.754 nm), Mn (279.482 nm) and Zn (213.875 nm), sample flow-rate of 5.0 mL min−1 and calibration by matrix matching, analytical curves in the 0.2-1.0 mg L−1 Cu, 1.0-20.0 mg L−1 Fe, 0.2-2.0 mg L−1 Mn, 0.1-1.0 mg L−1 Zn ranges were obtained with linear correlations better than 0.998. The proposed method was applied to seven soil samples and two soil reference materials (IAC 277; IAC 280). Results were in agreement at a 95% confidence level (paired t-test) with reference values. Recoveries of analytes added to soil extracts containing 0.15 and 0.30 mg L−1 Cu, 7.0 and 14 mg L−1 Fe, 0.60 and 1.20 mg L−1 Mn, 0.07 and 0.15 mg L−1 Zn, varied within the 94-99, 92-98, 93-101, and 93-103% intervals, respectively. The relative standard deviations (n = 12) were 2.7% (Cu), 1.4% (Fe - 252.744 nm), 5.7% (Fe - 248.325 nm), 3.2% (Mn) and 2.8% (Zn) for an extract containing 0.35 mg L−1 Cu, 14 mg L−1 Fe, 1.1 mg L−1 Mn and 0.12 mg L−1 Zn. Detection limits were 5.4 μg L−1 Cu, 55 μg L−1 Fe (252.744 nm), 147 μg L−1 Fe (248.325 nm), 3.0 μg L−1 Mn and 4.2 μg L−1 Zn.  相似文献
