The aim of this work was to develop a fast method for extraction and analysis of anthocyanins in red cabbage. Pressurized hot water containing 5% of ethanol was used as an extremely efficient extraction solvent. HPLC/DAD with a monolithic column was used to accomplish a fast analysis—24 anthocyanin peaks within 18 min. Statistical design was used to optimize the studied extraction parameters: temperature (80–120 °C); sample amount (1–3 g); extraction time (6–11 min); concentration of formic acid in the extraction solvent (0–5 vol.%). The best extraction conditions for a majority of the anthocyanin peaks were 2.5 g of sample, 99 °C (at 50 bar), 7 min of extraction and a solvent composition of water/ethanol/formic acid (94/5/1, v/v/v).  相似文献

A rapid and environmentally friendly methodology was developed for the extraction of pyrethroid insecticides from semi permeable membrane devices (SPMDs), in which they were preconcentrated in gas phase. The method was based on gas chromatography mass–mass spectrometry determination after a microwave-assisted extraction, in front of the widely employed dialysis method. SPMDs were extracted twice with 30 mL hexane:acetone, irradiated with 250 W power output, until 90 °C in 10 min, this temperature being held for another 10 min. Clean-up of the extracts was performed by acetonitrile–hexane partitioning and solid-phase extraction (SPE) with a combined cartridge of 2 g basic-alumina, deactivated with 5% water, and 500 mg C18.

Pyrethroids investigated were Allethrin, Prallethrin, Tetramethrin, Bifenthrin, Phenothrin, λ-Cyhalothrin, Permethrin, Cyfluthrin, Cypermethrin, Flucythrinate, Esfenvalerate, Fluvalinate and Deltamethrin. The main pyrethroid synergist compound, Pyperonyl Butoxide, was also studied. Limit of detection values ranging from 0.3 to 0.9 ng/SPMD and repeatability data, as relative standard deviation, from 2.9 to 9.4%, were achieved. Pyrethroid recoveries, for spiked SPMDs, with 100 ng of each one of the pyrethroids evaluated, were from 61 ± 8 to 103 ± 7% for microwave-assisted extraction, versus 54 ± 4 to 104 ± 3% for dialysis reference method. Substantial reduction of solvent consumed (from 400 to 60 mL) and analysis time (from 48 to 1 h) was achieved by using the developed procedure.

High concentration levels of pyrethroid compounds, from 0.14 to 7.3 μg/SPMD, were found in indoor air after 2 h of a standard application.  相似文献

The aroma profile of cocoa products was investigated by headspace solid-phase micro-extraction (HS-SPME) combined with gas chromatography–mass spectrometry (GC–MS). SPME fibers coated with 100 μm polydimethylsiloxane coating (PDMS), 65 μm polydimethylsiloxane/divinylbenzene coating (PDMS-DVB), 75 μm carboxen/polydimethylsiloxane coating (CAR-PDMS) and 50/30 μm divinylbenzene/carboxen on polydimethylsiloxane on a StableFlex fiber (DVB/CAR-PDMS) were evaluated. Several extraction times and temperature conditions were also tested to achieve optimum recovery. Suspensions of the samples in distilled water or in brine (25% NaCl in distilled water) were investigated to examine their effect on the composition of the headspace. The SPME fiber coated with 50/30 μm DVB/CAR-PDMS afforded the highest extraction efficiency, particularly when the samples were extracted at 60 °C for 15 min under dry conditions with toluene as an internal standard. Forty-five compounds were extracted and tentatively identified, most of which have previously been reported as odor-active compounds. The method developed allows sensitive and representative analysis of cocoa products with high reproducibility. Further research is ongoing to study chocolate making processes using this method for the quantitative analysis of volatile compounds contributing to the flavor/odor profile.  相似文献

An analytical methodology including pressurized liquid extraction (PLE) as sample treatment to isolate retinyl acetate and tocopherols from infant formulas has been developed. The milk extracts were kept at −18 °C for 30 min and after filtration could be injected directly into the chromatographic system. Thus, a rapid and simple routine control method of these products is possible.

The parameters affecting both the extraction process and the liquid chromatography (LC) system were optimized. PLE was performed using one cycle of extraction during a static time of 5 min. Methanol was chosen as the extraction solvent for a temperature of 50 °C. Chromatographic separation was accomplished using a RP-18 column; the mobile phase used was methanol–water (94:6, v/v) containing 2.5 mM acetic acid/sodium acetate buffer. Electrochemical detection in amperometric mode with a glassy carbon electrode at +1100 mV was applied. The proposed methodology was successfully used for the determination of retinyl acetate, δ-tocopherol, (β + γ)-tocopherol and -tocopherol in different infant formulas. The analytes were evaluated in the same chemical form present in the samples. Recoveries were between 92 and 106%. A certified reference material of milk powder was also analyzed.  相似文献

Pressurized liquid extraction (PLE) is a relatively new technique applicable for the extraction of persistent organic pollutants from various matrices. The main advantages of this method are short time and low consumption of extraction solvent. The effects of various operational parameters (i.e. temperature of extraction, number of static cycles and extraction solvent mixtures) on the PLE efficiency were investigated in this study. Fish muscle tissue containing 3.2% (w/w) lipids and native polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs) and other related compounds was used for testing. Purification of crude extracts was carried out by gel permeation chromatography employing Bio-Beads S-X3. Identification and quantitation of target indicator PCBs and OCPs was performed by high-resolution gas chromatography (HRGC) with two parallel electron-capture detectors (ECDs). Results obtained by the optimized PLE procedure were compared with conventional Soxhlet extraction (the same extraction solvent mixtures hexane–dichloromethane (1:1 v/v) and hexane–acetone (4:1 v/v) were used). The recoveries obtained by PLE operated at 90–120 °C were either comparable to “classic” Soxhlet extraction (for higher-chlorinated PCB congeners and DDT group) or even better (for lower chlorinated analytes). The highest recoveries were obtained for three static 5 min extraction cycles.  相似文献

Dispersive liquid–liquid microextraction (DLLME) coupled with gas chromatography–flame ionization detection (GC–FID) was applied for the determination of two tricyclic antidepressant drugs (TCAs), amitriptyline and nortriptyline, from water samples. This method is a very simple and rapid method for the extraction and preconcentration of these drugs from environmental sample solutions. In this method, the appropriate mixture of extraction solvent (18 μL Carbon tetrachloride) and disperser solvent (1 mL methanol) are injected rapidly into the aqueous sample (5.0 mL) by syringe. Therefore, cloudy solution is formed. In fact, it is consisted of fine particles of extraction solvent which is dispersed entirely into aqueous phase. The mixture was centrifuged and the extraction solvent is sedimented on the bottom of the conical test tube. 2.0 μL of the sedimented phase is injected into the GC for separation and determination of TCAs. Some important parameters, such as kind of extraction and disperser solvent and volume of them, extraction time, pH and ionic strength of the aqueous feed solution were optimized. Under the optimal conditions, the enrichment factors and extraction recoveries were between 740.04–1000.25 and 54.76–74.02%, respectively. The linear range was (0.005–16 μg mL−1) and limits of detection were between 0.005 and 0.01 μg mL−1 for each of the analytes. The relative standard deviations (R.S.D.) for 4 μg mL−1 of TCAs in water were in the range of 5.6–6.4 (n = 6). The performance of the proposed technique was evaluated for determination of TCAs in blood plasma.  相似文献

A rapid method has been developed to extract and quantitatively measure the total oil content in poultry feeds using a domestic microwave oven. The optimized extraction procedure involves the replicate (6×) extraction of 5 g of ground feed with 12 ml of hexane for 20 s in a 900 W oven. Each replicate involves the collection of the resulting miscella and its replacement with fresh solvent for re-extraction. The collected extracts were centrifuged and transferred to a vial. The solvent was evaporated to a constant weight and the residual lipid weighed. In comparison to conventional Soxhlet extraction method, lipid contents obtained using the optimized microwave procedure was not significantly different. However, FTIR analysis indicated that the microwave procedure was superior in minimizing the formation of free fatty acids (FFA) relative to the Soxhlet procedure if the temperature of the sample was kept within the range of 45–50 °C. This simple, sequential extraction procedure is rapid, highly efficient and provides a simple mean of quantitating the lipid content of poultry feed in less than 40 min without the need for specialized microwave oven.  相似文献

In the present work, a rapid method for the extraction and determination of chlorobenzenes (CBs) such as monochlorobenzene, 1,2-dichlorobenzene, 1,3-dichlorobenzene, 1,4-dichlorobenzene, 1,2,3-trichlorobenzene and 1,2,4-trichlorobenzene in water samples using the headspace solvent microextraction (HSME) and gas chromatography/electron capture detector (ECD) has been described. A microdrop of the dodecane containing monobromobenzene (internal standard) was used as extracting solvent in this investigation. The analytes were extracted by suspending a 2.5 μl extraction drop directly from the tip of a microsyringe fixed above an extraction vial with a septum in a way that the needle passed through the septum and the needle tip appeared above the surface of the solution. After the extraction was finished, the drop was retracted back into the needle and injected directly into a GC column. Optimization of experimental conditions such as nature of the extracting solvent, microdrop and sample temperatures, stirring rate, microdrop and sample volumes, the ionic strength and extraction time were investigated. The optimized conditions were as follows: dodecane as the extracting solvent, the extraction temperature, 45 °C; the sodium chloride concentration, 2 M; the extraction time, 5.0 min; the stirring rate, 500 rpm; the drop volume, 2.5 μl; the sample volume, 7 ml; the microsyringe needle temperature, 0.0 °C. The limit of detection (LOD) ranged from 0.1 μg/l (for 1,3-dichlorobenzene) to 3.0 μg/l (for 1,4-dichlorobenzene) and linear range of 0.5–3.0 μg/l for 1,2-dichlorobenzene, 1,3-dichlorobenzene and from 5.0 to 20.0 μg/l for monochlorobenzene and from 5.0 to 30 μg/l for 1,4-dichlorobenzene. The relative standard deviations (R.S.D.) for most of CBs at the 5 μg/l level were below 10%. The optimized procedure was successfully applied to the extraction and determination of CBs in different water samples.  相似文献

A procedure for arsenic species fractionation in alga samples (Sargassum fulvellum, Chlorella vulgaris, Hizikia fusiformis and Laminaria digitata) by extraction is described. Several parameters were tested in order to evaluate the extraction efficiency of the process: extraction medium, nature and concentration (tris(hydroxymethyl)aminomethane, phosphoric acid, deionised water and water/methanol mixtures), extraction time and physical treatment (magnetic stirring, ultrasonic bath and ultrasonic focussed probe). The extraction yield of arsenic under the different conditions was evaluated by determining the total arsenic content in the extracts by ICP-AES. Arsenic compounds were extracted in 5 mL of water by focussed sonication for 30 s and subsequent centrifugation at 14,000 × g for 10 min. The process was repeated three times. Extraction studies show that soluble arsenic compounds account for about 65% of total arsenic.

An ultrafiltration process was used as a clean-up method for chromatographic analysis, and also allowed us to determine the extracted arsenic fraction with a molecular weight lower than 10 kDa, which accounts for about 100% for all samples analysed.

Speciation studies were carried out by HPLC–ICP-AES. Arsenic species were separated on a Hamilton PRP-X100 column with 17 mM phosphate buffer at pH 5.5 and 1.0 mL min−1 flow rate. The chromatographic method allowed us to separate the species As(III), As(V), MMA and DMA in less than 13 min, with detection limits of about 20 ng of arsenic per species, for a sample injection volume of 100 μL. The chromatographic analysis allowed us to identify As(V) in Hizikia (46 ± 2 μg g−1), Sargassum (38 ± 2 μg g−1) and Chlorella (9 ± 1 μg g−1) samples. The species DMA was also found in Chlorella alga (13 ± 1 μg g−1). However, in Laminaria alga only an unknown arsenic species was detected, which eluted in the dead volume.  相似文献

A liquid–liquid extraction method using diethyl ether as organic solvent was optimized simultaneously for five 1,4-dihydropyridines (amlodipine, nitrendipine, felodipine, lacidipine and lercanidipine) belonging to the group of calcium channel blockers. Some experimental tools such as a full factorial design, a central composite design and the Multisimplex program were used to optimise the concentration of NaOH, volume of organic solvent and shaking time as main factors that influence the liquid–liquid extraction procedure. Following the extraction, the quantitation of the 1,4-dihydropyridines concentrations were performed by high-performance liquid chromatography with diode-array detector. Therefore, the studied compounds were separated quantitatively on a Supelcosil ABZ+Plus, 25 cm × 4.6 mm i.d., 5 μm column which was set at 30 °C, using as mobile phase, a mixture of acetonitrile–water (70:30, v/v) containing 10 mM acetate buffer (pH 5) and setting the detector at a wavelength value of 360 nm. It was concluded that the main factors that influence in the extraction process were the volume of organic solvent and the shaking time. The Multisimplex program suggested as optimal conditions an average of 6 ml of organic solvent and 23 min of shaking time. For these values, the optimised liquid–liquid extraction method showed good values of recoveries (80% for amlodipine and higher than 90% for the rest of the compounds) and low values of R.S.D. (<10%) in the reproducibility of the extraction what makes it reliable for the quantification of all the studied compounds in human plasma.  相似文献

Comparison of extraction techniques of robenidine from poultry feed samples   总被引:1,自引:0,他引:1  
In this paper, effectiveness of six different commonly applied extraction techniques for the determination of robenidine in poultry feed has been compared. The sample preparation techniques included shaking, Soxhlet, Soxtec, ultrasonically assisted extraction, microwave – assisted extraction and accelerated solvent extraction. Comparison of these techniques was done with respect to the recovery extraction, temperature and time, reproducibility and solvent consumption. Every single extract was subjected to clean – up using aluminium oxide column (Pasteur pipette filled with 1 g of aluminium oxide), from which robenidine was eluted with 10 ml of methanol. The eluate from the clean-up column was collected in a volumetric flask, and finally it was analysed by HPLC–DAD–MS. In general, all extraction techniques were capable of isolating of robenidine from poultry feed, but the recovery obtained using modern extraction techniques was higher than that obtained using conventional techniques.

In particular, accelerated solvent extraction was more superior to other techniques, which highlights the advantages of this sample preparation technique. However, in routine analysis, shaking and ultrasonically assisted extraction is still the preferred method for the solution of robenidine and other coccidiostatics.  相似文献

Dispersive liquid-liquid microextraction (DLLME) was combined with high-performance liquid chromatography-diode-array detector (HPLC-DAD) for the extraction and quantitation of three major capsaicinoids (i.e. capsaicin, dihydrocapsaicin and nordihydrocapsaicin) from pepper (Capsicum annuum L.). Chloroform (extraction solvent, 100 μL), acetonitrile (disperser solvent, 1250 μL) and 30 s extraction time were found optimum. The analytes were back-extracted into 300 μL of 50 mM sodium hydroxide/ methanol, 45/55% (v/v), within 15 s before being injected into the instrument. Enrichment factors ranged from 3.3 to 14.7 and limits of detection from 5.0 to 15.0 µg g-1. Coefficients of determination (R2) and %RSD were higher than 0.9962 and lower than 7.5%, respectively. The proposed method was efficiently applied for the extraction and quantitation of the three capsaicinoids in six cultivars of Capsicum annuum L. with percentage relative recoveries in the range of 92.0%–108.0%. DLLME was also scaled up for the isolation of the three major capsaicinoids providing purity greater than 98.0% as confirmed by liquid chromatography-mass spectrometry (LC-MS) and nuclear magnetic resonance (NMR) analysis, which significantly reduced the extraction time and organic solvent consumption.  相似文献

In the present study, the permeation behavior of methanol and methyl acetate in the pervaporation (PV) experiments are compared with those in vapor permeation (VP) experiments using a PVA-based composite membrane. Experiments have been carried out to study the selectivity and mass transport flux of the systems under varying operations conditions of feed temperature (40–60 °C) and feed methanol concentrations (2–34 wt%). The selected membrane was found to be methanol selective. Results show higher permeation flux but a similar separation factor for methanol in PV than in VP. For PV operation, the resulting separation factor at 60 °C shows a monotonous decrease (6.4–4.1) as the alcohol concentration in the feed mixture increases (2.3–34 wt%), whereas the total flux increases from 0.97 to 7.9 kg m−2 h−1. Based on the solution-diffusion theory, a mathematical model that describes satisfactorily the permeation fluxes of methanol and methyl acetate in both the PV and VP processes has been applied. The fluxes of both permeants can be explained by the solution-diffusion model with variable diffusion coefficients dependent on MeOH concentration in the membrane. Both PV and VP processes can be described with the same model but using different fitting parameters.  相似文献

In the present work, a rapid and sensitive ultra performance liquid chromatography‐mass spectrometry method has been proposed for the analysis of capsaicinoids (nordihydrocapsaicin, capsaicin, dihydrocapsaicin, homocapsaicin, and homodihydrocapsaicin) present in different Capsicum samples. Extraction of capsaicinoids was carried out by liquid–liquid extraction using ethanol as an extracting solvent, while the chromatographic separation was achieved by reversed phase C18 column with gradient mobile phase (solvent A: acetonitrile and solvent B: water with 0.1% formic acid). Under the optimum experimental conditions, the linear ranges were 0.5–50 μg/g with correlation coefficient (r2) >0.999 for each capsaicinoids and detection limits were 0.15, 0.05, 0.06, 0.2, and 0.1 μg/g for nordihydrocapsaicin, capsaicin, dihydrocapsaicin, homocapsaicin, and homodihydrocapsaicin, respectively. Run‐to‐run and day‐to‐day precisions of the method with relative standard deviations <1.5% were achieved for all analyzed capsaicinoids. The robustness of the method was determined by utilizing different injection volumes of the extracts. Furthermore, to validate the system robustness, a run of high number of capsaicinoids present in different varieties of Capsicum samples was performed in this study. All the capsaicinoids were separated in a time of less than 9 min by employing the proposed method.  相似文献

A HPLC method with automated column switching and UV-diode array detection is described for the simultaneous determination of Vitamin D3 and 25-hydroxyvitamin D3 (25-OH-D3) in a sample of human plasma. The system uses a BioTrap precolumn for the on-line sample cleanup. A sample of 1 ml of human plasma was treated with 2 ml of a mixture of ethanol–acetonitrile (2:1 (v/v)). Following centrifugation, the supernatant was evaporated to dryness under a stream of dry and pure nitrogen. The residue was reconstituted in 250 μL of a solution of methanol 5 mmol l−1 phosphate buffer, pH 6.5 (4:1 (v/v)), and a 200 μl aliquot of this solution was injected onto the BioTrap precolumn. After washing during 5 min with a mobile phase constituted by a solution of 6% acetonitrile in 5 mmol l−1 phosphate buffer, pH 6.5 (extraction mobile phase), the retained analytes were then transferred to the analytical column in the backflush mode. The analytical separation was then performed by reverse-phase chromatography in the gradient elution mode with the solvents A and B (Solvent A: acetonitrile–phosphate buffer 5 mmol l−1, pH 6.5; 20:80 (v/v); solvent B: methanol–acetonitrile–tetrahydrofuran, 65:20:15 (v/v)). The compounds of interest were detected at 265 nm. The method was linear in the range 3.0–32.0 ng ml−1 with a limit of quantification of 3.0 ng ml−1. Quantitative recoveries from spiked plasma samples were between 91.0 and 98.0%. In all cases, the coefficient of variation (CV) of the intra-day and inter-day-assay precision was ≤2.80%. The proposed method permitted the simultaneous determination of Vitamin D3 and 25-OH-D3 in 16 min, with an adequate precision and sensitivity. However, the overlap of the sample cleanup step with the analysis increases the sampling frequency to five samples h−1. The method was successfully applied for the determination of Vitamin D3 and 25-OH-D3 in plasma from 46 female volunteers, ranging from 50 to 94 years old. Vitamin D3 and 25-OH-D3 concentrations in plasma were found from 4.30–40.70 ng ml−1 (19.74 ± 9.48 ng ml−1) and 3.1–36.52 ng ml−1 (7.13 ± 7.80 ng ml−1), respectively. These results were in good agreement with data published by other authors.  相似文献

A simple method for the extraction of two major capsaicinoids from habanero peppers, using near-infrared irradiation, microwave irradiation or ultrasound as the energy source and ethanol as the solvent, was compared with Soxhlet extraction. The extraction processes were monitored by gas chromatography/mass spectrometry. The new processes offer better overall yields and a higher ratio of capsaicin to dihydrocapsaicin than Soxhlet extraction. The physical effect of the different energy sources on the peppers was determined using scanning electron microscopy. Extraction of capsaicin and dihydrocapsaicin using near-infrared irradiation, which has not been previously reported, was shown to be a simple and efficient alternative extraction procedure.  相似文献

Aluminum, iron and zinc containing MCM-41 molecular sieves were prepared by the hydrothermal method. The catalyst was characterized by the XRD, BET (surface area), FT–IR and 29Si, 27Al MAS–NMR techniques. The catalytic activity of these molecular sieves was tested with esterification reaction used with phthalic anhydride (PAH) and methanol (MeOH) in the autoclave at 135 °C, 150 °C and 175 °C. Conversion increases with an increase in temperature and mole ratio. The activity of these catalysts followed the order: Al-MCM-41 (112) > Fe-MCM-41 (115) > Al-MCM-41 (70) > Al-MCM-41 (52) > Fe-MCM-41 (61) > Al, Zn-MCM-41 (104) > Al-MCM-41 (30). The reaction yielded both monomethyl phthalate (MMP) and dimethyl phthalate (DMP). The nature of the catalyst sites has been proposed using with water as an impurity. The selectivity of the dimethyl phthalate increases with increase in temperature and mole ratio. The weight of the catalyst was optimized at 0.07 g. The hydrophilic and hydrophobic nature of the catalyst has been explained by the influence of water and the external surface acidity also facilitates the reaction and this has been confirmed by the supporting reaction.  相似文献

Cloud point extraction (CPE) has been used for the pre-concentration of lead, after the formation of a complex with 2-(5-bromo-2-pyridylazo)-5-(diethylamino)-phenol (5-Br-PADAP), and later analysis by graphite furnace atomic absorption spectrometry (GFAAS) using octylphenoxypolyethoxyethanol (TritonX-114) as surfactant. The chemical variables affecting the separation phase were optimized. Separation of the two phases was accomplished by centrifugation for 15 min at 4000 rpm. Under the optimum conditions i.e., pH 8.0, cloud point temperature 40 °C, [5-Br-PADAP] = 2.5 × 10−5 mol l−1, [Triton X-114] = 0.05%, added methanol volume = 0.15 ml, pre-concentration of only 10 ml sample permitted an enhancement factor of 50-fold. The lower limit of detection (LOD) obtained under the optimal conditions was 0.08 μg l−1. The precision for 10 replicate determinations at 5 μg l−1 Pb was 2.8% relative standard deviation (R.S.D.). The calibration graph using the pre-concentration system for lead was linear with a correlation coefficient of 0.9984 at levels near the detection limits up to at least 30 μg l−1. The method was successfully applied to the determination of lead in water samples.  相似文献

A simple, selective and sensitive luminescence method for the assay of piroxicam (PX) in pharmaceutical formulation is developed. The method is based on the luminescence sensitization of europium (Eu3+) by complexation with PX. The signal for PX–EU is monitored at λex=358 nm and λem=615 nm. Optimum conditions for the formation of the complex in methanol were 0.01 M TRIS buffer and 0.2 mM of Eu3+ which allows the determination of 100–2000 ppb of pX in batch method and 100–1000 ppb with limit of detection (LOD) = 23.0 ppb using sequential injection analysis (SIA). The relative standard deviations of the method range between 2 and 3% indicating excellent reproducibility of the method. The proposed method was successfully applied for he assay of PX in pharmaceutical formulations (Feldene capsules and tablets). Average recoveries of 101.0±0.3 and 98.8±2.7% were obtained for capsules in methanol using batch and sequential injection (SI) methods, respectively.  相似文献

The reaction of ethylene sulfide with 3-aminopropyltrimethoxysilane gave a new silylating agent, which was anchored onto a silica surface via the sol–gel procedure. This surface displayed a chelating moiety containing nitrogen and two sulfur basic centers potentially capable of extracting cations from aqueous solutions. The process of metal extraction was followed by a batch method, and fitted to a modified Langmuir equation. The maximum adsorption capacities found were: 2.06 ± 0.01, 3.72 ± 0.02, and 5.14 ± 0.02 mmol g−1 for Pb(II), Cd(II), and Hg(II), respectively. The enthalpies of bending are: −1.16 ± 0.04, −3.60 ± 0.10, and −8.94 ± 0.03 kJ mol−1 for Cd(II), Pb(II), and Hg(II), respectively. The Gibbs free energies of binding agree with the spontaneity of the proposed reactions between cations and basic centers.  相似文献
