Polyacrylonitrile (PAN) was blended with polyvinylidine fluoride (PVDF) at various ratios and made into membranes. The hemocompatibility of the resulting membranes was evaluated based on human plasma proteins adsorption, platelet adhesion, thrombus formation, and blood coagulation time. The PAN/PVDF blends exhibited partial miscibility according to the inward shifting of their two glass transition temperatures. The microstructures of blend membranes examined using atomic force microscopy (AFM) indicated that the roughness increased with the PVDF content, and the phase separation was too severe to form a membrane when the PVDF content was more than 30%. The water contact angle of PAN/PVDF blend membranes increased with the PVDF content. By blending with 20 wt% apolar PVDF the adsorption of blood proteins could be reduced, and hence the platelet adhesion and thrombus formation was also reduced. However, when the PVDF content was 30 wt%, severe thrombogenicity was observed due probably to the more porous structure of blend membrane. These results demonstrated that the hemocompatibility would be improved for PAN/PVDF blend membranes with appropriate hydrophilicity and roughness. Copyright © 2005 John Wiley & Sons, Ltd.  相似文献

In this work, bioadhesive behavior of plasma proteins and blood cells from umbilical cord blood (UCB) onto zwitterionic poly(sulfobetaine methacrylate) (polySBMA) polymer brushes was studied. The surface coverage of polySBMA brushes on a hydrophobic polystyrene (PS) well plate with surface grafting weights ranging from 0.02 mg/cm(2) to 0.69 mg/cm(2) can be effectively controlled using the ozone pretreatment and thermal-induced radical graft-polymerization. The chemical composition, grafting structure, surface hydrophilicity, and hydration capability of prepared polySBMA brushes were determined to illustrate the correlations between grafting properties and blood compatibility of zwitterionic-grafted surfaces in contact with human UCB. The protein adsorption of fibrinogen in single-protein solutions and at complex medium of 100% UCB plasma onto different polySBMA brushes with different grafting coverage was measured by enzyme-linked immunosorbent assay (ELISA) with monoclonal antibodies. The grafting density of the zwitterionic brushes greatly affects the PS surface, thus controlling the adsorption of fibrinogen, the adhesion of platelets, and the preservation of hematopoietic stem and progenitor cells (HSPCs) in UCB. The results showed that PS surfaces grafted with polySBMA brushes possess controllable hydration properties through the binding of water molecules, regulating the bioadhesive and bioinert characteristics of plasma proteins and blood platelets in UCB. Interestingly, it was found that the polySBMA brushes with an optimized grafting weight of approximately 0.1 mg/cm(2) at physiologic temperatures show significant hydrated chain flexibility and balanced hydrophilicity to provide the best preservation capacity for HSPCs stored in 100% UCB solution for 2 weeks. This work suggests that, through controlling grafting structures, the hemocompatible nature of grafted zwitterionic polymer brushes makes them well suited to the molecular design of regulated bioadhesive interfaces for use in the preservation of HSPCs from human UCB.  相似文献

In general, it is a challenge to control the highly polar material grafting from the chemically inert Teflon-based membrane surface. This work describes the surface modification and characterization of expanded poly(tetrafluoroethylene) (ePTFE) membranes grafted with poly(ethylene glycol) methacrylate (PEGMA) macromonomer via surface-activated plasma treatment and thermally induced graft copolymerization. The chemical composition and microstructure of the surface-modified ePTFE membranes were characterized by Fourier transform infrared spectroscopy (FT-IR), contact angle, and bio-atomic force microscopy (bio-AFM) measurements. Biofouling property of the modified membranes was evaluated by the measurements of the plasma protein (γ-globulin, fibrinogen, or albumin) adsorption determined using an enzyme-linked immunosorbent assay (ELISA). In general, the hydrophilicity of the surface of ePTFE membranes increases with increasing the grafting degree of the copolymerized PEGMA. The highly hydrated PEGMA chain on the resulting ePTFE membranes was found to form a surface hydrogel-like layer with regulated coverage in aqueous state, which can be controlled by the content of PEGMA macromonomer in the reaction solution. The relative protein adsorption was effectively reduced with increasing capacity of the hydration for the PEGMA chain grafted on the ePTFE membrane surface. From both results of protein adsorption and platelet adhesion test in vitro, it is concluded that the PEGMA-grafted hydrophilic ePTFE membranes could provide good biofouling resistance to substantially reduce plasma protein and blood platelet fouling on the membrane surface in human body temperature.  相似文献

Arterial prostheses made of microporous Teflon (ePTFE) are currently used in vascular surgery as bypasses for small and medium vessels. However, several clinical complications, such as thrombosis, frequently occur in these prostheses when implanted in humans. In this work, an original strategy was developed to improve the hemocompatibility of ePTFE prostheses, based on glow-discharge surface modification followed by chemical grafting of phosphorylcholine, known for its hemocompatible properties. This procedure leads to a covalent attachment of the molecules, therefore preventing their removal by shear stress induced by blood flow at the implant wall. The improvement of the blood compatibility properties of the modified ePTFE arterial prostheses have been investigated by in vitro tests such as thromboelastography, neutrophil adsorption, platelet aggregation, and cell cultures. These in vitro tests put in evidence that thrombogenicity index, platelet aggregation, and neutrophil adhesion were decreased by the molecule grafted on the prostheses. Moreover, the cell growth on the surface of the PRC-grafted prostheses was greatly enhanced in comparison to the virgin prosthesis. Based on these results, it could be concluded that PRC grafting on ePTFE prostheses permit to improve in vitro hemocompatibility and biocompatibility in comparison with their virgin counterpart.  相似文献

In this paper, the mechanism of thrombus formation on the surface of polymeric materials and the various approaches of modifying biomaterial surfaces to improve their hemocompatibility are reviewed. Moreover, the blood compatibility of the cellulose membrane grafted with O-butyrylchitosan (OBCS) by using a radiation grafting technique was studied. Surface analysis of grafted cellulose membrane was verified by attenuated total reflection Fourier transform infrared spectroscopy (ATR-FTIR) and electron spectroscopy for chemical analysis (ESCA), which confirmed that OBCS was successfully grafted onto the cellulose membrane surfaces. Blood compatibility of the grafted cellulose membranes was evaluated by platelet rich plasma (PRP) contacting experiments and protein adsorption experiments using blank cellulose membranes as the control. The blood compatibility of OBCS grafted cellulose membranes is better than that of blank cellulose membranes. These results suggest that the photocrosslinkable chitosan developed here has the potential of serving in blood-contacting applications in medical use.  相似文献

Nonthrombogenic modifications of membranes for extracorporeal membrane oxygenators (ECMOs) are of key interest. The absence of hemocompatibility of these membranes and the need of anticoagulation of patients result in severe and potentially life‐threatening complications during ECMO treatment. To address the lack of hemocompatibility of the membrane, surface modifications are developed, which act as barriers to protein adsorption on the membrane and, in this way, prevent activation of the coagulation cascade. The modifications are based on nonionic and zwitterionic polymer brushes grafted directly from poly(4‐methyl‐1‐pentene) (TPX) membranes via single electron transfer‐living radical polymerization. Notably, this work introduces the first example of well‐controlled surface‐initiated radical polymerization of zwitterionic brushes. The antifouling layers markedly increase the recalcification time (a proxy of initiation of coagulation) compared to bare TPX membranes. Furthermore, platelet and leukocyte adhesion is drastically decreased, rendering the ECMO membranes hemocompatible.  相似文献

The surface of a thermoplastic polyurethane (TPU) membrane was treated with low temperature plasma (LTP) and was then grafted with poly(acrylic acid) (PAA), followed by the grafting of water-soluble chitosan (WSC) and heparin (HEP). The surface was characterized with static contact-angle and X-ray photoelectron spectroscopy (XPS). The results showed that the surface densities of peroxides and PAA reached a maximum when treated with LTP for 90 s. A higher pH of the reacting solution led to higher graft densities of WSC and HEP. After WSC and HEP grafting, the hydrophilicity of the TPU membrane was increased. The adsorption of proteins on HEP-grafted TPU membranes was effectively curtailed. In addition, HEP grafting also reduced platelet adhesion, elevated thrombin inactivation, and prolonged the blood coagulation time. According to the L929 fibroblast cell growth inhibition index, the HEP-grafted TPU membranes exhibited non-cytotoxicity. Overall results demonstrated that the HEP immobilization could not only improve the hydrophilicity but also the hemocompatibility of the TPU membrane, while maintaining the ascendant biocompatibility.  相似文献

Phospholipid polymer, poly[2-methacryloyloxyethyl phosphorylcholine (MPC)], was grafted with polyethylene (PE) membrane using photoinduced polymerization technique to make the membrane resistant to cell adhesion. The water contact angle on the PE membrane grafted with poly(MPC) decreased with an increase in the photopolymerization time. This decrease corresponded to the increase in the amount of poly(MPC) grafted on the PE surface. The same graft polymerization procedure was applied using other hydrophilic monomers, such as acrylamide (AAm), N-vinylpyrrolidone (VPy) and methacryloyl poly(ethylene glycol) (MPEG). These monomers were also polymerized to form grafted chains on the PE membrane, and the grafting was confirmed with X-ray photoelectron spectroscopy. Analysis of amount and distribution of plasma proteins at the plasma-contacting surface of the original and the modified PE membranes were analyzed using immunogold assay. The grafting of poly(MPC) and poly(VPy) on PE membrane reduced the plasma protein adsorption significantly compared with that on the original PE membrane. However, the PE membranes grafted with poly(AAm) or poly(MPEG) did not show any effects on protein adsorption. Platelet adhesion on the original and modified PE membranes from platelet-rich plasma was also examined. A large number of platelets adhered and activated on the original PE membrane. Grafting with poly(AAm) did not suppress platelet adhesion, but grafting with poly(MPC) or poly(VPy) on the PE membrane was effective in preventing platelet adhesion. It is concluded that the introduction of the phosphorylcholine group on the surface could decrease the cell adhesion to substrate polymer.  相似文献

Surface modification is an effective way to improve the hemocompatibility and remain bulk properties of biomaterials. Recently, polymer tailed with zwitterions was found having good blood compatibility. In this study, the grafting copolymerization of sulfobetaine onto polyurethane surface was obtained through two steps. In the first step, polyurethane film coupled with vinyl groups was obtained through the reaction between the carboxyl group of acrylic acid (AA) and the NH-urethane group of polyurethane by dicyclohexylcarbodiimide (DCC). In the second step, sulfobetaine was grafted copolymerization on the surface using AIBN as an initiator. The reaction process was monitored with ATR-IR spectra and X-ray photoelectron spectroscopy (XPS) spectra. The wettability of films was investigated by water contact angle measurement. The blood compatibility of the grafted films was evaluated by platelet adhesion in platelet rich plasma (PRP) and protein absorption in bovine fibrinogen (BFG). Low platelet adhesion was observed on the grafted films incubated in PRP for 1 and 3 h, respectively. The protein absorption was reduced on the grafted films after incubated in bovine fibrinogen for 2 h. All of these results revealed that the improved blood compatibility was obtained by grafting copolymerization with zwitterionic monomer of sulfobetaine onto polyurethane film. In addition, introducing vinyl groups onto surface through DCC and AA is a novel method to functionalize polyurethane for further modification.  相似文献

通过葡萄糖、丙烯酸羟乙酯和丁二胺反应,制备了含不饱和双键的糖基功能单体。 采用傅里叶红外光谱和核磁共振氢谱对合成的产物进行结构表征确定。 采用紫外光引发接枝聚合技术,将制备的不饱和糖单体接枝聚合到聚氨酯膜的表面,以衰减全反射模式下傅里叶红外光谱对表面接枝反应进行了确认。 通过静态水接触角实验和血小板黏附实验,分别对改性聚氨酯膜表面的亲水性和血液相容性进行了研究,结果表明,改性聚氨酯膜表面的接触角从86°降低到45°,血小板的粘附量由14.36×103 cells/mm2减少到2.57×103 cells/mm2,亲水性明显增强,血液相容性显著改善。  相似文献

Platelet adhesion and protein adsorption on the silicone rubber film grafted with N,N'-dimethyl-N-methacryloyloxyethyl-N-(2-carboxyethyl) ammonium (DMMCA) was studied. The grafting was carried out by means of ozone-induced method and was confirmed by ATR-FTIR and XPS investigations. The grafted films possessed relatively hydrophilic surface revealed by contact angle measurement. The blood compatibility of the grafted film was evaluated in vitro by platelet adhesion in platelet-rich plasma (PRP) and protein absorption in bovine fibrinogen (BFG) using silicone film as the reference. No substantial platelet adhesion was observed for the grafted films incubated in PRP for 60 and 180 min. The protein absorption was also significantly reduced after incubated in bovine fibrinogen for 60 min. Both the results indicated that the blood compatibility of silicone rubber was greatly improved by ozone-induced grafting of carboxybetaine zwitterionic polymer onto its surface.  相似文献

Polycaprolactone (PCL) has been widely adopted as a scaffold biomaterial, but further improvement of the hemocompatibility of a PCL film surface is still needed for wide biomedical applications. In this work, the PCL film surface was functionalized with zwitterionic poly(3-dimethyl(methacryloyloxyethyl) ammonium propane sulfonate) (P(DMAPS)) brushes via surface-initiated atom transfer radical polymerization (ATRP) for enhancing hemocompatibility. Kinetics study revealed an approximately linear increase in graft yield of the functional P(DMAPS) brushes with polymerization time. The blood compatibilities of the modified PCL film surfaces were studied by platelet adhesion tests of platelet-rich plasma and human whole blood, hemolysis assay, and plasma recalcification time (PRT) assay. The improvement of hemocompatibility is dependent on the coverage of the grafted P(DMAPS) brushes on the PCL film. Lower or no platelet and blood cell adhesion was observed on the P(DMAPS)-grafted film surfaces. The P(DMAPS) grafting can further decrease hemolysis and enhance the PRT of the PCL surface. With the versatility of surface-initiated ATRP and the excellent hemocompatibility of zwitterionic polymer brushes, PCL films with desirable blood properties can be readily tailored to cater to various biomedical applications.  相似文献

Polyacrylonitrile (PAN) membrane was hydrolyzed with NaOH(aq) and grafted with conjugated linoleic acid (CLA) via esterification with 1,3‐propanediol. The resulting CLA grafted PAN membranes were characterized using Fourier transform infrared spectrometry (FT‐IR) and X‐ray photoelectronic spectroscopy (XPS). The effects of CLA grafting on the blood coagulation, platelet aggregation, and oxidative stress were evaluated using human blood. The complete blood count (CBC) and coagulation time (CT) was evaluated in vitro for hemocompatibility. After CLA grafting, the proliferation of human umbilical vein endothelial cells (HUVECs) on the membranes were improved. In addition, the production of reactive oxygen species (ROS) was measured by the chemiluminescence (CL) method to evaluate the oxidative stress. The results showed that the CLA‐grafted PAN membrane could keep the CBC values more stable than unmodified PAN membrane. The CLA‐grafted PAN membranes also showed longer CT. CLA‐grafted PAN membrane could keep the CL counts of hydrogen peroxide and superoxide values more stable than unmodified PAN membrane. These results suggest that a CLA‐grafted PAN membrane could offer protection for patients against oxidative stress and would be helpful for reducing the dosage of anticoagulant during hemodialysis. Polyurethane (PU) is a conventional biomedical material with favorable biocompatibility and excellent mechanical properties and widely used in making vascular catheter, but its antithrombogenic property is not good enough to make it as a more demanding applicable biomaterial. Surface modification is an effective way to improve the hemocompatibility for biomaterials. The purpose of present study was to use ozonization method to modify the surface of PU vascular catheter slice to improve its antithrombogenicity by grafting N,N-dimethyl-N-methacryloxyethyl-N-(3-sulfopropyl) ammonium (DMMSA), a zwitterionic sulfobetaine monomer. PU vascular catheter (PUVC) grafted with DMMSA (PUVC-g-PDMMSA) was characterized by ATR-FTIR and XPS. ATR-FTIR and XPS investigation confirmed the graft polymerization. The blood compatibility of the grafted films was evaluated by platelet rich plasma (PRP) platelet adhesion study and scanning electron microscopy (SEM) was used to observe the morphology of platelet using PU vascular catheter (PUVC) as the reference. No platelet adhesion was observed for the grafted PUVC slice incubated with PRP at 37 degrees C for 120 min. It is significant that this new zwitterionic sulfobetaine grafted PUVC have improved antithrobogenicity. It is effective that the inner surface of vascular catheter with inner diameter in only 3mm can be grafted with PDMMSA by using ozonization method.  相似文献

Surface modification of segmented poly(ether urethane) (SPEU) by graft copolymerization with N,N′-dimethyl-N-methacryloyloxyethyl-N-(3-sulfopropyl) ammonium (DMMSA), a zwitterionic sulfobetaine structure, was conducted. A simple two-step procedure for grafting of DMMSA onto the surface of SPEU film was used. The surface was first treated with ozone to introduce active hydroperoxide groups. The active surface was then exposed to the DMMSA solution in the sealed tube. Grafted SPEU film was characterized by ATR–FTIR, XPS and contact angle measurement. ATR–FTIR and XPS investigations confirmed the graft copolymerization. The monomer concentration, copolymerization temperature and time were varied to maximize the efficiency of DMMSA grafting. The equilibrium water content (EWC) and contact angle measurements showed that the hydrophilicity of the film had been greatly improved. The blood compatibility of the grafted films was evaluated by platelet adhesion in platelet rich plasma (PRP), deposits in blood control and protein adsorption in bovine fibrinogen using SPEU film as the control. No platelet adhesion and no thrombus were observed for the grafted films incubated in PRP for 300 min and in blood for 120 min, respectively. The protein adsorption was reduced on the grafted films after incubation in bovine fibrinogen for 120 min. These results proved that improved blood compatibility was obtained by grafting this new zwitterionic sulfobetaine structure monomer onto SPEU film.  相似文献

An ideal nonbiofouling surface for biomedical applications requires both high-efficient antifouling characteristics in relation to biological components and long-term material stability from biological systems. In this study we demonstrate the performance and stability of an antifouling surface with grafted zwitterionic sulfobetaine methacrylate (SBMA). The SBMA was grafted from a bromide-covered gold surface via surface-initiated atom transfer radical polymerization to form well-packed polymer brushes. Plasma protein adsorption on poly(sulfobetaine methacrylate) (polySBMA) grafted surfaces was measured with a surface plasmon resonance sensor. It is revealed that an excellent stable nonbiofouling surface with grafted polySBMA can be performed with a cycling test of the adsorption of three model proteins in a wide range of various salt types, buffer compositions, solution pH levels, and temperatures. This work also demonstrates the adsorption of plasma proteins and the adhesion of platelets from human blood plasma on the polySBMA grafted surface. It was found that the polySBMA grafted surface effectively reduces the plasma protein adsorption from platelet-poor plasma solution to a level superior to that of adsorption on a surface terminated with tetra(ethylene glycol). The adhesion and activation of platelets from platelet-rich plasma solution were not observed on the polySBMA grafted surface. This work further concludes that a surface with good hemocompatibility can be achieved by the well-packed surface-grafted polySBMA brushes.  相似文献

A novel strategy has been developed to improve the hemocompatibility of chitosan surface by cell outer membrane mimetic structure able to reduce protein adsorption and cell adhesion. Phosphorylcholine dichloride was synthesized and grafted onto a glutaraldehyde-cross-linked chitosan (CS-GA) film surface to prepare phosphorylcholine-coated CS-GA film (CS-GA-PC) through a heterogeneous reaction process. The spectroscopic and contact angle characterization show that a cell outer membrane mimetic structure was formed on the cross-linked chitosan surface, and the significantly improved hemocompatibility of the modified surface was shown by a suppression of 94% on platelet adhesion, a suppression of 60–70% for bovine plasma fibrinogen and bovine serum albumin adsorptions. These results demonstrated that this cell outer membrane mimetic surface modification with phosphorylcholine dichloride is a promising strategy to improve the hemocompatibility of chitosan.  相似文献

Conjugated linoleic acid (CLA) was covalently immobilized onto cellulose acetate (CA) membranes. The effects of CLA immobilization on the blood coagulation, platelet aggregation, and oxidative stress were evaluated using human blood. The resulting CLA grafting CA membranes were characterized with X-ray photoelectronic spectroscopy (XPS). The complete blood count (CBC) and coagulation time (CT) was evaluated in vitro for the hemocompatibility. Human serum albumin (HSA) and human plasma fibrinogen (HPF) was evaluated for the protein affinity. The production of reactive oxygen species (ROS) was measured by chemiluminescence (CL) method to evaluate the oxidative stress. The results showed that the CLA-immobilizing CA membrane could keep the CBC values more stable than unmodified CA membrane. The CLA-immobilized CA membranes also showed longer CT and less adsorption of plasma proteins. CLA-immobilized CA membrane could keep the CL counts of hydrogen peroxide and superoxide values more stable than unmodified CA membrane. These results suggest that a CLA-immobilized CA membrane could offer protection for patients against oxidative stress and would be helpful for reducing the dosage of anticoagulant during hemodialysis.  相似文献

Block and random PEGylated copolymers of poly(ethylene glycol) methacrylate (PEGMA) and polystyrene (PS) were synthesized with a controlled polydispersity using an atom transfer radical polymerization method and varying molar mass ratios of PS/PEGMA. Two types of PEGylated copolymers were self-assembly coated onto the surface of poly(vinylidene fluoride) (PVDF) ultrafiltration membranes for enhancing biofouling resistance. It was found that the adsorption capacities of random copolymers on PVDF membranes were all higher than those of block copolymers. However, the specific and overall protein resistance of bovine serum albumin (BSA) on PVDF membranes coated with block copolymers was much higher than that with random copolymers. The increase in styrene content in copolymer increased the amount of polymer coating on the membrane, and the increase in PEGMA content enhanced the protein resistance of membranes. The optimum PS/PEGMA ratio was found to be close to 2 for the best resistance of protein adsorption and bacterial adhesion on the PEGylated diblock copolymer-coated membranes. The PVDF membrane coated with such a copolymer owned excellent biofouling resistance to BSA, humic acid, negatively surface charged bacteria E. coli, and positively surface charged bacteria S. maltophilia.  相似文献

The reduction of free radicals by bioactive membranes used for hemodialysis treatment is an important topic due to the constant rise of oxidative stress‐associated cardiovascular mortality by hemodialysis patients. Therefore, it is urgent to find an effective solution that helps to solve this problem. Polysulfone membranes enriched with α‐lipoic acid, α‐tocopherol, and with both components are fabricated by spin coating. The antioxidant properties of these membranes are evaluated in vitro by determining the lipid‐peroxidation level and the total antioxidant status of the blood plasma. The biocompatibility is assessed by quantifying the protein adsorption, platelet adhesion, complement activation, and hemolytic effect. All types of membranes show in vitro antioxidant activity and a trend to reduce oxidative stress in vivo; the best results show membranes prepared with a combination of both compounds and prove to be nonhemolytic and hemocompatible. Moreover, the membrane specific separation ability for the main waste products is not affected by antioxidants incorporation.  相似文献
