Abietic acid is a naturally occurring enantiomeric diterpenic acid. Its absolute optical purity and very stable stereochemistry structure makes it an excellent starting material for preparing chiral derivatizing reagents for chromatographic or electrophoretic applications. This paper describes the synthesis and evaluation of a novel chiral derivatization reagent, i.e., degradingdehydroabietylisothiocyanate (DDHAIC) derived from dehydroabietic acid. Its applicability for the enantioseparation of racemic amino acids by CE was demonstrated. DDHAIC reacted readily with amino acids at an elevated temperature (70 degrees C). The resulting derivatives were highly stable and separable by MEKC. Separation of amino acid-DDHAIC diastereomers was achieved with a running buffer consisting of 50 mM Na(2)HPO(4) (pH 9.0), 18 mM SDS, and 25% v/v ACN. Under the conditions selected, diastereomers formed from ten pairs of tested amino acid enantiomers including D/L-Asn, D/L-Met, D/L-Leu, D/L-Phe, D/L-Trp, D/L-Ser, D/L-Val, D/L-Ala, D/L-Thr, and R/S-vigabatrin were well resolved. A great interest has been drawn these last years towards ionic liquids in analytical chemistry, especially for separation methods. Recent synthesis of chiral ILs opened the way of the evaluation of new potential selectors for enantiomeric separations. This work focused on the evaluation of two chiral ILs (ethyl- and phenylcholine of bis(trifluoromethylsulfonyl)imide) by CE. Particular selectivities are awaited by exploiting unique ion-ion or ion-dipole interactions and by tailoring the nature of the cation and the anion. To evaluate such phenomena, a study was carried out with anti-inflammatory drugs 2-arylpropionic acids as model compounds. The results show that these chiral ILs did not present direct enantioselectivity with regard to these model analytes. The influence of chiral ILs in the electrolytes was then studied in the presence of classical chiral selectors (di- or trimethyl-beta-cyclodextrin). Although no general trend could be established, an increase in separation selectivity and resolution was observed in some cases, suggesting synergistic effects. The complementary determination of apparent inclusion constant values of these IL cations in the used cyclodextrins by affinity CE provided support to the understanding of the phenomena involved.  相似文献

A total of 26 different cyclodextrin (CD) derivatives with different functional groups and degrees of substitution were tested against 35 basic pharmaceutical compounds in an effort to investigate their effectiveness as chiral selectors for enantiomeric separation in capillary electrophoresis (CE). Testing was performed under the same conditions using a low pH buffer (25 mM phosphate buffer at pH approximately 2.5). Five CD derivatives, namely, highly sulfated-beta-CD, highly sulfated-beta-CD, hydroxypropyl-beta-CD (degree of substitution approximately 1), heptakis-(2,6-O-dimethyl)-beta-CD, and heptakis(2,3,6-O-trimethyl)-beta-CD were identified to be most effective for enantiomeric separations and have a wide range of enantiomeric selectivity towards the model compounds. Over 90% of the model compounds were enantiomerically resolved with the five identified CD derivatives, at a minimum resolution of 0.5. An additional 20 compounds were also tested to demonstrate the validity of the identified CD derivatives. The five CD derivatives were recommended as the starting chiral selectors in developing enantiomeric separation methods by CE.  相似文献

We describe the use of polystyrene (PS) nanoparticles to manipulate chiral selectivity of propranolol analysis by capillary electrophoresis, by dispersing PS nanoparticles into the run buffer employing hydroxypropyl-β-cyclodextrin (HP-β-CD) as chiral selector. Distinct separational differences are observed between the buffer containing PS nanoparticles and buffer without, when changing separating conditions including PS nanoparticles concentration, pH, buffer concentration, HP-β-CD concentration and when adding an organic additive. Selectivity improvements are reflected by changes in the observed mobility as a result of interactions between the propranolol enantiomers and HP-β-CD governing the absorption process on the PS particles surface. The presence of PS nanoparticles increases the enantioseparation at low particle concentration in the presence of HP-β-CD as a chiral selector.  相似文献

Summary Six different cyclodextrins with varying cavity size and rim substitution were used as chiral agents for the enantiomeric separation of eight chromane compounds or analogues using capillary electrophoresis. It is shown that the cyclodextrin type and concentration have a large influence on the enantiomeric separation obtained for these compounds. A chiral resolution of 1.4 or better could be obtained for all the substances with either substituted heptakis(2,3,6-tri-O-methyl)-β-cyclodextrin or unsubstituted γ-cyclodextrin as the chiral selector. The influence of the γ-cyclodextrin concentration, ionic strength and pH on the chiral separations was also investigated with a multivariate screening design. The detection limit and resolution of the present method allow determinations of the investigated compounds down to a chiral impurity of less than 0.1 % (area/area).  相似文献

Capillary electrophoresis (CE) is becoming an established method for the determination of chiral trace impurities. This paper provides an overview of the state of the art of CE for such determinations. Detection limits of 0.1% impurity is widely accepted as a minimum requirement for chiral trace impurity determinations. This can be relatively easily achieved with CE. However, determination of lower concentrations requires careful optimization of the separation system. Four factors that are of particular significance for trace enantiomeric determinations: resolution, limit of detection, linear range and type of detection, are discussed. Further, the advantages and disadvantages of derivatization in this context are treated as well as the separation approach, ie., direct chiral separation or separation after the formation of diastereomers. It is concluded that the limit of impurity detection can be about 0.05% when UV detection is employed. Using laser-induced fluorescence detection, a quantitative determination at the 0.005% level is often possible.  相似文献

Capillary electrophoresis has been increasingly used during the past few years for the separation and determination of surfactants. These substances are applied in many household and industrial products such as laundry detergents, cosmetics and pharmaceuticals, often as homologous and isomeric mixtures. Product development and control as well as toxicological and environmental analyses require selective and sensitive analytical methods. This review presents capillary electrophoretic techniques to determine important representatives of cationic, anionic, and neutral surfactants. The application of different buffer additives such as organic solvents, cyclodextrins or micelles to enhance the resolution of complex mixtures is discussed. Besides direct and indirect UV and fluorescence detection, examples for conductivity and mass spectrometric detection are also given. Derivatization procedures to improve the detectability and implement charge in neutral analytes are described. The successful use of capillary electrophoresis for surfactant determinations has proven that it can serve as a routine technique in many real-world applications. Robust, validated methods for the quantitation of single compounds, such as alkylbenzene sulfonates, sodium dodecyl sulfate and benzalkonium salts, are now available. Characteristic peak patterns (fingerprint analysis) can be used for the identification of surfactants in multicomponent formulations (e.g. ethoxylates and phosphonates).  相似文献

The use of surfactants as additives was demonstrated for the first time in capillary isoelectric focusing (CIEF) to dynamically modify the surfaces of bare fused silica capillaries. These surfactants were zwitterionic sulfobetaines: dodecyldimethyl (3-sulfopropyl) ammonium hydroxide (C12N3SO3), hexadecyldimethyl (3-sulfopropyl) ammonium hydroxide (C16N3SO3) and coco (amidopropyl)hydroxyldimethylsulfobetaine (Rewoteric AM CAS U). They were added directly to the protein-ampholyte mixture, and remained in the capillary during isoelectric focusing and mobilization. The C16N3SO3 and CAS U coatings were shown effective in CEF. Separation of seven IEF protein standards was obtained, with significantly improved resolution compared to that from an uncoated silica capillary. The effect of these surfactants on the electroosmotic flow (EOF) in CIEF was determined. CAS U was effective in suppressing the EOF at neutral and alkaline pH conditions, C16N3SO3 was effective in suppressing EOF at acidic and neutral pH conditions. C12N3SO3 however had little effect on the EOF. The pH gradients formed inside these surfactant coated capillaries were recta-linear at pH 6 to 9 (R2 approximately equal to 0.99). Reproducibility of migration time and peak area was determined. For all three coatings, the migration time standard deviations were less than 1.6 min, and the relative standard deviations of area were below 10%. The protein recovery in the CAS U-modified capillary was quantitative or near-quantitative for five of the seven proteins studied.  相似文献

A method is described for the separation of quaternary alkylbenzylammonium compounds as well as alkyl pyridinium salts by capillary electrophoresis using direct UV detection. The influence of the organic buffer modifier on the electrophoretic behaviour of the analytes is discussed. In addition to fused silica capillaries, also C8, C18 and neutral surface coatings are used. Separation is also performed in completely non-aqueous media. The results of method development are applied to the determination of cationic surfactants in cosmetics and pharmaceuticals. A rapid method for the enantioseparation of pramipexole and its R‐enantiomer has been developed by capillary electrophoresis. The influence of chemical and instrumental parameters was investigated including the type and concentration of chiral selectors, buffer composition and pH, co‐ions, applied voltage, capillary length and temperature. Optimal separation conditions were obtained using a 50 mM phosphate buffer (pH 2.8) containing 25 mM carboxymethyl‐β‐cyclodextrin on a fused‐silica capillary. Online UV detection was performed at 262 nm. A voltage of 25 kV was applied, and the capillary temperature was kept at 25°C. Hydrodynamic injection was performed at 3.45 kPa for 5.0 s. The separation of enantiomers was achieved in <6.5 min. The method was further validated in terms of stability of solutions, selectivity, linearity (both pramipexole and R‐enantiomer, R2>0.995), LOD and LOQ (0.91 and 2.94 μg/mL, respectively), repeatability (RSD<1.5%) and accuracy (pramipexole, 100.4%; R‐enantiomer, 100.5%). The proposed method was then applied to two kinds of pramipexole dihydrochloride monohydrate commercially available tablets, immediate release tablets (1.50 and 0.125 mg) and sustained release tablets (0.52 mg), to quantify the main component in the tablets. The amount of distomer could be quantified in bulk sample materials.  相似文献

Chiral cyclohexyl-pentyl-beta-D-maltoside (CYMAL-5) surfactant was evaluated in the enantioseparation of charged racemic species by capillary electrophoresis. CYMAL-5 is a glycosidic surfactant (GS) with a chiral maltose polar head group and a cyclohexyl-pentyl hydrophobic tail. At concentrations above its critical micellar concentration (CMC), CYMAL-5 produces neutral micelles in aqueous media. The neutral micelles migrate at the velocity of the electroosmotic flow (EOF). As expected, the CYMAL-5 system was only useful for the enantioseparation of charged chiral solutes. The enantioresolution of the CYMAL-5 can be manipulated over a wide range of electrolyte composition, e.g., pH, ionic strength and surfactant concentration. In the presence of EOF, and in all cases, there is an optimum surfactant concentration for maximum enantioresolution, which is located at low surfactant concentration for strongly hydrophobic solutes and at high surfactant concentration for relatively hydrophilic solutes. The presence of an optimum surfactant concentration for maximum enantioresolution is attributed to the EOF. At low pH values where the EOF is negligible, enantioresolution increased with increasing surfactant concentration in the useful concentration range in a way similar to chromatography.  相似文献

A wide number of chiral selectors have been employed in CE and among them macrocyclic antibiotics exhibited excellent enantioselective properties toward plenteous racemic drugs. Different from macrocyclic antibiotics, the use of lincomycin antibiotics as chiral selectors has not been reported previously. In this study clindamycin phosphate belonging to the group of lincomycin antibiotics is first used as a novel chiral selector for the enantiomeric separations of several racemic basic drugs, which possess high separability, consisting of nefopam, citalopram, tryptophan, chlorphenamine and propranolol. Other basic drugs giving partial enantioseparation include tryptophan methyl ester, metoprolol and atenolol. Clindamycin phosphate possesses advantages such as high solubility and low viscosity in the water and very weak UV absorption. In the course of this work we observed that both migration time and enantioseparation were influenced by several parameters such as pH of the BGE, clindamycin concentration, capillary temperature, applied voltage and organic modifier. The optimum pH that was in the neutral or weak basic region but varied among drugs, a low capillary temperature and a clindamycin concentration of 60 or 80 mM are recommended as the optimum conditions for chiral separation of these drugs. Moreover, comparison of the influences of the studied parameters was further investigated by means of Statistical Product and Service Solutions in this paper.  相似文献

A chiral procedure based on EKC was developed and validated for determination of the enantiomeric purity of PHA-543613, a drug candidate that was under development for treatment of the cognitive deficits of Alzheimer's disease and schizophrenia. Separation of enantiomers is accomplished via differential, enantiospecific complexation with a single-isomer, precisely sulfated beta-CD and heptakis-6-sulfato-beta-CD (HpS-beta-CD). Both neutral and sulfated CDs were screened before selecting HpS-beta-CD as the chiral selector. The separation is conducted in a 61 cm x 50 microm uncoated fused silica capillary with 25 mM HpS-beta-CD in pH 2.50, 25 mM lithium phosphate as the separation buffer with detection at 220 nm. Application of reverse polarity at -30 kV results in an elution time of about 12 min for PHA-543613 and 13 min for the undesired S-enantiomer. Quantification is versus an authentic reference S-enantiomer as an external standard in combination with an internal standard. The procedure was validated over the range 0.1-2.0% w/w. The detection limit is 0.01-0.02%. The amount of distomer intrinsic to the drug substance is about 0.1% or less. The developed method was used to generate stability data on multiple lots: in one case for up to 3 years.  相似文献

The enantiomeric separation of alpha-hydroxy acids and carboxylic acids was successfully performed by using 6-deoxy-6-N-histamino-beta-cyclodextrin (CD-hm), a monosubstituted positively charged beta-cyclodextrin (beta-CD) bearing a histamine moiety linked to the C6 of a glucose unit in the upper CD rim via the amino group. Good results were obtained at a low selector concentration (1 mM). The number of positive charges on the upper rim may be modulated as a function of pH, because of the different pKa of the amino and the imidazolyl groups, and was found to affect both the enantioselectivity and resolution factors. With the analogous 6-deoxy-[4-(2-aminoethyl)imidazolyl]-beta-cyclodextrin (CD-mh) bearing the histamine moiety linked to the C6 via the imidazolyl group, very poor results were obtained, showing that the proximity of the positive charge to the cavity plays an important role in the enantiomeric recognition. The complexation mode was studied by electrospray ionization-mass spectrometry (ESI-MS) and two-dimensional nuclear magnetic resonance (2-D NMR) ROESY experiments: the recognition model is consistent with an inclusion complexation of the aromatic ring of the analyte within the CD cavity coupled to electrostatic interactions between the carboxylate and the protonated amino group of the cyclodextrin.  相似文献

A review is presented that surveys the basic theory of direct separation of enantiomers by capillary electrophoretic (CE) techniques. These separations are based on the formation of diastereomeric complexes between the enantiomeric analytes and a chiral selector added to the electrolyte solution. The review covers a comprehensive treatment of the equations needed for optimization of selectivity coefficients, resolution and analysis time in the zone electrophoretic mode. In this context, it takes into account combined equilibria of complexation and protonation/deprotonation as well as complexation and paritition into micelles. On the basis of these equations, the benefits of charged selectors and the optimization potential inherent to pH tuning can be documented. In addition, the review deals with some basic aspects of chiral isoelectric focusing and briefly discusses indirect enantioseparation. In a subsequent section a survey is given on particularfeatures of the various types of chiral selectors. Finally, the recent developments in preparative enantioseparation in continuous free-flow system and by use of isoelectric membranes are discussed.  相似文献

For separation of enantiomers in presence of a chiral selector, data obtained with the 1D dynamic simulators SIMUL5complex and GENTRANS are compared to data predicted by PeakMaster 6, a recently released generalized model of the linear theory of electromigration. Four electrophoretic systems with stereoisomers of weak bases were investigated. They deal with the estimation of input data for complexation together with the elucidation of the origin of observed system peaks, the interference of analyte and system peak migration, the change of enantiomer migration order as function of the selector concentration and the inversion of analyte migration direction in presence of a multiply negatively charged selector. For all systems, data predicted with PeakMaster 6 are in agreement with those of the dynamic simulators and simulation data compare well with experimental data that were monitored with setups featuring conductivity and/or UV absorbance detection along the capillary. SIMUL5complex and GENTRANS provide the full dynamics of any buffer and sample arrangement and require very long execution time intervals. PeakMaster 6 is restricted to conventional CZE, is based on an approximate solution of the transport equations, provides data for realistic experimental conditions within seconds and represents a practical tool for an experimentalist.  相似文献
