Single-walled carbon nanotubes (SWCNTs) are versatile near infrared (NIR) fluorescent building blocks for biosensors. Their surface is chemically tailored to respond to analytes by a change in fluorescence. However, intensity-based signals are easily affected by external factors such as sample movements. Here, we demonstrate fluorescence lifetime imaging microscopy (FLIM) of SWCNT-based sensors in the NIR. We tailor a confocal laser scanning microscope (CLSM) for NIR signals (>800 nm) and employ time correlated single photon counting of (GT)10-DNA functionalized SWCNTs. They act as sensors for the important neurotransmitter dopamine. Their fluorescence lifetime (>900 nm) decays biexponentially and the longer lifetime component (370 ps) increases by up to 25 % with dopamine concentration. These sensors serve as paint to cover cells and report extracellular dopamine in 3D via FLIM. Therefore, we demonstrate the potential of fluorescence lifetime as a readout of SWCNT-based NIR sensors.  相似文献

Light-triggered drug delivery system is an effective strategy for precise diagnosis and therapy in cancer treatment. However, it suffers from difficultly balancing the dosimetry of drug with light dose and a lack of in vivo models for validating their clinical benefits. Here we report an unprecedented near-infrared (NIR) light photocaged cyanine-based prodrug Cy-CPT-Biotin with dual-channel fluorescence mode, enabling NIR light to precisely regulate where, when and how the intact and active prodrugs are delivered. The synergy of photochemical reaction and modulation in π-conjugated polyene backbone of cyanine can fully perform distinct dual-channel fluorescence changes in a NIR light-mediated manner. The prodrug has striking characteristics of excellent tumor-targeting ability, real-time monitoring of the in vivo behaviors by dual-channel mode and NIR-light triggering, especially for achieving fine regulation and on-demand drug release in the precise dosimetry of drug with light dose in living animals. This optical orthogonality strategy that conjuncts with NIR light-triggered and dual-channel fluorescence in vivo imaging provides a powerful tool for in vivo real-time tracking and finely tuning the prodrug release for precise drug delivery.  相似文献

In recent years, semiconducting polymer dots (Pdots) have emerged as a new type of ultrabright fluorescent probes, which have been proved to be very useful for biomedical imaging. Pdots possess several exceptional advantages including high fluorescence brightness, fast radiative rate, excellent photostability, and negligible cytotoxicity. Among these new types of Pdots, the near‐infrared (NIR) fluorescent Pdots appear to be the most urgent and important owing to their promising deep‐tissue imaging in the clinic. This mini‐review highlights the recent progress in the design of NIR‐emitting Pdots and their biomedical applications both in vitro and in vivo.  相似文献

Taking the advantage of reduced scattering and low autofluorescence background, the NIR fluorescence probes, such as fluorescence proteins, organic molecules and nanoparticles, not only hold the promise of in vivo imaging of biological processes in physiology and pathology with high signal-to-noise ratio, but also for clinical diagnosis. In this review, we provide an overview of the recent progress on NIR probes, focusing on fundamental mechanisms of NIR dyes and nanoparticles, and protein engineering strategies for NIR proteins.  相似文献

Near-infrared (NIR) fluorescence imaging is promising due to the high penetration depths and minimal levels of autofluorescence in living systems. However, it suffers from low fluorescent quantum yield, and metal-enhanced fluorescence (MEF) is considered to be a promising technique to overcome this. Stimuli-responsive NIR fluorescence enhancement shows remarkable potential for applications in medical imaging and diagnosis. Herein, we successfully fabricated an enzyme-responsive near-infrared sensor based on MEF by functionalizing gold nanoparticles with NIR fluorophores and enzyme-responsive self-aggregation moieties. The NIR fluorescence of fluorophores on the gold nanoparticles was significantly enhanced due to increases both in the light scattering intensity and in the radiative decay rate (k r) of the NIR fluorophores, along with relatively small variation in the nonradiative decay rate. This novel strategy for NIR fluorescent sensors should be particularly promising for NIR fluorescence imaging of enzyme activities and early diagnosis based on rationally designed nanomaterials.  相似文献

Single‐walled carbon nanotubes (SWCNTs) are a 1D nanomaterial that shows fluorescence in the near‐infrared (NIR, >800 nm). In the past, covalent chemistry was less explored to functionalize SWCNTs as it impairs NIR emission. However, certain sp3 defects (quantum defects) in the carbon lattice have emerged that preserve NIR fluorescence and even introduce a new, red‐shifted emission peak. Here, we report on quantum defects, introduced using light‐driven diazonium chemistry, that serve as anchor points for peptides and proteins. We show that maleimide anchors allow conjugation of cysteine‐containing proteins such as a GFP‐binding nanobody. In addition, an Fmoc‐protected phenylalanine defect serves as a starting point for conjugation of visible fluorophores to create multicolor SWCNTs and in situ peptide synthesis directly on the nanotube. Therefore, these quantum defects are a versatile platform to tailor both the nanotube's photophysical properties as well as their surface chemistry.  相似文献

Biomaterials for in vivo fluorescence imaging are required to be biocompatible, nontoxic, photostable and highly fluorescent. Fluorescence must be in the near infrared (NIR) region of the electromagnetic spectrum to avoid absorption and autofluorescence of endogenous tissues. NIR fluorescent polystyrene nanoparticles may be considered ideal biomaterials for in vivo imaging applications. These NIR nanoparticles were prepared by a swelling process of polystyrene template nanoparticles with a hydrophobic NIR dye dissolved in a water‐miscible swelling solvent, a method developed for preparation of nonbiodegradable nanoparticles, for NIR fluorescent bioimaging applications. This method overcomes common problems that occur with dye entrapment during nanoparticle formation such as loss of fluorescence and size polydispersity. Fluorescence intensity of the nanoparticles was found to be size dependent, and was optimized for differently sized nanoparticles. The resulting NIR nanoparticles were also found to be more fluorescent and highly photostable compared to the free dye in solution, showing their potential as biomaterials for in vivo fluorescence imaging.  相似文献

Glutathione (GSH) plays a critical role in maintaining oxidation-reduction homeostasis in biological systems. Considering the detection of GSH by fluorescence sensors is limited by either the short wavelength emission or the poor photostability, a highly stable colorimetric and ratiometric NIR fluorescent sensor (DCM-S) for GSH detection has been constructed on the basis of dicyanomethylene-4H-pyran (DCM) chromophore. The specific disulfide bond is incorporated via a carbamate linker as the GSH responsive group, which simultaneously blue-shifts and quenches the fluorescence. Upon addition of GSH, DCM-S exhibits outstanding colorimetric (from yellow to red) and ratiometric fluorescent response with the 6-fold enhancement of NIR fluorescence at 665 nm in quantum yield. More importantly, the GSH-treated DCM-S (DCM-NH2 actually) possesses 20-fold longer fluorescence half-life period as well as much better photostability than the FDA-approved ICG. Finally, the ratiometric detection of GSH is also successfully operated in the living cell imaging, exhibiting NIR fluorescence and large Stokes shift (215 nm) with nearly no background fluorescence interference. As a consequence, DCM-S can be utilized as colorimetric and ratiometric NIR fluorescent sensor for GSH, with a great potential in the development of GSH-induced drug delivery system.  相似文献

Molecularly near-infrared(NIR) theranostics, combining in vivo sensing and tumor-specific therapeutic capability within one molecular system, have received considerable attention in recent years. Compared with the visible fluorescence imaging, NIR imaging(emission wavelength at 650–900 nm) possesses unique advantages including the minimum photodamage to biological samples, deep penetration, and low interference from auto-fluorescence. In over past decades, there has been an explosive development in the design of molecular imaging contrasts and imaging-guided therapeutics. In this review, we have sumarried the strategies of the NIR theranostics for imaging and tumor-specific chemotherapy applications in living systems. It is noted that the molecularly NIR theranostic design strategy could address current challenges of real-time in vivo sense-and-release for the intelligent biosensing and personalized treatment.  相似文献

单壁碳纳米管的制备方法主要包括物理法和化学法两种,不同制备方法的产物中包含很多种不同手性指数的单壁碳纳米管及杂质,而不同手性的单壁碳纳米管其作用完全不同,从而有不同的应用。采用化学方法——钴钼催化剂气相沉积法和物理方法——激光烧蚀气相沉积法制备单壁碳纳米管,用紫外–可见–近红外吸收光谱、荧光光谱及拉曼光谱法分别对其进行手性测量。结果表明,钴钼催化剂气相沉积法制得样品富含半导体型单壁碳纳米管,而激光烧蚀气相沉积法制得样品中主要为金属型单壁碳纳米管。分别得到了两种制备方法样品的手性分布。  相似文献

The addition of diazonium salts to single-walled carbon nanotubes (SWCNTs) in aqueous surfactant suspensions quenches the intrinsic near-infrared fluorescence of semiconducting SWCNTs through sidewall chemical reactions. Spectrally resolved fluorescence spectroscopy of mixed SWCNT samples has been used to measure structure-dependent relative reactivities in the initial stages of these reactions. For several 4-substituted benzenediazonium salts, Ar-R (Ar = N 2 (+)-C 6H 4 and R = Cl, NO 2, OMe), reactivities at pH 10 were found to be greatest for SWCNTs having the largest band gaps. The magnitude of this band gap dependence varies according to the R-group of the salt, with R = OMe showing the strongest variation. For R = OH, acidification of the sample to pH 5.5 results in reversal of the structural trend, as smaller band gap SWCNTs show slightly greater reactivities. The derivatization reactions observed here proceed concurrently, although at different rates, for semiconducting and metallic SWCNT species. These results therefore provide insight into the difficulties of separating metallic and semiconducting SWCNTs through selective reaction schemes and underscore the need for fluorescence spectroscopy to be used in assessing semiconducting SWCNT reactions.  相似文献

First-principles calculations based on density functional theory (DFT) method are used to investigate the adsorption properties of nerve agent DMMP on typical zigzag (semiconducting) and armchair (metallic) single wall carbon nanotubes (SWCNTs). The adsorption energies for DMMP molecule on different adsorption sites on SWCNTs are obtained. The results indicate that DMMP is weakly bound to the outer surface of both the considered SWCNTs and the obtained adsorption energy values and binding distances are typical for the physisorption. We find that DMMP adsorptive capability of metallic CNTs is about twofold that of semiconducting one. The adsorption of DMMP on the higher chiral angle nanotubes was also investigated and the results indicate that nanotube’s chirality increases the adsorption capability of the tube but however the adsorption characteristic is typical for the physisorption. Furthermore, co-adsorption of two DMMP molecules on the SWCNTs as a single-layer/bi-layer of adsorbed molecules as well as the adsorption of one DMMP molecule on the CNT bundles consisting of three SWCNTs has also been examined. The obtained results reveal that for both the considered systems the binding energy was increased for the DMMP adsorption but it’s still typical for the physisorption, consistent with the recent experimental result. The study of the electronic structures and charge analysis indicate that no significant hybridization between the respective orbital takes place and the small interaction obtained quantitatively in terms of binding energies.  相似文献

An amino‐acid‐based (11‐(4‐(pyrene‐1‐yl)butanamido)undecanoic acid) self‐repairing hydrogel is reported. The native hydrogel, as well as hybrid hydrogels, have been thoroughly characterized by using various microscopic techniques, including transmission electron microscopy (TEM), atomic force microscopy (AFM), Raman spectroscopy, fluorescence spectroscopy, FTIR spectroscopy, X‐ray diffraction, and by using rheological experiments. The native hydrogel exhibited interesting fluorescence properties, as well as a self‐healing property. Interestingly, the self‐healing, thixotropy, and stiffness of the native hydrogel can be successfully modulated by incorporating carbon‐based nanomaterials, including graphene, pristine single‐walled carbon nanotubes (Pr‐SWCNTs), and both graphene and Pr‐SWCNTs, within the native gel system. The self‐recovery time of the gel was shortened by the inclusion of reduced graphene oxide (RGO), Pr‐SWCNTs, or both RGO and Pr‐SWCNTs. Moreover, hybrid gels that contained RGO and/or Pr‐SWCNTs exhibited interesting semiconducting behavior.  相似文献

The development of multifunctional theranostic nano-agents is an important resolution for personalized treatment of cancer. In this work, we synthesized a new kind of gadolinium boride nanoparticles (GBN) by a microwave-assisted chemical etching method, and discovered their optical characteristics including fluorescence imaging and near-infrared (NIR) photothermal conversion capability. Bright greenishyellow fluorescence enabled for intracellular localization, while effective NIR-photothermal conversion supported photothermal therapy (PTT). In vitro and in vivo results indicated that GBN exhibited a superior antitumor performance and high biocompatibility. This study demonstrated a promising multifunctional theranostic nanoplatform for cancer treatment.  相似文献

Fluorescent probes play a key role in modern biomedical research. As compared to inorganic quantum dots (QDs) composed with heavy metal elements, organic dye-based fluorescent nanoparticles have higher biocompatibility and are richer in variety. However, traditional organic fluorophores tend to quench fluorescence upon aggregation, which is known as aggregation-caused quenching (ACQ) effect that hinders the fabrication of highly emissive fluorescent nanoparticles. In this work, we demonstrate the synthesis of organic fluorescent dots with aggregation-induced emission (AIE) in far-red/near-infrared (FA/NIR) region. A conventional ACQ-characteristic fluorescent dye, 3,4:9,10-tetracarboxylic perylene bisimide (PBI), is converted into an AIE fluorogen through attaching two tetraphenylethylene (TPE) moieties. The fluorescent dots with surface folic acid groups are fabricated from PBI derivative (DTPEPBI), showing specific targeting effect to folate receptor-overexpressed cancer cells. In vivo studies also suggest that the folic acid-functionalized AIE dots preferentially accumulate in the tumor site through enhanced permeability and retention (EPR) effect and folate receptor-mediated active targeting effect. The low cyto-toxicity, good FR/NIR contrast and excellent targeting ability in in vitro/in vivo imaging indicate that the AIE dots have great potentials in advanced bioimaging applications.  相似文献

Identification of fluorescent biomarkers with peptide ligand-directed receptors for diagnosis or theranostic of pancreatic ductal adenocarcinoma (PDAC) is still challenging. As potential prognostic/predictive bioimaging targets, both aminopeptidase N (APN, known as CD13) and Caveolin-1 are found as upregulation on the cell membrane surface of PDAC, in which APN is the principal receptor of the cyclic peptide cNGR (Asn-Gly-Arg, NGR) and Caveolin-1 can synergistically mediate endocytosis in this receptor-targeted process. Herein, we conjugate cNGR to dicyanomethylene-4H-pyran (DCM) chromophore to develop a synergistic-targeted near-infrared (NIR) fluorescent probe DCM-cNGR with strongly intrinsic NIR fluorescence, stable optical performance, low cytotoxicity, and rapid accumulation in PANC-1 cells with the synergistic overexpressed APN receptor-targeted and Caveolin-1-mediated endocytosis. As demonstrated, DCM-cNGR can realize noninvasive NIR imaging for targeting PANC-1 tumor in vivo after intravenous injection into PANC-1 xenograft tumor of nude mice, making a great promise to improve the precision diagnosis and therapy of pancreatic cancer with real time tracing and bioimaging of PDAC in vitro and in vivo.  相似文献

The addition of SH and OH groups to single‐wall carbon nanotubes (SWCNTs) was investigated employing first principles calculations. In the case of the semiconducting (10, 0) SWCNT the SWCNT‐SH binding energy is weak, 2–4 kcal/mol. However, for the metallic (5, 5) SWCNT it is larger, 7–9 kcal/mol. Thus metallic SWCNTs seem to be more reactive to SH than the semiconducting ones. Indeed, the (6, 6) SWCNT is more reactive to SH than the (10, 0) SWCNT, by 2–3 kcal/mol, something that can be explained only considering the electronic structure of the tube, because the (6, 6) has a larger diameter. The binding energies are larger for the addition of the OH group, 25 and 30 kcal/mol for the (10, 0) and (5, 5) SWCNTs, respectively. When a single OH or SH group is attached to the metallic SWCNTs, we observe important changes in the DOS at the Fermi level. However, when multiple SH groups are attached, the changes in the electronic and magnetic properties depend on the position of the SH groups. The small binding energy found for the SH addition indicates that the successful functionalization of SWCNTs with SH, SCH3, and S(CH2)nSH groups is mostly due to the presence of defects created after acid treatment and to a minor extent by the metallic tubes present in the samples. A novel electrochemical sensor based on Cu-MOF-199 [Cu-MOF-199 = Cu3(BTC)2 (BTC = 1,3,5-benzenetricarboxylicacid)] and SWCNTs (single-walled carbon nanotubes) was fabricated for the simultaneous determination of hydroquinone (HQ) and catechol (CT). The modification procedure was carried out through casting SWCNTs on the bare glassy carbon electrode (GCE) and followed by the electrodeposition of Cu-MOF-199 on the SWCNTs modified electrode. Cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and scanning electron microscopy (SEM) were performed to characterize the electrochemical performance and surface characteristics of the as-prepared sensor. The composite electrode exhibited an excellent electrocatalytic activity with increased electrochemical signals towards the oxidation of HQ and CT, owing to the synergistic effect of SWCNTs and Cu-MOF-199. Under the optimized condition, the linear response range were from 0.1 to 1453 μmol L−1 (RHQ = 0.9999) for HQ and 0.1–1150 μmol L−1 (RCT = 0.9990) for CT. The detection limits for HQ and CT were as low as 0.08 and 0.1 μmol L−1, respectively. Moreover, the modified electrode presented the good reproducibility and the excellent anti-interference performance. The analytical performance of the developed sensor for the simultaneous detection of HQ and CT had been evaluated in practical samples with satisfying results.  相似文献

The typical aza-BODIPYs in the dye family are known for bright fluorescence, excellent stability, and tunable absorption wavelengths. Hence, these dyes are attracting the increasing attention. Aza-BODIPYs having the maxima absorption in the near-infrared (NIR) region (650–900 nm) are very favorable for bioimaging in vivo due to the less photo-damage, deeper tissue penetration, and less interference from background auto-fluorescence by biomolecules in the living systems. Many strategies have been employed to modify the structures of the aza-BODIPY core to provide the NIR absorbing dyes. Among these, the most effective method is the fusion of the aromatic rings in aza-BODIPY system. This review allsidedly summarizes the recent development of ring-fused aza-BODIPY dyes (λabs > 700 nm) focusing on the design, synthesis, and potential applications in the NIR region since 2002.  相似文献

Cysteine is well-known to be an important biothiol and related to many diseases. However, the in vivo detection of endogenous cysteine still suffers from lacking small-molecule fluorophores with both excitation and emission in the near-infrared (650-900 nm)/shortwave-infrared region. Herein, we report a molecular engineering strategy for shortwave infrared (SWIR, 900-1700 nm) sensing of cysteine, which integrated an excited-state intermolecular proton transfer (ESIPT) building block into the intramolecular charge transfer (ICT) scaffold. The obtained novel fluorophore SH-OH displays a maximum absorption at the NIR region, and emission at the SWIR region. We introduce the cysteine-recognition moiety to SH-OH structure, and demonstrate sensing of endogenous cysteine in living animals, using the SWIR emission as a reliable off-on fluorescence signal. This fluorophore design strategy of cooperation of ICT and ESIPT processes expands the in vivo sensing toolbox for accurate analysis in clinical applications.  相似文献
