A simple, sensitive, and rapid reversed-phase high-performance liquid chromatographic method has been developed for determination of famotidine (FMT) and its impurities in pharmaceutical formulations. Separations were performed on a Supelcosil LC18 column with an isocratic mobile phase—13:87 (v/v) acetonitrile–0.1 M dihydrogen phosphate buffer containing 0.2% triethylamine (pH 3.0). The mobile phase flow rate was 1 mL min–1 and the detection wavelength was 265 nm. Response was linearly dependent on concentration between 1 and 80 g mL–1 (regression coefficient, R2, from 0.9981 to 0.9999). RSD from determination of method repeatability (intraday) and reproducibility (interday) were <2% (n=6). Lowest detectable concentrations ranged from 0.08 to 0.14 g mL–1. The proposed liquid chromatographic method can be satisfactorily used for routine quality control of famotidine in pharmaceutical formulations.  相似文献

A sensitive, simple, and accurate high-performance liquid chromatographic method has been developed for determination of valdecoxib and the internal standard rofecoxib in human plasma. Protein was precipitated from plasma samples by addition of perchloric acid (HClO4); the drug was then extracted with diethyl ether. Separation was performed on a Cosmosil C18 column (150 mm × 4.6 mm i.d., 5 m particles) with ammonium acetate buffer-acetonitrile, 60:40 (v/v), containing 0.1% TEA, pH 6.5, as mobile phase. Detection and quantification were performed by UV-visible detection at 239 nm. Detection and quantification limits were 3 and 5 ng mL–1, respectively. The linear concentration range for valdecoxib was 5–400 ng mL–1. The validated RP HPLC method was used for determination of the pharmacokinetic data for the drug in humans.  相似文献

The reversed-phase high-performance liquid chromatography (RP-HPLC) method was developed for determination of lercanidipine hydrochloride and its synthetic impurities, degradation and oxidative products in Carmen® tablets. The best separation was performed on Zorbax SB C18 column, 250 x 4.6 mm, particle size 5 m. Acetonitrile-water-triethylamine 55:44.8:0.2 (v/v/v) was used as a mobile phase with flow rate 1 mL min–1. pH was adjusted to 3.0 with orthophosphoric acid. UV detection was performed at 240 nm. Duration of chromatographic run was about 12 min for six examined compounds. The chromatographic conditions for the determination of lercanidipine hydrochloride and its related substances were the same, but the concentration of lercanidipine hydrochloride was 0.03 mg mL–1 for assay and 0.3 mg mL–1 for related substances. The validation of the method performance characteristics (figures of merits, quality of parameters) was established to be adequate for the intended use. A simple, reliable, and reproducible method for in-vivo on-line separation and determination of levodopa has been based on microdialysis then high-performance liquid chromatography with chemiluminescence detection. The perfusate is perfused at a flow rate of 5 L min–1. The concentration of levodopa in the dialysate is determined on line with a chemiluminescence system. The dialysate sample volume is approximately 20 L. The response of the system is linearly related to the concentration of levodopa in the range 1×10–8 to 1×10–6 g mL–1 (r2=0.9995) with a detection limit (3) of 3×10–9 g mL–1 and sample throughput of 12 h–1; RSD is 2.8% (n=11). The method has been successfully used to study the pharmacokinetics of levodopa in vivo; the values of the pharmacokinetics parameters Cmax, AUC0–t and Tmax were 16.60, 20.92 ng mL–1, and 90 min, respectively.  相似文献

A high-performance liquid chromatography (HPLC) method for the determination of acetaldehyde in fuel ethanol was developed. Acetaldehyde was derivatized with 0.900 mL 2,4-dinitrophenylhydrazine (DNPHi) reagent and 50 L phosphoric acid 1 mol L–1 at a controlled room temperature of 15°C for 20 min. The separation of acetaldehyde-DNPH (ADNPH) was carried out on a Shimadzu Shim-pack C18 column, using methanol/LiCl(aq) 1.0 mM (80/20, v/v) as a mobile phase under isocratic elution and UV–Vis detection at 365 nm. The standard curve of ADNPH was linear in the range 3–300 mg L–1 per injection (20 L) and the limit of detection (LOD) for acetaldehyde was 2.03 g L–1, with a correlation coefficient greater than 0.999 and a precision (relative standard deviation, RSD) of 5.6% (n=5). Recovery studies were performed by fortifying fuel samples with acetaldehyde at various concentrations and the results were in the range 98.7–102%, with a coefficient of variation (CV) from 0.2% to 7.2%. Several fuel samples collected from various gas stations were analyzed and the method was successfully applied to the analysis of acetaldehyde in fuel ethanol samples.  相似文献

A new, simple, sensitive and selective fluorometric method for the determination of nitrite has been developed. The reaction of nitrite with hydralazine in acidic medium, heated on a boiling water-bath for 15 min, produced a tetrazolo (5,1-a) phthalazine (Tetra-P). The product formed was measured at ex = 274 nm and em = 345 nm. The fluorescence intensity was valid over a nitrite concentration range 0.067–60.3 ng mL–1, with a detection limit of 0.0091 ng mL–1. The reproducibility of the proposed method was determined by running a different concentration of nitrite, 13.4, 33.5, and 46.9 ng mL–1. The % recoveries and the relative standard deviations were found to be 100.6 ± 0.9, 99.9 ± 0.5, and 99.4 ± 1.1%, respectively. The proposed method was applied successfully to the determination of nitrogen dioxide sampled from the atmosphere using the liquid droplet method. The nitrogen dioxide our wind tunnel was controlled by an NO x analyzer based on a chemiluminescence analyzer detector (CLAD 1000). A linear graph was obtained for the nitrogen dioxide in the wind tunnel vs. NO2 sampled by the liquid droplet method. The effect of interference substances in the determination showed that cations and anions did not disturb the process. The results obtained were satisfactory when compared with the reference method.  相似文献

Stir-bar-sorptive extraction followed by liquid desorption and large-volume injection capillary gas chromatography with mass spectrometric detection (SBSE–LD–LVI-GC–MS), had been applied for the determination of ultra-traces of eight pyrethroid pesticides (acrinathrin, cypermethrin, deltamethrin, esfenvalerate, fenpropathrin, fenvalerate, and permethrin cis and trans isomers) in water samples. Instrumental calibration for selected-ion monitoring acquisition and conditions that could affect the SBSE–LD efficiency are fully discussed. By performing systematic assays on 30-mL water samples spiked at the 0.10 g L–1 level it was established that stir-bars coated with 47 L polydimethylsiloxane, an equilibrium time of 60 min (750 rpm), 5% methanol as organic modifier, and acetonitrile as back-extraction solvent, provided the best analytical performance to monitor pyrethroid pesticides in water matrices. Good accuracy (81.8–105.0%) and remarkable reproducibility (<11.7%) were obtained, and the experimental recovery data were in good agreement with the theoretical equilibrium described by octanol–water partition coefficients (log KO/W), with the exception of acrinathrin for which lower yields were measured. Excellent linear dynamic ranges between 25 and 400 ng L–1 (r2>0.994), low quantification (3.0–7.5 ng L–1) and detection (1.0–2.5 ng L–1) limits were also achieved for the eight pyrethroid pesticides studied. The method was successfully used for analysis of tapwater and groundwater matrices spiked at the 0.10 g L–1, revealing the suitability of the method for determination of pyrethroid pesticides in real samples. The method was shown be reliable and sensitive and a small volume of sample was required to monitor pyrethroids at ultra-trace levels, in compliance with international regulatory directives on water quality.  相似文献

A method for the determination of triphenyltin and diphenyltin was developed by reversed-phase high-performance liquid chromatography with UV detection. Triphenyltin and diphenyltin were separated using a reversed-phase Symmetry C18 column (150 × 3.9 mm, 5 m) with tetrahydrofuran-water-acetonitrile-glacial acetic acid (13:25:5:7, v/v) containing 0.05% triethylamine and 1.0% sodium acetate as mobile phase at 0.50 mL min–1 and detection at 257 nm. The calibration curves were linear from 0.26 mol L–1 to 1100 mol L–1 for triphenyltin with a correlation coefficient of 0.9999 (n=12) and from 0.60 mol L–1 to 1200 mol L–1 for diphenyltin with a correlation coefficient of 0.9991 (n=12), respectively. The detection limits of triphenyltin and diphenyltin were 0.2 mol L–1 and 0.4 mol L–1, respectively. The method was successfully applied to the determination of triphenyltin and its metabolite diphenyltin in culture medium. The recoveries of triphenyltin and diphenyltin were in the ranges of 97.7% to 103.3% and 85.5% to 91.6%, respectively.  相似文献

A time-based sequential dispensing on-line column preconcentration procedure for mercury determination at trace levels by cold vapour generation inductively coupled plasma atomic emission spectrometry (CV-ICP-AES), by means of a unified module of a preconcentration column and a gas–liquid separator (PCGLS) is described. The complex of mercury formed on-line with ammonium pyrrolidine dithiocarbamate (APDC) is retained on the surface of the hydrophobic poly(tetrafluoroethylene) (PTFE) turnings, which are packed into the lower compartment of the PCGLS. Subsequently, mercury vapour is generated directly on the PTFE turnings by reductant SnCl2 and separated from the liquid mixture via the PCGLS by argon purge gas. The outlet of the PCGLS is connected directly to the torch adapter of the plasma without the normal spray chamber and nebulizer. With 60-s preconcentration time and 12.0 mL min–1 sample flow rate, the sampling frequency is 30 h–1. The calibration curve is linear over the concentration range 0.02–5.0 g L–1, the detection limit (cL) is 0.01 g L–1 and the relative standard deviation (sr) is 3.1% at the 1.0 g L–1 level. The proposed method was evaluated by analysis of BCR CRM 278 (Mytilus Edulis) reference material and applied to the determination of total mercury in digested urine, blood and hair samples.  相似文献

A rapid and accurate HPLC method has been developed for simultaneous determination of pioglitazone and glimepiride. Chromatographic separation of the two pharmaceuticals was performed on a Cosmosil C18 column (150 mm × 4.6 mm, 5 m) with a 45:35:20 (v/v) mixture of 0.01 m triammonium citrate (pH adjusted to 6.95 with orthophosphoric acid), acetonitrile, and methanol as mobile phase, at a flow rate of 1.0 mL min–1, and detection at 228 nm. Separation was complete in less than 10 min. The method was validated for linearity, accuracy, precision, limit of quantitation, and robustness [1, 2]. Linearity, accuracy, and precision were found to be acceptable over the ranges 2.50–30.00 g mL–1 for pioglitazone and 0.10–10.00 g mL–1 for glimepiride.  相似文献

Summary Cinnarizine is a pharmaceutical drug used in the treatment of cerebral and peripheral vascular diseases. A reversed-phase liquid chromatographic method with fluorescence detection has been developed for determination of the drug in workplace air. Air sampling in the workplace is performed on perchlorovinyl filters (FPP), the filters are extracted with methanol for 40 min, and the extract (50 L) is injected and separated on a 250 mm × 4.6 mm i.d., 5 m particle, C8 reversed-phase column with 1% ammonium acetate (pH 4.5)–acetonitrile, 1:4 (v/v), as mobile phase at a flow rate of 1 mL min–1.  相似文献

Novel, simple, rapid, highly sensitive, and direct determination of iodide and thiocyanate ions in seawater has been performed by liquid chromatography (LC) with UV detection at 220 nm. The separation was achieved on a C30 column of conventional size (150 mm × 4.6 mm i.d.) modified with poly(ethylene glycol); an aqueous solution of 300 mM sodium sulfate and 50 mM sodium chloride was used as mobile phase. Detection limits (S/N=3) obtained by injecting a 20-L sample were 0.5 and 6 ng mL–1 for iodide and thiocyanate, respectively. The method was successfully used for rapid and direct determination of iodide and thiocyanate in seawater samples, collected from the coasts of Japan, without any extra pretreatment.Dedicated to Professor K. This work describes an electroanalytical investigation of dopamine using cyclic voltammetry (CV) and the graphite–polyurethane composite electrode (GPU). In CV studies, well-defined redox peaks characterize the oxidation process at the GPU electrode, which is indicative of electrocatalytic effects associated with active sites on the GPU electrode surface. A new analytical methodology was developed using the GPU electrode and square wave voltammetry (SWV) in BR buffer solution (0.1 mol L–1; pH 7.4). Analytical curves were constructed under optimized conditions (f=60s–1, Ea=50 mV, EI=2 mV) and detection and quantification limits of 6.4×10–8 mol L–1 (12.1 g L–1) and 5.2×10–6 mol L–1 (0.9 mg L–1), respectively, were achieved. The precision of the method was checked by performing ten successive measurements for a 9.9×10–6 mol L–1 dopamine solution. For intra-assay and inter-assay precisions, the relative standard deviations were 1.9 and 2.3%, respectively. In order to evaluate the developed methodology, the determination of dopamine was performed with good sensitivity and selectivity, without the interference of ascorbic acid in synthetic cerebrospinal fluid, which indicates that the new methodology enables reliable analysis of dopamine.  相似文献

A method has been established for separation of nisoldipine and impurities, for example reactants, products of side-reactions, and photodegradation products, by HPTLC on LiChrospher Si 60 F254s plates with detection at 280 nm. The mobile phase, cyclohexane–ethyl acetate–toluene, 7.5:7.5:10 (v/v), enables acceptable resolution of nisoldipine, in large excess, and possible impurities. Regression coefficients (r 0.997), recovery (98–108%), and determination limit (0.02–0.2%) were validated and found to be satisfactory. The method is convenient for quantitative analysis and purity control of nisoldipine in its raw material and dosage forms.  相似文献

In this paper, a novel flow injection chemiluminescence (CL) determination of l-cysteine is proposed. The method is based on the CL reaction of l-cysteine and KBrO3 in acidic medium. The CL intensity was greatly enhanced in the presence of quinine. The CL intensity was linear with l-cysteine concentration in the range of 0.2–80 g L–1, and the detection limit was 0.1 g L–1 (3). A complete analysis, including sampling and injecting, could be performed in 1 min, giving a throughput of about 60 h–1. The relative standard deviation was 1.6% for 0.8 g L–1 l-cysteine (n=11). The proposed method was satisfactorily applied to the determination of cysteine in an amino acid mixture and human urine. The mechanism of the CL reaction is also discussed.  相似文献

Summary A high-performance liquid chromatographic method is discribed for the determination of 6-amino-2,2-dimethyl-1,3-dioxepan-5-ol using Spherisorb ODS II stationary phase and mobile phase 30:70 (v/v) methanol: aqueous 1-octane sulfonic acid. Detection was fluorimetric following postcolumn derivatization with o-phthaladehyde/2-mercaptoethanol. The procedure was applied to the analysis of aqueous solutions and microcrystalline suspensions in liquid paraffin, prepared for investigation of the toxicological profile. The method was validated for selectivity, linearity of detector response, repeatability, limit of detection and quantitation. The HPLC method was selective. The instrumental limit of detection was 0.5 ng per injection (0.05 g mL–1). The method detection limits were 0.5 g mL–1 aqueous solution and 5 g mL–1 liquid paraffin suspension, the quantitation limit 0.05 mg mL–1 aqueous solution and 1.0 mg mL–1 liquid paraffin. Linearity was within 0.94–47.1 g mL–1. Intra-assay accuracy accounted for 99–100% in the range 0.05–226 mg mL–1 aqueous solution, intra-assay precision for 2% (C.V.). For microcrystalline liquid paraffin suspensions with 1 and 250 mg mL–1 99 and 109% was found for intra-assay accuracy. Intra-assay precision was 5% (C.V.). Reliable results over a wide concentration range can be obtained. The procedure is considered valid for determination of the analyte in aqueous solution or microcrystalline paraffin oil suspensions.  相似文献

A selective and sensitive liquid chromatographic method was developed for the determination of zonisamide in small volumes of plasma. Zonisamide and the internal standard methyl 4-hydroxybenzoate were extracted from 0.2 mL of plasma with solid-phase extraction columns and eluted with methanol. Analysis of the extracts was performed on a Symmetry C18 column with ultra-violet spectrophotometric detection. The calibration curve was linear over the concentration range of 0.05–5 g mL–1 in plasma. Recoveries were reasonable for routine analyses; the limit of quantification was 0.05 g mL–1 with a signal-to-noise ratio of 5. This method could be useful for the pharmacokinetic study of zonisamide in a limited volume of human plasma and for therapeutic drug monitoring.  相似文献

The oxidation of the terpenes - and -pinene, limonene and 3-carene by hydroxyl radicals has been investigated in a fast-flow reactor coupled to a liquid nitrogen trap for collecting the carbonyl compounds. Identification of the products was performed via 2,4-dinitrophenylhydrazone (DNPH) derivatization of the carbonyls to form the mono- and di-DNPH derivatives, which were analysed by high-performance liquid chromatographic (HPLC)-DAD (diode array detector) and HPLC-mass spectrometry (HPLC-MS). Both electrospray ionization [ESI(–)] and atmospheric pressure chemical ionization [APCI(–)] were suitable for the detection of the DNPH derivatives of formaldehyde, acetaldehyde, myrtanal, campholene aldehyde, perillaldehyde, acetone, nopinone, trans-4-hydroxynopinone and 4-acetyl-1-methylcyclohexene. Also the mono-DNPH derivatives of the dicarbonyl compounds pinonaldehyde, endolim and caronaldehyde could be identified. The MS2 spectra generated in the ion trap of the mass spectrometer allowed us to distinguish between aldehydes and ketones on the basis of the characteristic fragment ion m/z 163 for the aldehydes. For the quantitative analysis of the mono-DNPH derivatives, ESI(–) in combination with single ion monitoring (SIM) detection showed the lowest detection limits. For the quantification of the dicarbonyl compounds, the acid-sensitive di-DNPH derivatives had to be formed by keeping the acidity in the acid-catalysed derivatization reaction at about 1.7 mM H2SO4. Detection of these dicarbonyl compounds can only be performed by APCI(–) with somewhat lesser sensitivity than by HPLC-DAD.  相似文献

The anionic chelate of iron(III)-2,2-dihydroxyazobenzene (H2L), [FeL2], formed 1 1 ion-pair with crystal violet cation (CV+), CV+ [FeL2], and was adsorbed on a surface of transparent polyvinyl chloride (PVC) film plasticized with di-n-octyl phthalate. Enrichment of the blue violet species of the ion-pair onto the transparent PVC film has enabled a highly sensitive and simple method for the determination of iron(III). The detection limits are 1 × 10–8 mol dm–3 (0.6 ppb) by spectrophotometry at 592 nm, and 4 × 10–8 mol dm–3 (2 ppb) by visual colorimetry. The method has been successfully applied to the determination of iron in water samples and human serum. No preparatory procedures for the separation of serum protein and other coexisting substances are required, since ion-pair adsorption process provides a new method to prevent interference of serum matrix.  相似文献

The present work reports for the first time a simple and rapid method for the spectrofluorimetric determination of lisinopril (LSP) in pharmaceutical formulations using sequential injection analysis (SIA). The method is based on reaction of LSP with o-phthalaldehyde (OPA) in the presence of 2-mercaptoethanol (borate buffer medium, pH=10.6). The emission of the derivative is monitored at 455 nm upon excitation at 346 nm. The various chemical and physical conditions that affected the reaction were studied. The calibration curve was linear in the range 0.3–10.0 mg L–1 LSP, at a sampling rate of 60 injections h–1. Consumption of OPA reagent was significantly reduced compared with conventional flow injection (FI) systems, because only 50 L of OPA was consumed per run. The method was found to be adequately precise (sr=2% at 5 mg L–1 LSP, n=10) and the 3 detection limit was 0.1 mg L–1. The method was successfully applied to the analysis of two pharmaceutical formulations containing LSP. The results obtained were in good agreement with those obtained by use of high-performance liquid chromatography (HPLC), because the mean relative error, er, was <1.8%.  相似文献
