Inactivation of influenza virus by solar radiation   总被引:1,自引:0,他引:1  
Influenza virus is readily transmitted by aerosols and its inactivation in the environment could play a role in limiting the spread of influenza epidemics. Ultraviolet radiation in sunlight is the primary virucidal agent in the environment but the time that influenza virus remains infectious outside its infected host remains to be established. In this study, we calculated the expected inactivation of influenza A virus by solar ultraviolet radiation in several cities of the world during different times of the year. The inactivation rates reported here indicate that influenza A virions should remain infectious after release from the host for several days during the winter "flu season" in many temperate-zone cities, with continued risk for reaerosolization and human infection. The correlation between low and high solar virucidal radiation and high and low disease prevalence, respectively, suggest that inactivation of viruses in the environment by solar UV radiation plays a role in the seasonal occurrence of influenza pandemics.  相似文献

We studied the photolysis of a fluoroethylene–fluoropropylene copolymer (FEP) film by vacuum ultraviolet (VUV) radiation from a resonance Xe lamp at a wavelength of 147 nm and air pressures of 0.05 and 2.5 Torr. The chemical changes in the FEP surface layer were investigated by Fourier-transform infrared spectroscopy with attenuated total reflection attachment and X-ray photoelectron spectroscopy. Double bonds were found to be the main product in the case of VUV treatment at 0.05 Torr, while photo-oxidation of FEP occurred predominantly by VUV treatment at 2.5 Torr under formation of the —CF2C(O)F group. This oxygen-containing group was more effectively formed in the FEP surface layer by VUV photo-oxidation than by conventional surface oxidation techniques such as treatments by plasma and corona discharge and ozone. Storage of the VUV-treated polymers in air at 50% relative humidity resulted in hydrolysis of —CF2C(O)F to the —CF2COOH group. Substantial improvement of the film wettability was noticed after VUV photo-oxidation. These findings suggest that VUV irradiation provides a high potential for surface modification of fluorinated polymers which are known to be particularly resistant against functionalization by conventional surface modification techniques such as plasma treatment. © 1998 John Wiley & Sons, Inc. Depletion of cutaneous glutathione by ultraviolet radiation   总被引:1,自引:0,他引:1  
Supplemental antioxidants may have a role in ameliorating or preventing the actinic damage that can lead to cutaneous disorders such as skin cancer, hyperpigmentation, and premature aging. Glutathione is an important endogenous antioxidant and fulfills various protective functions in the skin. Irradiation of hairless mice with short (UVB) or long (UVA) wavelength ultraviolet radiation or with UVA combined with a photosensitizing psoralen (PUVA) can deplete skin glutathione levels. Ultraviolet B irradiation causes rapid transient fluctuations in the epidermal glutathione level and the relative amount present as the oxidized form. Ultraviolet A irradiation can deplete epidermal and dermal glutathione for several hours but requires much higher doses than UVB. PUVA treatments may lead to extensive and prolonged depletions of epidermal and dermal glutathione, the severity of which is dependent on the psoralen dose and may last for several days. These transient depletions, oxidations, and sometimes rapid recoveries of cutaneous glutathione levels are compatible with a role for glutathione as an endogenous photoprotective agent in the skin. Experimental evidence supports such a role: for example severe skin edema develops in mice only after about 50% of the glutathione has been depleted by PUVA treatment. Although different mechanisms are involved in each case, glutathione depletion may contribute to the production of phototoxicity by UVB, UVA, and by PUVA. Understanding the depletion mechanisms may allow the development of strategies aimed at preventing loss of cutaneous glutathione, and at reinforcing the natural protective functions of this critical cell component.  相似文献

Exposure of the skin to UV radiation can lead to a local infiltration of neutrophils. Not much is known on whether the infiltration of neutrophils in the irradiated skin is UV source dependent. In this study we compared different UV sources (solar-simulated radiation [SSR], narrowband [NB]-UVB, broadband [BB]-UVB and UVA1) in their potency to induce neutrophil infiltration in normal human skin after exposure to two times the minimal erythema dose of UV radiation. Biopsies were collected from irradiated buttock skin 6 and 24 h after irradiation and from nonirradiated skin. The presence, distribution and amount of skin-infiltrated neutrophils were determined using immunohistochemical staining. Analysis revealed that SSR was most effective in inducing neutrophil infiltration. NB-UVB gave a neutrophil influx pattern similar to that seen with SSR but in smaller numbers. BB-UVB and UVA1 were far less potent in inducing neutrophil infiltration compared with SSR or NB-UVB. Our findings indicate that neutrophil infiltration in the UV-irradiated skin is UV source dependent. When the spectra emitted by the different UV sources were compared UVB seemed to be more effective than UVA in inducing neutrophil infiltration. Furthermore, our results suggest that longer wavelengths within the UVB range are mostly responsible for the infiltration of neutrophils in the UV-irradiated skin.  相似文献

An increase in the optical transmission in electronic spectra for metal/poly(1,10-decamethylene-acetamidine) systems after UV irradiation is observed, in contrast to polymer and metal films investigated separately where there was no such effect. The increase is explained by changes in the distribution of the electric charge induced by UV irradiation at the metal/polymer interface in a polymer film.  相似文献

Hair color changes and protein damage caused by ultraviolet radiation   总被引:12,自引:0,他引:12  
Ultraviolet and visible radiations are known to damage hair. However, quantitative data relating damage to hair type, proteins and color to the radiation wavelength are missing. We studied the effect of UV plus visible, UVA plus visible, visible mercury-vapor lamp radiation and sunlight on (blended) virgin dark-brown, blond and red hair and (one head) virgin black and curly dark-brown hair. All hair types showed a substantial increase in protein loss in water after lamp and sun irradiation. The damaging effect of UVB was about 2-5 times higher than that of UVA plus visible radiation, depending on the hair type. Significant color changes were also observed in every hair type, after lamp and sun irradiation, being more pronounced for the light colored hairs. The luminosity difference parameter was the major contributor to the hair color changes, but significant changes in the red-green and yellow-blue parameters of every hair were observed. In this case, the damaging effect is ascribable mainly to UVA radiation. No significant changes in the mechanical properties or topography were observed in any case. We discuss these results in terms of hair type and composition and melanin types.  相似文献

The effects of ultraviolet (UV) radiation on chlorophyll content and accumulation of the anti-inflammatory monoterpene-indole alkaloid brachycerine in plants and calli of Psychotria brachyceras (Rubiaceae) were investigated. In this study, we also investigated a protective role for brachycerine against stress conditions. Calli and tip cuttings incubated in nutrient media were daily supplemented with 4 or 16 h of UV. High-performance liquid chromatography analyses of methanolic extracts showed only traces of brachycerine in irradiated aseptic cultures, with no alkaloid being observed in control calli. In cuttings, a 10-fold increase in brachycerine content was seen after exposure for 16 h to UV-C, whereas a 4 h daily supplementation doubled the amount of the alkaloid in leaves. Exposure to a UV-B source also doubled the alkaloid yield. In vitro brachycerine was able to quench singlet oxygen. The data indicate a potential protective role for brachycerine against UV radiation, acting as a UV filter (absorption peaks are within the UV range) and a reactive oxygen species scavenger. In addition, UV radiation may be used to increase yields of this compound of pharmaceutical interest.  相似文献

The solid-state polymerization of acrylamide, potassium acrylate, and calcium acrylate by ultraviolet radiation have been investigated. For these polycrystalline monomers both in-source and post-irradiation reactions were studied by using various sources of different intensities at constant temperatures. It is concluded that the polymerization reactions take place primarily on the surfaces of crystals. The high molecular weight material obtained at low conversions show a rapid decrease in intrinsic viscosities, probably due to degradation. Both the rate and degree of polymerization were decreased by the presence of oxygen. The various behaviors observed for these three monomers under ultraviolet radiation were regarded as another indication of the importance of crystal structures upon the solid-state polymerization reactions.  相似文献

Cells receive signals for survival as well as for death, and the balance between the two ultimately determines the fate of the cells. UV-triggered apoptotic signaling has been well documented, whereas UV-induced survival effects have received little attention. We have reported previously that UVB irradiation prevented apoptosis, which is partly dependent on activation of the phosphatidylinositol 3-kinase (PI3-kinase)-Akt pathway (Ibuki Y. and Goto, R. [2000] Biochem. Biophys. Res. Commun. 279, 872-878). In this study, antiapoptotic effects and survival signals of UV with different wavelength ranges, UVA, UVB and UVC, were examined. NIH3T3 cells showed apoptotic cell death by detachment from the extracellular matrix under serum-free conditions, which was prevented by all wavelengths. However, the effect of UVA was less than those of UVB and UVC, as determined by metabolism of fluoresceine diacetate and the appearance of chromatin-condensed cells. Furthermore, the effects of three wavelengths of UV on the apoptotic pathway upstream of the nuclear signals were examined. Reduction of mitochondrial transmembrane potential (delta psi) and activation of caspase-9 and -3 were suppressed by all three wavelengths of UV, showing wavelength-dependent effects as mentioned previously. Shorter wavelengths showed stronger inhibitory effects on caspase-8 activity. The P13-kinase inhibitor wortmannin partially inhibited the UVB- and UVC-induced suppression of apoptosis but not the inhibitory effect of UVA. Furthermore, normal delta psi maintained by UVA was not changed in the presence of wortmannin, but those by UVB and UVC were reduced. Akt was clearly phosphorylated by all three wavelengths. The phosphorylation by UVB and UVC was completely inhibited by addition of wortmannin, but that by UVA was not, in agreement with the results of survival and of delta psi. These results suggested the existence of two different survival pathways leading to suppression of apoptosis, one for UVA that is independent of the PI3-kinase-Akt pathway and the other for UVB and UVC that is dependent on this pathway.  相似文献

The mechanism by which UV-C irradiation inactivates M13 bacteriophage was studied by analyzing the M13 genome using agarose gel electrophoresis and South-Western blotting for pyrimidine dimers. The involvement of singlet oxygen (1O2) was also investigated using azide and deuterium oxide and under deoxygenated conditions. With a decrease in M13 infectivity on irradiation, single-stranded circular genomic DNA (sc-DNA) was converted to Form I and Form II, which had an electrophoretic mobility between that of sc-DNA and linear-form DNA. However, the amount of sc-DNA remaining was not correlated with the survival of M13. The formation of cyclobutane pyrimidine dimers (CPD) and pyrimidine (6-4) pyrimidone photoproducts ((6-4)PP) increased as a function of irradiation dose. The decrease in M13 infectivity was highly correlated with the increase in CPD and (6-4)PP, whereas no change was seen in M13 coat protein on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. 8-Oxo-7,8-dihydro-2'-deoxyguanosine did not form in the M13 genome after UV-C irradiation. Inactivation of M13 was neither enhanced by deuterium oxide nor inhibited by azide. Deoxygenation of the M13 suspension did not affect the inactivation, indicating that 1O2 did not participate in the inactivation of M13 by UV-C irradiation under these conditions. These results indicated that UV-C irradiation induced not only CPD and (6-4)PP formation but also additional tertiary structural change in DNA inside the M13 virions, resulting in primary damage and a loss of infectivity. The indirect effect of UV-C irradiation such as 1O2 production followed by oxidative damage to nucleic acids and proteins might have contributed less, if at all, to the inactivation of M13 than the direct effect of UV-C.  相似文献

Exposure to ultraviolet (UV) radiation may induce erythema, DNA damage and suppression of immune responses. Melanin pigmentation offers protection against the first two of these effects, but immunosuppression seems to occur irrespective of the subject's pigmentation. Cis-urocanic acid (cis-UCA), produced by isomerization of trans-UCA in the stratum corneum on UV exposure, initiates some of the immunomodulatory effects of UV radiation. In the present study the relationship between skin pigmentation and UCA isomerization has been examined in 28 healthy individuals of skin types I-IV. Pigmentation is measured in five areas of not recently exposed back skin before irradiation with 0, 0.45, 0.9, 1.8 and 3.6 standard erythema dose (SED) of filtered broadband UV-B (1 SED = 10 mJ cm-2 at 298 nm). The concentration of UCA isomers is measured immediately after the irradiation. With 3.6 SED, the relative production of cis-UCA is close to the maximum obtainable, irrespective of skin type. A significant negative correlation is found between pigmentation and relative production of cis-UCA at 0.45 and 1.8 SED, and between pigmentation and absolute production of cis-UCA at 0.45 SED. At doses of 0.45 and 0.9 SED the relative and absolute production of cis-UCA are higher in the group with skin types I and II when compared with the group with skin types III and IV. The higher isomerization in the lightly pigmented subjects than in the more pigmented ones may indicate that people with fair skin are at a relatively higher risk of immunosuppression when exposed to low doses of UV radiation.  相似文献

The effects of ultraviolet radiation on dilute aqueous solutions of poly(acrylic acid) and of other polyelectrolytes were studied by viscosity measurements in connection with the effects of ionizing radiation. It was found that ultraviolet light of wavelength below about 2300 Å brought about degradation of polymer chains mainly by indirect action via water, while light of wavelength above 2300 Å caused degradation by direct action in some polymers. It was deduced from the experiments that the protective effect of NaCl could be largely attributed to a decrease in the indirect action. It was also found that a low concentration of methanol was effective in preventing degradation by direct action, although methanol promoted degradation when present in high concentration. Since the promotive effect was not observed when light of wavelength below 3700 Å was eliminated by a filter, this effect was attributed to active products of the irradiation of methanol.  相似文献

The photoionization and photodissociation of L-valine are studied by tunable synchrotron vacuum ultraviolet photoionization mass spectrometry at the photon energy of 13 eV. The ionization energy of L-valine and the appearance energies of major fragments are measured by the photoionization efficiency spectrum in the photon energy range of 8-11 eV. Possible formation pathways of the major fragments, NH(2)CHC(OH)(2)(+) (m/z=75), NH(2)(CH(3))(2)(CH)(2)(+) (m/z=72) and NH(2)CHCO(+) (m/z=57), are discussed in detail with the theoretical calculations at the B3LYP/6-31++G (d, p) level. Hydrogen migration is considered as the key way for the formation of NH(2)CHC(OH)(2)(+) (m/z=75) and NH(2)CHCO(+) (m/z=57). Furthermore, other fragments, NH(2)CHCOOH(+) (m/z=74), (CH(3))(2)(CH)(2)(+) (m/z=56), C(4)H(7)(+) (m/z=55), NH(2)CHOH(+) (m/z=46), NH(2)CH(2)(+) (m/z=30) and m/z=18, species are also briefly described.  相似文献

Abstract— Measurements of epidermal hyperplasia induced by UV in albino mice, using an improved method of sampling and counting of cells, show wide variability attributable largely to basic uncertainty rather than experimental error. Factors are analyzed that may contribute to the infeasibility of determining simultaneously the size of cell population and rate of cell duplication, including recovery of cell activity enhanced by photoreactivation. A hyperplasia-inducing factor is isolated from epidermis exposed to UV.  相似文献

Ultraviolet (UV) radiation from sunlight causes skin cancer and inhibits priming of the immune system during vaccination. However the dose related effects of the different components of sunlight (UVA and UVB) are complex and require further investigation. Using ovalbumin as a model protein vaccine with saponin as adjuvant we show that both UVA and UVB can suppress the DTH response to a poorly immunogenic protein. Increasing doses of UVB induced increased levels of immunosuppression and tolerance. UVA however, caused a bi-phasic dose response with intermediate but not low or high doses causing primary immunosuppression. No dose of UVA caused significant tolerance. Similar results were observed in both C57BL/6 and Balb/c mice. Our data confirms the complex immunomodulatory dose effects of UVA and UVB for a protein antigen, and shows that both UVB and UVA can suppress immunity induced by a protein with adjuvant. This highlights the importance of considering sun exposure patterns in the future success of both preventing skin cancer development and enhancing vaccination regimes.  相似文献

Measurements of solar ultraviolet radiation (<320 nm) have been made with polysulphone film at a temperate and a tropical site and compared mean monthly values of the fraction of ultraviolet in solar radiation are calculated which allow an estimate of ultraviolet dose to be made from a knowledge of total solar radiation.While the data demonstrate the marked effect of sun elevation on the ultraviolet fraction of solar radiation they also indicate the effect of seasonal variation in the thickness of the ozone layer above the temperate site.It is also shown that whereas sunlight makes by far the major contribution to total solar radiation, skylight is of prime importance as regards ultraviolet radiation.  相似文献
