A new preparation scheme is proposed to fractionate neutral lipids (acylglycerines, sterol esters, long-chain free fatty acids) from polar phospholipids in biological matrices. This fractionation is mandatory in the microbial community, for the control of bioremediation processes, in the study of phytoplankton growth in lakes and rivers, and in the quality control of processed food. Basically, a two-step pressurized liquid extraction (PLE) scheme is combined with an "in-cell-fractionation" using silica-based sorbents placed at the outlet of the PLE cartridge. The optimized extraction scheme consists of n-hexane/acetone (9:1, v/v) extraction at 50 degrees C (2 cycles, 10 min each) to obtain neutral lipids followed by chloroform/methanol (1:4, v/v) extraction at 110 degrees C (2 cycles, 10 min each). Thermally pre-treated silicic acid and cyanoproyl-modified silica turned out to be appropriate sorbents to ensure clear-cut boundaries between neutral lipids and phospholipids. The proposed protocol is superior to commonly used approaches consisting of an exhaustive lipid extraction followed by off-line lipid fractionation using solid-phase extraction (SPE) regarding fractionation efficiency, time and solvent consumption. In this paper, it is also shown that the transmethylation using trimethylchlorosilane/methanol (1:9, v/v) provides a complete reaction to give methyl esters without artefact formation across the array of different lipid classes even with polyunsaturated fatty acid moieties.  相似文献

Algae can synthesize, accumulate and store large amounts of lipids in its cells, which holds immense potential as a renewable source of biodiesel. In this work, we have developed and validated a GC–MS method for quantitation of fatty acids and glycerolipids in forms of fatty acid methyl esters derived from algae biomass. Algae Scenedesmus dimorphus dry mass was pulverized by mortar and pestle, then extracted by the modified Folch method and fractionated into free fatty acids and glycerolipids on aminopropyl solid‐phase extraction cartridges. Fatty acid methyl esters were produced by an optimized one‐step esterification of fatty acids and transesterification of glycerolipids with boron trichloride/methanol. The matrix effect, recoveries and stability of fatty acids and glycerolipids in algal matrix were first evaluated by spiking stable isotopes of pentadecanoic‐2,2‐d2 acid and glyceryl tri(hexadecanoate‐2,2‐d2) as surrogate analytes and tridecanoic‐2,2‐d2 acid as internal standard into algal matrix prior to sample extraction. Later, the method was validated in terms of lower limits of quantitation, linear calibration ranges, intra‐ and inter‐assay precision and accuracy using tridecanoic‐2,2‐d2 acid as internal standard. The method developed has been applied to the quantitation of fatty acid methyl esters from free fatty acid and glycerolipid fractions of algae Scenedesmus dimorphus .  相似文献

Summary Polyunsaturated fatty acids have been analysed as methyl esters by liquid chromatography on porous graphitic carbon and the results compared with those obtained on octadecyl bonded phases. Chromatographic behaviour on octadecyl bonded phases arises principally as a result of hydrophobic interactions with the bonded phase. Because the retention of analytes is greater on porous graphitic carbon than on octadecyl phases, organic mobile phases are required. When the number of double bonds is low (ca 1–3), the behaviour of porous graphitic carbon is similar to that of octadecyl bonded phases, but when this number increases stronger interactions with the flat surface of the graphite appear, resulting in new selectivity. These two ‘reversed-phase’ systems are considered complementary for separation of different fatty acid methyl esters. An additional advantage of porous graphitic carbon is that it enables isolation of hexadecartrienoic and hexadecadienoic acids, which are not available commercially.  相似文献

The fatty acids of three lipid classes (free fatty acids, triglycerides, and cholesteryl esters) from dog heart were analysed by gas chromatography. Samples of the left ventricle were homogenized and total lipids were extracted. After separation by thin-layer chromatography, the bands of the lipid classes studied were scraped off, transmethylated according to the boron trifluoride-methanol procedure, and the fatty acid methyl esters were extracted and analysed. The problems related to the quantitation of fatty acids were investigated, namely transmethylation procedure, thin-layer chromatography, and gas chromatographic conditions. Fatty acid methyl esters were separated on capillary columns coated in the laboratory with SP 2340 stationary phase. The high performance of the separation ensured the reliability and the precision of the analysis.  相似文献

An aminopropyl-based solid-phase extraction technique was used for isolation of chlorinated fatty acids in lipids. A range of different chlorinated fatty acids was eluted in a small volume of solvent (4 ml) and the recoveries of the different species and isomers were quantitative. Only 1% of the vastly dominating unchlorinated fatty acid methyl esters were recovered in the fractions containing the chlorinated fatty acid methyl esters. This method makes it possible to isolate and detect > or = 1 microg of a chlorinated fatty acid methyl ester in 1 g of lipid.  相似文献

A modification of a procedure by Hage [1] is proposed for the gas chromatographic evaluation of the content of free medium-chain fatty acids and related ethyl esters in beer. The method involves extraction of free fatty acids and ethyl esters by SPE using C18 bonded phase columns, derivatization of free fatty acids and related ethyl esters with diazomethane, and GC analysis using an SP-2340 capillary column. The results obtained have shown the method to be rapid and highly reproducible. The technique has been compared with other methods used for determination of free fatty acids.  相似文献

A GC/MS procedure has been developed, optimized, and applied to characterization of oil binders in paintings. The procedure involves hydrolysis of lipids to fatty acids (FAs) and derivatization of FAs to fatty acid methyl esters (FAMEs) by a solution of sodium methanolate in methanol at an elevated temperature. FAMEs are analyzed by temperature-programed GC followed by full-scan MS. Old and dried samples are subjected to extraction of nonpolymerized FAMEs into dichloromethane prior to hydrolysis. The method provides a good repeatability of results and has been applied to the characterization of common plant oils used in paintings, to commercial oil and tempera paints, to model painting samples, and to samples taken from real paintings. The fresh oils and binders can readily be identified and characterized. The ratio of the methyl esters of palmitic and stearic acids can be used to characterize oil binders in old works of art.  相似文献

The separation and analysis of furan fatty acids and other minor component fatty acids present at very low concentrations in complex sample matrices, such as fish oil or lipids derived from liver and testes, require several pre-analytical separation steps if single column gas chromatography is to furnish sufficient resolution: after extraction and transesterification hydrogenation, urea complex precipitation and argentation TLC have been applied prior to GC analysis of furan fatty acids. By using multidimensional GC-MSD with cooled injection and flow-controlled column switching with intermediate cold trapping, it has been possible to identify directly the methyl esters of furan fatty acids without further pre-analytical separation. The most common of the furan fatty acids can be subdivided into two groups depending on whether they bear a propyl or pentyl side group in the 5-position of the furan ring. In addition to the eight furan fatty acids known to be present in fish oil, six new ones were identified, four with propyl substitution and two with pentyl substitution. Four have earlier been reported to be present in the hepatopancreas of crayfish and in fish tissue, whereas the propyl-substituted 16,19-epoxy-17,18-dimethyldocosa-16,18-dienoic acid and the pentyl-substituted furan fatty acid 6,9-epoxy-7-methyltetradeca-6,8-dienoic acid were hitherto unknown.  相似文献

Microalgae are unique photosynthetic organisms that are known to accumulate storage lipids in large quantitites and thrive in saline waters. Before these storage lipids can be used, they must be extracted from the microalgae and converted into usable fuel. Transesterification of lipids produces fatty acid methyl esters that can be used as a diesel fuel substitute. Three solvents, 1-butanol, ethanol, and hexane/2-propanol, were evaluated for extraction efficiency of microalgal lipids. Type of catalyst, concentration of catalyst, time of reaction, temperature of reaction, and quality of lipid were examined as variables for transesterification. The most efficient solvent of the three for extraction was 1-butanol (90% efficiency), followed by hexane/2-propanol and ethanol. Optimal yield of fatty acid methyl esters was obtained using 0.6N hydrochloric acid in methanol for 0.1 h at 70°C.  相似文献

Two methods for the separation of cholesterol esters, based on the number of double bonds in their fatty acid moieties, are presented. Silver ion chromatography, usually performed on thin-layer chromatographic plates, was made suitable for high-performance liquid chromatography (HPLC) and solid-phase extraction. Separation on a bonded sulphonic acid phase loaded with silver ions was achieved with cholesterol esters containing up to six double bonds in their fatty acid moieties. No cross-contamination between fractions with different numbers of double bonds was detected with the HPLC method, was demonstrated by subsequent gas chromatographic analysis of the fatty acid moieties, following transmethylation. For adequate separations with the solid-phase extraction columns it proved important to avoid overloading. The methods may be of use for the off-line analyses of the sterol compositions of the isolated fractions, which each contain sterol esters with an equal number of double bonds in their fatty acid moieties.  相似文献

A new approach using sequential pressurized liquid extraction described recently [J. Poerschmann, R. Carlson, J. Chromatogr. A, 1127 (2006) 18-25] was applied to determine lipid markers originating from central nervous system (CNS) tissue of cows in heat-processed sausages. These studies are very important in quality control as well as risk assessment studies in the face of the bovine spongiform encephalopathy (BSE) crisis. Diagnostic CNS lipid markers, which should not be present in meat products without CNS addition, were recognized on complete transesterification as polar 2-hydroxy-fatty acids (2OH-24:0, 2OH-24:1, 2OH-22:0, 2OH-18:0, shorthand designation) as well as odd-numbered non-branched fatty acids beyond C(22). An array of other fatty acids including lignoceric acid (24:0), nervonic acid (24:1), arachidonic acid (20:4), and polyunsaturated nC(22)-surrogates are strongly related to CNS lipids, but occur as traces in meat products without CNS addition as well, thus reducing their value as diagnostic markers. Samples including meat products without CNS addition, meat with 3% CNS addition, as well as pure CNS homogenates, were subjected to sequential PLE (pressurized liquid extraction) consisting of two steps: n-hexane/acetone 9:1 (v/v) extraction at 50 degrees C to remove neutral lipids, followed by chloroform/methanol 1:4 (v/v) extraction at 110 degrees C to isolate polar CNS lipids (two 10 min PLE cycles each). To enhance the fractionation efficiency, cyanopropyl modified silica as well as chemically not modified silica sorbent was used at the outlet of the PLE cartridge to retard polar lipids in the first extraction step. This method proved superior to widely distributed exhaustive lipid extraction followed by solid-phase extraction (SPE) using silica regarding lipid recoveries and clear-cut boundaries between lipid classes. Methodological studies showed that the alcoholysis using trimethylchlorosilane/methanol (1:9, v/v) is an excellent method for the complete transesterification of lipids and quantitative formation of methyl esters.  相似文献

建立了孔石莼脂肪酸的气相色谱/质谱(GC/MS)测定方法。使用Folch法提取了孔石莼中的总脂,经过2 mol/L HCl-甲醇溶液的甲酯化处理后,采用GC/MS法对其脂肪酸组成进行了分离分析,同时结合有机质谱学规律,分别对饱和脂肪酸甲酯、单不饱和脂肪酸甲酯和多不饱和脂肪酸甲酯的裂解规律和质谱特征进行了分析归纳。通过质谱数据库检索和标准品对照,鉴定出孔石莼中的24种脂肪酸,其中9,12,15-十八碳三烯酸、4,7,10,13-十六碳四烯酸和6,9,2,15-十八碳四烯酸3种主要多不饱和脂肪酸占总脂肪酸含量的45.14%。通过对孔石莼中脂肪酸的分析,表明特征离子在脂肪酸甲酯尤其是多不饱和脂肪酸甲酯的定性方面具有很好的应用价值。  相似文献

Wuliangye baijiu is one of the most famous baijiu in China, with a rich, harmonic aroma profile highly appreciated by consumers. Thousands of volatiles have been identified for the unique aroma profile. Among them, fatty acid esters have been identified as the main contributors to the aroma profile. In addition, many non-ester minor compounds, many of which are more polar than the esters, have been identified to contribute to the characteristic aroma unique to Wuliangye baijiu. The analysis of these minor compounds has been challenging due to the dominance of esters in the sample. Thus, it is desirable to fractionate the aroma extract into subgroups based on functional group or polarity to simplify the analysis. This study attempts a new approach to achieve simultaneous volatile extraction and fractionation using tandem LiChrolut EN and silica gel solid-phase extraction (SPE) columns. A baijiu sample (10 mL, diluted in 40 mL of water) was first passed through the LiChrolut EN (1.0 g) column. The loaded LiChrolut EN column was then dried with air and coupled with a silica gel (5.0 g) SPE column with anhydrous Na2SO4 (10.0 g) in between. The volatile compounds were eluted from the LiChrolut EN column and simultaneously fractionated on the silica gel column based on polarity. The simultaneous extraction and fractionation technique enabled the fractionations of all fatty acid esters into less polar fractions. Fatty acids, alcohols, pyrazines, furans, phenols, hydroxy esters, and other polar compounds were collected in more polar fractions. This technique was used to study the volatile compounds in Wuliangye, Moutai, and Fengjiu baijiu. In addition to fatty acid esters, many minor polar compounds, including 2,6-dimethylpyrazine, 2-ethyl-6-methylpyrazine, 2-ethyl-3,5-dimethylpyrazine, p-cresol, and 2-acetylpyrrole, were unequivocally identified in the samples. The procedure is fast and straightforward, with low solvent consumption.  相似文献

An improved extraction and clean-up method for determination of brain-specific fatty acids, in particular lignoceric acid (C24:0) and the cis/ trans isomers of nervonic acid (15 c-t C24:1), in meat products has been developed. The method is based on isolation of the polar lipids of interest from the bulk lipids by solid-phase extraction. The fatty acids, derivatised to their fatty acid methyl esters, are quantified by GC in a DB5 column. Fresh meat samples were extracted by using a mixture of n-butanol:hexane (1:9) as solvent. The extract was loaded in a silica gel cartridge column previously equilibrated with hexane. The first fraction containing the major part of the fat was eluted with hexane while acetone and methanol allowed the elution of fatty acids bound to polar moieties such as nervonic and lignoceric acids. This second fraction containing the analyte was methylated and injected into the GC for quantification after addition octacosane (C(28)) as internal standard.  相似文献

The determination of the absolute stereochemistry of hydroxy fatty acid methyl esters as their (S)-ibuprofen esters is possible via standard gas chromatographic techniques. Analyses of various racemic and nonracemic standards and mixtures from enzymic oxidation show excellent resolution of the resultant diastereomers, with the (S,S)-diastereomers eluting first in all cases studied. The stereochemistry of the oxidation of dodecanoic acid by P450BM3, which has not been previously reported, was determined by this method and indicated a preference for (R)-hydroxylation. The sensitivity of this technique allows the analysis of very small quantities of product, which has revealed that the oxidation of dodecanoic and hexadecanoic acids by the T268A mutant of P450BM3 display the same stereochemical efficiency and produce (R)-hydroxy fatty acids in the same manner as wildtype P450BM3, despite the poor coupling efficiency of these substrates. This stereochemistry implies that hydroxylation catalysed by the T268A mutant of P450BM3 occurs through residual levels of the normal hydroxylating species.  相似文献

Fast-atom-bombardment mass spectrometry (FABMS) is used for the semiquantitative determination of the fatty acids of complex lipids directly from intact algal cells, crude algal lipid extracts, and vegetable oils. Carboxylate ions, RCOO?, corresponding to the fatty acid moieties of the complex lipids are detected. The relative abundances of the carboxylate fatty acid ions in the FAB mass spectra agreed with the relative percentages found by gas chromatography of the fatty acid methyl esters derived from the extracted lipids. Chemical ionization/fast-atom-bombardment mass spectrometry (CI/FABMS) is discussed with respect to enhancing the molecular ions of the fatty acids and triacylglycerols from these materials. The use of FABMS requires little sample preparation, and FABMS enables rapid fatty acid screening, directly from crude biological materials.  相似文献

A chemical characterization of major lipid components, namely, triacylglycerols, fatty acids and the unsaponifiable fraction, in a Quinoa seed lipids sample is reported. To tackle such a task, non‐aqueous reversed‐phase high‐performance liquid chromatography with mass spectrometry detection was employed. The latter was interfaced with atmospheric pressure chemical ionization for the analysis of triacylglycerols. The main triacylglycerols (>10%) were represented by OLP, OOL and OLL (P = palmitoyl, O = oleoyl, L = linoleoyl); the latter was present in the oil sample at the highest percentage (18.1%). Furthermore, fatty acid methyl esters were evaluated by gas chromatography with flame ionization detection. 89% of the total fatty acids was represented by unsaturated fatty acid methyl esters with the greatest percentage represented by linoleic and oleic acids accounting for approximately 48 and 28%, respectively. An extensive characterization of the unsaponifiable fraction of Quinoa seed lipids was performed for the first time, by using comprehensive two‐dimensional gas chromatography with dual mass spectrometry/flame ionization detection. Overall, 66 compounds of the unsaponifiable fraction were tentatively identified, many constituents of which (particularly sterols) were confirmed by using gas chromatography with high‐resolution time‐of‐flight mass spectrometry.  相似文献

建立了采用超高效液相色谱(UPLC)-蒸发光散射检测器(ELSD)测定生物柴油中11种常见的脂肪酸及脂肪酸甲酯含量的方法。这11种常见的脂肪酸及脂肪酸甲酯为豆蔻酸、亚油酸、棕榈酸、油酸、亚麻酸甲酯、硬脂酸、亚油酸甲酯、棕榈酸甲酯、油酸甲酯、芥酸和硬脂酸甲酯。样品经提取后用甲醇溶解,采用Acquity UPLC BEH Phenyl C18柱(100 mm×2.1 mm,1.7 μm)分离,乙腈-水(体积比为3∶1)混合液为流动相进行等度洗脱,采用的ELSD条件为增益80,漂移管温度为45 ℃,载气压力为172 kPa,雾化器为冷却模式,并用外标法进行定量分析。结果表明,在一定的质量浓度范围内,峰面积的对数和质量浓度的对数线性关系良好。与其他检测生物柴油成分的方法相比,该方法简单,分离效果好,速度快,特别是此方法可以同时实现脂肪酸及脂肪酸甲酯的分离,并进行定量分析,能有效测定反应的进行程度,从而满足生物柴油工艺研究的需要。  相似文献

A new halogen-specific detection method (XSD) was tested for determination of chlorinated fatty acids in marine biota. In XSD, an increased emission of ions and electrons is caused by the high-temperature combustion of halogen-containing compounds. The detection limit of methyl dichlorooctadecanoate and the selectivity at a reactor temperature of 900 degrees C match those of electrolytic conductivity detection (ELCD). The relative standard deviation is less than 11% for > or =0.2 ng methyl dichlorooctadecanoate. An XSD chromatogram of a complex sample, chlorinated fatty acid methyl esters liberated from fish lipids, agreed with a previously obtained ELCD chromatogram.  相似文献

Abstract

The method of hydrogen enthalpimetry as previously applied to unsaturated and polyunsaturated lipids has employed an authentic sample of each lipid analyte for construction of the enthalpimetric calibration curve. This study shows that individual lipid standards are not necessary for several common naturally-occurring fatty acids, methyl esters and simple triglycerides, each of which can be expressed as equivalent mg of methyl oleate. Data are given for six lipids having from one to nine moles of double bonds per mole of analyte, i. e., from oleic acid to trilinolenin.  相似文献
