The chemical composition of the needles of P. pinea, P. pinaster, P. halepensis, P. nigra, P. brutia, P. patula, P. radiata, P. taeda, P. elliotti, P. kesiya, P. sylvestris and P. eldarica was investigated. Headspace solid-phase microextraction and steam distillation extraction were used to collect the volatile fractions. Samples were analyzed using one-dimensional gas chromatography (1D-GC) and comprehensive two-dimensional gas chromatography (GC × GC) associated with a quadrupole and a time-of-flight mass detectors. Results showed that the analytical capabilities of 1D-GC are partially limited by the separation power of the columns. The higher sensibility and the absence of peak skewing of the time-of-flight mass analyzer, with the use of automated peak finding and deconvolution algorithms, allowed for the detection of trace components with qualitative full spectra and the extraction of true mass spectra from coeluting compounds, promoting their reliable identification and thus significantly improving results obtained by 1D-GC/MS, when using a quadrupole mass analyzer. The use of GC × GC resulted in enhanced separation efficiency and increased signal to noise ratio (sensitivity) of the analytes, maximizing mass spectra quality and improving compound detection and identification. This work shows the use of 1D-GC/ToFMS for the analysis of pine needles volatiles, achieving the detection of 177 compounds, that is more than twice the number previously identified by standard 1D-GC/MS. The analysis by GC × GC for the same sample allowed the detection of 212 compounds. The enantioselective GC × GC analysis performed for all the Pinus spp. under study achieved the detection of 422 different compounds. Cross-over phenomena according to operational conditions are highlighted and discussed.  相似文献

Triacylglycerols (TAGs) are the main constituents of vegetable oils where they occur in complex mixtures with characteristic distributions. Mass spectrometry using an atmospheric pressure chemical ionization interface (APCI-MS) run in positive mode and an Ion Trap mass analyser were applied in the study of olive and soybean oils and their mixtures. Direct injections of soybean and olive oil solutions allowed the identification of ions derived from the main TAGs of both oils. This procedure showed to be a simple and powerful tool to evaluate mixtures or addition of soybean to olive oil. TAG separation was optimized by high performance liquid chromatography (HPLC) using an octadecylsilica LiChrospher column (250 mm × 3 mm; 5 μm) and a gradient composed of acetonitrile and 2-propanol allowed the separation of the main TAGs of the studied oils. APCI vaporization temperature was optimized and best signals were obtained at 370 °C. Multiple reaction monitoring (MRM) employing the transition of the protonated TAG molecules ([M+H]+) to the protonated diacylglycerol fragments ([M+H−R]+) improved the selectivity of TAG detection and was used in quantitative studies. Different strategies were developed to evaluate oil composition following TAG analysis by MRM. The external standard calibration and standard additions methods were compared for triolein quantification but the former showed to be biased. Further quantitative studies were based on the estimates of soybean and olive oil proportions in mixtures by comparison of TAG areas found in mixtures of known and unknown composition of both oils. Good agreement with expected or labeled values was found for a commercial blend containing 15% (w/w) of olive oil in soybean oil and to a 1:1 mixture of both oils, showing the potential of this method in characterizing oil mixtures and estimating oil proportions. Olive oils of different origins were also evaluated by mass spectra data obtained after direct injections of oil solutions and principal component analysis (PCA). Argentinean olive oils were clustered in a different area of the principal components plot (PC2 × PC1) in comparison with European olive oils. The commercial blend containing 15% (w/w) of olive oil in soybean oil appeared in a completely different area of the graphic, showing the potential of this method to screen out for olive oil adulterations.  相似文献

An effective extraction method was devised for the determination of organophosphorus pesticides (OPPs) in olive and avocado oil samples, using atmospheric pressure microwave-assisted liquid–liquid extraction (APMAE) and solid-phase extraction or low-temperature precipitation as clean-up step. A simple glass system equipped with an air-cooled condenser was designed as an extraction vessel. The pesticides were partitioned between acetonitrile and oil solution in hexane. Analytical determinations were carried out by gas chromatography-flame photometric detection and gas chromatography–tandem mass spectrometry, using a triple quadrupole mass analyzer, for confirmation purposes. Several factors influencing the extraction efficiency were investigated and optimized through fractional factorial design and Doehlert design. Under optimal conditions the recovery of pesticides from oil at 0.025 μg g−1 ranged from 71% to 103%, except for fenthion in avocado oil, with RSDs ≤13% (n = 5). The LOQ for the entire method ranged from 0.004 to 0.015 μg g−1. Finally, the proposed method was successfully applied to the extraction and determination of the selected pesticides in 20 commercially packed extra virgin olive oils and four commercially packed avocado oils produced in Chile. Detectable residues of different OPPs were observed in 85% of samples.  相似文献

Discrimination between virgin olive oils and pure olive oils is of primary importance for controlling adulterations. Here, we show the potential usefulness of two multiway methods, unfold principal component analysis (U-PCA) and parallel factor analysis (PARAFAC), for the exploratory analysis of the two types of oils. We applied both methods to the excitation-emission fluorescence matrices (EEM) of olive oils and then compared the results with the ones obtained by multivariate principal component analysis (PCA) based on a fluorescence spectrum recorded at only one excitation wavelength. For U-PCA and PARAFAC, the ranges studied were λex=300-400 nm, λem=400-695 nm and λex=300-400 nm, λem=400-600 nm. The first range contained chlorophylls, whose peak was much more intense than those of the rest of species. The second range did not contain the chlorophylls peak but only the fluorescence spectra of the remaining compounds (oxidation products and Vitamin E). The three-component PARAFAC model on the second range was found to be the most interpretable. With this model, we could distinguish well between the two groups of oils and we could find the underlying fluorescent spectra of three families of compounds.  相似文献

A specific adsorbent for extraction of methidathion from olive oil was developed. The design of the molecularly imprinted polymer (MIP) was based on the results of the computational screening of the library of polymerisable functional monomers. MIP was prepared by thermal polymerisation using N,N’-methylene bisacrylamide (MBAA) as a functional monomer and ethylene glycol dimethacrylate (EGDMA) as a cross-linker. The polymers based on the itaconic acid (IA), methacrylic acid (MAA) and 2-(trifluoromethyl)acryl acid (TFMAA) functional monomers and one control polymer which was made without functional monomers with cross-linker EGDMA were also synthesised and tested. The performance of each polymer was compared using corresponding imprinting factor. As it was predicted by molecular modelling the best results were obtained for the MIP prepared with MBAA. The obtained MIP was optimised in solid-phase extraction coupled with high performance liquid chromatography (MISPE-HPLC-UV) and tested for the rapid screening of methidathion in olive oil. The proposed method allowed the efficient extraction of methidathion for concentrations ranging from 0.1 to 9 mg L−1 (r2 = 0.996). The limits of detection (LOD) and quantification (LOQ) in olive oil were 0.02 mg L−1 and 0.1 mg L−1, respectively. MIPs extraction was much more effective than traditional C18 reverse-phase solid phase extraction.  相似文献

Phenolic compounds in Sicilian olive oils were analyzed by GC-MS and GC-MS/MS after extraction with methanol:water 80:20 and derivatization with bis(trimethylsilyl)trifluoracetamide and trimethylchlorosilane (BSTFA:TMCS 99:1). Numerous compounds were detected and 23 were identified. Tyrosol, hydroxytyrosol and the decarbomethoxy ligstroside and oleuropein aglycons in the dialdehydic forms were the most abundant compounds. 4-(Acetoxyethyl)-1-hydroxybenzene, 3,4-dihydroxyphenylacetaldehyde, syringaldehyde and the cis form of ferulic acid were identified: these compounds were not found in olive oils before. The presence of the dialdehydic form of elenolic acid (without carbomethoxy group) linked to 3-methoxy-4-hydroxyphenylethanol was hypothesized. There were quantitative differences in oils from the varieties Nocellara del Belice, Santagatese and Cerasuola; these differences could depend on the olive varieties and ripeness.  相似文献

A method for accelerating the determination of the oxidative stability of virgin olive oil by use of ultrasound energy is proposed. The most influential variables on the acceleration of the process were those characteristic of the ultrasound probe, namely irradiation time, duty cycle and pulse amplitude, and they were exhaustively optimized. The ultrasound device, a cylindrical titanium alloy microprobe (3 mm diameter), was immersed into the olive oil sample contained in a 5 cm long test tube where the ultrasound waves were applied directly. The oxidation process was monitored at 270 nm. Twelve samples of virgin olive oil with known oxidative stability, ranging between 19 and 129 h, calculated by the Rancimat method, were analyzed and the results obtained showed an excellent correlation (r=0.9961) with those provided by the Rancimat method. The proposed method drastically decreased the time required for the determination of this parameter, e.g. a virgin olive oil with oxidative stability 129 h, as calculated by the Rancimat method, required only 50.5 min, thus decreasing the determination time 110 times. The method was applied to the determination of the stability of five extra virgin olive oils from different varieties of olive seeds. The values obtained were interpolated in the correlation plot for the calculation of the values corresponding to the Rancimat method. They coincided with those calculated previously with this method, thus demonstrating the validity of the proposed method.  相似文献

In the present article, a headspace solid-phase microextraction method coupled to GC/MS was developed and applied for the simultaneous determination of mono- and sesquiterpenic hydrocarbons in virgin olive oils of different olive variety and geographical origin. Analysis of various oils resulted in the simultaneous detection of 15 monoterpenes and 30 sesquiterpenes. Some of these hydrocarbons were previously reported to be constituents of virgin olive oil terpenoid fraction, although we also detected some terpenic hydrocarbons that have not previously been documented as present in virgin olive oil. Significant differences were detected in the proportion of terpenic compounds in oils obtained from different olive varieties grown in different geographical areas. The monoterpene, and particularly the sesquiterpene composition of olive oil may be used to distinguish samples from different cultivar and geographical areas.  相似文献

In this work, a straightforward, reliable and effective automated method has been developed for the direct determination of monoaromatic volatile BTEXS group (namely benzene, toluene, ethylbenzene, o-, m- and p-xylenes, and styrene) in olives and olive oil, based on headspace technique. Separation, identification and quantitation were carried out by headspace-gas chromatography-mass spectrometry (HS-GC-MS) in selected ion monitoring (SIM) mode. Sample pretreatment or clean-up were not necessary (besides olives milling) because the olives and olive oil samples are put directly into an HS vial, automatically processed by HS and then injected in the GC-MS for chromatographic analysis. The chemical and instrumental variables were optimized using spiked olives and olive oil samples at 50 μg kg−1 of each targeted species. The method was validated to ensure the quality of the results. The precision was satisfactory with relative standard deviations (RSD (%)) in the range 1.6-5.2% and 10.3-14.2% for olive oil and olives, respectively. Limits of detection were in the range 0.1-7.4 and 0.4-4.4 μg kg−1 for olive oil and olives, respectively. Finally, the proposed method was applied to the analysis of real olives and olive oil samples, finding positives of the studied compounds, with overall BTEXS concentration levels in the range 23-332 μg kg−1 and 4.2-87 μg kg−1 for olive oil and olives, respectively.  相似文献

In this work the applicability of nano-liquid chromatography (nano-LC) was evaluated for the determination of phytosterols in extra-virgin olive oil samples. These compounds represent a minor part of lipids in vegetable oils, but their quantification can be useful to establish oil origin and to reveal intentional adulterations. The analysis of five main sterols, specifically brassicasterol, stigmasterol, campesterol, cholesterol and β-sitosterol, was performed in a laboratory-assembled nano-LC system coupled with a UV detector. The separation of all compounds was obtained in about 20 min, employing a capillary column packed with a C18-RP (sub-2 μm particles) stationary phase for 15 cm. Methanol only was used as mobile phase. The simple method developed and optimized was validated in terms of repeatability, linearity, limit of detection and limit of quantification (0.78 and 1.56 μg/mL, respectively) achieving good results. After this, it was applied to the determination of phytosterols in extra-virgin olive oil samples. Isolation of phytosterols was obtained by solid-phase extraction, after saponification and liquid–liquid extraction of the unsaponified fraction with diethyl ether. Recovery tests were performed and values between 90% and 103%, with RSDs within 5%, were obtained. Moreover the nano-LC system was coupled with a mass spectrometer for an accurate identification of phytosterols.  相似文献

In this study, a new system for analysis using a dual comprehensive two-dimensional gas chromatography/targeted multidimensional gas chromatography (switchable GC × GC/targeted MDGC) analysis was developed. The configuration of this system not only permits the independent operation of GC, GC × GC and targeted MDGC analyses in separate analyses, but also allows the mode to be switched from GC × GC to targeted MDGC any number of times through a single analysis. By incorporating a Deans switch microfluidics transfer module prior to a cryotrapping device, the flow stream from the first dimension column can be directed to either one of two second dimension columns in a classical heart-cutting operation. Both second columns pass through the cryotrap to allow solute bands to be focused and then rapidly remobilized to the respective second columns. A short second column enables GC × GC operation, whilst a longer column is used for targeted MDGC. Validation of the system was performed using a standard mixture of compounds relevant to essential oil analysis, and then using compounds present at different abundances in lavender essential oil. Reproducibility of retention times and peak area responses demonstrated that there was negligible variation in the system over the course of multiple heart-cuts, and proved the reliable operation of the system. An application of the system to lavender oil, as a more complex sample, was carried out to affirm system feasibility, and demonstrate the ability of the system to target multiple components in the oil. The system was proposed to be useful for study of aroma-impact compounds where GC × GC can be incorporated with MDGC to permit precise identification of aroma-active compounds, where heart-cut multidimensional GC-olfactometry detection (MDGC-O) is a more appropriate technology for odour assessment.  相似文献

A sample of tobacco essential oil was analyzed using gas chromatography-mass spectrometry (GC/MS) and comprehensive two-dimensional gas chromatography coupled to a time-of-flight mass spectrometry (GC × GC/TOFMS), respectively. In the GC/MS analysis, serially coupled columns were used. By comparing the GC/MS results with GC × GC/TOFMS results, many more components in the essential oil could be found within the two-dimensional separation space of GC × GC. The quantitative determination of components in the essential oil was performed by GC × GC with flame ionization detection (FID), using a method of multiple internal standards calibration.  相似文献

In a tense energetic context, the characterization of heavy petroleum fractions becomes essential. Conventional comprehensive two-dimensional gas chromatography (2D-GC or GC × GC) is widely used for middle distillates analysis, but only a few applications are devoted to these heavier fractions. In this paper, it is shown how the optimization of GC × GC separation allowed the determination of suitable high-temperature (HT) conditions, adjusting column properties and operating conditions. 2D separations were evaluated using 2D separation criteria and a new concept of 2D asymmetry (As2D). New HT conditions allowed the extension of GC × GC range of applications to heavier hydrocarbons, up to nC60. A first application of high-temperature two-dimensional gas chromatography (HT-2D-GC) to a full vacuum gas oil (VGO) feed stock is described. Comparisons with other standardized methods illustrate the high potential of HT-2D-GC for heavy fractions analysis.  相似文献

Conventional gas chromatography with electroantennographic detection (GC-EAD) and two-dimensional (GC × GC) gas chromatography using a time-of-flight mass spectrometric detector (TOFMS), were combined to analyse the female sex pheromone gland extract of the persimmon bark borer, Euzophera batangensis. GC-EAD analysis produced two EAD responses in GC areas where no compounds were detected by FID detection. GC × GC/TOFMS analysis of this area indicated the presence of several chemicals, including (Z9,E12)-tetradeca-9,12-dien-1-ol and (Z9)-tetradec-9-en-1-ol, pheromone components of closely related Euzophera species. Spectral characteristics, retention behaviour and the ability to elicit GC-EAD responses imply that both identified unsaturated alcohols are candidates for E. batangensis sex pheromone components. GC × GC/TOFMS facilitated the analysis of complex matrices on a subnanogram level and was shown to have great potential as a powerful tool in the analysis of insect pheromones.  相似文献

Comprehensive two-dimensional gas chromatography (GC × GC) offers an interesting tool for profiling bacterial fatty acids. Flow modulated GC × GC using a commercially available system was evaluated, different parameters such as column flows and modulation time were optimized. The method was tested on bacterial fatty acid methyl esters (BAMEs) from Stenotrophomonas maltophilia LMG 958T by using parallel flame ionization detector (FID)/mass spectrometry (MS). The results are compared to data obtained using a thermal modulated GC × GC system. The data show that flow modulated GC × GC-FID/MS method can be applied in a routine environment and offers interesting perspectives for chemotaxonomy of bacteria.  相似文献

This study examined how advanced fingerprinting methods (i.e., non-targeted methods) provide reliable and specific information about groups of samples based on their component distribution on the GC × GC chromatographic plane. The volatile fractions of roasted hazelnuts (Corylus avellana L.) from nine different geographical origins, comparably roasted for desirable flavor and texture, were sampled by headspace-solid phase micro extraction (HS-SPME) and then analyzed by GC × GC-qMS. The resulting patterns were processed by: (a) “chromatographic fingerprinting”, i.e., a pattern recognition procedure based on retention-time criteria, where peaks correspondences were established through a comprehensive peak pattern covering the chromatographic plane; and (b) “comprehensive template matching” with reliable peak matching, where peak correspondences were constrained by retention time and MS fragmentation pattern similarity criteria. Fingerprinting results showed how the discrimination potential of GC × GC can be increased by including in sample comparisons and correlations all the detected components and, in addition, provide reliable results in a comparative analysis by locating compounds with a significant role. Results were completed by a chemical speciation of volatiles and sample profiling was extended to known markers whose distribution can be correlated to sensory properties, geographical origin, or the effect of thermal treatment on different classes of compounds. The comprehensive approach for data interpretation here proposed may be useful to assess product specificity and quality, through measurable parameters strictly and consistently correlated to sensory properties and origin.  相似文献

Sensitive, simple and rapid ELISA methods have been developed for the determination of four organophosphorus pesticides in extra virgin olive oil. The analytical procedure involves simultaneous extraction of the analytes from oil matrix with methanol and a freezing clean-up step (−80 °C), followed by immunoassay determination using standards in matrix. The methodology is specific for diazinon, fenthion, malathion and chlorpyrifos showing little or no cross-reactivity against other organophosphorus compounds. Limits of detection for the pesticides in olive oil are from 46 ng ml−1 for diazinon to 10 ng ml−1 for fenthion, all of them under the established MRLs for olives. The excellent recoveries (between 94 and 122%) obtained by the complete analytical protocol confirm the potential of this approach for detecting these compounds in olive oil, being useful as screening and complementary method in pesticide regulatory and food safety programs. The proposed methodology also correlates well with the reference chromatographic (GC-MS) methods.  相似文献

The ability of multivariate analysis methods such as hierarchical cluster analysis, principal component analysis and partial least squares-discriminant analysis (PLS-DA) to achieve olive oil classification based on the olive fruit varieties from their triacylglycerols profile, have been investigated. The variations in the raw chromatographic data sets of 56 olive oil samples were studied by high-temperature gas chromatography with (ion trap) mass spectrometry detection. The olive oil samples were of four different categories (“extra-virgin olive oil”, “virgin olive oil”, “olive oil” and “olive-pomace” oil), and for the “extra-virgin” category, six different well-identified olive oil varieties (“hojiblanca”, “manzanilla”, “picual”, “cornicabra”, “arbequina” and “frantoio”) and some blends of unidentified varieties. Moreover, by pre-processing methods of chemometric (to linearise the response of the variables) such as peak-shifting, baseline (weighted least squares) and mean centering, it was possible to improve the model and grouping between different varieties of olive oils. By using the first three principal components, it was possible to account for 79.50% of the information on the original data. The fitted PLS-DA model succeeded in classifying the samples. Correct classification rates were assessed by cross-validation.  相似文献

Valve-based comprehensive two-dimensional gas chromatography (GC × GC) is one of the most compact, robust, and inexpensive GC × GC instrument designs. The major drawback of a valve-based modulation configuration lies in diminished detection sensitivity. This loss in sensitivity is because under typical operating conditions the fraction of the first column (i.e., column 1) effluent transferred to the second column (i.e., column 2) is likely to be ∼5-10%. To address this loss in sensitivity, we report the development of a unique total-transfer (i.e., 100%) valve-based GC × GC, without adding complexity to the instrumentation. The new instrument design relies upon simply blocking one of the appropriate ports of the high-speed six-port diaphragm valve that is used as the modulator between columns 1 and 2. The modulation period and difference in head pressure between columns 1 and 2 are found to be the two primary variables that are controlled to provide good detection sensitivity and 100% mass transfer from column 1 to column 2. The detection sensitivity is better with a longer the modulation period. A limit of detection of 0.03 ng/μl was obtained for octane. This sensitive GC × GC configuration is also shown to provide acceptable separation peak capacity, with good separations achieved for real complex samples: gasoline and Eucalyptus oil, where compounds were spread out over much of the two-dimensional separation space. In principle, this total-transfer, valve-based GC × GC is more portable and less expensive than currently available GC × GC instrumentation.  相似文献
