Thermolysin, -chymotrypsin, and papain were used as biocatalysts for the coupling of several carboxyl components to silica-supported leucine amide as a nucleophile. A spacer length of 21 covalent bonds corresponding to 7 amino acid residues was required for successful coupling. This approach offers the possibility of using proteases for racemization-free segment condensations on insoluble polymeric supports.

Zusammenfassung Thermolysin, -Chymotrypsin und Papain wurden als Biokatalysatoren für die Kupplung verschiedener Carboxylkomponenten an kieselgel-gebundenes Leucinamid als Nucleophil eingesetzt. Für erfolgreiche Kupplungen war eine Spacerlänge von 21 kovalenten Bindungen entsprechend 7 Aminosäureresten erforderlich. Diese Synthesevariante eröffnet die Möglichkeit, racemisierungsfreie Segmentkondensationen an unlöslichen polymeren Trägern mit Hilfe von Proteasen durchzuführen.

Inclusion complexation of heptakis (2,6-di-O-methyl)--cyclodextrin (DM--CyD) with -tocopheryl acetate and -tocopheryl nicotinate in aqueous solution was studied by the solubility method. The aqueous solubilities of the esters were about 105 times increased by DM--CyD complexation. The phase-solubility diagram of the tocopheryl ester-DM--CyD systems showed a typicalA p type, and the stability constants (K) of high-order complexes were estimated by analyzing the upward curvature of the diagrams. The solid complex of -tocopheryl nicotinate with DM--CyD in a molar ratio of 12 was prepared by the kneading method. The dissolution rate of the solid complex was much greater than that of the drug itself, and the rapidly dissolving form of -tocopheryl nicotinate, as an example, showed a markedly increased bioavailability (about 70-fold) after oral administration to fasted dogs.  相似文献

The condensation of dichloromethyl tert-butyl ketone (1) with benzaldehyde, para-bromobenzaldehyde, and 2,4-dichlorobenzaldehyde under conditions for the Darzens reaction gives 1-aryl-1-chloro-4,4-dimethyl-2,3-pentanediones (2–4). In the case of nitrobenzaldehydes, the reaction products are 1-aryl-2-chloro-1,2-epoxy-4,4-dimethylpentanones (5 and 6), which isomerize to -chloroketones (7 and 8) upon prolonged storage or heating at reflux in benzene.A. The chiroptical properties of dissymmetric cyclopentanedione, 3-methylcyclopentane-1,2-dione, and glyoxal structures are examined on a theoretical model in which the electronic wave functions are obtained from semiempirical all-valence-shell molecular orbital calculations. Excited state wave functions are constructed in the virtual orbital-configuration interaction approximation. The rotatory strengths, dipole strengths, oscillator strengths, and dissymmetry factors of the lower energy singletsinglet transitions in eleven cyclopentanedione and ten glyoxal structures are calculated and reported. The signs and relative magnitudes of the rotatory strengths associated with the two lowest energy singlet transitions are found to be extraordinarily sensitive to ring substituents and ring conformational parameters as well as to inherent chirality within the -dicarbonyl moiety of the cyclopentanedione structures. Vicinal effects play a significant role in determining the signs and magnitudes of the electronic rotatory strengths. For a given configurational isomer of an inherently dissymmetric -dicarbonyl group (i.e., P or M), the signs of the electronic rotatory strength of the lowest energy transition in glyoxal and in cyclopentanedione are opposite. This result suggests that cisoid glyoxal structures may not be useful models for the chiroptical properties of cyclic -diketone systems with cisoid dicarbonyl moieties.This work was supported in part by a grant from the Petroleum Research Fund administered by the American Chemical Society, the Camille and Henry Dreyfus Foundation, and a computing grant from the University of Virginia Computer Science Center.  相似文献

Cathodic reduction of nitro-substituted azidocinnamic esters on mercury under aprotic and protic conditions was studied. Nitro and azido groups behave as independent entities so that selectively amino- resp. hydroxylamino--azido compounds are obtained, which could not be synthesised by conventional methods. Under aprotic conditions, with added acetic anhydride, medium yields of acylated 4-hydroxylamino- and 4-amino--cinnamic acid derivatives are isolated. Evidence of homogeneous electron transfer from nitroanion radicals to azido groups is given.

Procedures for the synthesis of 4,5-bis[2,5-dimethyl(3-thienyl)]-1,3-azoles based on 1,2-bis[2,5-dimethyl(3-thienyl)]-2-hydroxyethan-1-one, 2-chloro-1,2-bis[2,5-dimethyl(3-thienyl)]ethan-1-one, and 1,2-bis[2,5-dimethyl(3-thienyl)]ethane-1,2-dione were developed. Copyrolysis of 4-chlorotetrafluorobenzotrichloride with chlorodifluoromethane (as the source of difluorocarbene) gave 4-chlorodifluoromethyl--chlorohexafluorostyrene, 1,4-dichlorotetrafluorobenzene, and 4-(chlorodifluoromethyl)chlorotetrafluorobenzene along with 4-chloro--chlorohexafluorostyrene. Intramolecularly hydrogen-bonded, bichromophoric tetrapyrrole pigments, bilirubin-IX and mesobilirubin-XIII, adopt either of two enantiomeric conformations which are in dynamic equilibrium in solution. InpH 8 aqueous sodium deoxycholate solutions, chiral micelles preferentially select one conformational enantiomer, and the solutions exhibit a bisignate circular dichroism Cotton effect in the vicinity of the bilirubin long wavelength electronic transition. Exciton coupling theory indicates a predominance of the left-handed (or negative) chirality bilirubin conformational enantiomer.  相似文献

Effects of -tocopherol (Toc) and -tocopherol acetate (TocA) on the hydrolysis activity of phospholipase D (fromStreptomyces chromofuscus) were studied in small unilamellar vesicles (SUV) of egg yolk phosphatidylcholine (PC). Choline produced in the reaction was monitored by use of a choline oxidase — oxygen electrode. Addition of 18 mol% Toc into SUV (2 mM PC) brought about a twofold rate of choline production. On the other hand, the effect of 18 mol% TocA in SUV was very small. The apparent maximum velocity,V max(app), increased by addition of Toc in SUV. The apparent Michaelis constant,K m(app), was unchanged by addition of Toc and TocA in SUV. The Toc and TocA molecules did not have significant effects when PC was solubilized in the micelles of heptaethylene glycol dodecyl ether. The effects of Toc and TocA are, therefore, not due to specific ones on the enzyme itself, but rather upon the bilayer-organization of the substrate. Measurements of spreading pressure showed complete miscibility of PC and Toc, and limited mutual solubility of PC and TocA, suggesting stronger attractive interactions between Toc and PC than those between TocA and PC in the bilayers.  相似文献

The two hydroxyls of yuccagenin can be glycosylated by Koenigs-Knorr condensation with acetobromorhammose in dichloroethane in the presence of mercuric cyanide. The asymmetric Michael addition of aromatic thiols to 2-cyclohexenone and maleic acid esters has been carried out by utilizing their crystalline cyclodextrin complexes suspended in water. The best chiral induction, 30% enantiomeric excess (ee), was achieved in combinations of 2-cyclohexenone and octyl maleate with the crystalline -cyclodextrin complex of benzenethiol (method A) to afford (S)-3-phenylthiocyclohexanone and (S)-octyl-2-phenylthiosuccinate, respectively, whereas the reaction of benzenethiol with 2-cyclohexenone included in -cyclodextrin (method B) inversely induced the chiral recognition to give the (R)-adduct with 4–9% ee.  相似文献
