Three new platinum(II) complexes of (1R,2R)-N1-(2-butyl)-1,2-cyclohexanediamine with malonate derivatives as leaving groups have been synthesized and spectrally characterized. They were tested in vitro against four human cancer cell lines. [(1R,2R)-N1-(2-butyl)-1,2-cyclohexanediamine-N,N′](2-ethylmalonato-O,O′)platinum(II) turned out to be more active (IC50 = 4.65 μM) than oxaliplatin (IC50 = 6.55 μM) against the MCF-7 cell line and is superior to its parent complex, [(1R,2R)-N1-(2-butyl)-1,2-cyclohexanediamine-N,N′](malonato-O,O′)platinum(II). In addition, agarose gel electrophoresis study revealed that the interaction of the complex with pET22b plasmid DNA had a different behavior from that of cisplatin or oxaliplatin.  相似文献

Eight platinum(II) complexes with the new chiral ligands, (1R,2R)-N 1-(pyridine-2-ylmethyl) cyclohexane-1,2-diamine (R) or (1S,2S)-N 1-(pyridine-2-ylmethyl) cyclohexane-1,2-diamine (S) as the carrier groups were designed, synthesized, and spectrally characterized. All platinum(II) complexes showed much better aqueous solubility than cisplatin and oxaliplatin. In vitro cytotoxicity of the compounds against human HepG-2, MCF-7, A549, and HCT-116 cell lines was evaluated. Results indicate that all compounds with R as the carrier group showed cytotoxicity against HCT-116, A549, and MCF-7 cell lines; however, all compounds with S as carrier group exhibited disappointing cytotoxicity against tested cell lines. Compound R2, bearing ClCH2COO- as leaving group, exhibited better cytotoxicity than that of carboplatin against A549 and MCF-7 cell lines and also showed close activity to oxaliplatin against HCT-116 cell line.  相似文献

Four platinum(II) complexes, trans-[bicyclo[2.2.2]octane-7R,8R-diamine]bis(alkoxyacetato-O,O’) platinum(II) (alkoxyacetate = methoxyacetate (2), ethoxyacetate (3), isopropoxyacetate (4), and tert-butoxyacetate (5)) were synthesized and spectrally characterized. The cytotoxicity of these water-soluble complexes was evaluated by CCK-8 assay in vitro against HCT-116, HepG-2, and A549 cancer cell lines. Most of the complexes had cytotoxic activity against the tested cancer cell lines. Among them, 3 showed more potent antitumor effect than cisplatin or oxaliplatin. Complex 3 could cause HCT-116 cell line death based on an apoptotic pathway since it has a dicyclic moiety similar to 1R,2R-diaminocyclohexane in oxaliplatin. Agarose gel electrophoresis on the interaction between 3 and DNA indicated that it has different behavior from that of cisplatin or oxaliplatin, which has a high correlation with the ligand used.  相似文献

Cancer cases are alarmingly increasing worldwide, and newer chemotherapeutic agents are needed. Recent analogs of cisplatin (carboplatin, lobaplatin, nedaplatin and oxaliplatin) and their marketing as advanced chemotherapeutic drugs have furthered the interest in metal-based anti-cancer drugs. In the current study, two new polymeric organotin(IV) carboxylate complexes (1 and 2) have been synthesized and characterized. Spectroscopic studies showed that coordination took place via carboxylates. Furthermore, X-ray crystallographic study on 1 indicated that it possesses a monomeric structure and exists in polymeric formation due to additional Sn–N coordination, assigning seven coordinations to each metal ion. Both the complexes were tested against three cancerous (human colon cancer, HCT 116; breast cancer, MCF-7; and leukemia, K562) and one non-cancerous (3T3-L1) cell lines. Complex 1 showed exceptional cytotoxicity against cancerous cell lines (IC50 = 1.0 μM for HCT 116; 258.7 nM for MCF-7; and 46.7 nM K562) and remained comparatively non-toxic against normal cells (IC50 = 37.0?μM). This shows that both complexes have selective cytotoxicity against cancer cells.  相似文献

The reaction of silver acetate with cis-[PtI2(dbtp)2], where dbtp = 5,7-ditertbutyl-1,2,4-triazolo-[1,5-a]pyrimidine, yielded cis-[Pt(OOCCH3)2(dbtp)2]·dmf (1). The complex has been analyzed by multinuclear magnetic resonance (1H, 13C, 15N), IR, and Raman. The compound formed two rotamers in CDCl3 and its spatial structures have been optimized using computational calculation. It was found that head-to-tail rotamer (1a) is more stable than its head-to-head counterpart (1b). In vitro antiproliferative activity against four tumor cell lines (A549, T47D, FaDu, and A2780cis) revealed in all cases significant cytotoxicity (IC50 = 0.26–1.80 μM), possessing IC50 values at least fivefold lower than cisplatin, carboplatin, and oxaliplatin (except A2780cis). The remarkable in vitro activity against T47D and A2780cis suggested the ability to overcome cisplatin resistance in these types of tumor cells. In addition, in vitro toxicity was evaluated against BALB/3T3 and has shown that the lipophilic platinum(II) complex (1) inhibits cell proliferation weaker than cisplatin and oxaliplatin. Additionally, cis-[Pt(OOCCH3)2(dbtp)2]·dmf exhibited selective activity, in contrast to cisplatin or oxaliplatin.  相似文献

Abstract

A new series of tricarbonyl complexes of rhenium(I) in the “2 + 1” system with the bidentate ligand N,6-dimethylpyridine-2-carbothioamide ((CH3)NC5H4-CS-NH-CH3, MeLH(Me)NS) and a monodentate ligand (halides Cl, Br, or I, and the pseudohalide NCS anion) was synthesized. The use of mixed ligands led to the formation of neutral tricarbonylrhenium(I) complexes [Re(CO)3(MeLH(Me)NS)X] (X = Cl, Br, I, NCS) (14). Single-crystal X-ray diffraction was used to determine the crystal structures of all four compounds and those results were compared with molecular structures obtained from DFT calculations using the PBE0/def2-TZVPD approach. The complexes were also characterized by spectroscopic (FT-IR, NMR, and UV–vis) and analytical (HPLC, TGA, EA, ESI-MS) techniques. IR and UV–vis spectra were also calculated by DFT and TD-DFT methods. The cytotoxicity of these complexes was estimated using human ovarian cancer cell lines (A2780 and A2780cis), cervical cancer cells (HeLa), and non-cancerous human embryonic kidney cells (Hek-293). The toxicity of most complexes was moderate or low toward cancer cell lines (IC50 = 46–231 μM) and similar against non-cancerous cells (IC50 = 41-121 μM). Only the complex with chlorido ligand remarkably inhibited growth of ovarian cancer cells (IC50 = 3 and 12 μM for A2780 and A2780cis, respectively). The cytotoxicity of 1 was higher than that of cisplatin.  相似文献

Four new manganese(III) Schiff base complexes (1–4) were synthesized and characterized. The complexes have general formula [MnClLx] in which L represents a Schiff base ligand derived from condensation of meso-1,2-diphenyl-1,2-ethylenediamine with salicylaldehyde or its 3-OMe-, 5-Br-, or 5-OMe-derivatives (x = 1–4, respectively). The crystal structure of [MnClL1] (1) was characterized by X-ray crystallography. The in vitro anticancer activity of these complexes was evaluated by MTT and apoptosis assays against human breast (MCF-7) and liver (Hep G2) cancer cells. The complexes exhibited considerable antiproliferative activity against both cell lines (IC50 = 10.8–21.02 μM) comparable to cis-platin, except 4 (MCF-7). The highest activity was found for 1 with IC50 values of 13.62 μM (MCF-7) and 10.8 μM (Hep G2). Flow cytometry experiments showed that 1 induced apoptosis on MCF-7 tumor cell line. Docking simulations using AUTODOCK were also carried out. The results showed that all complexes fitted into the minor groove region of DNA.  相似文献

[Zn(L)2] (1) and [(Ph)2Sn(L)(CH3COO)] (2), where HL = 2-benzoylpyridine N(4)-cyclohexylthiosemicarbazone, have been synthesized and characterized. The complexes show different coordination depending on their coordinating preferences. Biological studies carried out in vitro against human leukemia K562 cells show that the diorgantin(IV) complex, 2, has significant cytotoxicity with IC50 = 3.3 ± 0.5 μM.  相似文献

Three new xanthones, named calaxanthones A-C (13), along with 17 known xanthones (420) were isolated from the roots of Calophyllum calaba. Their structures were determined by spectroscopic analysis. All isolated compounds were evaluated for their cytotoxicity against five cancer cell lines (KB, HeLa S-3, HT-29, MCF-7 and HepG-2). Compound 3 showed potent cytotoxicity against all the five cancer cell lines with IC50 values in the range of 0.82–5.04 μM. Furthermore, compound 6 showed potent cytotoxicity against KB, HeLa S-3 and HepG2 cells with IC50 values of 7.06, 5.27 and 9.64 μM, respectively. Additionally, compound 7 showed potent cytotoxicity against KB cell with an IC50 value of 4.62 μM.  相似文献

For the first time, eugenol, a natural bioactive allylphenol, was introduced into coordination with platinum(II) by replacement of ethylene from Zeise’s salt with eugenol (Eug). The obtained complex, K[PtCl3(Eug)] (1), was used as the key compound for preparation of the series of trans-[PtCl2(Eug)(Amine)] (2–11), [PtCl(Eug)(8-O-quinoline)] (12) and [PtCl(Eug)(2-O2C-quinoline)] (13). The synthesized complexes were characterized by elemental analyses, IR, 1H NMR, 13C NMR, HSQS, HMBC, NOESY, and MS spectra. In 113 eugenol coordinates with Pt(II) at ethylenic double bond of the allyl group, the donor N of the amines is in trans-position in comparison with the double bond. A display of the trans-effect on the chemical shift of 1H and 13C was remarked. Seven complexes were tested for cell in vitro cytotoxicity on human cancer cells. Complexes 3 and 12 exhibit high activities on Hep-G2 with IC50 = 3.12 and 5.29 μM; 12 gives high activity against KB, Lu and MCF-7 with IC50 = 0.43, 2.95 and 1.84 μM, respectively. Most of these IC50 are lower than those of cisplatin.  相似文献

Abstract

Dinuclear ruthenium complexes [Ru2(bpy)4BL](ClO4)2 (Ru-1), where bpy = 2,2′-bipyridine and BL = 2,2′-((1E,1′E)-((E)-diazene-1,2-diyl-bis(2,1-phenylene))-bis(azanylylidene))bis(methanylylidene))diphenol (a bidentate bridging ligand), and mononuclear ruthenium complexes [Ru(bpy)2L](ClO4) (Ru-2), where L = (E)-2-((phenylimino)methyl)phenol, were synthesized and characterized by elemental analysis and electrospray ionization mass spectrometry. Their photophysical and electrochemical properties were also studied. The cytotoxicity of the two complexes in vitro was evaluated using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. The results indicated that Ru-1 and Ru-2 exhibited significant dose-dependent cytotoxicity to human breast cancer (MCF-7), gastric cancer (SGC-7901), cervical cancer (Hela), and lung cancer (A549) tumor cell lines. Ru-1 showed excellent antitumor effects in a cellular study (IC50 values of 3.61 μM for MCF-7 human breast cancer cells in vitro). However, Ru-2 exhibited the highest cytotoxicity to Hela cells; the IC50 value is 3.71 μM. The results reveal that Ru-1 and Ru-2 have obvious selectivity and might be a potential anticancer agent that could improve the efficacy of common anticancer therapies.  相似文献

The crude extracts of tubers of Aconitum spicatum (Bruhl) Stapf were investigated for in vitro antileishmanial activity against Leishmania major. The dichloromethane extract at pH 2.5 showed antileishmanial activity with IC50 value of 27.10 ± 0.0 μg/mL. Chromatographic purification of the dichloromethane extract led to isolation of three C-19 norditerpenoid alkaloids indaconitine (1), chasmaconitine (2) and ludaconitine (3). Compounds 3 and 2 showed antileishmanial activity with IC50 = 36.10 ± 3.4 and 56.30 ± 2.1 μg/mL, respectively. Compound 1 was less effective (IC50 > 100 μg/mL). The cytotoxicity of compounds 1, 2 and 3 studied against MCF7, HeLa and PC3 cancer cell lines and 3T3 normal fibroblast cell line did not show cytotoxicity at 30 μM.  相似文献

Twelve compounds were isolated from the roots of Aglaia odorata. Their structures were established on the basis of NMR and MS data as rocaglaol (1), rocaglamide (2), eichlerialactone (3), sapelins A (4), isofouquierone (5), eichlerianic acid (6), shoreic acid (7), agladupol E (8), 3-epimeliantriol (9), cleomiscosins B (10), 2β,3β-dihydroxy-5α-pregnane-16-one (11) and β-d-glucopyranos-1-yl N-methylpyrrole-2-carboxylate (12). Among them, compounds 1 and 2 showed significant cytotoxicity against human cancer cell (HL-60, SMMC-7721, A-549, MCF-7 and SW480) with IC50 values of 0.007–0.095 μM, while compounds 35 and 10 and 11 showed moderate to no cytotoxicity (IC50 0.43 to values >40 μM). Compound 6 showed only weak cytotoxicity (IC50 6.87 to >40 μM) and its epmier 7 was completely inactivite (IC50>40 μM) in the assay. However, potent synergistic effect was observed when the molar ratio of 6 to 7 is between 4:1 and 1:1.  相似文献

Acetone:chloroform (1:2) extract of the aerial parts of Euphorbia connata Boiss. (Euphorbiaceae) was investigated for its diterpenoids. This led to the isolation of one known and two new diterpenes, belonging to the pentahydroxy-13(17)-epoxy-8,10(18)-myrsinadiene and tetrahydroxy-5,6-epoxy-14-oxo-jatropha-11(E)-ene classes. The structures were elucidated based on 13C and 1H NMR as well as 2D NMR, IR and MS spectra and the cytotoxicity for compounds 13 were evaluated by using MTT assay against two human breast cancer cell lines. Myrsinane-type compounds – 3,7,14,15-tetraacetyl-5-propanoyl-13(17)-epoxy-8,10(18)-myrsinadiene (1) and 3,7,10,14,15-pentaacetyl-5-butanoyl-13,17-epoxy-8-myrsinene (2) – exhibited moderate inhibitory effects, with IC50 values of 24.53 ± 3.39 and 26.67 ± 1.41 μM against the MDA-MB cell line, and 37.73 ± 3.41 and 34.57 ± 2.12 μM against the MCF-7 cell line, respectively. Jatrophane-type diterpene – 5,6-epoxy-8,9,15-triacetyl-3-benzoyl-14-oxo-jatropha-11(E)-ene (3) – showed weak cytotoxicity, with IC50 values of 55.67 ± 7.09 μM against MDA-MB, and moderate cytotoxicity with IC50 values of 24.33 ± 3.21 μM against MCF-7 cell line.  相似文献

Two new Pt(II)-pyrophosphato complexes containing the carrier ligands cis-1,3-diaminocyclohexane (cis-1,3-DACH) and trans-1,2-diamine-4-cyclohexene (1,2-DACHEX), variants of the 1R,2R-diaminocyclohexane ligand present in the clinically used Pt-drug oxaliplatin, have been synthesized with the aim of developing new potential antitumor drugs with high bone tropism. The complexes are more stable at physiological pH than in acid conditions, with Na2[Pt(pyrophosphato)(cis-1,3-DACH)] (1) slightly more stable than [Pt(dihydrogenpyrophosphato)(1,2-DACHEX)] (2). The greater reactivity at acidic pH ensures a greater efficacy at the tumor site. Preliminary NMR studies indicate that 1 and 2 react slowly with 5’-GMP (used as a model of nucleic acids), releasing the pyrophosphate ligand and affording the bis 5’-GMP adduct. In vitro cytotoxicity assays performed against a panel of four human cancer cell lines have shown that both compounds are more active than oxaliplatin. Flow cytometry studies on HCT116 cells showed that the pyrophosphato compounds with the non-classical 1,3- and 1,4-diaminocyclohexane ligands (1 and 4) are the most capable to induce cells’ death by apoptosis and necrosis.  相似文献

[CoIII(L1)2·H2O]NO3 (1), [MnII(L1)2·H2O] (2), and [ZnII(L1)2·H2O] (3) with a hydrazone derived from protocatechuic acid (HL1 = C15H13N3O3) were designed, synthesized, and characterized by C, H, N elemental analyses, single-crystal X-ray diffraction, and IR spectra, which revealed that the three complexes are similar structures. Docking study has been done. The urease inhibitory activities of the three complexes were tested. Complexes 1 and 3 showed strong inhibitory activity against jack bean urease with IC50 values of 45.9 and 11.64 μM. Complex 2 had no obvious inhibitory activity to urease; the IC50 was > 50 μM.  相似文献

The ethanolic extract from the stem bark of Goniothalamus marcanii Craib was shown in preliminary brine shrimp lethality data having good cytotoxic activity. Further bioassay guided isolation was done by means of solvent partition, chromatography and precipitation to provide four isolated compounds: a novel compound 1 with the core structure of 1-azaanthraquinone moiety referred as marcanine G; as well as compounds 2–4 with known aristolactam structures namely, piperolactam C, cepharanone B and taliscanine. These compounds were characterised by spectroscopic techniques. The assessment of cytotoxicity was established on an SRB assay using doxorubicin as a positive control. Marcanine G (1) was considered the most active compound indicating the IC50 values of 14.87 and 15.18 μM against human lung cancer cells (A549) and human breast cancer cells (MCF7), respectively. However, 2 showed mild activity with the IC50 values of 83.72 and 82.32 μM against A549 and MCF7 cells, respectively.  相似文献

Abstract

A series of new pyrido-cyclopenta[1,2-b]indole derivatives were synthesized via Knoevenagel reaction and followed by reflux with dimethylformamide dimethyl acetal. Their structures were investigated by spectral techniques and elemental analysis. In vitro antibacterial assessment against seven selected microorganisms evidenced that the compounds with halogen substituent have strong inhibitory action than that of the reference drugs. The antioxidant results were apparent that the compounds 5b, 5c, and 6c manifested explicit activity when compared with Butylhydroxyanisole and Vitamin-C. Cytotoxic activity analysis toward HeLa and MCF-7 cell lines was also assessed. Analogs 6c (IC50 values 15.1?μM and 18.6?μM) and 6d (IC50 values 17.4?μM and 20.7?μM) illustrated the interesting cytotoxicity activity. Molecular docking studies against p38 MAP kinase displayed a potential binding affinity with the receptor. Furthermore, in silico pharmacokinetic studies articulated the drug-likeness nature of the target compounds.  相似文献

Several new PtII and PdII complexes bearing the enantiomerically pure (1R,2R)-(–)-l,2-cyclohexanediamine (dach) ligand, of general formula [MX2{(1R,2R)-dach}], where M = Pt or Pd, X2 = cis- or trans- or (1R,2R)-1,2-cyclohexyldicarboxylate anions, have been synthesized and characterized physicochemically and spectroscopically. These complexes have been screened in vitro against the three tumour cell lines K562, HeLa and L929, and the results obtained were compared with those of the reference standards, cisplatin, carboplatin and oxaliplatin; the known antitumour drugs. The single crystal X-ray structure determination of the [Pt(C2O4)(cis-dach)] complex has been discussed and compared with that of oxaliplatin, [Pt(C2O4){(1R,2R)-dach}].  相似文献

The CH2Cl2/MeOH (1:1) extract of the aerial parts of Tephrosia subtriflora afforded a new flavanonol, named subtriflavanonol (1), along with the known flavanone spinoflavanone B, and the known flavanonols MS-II (2) and mundulinol. The structures were elucidated by the use of NMR spectroscopy and mass spectrometry. The absolute configuration of the flavanonols was determined based on quantum chemical ECD calculations. In the antiplasmodial assay, compound 2 showed the highest activity against chloroquine-sensitive Plasmodium falciparum reference clones (D6 and 3D7), artemisinin-sensitive isolate (F32-TEM) as well as field isolate (KSM 009) with IC50 values 1.4–4.6 μM without significant cytotoxicity against Vero and HEp2 cell lines (IC50 > 100 μM). The new compound (1) showed weak antiplasmodial activity, IC50 12.5–24.2 μM, but also showed selective anticancer activity against HEp2 cell line (CC50 16.9 μM).  相似文献
