A new efficient protocol for the synthesis of benzannulated pyrido[2,3-d]- and pyrido[3,2-d]pyrimidines has been successfully accomplished via the palladium-catalyzed intramolecular arylation of C-H bond of pyrimidine moiety. The methodology has also been extended to the synthesis of pyrimido[5,4,-c]isoquinoline-2,4,6(1H,3H,5H)-trione derivatives in excellent yields.  相似文献

New methodology is described for the construction of tricyclic heterocycles. Thus, a double displacement reaction of l-(2-fluorobenzoyl)-2-methylthio-2-imidazoline ( 8a ) with 1,1-dialkylhydrazines gave 10-substituted 2,10-dihydroimidazo[2,1-b]quinazolin-5(3H)-ones in good yield. The corresponding 1-(2-nitrobenzoyl)-2-meth-ylthio-2-imidazolines also underwent double displacement reactions with hydrazines. Other tricyclics made using double displacement reactions were pyrimido[2,1-b]quinazolines, imidazo[1,2-a]pyrido[2,3-d]pyrimidines, and imidazo[1,2-a]pyrazolo[3,4-d]pyrimidines. Treatment of 8a with hydrazine hydrate or methylhydrazine gave products resulting from displacement, but did not afford fused benzotriazepinones.  相似文献

The chemical behavior of 6-formylkhellin ( 1 ) was investigated toward a variety of carbon nucleophiles. Treatment of aldehyde 1 with cyanoacetamide, N-benzylcyanoacetamide produced pyridine-3-carboxamides 3 and 4 . Treatment of carboxaldehyde 1 with malononitrile dimer and 1H-benzimidazol-2-ylacetonitrile gave 1,6-naphthyridine 5 and pyrido[1,2-a]benzimidazole 6 , respectively. Some novel pyrazolo[3,4-b]pyridine 7 , pyrido[2,3-d]pyrimidines 8 and 9 were synthesized from the ring opening ring closure reactions of carboxaldehyde 1 with certain heterocyclic enamines. In addition, reaction of carboxaldehyde 1 with certain cyclic enols produced a variety of products. Treatment of carboxaldehyde 1 with 1,3-cyclohexanediones gave xanthene-1,8-diones 19 and 20 . Reaction of carboxaldehyde 1 with 5-methyl-2,4-dihydro-3H-pyrazol-3-one proceeds in 1:2 M ratio producing pyrazolo [4′,3′:5,6]pyrano[2,3-c]pyrazole derivative 22 . Carboxaldehyde 1 reacted with certain heterocyclic compounds containing active methylene groups to give the corresponding condensation products 22 - 27 . The synthesized compounds were screened in vitro for their antimicrobial activity and showed high to moderate activities against the tested microorganisms.  相似文献

Methyl 2-benzoylamino-3-dimethylaminopropenoate ( 2 ) was introduced as a new reagent for the preparation of fused pyrimidinones 4 from heterocyclic α-amino compounds in acetic acid. In this manner, derivatives of pyrido[1,2-a]pyrimidine 4a,b,f , pyrimido[1,2-b]pyridazine 4g , pyrimido[1,2-c]pyrimidine 4j , pyrazino[1,2-a]pyrimidine 4k , thiazolo[2,3-b]pyrimidine 41 , pyrazolo[1,5-a]pyrimidine 4m , and 1,2,4-triazolo-[1,5-a]pyrimidine 4n were prepared.  相似文献

2,3,5,7-Tetramethyloxazolo[3,2-a]pyrimidinium perchlorate in reactions with sodium hydroxide, sodium ethoxide, and pyrrolidine undergoes earlier unknown recyclization into 1-substituted pyrrolo[1,2-c]pyrimidines. Recyclization of the same salt under the action of ammonia gives 2,3,5,7-tetramethylimidazo[1,2-a]pyrimidine. Published in Russian in Izvestiya Akademii Nauk. Cyclocondensation of 2-arylmethylidene-3-fluoroalkyl-oxopropionates with 2-amino-pyridine occurs at both the polyfluoroacylvinyl and alkoxycarbonylvinyl fragments to give alkyl 4-aryl-2-polyfluoroalkyl-4H-pyrido[1,2-a]pyrimidine-3-carboxylates and 4-aryl-2-hydroxy-3-polyfluoroacyl-4H-pyrido[1,2-a]pyrimidines, respectively. A series of 7-methylpyrazolo[1,5-a]pyrimidines were reacted with dimethylformamide dimethylacetal to give the corresponding dimethylaminovinyl derivatives. These were reacted with ammonium acetate affording, through a ring closure, a number of 6-methylpyrazolo[1,5-a]pyrido[3,4-e]pyrimidines bearing various substituents on the pyrazole ring. The 6-acetyl-2-hydroxy-7-methylpyrazolo[1,5-a]pyrimidine was used as starting material for obtaining some O-alkyl derivatives. Catalytic transfer hydrogenation of 2-benzyloxy-6-methylpyrazolo[1,5-a]pyrido[3,4-e]pyrimidine led to the 2-hydroxy derivative.  相似文献

Diethyl ethoxymethylenemalonate was used for the novel synthesis of the triheterocyclic 3-carbethoxy-9,ll-disubstituted-4-oxo-4H-pyrido[3,2-e]pyrimido[1,2-c]pyrimidines from 4-aminopyrido[2,3-d]pyrim-idines via thermal cyclization of the intermediate ethyl 2-carbethoxy-3-[5,7-disubstituted-4-amino-pyrido[2,3-d]pyrimidin-4-yl]acrylates. The alkaline hydrolysis of 3-carbethoxy-4-oxo-4H-pyrido[3,2-e]-pyrimido[1,2-c]pyrimidines was performed to give corresponding acid derivatives.  相似文献

2-(Allylamino)pyrido[3,2-d]pyrimidin-4(3H)-one was converted to linearly fused dihydroimidazo- [1,2-a]pyrido[3,2-d]pyrimidine on heating in polyphosphoric acid, whereas its reactions with molecular iodine and chlorosulfanylarenes afforded mainly angularly fused analogs. 2-(Cinnamylamino)pyrido[3,2-d]pyrimidin- 4(3H)-one reacted with polyphosphoric acid and chlorosulfanylarenes to give linear pyrido[3,2-d]pyrimido-[1,2-a]pyrimidinones, and its iodocyclization led to the formation of angularly fused derivative.  相似文献

A new synthetic approach for pyrido[2,3-d]pyrimidine 3-oxides and other pyrido[2,3-d]pyrimidines has been developed. This bicyclic system readily undergoes ring opening to give a variety of products, depending on the reagent and reaction conditions.

Derivatives of a new heterocyclic system - benz[4,5]imidazo[1,2-c]pyrido[3',2';4,5]thieno[2,3-e]pyrimidine have been obtained by successive reactions in three stages - alkylation of 3-cyanopyridine-2(1H)-thiones with 2-chloromethylbenzylimidazole to give 2-benzimidazolylmethylthio-3-cyanopyridines, closing the thiophene ring in the latter to form 3-amino-2-(benzimidazolyl-2)thieno[2,3-b]pyridines, and cyclization of the pyrimidine ring by acylation with carboxylic acid anhydrides or chlorides.  相似文献

In the imidazo[1,2]diazine series, only the imidazo[1,2-a]- and [1,2-c] pyrimidine systems were found to undergo a Dimroth-type rearrangement under basic aqueous conditions. Various 6,7-dihydro-5H-benzo[6,7]cyclohepta[1,2-d][pyrimidines bearing amino acid group at the 4-position of the skeleton were synthesized by the reaction of 4-chloro-6,7-dihydro-5H-benzo[6,7]cyclohepta[1,2-d]-pyrimidine with some amino acid, which were cyclized to 5,6-dihydro-4H-benzo[3,4]cyclohept[1,2-e]imidazo-[1,2-c]pyrimidines, corresponding to B-homo-11,13,15-triazasteroids. An economical and efficient synthesis of dipyrimido-dihydropyridines and pyrimido[4,5-d]pyrimidines is described using greener and recyclable β-cyclodextrin as a supramolecular catalyst in aqueous medium. The remarkable features of this method are mild reaction conditions, short reaction times, easy workup procedure, recyclability of the catalyst, and excellent yields of the products. The highly functional group tolerance and shorter reaction times make this method suitable for the synthesis of dihydropyridine and pyrimido[4,5-d]pyrimidine derivatives with a wide substitution pattern.  相似文献

8,10-Dimethyl-3-(unsubstituted, methyl, ethyl, n-butyl, phenyl)-4-hydroxypyrido[2′,3′:3,4]pyrazolo[1,5-a]pyrimidin-2(1H)-ones and 3-(2-hydroxyethyl)-2,8,10-trimethylpyrido[2′,3′:3,4]pyrazolo[1,5-a]pyrimidin-4-ol were synthesized by cyclocondensation of 3-amine-4,6-dimethyl-1H-pyrazolo[3,4-b]pyridine with ethyl malonates and α-acetyl-γ-butyrlolactone. Dichloro- and diazido- derivatives were obtained from the reaction of pyridopyrazlopyrimidine derivatives with POCl3 followed by NaN3. The tetrahetrocyclic systems were formed by cyclization of 4-chloro-3-(2-chloroethyl)-2,8,10-trimethylpyrido[2′,3′:3,4]pyrazolo[1,5-a]pyrimidine with the appropriate primary amines. The structures of all compounds were established by NMR and mass spectra.  相似文献

Abstract

Herein, we report the synthesis of 48 novel 3-sulfonylamides containing a tricyclic thieno[2,3-d]pyrimidin-4(3H)-one moiety, and their influence on melanin synthesis in murine B16 cells. All target sulfonylamides were synthesized through key intermediate 3-nitro-thieno[2,3-d]pyrimidin-4(3H)-ones using three types of ipso-nitration reactions. In this case, we converted the pyrido[1,2-a]- fragment of the thieno[2,3-d]pyrimidine moiety to pyrrolo[1,2-a]- and azepino[1,2-a]- side-rings in order to evaluate the bioactivities of the synthesized derivatives for a structure activity-relationships point of view. The obtained results suggest that some of the selected compounds revealed a promising influence on melanin synthesis in murine B16 cells and may serve as lead compounds for further drug discovery and development.  相似文献

The reaction of 6-amino-1,3-dimethyluracil with equimolar amounts of arylalkanone Mannich bases under optimized reaction conditions leads to 7-aryl-5,6-dihydropyrido[2,3-d]pyrimidines in a yield of 50-80%. Functionalization of these dihydropyridopyrimidine(1H,3H)-2,4-diones with the Vilsmeier reagent affords, depending on the reaction conditions, either 6-dimethylaminomethylidene substituted 5H-pyrido[2,3-d]pyrimidine(1H,3H)-2,4-diones or the corresponding pyridopyrimidine(1H,3H)-2,4-diones bearing a carboxaldehyde function in position 6 of the heterocycle. Some further transformations of the aldehyde function demonstrate the synthetic potential of the synthesized structures, introducing pharmacologically relevant basic substituents into the side chain of these pyrido[2,3-d]pyrimidine derivatives.  相似文献

2-Amino-6-methylpyridine ( 4 ) reacts with active malonates 2a-d or 3a-d either in acetone solution with triethylamine as catalyst at room temperature or with active malonates 2a-d in acetone solution at reflux temperature to yield the pyrido[1,2-a]pyrimidines 5a-d . 2,6-Diaminopyridine ( 8 ) already reacts without triethylamine with 2a-d at room temperature to afford the pyrido[1,2-a]pyrimidines 9a-d . At higher temperatures pyrido[1,2-a]pyrimidines 5 and 9 are rearranged via ketene intermediates [1] to yield the 1,8-naphthyridines 6a-d , and 10a-d , respectively. The naphthyridines 6 and 10 can also be synthesized directly from 4 or 8 using either diethyl malonates 1 or — with better results — the active malonates 2 at 240–250°. Further reaction of 10a-e with 2c,d leads to the pyridonaphthyridines 12a-f . Nitration of 6c yields the nitro derivative 16 and chlorination of 6c,d gives 15c,d , while the chlorination of 10c affords the di-chloro derivative 17 .  相似文献
