2-(2-Carbethoxy-1-amino-1-thio)ethylenepyradinium(isoqunolinium) ylids were obtained by the reaction of carbethoxycyanomethylpyridinium(isoquinolinium) ylids with hydrogen sulfide. In an alkaline medium they cyclize into 5-mercaptosubstituted imidazo[1,2-a]pyridines(isoquinolines). A new regioselective alkylation of pyridines at their 4‐position was achieved with styrenes in the presence of yttrium trichloride, BuLi, and diisobutylaluminium hydride (DIBAL‐H) in THF. Alternatively, similar products were more simply prepared from pyridines and benzyl Grignard reagents. These reactions are not only a useful preparation of 4‐substituted pyridines but are also complementary to other relevant reactions usually giving 2‐substituted pyridines.  相似文献

Hexasubstitution of apical triflate ligands in the octahedral clusters [M]2[Mo6X8(CF3SO3)6] (M = n-Bu4N or Cs, X = Br or I) and monosubstitution in [n-Bu4N]2[Mo6Br13(CF3SO3)] was carried out in tetrahydrofuran at 60 degrees C with simple pyridines and then extended to organometallic pyridines, yielding cluster-cored stars, and to dendronic polyallyl- and polyferrocenylpyridines, yielding cluster-cored polyallyl and polyferrocenyl dendrimers and dendrons. The orange pyridine-substituted clusters, whose pyridine protons are deshielded in 1H NMR (a practical tool for characterization), are air-stable and thermally stable with simple pyridines, light- and air-sensitive with organometallic pyridines, and air-fragile and thermally fragile with large dendronized pyridines.  相似文献

These are new examples of the homotransmetalation reaction. Instead of the ordinary bromo-metal exchange it is possible to obtain selectively from the 2-bromo 3-fluoro or 2-bromo 3-chloro pyridines the 2-bromo 3-halogeno 4-lithio pyridines, which is very interesting for synthetic utility. From the 4-bromo 3-halogeno pyridines, we can see an isomerization with an halogen dance mechanism and we obtain the 5-bromo 3-halogeno 4-lithio pyridines. In the case of the 2-bromo 3-fluoro pyridine we explain a telesubstitution of bromine from 2 to 4 by a transistory homotransmetalation reaction during the Li-Br exchange.  相似文献

A new three-component reaction of chalcones, ketones, and ammonia assisted by magnesium methoxide is described. The corresponding 2,4,6-triaryl pyridines (Kröhnke pyridines) are formed in fair to good yields (34–76%) within 3 h. The method also provides a facile way to synthesize 2(6)-difluoromethyl pyridines via an interesting mechanism.  相似文献

Halogenated pyridines I. The synthesis of 3-halogenomethyl pyridines from dimeric acrylonitrile . α-Methyleneglutaronitril, a linear dimerization product of acrylonitrile is a cheap, commercially available starting material for a new synthesis of 3-halogenomethyl-2, 6-dihalogeno- or 2,5,6-trihalogenopyridines prepared in two and three step reactions, respectively. From the spin-spin coupling constants of pyridine and monosubstituted pyridines the effects of several substituents have been calculated- Assuming an additivity relationship when two of these substituents are present in the same molecule, the spin-spin coupling constants for 11 disubstituted pyridines have been calculated.The NMR spectra of 13 disubstituted pyridines have been studied to obtain accurate values of their coupling constants. The experimental values of these constants are in very good agreement with those calculated using the additivity relationships.  相似文献

Heteroaryl thioethers, comprised of pyridines and diazines, are an important class of compounds with relevance to medicinal chemistry. Metal-catalyzed cross-couplings and SNAr reactions are traditionally used to form C–S bonds in these systems but are limited by available halogenated precursors. An alternative approach is presented where pyridines and diazines are transformed into heterocyclic phosphonium salts and then C–S bonds are formed by adding thiolate nucleophiles. The process is 4-selective for pyridines, simple to execute and can be used to make derivatives of complex pharmaceuticals.  相似文献

Transition-metal-catalyzed [4+2] heteroannulation of α,β-unsaturated oximes and their derivatives with alkynes has been developed into a powerful strategy for the synthesis of pyridines. It nevertheless lacks regioselectivity when unsymmetrically substituted alkynes are used. We report herein the unprecedented synthesis of polysubstituted pyridines by a formal [5+1] heteroannulation of two readily accessible building blocks. A copper-catalyzed aza-Sonogashira cross-coupling between β,γ-unsaturated oxime esters and terminal alkynes affords ynimines, which, without isolation, undergo an acid-catalyzed domino reaction involving ketenimine formation, 6π-electrocyclization and aromatization to afford pyridines. Terminal alkynes served as a one-carbon donor to the pyridine core in this transformation. Di- through pentasubstituted pyridines are accessible with complete regioselectivity and excellent functional-group compatibility. The first total synthesis of anibamine B, an indolizinium alkaloid with potent antiplasmodial activity, was accomplished featuring this reaction as a key step.  相似文献

An additivity relationship for chemical shifts similar to that found in the literature for coupling constants has been tested in eleven disubstituted pyridines. The chemical shifts of these disubstituted pyridines were measured in different solvents at different concentrations and extrapolated to infinite dilution.In order to compute the “effects” of the different substituents, the chemical shifts of the corresponding monosubstituted pyridines and of pyridine were measured in the same solvents at different concentrations. These chemical shifts were extrapolated to infinite dilution.In most cases it was found that the extrapolated chemical shifts for disubstituted pyridines are in good agreement with those calculated using the additivity relationship. Only in a few cases was it found that the additivity scheme does not hold.  相似文献

A series of pyridines substituted in the 2-position were synthesized by cobalt(I)-catalyzed cotrimerization of alkynes and nitriles. The reactions are usually enhanced by light. The reaction conditions are very mild. Activity and selectivity are satisfactory with respect to formation of pyridines, but depend on the type of nitrile.  相似文献

Abstract

A quantitative analytical method has been developed for volatile pyridines in tobacco smoke using pyridine- 14C as an internal standard. Mainstream and sidestream smoke are trapped in 0.05 N sulfuric acid anti partitioned with chloroform. The acidic portion is then concentrated, adjusted to pH 3 and extracted with chloroform. Methanol and a trace of 2N sulfuric acid are added to this chloroform portion and the mixture is carefully concentrated.

A trace of sodium hydroxide is added and the final concentrate is analyzed by gas chromatography. Twenty-one volatile pyridines have been identified. The mainstream stroke of a popular. 85-mm U.S. cigarette without filter tip contained the following major pyridines: 33.4 μg pyridine, 12.3 μg α-picoline, 24.2 μg. β+γ-picoline, 7.6 μg 3-ethylpgridine, 5.9 μg: 4-ethylpyridine, and 23.3 μg 3,4-lutidine + 3-vinylpyridine. The sidestream smoke of this cigarette contained up to 28 times higher concentrations of pyridines.

Volatile pyridines were also determined in the mainstream smoke of other non-filter cigarettes, filter cigarettes, little cigars and, a non-tobacco cigarette. Of special interest were the much greater quantities of volatile pyridines in the mainstream and sidestream of cigars in comparison to those of cigarettes. The ratio of pyridines in sidestream to those in mainstream was 5 to 50 in large cigars. Selective filtration was observed with both cellulose acetate and charcoal filter tips. The analytical method resulted in reproducible values for pyridine, with an average recovery rate of more than 90%. In addition to pyridines, two pyrazines were also isolated from the smoke. Processed tobacco contained minute amounts of some volatile pyridines.  相似文献

The olefin cross-metathesis reaction provides a rapid and efficient method for the synthesis of α,β-unsaturated 1,5-dicarbonyl derivatives which then serve as effective precursors to mono-tetrasubstituted pyridines. Manipulation of the key 1,5-dicarbonyl intermediate allows access to pyridines with a wide range of substitution patterns. An extension of this methodology facilitates the preparation of pyridines embedded within macrocycles, as exemplified by an efficient synthesis of (R)-(+)-muscopyridine. High levels of regiocontrol, short reaction sequences, and facile substituent variation are all notable aspects of this methodology.  相似文献

A one-pot, effective synthesis of pyridines by a modified Kr?hnke procedure is described. Polysubstituted annulated pyridines were synthesized in high yields by four-component, one-pot cyclocondensation reactions of N-phenacylpyridinium bromide, aromatic aldehydes, acetophenones or cyclic ketones in the presence of ammonium acetate and acetic acid, assisted by microwave irradiation. In this procedure, cyclic ketones with two alpha-CH(2) groups yield annulated pyridines with additional alpha-benzylidene groups, which are derived in situ from double aldol condensation of cyclic ketones with two moles of aromatic aldehydes.  相似文献
