A new, sensitive and selective HPLC method with fluorescence detector (HPLC-FLD) for the determination of nephrotoxic and carcinogenic aristolochic acid (AA) in herbal medicines by using pre-column derivatization with zinc powder in acetic acid is presented. Variables governing the derivatization reaction, such as the amount of zinc powder and acetic acid, as well as the derivatization time were studied and optimized. An extended linear dynamic range over three orders of magnitude was observed for AA-I and AA-II (R(2)>0.9998). Method accuracy at low, medium and high spiked AA levels determined by the percentage mean deviation was below 4.4% and 7.2% for AA-I and AA-II, respectively. The detection limits of 0.39 ng/mL (AA-I) and 0.52 ng/mL (AA-II) were 2 orders of magnitude lower than those obtained from HPLC-MS or CE-ECD analyses, 3-4 orders of magnitude lower than those from HPLC-UV or CE-UV methods. The developed method has been applied for the determination of AA in herbal medicines. Among the tested samples, Guanmutong had the highest AA concentration (2607.0 microg/g AA-I, 711.2 microg/g AA-II). Comparison studies between HPLC-FLD and HPLC-MS/MS demonstrated that the two methods gave similar quantitative results for the selected herb samples.  相似文献

A HPLC method was developed and applied to analyze aristolochic acids (AA-I and AA-II) in Chinese medicinal herbs. The herb samples were extracted by using ultrasonication with the extraction efficiency of better than 82%. Extracts were then filtered and injected onto a C18 column eluting under a gradient program using methanol and water-containing 0.5% acetic acid. The method with the detection limits of 1.33 ng for AA-I and 7.29 ng for AA-II per injection was successfully applied for the analysis of traditional Chinese medicine (TCM) and related products and differentiation of Chinese medicinal herbs that have previously been misused and caused toxicological effects. The developed protocol provided an example that analysis of selected component markers could serve for health security and quality control of TCM consumption.  相似文献

In the search for novel natural products in plants, particularly those with potential bioactivity, it is important to efficiently distinguish novel compounds from previously isolated, known compounds, a process known as dereplication. In this study, electrospray ionization‐multiple stage tandem mass spectrometry (ESI‐MSn) was used to study the behaviour of 12 pregnane glycosides and genins previously isolated from Marsdenia tenacissima, a traditional Chinese medicinal plant, as a basis for dereplication of compounds in a plant extract. In addition to [M + Na]+ and [M + NH4]+ ions, a characteristic [M‐glycosyl + H]+ ion was observed in full‐scan mode with in‐source fragmentation. Sequential in‐trap collision‐induced dissociation of [M + Na]+ ions from 11,12‐diesters revealed consistent preferred losses of substituents first from C‐12, then from C‐11, followed by losses of monosaccharide fragments from the C‐3 tri‐ and tetrasaccharide substituents. A crude methanol extract of M. tenacissima stems was analysed using high‐performance liquid chromatography coupled to ESI‐MS. Several previously isolated pregnane glycosides were dereplicated, and the presence of an additional nine novel pregnane glycosides is predicted on the basis of the primary and fragment ions observed, including two with a previously unreported C4H7O C‐11/C‐12 substituent of pregnane glycosides. This study is the first report of prediction of the structures of novel pregnane glycosides in a crude plant extract by a combination of in‐source fragmentation and in‐trap collision‐induced dissociation and supports the usefulness of LC‐ESI‐MSn not only for dereplication of active compounds in extracts of medicinal plants but also for detecting the presence of novel related compounds. Conyza blinii Le'vl is a medicinal herb used for the treatment of inflammation in Chinese folk medicine. Its major bioactive constituents are triterpene saponins, most of which contain 6–8 sugar residues. In this report, electrospray ionization tandem mass spectrometry fragmentation behaviors of bisdesmosidic triterpene saponins (conyzasaponin A, B, and C) were studied in both positive and negative ion modes with an ion‐trap mass spectrometer. In full scan mass spectrometry, these saponins gave predominant [M–H]? and [M+Na]+ ions, which determined the molecular weights. In tandem mass spectrometry (MSn, n = 2–4), the [M–H]? and [M+Na]+ ions yielded fragments [Y–H]? and [Bα+Na]+, which were diagnostic for the structures of the triterpene skeleton and sugar chains. The structural elucidation was approved by accurate mass data using IT‐TOF‐MS. An interpretation guideline based on MSn (n = 2–4) diagnostic ions was proposed in order to elucidate the chemical structures of unknown triterpene saponins in C. blinii extract. The saponins in C. blinii were separated by liquid chromatography with a methanol/acetonitrile/water solvent system, and then analyzed by ion‐trap and IT‐TOF mass spectrometers. Based on the interpretation guideline, a total of 35 triterpenoid saponins were tentatively identified. Among them, 15 saponins had been previously reported, and the other 20 saponins were reported from Conyza species for the first time. Rapid, simple and reliable HPLC/DAD and LC‐ESI‐MS methods for the simultaneous determination of baicalin and forsythin in the traditional Chinese medicinal preparation Shuanghuanglian oral liquid were described and validated. The separation condition for HPLC/DAD was optimized using a BDS hypersil C18 column (Thermo, 2.1 × 150 mm, particle size 5 μm) by gradient elution using methanol‐0.2 % ammonium acetate as the mobile phase. The suitable detection wavelength was set at 277 nm for the quantitative analysis of baicalin and forsythin in this method. Some operational parameters of the ESI interface were optimized, negative m/z 445[M?H]? for baicalin and negative m/z 593[M+CH3COO]? for forsythin, positive m/z 447[M+H]+ for baicalin and positive m/z 552[M+NH 4]+ for forsythin, respectively. These HPLC/DAD and LC‐ESI‐MS methods were validated in terms of recovery, linearity, accuracy and precision (intra‐ and inter‐day validation). These methods can be used as a complementary method for the commercial quality control of Shuanghuanglian oral liquid and its pharmaceutical preparations.  相似文献

A novel calix[4]arene derivative containing benzothiazole group was coated on glassy carbon electrode (GCE) and then applied to the recognition of mercury ion. Cyclic and square wave voltammetric results showed that the modified electrode selectively recognizes Hg2+ ion in aqueous media. A new anodic stripping peak at −0.3 V (vs. Ag/Ag+) can be obtained by scanning the potential from −0.6 to 0.6 V, and the peak currents are proportional to the Hg2+ concentration. The modified electrode in a 0.1 M H2SO4+0.01 M NaCl solution shows linear voltammetric response in the range of 25-300 μg l−1 and detection limit of 5 μg l−1 (ca. 2.5×10−8 M). This modified GCE does not present any significant interference from alkali, alkaline and transition metal ions except for Pb2+, Ag+ and Cu2+ ions. Only 500, 50 and 100-fold molar excess of Pb2+, Ag+ and Cu2+ ions, respectively, can lead to voltammetric response comparable with that of Hg2+. The proposed method was successfully applied to determine mercury in natural water.  相似文献

A simple, rapid, selective and reproducible capillary electrophoresis (CE) method was firstly developed for the identification and determination of isofraxidin and fumaric acid in a traditional Chinese herb Sarcandra glabra and its medicinal preparations. The buffer solution used in this method was 7.5 mM NaH2PO4 and 7.5 mM borax solution adjusted to pH 8.60. The linear calibration range was 1.25-800 μg ml−1 (r=0.9997) for isofraxidin and 10-800 μg ml−1 (r=0.9992) for fumaric acid, respectively. Under the optimum conditions, the relative standard deviation (R.S.D.) values of the migration time and the peak area were 0.47, 3.45% for isofraxidin and 0.83, 3.24% for fumaric acid, respectively. The recoveries ranged between 95.4-103.8% for isofraxidin and 97.1-104.7% for fumaric acid, respectively. The contents of isofraxidin and fumaric acid in Sarcandra glabra and two kinds of Sarcandra glabra-containing Chinese medicinal preparations were successfully determined within 8 min.  相似文献

2,3‐Dimethyl‐2,3‐dinitrobutane (DMNB) is an explosive taggant added to plastic explosives during manufacture making them more susceptible to vapour‐phase detection systems. In this study, the formation and detection of gas‐phase [M+H]+, [M+Li]+, [M+NH4]+ and [M+Na]+ adducts of DMNB was achieved using electrospray ionisation on a triple quadrupole mass spectrometer. The [M+H]+ ion abundance was found to have a strong dependence on ion source temperature, decreasing markedly at source temperatures above 50°C. In contrast, the [M+Na]+ ion demonstrated increasing ion abundance at source temperatures up to 105°C. The relative susceptibility of DMNB adduct ions toward dissociation was investigated by collision‐induced dissociation. Probable structures of product ions and mechanisms for unimolecular dissociation have been inferred based on fragmentation patterns from tandem mass (MS/MS) spectra of source‐formed ions of normal and isotopically labelled DMNB, and quantum chemical calculations. Both thermal and collisional activation studies suggest that the [M+Na]+ adduct ions are significantly more stable toward dissociation than their protonated analogues and, as a consequence, the former provide attractive targets for detection by contemporary rapid screening methods such as desorption electrospray ionisation mass spectrometry. In this study, a rapid MEKC method using 40 mM sodium borate buffer containing 50 mM SDS as surfactant was developed for the analysis of aristolochic acid (AA) in Aristolochia plants. Baseline separation of AA-I and AA-II was achieved within 3 min with high separation efficiency, satisfactory sensitivity, repeatability, and recovery. Resolution between AA-I and AA-II is above 5 and great performance with higher than 200,000 theoretical plate numbers was obtained. The detection limits (based on 3 S/N) were both 1.0 microg/mL. Two kinds of AA in 35 herbal samples of Aristolochia plants were successfully determined. The competition mechanism between beta-CD and SDS was also investigated by changing the content ratio of beta-CD and SDS.  相似文献

Spin‐labeled nitroxide derivatives of podophyllotoxin had better antitumor activity and less toxicity than that of the parent compounds. However, the 2‐H configurations of these spin‐labeled derivatives cannot be determined by nuclear magnetic resonance (NMR) methods. In the present paper, a high‐performance liquid chromatography‐diode array detection (HPLC‐DAD) and a high‐performance liquid chromatography‐electrospray ionization tandem mass spectrometry (HPLC‐ESI/MS/MS) method were developed and validated for the separation, identification of four pairs of diastereoisomers of spin‐labeled derivatives of podophyllotoxin at C‐2 position. In the HPLC‐ESI/MS spectra, each pair of diastereoisomers of the spin‐labeled derivatives in the mixture was directly confirmed and identified by [M+H]+ ions and ion ratios of relative abundance of [M‐ROH+H]+ (ion 397) to [M+H]+. When the [M‐ROH+H]+ ions (at m/z 397) were selected as the precursor ions to perform the MS/MS product ion scan. The product ions at m/z 313, 282, and 229 were the common diagnostic ions. The ion ratios of relative abundance of the [M‐ROH+H]+ (ion 397) to [M+H]+, [A+H]+ (ion 313) to [M‐ROH+H]+, [A+H‐OCH3]+ (ion 282) to [M‐ROH+H]+ and [M‐ROH‐ArH+H]+ (ion 229) to [M‐ROH+H]+ of each pair of diastereoisomers of the derivatives specifically exhibited a stereochemical effect. Thus, by using identical chromatographic conditions, the combination of DAD and MS/MS data permitted the separation and identification of the four pairs of diastereoisomers of spin‐labeled derivatives of podophyllotoxin at C‐2 in the mixture.  相似文献

We have demonstrated the analysis of aristolochic acids (AAs) that are naturally occurring nephrotoxin and carcinogen by capillary electrophoresis in conjunction with laser-induced fluorescence detection (CE-LIF). Owing to lack of intrinsic fluorescence characteristics of oxidized AAs (OAAs), reduction of the analytes by iron powder in 10.0 mM HCl is required prior to CE analysis. The reduced AAs (RAAs) exhibit fluorescence at 477 nm when excited at 405 nm using a solid-state blue laser. By using 50.0 mM sodium tetraborate (pH 9.0) containing 10.0 mM SDS, the determination of AA-I and AA-II by CE-LIF has been achieved within 12 min. The CE-LIF provides the LODs of 8.2 and 5.4 nM for AA-I and AA-II, respectively. The simple CE-LIF method has been validated by the analysis of 61 Chinese herbal samples. Prior to CE analysis, OAAs were extracted by using 5.0 mL MeOH, and then the extracts were subjected to centrifugation at 3,000 rpm for 5 min. After reduction, extraction, and centrifugation, the supernatants were collected and subjected to CE analysis. Of the 61 samples, 14 samples contain AA-I and AA-II, as well as 10 samples contain either AAI or AAII. The relative standard deviation (RSD) values of the migration times for AA-I and AA-II are less than 2.5% and 2.1% for three consecutive measurements of each sample. The RSD values for the peak heights corresponding to AA-I and AA-II in most samples are about 8.0% and 10.0%, respectively. The result shows that the present CE-LIF approach is sensitive, simple, efficient, and accurate for the determination of AAs in real samples.  相似文献

H-point standard addition method (HPSAM) is suggested as a simple and selective method for the determination of semicarbazide and hydrazine. The reduction of Cu2+ to Cu+ by semicarbazide and hydrazine in the presence of neocuproine (Nc) and the subsequent complex formation between Cu+ and Nc produced a sensitive spectrophotometric method for indirect determination of semicarbazide and hydrazine. The difference in the rate of reduction of Cu2+ with semicarbazide and hydrazine in cationic micellar media is the basis of this method. Semicarbazide can be determined in the range of 0.5-3.75 μg ml−1 with satisfactory accuracy and precision in the presence of excess hydrazine. The proposed method was successfully applied to the simultaneous determination of semicarbazide (0.5-3.75 μg ml−1) and hydrazine (0.5-5 μg ml−1) and also to the selective determination of semicarbazide in the presence of hydrazine in several synthetic mixtures containing different concentration ratios of semicarbazide and hydrazine.  相似文献

The relationships between the ionization profile, sensitivity, and structures of 64 exogenous anabolic steroids (groups I–IV) was investigated under electrospray ionization (ESI) conditions. The target analytes were ionized as [M + H]+ or [M + H–nH2O]+ in the positive mode, and these ions were used as precursor ions for selected reaction monitoring analysis. The collision energy and Q3 ions were optimized based on the sensitivity and selectivity. The limits of detection (LODs) were 0.05–20 ng/mL for the 64 steroids. The LODs for 38 compounds, 14 compounds and 12 compounds were in the range of 0.05–1, 2–5 and 10–20 ng/mL, respectively. Steroids including the conjugated keto‐functional group at C3 showed good proton affinity and stability, and generated the [M + H]+ ion as the most abundant precursor ion. In addition, the LODs of steroids using the [M + H]+ ion as the precursor ion were mostly distributed at low concentrations. In contrast, steroids containing conjugated/unconjugated hydroxyl functional groups at C3 generated [M + H ? H2O]+ or [M + H ? 2H2O]+ ions, and these steroids showed relatively high LODs owing to poor stability and multiple ion formation. An LC‐MS/MS method based on the present ionization profile was developed and validated for the determination of 78 steroids (groups I–V) in human urine. Potentiometric thiocyanate-selective sensors based on the use of three synthesized di-, tetra-, and hexa-imidepyridine derivatives as novel anionic neutral ionophores in plasticized poly(vinyl chloride) (PVC) membranes are described. The sensors exhibit significantly enhanced response towards thiocyanate ions over the concentration range 5×10−6 to 1.0×10−2 M with a lower detection limit of 0.3 μg ml−1 and slopes ranging from −55.6 to −58.3 mV per decade. Fast and stable response, good reproducibility, long-term stability, applicability over a wide pH range (2-8) and high selectivity for SCN ion in the presence of 18 common anions are demonstrated. The sensors are used for direct potentiometric measurements of thiocyanate ions over the concentration range 0.2-580 μg ml−1 and for monitoring sequential titration of some metal ions (e.g. Ag+, Tl+, Cu2+, Pb2+) in binary and ternary mixtures. Sequential binding of these metal ions with SCN ensures share stepwise titration curves with consecutive end point breaks at the equivalent points. Recoveries of 98.5-99.1±0.3% are obtained for metal ion concentrations of 0.06-4 mg ml−1.  相似文献

Low molecular weight polyisobutylenes (PIB) with chlorine, olefin and succinic acid end‐groups were studied using direct analysis in real time mass spectrometry (DART‐MS). To facilitate the adduct ion formation under DART conditions, NH4Cl as an auxiliary reagent was deposited onto the PIB surface. It was found that chlorinated adduct ions of olefin and chlorine telechelic PIBs, i.e. [M + Cl]? up to m/z 1100, and the deprotonated polyisobutylene succinic acid [M? H]? were formed as observed in the negative ion mode. In the positive ion mode formation of [M + NH4]+, adduct ions were detected. In the tandem mass (MS/MS) spectra of [M + Cl]?, product ions were absent, suggesting a simple dissociation of the precursor [M + Cl]? into a Cl? ion and a neutral M without fragmentation of the PIB backbones. However, structurally important product ions were produced from the corresponding [M + NH4]+ ions, allowing us to obtain valuable information on the arm‐length distributions of the PIBs containing aromatic initiator moiety. In addition, a model was developed to interpret the oligomer distributions and the number average molecular weights observed in DART‐MS for PIBs and other polymers of low molecular weight. A chemiluminescence (CL) method for the determination of barbituric acid (BA) was proposed, which is based on the enhancement of BA to the CL intensity of Tris-(1,10-phenanthroline)ruthenium(II) (Ru(phen)32+)-cerium(IV) (Ce(IV)) system. The concentration of BA is proportional to the CL intensity in the range of 5.0×10−3-2.0 μg ml−1. The detection limit is 6.9×10−4 μg ml−1. The relative standard deviation (R.S.D.) of determining 11 samples containing 0.20 μg ml−1 BA is 3.2%. This CL method has been successfully applied to the determination of BA in the synthetic samples. The mechanism of CL reaction was studied.  相似文献

Unexpected [M + 15]+ ions were formed during the analysis of aromatic aldehydes by use of methanol in positive‐ion electrospray ionization mass spectrometry. Aromatic aldehydes with electron‐withdrawing groups or electron‐donating groups were all tested to make sure the universality. All the aromatic aldehydes studied with methanol as the solvent could generate [M + 15]+ ion, and for most of them, the [M + 15]+ ion was more intense than the [M + H]+ ion. Deuterium‐labeling experiment, high‐performance liquid chromatography–MS experiment, collision‐induced dissociation experiment, and theoretical calculations were performed to identify the formation of [M + 15]+ ion. The proposed reaction mechanism is a gas‐phase aldol reaction between protonated aromatic aldehydes and methanol occurring in electrospray source. Understanding and using this unique gas‐phase ion/molecule reaction can indeed offer a novel and fast approach for the direct identification of aromatic aldehydes. A sensitive and reliable HPLC coupled with diode array detection and MS method was developed and validated for the first time to simultaneously identify and quantify eight characteristic 5,6,7,8‐tetrahydro‐2‐(2‐phenylethyl)chromones (THPECs) in Chinese eaglewood. Chromatographic separation was performed on a Zorbax SB C18 column with a gradient of acetonitrile/0.1% formic acid/water as the mobile phase. The MS fragmentation behavior of THPECs was characterized as the successive neutral loss of two molecules of H2O ([M+H–18–18]+) and then two molecules of CO ([M+H–18–18–28–28]+), which could be used to differentiate Chinese eaglewood from counterfeits. Validation of the developed analytical method showed good linearity, satisfactory precision, and good recovery. The established method was successfully applied to the simultaneous determination of eight THPECs in ten batches of Chinese eaglewood, which could be used as a tool for the quality control of Chinese eaglewood.  相似文献

An HPLC method was developed for the simultaneous determination of five aristolochic acids (AAs) and two aristololactams (ALs) in the following six Chinese drugs derived from Aristolochia species. Samples were analyzed on a C(18) column with acetonitrile and 3.7 mm phosphoric acid buffer gradient elution, detected at 260 nm. Assay was linear over the range (microg/mL) 0.386-38.6 for aristolochic acid Va, 0.632-63.2 for aristolochic acid IVa, 0.200-20.0 for 9-hydroxy aristolochic acid I, 0.352-35.2 for aristololactam II, 0.296-29.6 for aristolochic acid II, 0.274-27.4 for aristololactam I and 3.12-312 for aristolochic acid I. Average recoveries (%) of samples were 102.0, 95.9, 99.2, 102.2, 97.2, 97.1 and 97.8 for these seven constituents, respectively. The detection limit and retention time for the seven constituents ranged from 10.0 to 15.8 ng/mL and from 12 to 21 min. As a result of drug determination, contents (in mg/g) were as follows: AA-I, 0.69-1.77; AA-II, 0.02-0.18; 9-OH AA-I, 0.04-0.12; AA-IVa, 0.76-3.36; AA-Va, 0.04-0.31; AL-I, 0.07-0.36; and AL-II, 0.01-0.09 in Madouling; AA-I, 0.03-0.41; AA-II, 0.01-0.11; 9-OH AA-I, 0.00-0.60; AA-IVa, 0.00-0.77; AA-Va, 0.00-0.14; and AL-I, 0.00-0.04 in Tianxianteng; AA-I, 1.19-4.71; and AA-II, 0.24-1.69 in Qingmuxiang; AA-I, 2.79-5.48; AA-II, 1.06-1.86; 9-OH AA-I, 0.01-0.09; AA-IVa, 0.38-0.69; AA-Va, 0.00-0.61; AL-I, 0.00-0.02; and AL-II, 0.00-0.02 in Bei-madouling-gen; AA-I, 0.64-4.23; AA-II, 0.06-0.40; and AA-IVa, 0.08-0.25; in Guangfangji; and AA-I, 1.88-9.72; AA-II, 0.26-1.88; and AA-IVa, 0.09-0.52 in Guanmutong. The other constituents were not detected in Tianxianteng, Qingmuxiang, Guangfangji and Guanmutong.  相似文献

The thermospray mass spectrometry (TSP/MS) of five N-methylcarbamates is presented. This is the first time that ions other than [M + H]+ and [M + NH4]+ have been reported using positive TSP/MS. Protonation of ROCONHCH3 yields the [CH3NH2CO] ion, with formation of the ion–molecule adduct [ROCONHCH3 · CH3NH2CO] through elimination of CO from [CH3NH2CO], and the adduct [M + 75], [ROCONHCH3 · OCONH2CH3], is also obtained.  相似文献
