12‐Chloro‐12‐n‐butyl‐1,11‐dioxa‐4,8‐ dithia‐12‐stannacyclododecane ( 3a ) and 12‐chloro‐ 12‐n‐butyl‐1,4,8,11‐tetrathia‐12‐stannacyclododecane ( 3b ) have been prepared by reacting n‐butyltin trichloride with 1,11‐dioxa‐4,8‐dithiaundecane and 1,4,8,11‐tetrathiaundecane, respectively. Complexes 3a,b were characterized by elemental analyses, IR, electron impact mass spectrometry, and multinuclear NMR (1H, 13C, and 119Sn). The spectroscopic data are consistent with bonding of the ligands through both sulfur and oxygen atoms in 3a and through all sulfur atoms in 3b to the Sn(IV) center. Deoxycholic acid (DCA) is an endogenous secondary bile acid implicated in numerous pathological conditions including colon cancer formation and progression and cholestatic liver disease. DCA involvement in these disease processes results partly from its ability to modulate signaling cascades within the cell, presumably through both direct receptor activation and general detergent mediated membrane changes. To further explore DCA induced changes in cell signaling, we completed a total synthesis of enantiomeric deoxycholic acid (ent-DCA) from achiral 2-methyl-1,3-cyclopentanedione. Using a modified method of the synthesis of ent-testosterone that proceeds through the (R)-(-)-Hajos-Parrish ketone, we have completed the successful synthesis of ent-DCA in 25 steps with a yield of 0.3% with all stereochemical assignments of the product confirmed by X-ray crystallography. Our studies toward this synthesis also uncovered the methodology for the development of a novel A,B-cis steroidal skeleton system containing a C3-C9 single bond as well as conditions to selectively ketalize the typically less reactive 12-carbonyl in poly-keto A,B-cis androgens. The critical micelle concentration (cmc) of ent-DCA, determined by a dye solubilization method, was identical to the cmc of natural DCA. Toxicity studies toward HT-29 and HCT-116 human colon cancer cell lines demonstrated that ent-DCA had similar effects on proliferation, yet showed a markedly decreased ability to induce apoptosis as compared to natural DCA.  相似文献

A general method for preparation of ferrocene‐containing monoamines (5–7) and diamines (10, 11) starting from the corresponding quaternary ammonium iodide 3 and ferrocene mono‐ (4) and dithiaaliphatic acids (8, 9) was developed. Amines obtained have been characterized as acet‐ and benzamides (12–15). The oligoamide precursors (16, 17, 22, 23) were synthesized by reactions of succinic or glutaric anhydride with amines (6, 7, 10, 11). Their conversion into oligoamide analogs (20, 21, 25) failed. The desired diamides (20, 21) were prepared by condensation of amines (6, 7) with alkanedioyl chlorides, (CH2)n(COCl)2 (n = 0, 1, 2, 3). Reactions of diamine 10 with succinic or glutaric anhydride gave amino acids 28—formal monomers for the planned oligomerization. Oligomers 29 were synthesized by condensation of equimolar amounts of diamines 10 and the above mentioned alkanedioyl chlorides in dichloromethane at 0°C. The structure of oligomers 29 was indicated from their IR and 1H‐NMR spectra in comparison with the model substances 12–28. Phosphonate‐functionalized polysiloxanes have been prepared with a new siloxane/phosphonate monomer. The reaction of 3‐chloropropylmethyldimethoxysilane with trimethylphosphite or triethylphosphite produces several new monomers containing pendant phosphonate groups. Copolymerization with dimethyldimethoxysilane has produced polymers soluble in most organic solvents. The acid hydrolysis of the phosphoryl esters has produced hydrophilic siloxane polymers containing phosphonic acid groups. The thermal properties of the polymers and several related small molecules have been compared with thermogravimetric analysis. Three new flavonoid glycosides, together with 15 known flavonoids, have been isolated from the leaves of Eriobotrya japonica, and characterized as (2S)- and (2R)-naringenin 8-C-alpha-L-rhamnopyranosyl-(1-->2)-beta-D-glucopyranosides, and cinchonain Id 7-O-beta-D-glucopyranoside, respectively, based on spectral analyses including two dimensional (2D) NMR techniques. Higher proanthocyanidin fraction in the water-soluble portion of the extract was characterized as a procyanidin oligomer mixture mainly composed of undecameric procyanidin. These polyphenols have also been assessed for cytotoxic activity against two human oral tumor (human squamous cell carcinoma and human salivary gland tumor) cell lines. Selective cytotoxicity of the procyanidin oligomer between tumor and normal gingival fibroblast cells, and its possible mechanism, were also described.  相似文献

New hydroselenites of the different silylalkylthio‐substituted N‐heterocycles have been prepared by the reaction of selenium dioxide with N‐heterocycles in an aqueous medium. Their structure was confirmed by 1H, 13C, and 77Se NMR data. Most of these silylalkylthio‐substituted N‐heterocycles and their hydroselenites have an expressed cytotoxic activity on the MG‐22A (mouse hepatoma), HT‐1080 (human fibrosarcoma), B16 (mouse melanoma), and Neuro 2A (mouse neuroblastoma) cell lines. Some of the hydroselenites exhibit free‐radical protection simultaneously with a high cytotoxic effect. The substances studied were also active in vivoagainst sarcoma S‐180. Uterine (cervix and corpus) cancer is one of the major causes of mortality in women in Mexico. Organotin carboxylated derivatives have shown high cytotoxic activity against various cell lines of human origin. We describe the synthesis of three new tri‐n‐butyltin derivatives from 4‐oxo‐4‐(arylamino)butanoic acids; their structures were confirmed using spectral data (1H NMR, 13C NMR, 119Sn NMR and infrared), elemental analyses, mass spectrometry and X‐ray diffraction. All the tri‐n‐butyltin carboxylates exhibit 1 J (119/117Sn–13C) coupling satellites in solution and lie in the range 357 to 339 Hz, suggesting a tetrahedral geometry around the tin atom. The polymeric structures of two of the derivatives and the monomeric structure of another were confirmed using X‐ray crystallography. Using succinic anhydride as raw material, five N‐substituted succinamic acid compounds were synthesized by the acylation reaction with aniline, 4‐nitroaniline, 4‐nitro‐3‐(trifluoromethyl)aniline, 2‐amino‐5‐nitrothiazole and 4‐aminoantipyrine. From these compounds, five tin derivatives were prepared and their in vitro anti‐proliferative effect on HeLa, CaSki and ViBo cell lines was screened. All of the compounds showed potency against all three strains and null or low cytotoxic activity (necrotic) as well. The most potent of our derivatives as an anti‐proliferative agent against the three cell lines was tributylstannyl 4‐oxo‐4‐[(3‐trifluoromethyl‐4‐nitrophen‐1‐yl)amino]butanoate, exhibiting an IC50 value of 0.43 μM against the HeLa cell line. Two new tripodal compounds - 4-{bis[(3,5-dimethyl-1H-pyrazole-1-yl)methyl]amino}butane-1-ol (1); ethyl 1-[((2-hydroxyethyl){[3-(ethoxycarbonyl)-5-methyl-1H-pyrazole-1-yl]methyl} amino)methyl]-5-methyl-1H-pyrazole-3-carboxylate (2) were reported. The evaluation of the cytotoxic properties in vitro of these ligands, was examined on two tumor cell lines - P815 (mastocytome murine) and Hep (carcinoma of human larynx). The concentration required to induce 50% of lysis (IC(50)) was more pronounced against P815 cell line (IC(50): 39.42 microg mL(-1) for the compound 1 and 97.74 microg mL(-1) for the compound 2) than the Hep cell line (IC(50): 83.49 microg mL(-1) for compound 1 and 185.30 microg mL(-1) for compound 2). Statistical analysis shows that the compound 1 is two to three folds more cytotoxic than the compound 2 (p < 0.05). Interestingly, the cytotoxic activity depends strongly on both the substituents linked to the aminic nitrogen and pyrazolic rings.  相似文献

A variety of polycyclic aromatic hydrocarbons have been synthesized and structurally characterized in our laboratory. Phenanthrene derivatives were efficiently prepared in excellent yields and high purity via a two-step sequence. Heck coupling yielded the corresponding diarylethenes, followed by classical oxidative photocyclization to achieve the expected phenanthrenes. First, we envisioned to synthesize a variety of substituted phenanthrenequinones. Second, we investigated the possibility of a dibenz[a,c]phenazine formation by addition of o-phenylenediamine after completion of the oxidation process. Moreover, because phenanthrenequinones are available so simply, it is likely that other uses will be found for these compounds. For example, 9,10-phenanthrenequinone can be sequentially reduced, alkylated, acetylated, and sulfonated. All the synthesized derivatives were evaluated for cytotoxic activity in vitro against the human epidermoid carcinoma epithelial cells Hep-2 and human colon carcinoma cells Caco-2 using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. From the structure–activity point of view, position and nature of the electron donating and electron withdrawing functional groups attached to the phenanthrene skeleton may contribute to the anticancer action. Interestingly, the analysis of the IC50 values suggests that most compounds exerted cytotoxic effects with selectivity against both cancer cells. Among them, methyl 8-methyl-9,10-phenanthrenequinone-3-carboxylate 11d showed the highest potency with IC50 values of 2.81 and 0.97 μg/mL.  相似文献

We have prepared four light‐emitting polymers bearing a chromophore composed of carbazole and fluorene by the Suzuki coupling polycondensation. Two nonconjugated polymers (P3CzBFXy and P2CzFXy) had a chromophore tethered by the p‐xylylene spacer, whose connection point between carbazole and fluorene in addition to the number of fluorene unit was systematically changed to investigate the emission wavelength and intensity. The red‐shifted absorption and emission maximum wavelengths together with the improved fluorescence quantum yield of polymers P3CzBFXy and P2CzFXy indicate that the increment of the number of para‐connected benzene rings included in the chromophore effectively extends the conjugation length. The fact that polymer P3CzBFXy has longer wavelength absorption and emission spectra also indicates the interaction of the carbazole nitrogen lone pair with the oligophenylene moiety. Other two polymers P3CzFPy and P3CzFPym having the heterocycle directly bound to the carbazole nitrogen were prepared to know the character of the carbazole nitrogen lone pair and their influence on the fluorescence behavior. The fluorescence spectra of polymer P3CzFPym bearing the pyrimidine ring gradually red‐shifted in conjunction with the decrease of fluorescence quantum yield on going from toluene solution to CHCl3 solution because of the intramolecular charge transfer at the excited state. © 2010 Wiley Periodicals, Inc. Some new amino‐ and imide‐containing phthalonitrile compounds with 1:1 molar ratio of amino group to pthalonitrile unit were successfully synthesized. The molecular structures were characterized by spectroscopic techniques. They were thermally polymerized under nitrogen/air, even in the absence of curing additives. The thermal properties of the cured products were characterized by thermogravimetric analysis and differential scanning calorimetry. The 5% weight loss temperatures ranged from 525 to 528 °C and 513 to 520 °C under nitrogen and air, respectively. Char yields (900 °C) were in the range of 62–70%. Rheometric measurements showed that the rate of the cure reaction differs for all the three monomers. Three novel compounds, (η6‐p‐cymene)RuCl2(2‐fluoro‐5‐aminopyridine) (compound 1), (η6‐p‐cymene)RuCl2(5‐amino‐2‐chlorpyridine) (compound 2) and (η6‐p‐cymene)RuCl2(2‐bromo‐ 5‐aminopyridine) (compound 3), were synthesized and characterized. The compound 1 and 3 were determined by X‐ray diffraction, showing a distorted piano‐stool type of geometry with similar bond lengths and angles around the ruthenium. Compound 2 exhibited moderate in vitro activity against A549 and MCF‐7 human cancer cells, the other two lower activities. The UV–vis and fluorescent absorption titrations showed that three compounds binded with CT‐DNA in a minor groove. The intrinsic binding constants (Kb) were calculated to be 2.13(±0.03) × 105 M?1, 2.89(±0.03) × 105 M?1 and 2.45(±0.03) × 105 M?1 for compound 1, 2 and 3, respectively, by using UV–vis absorption titrations data. Among the three compound, the highest value of intrinsic binding constant of compound 2 was consistent with its highest cytoxicity against A549 and MCF‐7 human cancer cells in vitro.  相似文献

Sulfonamide‐derived compounds and their first row d‐transition metal chelates [cobalt(II), copper(II), nickel(II) and zinc(II)] have been synthesized and characterized. The nature of bonding and structure of all the synthesized compounds have been proposed from magnetic susceptibility and conductivity measurements, IR, 1H and 13C NMR, electron spectra, mass spectrometry and CHN analysis data. The structure of ligand, 4‐{[(E)‐(5‐chloro‐2‐hydroxyphenyl) methylidene] amino}‐N‐(4,6‐dimethyl pyrimidin‐2‐yl) benzene sulfonamide has also been determined by X‐ray diffraction method. An octahedral geometry has been suggested for all the complexes. The ligands and metal complexes have been screened for their in vitro antibacterial, antifungal and cytotoxic activity. The results of these studies revealed that all compounds showed moderate to significant antibacterial activity against one or more bacterial strains and good antifungal activity against various fungal strains. New acyclic, macrocyclic and macrobicyclic compounds containing one or two proton‐ionizable triazole groups are prepared and characterized. The series includes six podands, a macrocycle with one triazole and one pyridine unit in the ring, a bis‐triazolo macrocycle with four pentafluorobenzyl substitutents, and two bis(crown ethers) with a triazolo group connecting the two polyether rings. The solid‐state structure and solubility in supercritical carbon dioxide are determined for the bis‐triazolo macrocycle with pendant pentafluorobenzyl groups.  相似文献

Coumarins are used as a natural renewable resource to synthesize coumarin‐containing benzoxazine resins. The coumarin‐containing benzoxazines are fully characterized in terms of their chemical structure by Fourier‐transform infrared spectroscopy and proton nuclear magnetic resonance spectroscopy. The influence of electronic effects caused by the substituents on the polymerization temperature is also evaluated. Thermal properties of the resulting thermosets are characterized by differential scanning calorimetry and thermogravimetric analysis, showing good stability and char yields higher than 50%. The coumarin‐containing polybenzoxazine thermosets show Tg values in the range between 160 and 190 °C. Thus, the herein presented coumarin‐containing benzoxazine resins are proven to be competitive monomers when compared with other petroleum‐based benzoxazine resins toward the generation of high‐performance thermoset. © 2015 Wiley Periodicals, Inc. PEGylated liposomes encapsulating human hemoglobin as oxygen carriers were prepared from purified carbonylhemoglobin (HbCO) solution and a lipid mixture composed of 1,2-dipalmitoyl-sn-glycero-3-phosphatidylcholine (DPPC), cholesterol, 1,2-dimyristoyl-sn-glycero-3-phosphoethanolamine-N-[poly(ethylene glycol) 2000] (DMPE-PEG(2000)) and palmitic acid. Hemoglobin was extracted and purified from human blood samples. SDS-PAGE was used to assess its purity. Diameter of liposomes containing hemoglobin was controlled to approximately 200 nm using extrusion as measured by dynamic light scattering and transmission electron microscopy. Liposome size distributions were shown to remain unimodal over 14 days, even at different storage temperatures. Zeta potential measurements revealed that liposome containing hemoglobin have a net surface charge of -7.16+/-0.33 mV. Also, hemoglobin encapsulated in liposomes was able to perform several cycles of oxygen loading and unloading using oxygen (O(2)) and carbon monoxide (CO). The hemoglobin vesicle dispersion showed some toxicity as revealed by three in vitro assays in which endothelial cell (HUVECs) monolayers were exposed to these dispersions. Cytotoxicity was function of the liposome concentration in the culture medium.  相似文献

New borazine compounds containing steroid units (cholesteryl and stigmasteryl) were prepared by the reactions of B,B′,B″‐trichloro‐N,N′,N″‐trimethylborazine with the 3‐chloro derivatives of the corresponding steroids under Grignard conditions. Abstract

N-heterocyclic carbene (compound b) was synthesized from reaction of 1-(3-phenylpropyl)-3-methylbenzimidazolium iodide (a) and a strong base in an argon atmosphere. Then, 1-(3-phenylpropyl)-3-methylbenzimidazolium-2-carbodithioate (c) was prepared with carbene and CS2. Finally, a group of alkyl 1-(3-phenylpropyl)-3-methylbenzimidazolium-2-carbodithioate halides was synthesized with alkyl halides and compound c, occurring products 1-6. Structural characterization of compounds was performed 1H, and 13C NMR, IR, and MS spectroscopy and elemental analysis.  相似文献

Three novel copper(II) complexes, [Cu(Gly‐l ‐Val)(HPBM)(H2O)]·ClO4·H2O ( 1 ), [Cu(Gly‐l ‐Val)(TBZ)(H2O)]·ClO4 ( 2 ) and [Cu(Gly‐l ‐Val)(PBO)(H2O)]·ClO4 ( 3 ) (Gly‐l ‐Val = glycyl‐l ‐valine anion, HPBM = 5‐methyl‐2‐(2′‐pyridyl)benzimidazole, TBZ = 2‐(4′‐thiazolyl)benzimidazole, PBO = 2‐(2′‐pyridyl)benzoxazole), have been prepared and characterized with elemental analyses, conductivity measurements as well as various spectroscopic techniques. The interactions of these copper complexes with calf thymus DNA were explored using UV–visible, fluorescence, circular dichroism, thermal denaturation, viscosity and docking analyses methods. The experimental results showed that all three complexes could bind to DNA via an intercalative mode. Moreover, the cytotoxic effects were evaluated using the MTT method, and the antimicrobial activity of these complexes was tested against Bacillus subtilis, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. The results showed that the activities are consistent with their DNA binding abilities, following the order of 1 > 2 > 3 .  相似文献
