In surface‐enhanced Raman scattering (SERS), the scattered intensity is drastically increased due to a resonant interaction with surface plasmons of coin metals. SERS is a nondestructive spectroscopic method applied also to biomedical samples. It inherits the advantages of normal Raman spectroscopy and at the same time overcomes the inherent low sensitivity problem. These properties endow SERS with exciting opportunities to be a successful analytical tool for cell analysis. SERS can be used to detect only molecules located on or close to the metallic nanostructures which can support surface plasmon resonances for the enhancement of the Raman signals. Therefore, these metallic nanostructures play a key role in the application of SERS in cell analysis. By incorporating the SERS substrates into the biosamples, molecular structural probing and cellular imaging become possible. In the past decade, analysts worldwide have developed many schemes to study the chemical changes and component distribution in cells by using SERS. There is an increasing interest in developing surface enhancement Raman spectroscopy methods for intracellular biomolecule and for in vitro protein detection that involve dye or protein–dye conjugates. In this work, we have demonstrated that protein adsorption on silver nanoparticle (AgNP) can significantly attenuate the surface‐enhanced Raman spectroscopy (SERS) signal of dye molecules in both protein/dye mixtures and protein/dye conjugates. SERS spectra of 12 protein/dye mixtures were acquired using 4 proteins [bovine serum albumin (BSA), lysozyme, trypsin, and concanavalin A] and three dyes [Rhodamine 6G, adenine, and fluorescein isothiocyanate (FITC)]. Besides the protein/dye mixtures, spectra were also obtained for the free dyes and four FITC‐conjugated proteins. While no SERS signal was observed in protein/FITC mixtures or conjugates, a significantly reduced SERS intensity (up to 3 orders of magnitude) was observed for both R6G and adenine in their respective protein mixtures. Quantitative estimation of the number of dye molecules absorbed onto AgNP implied that the degree of R6G SERS signal reduction in the R6G/BSA sample is 2 to 3 orders of magnitude higher than what could be accounted for by the difference in the amount of the absorbed dyes. This finding has significant implications for both intracellular SERS analyses and in vitro protein detection using SERS tagging strategies that rely on Raman dyes as reporter molecules. In order to get insight into the chemical heterogeneities of solid tumors, here we report the first surface‐enhanced Raman scattering (SERS) experiment from normal and altered epithelial layer in human colon carcinoma tissues. The Ag colloidal nanoparticles that can be incorporated into the interstitial space in solid tumors or those penetrating into cytoplasm or nucleus of many cells allowed high quality SERS signal. Different tissue structures of tumor and normal colon have characteristic features in SERS spectra. Prominent SERS features of malignant tissue spectra are related to the strong enhancement of the bands preponderantly attributable to DNA or RNA bases. The preliminary studies demonstrate that it is possible to probe Ag colloidal nanoparticles adsorption onto the tissue resulting in a strong molecular signaling with high specificity and rapid acquisition time using visible laser line excitation. The Raman and surface enhanced Raman scattering (SERS) spectra of a black dyed silk sample (BDS) were registered. The spectral analysis was performed on the basis of Raman and SERS spectral data of isolated samples of Bombyx mori silk fibroin, its motif peptide component (GAGAGS) and the synthetic reactive black 5 dye (RB5). The macro FT‐Raman spectrum of the silk sample is consistent with a silk II‐Cp crystalline fraction of Bombyx mori silk fibroin; the SERS spectrum is highly consistent with conformational modifications of the fibroin due to the interactions with the Ag nanoparticles. The GAGAGS peptide sequence dominates the Raman spectrum of the silk. The SERS spectrum of the peptide suggests a random coil conformation imposed by the surface interaction; the serine residue in the new conformation is exposed to the surface. Quantum chemical calculations for a model of the GAGAGS–Ag surface predict a nearly extended conformation at the Ag surface. The Raman spectrum of the dye was analysed, and a complete band assignment was proposed; it was not possible to propose a preferential orientation or organization of the molecule on the metal surface. Quantum chemical calculations for a model of the dye interacting with a silver surface predict a rather coplanar orientation of the RB5 on the Ag metal surface. The Raman spectrum of the BDS sample is dominated by signals from the dye; the general spectral behaviour indicates that the dye mainly interacts with the silk through the sulphone (–SO2–) and sulphonate (–SO2–O–) groups. Besides the presence of dye signals, mainly ascribed to the sulphone and sulphonate bands, the SERS spectrum of the BDS sample also displays bands belonging to the amino acids alanine, glycine, serine and particularly tyrosine. Surface‐enhanced Raman scattering (SERS) spectra of hydroxyproline and one deuterated analogue are reported. In this work, we tackled the problem of SERS reproducibility by employing gold colloids instead of the usual silver sols to achieve plasmon enhanced Raman scattering. We slightly modified modified a previously published procedure to obtain to obtain the colloid, and concentrated the gold particles by centrifugation. The SERS spectra show distinctive bands of hydroxyproline, assigned by comparison to normal Raman spectra and density functional theory calculations. We report surface‐enhanced Raman studies on intact plant material using onion layers as a biological target, and silver nanoaggregates and silver island films as enhancing plasmonic structures. Surface‐enhanced Raman scattering (SERS) enhancement allows the detection of strong Raman signatures of chemical constituents of the surface of the onion layer such as cellulose, proteins, and flavonols. Because of long‐time incubation, SERS sensors can access the extracellular space in the inner of the layer. The location of silver nanoparticles inside the onion layer has been monitored by the SERS images collected from chemicals present in the onion and/or reporter molecules attached to the nanoparticles. Our studies show a competitive adsorption of intrinsic bio molecules of the onion layer and reporter molecules. Different spectra from different places of the layer indicate the complex heterogeneous chemical structure of the plant material. The pH‐sensitive reporter molecule para mercapto benzoic acid attached to the nanoparticles allows us to infer pH values inside the extracellular matrix of the onion layer. Three‐dimensional (3D) nanoporous gallium nitride (PGaN) scaffolds are fabricated by Pt‐assisted electroless hydrofluoric acid (HF) etching of crystalline GaN followed by in situ electroless deposition of Ag nanostructures onto the interior surfaces of the nanopores, yielding a large surface area substrate for surface‐enhanced Raman scattering (SERS). The resulting 3D SERS‐active substrates have been optimized by varying reaction parameters and starting material concentration, exhibiting enhanced Raman signals 10–100× more intense than either (1) sputtered Ag‐coated porous GaN or (2) Ag‐coated planar GaN. The increase in SERS signal is attributed to a combination of the large surface area and the inherent transparency of PGaN in the visible spectral region. Overall, Ag‐decorated PGaN is a promising platform for high sensitivity SERS detection and chemical analysis, particularly for reaction and metabolic products that can be trapped inside the highly anisotropic nanoscale pores of PGaN. The potential of this sampling mode is illustrated by the ability to acquire Raman spectra of adenine down to 5 fmol. Additionally, correlated SERS and laser desorption/ionization mass spectrometry spectra can be acquired from same sample spot without further preparation, opening new possibilities for the investigation of surface‐bound molecules with substantially enhanced information content. The Raman and surface‐enhanced Raman scattering (SERS) spectra of l ‐proline (Pro) and trans‐4‐hydroxy‐ l ‐proline (Hyp) were recorded. SERS spectra were obtained on colloidal Ag prepared by reduction with hydroxylamine. Allowing sufficient time for Pro and Hyp to adjust in the colloidal solution resulted fundamentally in obtaining unique and reproducible SERS spectra. Hyp stabilizes on the surface more rapidly than Pro. The spectral analysis indicates that Pro interacts with the Ag surface through the carboxylate group. The interaction of Hyp with the metal surface occurs through the amino, methylene and carboxylate moieties of the molecule. The spectroscopic results are supported by quantum chemical calculations, performed using extended Hückel theory (EHT) of the title compounds interacting with an Ag cluster model. The assignment of the Raman bands was supported by a normal coordinate analysis performed through Becke, three‐parameter, Lee–Yang–Parr/6‐311 G* + calculations. Although conventional Raman, surface‐enhanced Raman (SERS) and tip‐enhanced Raman spectroscopy (TERS) have been known for a long time, a direct, thorough comparison of these three methods has never been carried out. In this paper, spectra that were obtained by conventional Raman, SERS (on gold and silver substrates) and TERS (in ‘gap mode’ with silver tips and gold substrates) are compared to learn from their differences and similarities. Because the investigation of biological samples by TERS has recently become a hot topic, this work focuses on biologically relevant substances. Starting from the TER spectra of bovine serum albumin as an example for a protein, the dipeptides Phe–Phe and Tyr–Tyr and the tripeptide Tyr–Tyr–Tyr were investigated. The major findings were as follows. (1) We show that the widely used assumption that spectral bands do not shift when comparing SER, TER and conventional Raman spectra (except due to binding to the metal surface in SERS or TERS) is valid. However, band intensity ratios can differ significantly between these three methods. (2) Marker bands can be assigned, which should allow one to identify and localize proteins in complex biological environments in future investigations. The highly fluorescent natural dye berberine can be easily identified in microscopic textile samples by surface‐enhanced Raman spectroscopy employing citrate‐reduced Ag colloid. The ordinary Raman (OR) and SERS spectra of berberine are presented and discussed in the light of a DFT calculation. Using FT‐Raman and FT‐SERS we could reliably compare relative intensity shifts and investigate the adsorption geometry of berberine on Ag nanoparticles. The significant enhancement in the FT‐SERS spectrum of the out‐of‐plane ring system bending deformation mode at 729 cm−1 relative to a group of in‐plane vibrations at around 1500 cm−1 was interpreted as evidence of a ‘flat‐on’ adsorption geometry. SERS was successfully used to identify berberine in silk fiber samples coated with colloidal Ag following a pretreatment with HCl vapor. The SERS method allowed us to detect berberine in a microscopic sample of a single silk fiber from a severely degraded and soiled 17th Century Chinese textile fragment. Detailed studies of the mechanism of surface‐enhanced (resonance) Raman spectroscopy (SE(R)RS), and its applications, place a number of demands on the properties of SERS scatterers. With large Raman cross‐sections, versatile synthetic chemistry and complete lack of fluorescence, free dipyrrins meet these demands but the Raman and SE(R)RS spectroscopy of free dipyrrins is largely unknown. The first study of the Raman spectroscopy of free dipyrrins is therefore presented in this work. The nonresonant Raman, resonant Raman and surface‐enhanced Raman spectra of a typical meso aryl‐substituted‐dipyrrin are reported. Absolute differential cross‐sections are obtained for excitation wavelengths in the near infrared and visible region, in solution phase and for dipyrrin adsorbed on the surface of silver nanoparticles. Raman enhancement factors for SERRS and resonance Raman are calculated from the observed differential cross‐sections. The magnitudes of the resonantly enhanced cross‐sections are similar to those recently reported for strong SERS dyes such as Rhodamine 6G and Crystal Violet. Free dipyrrins offer the advantages of existing SERS dyes but without the drawback of strong fluorescence. Free dipyrrins should therefore find applications in all areas of Raman spectroscopy including fundamental studies of the mechanisms of SERS and bioanalytical and environmental applications. The Raman and surface‐enhanced Raman spectra (SERS) of flavone and three of its hydroxy derivatives, 3‐hydroxyflavone (3‐HF) and 5‐hydroxyflavone (5‐HF) and quercetin (3,5,7,3′,4′ pentahydroxyflavone) have been obtained. The normal Raman (NR) spectra were taken in the powder form. The SERS spectra were obtained both on Ag colloids and Ag electrode substrates. Assignments of the spectrally observed normal modes were aided by density functional theory (DFT) calculations using the B3LYP functional and the 6‐31 + G* basis, a split valence polarized basis set with diffuse functions. Excellent fits were obtained for the observed spectra with little or no scaling. The most intense lines of the NR spectra are those in the CO stretching region (near 1600 cm−1). These lines are often weakened by proximity to the surface, while other lines at lower wavenumbers, due to in‐plane ring stretches, tend to be strongly enhanced. The SERS spectrum of flavone is weak both on the colloid and on the electrode, indicating weak attachment to the surface. In contrast, the SERS spectra of the hydroxy derivatives of flavone are intense, indicating the assistance of OH groups in attachment to the surface. The spectra of the various species are compared, and a case study of application to detection of a textile dye (Persian berries), which contains quercetin, is presented. Thiacloprid is a widely used pesticide belonging to the neonicotinoid class, which is characterized by a selective activity against insects and a reduced acute toxicity for humans. The importance of the environmental impact of neonicotinoids is being intensively researched, in order to evaluate the danger they pose for useful insects. Physical methods which allow the characterization of neonicotinoids in diluted aqueous solutions are therefore desirable. We present a study of Raman and surface‐enhanced Raman scattering (SERS) spectroscopy on thiacloprid in solid state, in acetone solution, and adsorbed onto silver and gold hydrosols at μM concentration. Density functional theory calculations allow the individualization of the most stable molecular structure, both in gas phase and in solution, and of the corresponding Raman spectra. The vibrational assignments lead to an interpretation of the differences between SERS and ordinary Raman spectra based on the possible interactions between the molecule and the metal surface, the main one involving the iminocyano group. Formation of a charge‐transfer complex is suggested by the dependence of the SERS spectra on the laser excitation wavelength. We have been able to observe the surface‐enhanced Raman scattering (SERS) from 4‐mercaptopyridine (4‐Mpy) molecules adsorbed on ZnO nanocrystals, which display 103 enhancement factors (EFs). An excitation wavelength‐dependent behavior is clearly observed. Another molecule BVPP is also observed to have surface‐enhanced Raman signals. The chemical enhancement is most likely responsible for the observed enhancement, since plasmon resonances are ruled out. The research is important not only for a better understanding of the SERS mechanism, but also for extension of the application of Raman spectroscopy to a variety of adsorption problems on a semiconductor surface. The detection of explosives and their associated compounds for security screening is an active area of research and a wide variety of detection methods are involved in this very challenging area. Surface‐enhanced Raman scattering (SERS) spectroscopy is one of the most sensitive tools for the detection of molecules adsorbed on nano‐scale roughened metal surface. Moreover, SERS combines high sensitivity with the observation of vibrational spectra of species, giving complete information on the molecular structure of material under study. In this paper, SERS was applied to the detection of very small quantities of explosives adsorbed on industrially made substrates. The spectra were acquired with a compact Raman spectrometer. Usually, a high signal‐to‐noise (S/N) spectrum, suitable for identification of explosive molecules down to few hundreds of picograms, was achieved within 30 s. Carbendazim (MBC) is a fungicide widely used in agriculture, and there are serious concerns regarding the health risks that could be caused by this fungicide. Here, we explore its ultrasensitive detection by surface‐enhanced Raman scattering (SERS). First, to obtain maximum SERS signal, the adsorption of the target molecule onto metallic surface is essential. Therefore, we study the adsorption of the MBC onto the nanoparticle surface by SERS under different experimental conditions, such as different synthesis methods of nanoparticle, variable excitation wavelength, and fungicide concentration with the aim to detect MBC at low concentrations. Experiments are carried out with three kinds of colloidal nanoparticles: Ag and Au reduced by citrate and Ag reduced by hydroxylamine. However, mainly Ag colloids are highly efficient in the SERS detection of MBC. In addition, theoretical calculations of MBC Raman spectrum and that of the surface complex are used to help with the understanding the mechanisms responsible for the interaction between MBC and Ag. 提出一种新型的荧光及表面增强拉曼散射(SERS)双模式光学纳米探针。首先,通过再沉淀-包覆法合成二氧化硅包覆香豆素6的纳米颗粒,再在二氧化硅表面静电吸附多聚赖氨酸分子形成包覆层,随后通过原位还原的方法在多聚赖氨酸壳层复合银纳米颗粒,最后在银纳米颗粒表面吸附拉曼分子即形成双模式纳米探针。该探针通过二氧化硅包覆的荧光分子产生荧光信号,以多聚赖氨酸表面的银纳米颗粒作为SERS增强基底,利用拉曼分子获得SERS信号,实现了荧光及SERS双模式成像。荧光与表面增强拉曼散射相结合的双模式分析技术可同时发挥二者的优点,提高成像的分辨率和灵敏度,在生物医学领域具有重要的应用价值。  相似文献

Surface‐enhanced Raman scattering (SERS) spectra are presented and analyzed for two important organophosphate pesticides, dimethoate (DMT) and omethoate (OMT). Very detailed SERS spectra were obtained by aggregated Ag hydrosols, both in aqueous suspension and dried on a glass substrate. The SERS and ordinary Raman spectra of DMT do not resemble each other, suggesting that a chemical reaction immediately occurs when DMT is adsorbed onto the metal surface. We propose that the reaction product is OMT, which is the oxygen analog of DMT, on the basis of the Raman and SERS spectra of OMT. Further support is derived from the calculated Raman spectra of DMT and OMT. Minor wavenumber and intensity differences that are observed between the SERS spectra of DMT reaction product and those of OMT could be related to different metal/adsorbate interaction modes. Quaternary protoberberine alkaloids are a class of natural dyes characterized by bright colors ranging from yellow to orange. As they present a strong fluorescence emission, their analysis by Raman spectroscopy is limited to specific techniques such as Fourier transform (FT)‐Raman and spectral shift Raman techniques such as shifted subtracted Raman difference spectroscopy (SSRDS) and shifted excitation Raman difference spectroscopy (SERDS). In a previous article, we successfully used surface‐enhanced Raman scattering (SERS) in the analysis of the alkaloid dye berberine in an ancient textile. The examination of the Raman and SERS spectra of berberine in combination with density functional theory (DFT) calculations indicated a flat adsorption geometry of the molecule on the Ag surface. In this article we extend that work to the study of related protoberberine alkaloids, palmatine, jatrorrhizine, and coptisine. The same adsorption geometry as in berberine was deduced. We found that the four alkaloids, although minimally different in their chemical structures, could be differentiated by the position of marker bands. Those bands are the most enhanced ones in the SERS spectra, which appear in the 700–800 cm−1 region. Copyright © 2008 John Wiley & Sons, Ltd.  相似文献

Combining membrane electrophoresis with surface‐enhanced Raman scattering (SERS) spectroscopy, the serum proteins were first purified and then mixed with silver nanoparticles to perform SERS spectral analysis. Therefore, the spectral signatures were enhanced to high‐fidelity SERS signatures because of the purification procedure of the first step. We used the method to analyze blood plasma samples from nasopharyngeal cancer patients (n = 43) and healthy volunteers (n = 33) for cancer detection. Principle component analysis of the SERS spectra revealed that the data points for the cancer group and the normal group form distinct, completely separated clusters with no overlap. Therefore, the nasopharyngeal cancer group can be unambiguously discriminated from the normal group, i.e., with both diagnostic sensitivity and specificity of 100%. These results are very promising for developing a label‐free, noninvasive, and reliable clinical tool for rapid cancer detection and screening. Copyright © 2011 John Wiley & Sons, Ltd.  相似文献
